Growth responses of Pithophora oedogonia (Mont.) Wittr. and Spirogyra sp. to nine combinations of temperature (15°, 25°, and 35°C) and photon flux rate (50, 100, and 500 μmol·m?2·s?1) were determined using a three-factorial design. Maximum growth rates were measured at 35°C and 500 pmol·m?2·s?1 for P. oedogonia (0.247 d?1) and 25°C and 500 μmol·m?2·s?1 for Spirogyra sp. (0.224 d?1). Growth rates of P. oedogonia were strongly inhibited at 15°C (average decrease= 89%of maximum rate), indicating that this species is warm stenothermal. Growth rates of Spirogyra sp. were only moderately inhibited at 15° and 35°C (average decrease = 36 and 30%, respectively), suggesting that this species is eurythermal over the temperature range employed. Photon flux rate had a greater influence on growth of Spirogyra sp. (31% reduction at 50 pmol·m?2·s?1 and 25°C) than it did on growth of P. oedogonia (16% reduction at 50 μmol·m?2·s?1 and 35°C). Spirogyra sp. also exhibited much greater adjustments to its content of chlorophyll a (0.22–3.34 μg·mg fwt?1) than did P. oedogonia (1.35–3.08 μg·mg fwt?1). The chlorophyll a content of Spirogyra sp. increased in response to both reductions in photon flux rate and high temperatures (35°C). Observed species differences are discussed with respect to in situ patterns of seasonal abundance in Surrey Lake, Indiana, the effect of algal mat anatomy on the internal light environment, and the process of acclimation to changes in temperature and irradiance conditions.  相似文献

Symbiodinium californium (#383, Banaszak et al. 1993 ) is one of two known dinoflagellate symbionts of the intertidal sea anemones Anthopleura elegantissima, A. xanthogrammica, and A. sola and occurs only in hosts at southern latitudes of the North Pacific. To investigate if temperature restricts the latitudinal distribution of S. californium, growth and photosynthesis at a range of temperatures (5°C–30°C) were determined for cultured symbionts. Mean specific growth rates were the highest between 15°C and 28°C (μ 0.21–0.26 · d?1) and extremely low at 5, 10, and 30°C (0.02–0.03 · d?1). Average doubling times ranged from 2.7 d (20°C) to 33 d (5, 10, and 30°C). Cells cultured at 10°C had the greatest cell volume (821 μm3) and the highest percentage of motile cells (64.5%). Growth and photosynthesis were uncoupled; light‐saturated maximum photosynthesis (Pmax) increased from 2.9 pg C · cell?1 · h?1 at 20°C to 13.2 pg C · cell?1 · h?1 at 30°C, a 4.5‐fold increase. Less than 11% of daily photosynthetically fixed carbon was utilized for growth at 5, 10, and 30°C, indicating the potential for high carbon translocation at these temperatures. Low temperature effects on growth rate, and not on photosynthesis and cell morphology, may restrict the distribution of S. californium to southern populations of its host anemones.  相似文献

This study establishes the bioenergetics budget of juvenile whitespotted bamboo shark Chiloscyllium plagiosum by estimating the standard metabolic rate (RS), measuring the effect of body size and temperature on the RS, and identifying the specific dynamic action (RSDA) magnitude and duration of that action in juvenile whitespotted bamboo sharks. The mean ±s .d . (RS) of six fish (500–620 g) measured in a circular closed respirometry system was 30·21 ± 5·68 mg O2 kg?1 h?1 at 18° C and 70·38 ± 14·81 mg O2 kg?1 h?1 at 28° C, respectively. There were no significant differences in RS between day and night at either 18 or 28° C (t‐test, P > 0·05). The mean ±s .d . Q10 for 18–28° C was 2·32 ± 0·06 (n = 6). The amount of oxygen consumed per hour changed predictably with body mass (M; 295–750 g) following the relationship: (n = 40, r2= 0·92, P < 0·05). The mean magnitude of RSDA was 95·28 ± 17·55 mg O2 kg?1 h?1. The amount of gross ingested energy (EI) expended as RSDA ranged from 6·32 to 12·78% with a mean ±s .d . of 8·01 ± 0·03%. The duration of the RSDA effect was 122 h. The energy content of juvenile whitespotted bamboo shark, squid and faeces determined by bomb calorimeter were 19·51, 20·3 and 18·62 kJ g dry mass?1. A mean bioenergetic budget for juvenile whitespotted bamboo sharks fed with squid at 18° C was 100C = 29·5G + 31·9RS+ 28·2RSDA+ 6·7F + 2·1E + 1·6U, where C = consumption, G = growth, F = egestion, E = excretion and U = unaccounted energy.  相似文献

