Distribution, population structure, and abundance of Amur kaluga Acipenser dauricus and Amur sturgeon A. schrenckii in the Lower Amur and Amur Estuary have been considered based on the materials of two surveys performed in 2011. Both species occur along almost the entire length of the Lower Amur and in major part of the Amur Estuary. The density of their aggregations has significantly decreased in most regions of the river if compared to the 1960s. In the Amur Estuary, A. dauricus and A. schrenckii are most abundant in freshwater and light salted water bodies of its western part. In 2011, the abundance and biomass of A. dauricus and A. schrenckii in the Lower Amur and Amur Estuary was 345000 specimens and 7110 tons and 289000 specimens and 1946 tons, respectively.  相似文献

A RAPD-PCR analysis of a sample of Amur sturgeon fish (46 individuals) is performed. An estimate of the genetic state of native populations of Amur sturgeon Acipenser schrenckii (Brandt, 1869) and kaluga fish Huso dauricus (Georgi, 1775) is given. Genetic evidence for the hybrid origin of two phenotypical hybrids is obtained. Estimates of the genetic distances between the species and the hybrids proved to be at the level of interspecific distances. It is shown that an Exact Test for differentiation of the populations is the most effective means of discriminating the two species, while multidimensional scaling (MDS) is the most effective means of discriminating the hybrids. It is concluded that the populations of sturgeon fish in the Amur River have maintained a substantial level of genetic diversity; the presence of hybrids in these populations is judged to be one of the risk factors. Multilocus RAPD-PCR markers are acknowledged as a convenient and reliable tool for conducting genetic monitoring of populations of Amur sturgeon fish for the purpose of preserving the gene pool of the fish.  相似文献

The complete mitochondrial genome of the Sakhalin sturgeon Acipenser mikadoi and two mitogenomes of the Amur sturgeon A. schrenckii were sequenced using Roche 454 technology. The mitogenomes of the green sturgeon A. medirostris (obtained from GenBank) and the Sakhalin sturgeon differ as much as the mitogenomes of two mtDNA haplogroups (SM and BG) found in the same population of the Amur sturgeon: 0.0042 ± 0.0006 and 0.0036 ± 0.0005 substitutions per site (Tamura–Nei distance, TrN), respectively. The differences of these mitogenome pairs from mitogenomes of sister species (kaluga A. dauricus and white sturgeon A. transmontanus) are 3–6 times larger: 0.0260 ± 0.0015 and 0.0102 ± 0.0008, respectively. Thus, the differences between the mitogenomes of the Sakhalin and green sturgeons can be attributed to the variability at the intraspecific level. The time that has passed since the divergence of the Sakhalin and green sturgeons is considered to be much shorter than was previously believed: approximately 0.16 rather than 9.60 million years.  相似文献

Three Acipenseridae species live in the rivers and marine waters of the Khabarovsk Territory, Russia: Sakhalin sturgeon, Acipenser mikadoi, Amur sturgeon, A. schrenckii, and kaluga, Huso dauricus. We review the general biology and life history of each species, including their historic and current distributions, and examine current paleogeographic theories to outline the possible origin and evolution of these three species in the Amur River, Sea of Okhotsk, and Sea of Japan biogeographic province. Apparently, these species have evolved during distinct geologic time periods, which has reinforced the reproductive isolation of these species although hybridization does occur. They have convergently adapted to the unique environmental conditions of the Amur River and Russian Maritime regions, and yet developed behavioral adaptations to reduce competition between species. Sakhalin sturgeon is the least studied species among anadromous sturgeon in the world. This species is highly migratory and spends the majority of its life in the ocean only returning to natal rivers to spawn. Amur sturgeon and kaluga are distributed throughout the Amur River basin and the estuary and share many life history traits. They are both represented by distinct morphs. Additionally, we present size and weight relationships to estimate the growth of Amur sturgeon and kaluga. All three species have suffered declines in abundance due to over fishing and their contemporary distributions have contracted compared to their historic ranges. We identify gaps in knowledge and suggest further research useful for guiding management of each species.  相似文献

