Summary The presence of soluble polymeric complexes in dilute solutions (10–30 M) of aluminium and phosphate at 25°C was demonstrated by paper electrophoresis and confirmed by chemical analyses and measurements of 32 P specific activities. Complexed aluminium and phosphate were not determined by colorimetric procedures without preliminary hydrolysis in 1 N HCl at 80–90°C. The formation of aluminophosphate complexes was maximal around pH 5 when, at an initial P/Al mole ratio of 1.6, 3 moles of P were complexed with 4 moles of Al. At pH 6.9, however, only 1 mole of P was removed for every 10 of Al, suggesting that hydroxyaluminium polymers predominated at higher ratios of OH-ions to H2PO4-ions. re]19750523  相似文献

The uptake of sulphate into roots of barley seedlings is highly sensitive to phenylglyoxal (PhG), an arginine-binding reagent. Uptake was inhibited by >80% by a 1-h pre-treatment of roots with 0.45 mol · m–3 PhG. Inhibition was maximal in pre-treatment solutions buffered between pH 4.5 and 6.5. Phosphate uptake, measured simultaneously by double-labelling uptake solutions with 32P and 35S, was less susceptible to inhibition by PhG, particularly at pH <6.5, and was completely insensitive to the less permeant reagent p-hydroxyphenylglyoxal (OH-PhG) administered at 1 mol · m–3 at pH at 5.0 or 8.2; sulphate uptake was inhibited in -S plants by 90% by OH-PhG-treatment. Root respiration in young root segments was unaffected by OH-PhG pre-treatment for 1 h and inhibited by only 17% after 90 min pre-treatment. The uptake of both ions was inhibited by the dithiol-specific reagent, phenylarsine oxide even after short exposures (0.5–5.0 min). Sulphate uptake was more severely inhibited than that of phosphate, but in both cases inhibition could be substantially reversed by 5 min washing of treated roots by 5 mol · m–3 dithioerythritol. After longer pre-treatment (50 min) with phenylarsine oxide, inhibition of the ion fluxes was not relieved by washing with dithioerythritol. Inhibition of sulphate influx by PhG was completely reversed by washing the roots for 24 h with culture solution lacking the inhibitor. The reversal was dependent on protein synthesis; less than 20% recovery was seen in the presence of 50 mmol · m–3 cycloheximide. Sulphate uptake declined rapidly when -S roots were treated with cycloheximide. In the same roots the phosphate influx was little affected, small significant inhibitions being seen only after 4 h of treatment. Respiration was depressed by only 20% in apical and by 31% in basal root segments by cycloheximide pre-treatment for 2 h. Similar rates of collapse of the sulphate uptake and insensitivity of phosphate uptake were seen when protein synthesis was inhibited by azetidine carboxylic acid, p-fluorophenylalanine and puromycin. Considering the effects of all of the protein-synthesis inhibitors together leads to the conclusion that the sulphate transporter itself, or some essential sub-component of the uptake system, turns over rapidly with a half-time of about 2.5 h. The turnover of the phosphate transporter is evidently much slower. Summary The growth and nitrogen uptake response of rape plants to nitrate concentration at the root surface were studied in solution culture in a controlled environment cabinet over a period of 24 days. NO3 was supplied at the rates of 10–5 M, 5×10–5 M, 10–4 M, 10–3 M and 10–2 M in solution and was maintained near these levels.With increasing mean N concentration in the tissues, the relative growth rate and leaf area ratio increased and unit leaf rate decreased slightly. Values of all three growth parameters decreased with plant age.The shoot: root dry weight ratios and their N content ratios increased with increasing mean per cent N in the plant. The length or surface area per unit dry weight of roots was correlated negatively with per cent N and positively with age.The maximum mean inflow of nitrate to rape roots decreased sharply with age. The concentration at which half maximal mean inflow was attained was 3.44×10–5 M NO3 .  相似文献

