The efficacy of an injectable microsphere formulation of ivermectin for control of the cattle tick, Boophilus annulatus (Say), was tested on 2 groups of 6 Hereford heifers held on separate 7-ha, tick-infested, buffel grass pastures. Cattle in one pasture were injected subcutaneously in the neck with a controlled-release microsphere formulation of ivermectin at the rate of 2.4 mg AI/kg body weight; the other group was injected with carrier only. Beginning 4 wk after injection and continuing throughout the remainder of the test (16 wk), no engorged ticks (> or = 5.5 mm) were found on any of the treated cattle, whereas large numbers of engorged ticks were found on the untreated controls. During this period, a few ticks were recovered from untreated sentinel animals placed in the treatment pasture during 7-8 wk after treatment, but none were recovered from animals exposed from 11-12 wk or 14-15 wk. Large numbers of B. annulatus ticks were found on untreated sentinel cattle placed in the control pasture during these same periods. Although the cattle, pastures, and tick habitat were approximately equal, the treated cattle gained an average of 77 kg compared with an average of 42 kg for the control group. This technology offers a possible alternative to the current official program of dipping and vacating pastures for eradication of Boophilus sp. infestations from the quarantine zone in southern Texas. Larger scale testing is needed to determine the potential of the injectable microsphere formulation and to optimize its use in eradication or control strategies.  相似文献

Abstract. The effect of the tick Amblyomma variegatum Fabricius on liveweight gain (LWG) on yearling cattle over 16 weeks was evaluated under quasi-natural conditions. Early in the exposure period when animals may have been in a state of suppressed metabolism, the effect of tick feeding on LWG was negligible. Overall, however, the difference in LWG between tick-free and tick-infested groups of cattle was 4.95 kg after 16 weeks. This difference between the groups was used to estimate a lower LWG of 48–63 g due to each engorging female tick.  相似文献

旨在探讨双酚A (BPA)对东亚三角涡虫的急性毒性及神经系统相关酶活性的影响。采用不同浓度的BPA处理涡虫24 h、48 h、72 h,求出半致死浓度,以此为基础,采用不同浓度的BPA处理涡虫24 h、72 h、144 h,测定AchE、ChAT及Na+~K+^-ATP酶活性。BPA对三角涡虫的24 h、48 h、72 h,LC50分别为12.18 mg/L、8.49 mg/L、6.43 mg/L。ChE、ChAT、Na+~K+^-ATP酶活性对BPA反应敏感,具有较好的规律性。24 h处理组,AChE酶活力随BPA浓度的升高而升高,72 h和144 h处理组则呈现先上升后下降的趋势,除0.643 mgBPA/L以外,其它处理组均表现出了时间-效应关系;在BPA胁迫下,ChAT酶活力均呈现降低趋势,在BPA浓度为0.643 mg/L、1.286 mg/L时,表现出了时间-效应关系;24 h、72 h处理组Na+~K+^-ATP酶活力随BPA浓度升高呈现先上升后下降的趋势,144 h处理组则呈现下降趋势,Na+~K+^-ATP酶活力随BPA胁迫时间的延长呈现先升高后下降的趋势。BPA对涡虫具有较强的生态毒性,AchE、ChAT、Na+~K+^-ATP酶活性可与其他敏感指标一起作为水体BPA污染的早期监测指标。  相似文献

Abstract Two experiments compared broods that were naturally tick-infested with an equal number that were rendered tick-free by application of an acaricide to their nests and of barriers against further infestation. In the first experiment conducted in 1991–92 nestlings in tick-infested broods had up to 159 larval ticks at once and a mean infestation of 23.6 larval ticks per chick per day. The chick's mean tick load was inversely correlated with its longevity. Chicks that survived to at least 18 days post-hatching had significantly lower larval tick loads than those that died by 18 days, excluding the third-hatched chick in each brood whose survival rate was low irrespective of tick-infestation. At 7 days post-hatching, tick-infested chicks had a lower haematocrit and higher polychromasia than tick-free chicks. I infer that blood loss anaemia caused the deaths of tick-infested chicks in their first week. None died in their second week and the demise of those in their third week may have been due to the paralysis manifested prior to their death. I conclude that heavy tick infestation of their chicks reduced the breeding success of the parent egrets below that of the egrets whose chicks were kept free of ticks. In the second experiment in the 1992–93 season the mean level of tick infestation was much lower (5.1 per chick) and these chicks survived equally with tick-free chicks through fledging.  相似文献

