Changes in resource availability and biotic and abiotic stress may alter the defensive mechanisms of pine trees. The effect of fertilisation on the resin canal structure of Pinus pinaster seedlings established in two trials in NW Spain, one attacked by Hylobius abietis and the other non-attacked, was studied. The leaders of 50 plants were destructively sampled and the resin canal density, the canal area and its relative conductive area in the phloem and xylem were assessed. Experimentally increased nutrient availability significantly decreased resin canal density in the phloem of the seedlings in the two analysed trials, where unfertilised seedlings presented up to 30% more resin canal density than the fertilised seedlings (mean value ± SEM = 0.32 ± 0.02 resin canals mm−2 in the fertilised plants versus 0.45 ± 0.04 resin canals mm−2 in the control plants). Fertilisation had no effect on the resin canal system in the xylem, but significantly increased tracheid size. Significant differences of resin canals among sites were observed mainly in the xylem; the resin canal density was 1.7-fold greater in the attacked site than in the non-attacked site. The similar structure of phloem resin canals in both sites supports that phloem resin canals are constitutive mechanisms of defence in P. pinaster, whereas xylem resin canals would be constitutive mechanisms but also inducible mechanisms of resistance following the attack of pine weevils or bark beetles. Ophiostomatoid fungi colonize the conducting tissues of conifer stems, the phloem and the xylem. These pathogenic fungi penetrate into the stem through injuries made by xylophagous insects vectoring these pathogens. In this study the response of the phloem of Scotch pine (Pinus sylvestris L.) to wounding (treatment 1) was compared with the response to wounding combined with application of high-molecular-weight compounds isolated from the mycelium of the ophiostomatoid fungus Ceratocystis laricicola Redfern & Minter (treatment 2). Both treatments induced the appearance of necrosis in the inner bark, the formation of periderm separating living and dead tissues, and formation of the callus alongside the wound perimeter. In addition, the bark accumulated lignin, bound proanthocyanidins, and resins, with a parallel decrease in the content of free proanthocyanidins, low-molecular-weight carbohydrates, and non-lignin components of the cell wall (P > 0.95). The size of necrotic spots, as well as changes in the content of most substances, were significantly higher in the treatment 2 than in the treatment 1 (P > 0.95). The accumulation of lignin in cell walls of phloem sieve cells was delayed in the treatment 2 as compared with that in the treatment 1. This suggested that the mycelial extract temporarily inhibited lignification at the early stage of the wound response. This disturbance of the cell wall protective transformation led to the hypothesis that the fungal suppressors retard the repair of inner bark injured by insects, thereby favoring the invasion of conifer tissues by ophiostomatoid fungi.  相似文献

Source water utilized by four families of loblolly pine (Pinus taeda L.) was assessed by comparing the H isotope composition ('D) of xylem sap and of soil water from four depths (0-20 cm, 20-40 cm, 1.2 m, and 2.1 m) across 1 year. Soil water 'D values varied with soil moisture content in the well-drained, sandy site and at each of the four soil depths. In September and November 1997 and May through November 1998, xylem sap 'D values closely matched the soil water 'D values of the upper soil horizons (0-20 and 0-40 cm) and, therefore, reflected significant water uptake from upper regions of the soil profile. In March 1998, xylem sap 'D values closely matched the soil water 'D values of the 1.2 m soil depth and, therefore, indicated that trees during this portion of the growing season were obtaining their water from deep in the soil profile. Analysis of source water use with a two-ended mixing model in the 3 months of collection that exhibited a range of soil water 'D values across the soil profile confirmed that trees utilized different sources of water depending upon season of the year. In September 1997 and November 1998, source water uptake was primarily from the upper soil profile while in March 1998, source water uptake was from deep in the soil profile. With few exceptions, we did not find striking differences in source water use between drought-hardy families and those that were locally adapted.  相似文献