Telemetered heart rate (fH) was examined as an indicator of activity and oxygen consumption rate (VO2) in adult, cultivated, Atlantic salmon, Salmo salar L. Heart rate was measured during sustained swimming in a flume for six fish at 10° C [mean weight, 1114 g; mean fork length (f. l.), 50·6 cm] and seven fish at 15° C (mean weight, 1119 g; mean f. l., 50·7 cm) at speeds of up to 2·2 body lengths/s. Semi–logarithmic relationships between heart rate and swimming speed were obtained at both temperatures. Spontaneously swimming fish in still water exhibited characteristic heart rate increases associated with activity. Heart rate and Vo2 were monitored simultaneously in a 575–1 circular respirometer for six fish (three male, three female) at 4° C (mean weight, 1804 g; mean F. L., 62· cm) and six fish (three male, three female) at 10° C (mean weight, 2045 g; mean f. l., 63·2 cm) during spontaneous but unquantified activity. Linear regressions were obtained by transforming data for both fH and Vo2 to log values. At each temperature, slopes of the regressions between fH and Vo2 for individual fishes were not significantly different, but in some cases elevations were. All differences in elevation were between male and female fish. There were no significant differences in regression slope or elevation for fish of the same sex at the two temperatures and so regressions were calculated for the sexes, pooling data from 4 and 10° C. There was no significant difference in the mean ± S. D. Vo2 between the sexes at 4° C (male, 66·0 ± 59·6 mgO2 kg?1 h?1; female, 88·0 ± 60·1 mgO2 kg?1 h?1) or 10° C (male, 166·2 ± 115·4 mgO2 kg?1 h?1; female, 169·2 ± 111–1 mgO2 kg?1h?1). Resting Vo2 (x?± s. d.) at 4°C was 36·7 ± 8.4 mgO2 kg?1 h?1, and 10° C was 72·8 ± 11·9 mgO2 kg?1 h?1. Maximum Vo2 (x?± S. D.) at 4° C was 250·6 ± 40·2 mgO2 kg?1 h?1, and at 10° C was 423·6 ± 25·2 mgO2 kg?1 h?1. Heart rate appears to be a useful indicator of metabolic rate over the temperature range examined, for the cultivated fish studied, but it is possible that the relationship for wild fish may differ.  相似文献

The temperature dependence of NADH:NR activity was examined in several marine phytoplankton species and vascular plants. These species inhabit divergent thermal environments, including the chromophytes Skeletonema costatum (12–15° C), Skeletonema tropicum (18–25° C), Thalassiosira antarctica (?2 to 4° C), and Phaeocystis antarctica (?2 to 4° C), the green alga Dunaliella tertiolecta (14–28° C), and the vascular plants Cucurbita maxima (20–35° C) and Zea mays (20–25° C). Despite the difference in growth habitats, similar temperature response curves were observed among the chromophytic phytoplankton, with temperatures optimal for NR activity being between 10–20° C. In contrast, the chlorophyll b‐containing alga and vascular plants exhibited optimal temperatures for NR activity above 30° C. Such dramatic differences in NR thermal characteristics from the two taxonomic groups reflect a divergence in NR structure that may be associated with the evolutionary diversification of chromophytes and chlorophytes. Further, it suggests a potential contribution of the thermal performance of NR to the geographic distributions, seasonal abundance patterns, and species composition of phytoplankton communities. NR partial activities, which assess the individual functions of Mo‐pterin and FAD domains, were evaluated on NR purified from S. costatum to determine the possible causes for high temperature (>20° C) inactivation of NR from chromophytes. It was found that the FAD domain and electron transport among redox centers were sensitive to elevated temperatures. S. costatum cells grown at 5, 15, and 25° C exhibited an identical optimal temperature (15° C) for NADH:NR activity, whereas the maximal NR activity and NR protein levels differed and were positively correlated with growth temperature and growth rate. These findings demonstrate that thermal acclimation of NO3? reduction capacity is largely at the level of NR protein expression. The consequences of these features on NO3? utilization are discussed.  相似文献

