The Cu/Zn superoxide dismutase (SOD1) catalyzes the dismutation of superoxide radicals produced in the course of biological oxidations. When placed under the control of the rat SOD1 gene promoter and transfected into human HepG2 hepatoma cells, the activity of a chloramphenicol acetyltransferase reporter gene was found to increase three- to four-fold in the presence of heavy metals (cadmium, zinc and copper). Functional analysis of mutant derivatives of the SOD1 gene promoter and the use of a heterologous promoter system confirmed that the induction of the SOD1 gene by metal ions requires a metal-responsive element (MRE) located between positions −273 and −267 (GCGCGCA). It was also shown by gel mobility shift assays that an MRE binding protein is induced by the exposure of the human liver cell line HepG2 to heavy metals. These results suggest that the MRE participates in the induction of the SOD1 gene by heavy metals. Received: 5 February 1999 / Accepted: 21 May 1999  相似文献

When expressed in heterologous microorganisms Vitreoscilla hemoglobin (VHb) acts as oxygen storage and causes a higher oxygen uptake. In this study, the effect of this protein on growth, sensitivity and antioxidant properties of Enterobacter aerogenes exposed to metal stress was investigated. The strain expressing VHb was more sensitive to mercury and cadmium as the minimal inhibitory concentration (MIC) for these metals was up to 2-fold lower in this strain than the host and the recombinant strain carrying a comparable plasmid. At lower concentrations than MIC, the metals partially limited growth and caused an inhibition proportional to metal concentration applied. The growth pattern of VHb expressing strain was also distinctly different from other two non-hemoglobin strains. The hemoglobin containing strain showed substantially higher superoxide dismuates (SOD) activity than the non-hemoglobin strains, while catalase levels were similar in all strains. All strains exposed to copper, however, showed similar MIC values, growth patterns, and SOD and catalase levels.  相似文献

Cadmium stress in sugar cane callus cultures: Effect on antioxidant enzymes   总被引:5,自引:0,他引:5  
Catalase (CAT) and superoxide dismutase (SOD) are antioxidant enzymes which are important in the metabolism of reactive oxygen species (ROS), and can be induced by environmental stresses including cadmium (Cd), a heavy metal toxic to living organisms. Sugar cane (Saccharum officinarumL.) in vitro callus cultures were exposed to CdCl2 and the activities of CAT and SOD were analysed. Lower concentrations of CdCl2, such as 0.01 and 0.1 mM caused a significant increase in callus growth, whereas 0.5 and 1 mM CdCl2 strongly inhibited growth of the callus cultures, but only after 9 days of CdCl2 treatment. Red-brown patches were also observed in calluses exposed to 0.5 and 1 mM CdCl2. Calluses grown in 0.01 and 0.1 mM CdCl2 did not exhibit any changes in CAT activity even after 15 days of growth in the presence of CdCl2. However, for calluses grown in higher concentrations of CdCl2 (0.5 and 1 mM), a rapid increase in CAT activity was detected, which was 14-fold after 15 days. Furthermore, up to five CAT isoforms were observed in callus tissue. Total SOD activity did not exhibit any major variation. One Mn-SOD and two Cu/Zn-SOD isoenzymes were observed in callus cultures and none exhibited any variation in response to the CdCl2 treatments. The results suggested that in sugar cane callus cultures, CAT may be the main antioxidant enzyme metabolizing H2O2.  相似文献

The oxidative status of liver of female rats exposed to lead acetate and cadmium acetate either alone or in combination at a dose of 0.05?mg/kg body wt intraperitoneally for 15 days was studied. After the administration of lead alone, the activity of superoxide dismutase (SOD) decreased in liver, whereas no changes were observed in catalase (CAT) activity, and glutathione (GSH) and thiobarbituric acid (TBARS) levels. Cadmium exposure and combined exposure to lead and cadmium led to decrease in GSH content and increased TBARS levels. Moreover, animals exposed to either cadmium alone or in combination with lead showed a decrease in SOD activity and an increase in CAT activity. The in vitro experiments showed that vitamin E failed to restore the antioxidant enzyme activities in metal treated postmitochondrial supernatant fraction of liver. But Mn2+ ions protected the mitochondria from lipid peroxidation and could completely restore Mn-superoxide dismutase (Mn-SOD) activity following metal intoxication. The results of this study indicate that despite the ability of lead and cadmium to induce oxidative stress the effect in liver is not intensified by combined exposure to both lead and cadmium. The observed changes in various oxidative stress parameters in the liver of rats co-exposed to lead and cadmium may result from an independent effect of lead and /cadmium and also from their interaction such as changes in metal accumulation and content of essential elements like Cu, Zn and Fe. These results suggest that when lead and cadmium are present together in similar concentrations, cadmium mediates major effects due to its more reactive nature.  相似文献

