The acute toxicity of lectin ML I from the toxic drug, mistletoe, was demonstrated in previous experiments. Because the reason for this extremely high toxicity is not yet clear, mice were studied histochemically at different times after treatment with various doses of ML I, ML I A or ML I B chain separately, or recombinations of ML I A and ML I B. Various plasma membrane-associated hydrolases as well as Golgi apparatus-and endoplasmic reticulum-linked hydrolases, peroxisomal and extraperoxisomal oxidases, lysosomal hydrolases, mitochondrial dehydrogenases, the cytoskeletal proteins keratin and vimentin as well as iron, glycogen and lipids were analysed in all organs and tissues of female mice. Irrespective of the dose, a clear-cut response was only observed in the liver. After ML I treatment, glycogen disappeared completely from all hepatocytes, and this effect did not depend on the ML I-concentration and exposure time. The increase in activity of Golgi-associated thiamine pyrophosphatase in hepatocytes and of non-specific alkaline phosphatase in the sinusoidal endothelial cells depended on the applied ML I concentration and the time of treatment. Doses of 600 or 900 ng ML I/kg drastically increased the phosphatase activities. These clear-cut changes of glycogen and enzyme activities were not observed after administration of the ML I B chain alone, and less so when the mice were treated only with the ML I A chain, or were treated with a recombination of ML I A and ML I B even at concentrations higher than that of ML I.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

In the oocyte two particular enzyme active zones exist depending on the state of development. In young oocytes acid phosphatase, succinate dehydrogenase and thiamine pyrophosphatase can be found only in the perinuclear zone. From the late secondary follicle on, activity of acid phosphatase, succinate dehydrogenase and non-specific esterases can be detected only in a peripheral area of cytoplasm, whereas thiamine pyrophosphatase is present in the entire ooplasm. In the cell cortex of the parasitic ciliate Ichthyophthirius multifiliis different kinds of cisternae were observed: the alveolar sacs, thick membrane cisternae and the endoplasmic reticulum. The thick membrane cisternae possess coated dilated rims and sometimes could be observed close to the endoplasmic reticulum. Using cytochemical techniques acid phosphatase, thiamine pyrophosphatase and nucleoside diphosphatase activities were detected in the thick membrane cisternae and in the alveolar sacs of trosphozoites. In the endoplasmic reticulum acid phosphatase activity was not detected and only very small amounts of thiamine pyrophosphatase and nucleoside diphosphatase reaction product were observed. After exit from the host, a reduction in acid phosphatase activity was evident in the alveolar sacs. At theront stage acid phosphatase activity is absent from these structures. However, high thiamine pyrophosphatase and nucleoside diphosphatase activities remain in the alveolar sacs during the whole life cycle. On the other hand, acid phosphatase, thiamine pyrophosphatase and nucleoside diphosphatase activities were detected in thick membrane cisternae of theronts. Based on the morphological aspects and enzymatic content the thick membrane cisternae of the cell cortex are designated as golgian-like cisternae. The cytochemical results point out a relationship between the alveolar sacs and the Golgi complex.  相似文献

Summary Electron microscopic cytochemistry was used to determine the localization of five phosphatase enzymes—glucose-6-phosphatase, inosine diphosphatase, thiamine pyrophosphatase, acid phosphatase, and adenosine triphosphatase—in control human testes. Glucose-6-phosphatase occurred in the endoplasmic reticulum and nuclear envelope of Sertoli cells, Leydig cells and primitive spermatogonia, but was not observed in more advanced spermatogenic cells. The presence of glucose-6-phosphatase activity paralleled the presence of glycogen in spermatogenic cells, i.e., both occurred in type AL and AD spermatogonia but not in type AP or B spermatogonia or in more advanced spermatogenic cells. Inosine diphosphatase activity was found in the endoplasmic reticulum, nuclear envelope, and Golgi complex of Sertoli cells and all spermatogenic cells except late spermatids. Additionally, inosine diphosphatase activity was localized at the junctions between Sertoli cells and late spermatids, but was not associated with any other plasma membrane. Thiamine pyrophosphatase reaction product was found in the Golgi bodies of Sertoli cells and in spermatogenic cells through immature spermatids. Neither inosine diphosphatase nor thiamine pyrophosphatase was observed in the Golgi bodies of spermatids during acrosomal formation. Acid phosphatase activity was found in lysosomes of spermatogonia, spermatocytes, and spermatids, in lysosomes of Leydig cells, and in lysosomes, lipofuscin bodies, and Golgi cisternae of Sertoli cells. It is thought that Sertoli lysosomes play a role in the phagocytosis of degenerating germ cells; however, the role of spermatogenic or Leydig lysosomes is unknown. Adenosine triphosphatase activity occurred at the interfaces between two spermatogonia, and between Sertoli cells and spermatogonia, but was not observed in the spaces between two Sertoli cells, two spermatocytes, two spermatids, or between Sertoli cells and spermatocytes, or between Sertoli cells and spermatids.Supported in part by a grant from the U.S. Atomic Energy Commission (AT-(40-1)-4002).  相似文献