Using exogenous 125I-ubiquitin, ubiquitin-lens protein conjugation was observed with supernatants of cultured rabbit lens epithelial cells and lens cortex tissue. Conjugation was ATP-dependent with the greatest variety and amount of conjugates larger than 150 kDa. In vivo production of ubiquitin-protein conjugates in cultured rabbit and beef lens epithelial cells and rabbit lens tissues of different developmental age was established using immunological detection. There were limited similarities between conjugates found in youngest as opposed to oldest tissue. Cultured rabbit cells contained 27 pmol/mg free ubiquitin and 18 pmol/mg conjugated ubiquitin. Levels of free ubiquitin in lens tissue epithelium, cortex, and core were 36, 5, and 5 pmol/mg, respectively. There were only 2 pmol/mg conjugated ubiquitin in each of these tissues. Hydrolysis of 125I-ubiquitin was catalyzed by supernatants of cultured lens cells, beef and human lens tissues, and reticulocytes. Degradation was greatest in epithelial tissues, and least in core. This corroborates studies which show that proteolytic capabilities are attenuated in older tissue. Decreased initiation of proteolysis by ubiquitination as well as diminished proteolysis in older lens tissue may be related to the accumulation of damaged proteins in aging lens tissue.  相似文献

Clones of Skeletonema costatum (Grev.) Cl. isolated from Narragansett Bay, R.I., during different seasons were grouped according to their electrophoretic banding patterns. The growth rates, pg chlorophyll · cell?1, carbon uptake · cell?1· h?1, and carbon uptake · pg chl?1· h?1 were measured at 20°C, in a 14:10 h L:D cycle at 180 μE · m?2· s?1. Statistically significant sources of variation were found among groups of clones in growth rate, pg chl · cell?1, and carbon uptake · pg chl?1· h?1. It was concluded that there is a significant relationship between the physiological characteristics of clones isolated from populations in different seasons and patterns of genetic variation inferred from the electrophoretic studies. However, genetic diversity detected by banding patterns tends to underestimate the total genetic diversity in natural populations. The groups of clones most common in summer bloom populations had significantly higher growth rates, lower values of pg chl · cell?1, and higher rates of carbon uptake · pg chl?1· h?1 at 20°C than did the group of clones most common in winter bloom populations. However, differences among groups in these parameters at 20°C alone cannot account for the seasonal cycling of genetically variable populations of Skeletonema in Narragansett Bay. The range of growth rates among clones of this species is 0.1–5.0 divisions · d?1 under a single set of temperature and light conditions. Chlorophyll concentrations range from 0.2–1.7 pg chl · cell?1 and carbon uptake · pg chl?1· h?1 varies by a factor of 7 among clones. The range of physiological variation in this species means that it is difficult to use laboratory studies of single clones to analyze the responses of natural populations of Skeletonema.  相似文献

