The ability of mebendazole and fenbendazole to bind to tubulin in cytosolic fractions from 8-day Ascaris suum embryos was determined by inhibition studies with [3H]colchicine. Colchicine binding in the presence of 1·10?6 M mebendazole was completely inhibited during a 6 h incubation period at 37°C. Inhibition of colchicine binding to A. suum embryonic tubulin by mebendazole and fenbendazole appeared to be noncompetative. The inhibition constants of mebendazole and fenbendazole for A. suum embryonic tubulin were 1.9·10?8 M and 6.5·10?8 M, respectively. Mebendazole and fenbendazole appeared to be competitive inhibitors of colchicine binding to bovine brain tubulin. The inhibition constants of mebendazole and fenbendazole for bovine brain tubulin were 7.3·10?6 M and 1.7·10?5 M, respectively. These values are 250–400 times greater than the inhibition constants of fenbendazole and mebendazole for A. suum embryonic tubulin. Differential binding affinities between nematode tubulin and mammalian tubulin for benzimidazoles may explain the selective toxicity. The importance of tubulin as a receptor for anthelmintic benzimidazoles in animal parasitic nematodes is discussed.  相似文献

Adult rat heart muscle cells obtained by perfusion of the heart with collagenase have been used to characterize the insulin receptors by equilibrium binding and kinetic measurements. Binding of 125I-labelled insulin to heart cells exhibited a high degree of specificity; it was dependent on pH and temperature, binding at steady increased with decreasing temperatures. About 70% of the radioactivity bound at equilibrium at 25°C could be dissociated by addition of an excess of unlabelled insulin. 54 and 40% of 125I-labelled insulin was degraded by isolated heart cells after 2 h at 37°C and 4 h at 25°C, respectively. This degrading activity was effectively inhibited by high concentration of albumin.Equilibrium binding studies were conducted at 25°C using insulin concentrations ranging from 2.5 · 10?11 mol/l to 10?6 mol/l. Scatchard analysis of the binding data resulted in a curvilinear plot (concave upward), which was further analyzed using the average affinity profile. The empty site affinity constant was calculated to be 9.5 · 107 l/mol with a total receptor concentration of 3.4 · 106 sites per cell.The presence of site-site interactions of the negative cooperative type among the insulin receptors has been confirmed by kinetic experiments. The rate of dilution induced dissociation was enhanced in the presence of native insulin (5 · 10?9 mol/l), both, under conditions of low and high fractional saturation of receptors.  相似文献

In our previous work we have shown that the age‐dependent decrease in the magnetosensitivity of heart muscle hydration is accompanied by a dysfunction of the Na+/K+ pump. The reciprocal relation between the Na+/K+ pump and Na+/Ca2+ exchange in development was suggested as a possible pathway for the age‐dependent decrease in the magnetosensitivity of heart muscle hydration (water content). Because high and low affinity ouabain receptors in cell membranes are involved in Na+/Ca2+ exchange and Na+/K+ pump functions, respectively, the effect of a 0.2 T static magnetic field (SMF) on dose‐dependent, ouabain‐induced hydration and [3H]‐ouabain binding with heart muscle tissues in young, adult and older rats was studied. Three populations of receptors in membranes with high (10?11–10?9 M), middle (10?9–10?7 M) and low (10?7–10?4 M) affinity to [3H]‐ouabain were distinguished, which had specific dose‐dependent [3H]‐ouabain binding kinetics and effects on muscle hydration. The magnetosensitivity of [3H]‐ouabain binding kinetics with high affinity receptors was prominent in all the three age groups of animals, while with low affinity receptors it was more expressed only in the young group of animals. All three types of receptors that caused modulations of muscle hydration were age dependent and magnetosensitive. Based on the obtained data we came to the conclusion that heart muscle hydration in young animals is more magnetosensitive due to the intense expression of high affinity ouabain receptors, which declines with aging. Colchicine was found to be taken up by adipose tissue and therein to bind to a soluble macromolecule not sedimented by centrifugation for 2 h at 100 000 × g. A similar binding occurred when soluble extracts of adipose tissue were incubated with colchicine. The binding reaction is temperature dependent and shows a pH optimum between 6.8 and 7.0. Double reciprocal plots of colchicine concentration versus amounts of colchicine bound to protein in the steady state disclosed an apparent Km of 0.250 to 1.5 ωM. The colchicine binding activity of soluble tissue extracts decreased when the extracts were incubated at 37°C. Addition of guanosine triphosphate and Mg2+ retarded the loss of colchicine binding activity. The molecular weight of the colchicine complex was estimated to be 115 000 and its sedimentation coefficient 5.8 S. All of these characteristics are remarkably similar to those of the protein tubulin which has been isolated from other tissues. Since it is now well known that tubulin is a protein subunit of cytoplasmic microtubules, it is suggested that the previously reported metabolic effects of colchicine on adipose tissue result from the dissolution of microtubules by colchicine.  相似文献