The trend of the total acid-soluble thiols (AST) levels including glutathione (both in reduced form, GSH, and in the oxidized one, GSSG) and free cysteine (CySH) has been studied in Euglena gracilis cells exposed to copper or cadmium up to the fifth day of growth.2. The copper does not affect the levels of AST and of glutathione. A limited increase of CySH is observed during the first days of growth. This is followed by a gradual depletion for longer times.3. The exposure to cadmium leads to an increase in the content of AST as well as of glutathione and CySH levels according to the metal dose and to the exposure time.4. The data are interpreted taking into account the previous observations on the responses of Euglena gracilis to exposure to heavy metals.  相似文献

The effect of lead, cadmium and cooper on protein pattern, free radicals and antioxidant enzymes in root of Lupinus luteus L. were investigated. Heavy metals inhibited growth of lupin roots, which was accompanied by increased synthesis and accumulation of a 16 kDa polypeptide (Przymusiński et al. 1991 Biochem. Physiol. Pflanzen., 187:51–57). This component has been earlier identified as immunologically related to Cu,Zn-superoxide dismutase (Przymusiński et al. 1995 Env.Exp.Bot., 35:485–495). However, more detailed study revealed that this stress-stimulated protein is composed of four to six polypeptides of different electrophoretic mobility. The most abundant polypeptides of the 16kDa region were found to be closely homologous to pathogen related proteins. The number and intensity of these polypeptides was highly variable in roots of individual seedlings, which suggests that they might represent separate allelic forms. Electron paramagnetic spectra revealed that at low lead concentrations the amplitude of the first derivative was similar to the control and distinctly increased at higher metal concentrations. On the other hand, at the lower lead concentrations the activity of antioxidant enzymes increased, whereas at higher metal doses the enzyme activities did not raise further (SOD) or even dropped (CAT, APOX). This implies that the responses of antioxidant system to lead is dose-dependent stimulated by low metal concentrations, whereas at the higher metal level the free radical emission is beyond the quenching capacity of antioxidant enzymes, which in turn might contribute to the reduced root growth. The effect of various heavy metals: Pb2+, Cd2+ and Cu2+ on phytochelatins and antioxidant enzymes depends on the kind of metal ion. Pb2+ and Cd2+ stimulated the PCs formation whereas Cu2+ was not effective. On the other hand, in root exposed to Cu the activity of catalase (CAT) was the highest as was the production of H2O2. The strong oxidative effect of Cu2+ ions which were not complexed by PCs suggests that these peptides might by involved in the cellular defense system by binding excessive heavy metal ions. On the basis of our results it can be concluded that in lupin roots exposed to heavy metals there is a complex defense system against metal phytotoxicity, which comprises of specific proteins, antioxidant enzymes and phytochelatins.  相似文献

There are increasing concerns over the environmental implications involved in battery plant due to the elevated metal concentrations in the surrounding soils. Earthworms are regarded as a model organism for evaluation of terrestrial environmental pollution. The biochemical responses in the earthworms, Eisenia fetida, exposed to polluted soils were investigated. Soil samples at 0, 50, 100, 200, 500 and 1000 m from a battery plant were collected and used to assess the relationship between sampling distance and biochemical responses in E. fetida. Biochemical alterations were carried out on 1, 3, 7 and 14 d of exposure. The metallothionein (MT), malondialdehyde (MDA) and antioxidant enzymes (superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD)) were measured. The results demonstrated that concentrations of heavy metals, especially Pb, decreased sharply with increase in distance, which may be due to the leaching and accumulation of Pb in soil during production processes. The concentrations of MT and MDA increased with exposure (dosage and time), indicating that E. fetida presented dose and time dependence after heavy metal exposure. The activities of SOD and CAT increased after heavy metal induction, while the activity of POD was lower than that of the control group. The SOD, CAT and POD present a cooperative effect for the protection of lipids and enzymes due to the pollution. The soils around the battery plant were polluted with metals giving rise to adverse biological effects.  相似文献