The eye lens is a useful tissue for studying phenomena related to aging since it can be separated into differentially aged or matured zones. This work establishes correlations between ubiquitin-lens protein conjugating capabilities and age, as well as the stage of maturation of bovine lens tissue. When exogenous 125I-ubiquitin was combined with supernatants of epithelial (least mature), cortex, and core (most mature) tissue, ATP-dependent conjugation of 125I-ubiquitin to lens proteins was most effective with the epithelial tissue preparation. Conjugate formation was greatest when lenses were obtained from young animals. Supernatants from cultured bovine lens epithelial (BLE) cells conjugated more 125I-ubiquitin to lens proteins than any tissue preparation. In all cases the predominant conjugates formed in these cell-free assays were of high molecular mass, although conjugates with masses in the 25-70 kDa range were also observed. Lens tissue and cultured BLE cell preparations were also probed with antibodies to ubiquitin to detect in vivo ubiquitin-lens protein conjugates. There was more free ubiquitin and ubiquitin conjugates in tissue from young as compared with older lenses. The greatest levels of conjugates were observed in cultured BLE cells. Specificity in the ubiquitination system is indicated since some of the conjugates formed in vivo appear identical to those formed in the cell-free assays and in reticulocytes using exogenous 125I-ubiquitin. Upon development and maturation of lens tissue (i.e., core as opposed to epithelium), there is accumulation of lower molecular mass conjugates.  相似文献

Ventilation was measured directly in the hagfish, Myxine glutinosa L., by means of an electro-magnetic blood flowmeter. Ventilatory flow and frequency increased from 0.86 ± 0.27 ml·min?, and 18.2 ± 5.1·min?, respectively, at 7°C to 1.70 ± 0.20 ml·min?, and 70.1 ± 9.5·min? at 15 ·C.Standard oxygen consumption,V?O2, was measured in non-buried hagfish. When severely dehydrated Bedouin goats were allowed to drink to satiation their plasma arginine vasopressin concentration immediately dropped from a value of 19.9±9.4 pmol·l-1 to 9.4±3.9 pmol·l-1 (P<0.05). It continued to drop further until a concentration of 1.8±2.9 pmol·l-1 was recorded, similar to that reported for goats allowed to drink freely. When the goats were shown the water but drinking was denied, plasma arginine vasopressin immediately dropped to 11.7±4.0 pmol·l-1 (P<0.05) and further decreased to 10.0±4.8 pmol·l-1 5 min following their sighting the water. This level, however, was not sustained and 2 h after the initial drop the high pre-trial concentration of plasma arginine vasopression was regained. Presumably, sighting of water by dehydrated goats induces an abrupt drop in their plasma arginine vasopressin level even before drinking commences. When rehydrated, by introducing water directly to the rumen, circumventing both the sensing of the water and the drinking proper, no immediate drop in the plasma arginine vasopression concentration of the newly rehydrated goats was observed. A delayed drop in the plasma arginine vasopressin levels took place slowly, concurrently with the drop in osmolality and concentration of Na+ in the plasma. It is suggested that sighting of water by dehydrated goats is involved in the modulation of plasma arginine vasopressin.  相似文献

In roach Rutilus rutilus growth ceases below a temperature threshold of 12° C. This cessation of growth is accompanied by a reduction in feeding. Do roach decrease feeding in the cold because of reduced energy demand, caused by the decelerating effect of low temperature on metabolism and growth, or is feeding directly limited by low temperatures, leading to reduced growth rates? It was found that at low temperatures the intake and digestion of food may be limited by reduced activities of digestive enzymes. Trypsin, amylase and γ‐glutamyl transferase showed a negative compensation with respect to temperature, resulting in very low activities at acclimation temperatures of ≤12° C. Trypsin activity, falling from 400·5 ± 131·2 U g?1 fresh mass of the gut at 27° C to 12·5 U g?1 fresh mass at 4° C, displayed the strongest linear correlation with growth rates, suggesting that trypsin activities may set a limit to growth in the low temperature range. If protein digestion is limiting at low temperatures, this should be reflected in reduced concentrations of amino acid in the white muscle. The size of the total amino acid pool was not affected by temperature acclimation and ranged between 19·2 ± 6·2 and 25·2 ± 3·6 µmol g?1 fresh mass of the white muscle. A decrease, however, was found of several amino acids, mainly of threonine and glutamine, in the low temperature range. Low concentrations of the essential amino acid threonine (0·14 ± 0·03 µmol g?1 fresh mass at 12° C and 0·12 ± 0·05 µmol g?1 fresh mass at 4° C) were probably due to nutritional or digestional limitations and may therefore have resulted from reduced trypsin activity in the cold. The non‐essential amino acid glutamine, however, can be endogenously synthesized and its low level observed at 4° C (0·16 ± 0·09 µmol g?1 fresh mass) was not necessarily a result of low trypsin activities. It is more likely that low temperatures impair glutamine synthesis. The possibility that glutamine concentrations may be down regulated under conditions when anabolic processes are not advantageous is discussed.  相似文献