Two new species of Pyxidiophora (Ascomycota, Laboulbeniomycetes, Pyxidiophorales) associated with bark beetles (Coleoptera, Scolytidae) colonising the sapwood and secondary phloem of conifers (Larix decidua and Picea abies) in Europe are described. Each of both fungal species appeared to be connected with a Gabarnaudia-like anamorph. Pyxidiophora corallisetosa could only be cultivated in the presence of the hyphomycete Clonostachys rosea, and a connection of P. corallisetosa with its Gabarnaudia-like anamorph was proven by the development of perithecia after axenic transfer of conidia to cultures of C. rosea. Pyxidiophora cuniculicola was found in cultures originating from bark beetles placed onto agar media. Conidia developing directly from germinating ascospores were identical to those developing from Gabarnaudia-like conidiophores also present in these cultures, which indicates an anamorph-teleomorph connection. The probable interactions between both species of Pyxidiophora with associated fungi are discussed.  相似文献

The internal transcribed spacer (ITS) region was sequenced in symbiotic dinoflagellates (zooxanthellae) from five morphospecies in the genus Madracis. The phylogeny of the symbionts is congruent with a companion phylogeny of the coral host. Comparison with known clade B symbiont ITS types reveals that M. mirabilis contains the B13 symbiont and that the other morphospecies contain the B7 symbiont. Madracis formosa also contains a previously undescribed type. The B7 and B13 symbionts appear to be highly specific to morphospecies in the genus Madracis. The host specificity between the B13 symbionts and its coral host may be the result of co-evolution of the coral-symbiont association and suggests that the brooding species, M. mirabilis, is reproductively isolated. Microhabitat differentiation associated with light utilization independent of depth is discussed.  相似文献

Plants of birdsfoot trefoil (Lotus corniculatus), alfalfa (Medicago sativa) and tobacco (Nicotiana tabacum) were transformed by Agrobacterium with binary vectors harbouring genes that code either for wild-type and %-zein:KDEL, or for #-zein:KDEL. The maize seed storage proteins %-zein and #-zein are rich in sulphur amino acids, and our long-term goal is to improve alfalfa and birdsfoot trefoil forage quality. Significant levels of zeins were detected in leaves of birdsfoot trefoil transformants, ranging up to 0.055% and 0.30% of total extractable protein for %-zein and #-zein:KDEL, respectively. In leaves of alfalfa, the %-zein:KDEL expression level was up to 0.026% of the total extractable protein. From a F1 population of transgenic tobacco, a plant was selected in which the amount of zeins (%-zein:KDEL plus #-zein:KDEL) accounted for 1.1% of the total extractable protein.  相似文献

Doubled haploid lines, which have high levels of genetic uniformity, are suitable for physiological or genetic studies. We established a transformation system using a doubled haploid line of cabbage. Hypocotyl explants that had been precultured for 3 days on Murashige and Skoog medium containing 50 µM acetosyringone were inoculated and cocultured with Agrobacterium tumefaciens strain LBA4404 (pIG121Hm) for 3 days. Kanamycin-tolerant shoots were regenerated onto shoot induction medium 3 months after Agrobacterium inoculation. The transformation efficiency was about 3% under optimal conditions. The segregation pattern of the self-pollinated transformant that carried one copy of the introduced transgene revealed that the #-glucuronidase gene was inherited in a 3:1 ratio.  相似文献

During the past decade, Chorioptes mites occupying the outer ear canals have been a common finding at routine necropsies of moose (Alces alces) in Sweden, but neither the taxonomy of the mites nor lesions from the infestation have been investigated. A group of 69 lactobacilli was isolated from caries lesions and root canals of early childhood caries (ECC) affected children treated in the Department of Pedodontics (Children’s Teaching Hospital, Brno, Czech Republic). Biochemical and physiological properties of all strains were characterized by API 50 CH kit and conventional tube tests. The rep-PCR fingerprinting with the (GTG)5 primer was used for genotypic grouping of the isolates. The (GTG)5-PCR fingerprinting grouped all analyzed strains into a few clusters in nearly full agreement with phenotype identification results and clarified the taxonomic position of 13 biochemically unidentified strains. In total, 20 strains of Lactobacillus fermentum, 17 L. rhamnosus, 14 L. casei/paracasei, 7 L. gasseri, 7 L. salivarius and 4 L. plantarum were identified. Mixtures of two or even three Lactobacillus spp. were isolated from a few root canal content samples. Results obtained by biotyping and (GTG)5-PCR were generally comparable except for L. gasseri strains that were not biochemically identified. The (GTG)5-PCR fingerprinting was shown to be quicker, easier to perform and more reliable than biotyping. Our results imply this molecular method as a good tool for screening and identification of Lactobacillus spp. inhabiting dental plaque.  相似文献