Abstract. Lactating Sanga cows of the Mashona breed from Zimbabwe, receiving either a low or a high level of nutritional supplement, were exposed to two levels of infestation of adults of the brown ear-tick, Rhipicephalus appendiculatus in the highveld of Zimbabwe. The effect of the ticks on the milk yield was measured over an 11-week period during the rainy season from January to April 1986. A technique in which calves were weighed before and after suckling was used to estimate milk yield. There were significant treatment effects of ticks (P < 0.05) on milk production but no significant differences in liveweight gain between calves from tick-free and tick-infested dams. The loss in milk production was poorly related to the number of female ticks that engorged, being 9 g (SEM 4) per tick. A Friesian x Hereford (Bos taurus) reference group of cattle carried 50% more ticks than the Mashona cows, illustrating a difference in resistance between the breeds. Thirteen screw-worm (Chrysomya bezziana) strikes were recorded amongst the thirty-two Mashona cows compared with twenty-one amongst the ten Friesian × Hereford animals between January and the end of March.  相似文献

Angiotensin-converting enzyme (ACE) activity has been determined in the semen of certain avian and mammalian species as well as its release during cold shock. The maximum and minimum levels of this enzyme were found in mammalian spermatozoa and in seminal plasma, respectively. It was found that ACE activity in mammalian spermatozoa was more pronounced than in the seminal plasma, whereas in the avian species a revers pattern was observed. However, there were no significant differences in ACE activity in spermatozoa and seminal plasma between layer and broiler strains of avian species. By contrast, ACE activity in the spermatozoa and seminal plasma of buffalo bulls was significantly higher (P/ 0.01) than in cattle bulls. Cold shock did not significantly alter semen characteristics in avian species, while a significant (P/ 0.01) decrease in sperm live counts and motility as well as a corresponding increase in morphological abnormalities were observed in the spermatozoa of cattle and buffalo bulls due to cold shock.  相似文献

Acetylcholinesterase (AchE) activity in erythrocytes and blood levels of cortisol and insulin were investigated in athletes training under different bioenergetic conditions (sprinters, middle-distance runners, and marathoners). The groups of sprinters and marathoners had a decreased enzyme activity compared to nonathletes (p < 0.05). In response to a standard exercise, load AchE activity increased in the groups of middle-distance runners and marathoners. A relationship was observed between the level of AchE activity and the cortisol-to-insulin ratio in the blood. This ratio is specific to the type of bioenergetic conditions and increases in the following order: controls, middle-distance runners, sprinters, and marathoners. In vitro experiments revealed an effect of insulin on AchE activity. This effect was significantly lower in sprinters than in the control group. A reduction in AchE activity and an increase in the cortisol-to-insulin ratio are considered as factors increasing metabolic turnover in athletes, mainly, lipid turnover. This mechanism ensures effective mobilization of substrates at the start of physical exercise and their recovery after. The observed relationship between the insulin level and the AchE activity may prove to be a mechanism of regulation of the insulin level. This relationship may change during adaptation to physical exercise, as in the case of sprinters, when the sensitivity of AchE to the inhibitory effect of insulin is decreased. A high blood level of cortisol and insulin is a distinctive feature of sprinters, which provides for a higher turnover of carbohydrates. In marathoners, low AchE activity leads to an increased effect of acetylcholine, which is manifested by an increased cortisol level and a decreased insulin level, thus providing for a higher lipid turnover.  相似文献

Cattle demonstrate divergent and heritable phenotypes of resistance and susceptibility to infestation with the cattle tick Rhipicephalus (Boophilus) microplus. Bos indicus cattle are generally more resistant to tick infestation than Bos taurus breeds although large variations in resistance can occur within subspecies and within breed. Increased tick resistance has been previously associated with an intense hypersensitivity response in B. taurus breeds; however, the mechanism by which highly resistant B. indicus cattle acquire and sustain high levels of tick resistance remains to be elucidated. Using the commercially available Affymetrix microarray gene expression platform, together with histological examination of the larval attachment site, this study aimed to describe those processes responsible for high levels of tick resistance in Brahman (B. indicus) cattle that differ from those in low-resistance Holstein-Friesian (B. taurus) cattle. We found that genes involved in inflammatory processes and immune responsiveness to infestation by ticks, although up-regulated in tick-infested Holstein-Friesian cattle, were not up-regulated in Brahman cattle. In contrast, genes encoding constituents of the extracellular matrix were up-regulated in Brahmans. Furthermore, the susceptible Holstein-Friesian animals displayed a much greater cellular inflammatory response at the site of larval R. microplus attachment compared with the tick-resistant Brahman cattle.  相似文献

