The lethal effects of high-intensity airborne sonic (9.9 kc/sec) and ultrasonic waves (30.4 kc/sec) on spores of Bacillus subtilis var. niger ATCC 9372 were determined. The spores, which were deposited on filter-paper strips, were exposed to sound waves for periods varying from 1 to 8 hr, at a temperature of 40 C and a relative humidity of 40%. Significant reductions in the viable counts of spores exposed to airborne sonic or ultrasonic irradiations were obtained. The antibacterial activity of airborne sound waves varied with the sound intensity level, the period of irradiation, and the distance of the sample from the sound source. At similar intensity levels, the amplitude of motion of the sound waves appeared to be a factor in acoustic sterilization.  相似文献

Biophysical wave stimulus has been used as an effective tool to promote cellular maturation and differentiation in the construction of engineered tissue. Pulsed electromagnetic fields (PEMFs) and sound waves have been selected as effective stimuli that can promote neural differentiation. The aim of this study was to investigate the synergistic effect of PEMFs and sound waves on the neural differentiation potential in vitro and in vivo using human bone marrow mesenchymal stem cells (hBM–MSCs). In vitro, neural‐related genes in hBM–MSCs were accelerated by the combined exposure to both waves more than by individual exposure to PEMFs or sound waves. The combined wave also up‐regulated the expression of neural and synaptic‐related proteins in a three‐dimensional (3‐D) culture system through the phosphorylation of extracellular signal‐related kinase. In a mouse model of photochemically induced ischemia, exposure to the combined wave reduced the infarction volume and improved post‐injury behavioral activity. These results indicate that a combined stimulus of biophysical waves, PEMFs and sound can enhance and possibly affect the differentiation of MSCs into neural cells. Our study is meaningful for highlighting the potential of combined wave for neurogenic effects and providing new therapeutic approaches for neural cell therapy. © 2016 American Institute of Chemical Engineers Biotechnol. The rates of propylene oxidation to propylene oxide by Methylococcus capsulatus (Bath) have been optimized in small shake flasks to establish the potential of this process for industrial application. It was shown that addition of the electron donors methanol, formaldehyde, formate or hydrogen stimulated the endogenous rate of propylene oxide formation 10 to 50 fold. Rates in excess of 500 nmol min-1 mg dry weight cells-1 have been obtained using methanol as the donor. These high rates could only be sustained for 3 to 4 min before loss of biocatalytic activity caused the rate of propylene oxide production to decline.  相似文献

Here, the effects of combinatorial cancer therapy including radiotherapy (RT) and radiofrequency (RF) hyperthermia in the presence of gold-coated iron oxide nanoparticles (Au@IONPs), as a thermo-radio-sensitizer, are reported. The level of cell death and the ratio of Bax/Bcl2 genes, involved in the pathway of apoptosis, were measured to evaluate the synergistic effect of Au@IONPs-mediated RF hyperthermia and RT. MCF-7 human breast adenocarcinoma cells were treated with different concentrations of Au@IONPs. After incubation with NPs, the cells were exposed to RF waves (13.56 MHz; 100 W; 15 min). At the same time, thermometry was performed with an infrared (IR) camera. Then, the cells were exposed to 6 MV X-ray at various doses of 2 and 4 Gy. MTT (3-[4,5-dimethylthiazol-2-y1]-2,5-diphenyltetrazolium bromide) assay was performed to evaluate cell viability and quantitative real-time polymerase chain reaction (qRT-PCR) was used to determine the expression ratio of Bax/Bcl2. Cellular uptake of nanoparticles was confirmed qualitatively and quantitatively. The results obtained from MTT assay and qRT-PCR studies showed that NPs and RF hyperthermia had no significant effect when applied separately, while their combination had synergistic effects on cell viability percentage and the level of apoptosis induction. A synergistic effect was also observed when the cancer cells were treated with a combination of NPs, RF hyperthermia, and RT. On the basis of the obtained results, it may be concluded that the use of magneto-plasmonic NPs in the process of hyperthermia and RT of cancer holds a great promise to develop a new combinatorial cancer therapy strategy.  相似文献