The aquiferous systems of two Indopacific Oceanapia species (Oceanapiidae) were studied by corrosion casts: O. amboinensis living in shallow lagoons and O. fistulosa living at the base of the reef slope. Both species show a massive, entirely buried body, emerging from the sediment only by long, completely close fistules. Particularly in O. fistulosa the corrosion casts revealed a complex, grape-like structure of the choanosome organised in anatomical and functional units composed by an incurrent web whose anastomosed meshes are crossed by a central excurrent canal. A system of thin canals connects the two systems giving rise to an area of choanocyte chambers. The corrosion casts revealed that in both species incurrent water penetrates into the sponge body by the fistules and that it is expelled through specialised structures buried in the sediment. This observation is in accordance with field experiments performed on O. fistulosa. In some specimens of this species, a solution of china ink injected into plastic bags enveloping the external fistules was observed, after waiting for a while, to flow through the buried structures.  相似文献

Summary. One of the most species-rich ant-plant mutualisms worldwide is the palaeotropical Crematogaster-Macaranga system. Although the biogeography and ecology of both partners have been extensively studied, little is known about the temporal structuring and the dynamics of the association. In this study we compared life-history traits of the specific Crematogaster (Decacrema) partner-ants and followed the development of ant colonies on eight different Macaranga host plant species, from colony founding on saplings to adult trees in a snapshot fashion. We found differences in the onset of alate production, queen number and mode of colony founding in the ant species and examined the consequences of these differences for the mutualism with the host plant. The lifespan of some host plants and their specific ant partners seemed to be well matched whereas on others we found an ontogenetic succession of specific partner ants. The partner ants of saplings or young plants often differed from specific partner ants found on larger trees of the same species. Not all specific Crematogaster species can re-colonize the crown region of adult trees, thus facilitating a change of ant species. Therefore lifespan of the ant colony as well as colony founding behaviour of the different partner ant species are important for these ontogenetic changes. The lifespan of a colony of two species can be prolonged via secondary polygyny. For the first time, also primary polygyny (pleometrosis) is reported from this myrmecophytic system.  相似文献

Lactic acid bacteria (LAB) are generally sensitive to hydrogen peroxide (H2O2), Lactobacillus sakei YSI8 is one of the very few LAB strains able to degrade H2O2 through the action of a heme-dependent catalase. Lactobacillus rhamnosus strains are very important probiotic starter cultures in meat product fermentation, but they are deficient in catalase. In this study, the effect of heterologous expression of L. sakei catalase gene katA in L. rhamnosus on its oxidative stress resistance was tested. The recombinant L. rhamnosus AS 1.2466 was able to decompose H2O2 and the catalase activity reached 2.85 μmol H2O2/min/108 c.f.u. Furthermore, the expression of the katA gene in L. rhamnosus conferred enhanced oxidative resistance on the host. The survival ratios after short-term H2O2 challenge were increased 600 and 104-fold at exponential and stationary phase, respectively. Further, viable cells were 100-fold higher in long-term aerated cultures. Simulation experiment demonstrated that both growth and catalase activity of recombinant L. rhamnosus displayed high stability under environmental conditions similar to those encountered during sausage fermentation.  相似文献

We observed a weed (Ambrosia artemissifolia)–beetle herbivore (Ophraella communa) system for three years in a spatially continuous field (≈200 ha). We analyzed our field data in the light of two contrasting theories: the resource-concentration hypothesis and reaction–diffusion theory. For the resource-concentration hypothesis, we calculated the correlation coefficients between weed and beetle abundances for every season in each year. Although we found weak support for resource concentration in some seasons, we could not find any clear relationships in other seasons. We discuss a dispersal-based mechanism to explain the differences observed among seasons in lieu of the resource-concentration hypothesis. For the reaction–diffusion theory, we estimated the nonparametric spatial covariance functions for the spatial autocorrelation of weeds and beetles. Although we could not find any strong spatial structure for the individual species, we found evidence of spatial interactions between weeds and beetles using time lagged cross-correlation functions. Weed abundance enhanced local beetle abundance. Through time, there was evidence of beetle spillover to adjacent locations at roughly the one beetle-generation time scale. Sites with large number of beetles did not seem to reduce subsequent weed abundance.  相似文献