The maintenance requirement(s) exhibited by Bacillus sphaericus1593M was clearly established from the data on the steady state continuous cultures wherein the growth yield was found to be increasing with increase in dilution rate (4). In the present study we have attempted to estimate the maintenance coefficient of B. sphaericus 1593M by achieving zero growth rate in a total cell retention culture (TCRC). Zero growth rate was achieved at a cell density of 5.57?g?l?1 which remained constant reflecting a steady state in the TCRC. Based on the substrate flux calculations, maintenance coefficient was found to be 0.087?h?1. Further, using this data of maintenance coefficient, steady state cell density and substrate profiles in continuous cultures of B. sphaericus 1593M were predicted by simulation, which showed a close relationship to that of the experimental data.  相似文献

Glucokinase, the organ specific key enzyme of glucose metabolism in liver, was studied in primary cultures of adult rat hepatocytes during the first two days after cell preparation. In the presence of dexamethasone low concentrations of insulin (10?9 mol/l) prevented the observed time dependent decrease of glucokinase activity while higher insulin concentrations (10?8 and 10?7 mol/l) led to a twofold increase of enzyme activity. The enhancement of glucokinase activity was completely blocked by either actinomycin D or cycloheximide. The degree of this insulin dependent induction was correlated with the concentration of added dexamethasone, which seemed to perform a permissive function. The induction of glucokinase activity could be prevented by addition of glucagon (2 × 10?7 mol/l).  相似文献

The microtubule-disruptive drugs colchicine and vinblastine alter ligand-induced redistribution of cell surface immunoglobulins and lectin receptors. These effects can be duplicated by treatment of cells with the divalent cation ionophores A23187 and X537A. Ionophore activity was dependent upon the presence of Ca2+ (1.8·10?3?4·10?4 M) in the culture medium. The K+-selective ionophore valinomycin had no effect on ligand-induced redistribution of surface receptors. It is suggested that A23187 and X537A impair membrane-associated microtubules involved in transmembrane control of receptor mobility and topography. In contrast to the action of colchicine and vinblastine that bind directly to microtubules, it is proposed that ionophores indirectly affect microtubules by raising the concentration of Ca2+ in the cytoplasm to levels that favor microtubule depolymerization and inhibit microtubule assembly.  相似文献

Copper (II) accumulation has been investigated in the green alga Scenedesmus subspicatus G. Brinkmann considering both adsorption and uptake kinetics. Experiments were conducted in a Cu- and PH-buffered medium at different free Cu2+ concentrations that were neither growth limiting nor toxic. We distinguished between adsorption on the cell surface and intracellular uptake by extracting copper from the cells with EDTA. Data from short-term experiments were compared with data obtained from experiments under steady state conditions. The accumulation of Cu can be described by two processes, an initial fast adsorption occurring within a minute followed by a slower intracellular uptake. Metal uptake followed Michaelis-Menten kinetics and is mediated by two systems, one with low and the other with high affinity. The maximum uptake rates (1.30 × 10?-10 mol·[g dry wt algae]?1· min?1, 3.67 × 10?-9 mol·[g dry wt algae]?1·min?1), and the half-saturation constants (6.84 × 10?-14 M, 2.82 × 10?-12 M) for the two uptake systems were determined using the Lineweaver-Burk plot. The calculated maximum concentration of binding sites on the surface of the algae is initially higher (9.0 × 10?-6 mol Cu.[g dry wt algae]?1) than under steady state conditions (2.9 × 10?-6 mol Cu·[g dry wt algae]?1). This suggests that the initial binding to the algal surface comprises the binding to specific transport ligands as well as to inert adsorption sites. The conditional stability constant of the Cu binding to surface ligands was calculated as log KCu= 11.0 at pH 7.9. This freshwater alga has a high ability to accumulate Cu, reflecting its adaptation to the bioavailable concentration of copper.  相似文献

