Dineen J. K. and Windon R. G. 1980. The effect of acquired resistance on adult worms of Trichostrongylus colubriformis in Lambs. International Journal for Parasitology10: 249–252. Wether lambs were classified as either responders or non-responders to vaccination and challenge with Trichostrongylus colubriformis. Worms recovered from these animals and unvaccinated controls were measured, eggs in utero counted and sex ratios (male/female) were determined for individual burdens. The results showed that all three parameters were reduced in responder lambs compared with non-responder and unvaccinated lambs and were positively correlated with worm counts in responder lambs whereas they tended to be negatively correlated with worm counts in non-responders and unvaccinated animals.These various effects of resistance on parasite development in responder lambs may be due to the action of endogenous inflammatory agents generated immunologically. On the other hand negative correlation between developmental parameters and worm burdens in non-responders and unvaccinated lambs suggests that development of parasites in heavy infections may be restricted by competition for a limiting physiological, spatial or nutritional resource of the host.  相似文献

Groups of lambs were infected daily with either 250, 500 or 1000 Ostertagia circumcincta larvae and the course of their worm burdens by post mortem examination at five intervals of time was studied. The number of worms appeared to be related to the rate of intake of larvae. A morphometric study of female worm lengths and observations on the incidence of reduced vulval flaps indicated that the population of worms turned over rapidly. Mapes C.J. and Coop R.L. 1973. The fate of multiple doses of infective larvae of Nematodirus battus in 8-month-old lambs and their effect on intestinal enzyme activity. International Journal for Parasitology3: 363–370. The fate of five daily doses of 60,000 infective larvae of Nematodirus battus was studied in 8-month-old lambs. On 18, 26 and 34 days after the last larval dose 4.0, 3.3 and 1.0 per cent of the total infective dose was recovered. Approximately 50 per cent of the worms recovered on these days were fourth-stage larvae. It is suggested that L5 and adult stages were preferentially lost from the hosts and that male worms were developing at a faster rate than the females. The populations of N. battus were smaller, contained higher proportions of fourth-stage larvae and shorter L5 and adult worms than those developing from similar infective doses in 3-month-old lambs. A transient decrease in alkaline phosphatase and maltase levels was found in the mucosa of the small intestine and was compared with the marked and persistent changes in mucosal enzyme activities found with similar infective doses in 3-month-old hosts.  相似文献

Barker I. K. and Titchen D. A. 1982. Gastric dysfunction in sheep infected with Trichostrongylus colubriformis, a nematode inhabiting the small intestine. International Journal for Parasitology12: 345–356. Six of 12 lambs infected with Trichostrongylus colubriformis had reduced abomasal acidification (pH 4.0–8.1) in comparison with uninfected pair-fed and replete controls (pH <3.5), though less than 0.8% of the worm burden was in the abomasum. Loss of prominence of parietal cells and encroachment of mucous cells deep into fundic glands was seen by light microscopy. Under the electron microscope, parietal cells had little canalicular or tubulovesicular development, had large vacuoles, many polyribosomes and few mitochondria in comparison with those in controls. In a further 8 sheep prepared with abomasal fistulae and separated fundic pouches and inoculated orally with T. colubriformis, the volume of fundic pouch secretion declined as feed intake dropped and in 7 out of 8 animals H+ concentration in fundic pouch secretion also fell. Infection generally reduced volume and acidity of pouch secretion more than did a pre-inoculation fast. In 5 sheep, abomasal content exceeded pH 4. Inoculation of T. colubriformis by enterotomy and Ostertagia circumcincta per os, in a lamb with a separated fundic pouch, caused depression of volume and acidity of pouch secretion characteristic of T. colubriformis infection, rather than the hypersecretion typical of abomasal infection with Ostertagia. Factors inhibitory to parietal cell differentiation and gastric acid secretion may be released from the small intestine of some sheep in response to changes in the gut induced by the presence of T. colubriformis. Abomasal dysfunction is a manifestation of severe intestinal trichostrongylosis.  相似文献