微生物在修复重金属污染土壤中发挥重要作用。为阐明假单胞菌TCd-1降低水稻镉吸收的机理,通过加镉土培盆栽试验,在10 mg/kg镉处理下,对比研究了接种菌株对2种镉耐性不同水稻品种(特优671、百香139)镉含量、根系活力、光合作用、抗氧化酶活性及抗氧化物质含量的影响。结果表明:与10 mg/kg镉处理相比,接种假单胞菌TCd-1后水稻“特优671”和“百香139”的根、茎、叶、糙米的镉含量分别依次降低了30.4%、39.1%、40.7%、29.2%和52.2%、51.7%、18.4%、38.8%;提高了2种水稻的根系活力、光合作用,促进了水稻的生长;“特优671”、“百香139”叶片的超氧化物歧化酶(Superoxide dismutase, SOD)活性分别提高了7.3%、138.5%,过氧化物酶(Peroxidase, POD)活性提高了82.0%、106.2%,过氧化氢酶(Catalase, CAT)活性提高了58.8%、172.7%;叶片的类黄酮含量提高了139.4%、73.6%,总酚含量提高了27.2%、23.1%;超氧阴离子含量降低了44.2%、29.0%,丙二醛(Mal...  相似文献

The results of experiment clearly reveal that cadmium inhibited root penetration by the second stage juveniles (J2) of Meloidogyne incognita which subsequently affected the development of root galls in tomato. The heavy metal was highly injurious to tomato plants at all the concentrations tested for (7.5, 15.0, 30.0 and 60.0?ppm). The inhibitory effect on plant growth and other parameters (fresh and dry weight of plant, chlorophyll content of leaves, water absorption capability of roots) significantly increased with an increase in the concentration of the metal. Abstract

Aquatic plants have been identified as potentially useful for accumulating and bioconcentrating heavy metals. This study was developed to test the hypothesis that nutrient enrichment enhances the metal tolerance of floating macrophytes. Relative growth rates (RGR), photosynthetic pigments (chlorophyll a, b and carotenoid), malondialdehyde (MDA) content, and electrical conductivity (EC) were measured in Lemna gibba exposed to different cadmium and copper concentrations in laboratory conditions. Relative growth rates were negatively correlated with metal exposure, but nutrient addition suppressed this effect. Photosynthetic pigment levels were negatively correlated with metal exposures, and nutrient addition attenuated chlorophyll decrease in response to metal exposures. MDA content and EC also showed sharp increases at higher concentrations, indicating oxidative stress. This study indicates that nutrient enrichment increases the tolerance of Lemna gibba to metals, and that Lemna gibba is a suitable candidate for the phytoremediation of low-level copper and cadmium pollution.  相似文献

Increased heavy metal pollution generated through anthropogenic activities into the environment has necessitated the need for eco-friendly remediation strategies such as mycoremediation. With a view to prospecting for fungi with heavy metal remediation potentials, the tolerance of five Aspergillus species isolated from soils of three active gold and gemstone mining sites in southwestern Nigeria to varied heavy metal concentrations was investigated. Isolated Aspergillus strains were identified based on the internal transcribed spacer 1 and 2 (ITS 1 and ITS 2) regions. Growth of Aspergillus strains were challenged with a range of varied concentrations of heavy metals: cadmium (Cd) (0–100), copper (Cu) (0–1000), lead (Pb) (0–400), arsenic (As) (0–500), and iron (Fe) (0–800) concentrations (ppm) incorporated into Malt Extract Agar (MEA) in triplicates. Mycelial radial growths were recorded at intervals of 3 days during a 13-day incubation period. Aspergillus strains were identified as A. tubingensis, A. fumigatus, A. terreus, A. nidulans, and A. nomius. A. tubingensis tolerated Cd, Cu, Pb, As, and Fe at all test concentrations (100–1000 ppm), showing no significant (p > .05) difference compared with the control. Similarly, A. nomius tolerated all concentrations of Cu, Pb, As, and Fe and only 50 ppm Cd concentrations. A. nidulans, A. terreus, and A. fumigatus, on the other hand, tolerated all concentrations of Cu, Pb, and Fe with no statistical significance (p > .05) difference from the controls. Overall, the Aspergillus species showed tolerance to heavy metal concentrations above permissible limits for contaminated soils globally. These heavy metal tolerance traits exhibited by the Aspergillus isolates may suggest that they are potential candidates for bioremediation of heavy metal–polluted environments.  相似文献

