The fine structure of the red cells in Arabian camels was investigated and certain characteristic features were noted. The plasmalemma of camel erythrocytes are tri-laminar, the inner and outer membranes are of high electron density between which is a zone of lesser electron density. No intracellular organelles were observed with the occasional exception of a small number of mitochondria. In the camel erythrocyte, a marginal band consisting of 30-45 microtubules was observed in many cells. Some of the possible functions of the marginal band in camel erythrocytes are discussed.  相似文献

The granular leucocytes of an active, mature female tuatara, Sphenodon punctatus (Gray) were examined in the electron microscope. Eosinophils contained a lobulated nucleus, homogeneous, dense, irregularly shaped granules, assorted smaller granular inclusions, mitochondria and beta-glycogen. Endoplasmic reticulum, Golgi complexes and ribosomes were scanty. Immature neutrophils (myelocytes) were regular in outline and contained a compact nucleus. In the adjacent centrosomal region were paired centrioles with connected microtubules, and Golgi complexes. Ovoid electron-dense granules, mitochondria, lipid droplets and numerous microfilaments arranged randomly or in bundles, lay in the cytoplasm. Mature neutrophils were often highly irregular in outline, had a segmented nucleus and contained possibly a second type of granular inclusion. The basophils were regular in outline with a compact nucleus. Numerous ovoid homogeneous, electron-dense granules, mitochondria, beta-glycogen particles and some microfilaments were seen in the cytoplasm. The granules in many basophils appeared 'altered' or degenerate and most of these contained microtubules. The cytology of the granulocytes of the tuatara is compared with that in other vertebrates.  相似文献

An electron microscopic study revealed that during aggregation the cytoplasm of a number of cells increases in electron density. Increased electron density is shown to be the consequence of cell shrinkage, which causes a closer packing of cytoplasmic components. Originally electron-dense cells are spread randomly over the aggregate. The anterior prestalk region of the slug is almost devoid of electron-dense cells. In the posterior prespore region, cells with varying degrees of electron density are intermixed with 15–20% electron-lucent cells. During culmination all cells of the prespore region become very electron dense. Besides introducing a new criterion to recognize prespore cells at an early stage of development, our data give further evidence that induction of prespore cell differentiation is not necessarily position dependent.  相似文献

Summary This study reconstructed heterogeneous embryos using camel skin fibroblast cells as donor karyoplasts and the bovine oocytes as recipient cytoplasts to investigate the reprogramming of camel somatic cell nuclei in bovine oocyte cytoplasm and the developmental potential of the reconstructed embryos. Serum-starved skin fibroblast cells, obtained from adult camel, were electrically fused into enucleated bovine metaphase II (MII) oocytes that were matured in vitro. The fused eggs were activated by Inomycin with 2 mM/ml 6-dimethylaminopurine. The activated reconstructed embryos were cocultured with bovine cumulus cells in synthetic oviduct fluid supplemented with amino acid (SOFaa) and 10% fetal calf serum for 168 h. Results showed that 53% of the injected oocytes were successfully fused, 34% of the fused eggs underwent the first egg cleavage, and 100% of them developed to four- or 16-cell embryo stages. The first completed cleavage of xenonuclear transfer camel embryos occurred between 22 and 48 h following activation. This study demonstrated that the reconstructed embryos underwent the first embryonic division and that the reprogramming of camel fibroblast nuclei can be initiated in enucleated bovine MII oocytes.  相似文献

Oocytes from the land hermit crab, Coenobita clypeatus, in various stages of vitellogenesis were examined by light and electron microscopy. Early vitellogenic oocytes are characterized by accumulations of discrete vesicles of endoplasmic reticulum in the perinuclear cytoplasm. As oocytes develop, the endoplasmic reticulum becomes abundant, and numerous Golgi complexes are seen. There is a well developed Golgi-endoplasmic reticulum interaction. Within the confines of the reticulum are discrete intracisternal granules, which can be seen coalescing into electron-dense yolk bodies. Lipid accumulation is seen throughout the cytoplasm. Coincident with the burst of intra-oocytic metabolism are oolemma modifications and micropinocytosis, which provide ultrastructural evidence for extra-oocytic yolk production. The mature oocyte contains numerous yolk and lipid vesicles of varying electron density that comprise both intra- and extra-oocytic substrates.  相似文献