The propagation of sound waves along relaxed single fibres of glycerinated rabbit psoas muscle has been observed using Brillouin scattering at frequencies up to 1.6 GHz. Two types of waves were observed: one with a velocity of 1508±7 m s–1, which is attributed to sound waves in intra-cellular saline, the other with a velocity of 912±25 m s–1, which is attributed to waves propagating along the protein filaments within individual sarcomeres. The latter sound velocity is much higher than that which has been reported by Stienen and Blangé (1985) for 50 s tension transients, and the difference is attributed to the much higher stiffness of the protein filaments compared to the cross-bridges which determine the low-frequency elasticity of muscle fibres.  相似文献

The effects of salicylic acid (SA), sodium nitroprusside (SNP), a nitric oxide donor, and their combination (SA+SNP) on some physiological parameters of 23-day-old soybean seedlings grown under saline and nonsaline conditions were studied. The changes in the leaf area, shoot fresh and dry weights, contents of chlorophylls and carotenoids, amounts of MDA and hydrogen peroxide showed that the addition of 100 μM SA and/or 100 μM SNP markedly declined the oxidative damage to soybean plants induced by 50 and 100 μM NaCl. Our results proved that combined action of SA and nitric oxide donor significantly activated catalase (CAT), ascorbate peroxidase (APX), and guaiacol peroxidase (GPX), which contributed to the decay of H2O2 in soybean leaves under NaCl toxicity. The protective action of (SA+SNP) against saltinduced oxidative damage was often more efficient than effects of SA and SNP alone. We also observed that the accumulation of proline was apparently accelerated by these substances under salt stress. As well, it was observed that the interaction between SA and nitric oxide had synergistic effects in decreasing of the damages induced by NaCl salinity.  相似文献

Methylococcus capsulatus (Bath) possesses methane monooxygenases (soluble - (sMMO) and particulate - (pMMO)) which are able to catalyse the epoxidation of propylene to propylene oxide. In a previous paper we have shown that the production of the epoxide caused a rapid inactivation of the bioconversion process (Stanley et al, 1992). This paper shows that cultures containing pMMO, inactivated by propylene oxide production, could be completely reactivated in the presence of growth substrates within 5 h after the removal of propylene oxide so long as the propylene oxide production rate was below 150 nmol min-1 [mg dry weight cells]-1. Reactivation under these conditions was detectable within 30 min of propylene oxide removal. On the other hand, cells inactivated by propylene oxide production rates in excess of 150 nmol min-1 [mg dry weight]-1 did not begin to recover activity within the 30 min period. Furthermore a lag period was observed before reactivation began which was dependent upon the initial production rate. Cultures possessing sMMO took twice as long to recover their activity compared with cells containing pMMO.

Reactivation of propylene oxide production could occur without growth, but the process required the presence of a carbon and energy source (methane or methanol), sulphur, nitrogen and oxygen, although copper (which is normally involved in pMMO activity) was not required. It was shown that de novo protein synthesis was required for reactivation of activity.

Production rates of 12 g 1-1 d-1 could be maintained for longer than three weeks in a single phase production process and rates up to 30 g 1-1 d-1 were achieved in a two stage process. Using Methylocystis parvus (OBBP) rates of up to 90 g 1-1 d-1 were attained over a one week period.  相似文献

