The endopeptidaees (Eps) In wheat endosperm during seed germination and subsequent seedling growth were character-Izad by gradiant-polyacrylamide gel electrophoresis with gelatin copolymerized into the gel. Four cysteine Eps (EP1, EP2, EP3 and EP4) ware detected in wheat endosperm during the 7 d growth after seed imbibition. The results also showed that the activities of all of these Eps increased continuously, and EP2 first appeared and had the highest proteolytic activity among the four Eps in this experimental process. The optimum pH and temperature of all four Eps were 4.0 and 40.0℃. All Eps were completely inhibited by 25 μmol/L E-64 and had no good thermal stabilities, especially EP1. In addition, these Eps had different substrate apecificities to albumins, globulins, gliadins and glutanins; the main storage proteins of mature wheat endosperm. Among them, EP2 had the highest proteolytic activities on globulins, gliadins and glutenins, and might be the most important and specific EP with potential to be tightly correlated with seedling devalopment.  相似文献

The ovule is anatropous and bitegmic. The nuceIlar cells have disorganized except the chalazal proliferating tissue. The curved embryo sac comprises an egg apparatus and a central cell with two palar nuclei and wall ingrowths on its micropylar lateral wall. The antipodal cells disappear. Embryo development is of the Onagrad type. The filament suspensor grows to a length of 785 μm and degenerats at tarpedo embryo stage. The basal cell produces wall ingrowths on the micropylar end wall and lateral wall. The cells of mature embryo contain many globular protein bodies, 2.5–7.5 μm in diameter, composed of high concentration of protein and phytin, insoluble polysaccharide and lipid. The cells, except procambium, also contain many small starch grains. Some secretory cavities scattered in the ground tissue have liquidlike granules composed of protein, ploysacchaide and lipid. Endosperm development follows the nuclear pattern. At the late heart embryo stage, the endosperm around the embryo and the upper suspensor and the peripheral endosperm of the basal region of the U-shaped embryo sac becomes cellular. The endosperm at micropylar and chalazal ends remains free nuclear phase until the late bended cotyledon stage. Wall ingrowths at both micropylar and chalazal end wall and lateral wall of the embryo sac become more massive during endosperm development. Wall ingrowths also occur on the outer walls of the outer layer endosperm cells at both ends and lateral region of the embryo sac. When the embryo matures, many layers of chalazal endosperm ceils including 2–4 layers of transfer cells, a few of micropylar endosperm cells and 1–5 layers of peripheral endosperm cells are present. The nutrients of the embryo and endosperm at different stages of development are also discussed.  相似文献

Dad1是一个高度保守的细胞程序性死亡抑制基因。以拟南芥Dad1 (AtDad1) cDNA为模板,利用体外RT-PCR对其进行地高辛标记。以产物反义RNA作探针 (顺义RNA为对照),对Dad1在玉米种子发育过程中的时空表达图谱进行了研究。结果表明Dad1在一些衰老组织如子房壁、珠被(Fig.1, B&C)、成熟的胚乳 (Fig.1, E) 以及脱落的根冠细胞 (Fig.2 A)中表达量均较高,同时在新陈代谢活跃的组织如根尖分生组织细胞 (Fig.2, A&B)、发育的子叶(Fig.2, C&D)、雌性生殖细胞 (Fig.1 C)以及发育的胚乳细胞(Fig.1 C)中表达量也较高。结论:同其它植物中的Dad1同源基因一样,玉米中的Dad1基因在胚胎发育及胚乳PCD等多种发育过程中也可能起重要作用。  相似文献

Abstract: The embryology and seed structure of Paepalanthus sect. Actinocephalus species were studied. The embryological and structural seed characters fit well with those of the other commelinaceous families. Within the Commelinales sensu Dahlgren, Eriocaulaceae and Xyridaceae represent two embryologically close families. In Paepalanthus sect. Actinocephalus the ovule is orthotropus, bitegmic, and tenuicellate with a micropyle formed by the inner integument. The seeds are endotestal. The outer cell layer of the testa and the outer periclinal wall of the endotesta disintegrate during development. The endotegmen is tanniniferous. The outer layer of the tegmen becomes compressed and is no longer recognizable in the mature seed. The seeds are operculate.  相似文献