The circulating amoebocytes of the freshwater mussels, Indonaia caerulia and Parreysia favidens have been studied. The total number of amoebocytes is low and variable, and the individual to individual variation in total counts is particularly significant. The amoebocytes are of 3 main types: Acidophils, Large Basophils and Small Basophils. Acidophils contain large acidophilic granules and basophils fine basophilic granules within the cytoplasm. Acidophils are the largest and small basophils the smallest. The greater volume of the cell is occupied by cytoplasm in acidophils and large basophils and by the nucleus in small basophils. Large basophils are the most and small basophils the least abundant. Amoebocytes clump together by means of filipodia. Large basophils possess these protoplasmic processes in the greatest abundance, and partake most actively and in largest numbers in clumping; while small basophils lack filipodia and play an absolutely passive role. Clumping progressively becomes more extensive with passage of time following cardiac injury inflicted during blood collection. I. caerulia appears to be generally more efficient than P. favidens with regard to clumping reaction. The different structural, numerical and behavioural aspects of the amoebocytes have been considered with reference to their probable causative factors and functional import.  相似文献

We used ultrastructural autoradiographic and cytochemical methods to localize esterase activities in unstimulated guinea pig basophils and in basophils undergoing degranulation or recovery from degranulation. We used tritium-labeled diisopropylfluorophosphate (DFP) as a probe for serine enzymes and localized this probe by ultrastructural autoradiography to cytoplasmic granules of immature or mature unstimulated basophils, as well as to granules released by degranulating basophils. Ultrastructural cytochemistry using alpha naphthyl acetate (ANA) as substrate localized nonspecific esterase activity to extruded granules, either within the interiors of degranulation sacs or within granules completely separated from degranulating basophils. Extruded granules retained their esterase activity for as long as 24 hr after antigen-induced degranulation. The plasma membranes of unstimulated or degranulating basophils, as well as of basophils recovering from degranulation, displayed prominent cell surface ANA esterase ectoenzyme activity. Lipid bodies, organelles present in the cytoplasm of both control and recovering basophils, were also alpha naphthyl acetate esterase (ANAE)-positive. Thus, cytochemical and autoradiographic techniques localized esterase and/or [3H]-DFP-binding activities to cytoplasmic granules, lipid bodies, and cell surface of basophils, and these enzyme activities persisted during both degranulation and recovery from degranulation.  相似文献

Spermatogenesis was studied with the aid of the light and electron microscopes in fourteen sexually mature camels slaughtered at different times of the year. The testes were fixed by vascular perfusion with glutaraldehyde. Spermatogenesis in the camel was generally similar to that of most mammalian species, although some features specific for the camel were observed. Spermatogenesis was found to be continuous throughout the year.  相似文献

Intracellular localization of serotonin (5-HT) in the mast cells of two phenotypes in normal rat colon and dextran sodium sulphate-induced colitis was studied by immunoelectron microscopy with a quantitative analysis of the distribution of immunogold labelling. Mucosal mast cells in normal rats contained round shape secretory granules with varying electron density. Immunogold labelling for 5-HT was concentrated over the secretory granules. In mucosal mast cells from colitis rats, vacuolated granules without 5-HT labelling were frequently observed and immunogold labelling over the secretory granules was significantly increased compared to controls. On the other hand, connective tissue mast cells in normal rats contained oval shape secretory granules with homogeneous electron density. Their immunogold labelling was diffusely scattered over the secretory granules as well as over the cytoplasm. In connective tissue mast cells from colitis rats, secretory granules with high electron density were increased and the immunogold labelling over the secretory granules was much higher than that in controls. The present results suggest that intracellular localization of 5-HT is different in two phenotypes of mast cells and they may release 5-HT in a different manner. Mucosal mast cells may release 5-HT by a degranulation or exocytosis, while connective tissue mast cells may release 5-HT by a diacrine manner of secretion.  相似文献