Evidence for a requirement for CO2 in the productive metabolism of aliphatic alkenes and epoxides by the propylene-oxidizing bacterium Xanthobacter strain Py2 is presented. In the absence of CO2, whole-cell suspensions of propylene-grown cells catalyzed the isomerization of propylene oxide (epoxypropane) to acetone. In the presence of CO2, no acetone was produced. Acetone was not metabolized by suspensions of propylene-grown cells, in either the absence or presence of CO2. The degradation of propylene and propylene oxide by propylene-grown cells supported the fixation of 14CO2 into cell material, and the time course of 14C fixation correlated with the time course of propylene and propylene oxide degradation. The degradation of glucose and propionaldehyde by propylene-grown or glucose-grown cells did not support significant 14CO2 fixation. With propylene oxide as the substrate, the concentration dependence of 14CO2 fixation exhibited saturation kinetics, and at saturation, 0.9 mol of CO2 was fixed per mol of propylene oxide consumed. Cultures grown with propylene in a nitrogen-deficient medium supplemented with NaH13CO3 specifically incorporated 13C label into the C-1 (major labeled position) and C-3 (minor labeled position) carbon atoms of the endogenous storage compound poly-beta-hydroxybutyrate. No specific label incorporation was observed when cells were cultured with glucose or n-propanol as a carbon source. The depletion of CO2 from cultures grown with propylene, but not glucose or n-propanol, inhibited bacterial growth. We propose that propylene oxide metabolism in Xanthobacter strain Py2 proceeds by terminal carboxylation of an isomerization intermediate, which, in the absence of CO2, is released as acetone.  相似文献

The effects of ethylene oxide and propylene oxide block copolymers (pluronics) on the permeability of several weak acids and bases through bilayer lipid membranes have been studied by the methods of monitoring (1) pH shifts near planar bilayers, (2) doxorubicin fluorescence quenching inside liposomes, and (3) current transients in the presence of hydrophobic anions. It has been shown that pluronics facilitate the permeation of comparatively large molecules (such as 2-n-undecylmalonic acid and doxorubicin) across lipid bilayers, while the permeation of small solutes (such as ammonium and acetic acid) remains unaffected. Pluronics also accelerate the translocation of large hydrophobic anions (tetraphenylborate). The effect of pluronics correlates with the content of propylene oxide units: it is enhanced when the portion of polypropylene oxide block in the copolymer is increased. The action of the pluronic on lipid membrane permeability differs from the effect of the conventional detergent Triton X-100, which does not affect doxorubicin transport if added at concentrations similar to those used for pluronics. It has been proposed that pluronics accelerate the processes of solute diffusion within lipid bilayers (in a structure-dependent manner) rather than influencing the rate of solute adsorption/desorption on the membrane surface. We suppose that the effect of pluronics on doxorubicin permeation across lipid bilayers along with the known effect on the multidrug resistance protein determines its influence on the therapeutic activity of anthracycline drugs.  相似文献

New methods of sterilization employing a chemical with moderate heat and ultrasonic energy have been devised. Inactivation of high-density bacterial spore suspensions is achieved by treatment with low concentration aqueous acid glutaraldehyde solutions at temperatures above or about 54 C. Low (20 kHz) or high (250 kHz) frequency ultrasonic energy is synergistic with glutaraldehyde. Rapid inactivation may also be achieved by using ultrasonic energy and aqueous alkalinized glutaraldehyde solutions at low (25 C) or moderate (55 C) temperatures. If compared to present room temperature techniques, "surface sterilization" time for contaminated objects can be reduced from hours to minutes.  相似文献

The effect of three concentrations, 1, 2 and 4 per cent, of propylene glycol has been investigated on human lymphocytes in vitro. An increased ratio of anaphase cells was observed at 4 per cent propylene glycol in the culture medium, when added for the last 24 h in 72 h blood cultures. No change in chromosome number has concomitantly been observed. A weak stathmokynetic action was induced by the same concentration of propylene glycol if administered for the final two hours of cell cultivation. These effects could be due to a delay in the normal sequence of the sister chromatid separation during mitosis, induced by the highest drug concentration.  相似文献

