Summary Plastid and mitochondrial DNAs from Hedysarum species of the western Mediterranean basin, H. spinosissimum ssp eu-spinosissimum, H. spinosissimum ssp capitatum, H. carnosum, H. coronarium and H. flexuosum, were compared by restriction endonuclease fragment analysis. ctDNA fragment patterns for ssp eu-spinosissimum and ssp capitatum were indistinguishable in different enzyme digests. An identical ctDNA variation was found in Hpa II digests with two Sardinian populations of ssp capitatum. Each of the two subspecies was characterized by specific mt DNA patterns with Pst I, Bam HI, Sma I and EcoRI. No variation was detected in populations of different geographical origins for a given subspecies. H. carnosum, H. coronarium and H. flexuosum generated specific ct and mt DNA patterns. Comparison of mitochondrial fragments indicated: — a strong homology between the two subspecies, — a closer homology among the three other diploids, each being closer to the other two than to H. spinosissimum subspecies — as was also the case for the plastid genomes.  相似文献

The semiterrestrial crab Neohelice (=Chasmagnathus) granulata (Dana 1851) is a predominant species in brackish salt marshes, mangroves and estuaries. Its larvae are exported towards coastal marine waters. In order to estimate the limits of salinity tolerance constraining larval retention in estuarine habitats, we exposed in laboratory experiments freshly hatched zoeae to six different salinities (5–32‰). At 5‰, the larvae survived for a maximum of 2 weeks, reaching only exceptionally the second zoeal stage, while 38% survived to the megalopa stage at 10‰. Shortest development and negligible mortality occurred at all higher salt concentrations. These observations show that the larvae of N. granulata can tolerate a retention in the mesohaline reaches of estuaries, with a lower limit of ca. 10–15‰. Maximum survival at 25‰ suggests that polyhaline conditions rather than an export to oceanic waters are optimal for successful larval development of this species. In another experiment, we tested the capability of the last zoeal stage (IV) for reimmigration from coastal marine into brackish waters. Stepwise reductions of salinity during this stage allowed for moulting to the megalopa at 4–10‰. Although survival was at these conditions reduced and development delayed, these results suggest that already the zoea-IV stage is able to initiate the reimmigration into estuaries. After further salinity reduction, megalopae survived in this experiment for up to >3 weeks in freshwater, without moulting to juvenile crabs. In a similar experiment starting from the megalopa stage, successful metamorphosis occurred at 4–10‰, and juvenile growth continued in freshwater. Although these juvenile crabs showed significantly enhanced mortality and smaller carapace width compared to a seawater control, our results show that the late larval and early juvenile stages of N. granulata are well adapted for successful recruitment in brackish and even limnetic habitats.  相似文献

Two sympatric subspecies of the xerohalophyte Atriplex canescens Pursh. (Nutt.) were compared for 84 d in outdoor salinity trials in their native coastal desert environment in Sonora, Mexico. Subspecies linearis grows naturally on sea water in the high intertidal zone of estuaries while subspecies canescens grows on dunes. In lysimeter pot experiments, ssp. linearis exhibited 50% growth reduction when the mean root zone salinity reached 1160 mol m−3 NaCl compared to just 760 mol m−3 for ssp. canescens. When irrigated with sea water in a flood plot, ssp. linearis had 50% higher growth rates than ssp. canescens. The specialization of ssp. linearis for a saline environment was associated with greater net transport of Na+ from root to shoot, greater Na+ accumulation in the leaves and a higher Na:K ratio in the leaves compared to ssp. canescens. On the other hand, the two subspecies achieved approximately the same degree of osmotic adjustment in the leaves, equal to two to three times the external salinity, and had similar water use efficiencies. Even at relatively low salinities, both subspecies accumulated larger quantities of Na+ for osmotic adjustment than K+. The results suggest that breeding for Na+ accumulation rather than exclusion might be the more effective strategy for improving salt tolerance of conventional crop plants.  相似文献

