Dicarboximide fungicides have been used for the control of grey mould in protected crops in Crete since 1977. During the 1980 growing season a decline of their efficacy was observed. In successive surveys carried out in May 1980, February 1981 and May 1981 in 28, 10 and 13 plastic houses repectively, a considerable proportion of resistant strains was found. From each of the plastic houses sampled mostly either only resistant or only sensitive strains were isolated. In three of the plastic houses with resistant strains there was an acute disease control problem. The ED50 of 15 resistant strains studied was in the area of 3·5 μg/ml vinclozolin as compared with 0·2 μg/ml for the wild type strains. The vinclozolin-resistant strains were also resistant to procymidone, iprodione, and dicloran. In most of the cases strains resistant to vinclozolin were also resistant to benomyl and strains sensitive to vinclozolin were also sensitive to benomyl. In the absence of fungicide, resistant strains grew more slowly on PDA than sensitive ones, but spores germinated equally well. Vinclozolin (0·75 mg a.i./ml) did not protect eggplant seedlings against resistant strains but gave satisfactory control of sensitive ones.  相似文献

Dicarboximide fungicides are widely used to control various fungal species. Their primary action is not known, due to a lack of knowledge concerning the mechanism of action of the dicarboximide group. The cytotoxicities of vinclozolin and iprodione in rat hepatoma-derived Fa32 cells were investigated. Cytotoxicity was measured by neutral red uptake inhibition after treatment for 24 hours. Iprodione was more toxic than vinclozolin. Vinclozolin was less toxic in glutathione-depleted cells than in control cells. This was also true for iprodione at lower concentrations, but iprodione became more toxic at higher concentrations. Both the fungicides increased the endogenous glutathione content by 20% after 1 hour. After 24 hours, the glutathione content was doubled by vinclozolin, but was not affected by iprodione. No effect on glutathione S-transferase activity or reactive oxygen species formation could be observed. Cytochrome P450-dependent ethoxyresorufin-O-deethylase and pentoxyresorufin-O-depentylase activities were moderately activated by iprodione and strongly activated by vinclozolin. A glutathione-related cytochrome P450-dependent metabolic attack of vinclozolin and iprodione could be responsible for their cytotoxicity in Fa32 cells. Further research is needed to fully elucidate these (or other) mechanisms.  相似文献

Smoulder, caused by Botrytis narcissicola, is the most widespread foliar disease of narcissus (Narcissus cultivars) in the UK and causes significant losses in bulb and flower yield. A range of current and novel fungicide treatments was examined to determine if control of the disease could be improved and to measure the effect of treatments on bulb yield. In assays on pot-grown plants, carbendazim+flusilazole, epoxiconazole, iprodione + thiophanate-methyl, tebuconazole and vinclozolin reduced lesion size by more than 90%, compared with untreated plants, when applied 1 day before inoculation with the fungus; vinclozolin and tebuconazole were also effective when applied 2 days after inoculation. In field experiments in Cambridgeshire and Lincolnshire, significant reductions were observed in secondary smoulder symptoms (leaf lesions and stem rot) following fungicide sprays. Spray programmes of the novel fungicides azoxystrobin, cyprodinil, kresoxim-methyl, tebuconazole and pyrimethanil were as effective, or more so, than the standard treatments (chlorothalonil, vinclozolin) used by growers. Mixtures of carbendazim + tebuconazole and azoxystrobin + tebuconazole also gave effective control of smoulder. Programmes of four to six sprays, using two or three fungicides with different modes of action, applied alternately, reduced smoulder by 35–69% and increased bulb yields by 7–59%. Although treatment around and after flowering resulted in the greatest control of secondary smoulder, treatment before flowering was required for the highest bulb yields. The choice of fungicides for use in spray programmes to provide effective and reliable control of smoulder, and the possibility of control using fewer sprays, are discussed.  相似文献

Lettuce drop caused by Sclerotinia minor Jagger is the most common disease affecting lettuce crops during the fall and winter in Roussillon. Spraying of cyclic imide fungicides (iprodione, vinclozolin, and less commonly procymidone) generally provides satisfactory protection. However, since about 1985, the mean field efficacy of iprodione and vinclozolin has significantly decreased, whereas that of procymidone is currently not in question. The appearance of fungicide-resistant isolates of S.minor could explain the reduced efficacy of these fungicides. We thus looked for iprodione and procymidone-resistant isolates of S.minor in these problem fields. The investigations were earned on for over 3 years. Most of the isolates showed fungicide sensitivity (iprodione IC 50 = 0.17 mg 1−1), about one quarter showed average resistance (iprodione IC 50 = 0.59 mg 1−1) and a few were found to be highly resistant. Resistant isolates were also obtained in vitro when using high fungicide concentrations. Moreover, isolates resistant to iprodione were found to be resistant to procymidone. The most highly resistant isolates produced fewer and larger-sized sclerotia than the sensitive isolates, they were also more quickly inhibited by high osmotic pressures. Field variability was quite low: 73 of 74 isolates obtained from one field were found to be sensitive, with only one showing mild resistance. There did not seem to be any correlation between the observed decrease in the field efficacy of iprodione and the presence of resistant isolates.  相似文献