Cytochemically peroxidase activity has been examined on the light optical and ultrastructural levels in blood granulocytes of the rabbits after a single (5 mg/kg) and multiple (1 and 5 mg/kg every 24 hrs during 4 weeks) administrations of hydrocortisone. Under electron microscope peroxidase activity was detected in the blood of intact rabbits into typical primary granules (TPG) and small polymorphic granules (SPG) of neutrophils as well as into specific granules of basophils sometimes in perinuclear space and GER channels. 6 h after hydrocortisone injection peroxidase activity in neutrophils increased, the reaction product in both kinds of cytoplasmic granules was electron denser than in the controls. After multiple hydrocortisone (1 mg/kg) administrations peroxidase general activity in granulocytes has not considerably changed, but the number of TPGs and SPGs was decreased in neutrophils. Multiple administrations of a higher dose of hydrocortisone (5 mg/kg) have induced peroxidase activity decreasing in neutrophils and a decrease in the number and electron density of TPGs and SPGs in them. In basophils there was a significant accumulation of the reaction product of high electron density in perinuclear space, in specific granules and GER channels. The conclusions has been drawn that a short-term raising of hydrocortisone level stimulates and prolonged hypercorticism inhibits peroxidase activity in neutrophils and, consequently, their function.  相似文献

The nasal septal island (NSI) is a sensory patch of neuroepithelium located within the soft tissue of the nasal septum in dromedaries. The island has unique anatomical features, including the specialized subepithelial glands. The aim of the present study was to describe the microscopic features and ultrastructure of these subepithelial glands and to speculate the possible functions. A total of 10 camel heads were used for the study. Unlike the serous and mucous airway glands, the NSI glands’ ultrastructural features were typical for cells of the (Amine Precursor Uptake and Decarboxylation, APUD) system. These features were included, membrane bound secretory vesicles of varying electron density, smooth endoplasmic reticulum in the form of vesicles; electron dense mitochondria, abundant rough endoplasmic reticulum and free ribosomes. Alcian-PAS identifiable mucus granules were not observed, except for few clusters of cells, located at the luminal surface. The probable functions were discussed on basis of cellular morphology and context. In a conclusion, the NSI subepithelial glands in dromedaries had unique anatomical structures, and as many other APUD cells, they had the machinery required for synthesis of a variable number of biologically active peptides, amines and chemical mediators.  相似文献

The cardiac and pyloric glands in the gastric mucosa of the South African hedgehog, Atelerix frontalis, are described. The cardiac area of the stomach contains proper cardiac glands and lacks undifferentiated fundic glands. The cardiac glands are simple tubular, coiled, and lined with columnar cells ultrastructurally similar to those of the gastric surface epithelium. Secretory granules with varying electron densities fill the apical cytoplasm of these cells. In contrast to other mammals, these glands lack mucous neck cells. The neck of the pyloric glands contains only a single cell type, whereas the basal regions of these glands contain “light” and “dark” cells. The secretory granules in the “dark” cells and the pyloric neck cells have a moderate electron density and often contain an electron dense core. An electron-lucent cytoplasm with numerous polysomes is characteristic of the “light” cells. Some “light” cells contain electron-dense granules in the apical cytoplasm. The presence of only neutral mucins in the cardiac gland cells denotes the absence of mucous neck cells. The acidic mucins within the pyloric neck cells seem to indicate that these cells are mucous neck cells, whereas the neutral mucins within the basally located pyloric gland cells show at least a partial functional difference from the pyloric neck cells. © 1993 Wiley-Liss, Inc.  相似文献