The aim of this work was to study the influence of storage in liquid nitrogen on the viability of seeds of the hybrid orchid Bratonia and further development of its protocorms in vitro. Seeds were frozen in ampoules by direct immersion in liquid nitrogen and stored in the cryobank for a month. The germination rates of cryopreserved and control (nonfrozen) seeds did not differ and remained as high as 100%. The protocorms derived were cultured on the agar-solidified Murashige and Skoog nutrient medium (MS), half-strength MS and Knop media and also in Morel liquid medium. During the first 45 days of culturing, protocorms derived from cryopreserved seeds grew faster than control protocorms on the MS and half-strength MS media but, at longer culturing (496 days), the size of control protocorms was significantly larger. After 639 days of culturing, there was no difference in the amount of perished, budding, and newly formed protocorms obtained from cryopreserved and control seeds, except half-strength MS medium where the number of budding protocorms in the case of cryopreserved seeds was a little greater than in the control treatment. After seed cryopreservation, the frequency of budding and newly formed protocorms was greater on the agarized MS and in liquid Morel media. Cryopreservation had little effect on the subsequent growth of protocorms in vitro. The preferable nutrient media for culturing the protocorms have been suggested.  相似文献

对金丝桃属5组9种植物的种子进行了宏观及微观形态学研究,以探讨岐山金丝桃的系统位置。结果显示,种子形态特征在本实验观察的5个组之间有明显差异。其中金丝桃组种子外形较为细长,种皮纹饰为狭长而规则的矩形网纹;黄海棠组种子稍粗,种皮纹饰为长宽近等的多边形(稀矩形)网纹;元宝草组的元宝草、贯叶连翘组的贯叶连翘以及挺茎遍地金组的云南小连翘种子均较小,元宝草种皮纹饰为负网纹,贯叶连翘种皮纹饰为相邻网纹间有间隙的近圆形网纹,云南小连翘种皮纹饰为复网纹。研究显示,岐山金丝桃的种子形态与黄海棠组最为接近,支持岐山金丝桃归于黄海棠组。  相似文献

目的:探究Mi R-935调控胃癌SGC7901细胞的增殖和浸润与Notch1基因表达的关系。方法:分别检测40例正常人胃粘膜组织与40例胃印戒细胞癌的Notch1表达情况,并分析胃印戒细胞癌组织中Notch1表达与患者年龄、性别、组织进展程度、TNM分期、有无淋巴结转移的关系;采用Mi R-935转染体外培养的SGC7901细胞系,检测Notch1的表达情况,其后采用Mi R-935抑制剂处理,通过Transwell实验检测胃癌细胞的侵袭能力,细胞划痕实验检测胃癌细胞迁移能力。结果:正常人胃粘膜组织中Notch1表达呈阴性,而胃印戒细胞癌组织中Notch1表达呈阳性;Notch1的表达与胃印戒细胞癌的TNM分期、有无淋巴结转移存在着显著的相关性;转染Mi R-935的SGC7901细胞Notch1表达明显上调,采用Mi R-935抑制剂处理后,Notch1的表达显著下降。结论:Mi R-935可能通过调控Notch1的表达调控胃癌的扩增和浸润。  相似文献

We report here on a comparative developmental profile of plant hormone cytokinins in relation to cell size, cell number and endoreduplicaUon in developing maize caryopsis of a cell wall invertase-deficient miniature1 (mn1) seed mutant and its wild type, Mn1, genotype. Both genotypes showed extremely high levels of total cytokinins during the very early stages of development, followed by a marked and genotype specific reduction. While the decrease of cytokinins in Mn1 was associated with their deactivation by 9-glucosylation, the absolute and the relative part of active cytokinin forms was higher in the mutant. During the exponential growth phase of endosperm between 6 d after pollination and 9 d after pollination, the mean cell doubling time, the absolute growth rate and the level of endoreduplication were similar in the two genotypes. However, the entire duration of growth was longer in Mnl compared with mnl, resulting in a significantly higher cell number in the Mnl endosperm. These data correlate with the previously reported peak levels of the Mn1-encoded cell wall invertase-2 (INCW2) at 12 d after pollination in the Mn1 endosperm. A model showing possible crosstalk among cytokinins, cell cycle and cell wall invertase as causal to increased cell number and sink strength of the Mn1 developing endosperm is discussed.  相似文献

EMBRYONIC FLOWER (EMF) genes are required to maintain vegetative development via repression of flower homeotic genes in Arabidopsis. Removal of EMF gene function caused plants to flower upon germination, producing abnormal and sterile flowers. The pleiotropic effect of ernfl mutation suggests its requirement for gene programs involved in diverse developmental processes. Transgenic plants harboring EMF1 promoter::glucuronidase (GUS) reporter gene were generated to investigate the temporal and spatial expression pattern of EMF1. These plants displayed differential GUS activity in vegetative and flower tissues, consistent with the role of EMF1 in regulating multiple gene programs. EMFI::GUS expression pattern in emf mutants suggests organ-specific auto-regulation. Sense- and antisense (as) EMF1 cDNA were expressed under the control of stage- and tissue-specific promoters in transgenic plants. Characterization of these transgenic plants showed that EMF1 activity is required in meristematic as well as differentiating tissues to rescue emf mutant phenotype. Temporal removal or reduction of EMF1 activity in the embryo or shoot apex of wild-type seedlings was sufficient to cause early flowering and terminal flower formation in adult plants. Such reproductive cell memory is reflected in the flower MADS-box gene activity expressed prior to flowering in these early flowering plants. However, temporal removal of EMF1 activity in flower meristem did not affect flower development. Our results are consistent with EMF1's primary role in repressing flowering in order to allow for vegetative growth.  相似文献