When the cattle tick Boophilus microplus, after dropping from its host, was maintained at 27 C, the digestion of hemoglobins in the gut proceeded at a steady rate and was virtually complete by the 13th day. The rate was essentially the same whether the ticks were strains susceptible (Yeerongpilly) or resistant (Biarra) to organophosphorous compounds or whether the hosts were British breed, Brahman or banteng cattle, or buffalo. Ferrihemoglobin appeared in the gut contents and hematin was deposited. About 10% of the hematin released from the hemoglobin was transferred to the eggs and from them to the larvae. Translucent ticks, sometimes found on heavily infested hosts and referred to as “serum” ticks, had about half the normal hemoglobin content.  相似文献

Two herds of experimental Sanga cattle were maintained under traditional savanna grazing management for three years in the Central Province of Zambia. One herd was kept free of ticks by regular acaricide treatment, while the other was given no tick control. Milk production, growth rate, fertility and mortality were monitored throughout the trial. The tick-free herd performed significantly better than the tick-infested herd, but the value of the additional production was much less than the cost of the acaricide used. it is concluded that there is no economic justification for intensive tick control under these conditions. However, strategic tick control would be justified if the quantity of acaricide used could be reduced by 50% without any major reduction in benefits. The results indicate that treating calves below 45 days of age reduces their performance, as does treating cows during periods of very low tick challenge. Therefore, it seems likely that economically beneficial strategic tick control policies could be developed.  相似文献

Field collections of ticks from two breeds of cattle showed that the common species of ticks wereAmblyomma hebraeum, Rhipicephalus appendiculatus andRhipicephalus evertsi evertsi. The density of these species was higher on Hereford than on Brahman cattle. The results also indicated that the density of immature stages of the above-mentioned ticks is higher on Hereford than on Brahman cattle.A positive correlation was found between the number of ticks on the cattle and the serum gamma globulin levels, an indication of an increase in the production of antibodies. An inverse relationship was found between tick burden and red blood cell count and hemoglobin concentration. Other ablood components, such as basophils, eosinophils and lymphocytes were not affected by the changes in the density of the tick populations. It appears that resistance may have been acquired by the hosts and that Brahman cattle may acquire resistance to a higher degree than Herefords.  相似文献

The actions of ethanol on the structural stability of acetylcholine receptor (AchR)-enriched membrane vesicles and the activity of various molecular forms of acetylcholinesterase (AchE) were investigated, using the receptor and the enzyme isolated from the electric organ of Torpedo californica. In the presence of ethanol up to 200 mM, the thermogram of AchR-enriched membranes exhibited no significant decrease in the temperature (td) of receptor transition at 57 degrees C, but a decrease in the enthalpy change (delta Hd) indicated a slight ethanol-induced structural perturbation. The presence of 12.5 nmol alpha-bungarotoxin also caused a decrease in delta Hd. A complete loss of the receptor transition was observed at a higher concentration 500 nmol of alpha-bungarotoxin and no recovery of the transition was found with the addition of 200 mM ethanol. The results suggested a noncompetitive interaction of ethanol with the receptor. In the presence of 200-1000 mM ethanol, the activity of two soluble forms of AchE, a higher (117 S) aggregate and a lower (10 S) aggregate was not significantly affected. Comparing the activity of these two aggregates over a wide concentration range of ethanol (200-2000 mM) revealed no obvious difference in the level of ethanol effect between them. However, after removal of ethanol, the higher aggregate form of AchE exhibited a greater recoverability of the activity, suggesting a possible slightly greater structure-functional stability for it. Studies of soluble AchE and membrane-bound AchE showed that the presence of 200 or 600 mM ethanol caused a greater level of inhibition in membrane-bound enzyme than in soluble enzyme, possible due to a disruption of protein-lipid interaction needed to maintain the conformation of membrane-bound AchE. Interestingly, at a much higher concentration of ethanol (2.0 M), membrane-bound AchE became more resistant to ethanol than did the soluble forms of AchE. In this case, the effective concentration of ethanol felt by the enzyme was expected to be less for membrane-bound AchE, owing to ethanol's solubility in lipids.  相似文献