We studied how the relationship between cadmium (Cd) toxicity and oxidative stress influenced the growth, photosynthetic efficiency, lipid peroxidation, and activity of ntioxidative enzymes in the roots and leaves of rice(Oryza sativa L Dongjin). Plants were exposed to Cd for 21 d. Both seedling growth and photosynthetic efficiency decreased gradually with increasing cadmium concentrations. Lipid peroxidation increased slowly in both roots and leaves, causing oxidative stress. However, each tissue type responded differently to Cd concentrations with regard to the induction/ inhibition of antioxidative enzymes. The activity of Superoxide dismutase (SOD) increased in both roots and leaves. Ascorbate peroxidase (APX) activity increased in leaves treated with up to 0.25 μM Cd, then decreased gradually at higher concentrations. In contrast, APX activity in roots increased and remained constant between 0.25 and 25 μM Cd. Enhanced peroxidase (POD) activity was recorded for treatments with up to 25/M Cd, gradually decreasing at higher concentrations in the leaves but remaining unchanged in the roots. Catalase (CAT) activity increased in the roots, but decreased in the leaves, whereas the activity of glutathione reductase (GR) was enhanced in both roots and leaves, where it remained elevated at higher Cd concentrations. These results suggest that rice seedlings tend to cope with free radicals generated by Cd through coordinated, enhanced activities of the antioxidative enzymes involved in detoxification.  相似文献

Respirometric experiments demonstrated that the oxygen uptake by Thiobacillus ferrooxidans strain LR was not inhibited in the presence of 200 mM copper. Copper-treated and untreated cells from this T. ferrooxidans strain were used in growth experiments in the presence of cadmium, copper, nickel and zinc. Growth in the presence of copper was improved by the copper-treated cells. However, no growth was observed for these cells, within 190 h of culture, when cadmium, nickel and zinc were added to the media. Changes in the total protein synthesis pattern were detected by two-dimensional polyacrylamide gel electrophoresis for T. ferrooxidans LR cells grown in the presence of different heavy metals. Specific proteins were induced by copper (16, 28 and 42 kDa) and cadmium (66 kDa), whereas proteins that had their synthesis repressed were observed for all the heavy metals tested. Protein induction was also observed in the cytosolic and membrane fractions from T. ferrooxidans LR cells grown in the presence of copper. The level of protein phosphorylation was increased in the presence of this metal.  相似文献

Cadmium is a non-essential heavy metal that can be harmful even at low concentrations to plants. Colocassia esculentum (Araceae) plant was studied to know its tolerance capability to cadmium. Colocassia esculentum plants grown in pots containing different concentrations of cadmium (Cd) were analyzed for dry matter, fresh weight and total metal content. Cadmium depressed dry matter production of the plant up to 33%. Plant accumulated larger portion of the heavy metal in the roots followed by stem and leaf. Chlorophyll content of the plant declined on treatment with heavy metal while the activity of antioxidative enzymes catalase and peroxidase increased. Colocassia esculentum also showed an increase in total protein along with greater A250/A280 value suggesting an increase in metal protein complexes. Cadmium elicited anatomical changes in the root of C. esculentum. These changes under heavy metal stress indicate the adaptive properties of this plant species.  相似文献

Euglena gracilis is susceptible to cadmium (Cd) at high concentrations. There are no comparative data on cytotoxicity or abnormality of CdCl2 to E. gracilis Z and its achlorophyllous mutant SMZ. The present study examined the cytotoxicity of CdCl2 under continual exposure at levels ranging from sub-ppm to ppm, and assessed the effects of zinc (Zn) or cyanocobalamin (VB12) supplementation on the suppression of Cd-induced abnormal cell proliferation and hypertrophy. With Zn levels restricted to 1 ppm [as Zn++], cell growth of both E. gracilis strains was reduced in proportion to Cd concentration. More abnormal cells (hypertrophied, V-shape and starfish-shape) were observed in both strains at sub-ppm levels of Cd. ZnSO4 supplementation from 2 to 63 ppm significantly suppressed the incidence of Cd-induced abnormality. However, a significant increase in abnormal cells was observed following Zn supplementation at levels of 125 and 250 ppm, which produced remarkable differences in cell morphology. The incidence of abnormal cells varied with supplemented VB12 levels ranging from 4 to 250 ppb in both E. gracilis strains.  相似文献