The action of interferential current (IFC) upon intracellular content of cyclic adenosine monophosphate (cAMP) after prestimulation with the chemotactic peptide N-formyl-methionyl-leucyl-phenylalanine (fMLP) has been investigated. Human promyelocytes (HL-60) differentiated to granulocytes by dimethylsulphoxide (DMSO) have been treated with different concentrations of fMLP. This enhances their cAMP content. The half maximal effective concentration (EC50) was about 4 nM. Exposure to IFC with modulation frequencies of 35 and 125 Hz (5 min, 250 microA/cm2) after prestimulation with various concentrations of fMLP had no enhancing effect at low or high concentrations of fMLP. In contrast, at medium concentrations in the range of about 100 pM fMLP, a significant enhancement of cAMP could be observed. This synergistic effect of fMLP and IFC has been examined in detail by varying the modulation frequency, current density, and exposure time. Enhancement of cAMP content could be observed at certain modulation frequencies and exposure times suggesting a window effect, whereas for the current density in the range measured (8.5 microA/cm2-2.5 mA/cm2) a constant enhancement could be observed. The synergistic effect of fMLP and IFC could only be observed in the treatment sequence of fMLP followed by IFC; an inverse sequence had no effect on the cAMP content. .  相似文献

Worldwide, cyanobacterial blooms have been increasing in intensity and frequency, with toxic cyanobacteria sometimes dominant throughout the year in many freshwater bodies. Since the coexistence of more than one type of cyanotoxins in freshwater environments is a common phenomenon, studies on the joint effects of these toxins would be very useful. In this study, the single and combined effects of two cyanotoxins with different modes of action (hepatotoxic and neurotoxic) on the survival (lethal exposure) and feeding (sublethal exposure) of the cladoceran Daphnia magna were investigated. With the single exposures, it was observed that both the survival and feeding activity of the daphnids were impaired by the hepatotoxic and neurotoxic extracts at environmentally relevant concentrations. In the combined exposures, both survival and feeding rate endpoints showed a good fit to the independent action model. For the acute assay and 24 h exposure period in the feeding inhibition test, there was no interaction between components of the hepatotoxic and neurotoxic extracts, although a slight tendency to a synergistic deviation could be seen in the feeding rates. On the other hand, for the 4 h post-exposure period, a synergistic deviation was found in feeding rates at all mixture concentrations tested. Hence, the combined exposure of hepatotoxins and neurotoxins should also be taken into account in risk assessments of freshwater bodies, since the mixture of these toxins can result in more severe post-exposure effects on the feeding of daphnids than the sum of those expected for single exposures.  相似文献

Biocatalytic resolution of 3-(2′-nitrophenoxy)propylene oxide (1a), 3-(3′-nitrophenoxy)propylene oxide (1b) and 3-(4′-nitrophenoxy)propylene oxide (1c) were exploited by using lyophilized cells of yeast Trichosporon loubierii ECU1040 with epoxide hydrolase (EH) activity, which preferentially hydrolyzes (S)-enantiomers of the epoxides (1a–c), yielding (S)-diols and (R)-epoxides. The activity increased as the nitro group in the phenyl ring was shifted from 4′-position (1c) to 2′-position (1a). When the substrate concentration of 1a was increased from 10 to 80 mM, the E-value increased at first, until reaching a peak at 40 mM, and then decreased at higher concentrations (>40 mM). The optically active epoxide (R)-1a was prepared at gram-scale (97% ee, 41% yield). Furthermore, a simple method was developed to predict the enantiomeric excess of substrate (ees) at any time of the whole reaction course based on the ees value determined at a certain reaction time at a relatively lower substrate concentration. This will be helpful for terminating the reaction at a proper time to get both higher optical purity and higher yield of the remaining epoxides.  相似文献