Dineen, J. K. and Windon, R. G. (1980). The effect of sire selection on the response of lambs to vaccination with irradiated Trichostrongylus colubriformis larvae. International Journal for Parasitology10: 189–196. Rams selected for responsiveness and unresponsiveness to vaccination with irradiated T. colubriformis larvae at an early age were mated to unselected random bred ewes. Progeny were vaccinated with 20,000 irradiated larvae at 8 and 12 weeks of age, given anthelmintic treatment at 16 weeks and challenged with 20,000 normai larvae at 17 weeks. The results, based on wether worm counts and ewe faecal egg counts, showed significant differences between responder and non-responder progeny. There was a significant correlation between worm counts and faecal egg counts of half-sibs from the same sire group. The occurrence of globule leucocytes was inversely related to worm burdens of wether progeny, however, no clear relationship was found with eosinophils. In vitro lymphocyte stimulation using T. colubriformis L3 antigen, concanavalin A and lipopolysaccharide showed that statistically defined responder progeny, pooled from both responder and non-responder sire groups, gave higher responses than non-responder lambs after vaccination. The results confirm that genetically-determined factors are involved in the response of lambs to vaccination at an early age, and indicate that rapid genetic progress may be achieved in the type of mating usually carried out under fleld conditions.  相似文献

The response of lambs to vaccination and challenge with Trichostrongylus colubriformis: effect of plane of nutrition on, and the inter-relationship between, immunological responsiveness and resistance. International Journal for Parasitology14: 39–44. Merino lambs weaned at 8 weeks of age were fed either ground and pelleted (high plane, HP) or chopped (low plane, LP) lucerne hay ad libitum to achieve an approximate 2-fold difference in liveweight gain. When aged 17 and 21 weeks, 15 of the 20 lambs in each diet group were vaccinated with 80,000 irradiated T. colubriformis larvae. At 25 weeks, vaccinated and unvaccinated lambs were treated with anthelmintic and one week later challenged with 30,000 normal larvae. Four weeks after challenge the animals were killed for worm counts. After vaccination HP lambs had higher titres of antibodies to the parasite and after challenge had lower worm egg outputs, and lower worm burdens than LP lambs. Immunological responsiveness (serum titre of complement-fixing antibodies against worm antigen) and manifestations of resistance (eggs produced per female worm per day and percent protection calculated from worm counts) were significantly correlated within dietary groups. Percent protection and egg production per female worm were highly correlated (r = ?0.81) in individual animals pooled over dietary groups, suggesting that both manifestations of resistance respond to essentially the same immunological mechanism. Failure to obtain significant correlation between weight gain pre-vaccination and immunological and resistance parameters indicated that animal production and resistance to infection are not genetically linked. Negative correlation of weight gain during the vaccination period with serum antibody titre at challenge suggests that the developing immune response competes with weight gain for limited physiological resources of the animal.  相似文献

The response of lambs to vaccination at weaning with irradiated Trichostrongylus colubriformis larvae: segregation into ‘responders’ and ‘non-responders’. International Journal for Parasitology8: 59–63. Groups of colostrum fed and colostrum deprived lambs were vaccinated with irradiated Trichostrongylus colubriformis larvae at weaning (3 months) and challenged with normal infective larvae. As there was no significant difference between the worm counts of these groups after challenge, it was concluded that the failure of lambs, generally, to respond to vaccination as vigorously as mature sheep, was not due to ‘feed-back inhibition’ of the immune response by the action of maternal antibody.However the results showed that the lambs segregated into ‘responders’ and ‘non-responders’. The mean worm count of responders was 1560 whereas for non-responders this was 24,340. A responder was defined as an animal whose worm count was less than the lower limit of the 99 % confidence interval of unvaccinated controls. These results suggest that genetically-determined factors play an important role in the responsiveness of lambs to vaccination.Whereas counts of globule leucocytes in duodenal tissues were negatively correlated with worm counts (i.e. positively correlated with resistance), counts of both eosinophils and neutrophils were positively correlated with worm counts. These findings suggest that either globule leucocytes are involved in the resistance mechanism, or they are by-products of cellular events involved in resistance. On the other hand it is unlikely that either eosinophils or neutrophils are involved in the mechanism of resistance.  相似文献

Sheep infected with the nematode parasite Trichostrongylus colubriformis showed anti-T. colubriformis acetylcholinesterase. (AChE) antibodies in the IgG1 but not the IgG2 or IgM fractions prepared from their serum. Using the fluorescent antibody technique with representative sera, antibodies in the IgG1 fraction exhibited specificity for antigens in the subventral glands of the worm excretory system. IgA antibody specificity for antigens in the excretory glands and intestine of the worm was also demonstrated.  相似文献

Immunity was successfully transferred by ‘Transfer Factor’ prepared from leucocytes of two adult Scottish Blackface donor rams infected with O. circumcincta and T. colubriformis to 4-month-old susceptible Fin X Dorset lambs. The immunity was expressed by a significantly reduced faecal egg count and worm burden compared to challenged, untreated controls. The immunity was comparable to that produced in another group of lambs given an initial infection prior to challenge with both parasites.  相似文献