This study investigates muscle proteins in Russian (Acipenser gueldensiaedtii Brandt), Siberian (Acipenser baerii Brandt), and Amur (Acipenser schrenckii Brandt) sturgeons using the method of polyacrylamide gel electrophoresis. A group of fractions, which is presumably the product of a polyallelic duplicated MY-1* locus, is recorded among the general pool of proteins. Data from densitometric analysis makes it possible to put forward a hypothesis about an eight-gene determination of this system of proteins. The highest heterogeneity and polymorphism was revealed in the Russian sturgeon. Analysis of the genotype distribution in samples of the studied species allowed us to make an assumption that the genetic structure of spawning sturgeon stocks is significantly metamorphosed under the influence of anthropogenic and natural factors.  相似文献

Genetic characterization was performed in five individuals of wild Amur sturgeon Acipenser schrenckii, and/or its presumed hybrid caught around Hokkaido, using a mitochondrial DNA (mtDNA) marker and two markers of nuclear DNA (nDNA). Genetic analyses indicated that two of the five fish had the mtDNA haplotype of Kaluga, Huso dauricus, whereas the nDNA markers indicated signs for both A. schrenckii and H. dauricus genotypes, referring to a hybrid origin. The other three fish were plausibly pure A. schrenckii. The results indicated the importance of combined usage of mtDNA and nDNA markers for correct species identification in sturgeon.  相似文献

Present study was performed to identify the species of ascarids from macaw parrot, Ara chloroptera, in China. Total 6 ascarids (3 males and 3 females) were collected in the feces of 3 macaws at Guangzhou Zoo in Guangdong Province, China. Their morphological characteristics with dimensions were observed under a light microscope, and their genetic characters were analyzed with the partial 18S rDNA, ITS rDNA and nad4 gene sequences, respectively. Results showed that all worms have no interlabia but male worms have two alate spicules, well-developed precloacal sucker and a tail with ventrolateral caudal alae and 11 pairs of papillae. The partial 18S rDNA, ITS rDNA and nad4 sequences were 831 bp, 1015 bp and 394 bp in length, respectively. They showed the highest similarity of 99.8% (18S rDNA) with Ascaridia nymphii, 93.8% identities (ITS rDNA) with A. columbae and 98.5% to 99.5% identities (nad4) with Ascaridia sp. from infected parrot. All Ascaridia nematodes from the macaws were clustered into one clade and formed monophyletic group of Ascaridia with A. columbae and A. galli in two phylogenetic trees. It is observed that the combining morphological and sequencing data from three loci, the present Ascaridia species was identified as Ascaridia nymphii, which is the first record of A. nymphii from macaw parrot in China.  相似文献

The six North Pacific members of the genus Acipenser have generally similar environmental requirements and share several specific morphological features. This Pacific group consists of green sturgeon, Acipenser medirostris, Sakhalin sturgeon, A. mikadoi, white sturgeon, A. transmontanus, Chinese sturgeon, A. sinensis, Dabry’s sturgeon, A. dabryanus and Amur sturgeon, A. schrenkii. A qualitative morphological examination of these species revealed synapomorphic characters that suggest close phyolgenetic relationships resulting from common ancestry of eastern and western species within the Pacific Rim. An evolutionary taxonomic approach, considering a reduction of characters from east to west, would suggest an Asian ancestry for all Pacific sturgeons. In contrast however, a phylogenetic approach using derived character states suggests a North American ancestry. Besides a common ancestry for Chinese and American sturgeons, the taxonomic status of A. mikadoi is discussed.  相似文献

