By means of plastic reconstruction peculiarities in structure and spatial organization of the palatal glands have been revealed in newborns and mature persons. Morphometrical analysis has been performed in serial semithin sections stained with toluidine++ blue. The palatal gland ducts in the newborn are presented as a system of epithelial tubules with a gradually enlarged external and internal diameters and wall thickness towards direction of saliva flow. In the excretory ducts of the newborn sharp dilatations, facilitating to delay and accumulation of the secretion are not revealed. In the excretory ducts system in the mature person certain changes in the wall thickness are revealed and a number of narrowed and dilated areas have been found; they probably effect the secretion rate. This, in its turn, should produce some effect on the definite composition of saliva.  相似文献

The lymphatic bed of the thyroid gland has been studied in 24 intact rats. Three techniques facilitating to reveal lymphatic vessels in the organ have been used: preparation of semithin sections, injection of the blood bed with methyl methacrylate with a successive chemical extraction of the preparations, injection of the blood bed with liquid solution of methyl methacrylate with a consecutive study of the preparations in the scanning electron microscope. Methods of electron histochemistry (revealing horseradish peroxidase) and kryofractography have been applied. Construction of the thyroid lymphatic bed, structure of the wall are described, fibroblastic membrane (F-membrane) is revealed and the signs are presented, that allow to differentiate F-membrane from endothelium of the lymphatic capillaries. The pathways of lymph outflow in the rat thyroid gland consist of the following links: interstitial space in the interlobular spaces of the gland, into them the tissue liquor (lymph) is filtered ; plates of the F-membrane regulating direction of excessive albumin-containing liquor in the lymphatic capillaries, surrounding groups of 5-11 follicles, embracing the microlobule of the gland and situating in the interlobular spaces, deferent lymphatic vessels.  相似文献

The influence of various histologic techniques on the results obtained by morphometric analysis of the rat thyroid gland was studied. The limits of thyroid follicles were more clearly defined in both silver-impregnated paraffin-embedded sections and resin-embedded semithin sections than in routinely stained paraffin-embedded sections, thus enabling more accurate measurements of thyroid structures. Due to its simplicity, the silver impregnation method is clearly useful for histomorphometric studies when large numbers of measurements are involved. C cells were easily identified in paraffin-embedded sections by immunohistochemical staining. The measurement of interstitial tissue in sections without immunostaining of C cells led to an overestimation of the volume fraction of interstitial tissue.  相似文献

By means of the multilayer graphic and plastic reconstruction methods using series of semithin sections, spatial tridimensional organization of the epithelial complexes and blood microcirculatory bed in the rat palatal salivary glands and the lacrimal gland of the human newborn have been studied. Since their ducts serve not only for discharging their secrete into the external medium, but also for accumulation (as collectors), the sublobular unit--adenomere should be referred to as a part of elementary level of organization of the epithelial complexes. The adenomere has in its composition a collecting centrally situating duct. However, while studying structure of the blood microcirculatory bed, it is found out that there is not any strict territorial correspondence between its functional units and structural units of the glandular epithelium. Nevertheless, giving a great importance to a tight syntopic connection of the collecting ducts of the adenomeres with the postcapillary venules (that belong to filtrating microvessels), these are sublobular units--adenomeres that are distinguished as structural-functional units in the glands.  相似文献

Morphological and histochemical changes of ultimobranchial follicles of thyroid have been investigated in rats from newborn to 18 months of age. The first well-delimited ultimobranchial follicles, though with no lumen, were detected in the thyroid gland of 10-day-old rats. At 30 days of age, follicles possessing regular lumina were present in the thyroid. With age, the follicles gradually increased in volume assuming extreme dimensions in adult age. The follicles displayed varying shapes from simple cysts to bizarre forms. From the age of 50 days the cells of the follicular wall are separated from the cell debris contained in the lumen. The latter gave a PAS positive reaction. The cells of the ultimobranchial follicles did not exhibit argyrophilia and metachromasia showing that they differ considerably from the C-cells likewise of ultimobranchial origin, which are known to give marked argyrophilic and metachromatic reactions.  相似文献

By means of the histostereometrical method, peculiarities of the thyroid gland formation have been studied in the lemming (Dicrostonyx torquatus) fetuses and newborns at the stage of the population quantity drop, as well specificity of the organ's morphofunctional state during the postnatal period. On the 14th prenatal day the thyroid gland already has the follicular structure. Up to the 19th day progressive follicular growth, accumulation of colloid and increase of its density occur. On the 19th-20th days, as well as in newborn animals, there are certain signs, demonstrating as essential activation of the thyroid gland function. In the newborn animals cavities of the completely formed follicles are devastated. During the postnatal period again growth of follicles, accumulation of colloid are observed, signs of hypersecretion of the hypophysis appear.  相似文献