Protium heptaphyllum is a Burseraceae species known by the production of aromatic resin with medicinal, economic, and ecological values. Information on the development, architecture, and lifetime of the secretory system are crucial to understand the resin production and contribute to a more sustainable tapping regime. We investigated the histology and ultrastructure of the secretory canals under a developmental point of view. Stem samples were analyzed under light and transmission electron microscopy by conventional and cytochemical methods. Secretory canals, originated from procambium and cambium, occurred immersed in the primary and secondary phloem. Mature canals have a secretory epithelium and a wide lumen where the exudate is accumulated. A sheath of parenchyma cells with meristematic features surrounds the epithelium. The canals originate by schizogenesis and develop by schyzolysigenesis. Canals active in secretion occurred since the shoot apex and near the cambium. In the dilation zone of the secondary phloem, secretory canals exhibit sclerified epithelial and sheath cells and are inactive in secretion. Secreting epithelial cells have subcellular apparatus consistent with oleoresin, polysaccharides, and enzymes secretion. Pectinase and cellulase were cytochemically detected in developing canals and are involved in cell wall changes associated to canal growth and release of exudate. In P. heptaphyllum, the secretory system has a complex structure resultant from longitudinal growth, lateral ramification, and fusion of the adjacent canals, in addition to intrusive growth of both epithelial and sheath cells. Although some anatomical results are already known, ultrastructural data represent the novelty of this work. Our findings can contribute to the establishment of more efficient and sustainable techniques for resin extraction in this species.  相似文献

We describe the diet of introduced European hedgehogs Erinaceus europaeus (Linnaeus, 1758) in a New Zealand dryland system and provide the first quantitative analysis of food selectivity for this species. We also describe and compare the diets of nine hedgehogs and measure dietary overlap between these individuals. The most commonly eaten foods were beetles, including rare native species (in 94% of droppings), earwigs (92%), spiders (25%) and native skinks (14%). Remains of at least three skinks were found in one dropping. Earwigs and darkling beetles (Tenebrionidae) were the most preferred food types, and Hymenoptera and cylindrical bark beetles (Colydiidae) were least preferred. Consumption of most foods mirrored their availability. Most individuals’ diets conformed to the pattern seen at the population scale, with 21–54% of dry faecal mass derived from beetles and 13–39% from earwigs. One animal frequently consumed large amounts of fruit. Dietary overlap between pairs of individuals was high (mean Horn’s index, 0.84). This may be the result of limited opportunities to diversify in a very moisture-limited and low diversity habitat.  相似文献

We have recently indicated the plant growth promoting activities of Pseudomonas sp. as well as their alleviating effects on some soil stressors such as salinity. This is because in recent years, biological fertilizers have received special attention by scientists in sustainable agriculture. Accordingly, it is pertinent to specify the beneficiary level of such soil bacteria on plant growth including phosphorous (P) uptake. Hence, the objectives were to determine: (1) the plant growth promoting effects of the tested Pseudomonas sp., and (2) its combined effects with different P fertilization rates on the nutrient uptake (N, P, and K) and yield of wheat (Triticum aestivum L.) under greenhouse and field conditions. The experiments were factorially arranged on the basis of a completely randomized block design with three replicates and were conducted at the Research Farm of Agriculture and Natural Resources Research Center of Khorasan, Mashhad, Iran. P was fertilized at three levels including 0, 25 and 50 kg/ha P2O5. Pseudomonas sp. including Pseudomonas fluorescens 153, P. fluorescens 169, P. putida 4, and P. putida 108 were tested. Activities such as production of ACC deaminase and IAA-like products, as well as P solubilization were among the most important activities of the tested Pseudomonas sp. Such bacterial effects greatly enhanced wheat growth and yield under greenhouse and field conditions. The results also showed that the effects of Pseudomonas sp. on wheat nutrient uptake and the effects of bacteria as well as P fertilization on wheat yield were significant. P. putida 108 was the most effective strain enhancing wheat P uptake and grain yield under greenhouse (96 and 58%) and field (80 and 37%) conditions, respectively. Hence, although Pseudomonas sp. could be a suitable replacement for high P fertilization, however, the optimum wheat yield resulted when the bioinoculants are combined with 50% (25 kg/ha P2O5) P fertilization. The present study was designed to expand genetic knowledge of myo -inositol (MI) metabolism in Lactobacillus casei. Twenty-four L. casei isolates of dairy origin were tested for the presence of iol cluster. PCR screening revealed eight strains encoded functions involved in MI utilization, of which one strain was able to use MI as carbon source. To gain a deeper understanding of the function of iol genes, four of the eight observed iol clusters were subjected to the full sequencing procedure. The results showed that the iol cluster was not a common feature among dairy L. casei strains. In addition, the four iol clusters were highly similar to one another in terms of sequence similarity and operon architecture. However, abundant polymorphisms that comprised a majority of synonymous mutations were detected throughout the full sequences. Three of them distributed among iolB, iolC, and iolT genes were found in linkage to MI-negative phenotype. Compared with other bacterial iol clusters, the iol cluster of L. casei showed a high similarity with that of Bacillus subtilis.  相似文献