The method of polymerase chain reaction with random primers (RAPD PCR) was used to identify the progeny of the crosses between three sturgeon species, Amur sturgeon (Acipenser schrenckii Brandt, 1869), Siberian sturgeon (A. baerii Brandt, 1869), and sterlet (A. ruthenus Linnaeus, 1758). Using ten primers, genetic variation in 70 yearlings, produced in seven individual crosses: Acipenser schrenckii × A. schrenckii, A. baerii × A. baerii, A. ruthenus × A. ruthenus, A. schrenckii × A. baerii, A. baerii × A. schrenckii, A. schrenckii × A. ruthenus, and A. ruthenus × A. schrenckii was described and evaluated. It was demonstrated that the samples composed of hybrids from individual crosses were more variable than the samples of parental species. On the other hand, pooled samples of hybrids from two cross directions were genetically less variable than the pooled samples of their parents. The three main features of the hybrid RAPD profiles identified included: (1) preservation of marker DNA fragments of both parents in one genome; (2) presence of specific DNA fragments, absent from both parents; and (3) dependence of the frequency of some DNA fragments from the cross direction. Multidimensional scaling clearly distinguishes in the space of three coordinates the individuals of original species and the hybrid progeny with differentiation in the groups of direct and backcross hybrids. Analysis of relationships (UPGMA and NJ) pointed to substantial differentiation between the species, as well as between the species and hybrid progeny. Close genetic relationships between direct and backcross hybrids were demonstrated. Multilocus RAPD markers in association with statistical methods are considered to be the useful tool for discrimination of interspecific hybrids of sturgeon. In this study, six candidate female-specific DNA sequences of octaploid Amur sturgeon (Acipenser schrenckii) were identified using comparative genomic approaches with high-throughput sequencing data. Their specificity was confirmed by traditional PCR. Two of these sex-specific sequences were also validated as female-specific in other eight sturgeon species and two hybrid sturgeons. The identified female-specific DNA fragments suggest that the family Acipenseridae has a ZZ/ZW sex-determining system. However, one of the two DNA sequences has been deleted in some sturgeons such as Sterlet sturgeon (Acipenser ruthenus), Beluga (Huso huso) and Kaluga (H. dauricus). The difference of sex-specific sequences among sturgeons indicates that there are different sex-specific regions among species of sturgeon. This study not only provided the sex-specific DNA sequences for management, conservation and studies of sex-determination mechanisms in sturgeons, but also confirmed the capability of the workflow to identify sex-specific DNA sequences in the polyploid species with complex genomes.  相似文献