Summary The rate of potassium uptake by two varieties of burley tobacco (Nicotiana tabacum) differed by as much as two-fold when excised roots were absorbing K from solutions of 2.5 × 10–4 M KCl. With increasing substrate levels of KCl the difference between varieties decreased and no difference was observed at a KCl concentration of 1.6 × 10–2 M. The K-absorption by excised roots of several varieties from solutions of 5 × 10–4 M KCl was compared.Contribution of the Department of Agronomy, Kentucky Agr. Exp. Surface areas and energetic properties of the shooting stage roots of rye (Secale L.), triticale (Triticale), barley (Hordeum L.) and four wheat (Triticum L.) varieties were estimated from experimental water vapor adsorption data. Roots stressed during 10 days at pH 4 with aluminium concentrations ranging from 0 to 40 mg dm–3 were studied. Roots grown continuously at pH 7 were taken as controls. The surface properties of the roots grown at pH 4 without Al addition were apparently the same as those of the control roots. With the increase of the concentration of the aluminium treatment the surface area of the roots increased for all of the plants, beginning at 5 mg Al dm–3 for barley, at 10 mg Al dm–3for wheat and triticale, and at 40 mg Al dm–3 for rye. The average water vapor adsorption energy of the root surface decreased in general with the increase of Al stress concentration for all plants but triticale, for which this increased. The sensitive cereal varieties seem to have greater amount of high energy adsorption centers (more polar surface) than the resistant ones (lower surface polarity), however more data is needed to justify this hypothesis. For Al-sensitive roots, fraction of high energy adsorption sites decreased and fraction of low energy sites increased under the Al stress. Smaller changes in adsorption energy sites were noted for roots of Al-resistant plants.  相似文献

Plexiglass pot growth chamber experiments were conducted to evaluate the chemical alterations in the rhizosphere of mycorrhizal wheat roots after inoculation with Glomus intraradices [arbuscular mycorrhizal fungus (AMF)]. Exchange resins were used as sinks for nutrients to determine whether the inoculated plant can increase the solubility and the uptake of P and micronutrients. Treatments included: (1) soil (bulk soil); (2) AMF inoculation no P addition (I–P); (3) no inoculation with no P addition (NI–P); (4) AMF inoculation with addition of 50 mg P (kg soil)–1 (I+P), and (5) no inoculation with addition of 50 mg P (kg soil)–1 (NI+P). The AMF inoculum was added at a rate of four spores of G. intraradices (g soil)–1. The exchange resin membranes were inserted vertically 5 cm apart in the middle of Plexiglass pots. Spring wheat (Triticum aestivum cv. Len) was planted in each Plexiglass pot and grown for 2 weeks in a growth chamber where water was maintained at field capacity. Rhizosphere pH and redox potential (Eh), nutrient bioavailability indices and mycorrhizal colonization were determined. Mycorrhizal inoculation increased the colonization more when P was not added, but did not increase the shoot dry weight at either P level. The rhizosphere pH was lower in the inoculated plants compared to the noninoculated plants in the absence of added P, while the Eh did not change. The decrease in pH in the rhizosphere of inoculated plants could be responsible for the increased P and Zn uptake observed with inoculation. In contrast, Mn uptake was decreased by inoculation. The resin-adsorbed P was increased by inoculation, which, along with the bioavailability index data, may indicate that mycorrhizal roots were able to increase the solubility of soil P.  相似文献