In vitro regeneration of black nightshade (Solanum nigrum L.) plants was achieved through callus-mediated shoot organogenesis followed by 30 d indoor ex vitro adaptation to nutritional stress under environmental ambience and thereafter 6-d outdoor acclimatization in pots prior to field establishment. Relevant physiological parameters including pigment content, chlorophyll a fluorescence, net photosynthetic rate (P N), transpiration rate (E), and stomatal conductance (g s) of in vitro-regenerated plants were investigated during the course of ex vitro adaptation. During the first 4 d of indoor transplantation to potting substrate, there was a marginal reduction in the leaf chlorophyll and carotenoid contents but P N and E were strongly reduced. The stomatal conductance and E/P N ratio were significantly higher in plants up to 20 d of indoor adaptation than those of comparable age grown naturally from seeds. The shape of the OJIP fluorescence transient varied significantly with acclimatization, and the maximum change was observed at 2.0 ms. The 2.0 ms variable fluorescence (V j), 30 ms relative fluorescence (M 0), photon trapping probability (TR0/Abs), and photosystem II (PSII) trapping rate (TR0/RC) showed initial disturbance and subsequent stabilization during 30 d of indoor acclimatization. Energy dissipation (DI0/RC) and electron transport probability (ET0/TR0) showed an initial phase of increase during the 4 d after plants were transplanted outdoors. During the 6-d outdoor acclimatization after transfer of plants to soil, no significant change in total chlorophylls and carotenoids, E, and g s were observed, but P N improved after reduction on the first d. The OJIP-derived parameters experienced change on the first d but were stabilized quickly thereafter. There was no significant difference between outdoor acclimatized plants and those of the seed-grown plants of comparable age with respect to photosynthetic and fluorescence parameters. Direct transfer of plants without indoor acclimatization, however, showed a completely different trend with respect to P N, E, and OJIP fluorescence transients. The bearing of this study on optimizing micropropagation is discussed.  相似文献

Columba livia is an important reservoir and carrier of Cryptococcus neoformans, Cryptococcus uniguttulatus, Cryptococcus laurentii and Cryptococcus albidus. Upper digestive tract of this species is also known as a habitat for Cryptococcus neoformans. Given the increasing clinical interest of this microorganism, 331 swabs from crop and 174 dropping samples from pigeon lofts in Grand Canary Island have been studied. The obtained results show an extensive presence samples 81 positive (24.47%) of Cryptococcus spp. in analysed crops: 32 (9.66%) for C. neoformans, 24 (7.2%) for C. uniguttulatus, 23 (6.9%) for C. albidus and 2 (0.6%) for C. laurentii. In the same way, Cryptococcus spp was also isolated in 82 (47.13%), dropping samples: C. neoformans in 59 (33.9%), C. uniguttulatus, in 9 (5.17%), C. laurentii in 8 (4.59%) and C. albidus in 6 (3.44%) of the investigated samples, respectively. The cryptococcosis produced by species of cryptococci other than C. neoformans has become more important during the last decade, supporting the study on the role of pigeon in the epidemiology of this disease.  相似文献