Summary Uptake and assimilation of nitrate by two subspecies ofHypochaeris radicata L. were investigated under laboratory conditions as well as in the field.H. radicata ssp.radicata grows on relatively nitrogen-richer soils thanH. radicata ssp.ericetorum. Attempts were made to relate nitrate uptake and nitrate assimilation in the two subspecies to their different distribution in the field.No differences between the two subspecies with respect to nitrate uptake and nitrate assimilation were observed under laboratory conditions. In plants from the field intact tissue nitrate reductase was higher in ssp.radicata than in ssp.ericetorum. The relationship between Na+ accumulation and salt tolerance was tested by comparing subspecies of the halophyte, Atriplex canescens (fourwing saltbush), that differed markedly in Na+ content and Na:K ratios. Above ground tissues of one low-sodium and two high-sodium subspecies were compared with respect to cation accumulation, osmotic adjustment and growth along a salinity gradient in greenhouse trials. Plants of each subspecies were grown for 80 d on 2.2, 180, 540 and 720 mol m?3 NaCl. At harvest, A. canescens ssp. canescens had significantly lower Na+ levels, higher K+ levels and lower Na:K ratios in leaf and stem tissues than A. canescens ssp. macropoda and linearis over the salinity range (P < 0.05 or 0.01). Na:K ratios in leaves of the latter two, high-sodium, subspecies were approximately 2 on the lowest salinity treatment and ranged from 5 to 10 on the more saline solutions. By contrast, Na:K ratios in leaves of the low-sodium subspecies canescens, were only 0.4 on the lowest salinity and ranged narrowly from 1.7 to 2.3 at higher salinities. However, despite different patterns of Na+ and K+ accumulation, all three subspecies exhibited equally high salt tolerance and had similar osmotic pressures in their leaves or stems over the salinity range. Contrary to expectations, high salt tolerance was not necessarily dependent on high levels of Na+ accumulation in this species.  相似文献

An endemic brackish water faunule consisting of three taxa of bivalves,Hudsonella sinensis (Chen) n. gen.,Hudsonella sp., andProtocardia hepingxiangensis Chen & Lin occurs in the Bajocian of the southern Qinghai province, China.Hudsonella sinensis is a very variable species with a hinge structure characteristic of the subfamily Pachydontinae of the family Corbulidae. According to both geochemical data (stable carbon isotopes and the strontium/barium ratio) and ecological data the faunule lived in a brackish water embayment of mesohaline salinity (5#x2013;18 per mil) burrowing in soft substrate.  相似文献

Coincya monensis is represented in the British flora by two, cytologically distinct subspecies. Coincya monensis ssp monensis is an endemic diploid with a coastal sand dune distribution that includes a number of isolated populations. Coincya monensis ssp cheiranthos is a tetraploid alien, well established in South Wales in early successional habitats. Both subspecies share similar life form traits, flowering times and pollinators. Cluster analysis and phylogenetic reconstruction based on sequences of the mitochondrial nad4 gene confirmed the distinction between alien and endemic taxa. Tetraploid populations carry more polymorphic RAPDs loci and their genetic diversity is partitioned more within than among populations. In contrast, C. monensis ssp monensis has a distinct population genetic structure. Analysis of the multilocus genetic data confirmed a structure of genetically isolated, endemic population clusters in Scotland, Arran, the Isle of Man and South Wales. Experimental hybridisation showed the two subspecies are interfertile. Multivariate analysis of RAPDs data resolved hybrids between alien and endemic clusters and hybrids contained a proportion of alien-specific polymorphic loci. Hybrids of alien maternal parentage contained the mitochondrial nad4 sequence characteristic of the alien subspecies. Since the alien subspecies can invade mobile sand dune communities from urban sites and compete for pollinators, there is a risk that alien and endemic populations will mix and introgress. Conservation of endemic genetic diversity in Britain will require protection for all C. monensis ssp monensis populations. Currently, the most disjunct endemic population in South Wales is most at risk from introgression.  相似文献