The effects of acoustic and visual stimuli and their synergistic effects on heart rate variability including gender differences were investigated. Of particular interest was the influence of visual stimulus on heart rate variability during listening to simple sounds of different characters. Twelve male and 12 female university students were selected as subjects. The subjects listened at rest to 7 different figures of sound at loudness levels averaging 60 dB. Beat-to-beat R-R intervals were continuously recorded under the closed-eye condition (CEC) and the open-eye condition (OEC) prior to, during, and immediately after the exposure to acoustic stimuli. Low frequency (LF) power was defined over 0.04-0.15 Hz and high frequency (HF) power over 0.15-0.40 Hz. Cardiac autonomic function was estimated by plotting LF/HF in standard measure against HF in standard measure and by plotting LF/HF (%) against HF (%), accompanied by a demarcated central area. Values of LF/HF tended to be smaller under CEC than under OEC. Values of HF while listening to a 110 Hz sine wave under CEC were significantly greater than values for 880 Hz and 3520 Hz sine waves, or for 110 Hz or 880 Hz sawtooth waves, under OEC. Under CEC, values of HF for 7 figures of sound were greater in females than in males. The value of HF of sine wave for 110 Hz under CEC and OEC was significantly greater than that for white noise under the OEC. The results suggest that the cardiac parasympathetic nervous activity during auditory excitation increases with elimination of visual stimuli and tends to be greater in females than in males.  相似文献

Propylene oxide is used extensively in the chemical and food manufacturing industries, but relatively little is known of its ability to interact with genetic material. Studies were undertaken to investigate its ability to induce gene mutations and primary DNA damage in bacteria and chromosomal damage in mammalian cells. The induction of base-substitution mutations was demonstrated in spot tests with strains of Salmonella typhimurium and Escherichia coli at 700 micrograms/plate of propylene oxide; inclusion of a preparation of rat-liver microsomes and cofactors (S9 mix) was without significant effect on this response. A linear dose--response relationship was recorded in plate tests with S. typhimurium strains TA100 and TA1535 over the range 100--750 micrograms/plate. After addition to dividing lymphocytes in cultures established from human peripheral blood, propylene oxide caused dose-related chromosomal damage, detected at 1.85 and 9.25 micrograms/ml. Oral administration of propylene oxide at 2 x 100, 2 x 250 or 2 x 500 mg/kg to male mice produced no detectable increases in the incidence of micronucleated, polychromatic erythrocytes in bone marrow. A male mouse dominant lethal test employing oral doses of 50 or 250 mg/kg/day for 14 days gave no evidence of mutagenic action on sperm. Intraperitoneal injections of propylene oxide at 2 x 300 mg/kg induced increased numbers of micronucleated erythrocytes in mice, but lower doses given by this route had no such effect. Possible reasons for the contrasting findings in vitro and in vivo are discussed.  相似文献

Beneficial actions of nitric oxide (NO) in failing myocardium have frequently been overshadowed by poorly documented negative inotropic effects mainly derived from in vitro cardiac preparations. NO's beneficial actions include control of myocardial energetics and improvement of left ventricular (LV) diastolic distensibility. In isolated cardiomyocytes, administration of NO increases their diastolic cell length consistent with a rightward shift of the passive length-tension relation. This shift is explained by cGMP-induced phosphorylation of troponin I, which prevents calcium-independent diastolic cross-bridge cycling and concomitant diastolic stiffening of the myocardium. Similar improvements in diastolic stiffness have been observed in isolated guinea pig hearts, in pacing-induced heart failure dogs, and in patients with dilated cardiomyopathy or aortic stenosis and have been shown to result in higher LV preload reserve and stroke work. NO also controls myocardial energetics through its effects on mitochondrial respiration, oxygen consumption, and substrate utilization. The effects of NO on diastolic LV performance appear to be synergistic with its effects on myocardial energetics through prevention of myocardial energy wastage induced by LV contraction against late-systolic reflected arterial pressure waves and through prevention of diastolic LV stiffening, which is essential for the maintenance of adequate subendocardial coronary perfusion. A drop in these concerted actions of NO on diastolic LV distensibility and on myocardial energetics could well be instrumental for the relentless deterioration of failing myocardium.  相似文献