This study demonstrates gross arterial supply, venous drainage and microvascular patterns of larval and adult thyroid glands in the African Clawed Frog, Xenopus laevis by scanning electron microscopy of vascular corrosion casts and light microscopy of stained serial tissue sections. Results confirm published findings gained by microscopical dissections with respect to gross arterial supply. However, in adult frogs one rather than two thyroid veins are found. This study reveals for the first time that bilaterally located thyroid glands in premetamorphosis have immature capillary networks, lack a clear hierarchy of blood vessels, and show many signs of sprouting angiogenesis. During metamorphic climax, blood vessels gain a clear hierarchy and capillaries form closed networks around thyroid follicles. From climax onwards, non‐sprouting angiogenesis (intussusceptive microvascular growth) becomes the prevailing mode of angiogenesis intensifying follicle capillarization. Due to narrow interfollicular spaces, thyroidal arterioles remain superficial while draining venules are located interfollicularly. In contrast with the mammalian thyroid gland where most thyroid follicles have their own capillary beds, most thyroid follicles in Xenopus share their capillary beds with neighbouring follicles. Consequently, the concept of individual morphological and functional angiofollicular units applicable to adult mammalian and human thyroid glands is not applicable to larval and adult amphibian thyroid glands.  相似文献

Summary The cholinergic innervation of the thyroid gland has been studied in the human. AChE positive nerve fibers are localized in the wall of the thyroid artery, arranged in two plexuses, a superficial (adventitial) and a deep one (medial). The glandular tissue is provided with cholinergic nerve fibers, localized between and around thyroid follicles. The present results suggest that the endocrine activity of the thyroid gland is also under the control of the autonomic nervous system.  相似文献

A comparative study of the thyroid gland development in the human male and female embryos and prefoetuses was carried out. The development of incretory part of the testicles was shown to occur earlier than the differentiation of follicles in the thyroid gland and the appearance of colloid in it. Sexual differences were noted in the appearance of follicles, connective tissue, nerve fibres in the thyroid gland (earlier in the male foetuses). The processes of organ differentiation in the male foetuses were more active than in the female ones during the whole prenatal period of development. A suggestion is put forward that the sexual hormones of embryonic testicles influence the thyroid gland differentiation.  相似文献

By means of macro-microscopical preparation methods, horseradish peroxidase injection, semithin sections, electron microscopy histotopography and composition of microvessels of the sciatic nerve have been studied in 20 mature male white rats. The perineural membrane has been stated to have no its own vascular network. In composition of the sciatic nerve and its large branches muscular venules are revealed; they run longitudinally along the whole length of the endoneural space and are tributaries of the epineural veins. A suggestion is made that presence of the contractile apparatus in the venular wall can play an important role in formation of the postcapillary resistance, regulating the hydrostatic pressure value in the lumen of endoneural capillaries ad intensity of liquor filtration into the interstitial space.  相似文献

The three dimensional morphology of the thyroid tissue in juvenile rainbow trout Oncorhynchus mykiss was determined using serial transverse and longitudinal sections of lower jaws stained with haematoxylin and eosin. The thyroid tissue comprised interconnecting tubular units not the follicles that are widely described in the literature. Some of these tubules were subdivided by branching, giving the appearance of follicles in the sections. The thyroid tissue was aggregated into two major concentrations of tissue, associated with the first and second branchial arteries, with the majority of the tissue located dorsal and lateral to the aorta. These observations in juvenile rainbow trout suggest that thyroid tissue of some species of fishes may not be follicular, as previously assumed. The physiological and evolutionary implications have yet to be determined.  相似文献

Summary To solve the unsettled problem of the identification of the gastrin cells, a study has been undertaken on the electron microscopical characteristics of the gastrin-containing cells of the cat pyloric mucosa. Cells which, on semithin sections, were shown by an immunofluorescence method to contain gastrin, have been identified on serial ultrathin sections. The ultrastructural features of these cells are those which have been described as characteristic of the G cells of the antropyloric mucosa. Other non-entero-chromaffin endocrine cells, which have been recognized as the D cells of the gastro-intestinal mucosa, did not result to contain gastrin.  相似文献

The literature concerning the problems of myocyrdial sinusoids has been analysed and morphological investigation of 98 human hearts has been performed. As demonstrates the comparison of the literature data, the term "sinusoid" in the human heart is applied to different structures. In some cases--to designate dilated areas of the venous part of the blood bed in the ventricular walls, in others--"hollow spaces", "intercellular spaces" and Vieussens--Thebesius vessels. Combining methods of intravascular injections and studying serial histological sections, it has been found that the sinusoids--the "hollow spaces", the "intercellular spaces", the "Veussens--Thebesius vessels" present the areas of intertrabcular spaces, which get into the planes of some sections, not being components of the myocardial blood bed. Spatial angioarchitectonics of the sinusoids--dilated areas in the venous part of the myocardial blood bed have been investigated by means of the plastic reconstruction method. It is suggested to apply the term "sinusoids" only to the given parts of the cardiac blood bed.  相似文献