Waller P. J. and Thomas R. J. 1978. Host-induced effects on the morphology of Ostertagia circumcincta in grazing sheep. International Journal for Parasitology8: 365–370. Morphological changes are described in adult Ostertagia circumcincta recovered from lambs slaughtered monthly from June to November. Comparisons were made between continuously grazed principal lambs exposed to infection for increasingly long periods, and tracer Iambs exposed for only four weeks. Lambs were either slaughtered immediately off pasture or held 2 weeks under worm free conditions before slaughter. Although no resistance to establishment of infection occurred, shown by comparable worm burdens of principals and tracers, a rapid parasite population turnover was observed in the principal lambs which ceased once the lambs were removed from the infected pasture. Significantly fewer adult female Ostertagia with fully developed vulval flaps were recorded in principals compared to tracers. Such differences are considered to be primarily host induced responses which rapidly come into play, evidenced by the entire populations of developing female worms in tracers slaughtered immediately off pasture showing arrested vulval flap development after they had completed development in those tracers held for a further two weeks. A progressive reduction in the proportion of adult male worms also occurred in the principal lambs. Population density was shown to have an inverse correlation with worm size. By analogy, it is suggested that seasonal change in the presence of cuticular inflations in Haemonchus contortus females with linguiform vulval flaps may be primarily host induced, rather than a change in the relative fitness of discrete genotypes.  相似文献

Jones W. O. and Symons L. E. A. 1982. Protein synthesis in the whole body, liver, skeletal muscle and kidney cortex of lambs infected by the nematode Trichostrongylus colubriformis. International Journal for Parasitology12: 295–301. Tyrosine flux and the synthesis of protein in the whole body, liver, skeletal muscle and kidney cortex and of albumin in lambs infected with Trichostrongylus colubriformis and uninfected lambs fed ad libitum or pair-fed with the infected group, were measured by constant infusion of 14C-l-tyrosine. Live weight gain was lower in the infected than in pairfed lambs, but rates of whole body protein synthesis were similar in both groups. On the other hand, compared with control lambs, there was a faster rate of protein synthesis per unit of protein consumed in infected but not in pair-fed lambs. Rates of protein synthesis per unit of body weight in infected were higher than in pair-fed lambs, but similar to the rate in control lambs. The fractional synthetic rates (FSR) of albumin and liver proteins and the amount of liver protein synthesized per day were increased by infection. The FSR and amount of protein synthesized per day were depressed in skeletal muscle and kidney cortex. Anorexia did not explain any of these changes. Infection caused a loss of protein from each of these tissues, but this loss was due to anorexia in only the liver. There was generally good correlation between concentration of RNA per g fresh weight or per mg nitrogen and the FSR of protein. However, although the RNADNA ratio correlated well with synthesis in skeletal muscle, it was poorly correlated for liver proteins. The relationship between the rate of growth and protein synthesis in infected lambs is discussed.  相似文献

Unusually high levels of acetylcholinesterase (AChE) were found in the nematode parasites Trichostrongylus axei, T. colubriformis and T, retortaeformis. In T. colubriformis the enzyme was located in the oesophageal and excretory glands of the parasitic stages. The highest level/unit wt was found in the fourth-stage larvae, which per worm had a comparable level to that in adult worms because the excretory gland was fully developed in the fourth-stage larvae. In acrylamide gel electrophoresis, T. axei and T. colubriformis AChE and esterases were similar but differed from those present in T. retortaeformis. Globulins prepared from the sera of sheep and guinea-pigs infected with T. colubriformis complexed with T. colubriformis and T. axei AChE, but not with esterases nor with AChE from T. retortaeformis, Nippostrongylus brasiliensis, Oesophagostomum radiatum or O. venulosum. Complexing of AChE to globulins did not inhibit the enzymic function of this enzyme.  相似文献

Wedrychowicz H., Maclean J. M. and Holmes P. H., 1984. The influence of Trypanosoma brucei infection on local immunoglobulin responses of rats to Nippostrongylus brasiliensis. International Journalfor Parasitology14: 453–458. Serum, intestinal and lung immunoglobulin and antibody isotype responses to Nippostrongylus brasiliensis infection were studied in normal and trypanosome-infected Hooded Lister rats. Rats which received trypanosomes 7 days before N. brasiliensis infection had impaired responses of serum IgG and IgA. Bronchial and intestinal mucosal IgG was not reduced whilst IgA concentration in these sites was markedly diminished. Total immunoglobulin M levels in T. brucei parasitised rats were higher in both sera and mucosal sites. However, tests with radiolabelled adult nematode excretory-secretory antigens indicated that specific lung and intestinal IgM responses were reduced. Immunoglobulin A antibody responses were diminished most markedly in sera and lungs and also in the intestine while IgG antibodies were decreased in sera and intestine mucosae T. brucei infected rats had higher worm burdens than rats infected with N. brasiliensis alone but worm expulsion was not delayed. The results indicate that local as well as systemic antibody responses are reduced in trypanosome infected animals.  相似文献