The effect of different dosages of thymol alone, iprodione alone and combinations of thymol and iprodione on white rot disease of garlic and its impact on soil microbial community structure were investigated under greenhouse conditions. Thymol alone or in combination with the fungicide iprodione did not appear to reduce either white rot incidence or soil sclerotia density as compared to an infected control. However, iprodione alone or in combination with thymol reduced soil fungal biomass. In addition, iprodione alone decreased soil microbial activity as estimated by fluorescein diacetate (FDA). Soil bacterial community structure as estimated by phospholipid fatty acid (PLFA) profiles was also was affected by both thymol and iprodione applications. The correlation biplot of the individual PLFAs and biocide treatment indicated that the treatments with thymol alone increased cyclopropyl fatty acid (cy17:0 and cy19:0), while the treatments with iprodione alone increased some saturated and branched fatty acids (principally i16:0, a15:0 and 18:0). In addition, taking into account PLFA biomarkers, thymol applications reduced Gram-negative bacteria in soil. To our knowledge, this research is the first report about the effect of a monoterpene (thymol) on soil microflora.  相似文献

The new fungicides iprodione, vinclozolin and prochloraz, and also a mixture containing carbendazim and maneb, were compared with the established protectant fungicides dichlofluanid and chlorothalonil for effectiveness against grey mould ( Botrytis cinerea ) in an unheated tomato crop. Iprodione and vinclozolin gave the best control of lesions on leaves and stems but dichlofluanid was the most effective in controlling ghost-spotting of fruit. The carbendazim/maneb mixture was effective against a carbendazim-sensitive isolate but not against an insensitive isolate of B. cinerea . When applied 1 or 2 days after inoculation, all five protectant fungicides controlled the insensitive isolate better than the sensitive isolate.  相似文献

Catabolic dehydroquinase, which functions in the inducible quinic acid catabolic pathway of Neurospora crassa, has been purified from wild type (74-A) and three mutants in the qa gene cluster. The mutant strains were: 105c, a temperature-sensitive constitutive mutant in the qa-1 regulatory locus; M-16, a qa-3 mutant deficient in quinate dehydrogenase activity; and 237, a leaky qa-2 mutant which possess very low levels of catabolic dehydroquinase activity. The enzymes purified from strains 74-A, 105c, and M-16 are identical with respect to behavior during purification, specific activity, electrophoretic behavior, stability, molecular weight, subunit structure, immunological cross-reactivity, and amino acid content. The mutant enzyme from strain 237 is 1,500-fold less active and appears to have a slightly different amino acid content. It is identical by a number of the other criteria listed above and is presumed to be a mutant at or near the enzyme active site. These data demonstrate that the qa-1 gene product is not involved in the posttranslational expression of enzyme activity. The biochemical identity of catabolic dehydroquinase isolated from strains 105c and M-16 with that from wild type also demonstrates that neither the inducer, quinic acid, nor other enzymes encoded in the qa gene cluster are necessary for the expression of activity. Therefore the combined genetic and biochemical data on the qa system continue to support the hypothesis that the qa-1 regulatory protein acts as a positive initiator of qa enzyme synthesis.  相似文献

Bacillus licheniformis N1 is a biological control agent to control gray mold diseases caused by Botrytis cinerea. Various formulations of B. licheniformis N1 were generated and evaluated for the activity to control strawberry gray mold. The wettable powder type formulation N1E was selected in pot experiments with remarkable disease control activity on both strawberry leaves and flowers. The N1E formulation contained 400 g of corn starch, 50 ml of olive oil, and 50 g of sucrose per a liter of bacterial fermentation culture. Optimum dilution of N1E to appropriately control the strawberry gray mold appeared to be 100-fold dilution in plastic house artificial infection experiments. The significant reduction of symptom development in the senescent leaves was apparent by the treatment of N1E at 100-fold dilution when N1E was applied before Bo. cinerea inoculation, but not after the inoculation. Both artificial infection experiments in a plastic house and natural infection experiments in the farm plastic house under production conditions revealed that the disease severity of gray mold on strawberry leaves and flowers was significantly reduced by N1E treatment. The disease control value of N1E on strawberry leaves was 81% under production conditions, as compared with the 61.5% conferred by a chemical fungicide, iprodione. This study suggests that our previously generated formulation of B. licheniformis N1 will be effective to control strawberry gray mold by its preventive activity.  相似文献