The effects of vinblastine and colchicine on the Golgi apparatus of stomach surface mucoid and absorptive intestinal cells were compared by cytochemical analysis. The two epithelial cells were chosen because of their different specific functions in the formation of secretory granules, the production of lysosomes and the intensity of membrane traffic in the cytoplasm. For the analysis, adult mice were injected with 1 mg/100 g b.w. of vinblastine and 1 mg/100 g b.w. of colchicine. For the demonstration of cis and trans cisternae of the Golgi apparatus, prolonged osmification, thiamine pyrophosphatase and acid phosphatase activity identification were applied. After treatment with vinblastine or colchicine, polarity of stacks in the Golgi apparatus of surface mucoid cells is preserved although the number of cisternae with thiamine pyrophosphatase or acid phosphatase activity decreases. However, the Golgi apparatus of intestinal absorptive cells completely disintegrates and only a few separated cis or trans cisternae can be identified. The main effect seems to be a reduction of vesicles which can be cytochemically identified as parts of the Golgi apparatus and an accumulation of vesicles which probably originate from budding ER. Communication between the ER and the Golgi apparatus seems to be interrupted.  相似文献

Summary S.W.R. adult male mice received one injection of colchicine and were killed at various time-intervals from 4 to 24 hours. Ultrastructural morphology and cytochemical activity of acid phosphatase, arylsulfatase and thiamine pyrophosphatase were recorded in the duodenal epithelium. Extensive damage was observed in the undifferentiated cells with formation of large heterophagosomes and in the absorptive cells with apparition of cytolysomes. The activity of the acid phosphatase and thiamine pyrophosphatase decreased after 6 hours of treatment. Moreover, the migration of the acid phosphatase content of the Golgi vesicles was restraint. The activity of acid hydrolases in skeletal muscles of normal rats and of rats after subcutaneous administration of dimethyl-para-phenylene diamine (DPPD) was studied with a combined histochemical and biochemical investigation. In this communication the histochemical findings are presented. After 4 days of DPPD treatment, coagulation necrosis, fragmentation and disintegration of fibres were seen in the muscles. An inflammatory infiltrate was seen between the muscle fibres. These pathological changes reached maximum intensity after 7 to 9 days. After 11 days the changes became less, despite continued treatment with DPPD. From the histochemical findings it appeared that the activity of acid phosphatase, beta-glucuronidase and E600 resistant non-specific esterase was increased in both a granular and a diffuse pattern in the skeletal muscles of the DPPD rats. The increase in activity of leucine aminopeptidase was much less pronounced and was mainly granular. The increase in the activity of acid hydrolases ran parallel to the severity of the pathological changes and reached a maximum after 7 to 9 days of DPPD treatment. The statistical calculations of the histochemical findings revealed that the increased activity of one acid hydrolase was significantly paralleled by an increased activity of a second hydrolase. There was a moderate probability that the activity of all other histochemically studied acid hydrolases, with the exception of leucine aminopeptidase, was increased. There was no difference in activity and localization of the acid hydrolases studied in aerobic type I and anaerobic type II fibres. The localization of acid phosphatase and beta-glucuronidase activity in muscle fibres and in inflammatory infiltrate mostly coincided. In cases where these enzymes were localized both centrally and in the subsarcolemnal areas of the muscle fibres, the activity of E600 resistant non-specific esterase was usually, and the activity of leucine aminopeptidase was exclusively located in the subsarcolemnal areas. All of the acid hydrolases examined were found to be present in the inflammatory exudate and in the connective tissue.  相似文献