Investigations on the effects of low levels of Al on P adsorption,uptake and translocation in seedlings of the indigenous grassAgrostis capillaris were undertaken. Apparent uptake and transportof H2 32PO4 from nutrient solutions containing 10 or 100mmolm–3 phosphate were characterized as functions of timeand concentration. Experiments on 32P uptake and transport insolutions containing no Al (control) or Al ranging from 3.7to 185 mmol m–3 at pH ranging from 4.3 to 4.6, showedthat in 10 mmol m–3 P, effects of Al at 3.7 and 37 mmolm–3 on the size of the initial uptake shoulder were small,but some increase in subsequent P uptake to the roots was observed,though transport to the shoots was suppressed. With 37 mmolm–3 Al in nutrient solution containing 100 mmol m–3P, the uptake shoulder was much increased above the control.Subsequent root uptake was stimulated but transport was unaffected.Lack of toxicity of the Al concentrations used was indicatedby a lack of significant effect on plant fresh weight. AbsorbedAl was almost totally retained in the root in all treatments.Speciation calculations showed that the major species in Alamended nutrient solution at pH 4.4 were H2PO4, AI3+and AIHPO4+, together with substantial amounts of AISO4+ andsoluble aluminium hydroxy complexes (AIOH2+, AI(OH)2+), dependingon the relative concentrations of P and Al. The effects of Al,with 10 mmol m–3 P, on adsorption of complexed P werepartly accounted for in terms of preferential cell wall adsorptionof Al complexes not containing P. The influence of base cation concentrations on pH and aluminium sensitivity of the woodland grasses Bromus benekenii and Hordelymus europaeus was studied in flowing solution culture experiments. Plants were exposed to low pH (3.9, experiment 1) and Al concentrations of 19 and 37 M (experiment 2) at two base cation (Ca+Mg+K) levels, all within the ranges measured in natural forest soil solutions. Elevated base cation concentrations ameliorated both H and Al toxicity, as indicated by increased root and shoot growth. In the third experiment, interactions between pH (4.3 and 4.0) and Al (0 and 19 M) were investigated. It was shown that the combined toxicity effects of H and Al were not greater than the separate H or Al effects. Tissue concentrations of base cations and Al increased with increasing concentrations in the solution, but were also influenced by the base cation : Al ratio. Relating the experimental evidence with the composition of forest soil solutions suggests an important role of soil pH and Al in controlling the distribution of the two species. Growth conditions also differ at various soil depths. Concentrations of free cationic Al were higher and base cation concentrations lower at 5–10 cm than at 0–5 cm soil depth. Increasing base cation concentrations may protect roots from both H and Al injury during periods of drought when concentrations of most elements increase in the soil solution, whereas molar ratios between base cations, H and Al remain unchanged.  相似文献

Norway spruce (Picea abies) was exposed to nutrient solutions containing a range of aluminium (Al) concentrations at several pH levels (3.2, 4 and 5). Root growth was reduced by 100 µM and 400 µM Al at pH 4 and 5, but at pH 3.2 only by 400 µM Al. The Al content of the roots increased with increasing pH. The Al content of the roots was higher at the root tips than at the older root parts at all pH values. Using X-ray microanalysis it could be shown that higher levels of Al at increased pH were mainly due to increased Al contents in root cortex cell walls. In seedlings, mycorrhizal with Pisolithus tinctorius or Lactarius rufus, the Al concentration of cortex cell walls was higher when nitrate (NO3) rather than ammonium (NH4) was the nitrogen (N) source.  相似文献

Summary Ryegrass plants were grown in pots of Horotiu sandy loam, either singly or as a dense sward, with a range of P fertiliser rates and regular harvests. Plants were non-mycorrhizal. P inflows into roots increased with P fertiliser rate. Sward plants absorbed up to 14.3% of the P fertiliser added, and single plants up to 7.6%. Sward plants absorbed most of the fertiliser P available to them within two weeks of germinating. After that, root growth ceased except at the highest P fertiliser rate used, and P inflow into roots decreased from 10–15 to 10–16 mole/cm/sec. Single plants, however, had continuous root growth and P uptake throughout the two month experiment, except for those at the two lowest fertiliser rates. Singly grown plants absorbed much more P than each plant in the dense swards. In single plants, root length/root weight ratio increased with increasing P fertiliser. Plant growth was dependant on continued P uptake by the roots, which only occurred if new roots were continally produced and new volumes of soil tapped for P. Miniature heat balance-sap flow gauges were used to measure water flows in small-diameter roots (3–4 mm) in the undisturbed soil of a mature beech–oak–spruce mixed stand. By relating sap flow to the surface area of all branch fine roots distal to the gauge, we were able to calculate real time water uptake rates per root surface area (Js) for individual fine root systems of 0.5–1.0 m in length. Study aims were (i) to quantify root water uptake of mature trees under field conditions with respect to average rates, and diurnal and seasonal changes of Js, and (ii) to investigate the relationship between uptake and soil moisture θ, atmospheric saturation deficit D, and radiation I. On most days, water uptake followed the diurnal course of D with a mid-day peak and low night flow. Neighbouring roots of the same species differed up to 10-fold in their daily totals of Js (<100–2000 g m−2 d−1) indicating a large spatial heterogeneity in uptake. Beech, oak and spruce roots revealed different seasonal patterns of water uptake although they were extracting water from the same soil volume. Multiple regression analyses on the influence of D, I and θ on root water uptake showed that D was the single most influential environmental factor in beech and oak (variable selection in 77% and 79% of the investigated roots), whereas D was less important in spruce roots (50% variable selection). A comparison of root water uptake with synchronous leaf transpiration (porometer data) indicated that average water fluxes per surface area in the beech and oak trees were about 2.5 and 5.5 times smaller on the uptake side (roots) than on the loss side (leaves) given that all branch roots <2 mm were equally participating in uptake. Beech fine roots showed maximal uptake rates on mid-summer days in the range of 48–205 g m−2 h−1 (i.e. 0.7–3.2 mmol m−2 s−1), oak of 12–160 g m−2 h−1 (0.2–2.5 mmol m−2 s−1). Maximal transpiration rates ranged from 3 to 5 and from 5 to 6 mmol m−2 s−1 for sun canopy leaves of beech and oak, respectively. We conclude that instantaneous rates of root water uptake in beech, oak and spruce trees are above all controlled by atmospheric factors. The effects of different root conductivities, soil moisture, and soil hydraulic properties become increasingly important if time spans longer than a week are considered.  相似文献