Fludioxonil is a new non-systemic phenylpyrrole fungicide. It is a derivative of the antibiotic pyrrolnitrin and is highly active against Ascomycetes. Basidiomycetes and Deuteromycetes. In the present study we determined the baseline sensitivity of B. fuckeliana for fludioxonil in comparison to the dicarboximide vinclozolin. Subsequently these baselines were compared with the sensitivity of dicarboximide-resistant field isolates and with resistant laboratory strains selected on media amendedwith vinclozotin or fludioxonil, respectively. Additionally, sensitivities of genetically homogeneous ascospore progenies of sexual crosses of sensitive isolates with dicarboximide-resistant field isolates or resistant laboratory strains were compared to, the baselines. Baseline ECst, values for the inhibition of germination ranged from 0.07 to 0.20 μg ml-1 of vinclozolin and for mycelial growth from 0.003 to 0.016 μg ml’ of fludioxonil and from 1.7 to 2.0 μg ml-1 of vinclozolin and for mycelial growth from 0.003 to 0.016 μg ml-1 of fludioxonil and from 0.13 to 0.27 μg ml-1 of vinclozolin. Sensitivity to fludioxonil of all dicarboximide-resistant field isolates did not differ from their baseline. However, resistant laboratory strains that were selected either on media amended with fludioxonil or vinclozolin showed decreased sensitivities to both active ingredients. All sexual progenies of the crosses described above were of the parental phenotypes. We found no independent segregation of the resistance markers.  相似文献

The stability of microsatellite markers was investigated in the spore-producing fungus Botrytis cinerea exposed to four growth conditions. This knowledge is essential in order to differentiate mutations from genetic exchanges or recombination in population genetics studies. It is also important when using strains from collections that need to be regularly propagated on medium. Successive spore generations of four isolates of the fungus were realised in plates on different agar media: a nutrient-rich medium, a nutrient-poor medium, a medium supplemented with the antibiotic pyrrolnitrin and a medium supplemented with the fungicide iprodione. The stability of nine microsatellite markers was studied by comparing the molecular pattern obtained between the wild type parent strains and the final generations obtained. The results showed that, despite the phenotypic changes observed in some generations, no changes were observed in the allele size at nine microsatellite loci whatever the selective pressure endured by the fungus. This is the first study that reveals long-term stability of microsatellite markers of a spore-producing fungus exposed to different stresses.  相似文献

根据几种丝状真菌Hog1 MAPK(丝裂原活化蛋白激酶)的保守氨基酸序列设计简并引物,从番茄早疫病菌Alternaria solani中扩增出MAPK同源基因的部分片段,命名为AsHog1。用Real-timeRT-PCR技术比较了AsHog1在异菌脲敏感菌株和抗性菌株中的表达特性。结果表明,敏感菌株QX21经0.8mol/L NaCl和2mg/L异菌脲处理后,AsHog1相对表达量持续升高,在12h达到最大值,分别为对照的4.12倍和25.23倍,16h略有降低;而室内诱导抗性突变体和田间抗性菌株经相同处理后的AsHog1表达量变化不大,且明显低于敏感菌株。由此推测,番茄早疫病菌AsHog1基因表达特征与其对异菌脲抗药性相关。研究结果为深入研究病原菌对异菌脲抗药性的分子机理、建立抗药性分子监测技术、延缓和防止抗药性的产生及发展奠定了一定的理论基础。  相似文献

A ready-to-fog' formulation of iprodione applied with themal foging equipment was a satisfactory alternative to high-volume (HV) sparys of iprodione for the control ofnatural infecton with grey mould (Botrytis cinerea) in an unheated tomato crop. There were no significant differences in numbers of droped fruit nor in the control of naturally-occurring lesions in stems or leaves, nor of ghost-spotting on fruit, between the HV sprays and fogs applied every fortnight. Except for ghost-spotting, control by fogs applied weekly was better than by eihter for nightly treatment. In contrast, the control of infection from B.cinerea inoculations on peitoles was significantly better by the fortnightly HV treatment than by either frequency of foging. Control of infection imporved with increase in the number of fungicide applications to the peitole before inoculation; two HV sprays gave complete control but six fog applications did not. The variation in the conrrol of infection by fogs and presumbly therefore in the distribution of fungicide within the 61 m2 glasshouse compartments was small. Application of the iprodione fogging formulation significantly reduced the number of fruits see by about 5% compared with the HV treatment.  相似文献