The fate of 300,000 and 5 × 60,000 infective larvae of Nematodirus battus during the 2–3 weeks after patency was studied in 3-month-old lambs. The proportions of the infective doses recovered after 18, 26 and 34 days were 37, 6 and 3.5 per cent in the single dose and 29,12 and 4.7 in the multiple dose groups. Fifth-stage larvae and adults, especially females, were preferentially lost from the hosts. The numbers of worms present and the percentage of fourth-stage larvae present were similar in both the single and multiple dose groups.  相似文献

The distributions of Haemonchus contortus, Ostertagia circumcincta and Trichostrongylus axei during their development in lambs were studied by freezing and sectioning the abomasum and then recovering the worms from the contents and mucosa of each section. In previously worm-free lambs, the distribution of fourth stage H. contortus was very regular, the frequency of larvae being highest in the anterior fundic area and then decreasing in each section towards the pylorus. The distributions of the early larval stages of O. circumcincta and T. axei were variable but tended to be biomodal, with one peak of larvae in the fundic area and a second in the pyloric area. It is suggested that the difference in distribution between H. contortus and the other two species may be due to a difference in the rate of response of exsheathed third stage larvae to stimuli in the abomasum. In the later stages of development, H. contortus and O. circumcincta tended to migrate towards the pyloric area. In previously infected lambs, the distribution of inhibited early-fourth stage H. contortus was similar to that of normally developing fourth stage larvae in previously worm-free lambs and appeared to be unaffected by the host response which caused an altered distribution and rejection of late-fourth stage and adult worms. The rate of rejection of adult O. circumcincta differed between the fundic and pyloric areas of the abomasum, which suggests that the host response in these two areas may be different.  相似文献

Windon R. G. and Dineen J. K. (1981). The effect of selection of both sire and dam on the response of F1 generation lambs to vaccination with irradiated Trichostrongylus colubriformis larvae. International Journal for Parasitology11: 11–18. Rams and ewes, tested for responsiveness to vaccination with irradiated T. colubriformis larvae at an early age, were mated on the basis of responder × responder and non-responder × non-responder. Progeny were vaccinated at 8 and 12 weeks of age with 20,000 irradiated larvae, treated with anthelmintic at 16 weeks and challenged with 20,000 normal larvae at 17 weeks. Faecal egg counts of progeny from responder matings were significantly lower than progeny from non-responders, and within each mating type, ewe lambs had markedly lower egg counts than ram lambs. The level of circulating complement-fixing antibodies to T. colubriformis larval extract were inversely related to egg counts. Thus, ewe progeny from responder matings had the highest serum antibody levels, non-responder ram progeny had the lowest levels and responder rams and non-responder ewes had similar intermediate levels. In vitro responses of cells stimulated with T. colubriformis L3 antigen were greater in progeny from responder matings, whereas responses to bacterial lipopolysaccharide were higher in progeny from non-responder matings. The results confirm that the response to vaccination at an early age is genetically determined, and show that the response of progeny is most vigorously expressed when both sires and dams have been selected.  相似文献

Commencing in December 1970, paddocks of a uniform series of sheep pastures were grazed for 6, 12 or 24 weeks by either yearling steers is or yearling ewes. Cattle pastures were treated similarly. All ewes and steers were pre-dosed with anthelmintic.At the conclusion of alternate grazing the effectiveness of the grazing treatments was evaluated by grazing each paddock for 1 month with either ten worm-free lambs (sheep pastures) or three wormfree calves (cattle pastures). The test animals were then slaughtered for total worm counts. The grazing of sheep pastures with cattle for 6, 12 or 24 weeks from December onwards resulted in reductions in numbers of Haemonchus contortus and Trichostrongylus colubriformis in test lambs. In comparison with continuous grazing by sheep, Nematodirus spp was only reduced after 24 weeks grazing by cattle. Cattle pastures grazed by sheep for 6 weeks showed no reduction in numbers of Ostertagia ostertagi or Cooperia oncophora in test calves. However after 12 weeks with sheep, numbers of O. ostertagi though not C. oncophora were reduced and after 24 weeks of alternate grazing both these species were reduced.Calves following the 6 week sheep treatments acquired both Haemonchus contortus and Trichostrongylus colubriformis and the calves from the 12 week sheep treatment paddock also carried H. contortus. For sheep the only evidence of cross-transmission was the occurrence of small numbers of Cooperia oncophora in test lambs from the 24 week cattle grazing treatment.The results provide evidence that sequential stocking with cattle and sheep in conjunction with anthelmintic treatment is an effective management strategy for preparing parasitologically safer pastures, but further information is required to determine the optimum timing of sequential stocking in farming situations.  相似文献