The cadmium (Cd2+) and lead (Pb2+)-induced changes in Cu,Zn-SOD gene expression on the level of mRNA accumulation and enzyme activity were analyzed in roots of soybean (Glycine max) seedlings. The Cd2+ caused the induction of copper–zinc superoxide dismutase (Cu,Zn-SOD) mRNA accumulation, at each analyzed metal concentration (5–25 mg/l), whereas in Pb2+-treated roots this effect was observed only at the medium metal concentrations (50–100 mg/l of Pb2+). The analysis of Cu,Zn-SOD activity proved an increase in enzyme activity during Cd2+/Pb2+ stresses, however in Pb2+-treated plants the activity of enzyme was not correlated with respective mRNAs level. Presented data suggest that different metals may act on various level of Cu,Zn-SOD expression in plants exposed to heavy metals stress.  相似文献

High concentrations of heavy metals have been previously detected in Antarctic sponge tissues, but their effect on the associated bacterial assemblages has been never investigated. Metal tolerance is often linked to antibiotic resistance and can also affect biochemical activities within microbial populations. In the present work, the response to heavy metals and antibiotics, as well as the enzymatic profile, of bacteria associated with the sponge Hemigellius pilosus, was analyzed. Tolerance to mercury, cadmium and zinc (at concentrations between 10 and 10,000 ppm) was tested by the plate diffusion method. Almost all isolates completely tolerated zinc and cadmium up to 1,000 and 2,500 ppm, respectively, whereas complete tolerance to mercury was generally observed at concentrations between 10 and 500 ppm. As bacteria can develop resistance in the growing presence of toxic compounds in the environment, this finding could be related to the concentrations of metals in the sponge tissues. The susceptibility assay to 11 antibiotics revealed that multiple antibiotic resistance was generally exhibited, with gentamicin that inhibited all Antarctic isolates. The comparison of the heavy metal and antibiotic resistance patterns at phylogenetic level revealed some distinctive features, suggesting that the dissemination of heavy metal tolerance and antibiotic resistance may possess great relevance for the population dynamics. Additionally, growth patterns often highly differed among strains in the same species, thus appearing to be more likely strain specific rather than species specific. The enzyme expression by the isolates was not really affected by the heavy metal tolerance they showed, as variation in the enzymatic profiles was observed in strains within the same genus that showed different/similar heavy metal tolerance patterns.  相似文献

In this study, we have analyzed superoxide dismutase (SOD), ascorbate peroxidase (APX) and glutathione reductase (GR) activities, biomass accumulation and chlorophyll‐a content in the Arthrospira platensis ‐M2 strain grown at different concentrations of zinc (Zn), tin (Sn) and mercury (Hg). We found that there is a close relationship between chlorophyll‐a content and biomass accumulation in A. platensis ‐M2 strain as a result of Zn, Sn and Hg exposures. Sn was found to be the most toxic heavy metal among others because of the continious inhibition of both biomass and chlorophyll‐a accumulation at 500 and 1000 μg mL?1 concentrations after the third day of the study, while they represented continuous increases at each Zn and Hg concentration over 7 days. Lower concentrations of Zn and Sn stimulate SOD and GR activities remarkably, probably due to oxidative stress caused by heavy metal toxicity. APX activity was significantly lowered by higher concentrations of the three metals used in this study. Our results suggest that higher heavy metal concentrations inhibited SOD, APX and GR activities but biomass and chlorophyll‐a accumulation endured in a time‐dependent manner, possibly due to some different defence mechanisms, which remain to be investigated.  相似文献

Through preliminary plate tests,Kluyveromyces marxianus was found to be much more resistant to toxic heavy metals compared to aCUP1 R strain ofSaccharomyces cerevisiae. Specific growth rate and maximum dry weights affected by increasing metal concentrations were determined to obtain precise patterns of resistance. Metal biosorption was also monitored during the course of growth in synthetic media containing respective metals at 0.5 mM final concentration. Although Zn- and Co-binding was negligible, as much as 90% of silver, 60% of copper, and 65% of cadmium were found to be absorbed by the end of active growth. Analysis of the protein profiles ofS. cerevisiae andK. marxianus on metal exposure suggested constitutive production of metallothionein inK. marxianus. Furthermore, a smaller protein synthesized byK. marxianus on induction by silver or cadmium accounts for the high resistance of the organism to these metals.  相似文献