Microtubule-disrupting alkaloids and protein fixatives were used to investigate the nature of an active process that must occur within stimulator cells in order for them to intiate a unidirectional mixed lymphocyte response (MLR). Brief treatment of the stimulator cells (SC) with glutaraldehyde (0.15%), formalin (0.6%), or lanthanum chloride (10?3 M) abolished their capacity to activate responder cells (RC). Pre-treatment of SC with the microtubule-disrupting alkaloids, colchicine (c) (10?4 to 10?6) or clochicine + vincristine (c+v) (10?4 to 10?6 M) also abrogated their stimulating capacity. This capacity was not restored by the addition of supernates from untreated cultures, thereby excluding the possibility that the alkaloids acted by decreasing the release of soluble stimulatory factors from SC. The introduction of alkaloid-inactivated, mitomycin-treated RC as drug carriers did not affect the mitogenic response of untreated RC to concanavalin A. This excluded a significant leakage of alkaloids from the treated SC and uptake by RC during culture. Lumicolchicine produced no decrease in the stimulating capacity of SC. This suggested that the suppression induced by low concentrations of colchicine resulted from its specific disruption of microtubules. None of the above treatements quantitatively reduced the antigenicity of SC, as evaluated by humoral and cell-mediated lysis of the treated cells. Also, these treatements produced no significant changes in the specific binding of concanavalin A by SC. These indicate there is a functional interaction of microtubular structures with cell surface antigens that appears to regulate either the capacity of SC to associate with RC, or the ability of SC to form and stabilize stimulatory antigenic configurations on the cell surface.  相似文献

Cellular proliferation and generation of cytotoxic lymphocytes in mixed lymphocyte culture reactions (MLC) results from cellular interactions initiated by individual differences in cell surface structures determined by the HLA-D locus. Cellular microtubular assemblies (MTA) modulate cell shape and surface architecture and have also been implicated in mediating stimulatory signals from the lymphocyte plasma membrane to intracellular sites. To assess the role of MTA in alloactivation, we tested the effect of colchicine, a microtubule-disrupting alkaloid, on the MLC. Diametrically opposite results were observed depending upon the colchicine treatment protocol. Brief exposure of stimulating cells to 10?6M colchicine resulted in an increase in [3H]thymidine incorporation from 30, 694 ± 2787 to 47,345 ± 4361 cpm/culture (mean ± SEM) (P < 0.001), exposure of responding cells to 10?6M colchicine resulted in an increase from 33,054 ± 4012 to 46,790 ± 5458 cpm/culture (P < 0.01), and exposure of both cells to 10?6M colchicine resulted in an increase from 33,054 ± 4012 to 52,685 ± 6720 cpm/culture (P < 0.01). However, direct addition of colchicine to a final concentration of 10?6M to MLCs at different times resulted in complete suppression of proliferation when added as late as 96 hr, and 63% suppression when added at 120 hr. Pretreatment of stimulating, responding, or both cells with lumicolchicine did not enhance proliferation. Pretreatment of cells with 10?6 and 10?4 M colchicine enhanced proliferation, while pretreatment with 10?2M colchicine prevented blastogenesis. The potentiation of proliferation induced by colchicine was evident as early as 48 hr after the initiation of the MLC. Generation of specific cytotoxic cells in the MLC was also enhanced by exposing lymphocytes to colchicine prior to the proliferative phase in six out of eight experiments (specific chromium release = 168% of control) despite constant effector:target ratios. These findings indicate that early disruption of microtubules leads to enhanced cellular proliferation and generation of cytotoxic lymphocytes in allogeneic one-way MLCs and suggest that the state of polymerization of the MTA may modulate immune responses involving cell-cell interactions.  相似文献

The role of microtubules and microfilaments in the control of random movement of mouse peritoneal macrophages was examined by studying the colchicine and cytochalasin B (CB) effects. Colchicine in the concentration range of 10?8–10?4 M enhances the random movement of these cells. Enhanced movement of macrophages is observed only at colchicine concentrations which cause inhibition of their spreading and adhesion. CB does not enhance random movement at any concentration; it inhibits movement at 10?7 M and higher concentrations. Furthermore, though 10?8 M CB alone has no effect on the migration of macrophages, when present along with colchicine, the two drugs act synergistically and enhance random movement to a greater extent than colchicine alone. These findings suggest a cooperative interaction between microtubules and microfilaments in the control of movement of macrophages. The role of microtubules in silicon metabolism leading to valve formation was investigated in the pennate diatom Navicula saprophila Lange-Bertalot & Bonik. By using synchronized cells blocked after mitosis and cytokinesis but prior to cell wall formation, effects due to inhibition of mitosis were eliminated. Cells were treated with three anti-microtubule drugs to assess the role of microtubules. Chemical analogs to two of the drugs provided controls for inhibition not related to microtubule disruption. Although all three anti-microtubule drugs reduced cell separation at high concentrations (1 × 10?3 M), podophyllotoxin was the only drug which reduced cell separation at concentrations lower than 1 × 10?5 M. None of the drugs at any concentration tested affected cell viability. There was no differential inhibitory effect between the active and inactive drugs on silicic acid transport, total uptake, incorporation, or pool formation. There was no qualitative difference between silica incorporated in treated and untreated cells. A colchicine binding component was isolated from N. saprophila. The characteristics of colchicine binding suggest this component may be tubulin. Microtubules do not appear to be involved in any of the steps of silicon metabolism leading to valve formation and yet they have profound influence on the symmetry and pattern of the mineralized product, the siliceous valve.  相似文献