A method based on solid-phase extraction (SPE) and high-performance liquid chromatography (HPLC) was developed for the simultaneous determination of 3-(3,5-diclorophenyl)-5-ethenyl-5-methyl-2,4-oxazolidinedione (vinclozolin) and 3-(3,5-diclorophenyl)-N-(1-methylethyl)-2,4-dioxo-1-imidazolidinecarboxamide (iprodione) in human urine. Urine samples containing vinclozolin and iprodione were collected by solid phase extraction using C(18) cartridges. The chromatographic separation was achieved on a Spherisorb ODS2 (250 mm x 4.6 mm, 5 microm) column with an isocratic mobile phase of acetonitrile-water (60:40, v/v). Detection was UV absorbance at 220 nm. The calibration graphs were linear from 30 to 1000 ng/mL for the two fungicides. Intra- and inter-day R.S.D. did not exceed 2.9%. The quantitation limit was 50 ng/mL for vinclozolin and 30 ng/mL for iprodione, respectively.  相似文献

The somatic recombinogenic activity of the phenylpyrrole fungicide fludioxonil, in diploid Aspergillus nidulans was found similar to that caused by aromatic hydrocarbon and dicarboximide fungicides (AHDFs), such as iprodione, chlozolinate and tolclofos–methyl. All these fungicides not only increased the number of mitotic recombinants but also provided similar appearance, small sectors, of white and yellow mitotic recombination products. Fludioxonil highly resistant strains (resistant factor approximately 5000) of Botrytis cinerea were isolated at high frequency (1.08 × 10−5). Study of cross-resistance patterns of mutant strains to other fungicides, revealed cross-resistance of fludioxonil with dicarboximides (iprodione, procymidone, and chlozolinate) and aromatic hydrocarbons, such as tolclofos–methyl, pentachloronitrobenzene (PCNB), tecnazene and chloroneb. The positive cross-resistance relationships found between phenylpyrroles and members of the AHDFs and their ability to increase mitotic instability in diploid A. nidulans , indicate that phenylpyrroles should be included with AHDFs. A study of fitness parameters in wild-type and representative fludioxonil-resistant mutants of B. cinerea , showed that the mutation(s) leading to fludioxonil resistance may or may not affect some fitness-determining characteristics, such as sensitivity to high osmolarity, growth rate, conidial germination and germ-tube elongation. Pathogenicity tests on cucumber seedlings showed that an osmosensitive representative strain of B. cinerea , resistant to fludioxonil, was as virulent as the wild-type strain. The phenylpyrrole fungicide was ineffective, even in high concentrations, to control grey mould caused by this isolate.  相似文献

Laboratory studies showed that mixtures of preparations of iprodione and carbaryl exerted a strong synergistic inhibitory effect on conidiospore germination of the insect-pathogenic fungus Verticillium lecanii. Such synergism was not observed with iprodione and pirimicarb, or vinclozolin and carbaryl. No synergistic effect was observed on the germination of Botrytis cinerea conidia on agar.  相似文献

Sclerotinia sclerotiorum is an important plant pathogen with worldwide distribution that causes severe economic losses of various agricultural crops such as soybean. This fungus is normally controlled with synthetic chemical fungicides that pose risks to the environment, and can be harmful to human health, and they can also induce resistance in pests. The aim of this study was to investigate the potential of Trichoderma asperelloides as a biocontrol agent towards white mold disease on soybeans crops. The antagonism of two strains of T. asperelloides (T25 and T42) isolated from soil samples was determined in-vitro by dual-culture confrontation testing on nine S. sclerotiorum strains obtained from sclerotia collected on diseased soybean plants. The mycelial growth and inhibition of carpogenic and ascospore germination by T. asperelloides extracts, as well as the efficacy of these on white mold control in soybeans were evaluated. Both strains of T. asperelloides exhibited high potential of antagonism. Methanolic and ethyl acetate extracts of the two T. asperelloides strains showed excellent growth inhibition (60–100%) on all of the pathogens tested. The ethyl acetate extracts of both T. asperelloides strains exhibited the highest efficacy against carpogenic germination, decreasing by 20–30% the number of ascospores per apothecium. Strains of T. asperelloides tested were more efficient in controlling white mold than two commercial products made from Trichoderma harzianum. The new strains of T. asperelloides have potential for successful biological control of white mold disease of soybean crops in the field.  相似文献