The uptake of orthophosphate (32P) by excised corn roots, Zea mays L. was studied using roots grown on 0.2 mm CaSO4. Nine concentrations of KH2PO4 from 1 to 256 μm were used at temperatures of 20°, 30°, and 40°. Enzyme kinetic analysis was applied to the data obtained. Two apparent mechanisms (sites) of phosphate uptake were observed, 1 dominating at high P concentrations and 1 at low P concentrations. A Km of 1.36 × 10−4 and a Vmax of 177 × 10−9 moles per gram of roots per hour at 30° was calculated for the mechanism dominating at high P concentrations. Similar calculations gave a Km of 6.09 × 10−6 and a Vmax of 162 × 10−9 moles per gram of roots per hour at 30° for the mechanism dominating at low P concentrations. The Q10 for both mechanisms was approximately 2. Calculation of thermodynamic values from the data gave ΔF of − 5200 cal, ΔH of − 950 to − 1400 cal, and a enthalpy of activation (A) of 10,300 to 13,800 cal per mole for the mechanism dominating at high P concentrations. Similar calculations from the data for the mechanism dominating at low P concentrations gave a ΔF of − 7300 cal, ΔH of − 10,700 to − 8200 cal, and a A of 9300 to 18,900 cal per mole. If the dual mechanism interpretation of this kind of data adequately describes this system, then both mechanisms of P absorption by corn roots involve chemical reactions.  相似文献

The ecophysiological characteristics of fine roots of mature forest plants are poorly understood because of difficulties of measurement. We explored a root in-growth approach to measure respiration and nitrate uptake of woody plant roots in situ. Roots of seven species were grown into sand-filled chambers. Root-associated respiration was measured as CO 2 emission on four dates and nitrate uptake was quantified using 15N. All the roots were younger than 3 months at the time of measurement. Fine root respiration measured over the temperature range of 14.5–15.5 °C averaged 18.9–36.5 nmol gDM –1 s –1 across species. Nitrate uptake rates by these fine roots (1.3–6.8 nmol gDM –1 s –1) were comparable to other studies of forest trees. The root respiration rates were several times higher than measurements on detached roots of mature trees, concurring with literature observations that young roots respire much more rapidly than older roots. The root in-growth approach appears promising for providing information on the metabolic activity of fine roots of mature forest trees growing in soil.  相似文献

Total, organic and extractable P were measured in the humus and underlying soil to 10 cm depth beneath Sitka spruce (SS) and mixed Sitka spruce and Scots pine (SS+SP) stands planted on upland heath. The humus beneath SS+SP contained significantly (p<0.01) greater amounts of total and organic-P than that in SS and the mixed stands had more effectively retained approximately 87 per cent of previously applied fertilizer-P, totalling 100 kg P ha–1, compared with 70 per cent in SS. Despite the larger amounts of total-P in the mixed plots 0.01 M CaCl2 extractable molybdate reactive phosphorus (MRP) was significantly (p<0.05) greater in SS+SP humus only during March and April. Greater concentrations of MRP were released from the humus and soil during July and August at a mean rate of 58 g P ha–1 day–1. This coincided with drying of the soil during the summer and the rate of release, attributed to death of fine roots and microorganisms, was 4 to 30 times greater than reported values for rates of net mineralization of P from forest soils.  相似文献