The glycoproteinic nature of the insulin receptor was indicated using two different approaches: 1. [125I]insulin binding to soluble receptors from mouse liver was inhibited by digestion with β-galactosidase or pretreatment with Ricinus communis I or concanavalin A. An other enzyme (neuraminidase) and lectins (wheat germ agglutinin, Dolichos biflorus) did not affect the binding reaction. These data confirmed that insulin directly interacts with the galactoglycoproteins of liver membranes. 2. The galactose oxidase-sodium boro[3H]hydride technique, previously used for labeling accessible membrane galactoglycoproteins, was again utilized to discern the components that interact with insulin. When liver membranes were equilibrated with 10?7 M insulin prior to labeling, the SDS gel radioactive profiles were specifically modified within two galactoglycoproteins of apparent molecular sizes 195 000 and 145 000, compatible with their participation in the insulin binding interaction. Membrane pretreatment with β-galactosidase or Sophora japonica lectin reduced the labeling in most peaks, thus supporting the argument for labeling sensitivity. Preincubation of membranes with 10?7 M proinsulin slightly hindered labeling while pretreatment with 10?7 M glucagon was ineffective, suggesting a specificity of the insulin effect. These data indicate the glycoprotein nature of the insulin receptor for two reasons: alteration of insulin binding after modification of the galactoglycoproteins, and alteration of galactoglycoprotein labeling after insulin binding. Two galactoglycoproteins, with apparent molecular weights 145 000 and 195 000, respectively, were identified and they are suggested to have insulin binding properties.  相似文献

The effects of colchicine and 2-Br-α-ergocryptine-methane-sulfonate (CB 154) on the release of prolactin and growth hormone have been studied in a clonal strain of rat pituitary tumor cells (GH3) in monolayer culture. These cultures produce both prolactin and growth hormone and release both proteins spontaneously into the medium without storing them in large amounts. Immunological methods were used to measure both intracellular and extracellular concentrations of the hormones. Colchicine (5 × 10?6 M for 3 hours) caused a 2- to 3-fold increase in intracellular concentrations of prolactin and growth hormone but, under basal conditions, had little or no measurable effect on the amounts of hormone accumulated in the medium during the course of the standard three hour treatment period. This latter finding evidently is due to a lag in the onset of drug action. Colchicine had little or no effect on accumulation of extracellular prolactin during the first two hours of treatment whereas such accumulation was depressed by over 60% during the third hour of treatment. Previous studies have shown that treatment of GH3 cells with thyrotropin releasing hormone (TRH) and hydrocortisone (HC) increases both intra and extracellular levels of prolactin and growth hormone, respectively. In cultures treated with TRH (5 × 10?8 M), colchicine (5 × 10?6 M for 3 hours) increased intracellular prolactin by about 70% and decreased extracellular hormone by 10%. In cultures treated with HC (3 × 1O?6 M), colchicine increased intracellular growth hormone by more than 100% and decreased medium concentrations of the hormone by 15%. Colchicine did not significantly alter total hormone (intracellular + extracellular) accumulation, cellular uptake of 3H-amino acids, or total cell protein synthesis. The synthetic ergot alkaloid, CB 154, (3.3 × 10?6 M for 3 hours) caused an 80% increase in intracellular, and a nearly 50% decrease in extracellular, prolactin without affecting the accumulation of growth hormone, the uptake of 3H-labeled amino acids, or overall protein synthesis in the cultures. Elevation of medium potassium concentration from a basal value of 5.3 mM to 3–5 × 10?2 M (by addition of KCl) decreased intracellular levels of prolactin by 85% and growth hormone by 55%. These effects of high potassium were blocked by colchicine and by CB 154. We conclude that colchicine, after a lag period of two hours, acts to inhibit the release of prolactin and growth hormone from GH3 cells. By the end of three hours of treatment, this inhibition is over 60% complete in the case of prolactin. The qualitatively different effects of colchicine and CB 154 on prolactin and growth hormone release suggest that these two secretory blocking agents probably act on GH3 cells by different mechanisms.  相似文献