In vitro dosage response data with different isolates of Penicillium digitatum and the fungicide guazatine indicated an approximate 10-fold shift in tolerance when compared with wild type strains. ED50 values for resistant strains were approximately 0.5 μg/ml compared to approximately 0.05, μg/ml for the wild type strains. Colony growth of guazatine resistant isolates on selective media containing carbendazim showed that they were also resistant to the benzimidazole group of fungicides. In vivo tests in inoculated oranges with strains previously characterised by in vitro tests confirmed resistance to guazatine and benomyl. A combined treatment of these fungicides at 400 /μ/ml and 500 μg/ml respectively, which normally gives protection against decay, also failed to provide adequate mould control. Growth and pathogenicity of the resistant strains in these tests in oranges were indistinguishable from that of wild type strains.  相似文献

In this study, sensitivities of 156 Sclerotinia sclerotiorum isolates collected from sunflower fields of West Azarbaijan province, Iran, were assessed to carbendazim and iprodione, and the baseline sensitivities were established for azoxystrobin and tebuconazole. Resistance to carbendazim and iprodione was observed in 53.85% and 4.49% of the isolates, respectively. The 50% effective concentration (EC50) values of azoxystrobin for the isolates ranged from 0.017 to 3.515 μg/ml with a mean of 0.330 μg/ml, and 8.97% of the strains showed low levels of resistance to the fungicide. However, in the presence of salicylhydroxamic acid, all isolates were sensitive to azoxystrobin and EC50 values ranged from 0.015 to 0.263 μg/ml with a mean of 0.086 μg/ml. All isolates were found to be sensitive to tebuconazole, and EC50 values ranged from 0.003 to 0.177 μg/ml with a mean of 0.036 μg/ml. Among the multiple-resistant isolates, the strains exhibiting resistance to both carbendazim and iprodione were detected in the highest frequency (4.49%). No correlation was observed between mycelial growth and aggressiveness with fungicide sensitivity of the isolates suggesting the absence of fitness cost associated with resistance to the studied fungicides. The results indicated that iprodione, azoxystrobin and tebuconazole could be effectively used in rotation or mixture in spray programmes to manage S. sclerotiorum in the region. The baselines established for azoxystrobin and tebuconazole would be useful in monitoring the fungal populations in the province to assess possible shifts in fungicide sensitivity of S. sclerotiorum isolates in the future.  相似文献

Vinclozolin, an endocrine disrupting chemical, is a chlorinated fungicide widely used to control fungal diseases. However, its metabolite 3,5-dichloroaniline is more toxic and persistent than the parent vinclozolin. For the biodegradation of vinclozolin, vinclozolin- and/or 3,5-dichloroaniline-degrading bacteria were isolated from pesticide-polluted agriculture soil. Among the isolated bacteria, a Rhodococcus sp. was identified from a 16S rDNA sequence analysis and named Rhodococcus sp. T1-1. The degradation ratios for vinclozolin or 3,5- dichloroaniline in a minimal medium containing vinclozolin (200 microg/ml) or 3,5-dichloroaniline (120 microg/ml) were 90% and 84.1%, respectively. Moreover, Rhodococcus sp. T1-1 also showed an effective capability to biodegrade dichloroaniline isomers on enrichment cultures in which they were contained. Therefore, these results suggest that Rhodococcus sp. T1-1 can bioremediate vinclozolin as well as 3,5-dichloroaniline.  相似文献

Summary Eight ineffective mutant strains were isolated from N-methyl-N'-nitro-N-nitrosoguanidine mutagenized cultures of cowpea Rhizobium strain 32H1. Strains CR1, CR2, CR3, CR4, CR5 and CR6 induced more, but smaller, nodules than the wild type. With the exception of strain CR2, these mutant strains reduced less than 1% of the amount of acetylene reduced by the wild type, in both the free-living and symbiotic assays. Strain CR2 reduced acetylene in the free-living assay but not in the symbiotic assay. Strains CR7 and CR8 responded variably (5–20% of the wild type) in free-living and symbiotic acetylene reduction assays. Nodules also varied from small white to normal-sized pink nodules. The phenotypic characteristics of the mutant strains were consistant with all leguminous plants tested and were stable upon reisolation from nodules. Fully effective revertants were selected from 4 of the ineffective mutant strains by the use of the leguminous plant,Macroptilium lathyroides.