N-acetylglucosaminyl transferases from pupae of Stomoxys calcitrans (L.) were studied in 10,000g pellet suspensions. Characterization of these enzymes was based on formation of glycolipids (ie, Dol·PP-GlcNAc and Dol·PP-(GlcNAc)2), oligosaccharide lipids, and giycoproteins. Studies on transferase activity during the pupal instar showed that there were two peaks of activity; the first peak was on day 0 (prepupae) and the second at 3 days after pupation. Subcellular fractionation indicated that 10,000g and 100,000g pellets contained most of the transferase activities. The transferases required divalent cations (either Mn2+ or Mg2+). The pH optimum, which varied for each of the products formed, was 7.5 for glycolipids, 7.0 for oligosaccharide lipids, and 6.5 for glycoprotein. Inclusion of dolichol monophosphate doubled the amount of Dol·PP-GlcNAc and Dol·PP·(GlcNAc)2 formed, but had little effect on oligosaccharide lipid and glycoprotein formation. Tunicamycin was a potent inhibitor of glycolipid formation with an I50 of 1.8–4.8 nM. It was confirmed that tunicamycin acts by preventing the transfer of GlcNAc-1-P from UDP-GlcNAc to Dol·P. UMP reverses glycolipid formation, yielding UDP-GlcNAc. Some characterization of the products was performed. Glycolipids were shown to be Dol·PP-GlcNAc and Dol·PP-(GlcNAc)2. Glycoprotein was rapidly solubilized by protease and detergent treatments, whereas oligosaccharide lipids appeared to be acid-labile, pyrophosphate-containing lipids. The apparent kinetic constants for the formation of glycolipids were as follows: UDP-GlcNAc Km = 1.55 ± 0.47 μM, Vmax = 0.66 ± 0.21 pmol·min?1·mg?1; Dol·P Km = 2.08 ± 0.85 μM, Vmax = 0.13 ± 0.06 pmol·min?1·mg?1 protein.  相似文献

Growth and sexual reproduction of the marine littoral diatom Cocconeis scutellum Ehrenb. var. ornata Grun. were investigated at 30 different combinations of temperature (5, 10, 14, 18, 22° C), irradiance (20, 60, 100 μE·m?2·s?1) and daylength (14:10 and 10:14 h LD cycle). Growth occurred at all combinations. The optimal growth was observed at 14–18° C, long daylength and highest-to-moderate irradiance, and at 18° C, short daylength and highest irradiance. Sexual reproduction on the other hand occurred between 5 and 18° C, and the optimal condition was 10–14° C and short daylength. Annual cyclic, and sesonal changes in the distribution of cell size (valve length) were observed in a field population. These changes were characterized by an annual minimum in mean cell size in autumn, an annual maximum in winter, a slight decrease from the mean in spring–middle summer, a rapid decrease from the mean in late summer–early autumn, and appearance of bimodal distribution of cell size in winter. These changes were caused by sexual reproduction in autumn, rapid growth in late summer–early autumn and slow growth in other seasons, and poor viability of small cells near the lower end of the size range.  相似文献