In the context of a main project that aims to recover modern data on diatom distribution applicable to paleosalinity reconstructions in coastal areas of Southern South America, the composition and distribution of dead diatom assemblages in the littoral zone of the Quequén Salado estuary (Argentina) were studied. Diatom zones were defined along the estuarine gradient by cluster analysis and related to the salinity range and sediment composition by Canonical Correspondence Analysis. Four diatom zones were identified. A mixture of marine, brackish and freshwater diatoms, probably allochthonous, characterized the inlet (zone I). Marine/brackish taxa, represented mainly by Paralia sulcata dominated zone II, characterized by polyhaline conditions and sandy sediments. Zone III was characterized by mesohaline conditions, muddy sediments and the dominance of the estuarine diatom Amphora helenensis. Brackish/freshwater and freshwater diatoms dominated the headwaters (zone IV), where salinity was always below 5‰. The comparison of Quequén Salado diatom assemblages with previous results from the Quequén Grande estuary showed a similar taxonomic composition between both estuaries. However, differences in the salinity ranges of the estuaries (related to differences in the degree of human impact and tidal range) lead to a displacement in their spatial distribution along the longitudinal estuarine axis. This paper contributes to the knowledge of the ecological requirements of South American estuarine diatoms and provides useful data for paleosalinity reconstructions in the region. In this study, we investigated the germination behavior, and the differences in desiccation sensitivity and water uptake (WU), of the acorns of the two subspecies of Quercus ithaburensis, inhabiting different ecological niches. Sensitivity of acorns to drying was assessed by subjecting them to several desiccation treatments. The results showed that acorn water loss due to desiccation follows an almost linear pattern for the subspecies macrolepis, while in the case of subspecies ithaburensis, it follows a negative exponential model. Accordingly, the germination behavior was found highly dependent on water loss for both subspecies. The low acorn water content (15%) achieved after 15 days of drying was crucial for the ssp. macrolepis, while it had no effect on seed viability in ssp ithaburensis, that needed a subtle further water loss to reduce its germinability (i.e. <14.5% mc). Our results suggest that intraspecific differences in seed responses to drying may be due to different adaptation to survive drought after dispersal and contribute to explain the ability of ssp. ithaburensis to grow in the hottest and driest Mediterranean bioclimates. Despite being recalcitrant, the ssp. ithaburensis can be considered as one of the most tolerant oaks to desiccation worldwide.  相似文献

The native area of gammarids from the so-called ‘Caspian complex’, Pontogammarus robustoides (G.O. Sars, 1894), Obesogammarus crassus (G.O. Sars, 1894), Dikerogammarus haemobaphes (Eichwald, 1841) and D. villosus (Sowinsky, 1894), is associated with brackish waters. Over the last several decades they have colonized the European inland waters and part of the brackish Baltic Sea. It is believed that anthropogenic increase in the salinity of inland waters facilitated their expansion. However, the influence of salinity on the dispersal of gammarid species outside their native area is not fully understood. We tested the hypothesis that salinity was a major factor in determining distribution, based on the abundance of Gammaridae in three coastal areas of low salinity (brackish Baltic), i.e. 0.3, 3.4 and 7.3 PSU, successfully inhabited by them. Additionally, for the first time, the effect of water salinity on the osmoregulatory capacity of O. crassus was examined under laboratory conditions, for the salinities given above. The experiments showed that similarly as in the case of other Caspian complex species, salinity values of about 7 PSU create better conditions for osmoregulation in O. crassus than lower salinities (i.e. 0.3 and 3.4 PSU). In the environmental part of the study, we observed that only D. villosus achieved a significantly higher abundance in the area of 7.3 PSU. Thus, we concluded that in the range of 0.3–7.3 PSU, salinity is not a key factor governing the distribution of Ponto-Caspian gammarids.  相似文献

The North Sea–Baltic Sea transition zone constitutes a boundary area for the kelp species Saccharina latissima due to a strong salinity gradient operating in the area. Furthermore, the existence of S. latissima there, along Danish waters, is fairly patchy as hard bottom is scarce. In this study, patterns of genetic diversity of S. latissima populations were evaluated along the salinity gradient area of Danish waters (here designated brackish) and were compared to reference sites (here designated marine) outside the gradient area, using microsatellite markers. The results showed that the S. latissima populations were structured into two clusters corresponding to brackish versus marine sites, and that gene flow was reduced both between clusters and between populations within clusters. In addition, results provided empirical evidence that marginal populations of S. latissima in the salinity gradient area exhibited a distinct genetic structure when compared to marine ones. Brackish populations were less diverse, more related, and showed increased differentiation over distance compared to marine populations. The isolation of the brackish S. latissima populations within the salinity gradient area of Danish waters in conjunction with their general low genetic diversity makes these populations vulnerable to ongoing environmental and climate change, predicted to result in declining salinity in the Baltic Sea area that may alter the future distribution and performance of S. latissima in the area.  相似文献