该研究在前期对‘吴旗黄芥’黄籽性状遗传图谱研究的基础上,利用白菜及拟南芥基因组信息,在芸薹属白菜A基因组BAC克隆KBrH105I15上设计了5对SCAR引物,在拟南芥第3染色体黄籽基因的同源区域At3g14120与At3g29615附近设计了6对IP引物,11对引物分别扩增F2群体(‘吴旗黄芥’ב武功褐芥’)的1 212个单株,开发与‘吴旗黄芥’黄籽基因更近的分子标记。结果表明:来自于白菜A基因组BAC克隆KBrH105I15上的Y12(Y12为共显性标记),以及来自于拟南芥第3染色体同源区域At3g24180的IP-6表现与‘吴旗黄芥’黄籽基因紧密连锁,其遗传距离分别为0.2和0.1cM,较之前最近的标记距离分别缩短0.3和0.2cM。这2个标记的开发对开展‘吴旗黄芥’黄籽基因的克隆奠定了基础。  相似文献

The effectiveness of a seed disperser depends on the quantity and quality of dispersal. The quality of dispersal depends in large part on factors that affect the post–dispersal fate of seeds, and yet this aspect of dispersal quality is rarely assessed. In the particular case of seed dispersal through endozoochory, the defecation pattern produced has the potential of affecting the fate of dispersed seeds and consequently, dispersal quality and effectiveness. In this study, I assessed the effects of dung presence and dung/seed densities on seed predation by rodents and secondary dispersal by dung beetles. In particular, I compared seed fates in clumped defecation patterns, as those produced by howler monkeys, with seed fates in scattered defecation patterns, as those produced by other frugivores. I also determined the prevalence of red howler monkeys (Alouatta seniculus) as seed dispersers at the plant community level in Central Amazonia by determining the number of species they dispersed in a 25–month period. I found that dung presence and amount affected rodent and dung beetle behavior. Seed predation rates were higher when dung was present, and when it was in higher densities. The same number of seeds was buried by dung beedes, in dumped versus scattered defecation patterns, but more seeds were buried when they were inside large dung–piles versus small piles. Seed density had no effect on rodent or dung beetle behavior. Results indicate that caution should be taken when categorizing an animal as a high or low quality seed disperser before carefully examining the factors that affect the fate of dispersed seeds. Red howler monkeys dispersed the seeds of 137 species during the study period, which is the highest yet reported number for an Alouatta species, and should thus be considered highly prevalent seed dispersers at the plant community level in Central Amazonian terra firme rain forests.  相似文献

ES细胞系统与基因定位致变相结合,进行基因敲除(knockout)已成为研究基因在生物体内功能的重要手段。在ES细胞系的建立、外源基因导入ES细胞、种系嵌合鼠的获得等三个重要环节中,种系嵌合鼠的获得是最关键的一环。由于ES细胞系统技术复杂、实验条件要求很高,尽管国际上已报导了上百例的基因敲除(knockout)实验,但是到目前为止,我国还无一例在国内条件下获得种系嵌合鼠的正式报道。本研究对影响种系嵌合鼠获得的两种因素(饲养层细胞、受体胚胎种类)进行了比较研究,成功地获得了种系嵌合鼠。将HM1细胞在STO或MEF培养层上培养至2133代,注射到不同小鼠的囊胚里,经过恢复培养,移植到假孕的昆明白雌鼠子宫内。由于HM1细胞来源于粟色的的129品系,而胚胎供体鼠的毛色为黑或白色,仔鼠出生一周后即可辨别是否为毛色嵌合鼠。用成年嵌合鼠与其受体胚胎相同品系的小鼠交配,进行种系嵌合鼠鉴定。曾有报导:STO培养层会导致ES细胞发生核变。我们改用MEF培养层,获得嵌合鼠的比率高达48.6%(Table1)。不同小鼠胚胎之间存在差异,C57BL/6J、ICR和昆明白三者提供的受体胚胎产生嵌合鼠的比率分别为71.4%、55%  相似文献