Based on a taxonomic approach, combining morphological characters with DNA sequences (i.e.,18S rDNA, ITS1, 5.8S rDNA and ITS2), Susanlimocotyle n. gen. is proposed to accommodates Susanlimocotyle narina n. sp. from the nostrils of the ariid Sciades herzbergii (Bloch) from the coast of the state of Pará, Brazil. Susanlimocotyle n. gen. is characterized by species possessing: an intestinal ceca confluent posteriorly; a male copulatory organ, comprising a variable tube, articulated with the accessory piece; a sclerotized vagina, vaginal aperture dextro-ventral; an onchium; a robust ventral bar; two dorsal bars; a ventral anchor with elongated shaft and a dorsal anchor with deep root expanding into wings. In addition, new molecular data of Chauhanellus spp. are also provided and used for the evaluation of the phylogenetic relationships among monogenoids parasitizing siluriforms. Susanlimocotyle n. gen. exhibited a higher genetic divergence level for 18S rDNA (4.6 to 7.2% [83–130 bp]) with respect to Chauhanellus spp. despite sharing S. herzbergii as a host, than Hamatopeduncularia spp., (4.1 to 5.8% [75–110 bp]) from Oriental ariids. For the 18S rDNA, 5.8S rDNA, ITS1 and ITS2 regions, C. boegeri and C. susamlimae were observed to have the smallest interspecific distances, and C. velum was revealed to be the most genetically distant species to Chauhanellus. The proposal for Susanlimocotyle n. gen. is also supported by phylogenetic analysis based on the 18S rDNA gene, which supports the close relationship between the new genus and Hamatopeduncularia and Chauhanellus from ariids from the South America and Oriental regions. Acipensericola glacialis n. sp. infects the heart of lake sturgeon, Acipenser fulvescens (Rafinesque), in the Lake Winnebago System and differs from its only congener, Acipensericola petersoni Bullard, Snyder, Jensen & Overstreet, 2008, by having a dendritic intestine, deeply-lobed testes, a post-ovarian oötype, and a common genital pore that is medial to the dextral caecum. Acipensericola petersoni has a non-dendritic intestine, testes that are not deeply lobed, an oötype that is at level of the ovary (ventral to the ovary), and a common genital pore that is dorsal to the dextral caecum. Comparison of the large (28S) and small (18S) sub-unit ribosomal DNA and internal transcribed spacer 2 (ITS2) regions between specimens of A. glacialis n. sp. and A. petersoni revealed 13 (of 1,621 nt; 99.2% similarity in the 28S), 8 (of 1,841 nt; 99.9% similarity in the 18S), and 11 (of 442 nt; 97.5% similarity in the ITS2) nucleotide differences. Collectively, these results comprise an unexpectedly high degree of morphological and molecular similarity given the geographical (Mississippi River Basin vs Great Lakes Basin) and phylogenetic (Polyodontidae vs Acipenseridae) separation of these hosts but seemingly did not reject a previous hypothesis concerning lake sturgeon dispersal from the Mississippi Refugium following the Wisconsin glaciation ~18,000 years ago. The karyology of species of sturgeon from the Russian Far East demonstrates that the karyotype of the Sakhalin sturgeon (Acipenser mikadoi) includes 262 ± 4 chromosomes with 80 biarmed chromosomes and the number of chromosome arms (NF) 342 ± 4, the karyotype of the Amur sturgeon (A. schrenckii) includes 266 ± 4 chromosomes with 92 biarmed chromosomes and NF 358 ± 4, and the karyotype of the kaluga (A. dauricus) consists of 268 ± 4 chromosomes with 100 biarmed chromosomes and NF 368 ± 4. These results prove that all western Pacific sturgeon species are from a tetraploid origin, based on a recent ploidy scale. This suggests that at least three polyploidization events have occurred during the evolution of Acipenseridae. However, if polyploid species originated by hybridization between diploid species, there may have been more polyploidization events in this group of fishes.  相似文献

The 5S rDNA coding region and its spacer have been successfully utilized in phylogenetic studies of plants. However, it has not been utilized in the phylogenetic analysis of Cucurbitaceae. Here, we obtained the 5S rDNA sequences of 12 Cucurbitaceae species by direct PCR or cloning. The 5S rDNA sequences ranged from 275 to 359 bp, and the coding regions of all species were 120 bp long, except for that of Cucurbita, which was 119 bp. Some genus-specific SNPs were observed in the coding regions of Cucurbita, Lagenaria, Melothria, and Tricosanthes. The GC content of the coding regions was generally higher than that of the NTS regions, and the difference in GC content between the coding and NTS regions varied among species, with Gynostemma pentaphyllum having the greatest difference of 20.3. The phylogenetic trees generated using maximum parsimony and maximum likelihood were congruent and well supported by the recently published classification of Cucurbitaceae. These results demonstrated the utility of the 5S rDNA sequence in inferring phylogenetic relationships among 12 Cucurbitaceae species, and its utility could be extended by using a greater number of species in future studies.  相似文献

We investigated seasonal variation in the diet of hatchery-reared juvenile Amur sturgeon Acipenser schrenckii in the Tongjiang reach of the Songhua River, Northeast China. The results indicated that Amur sturgeon fed mainly on Chironomidae and Ephemeroptera and secondarily on unspecified bony fish, Odonata, and Trichoptera. Diet changed throughout the year, with autumn 2017 and spring 2018 dominated by Ephemeroptera larvae, while summer 2018 prey comprised primarily Chironomidae larvae. Unspecified bony fish were observed at greater frequency in autumn 2017. Amur sturgeon captured in summer contained a greater quantity of Trichoptera larvae compared to those from autumn and spring sampling. Results were consistent with previous reports of Amur sturgeon diet in other areas of its native distribution and suggested that they may be opportunistic feeders throughout the year, although larval Chironomidae and Ephemeroptera appear to be important prey. These results represent the first reported diet composition data for juvenile Amur sturgeon in the lower Songhua River. Results of the study form part of a comprehensive investigation of feeding habits of juvenile Amur sturgeon that provides basic data for study of the biological characteristics of the species.  相似文献