This study reconstructed heterogeneous embryos using camel skin fibroblast cells as donor karyoplasts and ovine oocytes as recipient cytoplasts for investigating the developmental potential of the reconstructed embryos. Serum-starved adult camel skin fibroblast cells were used as donor somatic cells. Ovine oocytes matured in vitro were employed as recipient cytoplasts. The fusion of fibroblast cells into recipient cytoplasm was induced by electrofusion. The fused oocytes were activated by 5mM/ml inomycin with 2mM/ml 6-dimethylaminopurine (6-DMAP). The activated reconstructed embryos were co-cultured with ovine cumulus cells in synthetic oviduct fluid supplemented with amino acid (SOFaa) and 10% fetal calf serum (FCS) for 168h. A total of 300 enucleated ovine oocytes were available for xenonuclear embryo reconstruction. The results showed that 71% of the nuclear transfer couplets were successfully fused, 55% of the fused oocytes cleaved within 48h after activation, 82% of the cleaved oocytes developed to 2-16-cell embryo stages and 18% of the cleaved nuclear transfer zygotes developed to the morula stage. This study demonstrated that the xenonuclear transfer camel embryos can undergo the first embryonic division and subsequent development to morula stage in vitro.  相似文献

The formed elements of the blood of L. corrianus are comprised exclusively of amoebocytes, whose total number is approximately 758 per cubic mm of blood. The blood cells are of three types, viz. acidophils, large basophils and small basophils, which are respectively 26.32%, 60.20% and 13.48% of the total blood cell population. The cytoplasm of the acidophils is filled with large acidophilic granules, while that of the basophils contains fine basophilic granules. Acidophils are the largest (7.6 μ × 6.7 μ), large basophils are smaller (6.5 μ × 4.9 μ), and small basophils are the smallest (4.1 μ × 3.8 μ). The nucleus occupies a very small volume of the cell (approx. 1/9th) in acidophils, a larger volume (approx. 1/4th) in large basophils, and a major volume (approx. 3/4th) in small basophils. The blood cells show amoeboid movement, which is brought about by means of broad lamellate pseudopodia. Large basophils move very slowly, acidophils move faster, and small basophils move quite fast. Fine bristle-like filipodia help the blood cells in getting entangled to form clumps. Filipodia are more abundant in large basophils and acidophils which pertake more actively in clumping than small basophils which rarely possess filipodia. Clumping is a reversible process which is promoted by mechanical agitation of blood.  相似文献

Icosahedral viral particles were found in the cytoplasm of erythrocytes and splenic reticular cells of a marine toad (Bufo marinus) collected from Costa Rica. Capsids had a maximum diameter of 312 nm and a spherical core with biphasic electron density. Viruses in erythrocytes were associated with cytoplasmic assembly areas and vacuoles in cytoplasm. Nuclei had finely granular material of decreased electron density located centrally, but contained no viral particles. A group of unenveloped viral particles was seen extracellularly in a splenic vessel. The virus was consistent with an iridovirus. In a blood smear stained with Giemsa round basophilic bodies with average diameters of 1.70 microns and morphologically similar to Pirhemocyton sp. were seen in the cytoplasm of erythrocytes and occasionally in the cytoplasm of monocytes or extracellularly. Erythrocytes containing these bodies had vacuoles and irregular pale-staining areas in the cytoplasm and pale-staining areas in the nucleus. These changes corresponded to the viral particles, assembly areas, vacuoles and nuclear changes at the ultrastructural level.  相似文献

Large cells, generally thought to be associated with neoplastic disorders in bivalve mollusks, were studied with the electron microscope. The atypical cells had an average diameter of 15 μm and a nuclear to cytoplasmic ratio of about 1–1.5. Nuclei were extensively pleomorphic and bizarre shapes were common. Nucleoli were prominent and often multiple. In the cytoplasm of the large cells, there was a wide range of variability in anomalous organelles. Two cell types were originally thought to exist; however, it is now thought that the two cell types represent the two extreme morphological expressions of a single cell line. Their varying appearance is correlated with the density of ribosomes and abundance of cellular organelles. The large Mytilus edulis cells possess many ultrastructural properties that are characteristic of certain malignant vertebrate cells. However, alternative explanations for their structure and function are also possible.  相似文献