The effect of tick infestations on body weight and various blood parameters was monitored in juvenile northern brown bandicoots (Isoodon macrourus) after release into tick-infested or tick-free enclosures. Three species of ticks were observed in the enclosures, Haemaphysalis humerosa, Ixodes tasmani and Ixodes holocyclus. Bandicoots released into tick-infested enclosures showed a reduced growth rate (1.8 versus 2.5 g/day increase in body weight), a reduced haematocrit value (27.4 versus 40.0%) and an increased number of white blood cells when compared with bandicoots released into tick-free enclosures. These results suggest that tick infestations may influence the health of juvenile I. macrourus.  相似文献

The postthaw motility and fertility of buffalo and cattle semen is reduced when they are cryopreserved for artificial insemination. In the present study, an attempt was made to characterize the cryogenic changes in proteases and antiprotease activities (APA) of buffalo and cattle semen because these proteolysis regulators have been reported to be associated with sperm motility and fertility. Buffalo sperm demonstrated at least two major proteases of 45 and 42 kDa and three minor proteases of 95, 52, and 33 kDa. Similarly, cattle sperm demonstrated three major proteases of 62, 45, and 42 kDa and two minor proteases of 85 and 78 kDa. Buffalo seminal plasma demonstrated at least three major proteases of 78, 68, and 62 kDa and one minor protease of 98 kDa and cattle seminal plasma demonstrated one major protease of 68 kDa and two minor proteases of 78 and 75 kDa. Except for the 45 kDa protease, most of the previously mentioned proteases were found to be metalloproteinases. Compared with fresh sperm, cryopreserved buffalo and cattle sperm demonstrated a major protease band of 52/49 kDa and the activity of this protease reduced progressively with the duration of cryopreservation. On the contrary, compared with the fresh seminal plasma, cryopreserved buffalo and cattle semen extenders displayed the presence of a new protease band of 45 kDa and demonstrated that this protease activity was leaked from buffalo and cattle cryopreserved spermatozoa. Buffalo and cattle seminal plasmas displayed at least two major APA of 86 and 26 kDa. Compared with buffalo, cattle seminal plasma demonstrated significantly greater APA. Thus, the present study demonstrated the presence of an array of proteases and APA in buffalo and cattle semen and the activities of which changed during cryopreservation. The leakage of the specific protease activity and changes in the proteases and APA might be attributed to reduced motility and fertility of cryopreserved semen in these species.  相似文献

Early experiments performed by our group with the phage display technique revealed the potential for using epitope-displaying phages (mimotopes) as a tool for tick antigen discovery. Thus, as a preliminary study, inflammatory reactions induced by phage display tick-borne candidates were investigated by using the cutaneous hypersensitivity test. The profile of selected Rhipicephalus microplus mimotopes was assessed on tick field-exposed cattle and our data indicated a pattern similar to immediate hypersensitivity reaction and not a delayed immune response as expected. However, the wild-type phage inoculation surprisingly induced a strong immediate response on its own. Such reactions indicate that the wild-type phage may have hidden many of the potential reactions raised by the mimotopes. The study of the inflammatory reactions to these phage mimotopes in tick-infested hosts may provide basic information about the immune reaction. Finally, this work is of relevance for when considering research alternatives for finding and characterization of antigens by the phage display technique.  相似文献

A modified high pressure liquid chromatographic method using lactose (Gal beta 1----4Glc) as an exogenous acceptor has been used to characterize the sialyltransferases known to increase in the serum of colchicine-treated rats. The results show a 10-fold increase of Gal beta 1----4GlcNAc alpha 2----6 sialyltransferase (alpha 2----6 ST), whereas the Gal beta 1----3GlcNAc alpha 2----3 sialyltransferase showed only 1.6-fold increase in the serum after 17 h of colchicine treatment. The sialyltransferase activity in serum using exogenous desialylated, alpha 1-acid glycoprotein as acceptor also showed an eightfold increase. In liver homogenate and Golgi membrane, the sialyltransferase activity when assayed with desialylated alpha 1-acid glycoprotein as acceptor showed a slight decrease after 4 h, but returned to normal level after 17 h. A similar trend was seen when the two transferases were assayed with lactose as acceptor. The antiserum to rat alpha 2----6 ST inhibited the sialyltransferase activity in serum, liver, and jejunal incubation medium. Jejunal sections from rats treated with colchicine for 4 h in presence of heated serum showed a decrease of sialyltransferase, with consequent increase of the alpha 2----6 ST enzyme activity in the medium. This result suggests that intestinal tissue could be a source of increased serum enzyme activity in colchicine treatment.  相似文献