Photosynthesis and respiration of three Alaskan Porphyra species, P. abbottiae V. Krishnam., P. pseudolinearis Ueda species complex (identified as P. pseudolinearis” below), and P. torta V. Krishnam., were investigated under a range of environmental parameters. Photosynthesis versus irradiance (PI) curves revealed that maximal photosynthesis (Pmax), irradiance at maximal photosynthesis (Imax), and compensation irradiance (Ic) varied with salinity, temperature, and species. The Pmax of Porphyra abbottiae conchocelis varied between 83 and 240 μmol O2 · g dwt?1 · h?1 (where dwt indicates dry weight) at 30–140 μmol photons · m?2 · s?1 (Imax) depending on temperature. Higher irradiances resulted in photoinhibition. Maximal photosynthesis of the conchocelis of P. abbottiae occurred at 11°C, 60 μmol photons · m?2·s?1, and 30 psu (practical salinity units). The conchocelis of P. “pseudolinearis” and P. torta had similar Pmax values but higher Imax values than those of P. abbottiae. The Pmax of P. “pseudolinearis” conchocelis was 200–240 μmol O2 · g dwt?1 · h?1 and for P. torta was 90–240 μmol O2 · g dwt?1 · h?1. Maximal photosynthesis for P. “pseudolinearis” occurred at 7°C and 250 μmol photons · m?2 · s?1 at 30 psu, but Pmax did not change much with temperature. Maximal photosynthesis for P. torta occurred at 15°C, 200 μmol photons · m?2 · s?1, and 30 psu. Photosynthesis rates for all species declined at salinities <25 or >35 psu. Estimated compensation irradiances (Ic) were relatively low (3–5 μmol · photons · m?2 · s?1) for intertidal macrophytes. Porphyra conchocelis had lower respiration rates at 7°C than at 11°C or 15°C. All three species exhibited minimal respiration rates at salinities between 25 and 35 psu.  相似文献

Ulothrix zonata (Weber and Mohr) Kütz. is an unbranched filamentous green alga found in rocky littoral areas of many northern lakes. Field observations of its seasonal and spatial distribution indicated that it should have a low temperature and a high irradiance optimum for net photosynthesis, and at temperatures above 10°C it should show an increasingly unfavorable energy balance. Measurements of net photosynthesis and respiration were made at 56 combinations of light and temperature. Optimum conditions were 5°C and 1100 μE·m?2·s?1 at which net photosynthesis was 16.8 mg O2·g?1·h?1. As temperature increased above 5° C optimum irradiance decreased to 125 μE·m?2·s?1 at 30°C. Respiration rates increased with both temperature and prior irradiance. Light-enhanced respiration rates were significantly greater than dark respiration rates following irradiance exposures of 125 μE·m?2·s?1 or greater. Polynomials were fitted to the data to generate response surfaces. Polynomial equations represent statistical models which can accurately predict photosynthesis and respiration for inclusion in ecosystem models.  相似文献

Feeding rates of Brachionus plicatilis were studied for two types of food — algae Monochrysis lutheri and baker's yeast Saccharomyces cerevisae. The main regularities of changes in filtration rate and ration were studied in small culture volumes (1 ml) for adult amictic females depending on food concentration (1, 2, 4, 8 and 16 · 106 cells · ml−1), ambient temperature (16 and 26 °C), and salinity (5, 10, 15, 20, 25 and 30 ppt). B. plicatilis ration did not depend on the salinity, but was largely determined by temperature and food concentration. It was found that at 16 and 26 °C the dependence of the ingestion rate (ration) on food concentration differed greatly. A hypothesis was suggested to explain this phenomenon. A critical concentration of both types of food at which the increase in the rotifer ration ceased is 4 · 106 cells · ml−1. This is the minimum “background” food concentration for B. plicatilis mass cultivation. The average rations measured at the concentration of M. lutheri and S. cerevisae of 4 · 106 cells · ml−1 where 1.3 ± 0.1 and 4.8 ± 1.3 μg dry weight. · ind−1 · day−1 at 26 °C and 0.54 ± 0.1 and 1.9 μg d. w. · ind−1 · day−1 at 16 °C, respectively. The rations obtained in the laboratory were corrected for the conditions of rotifer commercial production in the open field in summer time. The correct values were 0.86 and 0.72 μg d. w. · ind−1 · day−1 for algae and yeast, respectively.  相似文献