Deposits of bile in the cytoplasm of hepatocytes from six patients with cholestasis were studied by electron microscopy and ultrastructural cytochemistry. The deposits were identified in vacuoles of varying size and complexity. The demonstration of acid phosphatase activity in these vacuoles suggests that lysosomes participate in the elimination of bile deposits retained in hepatocytes.  相似文献

Selective growth of human basophilic granulocytes was obtained in suspension cultures of mononuclear cells from umbilical cord blood. Approximately 50 to 80% of nonadherent cells recovered from 2- to 3-wk-old cultures contained metachromatic granules, and these cells were identified as human basophilic granulocytes by electron microscopy. Histamine content of cultured human basophils was comparable to that in peripheral blood basophils. Cultured basophils bear 2.7 to 3.7 X 10(5) IgE receptors per cell that bind both human IgE and rodent IgE with comparable affinity. Average equilibrium constants of the receptors for human IgE and mouse IgE were 2.56 +/- 0.88 X 10(9) M-1 and 1.85 +/- 0.86 X 10(9) M-1, respectively. The cell-surface component of the IgE receptors on cultured basophils has a m.w. of 64,000. Cultured basophils could be passively sensitized with human IgE and mouse IgE monoclonal antibody, and sensitized basophils released characteristic cytoplasmic granules and both histamine and arachidonate upon challenge with either anti-human IgE or antigen. Incubation of cultured basophils with ionophore A23187 or F-Met-Leu-Phe resulted in histamine release. However, compound 48/80 failed to induce histamine release from the cells.  相似文献

Dromedary camels have been implicated consistently as the source of Middle East respiratory syndrome coronavirus (MERS-CoV) human infections and attention to prevent and control it has focused on camels. To understanding the epidemiological role of camels in the transmission of MERS-CoV, we utilized an iterative empirical process in Geographic Information System (GIS) to identify and qualify potential hotspots for maintenance and circulation of MERS-CoV, and produced risk-based surveillance sites in Kenya. Data on camel population and distribution were used to develop camel density map, while camel farming system was defined using multi-factorial criteria including the agro-ecological zones (AEZs), production and marketing practices. Primary and secondary MERS-CoV seroprevalence data from specific sites were analyzed, and location-based prevalence matching with camel densities was conducted. High-risk convergence points (migration zones, trade routes, camel markets, slaughter slabs) were profiled and frequent cross-border camel movement mapped. Results showed that high camel-dense areas and interaction (markets and migration zones) were potential hotspot for transmission and spread. Cross-border contacts occurred with in-migrated herds at hotspot locations. AEZ differential did not influence risk distribution and plausible risk factors for spatial MERS-CoV hotspots were camel densities, previous cases of MERS-CoV, high seroprevalence and points of camel convergences. Although Kenyan camels are predisposed to MERS-CoV, no shedding is documented to date. These potential hotspots, determined using anthropogenic, system and trade characterizations should guide selection of sampling/surveillance sites, high-risk locations, critical areas for interventions and policy development in Kenya, as well as instigate further virological examination of camels.  相似文献

The morphological changes of gingival mast cells of alloxan-induced diabetic rats were studied by electron microscopy. The following observations were made. The cell nucleus and cytoplasm degenerated. The electron density of the granules in the cell cytoplasm clearly decreased. Some granules had dense irregular threads and the granules were surrounded by a thin vacuole. A ghost vacuole formed in some mast cells and disorganized materials accumulated in the cytoplasm. The mast cell nuclei were generally irregular and degenerating mast cells had pyknotic nuclei. General destruction of the cell membrane and granule shedding in some samples was noted and mitochondria with atypical cristae in the cytoplasm of the mast cells were seen. We conclude that the characteristics of the ultrastructure of gingival mast cells in diabetics are distinctive and should be used as criteria for pathogenesis of gingival inflammation.  相似文献