Three-spined sticklebacks (Gasterosteus aculeatus L.) living at the southern limit of the species distribution range could possess specific morphological and physiological traits that enable these fish to live at the threshold of their physiological capacities. Morphological analysis was carried out on samples of sticklebacks living in different saline habitats of the Camargue area (Rhone delta, northern Mediterranean coast) obtained from 1993 to 2017. Salinity acclimation capacities were also investigated using individuals from freshwater-low salinity drainage canals and from mesohaline–euryhaline lagoons. Fish were maintained in laboratory conditions at salinity values close to those of their respective habitats: low salinity (LS, 5‰) or seawater (SW, 30‰). Fish obtained from a mesohaline brackish water lagoon (BW, 15‰) were acclimated to SW or LS. Oxygen consumption rates and branchial Na+/K+-ATPase (NKA) activity (indicator of fish osmoregulatory capacity) were measured in these LS or SW control fish and in individuals subjected to abrupt SW or LS transfers. At all the studied locations, only the low-plated “leiurus” morphotype showed no spatial or temporal variations in their body morphology. Gill rakers were only longer and denser in fish sampled from the LS–freshwater (FW) drainage canals. All fish presented similar physiological capacities. Oxygen consumption rates were not influenced by salinity challenge except in SW fish transferred to LS immediately and 1 h after transfer. However, and as expected, gill NKA activity was salinity dependent. Sticklebacks of the Camargue area sampled from habitats with contrasted saline conditions are homogenously euryhaline, have low oxygen consumption rates and do not appear to experience significantly greater metabolic costs when challenged with salinity. However, an observed difference in gill raker length and density is most probably related to the nutritional condition of their habitat, indicating that individuals can rapidly acclimatize to different diets.  相似文献

Caridina gracilipes, an atyid shrimp of Taiwan, inhabits the fresh and brackish waters. Its life history can be classified into amphidromous and landlocked types. Recent human’s transportation has caused concerns of genetic contamination between these long evolved lineages and ecological instabilities. Molecular markers with high sensitivities are required for detecting the possible introgression due to human disturbance. In the study, we described the development of 15 polymorphic microsatellite loci in C. gracilipes for genetic and ecological studies. These new markers were tested in 20 individuals of three populations. The number of alleles ranged from 6 to 17. Expected (H E) and observed (H O) heterozygosities ranged from 0.653 to 0.954, averaged at 0.865, and from 0.000 to 0.4, averaged at 0.146, respectively. All loci are significantly deviated from Hardy–Weinberg expectations due to the heterozygote deficiency. Significant genetic differentiation among populations was detected based on F ST values, which were estimated from 0.82 to 0.88. Summary Intraspecific variation of a wild potato species, Solanum acaule Bitt., was analyzed by RFLPs of genomic DNA. One hundred and five accessions were selected throughout the distribution area, including all subspecies, i.e., ssp. albicans (hexaploid), ssp. punae (tetraploid), ssp. acaule (tetraploid) and ssp. aemulans (tetraploid). Twenty-seven low-copy DNA clones (probes) were Southern hybridized with EcoRI, EcoRV, HindIII, and XbaI digests of total DNA of all accessions. In total, 238 RFLPs were detected from 94 enzyme x probe combinations. Among them, 49 RFLPs were specific to ssp. albicans, suggesting that the additional third genome is distinct from its two other genomes. RFLPs between and within subspecies were analyzed by principal component analysis. DNA similarities between subspecies coincided with a former taxonomic treatment in the sense that ssp. albicans is the most distantly related to ssp. acaule and ssp. aemulans is distantly related. Subspecies acaule and ssp. punae were indistinguishable. In addition, RFLPs could be used to distinguish groups within subspecies. Subspecies aemulans, confined to Argentina, was divided into two populations, one from the province of La Rioja and the other from the province of Jujuy. In ssp. acaule, some accessions from the southernmost distribution area were clearly distinguishable, while the others varied continuously, showing a geographical cline from Peru to Argentina.Reference to a specific brand or firm name does not constitute endorsement by the US Department of Agriculture over others of similar nature not mentioned  相似文献