Aims: To study the accumulation of vegetative cells and endospores of Alicyclobacillus, as well as viable aerobic counts during the continuous production of apple juice concentrate. Methods and Results: Apples were processed for a continuous process running time of 108 h (processing rate 1·8–2·0 t h?1) without clean‐in‐place (CIP) procedures in‐between different batches. Samples from single‐strength apple juice, concentrate after evaporation (±30°Brix), the final product (concentrate pasteurized at 102–104°C for 90 s) and condensate water (by‐product of the juice concentration process) were collected every 12 h. From 12 to 84 h of processing, vegetative Alicyclobacillus counts in single‐strength apple juice increased significantly (P < 0·05) from 1 to 3·15 log10 CFU ml?1. Accumulation patterns of vegetative cells in apple concentrate and the final product were similar from 24 to 84 h of processing, with the respective counts increasing from 0·13 to 1·63 and 0·01 to 1·69 log10 CFU ml?1. The highest Alicyclobacillus endospore counts in single‐strength juice, concentrate and the final product was at 84 h of processing with 1·32, 1·59 and 1·64 log10 CFU ml?1, respectively. Conclusions: Alicyclobacillus vegetative cells and endospores accumulate in fruit concentrates during a continuous process running time of 108 h. Zinc, cadmium, and copper are known to interact in many transport processes, but the mechanism of inhibition is widely debated, being either competitive or noncompetitive according to the experimental model employed. We investigated the mechanisms of inhibition of zinc transport by cadmium and copper using renal proximal cells isolated from rabbit kidney. Initial rates of65Zn uptake were assessed after 0.5 min of incubation. The kinetics parameters of zinc uptake obtained at 20°C were a Jmax of 208.0±8.4 pmol· min−1·(mg protein)−1, aK m of 15.0±1.5 μM and an unsaturable constant of 0.259±0.104 (n=8). Cadmium at 15 μM competitively inhibited zinc uptake. In the presence of 50 μM cadmium, or copper at both 15 and 50 μM, there was evidence of noncompetitive inhibition. These data suggest that zinc and cadmium enter renal proximal cells via a common, saturable, carrier-mediated process. The mechanisms of the noncompetitive inhibition observed at higher concentrations of cadmium or with copper require further investigation, but may involve a toxic effect on the cytoskeleton.  相似文献

This is the first study investigating the plant–herbivore interaction between Sarpa salpa, which has overgrazed seagrass transplants in Portugal, and the seagrasses Cymodocea nodosa, Zostera marina and Zostera noltii, which have been considered for restoration. When offered the choice between the three seagrasses in outdoor tanks, adult S. salpa clearly preferred Z. noltii. Testing the seagrasses separately, mean ± s.d. feeding rates ranged from 21 ± 11 g seagrass fresh mass kg?1 fish mass day?1 for Z. marina to 32 ± 9 g seagrass fresh mass kg?1 fish mass day?1 for C. nodosa and 40 ± 11 g seagrass fresh mass kg?1 fish mass day?1 for Z. noltii (temperature = 16° C). Food‐processing rate in S. salpa did not differ between seagrasses, and there was no evidence of a regulation of processing rate according to food intake. Seagrasses differed substantially in nitrogen content and C:N, with C. nodosa containing the highest nitrogen content and lowest C:N (2·5 ± 0·1% and 14·0 ± 1·0), followed by Z. noltii (2·1 ± 0·1% and 17·0 ± 1·0) and Z. marina (1·4 ± 0·1% and 26·0 ± 2·0). Food‐processing rate in S. salpa and the nutritional value of the seagrasses were not correlated with the observed feeding preference and rate. The study suggests that C. nodosa and Z. marina are less at risk of overgrazing by S. salpa and might thus be preferable to Z. noltii for seagrass restoration in areas with noticeable abundances of this fish.  相似文献

Two replicate experiments were conducted to investigate the effect of light intensity on the growth and nutrient uptake of Skeletonema costatum (Grev.) Cleve in silicate-limited continuous culture. Each experiment began with 4 identical chemostat cultures of S. costatum growing at the normal laboratory light (0.14 ly · min?1, continuous illumination) under strong silicate limitation. Screens were placed over 3 cultures reducing them to light intensities of 0.042, 0.021 and 0.0018 ly · min?1. Based on growth rules, nutrient uptake rates, cell morphology and chemical composition, the cultures receiving 0.021, and 0.0018 ly · min?1 appeared to he light-limited, whereas the culture receiving 0.14 ly.  相似文献