Exposure to propylene oxide was determined previously by the degree of alkylation of hemoglobin measured on the histidine residue as N-3-(2-hydroxypropyl) histidine, using blood samples from 8 propylene oxide-exposed employees and 13 unexposed referents. Mononuclear leukocytes isolated from the same blood samples were used to quantify DNA repair proficiency following an in vitro challenge with the carcinogen, N-acetoxy-2-acetylamino-fluorene. Decreases in the DNA repair proficiency index correlated significantly to in vivo exposure levels to propylene oxide (r = –0.64, p <0.03). 1. Sound production by aquatic insects is found in four orders — Trichoptera, Odonata, Heteroptera and Coleoptera. 2. Immature aquatic insects that produce sound are rare, stridulation being present in one family of Trichoptera (Hydropsychidae) and one genus and species in a relic suborder of Odonata (Anisozygoptera) - Epiophlebia superstes. Hydropsychid larvae produce sound with a head/fore femur mechanism and use sound as part of aggressive behaviour for defence of feeding nets. Larval E. superstes use a hind femur/abdominal mechanism to dissuade predators. 3. Sound production has been documented in adults of all families of aquatic Heteroptera except Helotrephidae. In corixids and notonectids, acoustic signals play a role in mating. Members of the genus Buenoa (Notonectidae) are unique in having two stridulatory mechanisms in the same individual. Sound production has been most intensively studied in the Corixidae. Although sounds are used in mating by all singing corixids, their use seems to be facultative in some species and obligatory in others. Recent experiments by Theiss (1982) have shown that the air stores carried by corixids are used for both sound radiation and reception. 4. The adephagan beetle families Hygrobiidae, Dytiscidae and Haliplidae have all been shown to produce sound. Mechanisms of sound production have been established for haliplids and hygrobiids but have yet to be for most dytiscids. Sound production is used by beetles as part of sequences of aggressive/defensive and reproductive behaviour. 5. Sound production is especially well documented in the Hydrophilidae (Polyphaga). Hydrophilids use an abdominal/elytral mechanism and sound appears to be used in the same contexts as in adephagans. 6. Insects that produce sound under water must contend with the physical problems of sound transmission in a relatively dense, viscous medium with sharp boundaries. Because of potential distortion of the frequency components in a signal by reflection from the air/water interface in very shallow water, frequency is unreliable for encoding information. Aquatic insects use instead amplitude modulation and temporal patterning of signals. 7. For aquatic invertebrates, sound fields are different than those in air because the extent of the near field is approximately four times greater in water. This near field, a region in which displacement waves are predominant over pressure waves, extends to a greater distance than most aquatic insects communicate over. Such displacement waves could have important but as yet unconsidered effects. 8. The mass and viscosity of the water dictates that sound producing structures of aquatic insects should be heavier and more massive than those of terrestrial insects. A survey of stridulatory organs of aquatic insects reveals this to be true and reveals that the relatively fragile, membranous stridulatory organs of some terrestrial insects (especially Orthoptera) are absent. 9. The elaboration of sound producing structures in aquatic insects probably occurred at the family or subfamily level and for Heteroptera, Trichoptera and Odonata evolved after the invasion of the water. Acoustic signals used reproductively would probably be more closely associated with the emergence of new taxa. 10. Stridulatory structures have been derived from either structures devoted to some other function or from structures involved in the behaviour currently enhanced by sound production.  相似文献

Diethylcarbamazine is a drug that is used for the treatment of filariasis in humans and animals; it also has effects on intestinal nematodes, but its mechanism of action remains unclear. Emodepside is a resistance-busting anthelmintic approved for treating intestinal parasitic nematodes in animals. The novel mode of action and resistance-breaking properties of emodepside has led to its use against intestinal nematodes of animals, and as a candidate drug for treating filarial parasites. We have previously demonstrated effects of emodepside on SLO-1 K+-like currents in Ascaris suum. Here, we demonstrate that diethylcarbamazine, which has been proposed to work through host mediated effects, has direct effects on a nematode parasite, Ascaris suum. It increases activation of SLO-1 K+ currents and potentiates effects of emodepside. Our results suggest consideration of the combination of emodepside and diethylcarbamazine for therapy, which is predicted to be synergistic. The mode of action of diethylcarbamazine may involve effects on parasite signaling pathways (including nitric oxide) as well as effects mediated by host inflammatory mediators.  相似文献