The silver‐studded blue, Plebejus subsolanus, is widely distributed in the Russian Altai mountains, northeastern China, the Korean Peninsula, and the Japanese archipelago. In Japan, the species is distributed across wide elevation ranges from the lowlands of Hokkaido to the subalpine zone of Honshu. Current subspecies classification in Japan is as follows: ssp. iburiensis, occurring in lowland grasslands in Hokkaido; ssp. yaginus in lower mountain grasslands in Honshu; and ssp. yarigadakeanus in higher mountain grasslands in Honshu. The habitat of this species has been markedly reduced due to recent habitat destruction and land‐use changes. Here, we undertook phylogeographic analyses of two subspecies, ssp. yaginus and yarigadakeanus in the central mountainous regions of Japan, based on two mitochondrial gene sequences, in order to collect information for establishing effective conservation strategies. From 57 samples from the four mountain ranges, we obtained a haplotype network comprised of 12 haplotypes. Because of the haplotype network topology, the geographic distribution of haplotypes and the correspondence of haplotype divergence to subspecies taxonomy, we provisionally divided the haplotypes into three haplogroups: YR1 and YR2, which comprised ssp. yarigadakeanus, and YG, which comprised ssp. yaginus. Mitochondrial DNA genetic differentiation generally agreed with morphological subspecies classification. The haplotype network suggested that ssp. yarigadakeanus populations had multiple origins, and the subspecies character of “bright blue of the male's wings” was assumed to have evolved independently in each subalpine meadow. We found that P. subsolanus was genetically differentiated depending upon the elevation at each mountain region, suggesting that each haplogroup should be a conservation unit.  相似文献

Heteropappus hispidus ssp. leptocladus is an edaphic endemic taxon that is confined to serpentine and limestone-derived soils and is allopatrically distributed in three regions of western Japan. In this study, we attempted to detect genomic signatures of seven H. hispidus ssp. leptocladus populations along with eight other subspecies populations using eight nuclear microsatellite loci. The Mantel test supported an isolation by distance model across all H. hispidus populations, thus implying the possibility of parallel evolution for each subspecies. Results from AMOVA recognized relatively larger differentiations in geographic distribution compared to intraspecific taxonomy. Relationships indicated by neighbor-joining phylogenetic tree analysis and population structure generally did not reflect an intraspecific taxonomy. Populations from limestone-derived soil harbored a homogeneous genetic structure with neighboring populations from serpentine-derived soils. These results suggest that the edaphic ecotype may have derived allopatrically while a lack of edaphic constraint existed between serpentine and limestone soils.  相似文献

Forest trees frequently form species complexes, complicating taxonomic classification and gene pool management. This is certainly the case in Eucalyptus, and well exemplified by the Eucalyptus globulus complex. This ecologically and economically significant complex comprises four taxa (sspp. bicostata, globulus, maidenii, pseudoglobulus) that are geographically and morphologically distinct, but linked by extensive “intergrade” populations. To resolve their genetic affinities, nine microsatellites were used to genotype 1200 trees from throughout the natural range of the complex in Australia, representing 33 morphological core and intergrade populations. There was significant spatial genetic structure (FST = 0.10), but variation was continuous. High genetic diversity in southern ssp. maidenii indicates that this region is the center of origin. Genetic diversity decreases and population differentiation increases with distance from this area, suggesting that drift is a major evolutionary process. Many of the intergrade populations, along with other populations morphologically classified as ssp. pseudoglobulus or ssp. globulus, belong to a “cryptic genetic entity” that is genetically and geographically intermediate between core ssp. bicostata, ssp. maidenii, and ssp. globulus. Geography, rather than morphology, therefore, is the best predictor of overall genetic affinities within the complex and should be used to classify germplasm into management units for conservation and breeding purposes.  相似文献