Strategic tick control is an attempt to control ticks and reduce losses in animal production due to tick infestations while decreasing the cost for this control. Tick control is understood as a necessity to achieve maximum animal production from animals raised in tick-infested areas. At the same time, it has become difficult to justify intensive, year-round tick control for local breeds and their crosses. The increasing costs of acaricides, maintenance of the infrastructures, salaries for personnel, etc. have created a tick control programme where the benefits may not be adequate to justify the expense. This has been the case in Burundi. This programme with adequate participation from the cattle owners of Burundi and organizational support from government personnel can help solve this economic problem. The strategic programme reduces the period of tick control to four months each year. Once optimal participation is achieved, this period can probably be reduced to three months (Kaiser et al., 1988). The period of treatment corresponds to when there is maximal feeding activity of adult female ticks on the bovine population within a certain region. Cattle should be treated once each week throughout the four-month period in order to prevent female tick engorgement (based on the life cycle of R. appendiculatus). This will greatly reduce the number of female ticks which are available to recycle the population of ticks in the participating area. At the same time, a certain population of ticks must be maintained in order to assure contact between the cattle, ticks, and tick-borne diseases of this particular region.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Ever since their accidental introduction, cattle ticks, gastrointestinal nematodes and buffalo flies have been major parasites of cattle in northern Australia. Enormous effort and resources have been directed at chemical control of these parasites but the problem persists in undiminished form. The principal control measure remains the use of breeds that have some degree of parasite resistance. No breed is completely resistant and all are at times adversely affected by parasites. Complete resistance is the ultimate solution but has been generally ignored as a commercial reality. Studies at Rockhampton have demonstrated that completely resistant lines can be developed by genetic means within a commercially acceptable timeframe from even the most parasite-susceptible breeds. Genetic changes in tick and worm resistance over 15 years in response to selection for increased tick resistance in the Belmont Adaptaur (Bos taurus) line are reported. Costs and benefits of achieving increased tick resistance are examined and the applicability of the results to other breeds is discussed. Breed differences in resistance to buffalo flies and their effects on live weight are also reported and the possibility of selecting for increased buffalo fly resistance is explored.  相似文献

Thirteen restriction endonucleases were used to investigate nuclotide sequence variation in the 18S rRNA DNA of 88 individuals from ten Sarcocystis taxa collected as cysts from their intermediate hosts, swine, cattle and water buffalo. A DNA sequence of approximately 900 bp was used. A total of 26 electromorphs were detected. The electromorphs were sorted into seven different haplotypes that coincided with the six named species and an unidentified species from cattle. These findings support those of our morphological examinations, which suggested that the taxa resembling Sarcocystis hirsuta, S. hominis, both found in water buffalo, and S. sinensis found in cattle, are not new species but are in fact S. hirsuta and S. hominis as found in cattle, and S. sinensis as found in water buffalo; this finding supports the idea that these species can utilize both cattle and water buffalo as intermediate hosts and are not restricted to one or the other host group as previously thought. PCR-RFLP resolved by agarose gel electrophoresis is shown to be an easy and rapid method of discriminating between these species.  相似文献

Karminomycin effect on the activity of some serum enzymes, such as hexokinase (HK), lactate dehydrogenase (LDG), its isoenzymes and glucose-6-phosphatase (G-6-P-ase) was studied. Biochemical assays were applied to 52 patients with neglected malignant tumors. The course dose of the drug was on the average 72mg. The objective antitumor effect was registered in 15 patients. A reliable increase in the values of LDG-5 and G-6-P-ase was observed after the treatment course in the combined group consisting of all the patients subjected to the biochemical assay. Normalization of the serum enzyme spectrum was observed in 15 patients effectively treated with karminomycin: activity of HK and the cathode fractions of LDG decreased. When treatment with karminomycin was ineffective (37 cases), the changes in the enzymatic activity recorded before the treatment further aggraviated. It was found that the level of G-6-P-ase in the patients' treated with karminomycin increased independent of the treatment effect which was probably associated with its toxic effect on the liver.