Ultrastructural characteristics of corticotropic cells have been studied on serial semithin and ultrathin sections of the hypophysis from intact rats by the immunofluorescence and electron microscopic methods. It is shown that the population of corticotropic cells is heterogeneous and consists of degranulated, moderate granulated, hypergranulated cells and transitional forms among them. Ultrastructurally corticotropic cells correspond to Siperstein-Nakayama's ACTH cells. Kurosumi-Kobayashi "corticotrophs" fail to react with the antiserum to ACTH, so they have not regard for its production.  相似文献

In this research the TFR localization in non functioning human thyroid nodules and in the extranodular thyroid tissue, using an immunohistochemical technique, has been studied. For this study a monoclonal antibody (B3/25) against TFR and the peroxidase technique have been utilized. Moreover a morphometric comparative analysis was carried out based on the following parameters: 1) mean immunoreactive area for microscopic field, 2) mean value of immunoreactive follicular perimeter, 3) integrated optical density, 4) % of immunoreactive area on total examined area in nodular and extranodular tissue. The immunoreactivity was detected in some follicular cells in a number of follicles randomly distributed in the extra nodular tissue. As concern the non functioning thyroid nodules, the positivity was localized in the generality of the follicles both in the flattened epithelial cells of the larger follicles and in the cuboidal cells of the smaller ones. The morphometric parameters confirm a statistically significant difference of immunoreactivity between extranodular and nodular tissue. These results suggest that TF might play a role in the cellular proliferation of thyroid gland.  相似文献

Rat thyroid follicles in both the normal and the inverted configuration have been cultured in suspension. It has been found that normal follicles (i.e., those having a structural organization similar to the follicles in vivo) express most functional properties of the thyroid gland. They trap iodide, synthesize, secrete, and iodinate thyroglobulin, and secrete thyroid hormone into the culture medium. On the contrary, in inverted follicles (i.e., those having a cell polarity reversed with respect to normal) we were unable to detect expression of some of these functions. We conclude that both the follicular organization and the proper cell polarity are needed for the full expression of thyroid functional properties.  相似文献

A study on thyroid glands of Wistar rats of ages ranging from 1 to 120 days was carried out. The glands were serially sectioned and stained for calcitonin using the peroxidase antiperoxidase method. All the thyroids contained ultimobranchial follicles (UBF) located partially embedded among the usual follicles but in a 5-day-old rat this structure showed an unusual position in the interstitium of connective tissue between the cartilage of the trachea and the thyroid gland. We have observed in the wall of that UBF the presence not only of resting C cells but also mitotic figures of C cells. Furthermore, on the opposite side of the same UBF an active area of formation of thyroid follicles was found. These observations provided the first evidence of the contribution of the UBF in the formation of C cells during the postnatal life of the rat. Furthermore, it is suggested that some C cells may share a common origin with ultimobranchially derived follicular cells.  相似文献

The thyroid is a bilobated endocrine gland localized at the base of the neck, producing the thyroid hormones T3, T4, and calcitonin. T3 and T4 are produced by differentiated thyrocytes, organized in closed spheres called follicles, while calcitonin is synthesized by C-cells, interspersed in between the follicles and a dense network of blood capillaries. Although adult thyroid architecture and functions have been extensively described and studied, the formation of the “angio-follicular” units, the distribution of C-cells in the parenchyma and the paracrine communications between epithelial and endothelial cells is far from being understood.This method describes the sequential steps of mouse embryonic thyroid anlagen dissection and its culture on semiporous filters or on microscopy plastic slides. Within a period of four days, this culture system faithfully recapitulates in vivo thyroid development. Indeed, (i) bilobation of the organ occurs (for e12.5 explants), (ii) thyrocytes precursors organize into follicles and polarize, (iii) thyrocytes and C-cells differentiate, and (iv) endothelial cells present in the microdissected tissue proliferate, migrate into the thyroid lobes, and closely associate with the epithelial cells, as they do in vivo.Thyroid tissues can be obtained from wild type, knockout or fluorescent transgenic embryos. Moreover, explants culture can be manipulated by addition of inhibitors, blocking antibodies, growth factors, or even cells or conditioned medium. Ex vivo development can be analyzed in real-time, or at any time of the culture by immunostaining and RT-qPCR.In conclusion, thyroid explant culture combined with downstream whole-mount or on sections imaging and gene expression profiling provides a powerful system for manipulating and studying morphogenetic and differentiation events of thyroid organogenesis.  相似文献

ABSTRACT. The population of organelles in the periphery of the contractile vacuole of Amoeba proteus has been studied throughout the cycle using proper fixation and dehydration procedures on serial semithin and thin sections. Three distinct types of perivacuolar vesicles which arise successively from one another in the course of the cycle have been described.