Astroglial cultures from newborn mouse cerebral cortex contain [125I]insulin binding sites. Binding was specific reversible, time dependent and reached equilibrium after 45 min. Insulin analogues compete for this [125I]Insulin binding. Incubation of cerebral cortex astroglial cultures with insulin induced a time-and dose-dependent inhibition of the [3H]GABA high affinity uptake. A decrease in theV max rather than, an effect on theK m was observed. This effect was dose-dependent and effective at 10–10 M. Autoradiographic observations on the cell monolayer showed the presence of two groups of cells: one which strongly takes up [3H]GABA and consist in smaller GFAP positive process-bearing cells and another group of much flatter and larger GFAP positive cells which uptake was lower. The smaller stellate cells were apparently the most sensitive to insulin effect. These results: 1) confirm the presence of insulin binding sites on astroglial primary cultures, 2) show an effect of insulin of [3H]GABA high affinity uptake of these cells; this effect being optimal on a stellate-like population of astrocytes, and 3) indicate, that insulin may interfere in neuromodulation through astroglial signals.  相似文献

Adipose cells have been recognized as an integral component of the bone marrow hematopoietic microenvironment in vivo and as an essential cell type required for in vitro maintenance of stem cells. Four stromal cell lines obtained from the adherent cell population of murine bone marrow cultures have been enriched and purified by multiple trypsinizations. We noted that these cell lines exhibited an accumulation of vacuoles of lipid, the extent of which varied be-tween cell lines in response to a change from medium containing 10% fetal calf serum to medium containing 20% horse serum. The lipid was lost when the cell lines were transferred back into the medium supplemented with fetal calf serum. In light of the reported lipogenic and antilipolytic effects of insulin on fibroblasts and adipocytes, we investigated the ability of insulin to induce adipocyte transformation of these bone marrow stromal cell populations. Three cell lines were exposed to bovine insulin at concentrations ranging from 10?9 to 10?6 M. All three cell lines responded to the insulin by accumulating lipid, but the extent of accumulation and the insulin concentration at which maximum lipid content was attained were population specific. One cell line (MC1) responded fully at physiological levels of insulin (10?9 M), whereas the other two showed lipid accumulation only at pharmacological concentrations. The initial growth of MC1 was inhibited in the presence of 10?9 M insulin which is compatible with the observed differentiation to adipocytes. The growth of MC3 was unaltered in the presence of physiological concentrations of insulin, whereas that of MC4 was accelerated. Grafts of organ cultures of the cell lines under the kidney capsule of syngeneic mice developed specific characteristics rep-resentative of the different cell lines. In particular, the majority of the grafts of MC1 consisted primarily of fat cells which were not observed in the grafts of MC3 and MC4. These data strongly suggest that these cell lines comprise cells with different potentialities and that the MC1 line represents a preadipocyte stromal cell of bone marrow.  相似文献

The addition of insulin (4.0 × 10?11 M) or acetylcholine (10?6 M) to isolated hepatocytes stimulated glycogen accumulation and this stimulation was more pronounced when the medium glucose was raised from 50 to 300 mg percent. Studies with [14C]-glucose showed a two-fold stimulation in glycogen synthesis by the addition of insulin (4.0 × 10?11 M) or acetylcholine (10?6 M). A sixteen percent increase in the activity of glycogen synthase was observed in cells incubated for 10 minutes with insulin (4.0 × 10?11 M) or acetylcholine (10?6 M), whereas at one hour incubation a 40 percent increase in activity was observed with the same concentration of insulin or acetylcholine. Cells dissociated from the R3230AC mammary adenocarcinoma from intact and diabetic rats were examined for insulin binding and glucose transport. The Kd for insulin binding, ~ 10?10 M, was similar in all tumors studied. However, the apparent number of receptor sites per cell increased in cells from diabetic rats. Kinetic analysis of 3-0-methyl glucose (3-OMG) entry showed both diffusional and passive carrier characteristics. Insulin (4 × 10?9 M) in vitro did not affect diffusional entry, whereas the hormone altered the passive carrier system, as reflected by an increase in Km and Vmax. Insulin decreased initial velocity of glucose transport at 4–6 mM glucose levels but increased initial velocity of glucose transport at 20 mM glucose. An explanation of the role of insulin on tumor growth in vivo from effects on glucose transport in vitro is proposed.  相似文献