The relation of fecundity of females to size-weight and age indices is shown. Stocking density and hypoxia are considered priority issues in aquaculture research. In this study, two experiments were carried out in order to investigate the effects of chronic stress (stocking density) and acute stress (hypoxia) on the immune physiology responses (hematology, serum cortisol, glucose, total protein and the mRNA expression of CYP 1A) of juvenile Amur sturgeon (Acipenser schrenckii). In the chronic stress study, three triplicate groups of Amur sturgeon (42.0 ± 2.3 g) were reared in nine square concrete ponds (4.4 × 4.4 × 0.45 m3) at three stocking densities (3.7, 6.9 and 9.0 kg/m3) for 50 days. In the acute stress study, three triplicate groups: normal group (7 mg/l), hypoxia group 1 (5 mg/l) and hypoxia group 2 (3 mg/l) were used in nine 100 L indoor tanks. Sampling was performed at the end of the stocking density experiment (50 days) and at 0, 0.5, 1.5, 3 and 6 h after hypoxia stress. The results showed that increased stocking density reduced the morphological indexes (hepatosomatic index, spleen-somatic index and kidney-somatic index), while total protein and hemoglobin increased significantly in the stressed group. In response to hypoxia, the levels of cortisol, glucose and hematological parameters elevated significantly after this stress. As for spleen-somatic index, there was a decline after hypoxia though H1 group returned to the normal level at 3 h and 6 h after hypoxia stress. Additionally, In order to better understand the immune response of Amur sturgeon to chronic and acute stressors, we cloned the complete coding sequence of Amur sturgeon CYP 1A for the first time and investigated its tissue-specific expression and stress-induced expression. CYP 1A mRNA in liver showed over expressions both in crowding condition and in hypoxia stress. The same trend was also found in spleen and kidney which may provide evidence that CYP 1A could serve as a good indicator of immune response in Amur sturgeon. In addition, the result suggested a typical immune response both in high stocking density and hypoxia stress. But the chronically stressed fish might have an adaptation capability to survive under a stable crowding condition without a change in some immune parameters (cortisol, glucose, WBCs and RBCs).  相似文献

The 5S rDNA of plant is organized into clusters of tandem repeat units which include a coding region of 5S rRNA gene and variable sequences of nontranscribed spacer (NTS). In this study, we investigated sequence polymorphism and chromosomal localization of 5S rDNA in three cultivated varieties of sweet potato (Ipomoea batatas Lam.). Two different PCR products of 5S rDNA were amplified from all three varieties, as approximately 0.25 kb and 0.34 kb with multiples. In sequence analysis, the 5S rDNA ofI. batatas were discriminated from four consensus sequences by in reasonable sizes and molecular informative factors. Four consensus sequences were divided into three short sequences, including 263, 253, and 243 – 283 bp by sequence variation between 160 and 186 bp in NTS region, and one long sequence with 340 bp. To identify molecular relationship among varieties, phylogenetic analysis was applied. A total of 35 sequenced clones in this study were classified into four groups in phylogenetic tree. Interestingly, two varieties included all four groups, but one variety only two groups. To localize the physical map of 5S rDNA, fluorescencein situ hybridization (FISH) was performed in metaphase chromosomes of each varieties. In 90 chromosomes ofI. batatas, 6 loci of 5S rDNA were detected in chromosomes for all varieties. Our results will help to further more understand the genomic relationship inI. batatas, to investigate molecular relationship among varieties.  相似文献