Two Sphagnum moss species occupying hummock areas (Sphagnum capillifolium) and wetter hollows (Sphagnum recurvum) on a raised bog in north east Scotland were treated every two weeks with NH4NO3 solutions to supply 3g N m–2 yr–1. Although S. recurvum moss contained a greater concentration of total P than S. capillifolium the amounts and N:P ratios were similar in both species. Larger amounts of total dissolved P (TDP) and molybdate reactive P (MRP) were extracted from beneath S. recurvum to 25 cm below the moss. Additions of N both increased and decreased the amounts of TDP at different times, and decreased MRP. The MRP fraction accounted for about 20 per cent of TDP and the difference was assumed to be in organic forms (DOP). Nitrogen addition had no effect on the amounts of DOP, but C:P ratios of this fraction changed with species, depth and N addition. Microbial P accounted for as much as 70 per cent of total P and showed seasonal variations, but no differences between the two moss species and N addition.  相似文献

Summary Two acid soils showing different Al solubility as a function of pH were limed to a range of pH values (in 10–2 M CaCl2) between 4.1 and 5.6. The apparent critical pH for the growth of barley in pots was 0.25 lower in the soil showing lower Al solubility. The addition of phosphate reduced exchangeable and soluble Al in the soils, and lowered the apparent critical pH by 0.35 while maintaining the difference between the soils. The Al concentration at the critical pH, measured after cropping to take account of the treatment effects on soil Al, also varied with soil and with phosphate addition. These apparent critical values of both pH and soluble Al varied linearly with available phosphate, over the range 18 to 73 mg P/kg soil, as follows: pH from 4.9 to 4.3; soluble Al, from 0.010 mM to 0.056 mM; and the soluble Ca/Al mole ratio, from 1270 to 214.  相似文献

The effects of different concentrations of lead nitrate (10–5 to 10–3 M) on root, hypocotyl, and shoot growth of Indian mustard (Brassica juncea L. var. megarrhiza), and the uptake and accumulation of Pb2+ by its roots, hypocotyls, and shoots were investigated. Lead had no significant inhibitory effect on the root growth at concentrations of 10–5 to 10–4 M during the entire treatment, while at 10–3 M, Pb slightly inhibited the root and shoot growth. B. juncea has ability to take up Pb from solutions and accumulate it in its roots, and transport and concentrate it. The Pb contents in the parts of plants treated with 10–3 M Pb were greater than those of untreated plants, by factors of 230 in the roots, 170 in the hypocotyls, and 3 in the shoots.  相似文献

Summary The properties of a system consisting of a thin phospholipid film separating two electrolyte solutions containing 1mm 2,4-dinitrophenol have been studied. Both the variation of electrical conductance as a function of pH, keeping the pH the same on both sides of the membrane, and the nonlinear variation of electrical potential difference as a function of pH difference across the membrane have been explained in terms of lipid-soluble complexes of the typeXP 2 whereX is a cation andP dinitrophenate. Summary A study was made of the effects of the herbicides 2,4-D (2,4-dichlorophenoxyacetic acid) and MCPA (4-chloro-2-methyl-phenoxyacetic acid) on ion uptake, leakage and growth of rice seedlings. Using isotopically-labelled solutions containing different concentrations of 2,4-D or MCPA, it was established that 10–4 M 2,4-D or MCPA effectively inhibited potassium ion uptake, while K-ion leakage from the roots occurred only at 10–3 M. The growth of the rice seedlings was markedly retarded even at low (10–6 M) concentrations, and the roots and shoots showed different tolerances to the herbicide. At 10–8 M herbicide, the effects were not injurious, but rather favourable. Reduction in root length by herbicides was not in accordance with dry-matter production.  相似文献

10–7 M cycloheximide inhibited bean (Phaseolus vulgaris)root elongation by about 20 per cent but it inhibited absorptionof rubidium, sodium, and phosphate ions to a much greater extent(34–71 per cent). Tips of intact plant roots grown inthe inhibitor showed more inhibition in ion uptake than adjacentproximal portions of the same roots and this is taken to indicatethat 10–7 M cycloheximide does not exert its effect onion uptake by any uncoupling action. Sodium uptake from 0.5 or 10 mM NaCl solutions by root tipswas inhibited by 10–7 M cycloheximide to twice the extentthat it was in the elongating region of the root. Assuming thatthe inhibitor affects the plasmalemma more than the tonoplast,Epstein's model of parallel operation of system 1 and system2 at the plasmalemma is supported.  相似文献