Echinostomes are intestinal trematodes that infect a wide range of vertebrate hosts, including humans, in their adult stage and also parasitize numerous invertebrate and cold-blooded vertebrate hosts in their larval stages. The purpose of this study was to compare Echinostoma malayanum parasite growth, including worm recovery, body size of adult worms, eggs per worm, eggs per gram of feces, and pathological changes in the small intestine of experimental animals. In this study, 6-8-week-old male hamsters, rats, mice, and gerbils were infected with echinostome metacercariae and then sacrificed at day 60 post-infection. The small intestine and feces of each infected animal were collected and then processed for analysis. The results showed that worm recovery, eggs per worm, and eggs per gram of feces from all infected hamsters were higher compared with infected rats and mice. However, in infected gerbils, no parasites were observed in the small intestine, and there were no parasite eggs in the feces. The volume of eggs per gram of feces and eggs per worm were related to parasite size. The results of histopathological changes in the small intestine of infected groups showed abnormal villi and goblet cells, as evidenced by short villi and an increase in the number and size of goblet cells compared with the normal control group.  相似文献

Background

Stool examination by counting eggs per gram of feces (EPGs) is the best method to estimate worm burden of Clonorchis sinensis in infected humans. The present study investigated a correlation between EPGs and worm burden in human clonorchiasis.

Methods and Findings

A total of 60 residents, 50 egg-positive and 10 egg-negative, in Sancheong-gun, Korea, participated in this worm collection trial in 2006–2009. They were diagnosed by egg positivity in feces using the Kato-Katz method. After administration of praziquantel, they were purged with cathartics on the next day, and then discharged adult worms were collected from their feces. Their EPGs ranged from 0 to 65,544. Adult worms of C. sinensis were collected from 17 egg-positive cases, and the number of worms ranged from 1 to 114 in each individual. A positive correlation between EPGs and numbers of worms was demonstrated (r = 0.681, P<0.001). Worm recovery rates were 9.7% in cases of EPGs 1–1,000 and 73.7% in those of EPGs over 1,000. No worms were detected from egg-negative subjects. Maximum egg count per worm per day was roughly estimated 3,770 in a subject with EPGs 2,664 and 106 collected worms.

Conclusions

The numbers of the worms are significantly correlated with the egg counts in human clonorchiasis. It is estimated that at least 110 worms are infected in a human body with EPGs around 3,000, and egg productivity of a worm per day is around 4,000.  相似文献

Sangster N. C. and Prichard R. K. 1984. Uptake of thiabenzadole and its effects on glucose uptake and carbohydrate levels in thiabendazole-resistant and susceptible Trichostrongylus colubriformis. International Journal for Parasitology14:121–126. Some possible mechanisms of thiabenzadole (TBZ) resistance have been investigated in Trichostrongylus colubriformis. Uptake of TBZ and 3-0-methyl glucose and changes in carbohydrate levels in a TBZ-resistant and a TBZ-susceptible strain of T. colubriformis were measured after in vitro incubation, in the absence or presence of TBZ. The rate of TBZ uptake in adult worms was greater in the resistant strain but the difference was not statistically significant. Final concentrations of TBZ in resistant worms were significantly greater than in the susceptible strain, but there was no difference in TBZ uptake by eggs of either strain. A mechanism of resistance based on decreased drug uptake by resistant parasites is thus unlikely. Resistant worms absorbed 3-0-methyl glucose faster than susceptible parasites, however, this rate was unchanged in either strain when the worms were exposed to TBZ and indicates that the drug does not affect glucose uptake. Levels of glucose, trehalose and glycogen fell during the incubation with the decrease being more pronounced in the resistant strain. When the worms were exposed to TBZ the same changes in carbohydrate levels occurred except for resistant worms maintained anaerobically where levels were higher, showing that TBZ does not affect the rate of carbohydrate utilization.  相似文献