Summary Electron microscopic cytochemical studies on the rat choroid plexus epithelium have revealed enzymatic sites for the activities of acid phosphatase, glucose-6-phosphatase and thiamine pyrophosphatase on different organelles. Only the activity of acid phosphatase has been previously described. Acid phosphatase, glucose-6-phosphatase and thiamine pyrophosphatase were respectively situated mainly in the lysosomes, in the endoplasmic reticulum and nuclear envelope, and in the Golgi complex. These three enzymes can thus be considered as marker enzymes for their respective organelles in the choroid plexus epithelial cells as well as in other tissue cells. The possible function of these enzymes in the choroid plexus epithelial cells is also briefly discussed.  相似文献

Summary The central nervous systems of web-building spiders (Araneidae, Agelenidae) and hunting spiders (Lycosidase, Salticidae) were tested for non-specific and specific phosphatases. Acid phosphatase exhibited weakly to moderately positive reactions in the neuronal cell bodies and in the neuropile fibre mass of all species investigated. Alkaline phosphatase could only be demonstrated in the external and internal neural lamellae of the brain and ventral cord of several specimens of the araneid species investigated. Tests for thiamine pyrophosphatase were negative with both the lead and calciumcobalt methods. Distinctive positive reactions for adenosine triphosphatase were visible in the nervous system of all the species used, being especially strong in the optic ganglia of the hunting spiders. The demonstration of adenosine triphosphatase was only possible when applying the calcium-cobalt method after Padykula and Herman, while the lead method after Wachstein and Meisel did not produce any staining reaction at all. Controls of the histochemical reaction showed that the enzyme was activated by Ca2+ and inhibited by sulphydryl destroying reagents (e.g. PCMB), but was insensitive to ouabain. It could be probably classified as a mitochondrial proton-translocating adenosine triphosphatase.  相似文献

The properties of thiamine pyrophosphatase in the Golgi apparatus of rat liver were studied. Thiamine pyrophosphatase in an extract of the Golgi apparatus was separated into 6 bands of between pH 5.4 and 6.3 by isoelectric focusing on polyacrylamide gel. On the gels all these subforms catalyzed the hydrolyses of GDP, IDP, UDP, and CDP as well as that of thiamine pyrophosphate. The characteristics resembled those of Type B nucleoside diphosphatase of rat brain, though the enzyme did not have 3 subforms of Type B nucleoside diphosphatase in the higher pH region on isoelectric focusing. Thiamine pyrophosphatase of the Golgi apparatus was separated from microsomal nucleoside diphosphatase by DEAE-cellulose column chromatography. The properties of the enzyme were quite similar to those of Type B nucleoside diphosphatase with respect to its substrate specificity, optimum pH for activity, and inhibition by ATP. These findings suggest that thiamine pyrophosphatase in the Golgi apparatus is different from microsomal nucleoside diphosphatase and that it might be basically the same enzyme as Type B nucleoside diphosphatase except for different extents of modification.  相似文献

Cytochemical distributions of acid phosphatase, thiamine pyrophosphatase, and ATP-diphosphohydrolase activities have been examined on thin sections of rat pancreas and on isolated zymogen-granule membranes. Acid phosphatase was found in the rigid lamellae separated from the Golgi stacked cisternae, in condensing vacuoles, and in the trans-saccules of Golgi apparatus; it was not detected in purified zymogen-granule membranes. Thiamine pyrophosphatase was detected in trans-saccules of the Golgi apparatus, in purified zymogen-granule membranes, and in the plasmalemma of the acinar cell. It was absent in condensing vacuoles. The ATP-diphosphohydrolase activity has a distribution similar to thiamine pyrophosphatase. These observations illustrate the similarity between the trans-saccules of the Golgi apparatus and the membrane of mature zymogen granules and the disparity between the latter membrane and the membrane of the condensing vacuole. They suggest that the condensing vacuole might not be the immediate precursor of the zymogen granule as commonly assumed. An alternative possibility would be that condensing vacuoles would fuse with the trans-saccule (transition) of the Golgi apparatus which in turn would form mature zymogen granules.  相似文献

Summary In motoneurons of the spinal cord of mice, the structure of lysosomes and Golgi zone was investigated using light and electron microscopy, under the influence of the cornpulsatory motor activity and tetanus toxine. In the motoneurons the activity of acid phosphatase according Gomori and activity of thiamine pyrophosphatase according Novikoff and Goldfischer were estimated.In animals after compulsatory motor activity or tetanus toxine the enlargement of Golgi zone and the increase of lysosomes activity was shown. The results of experiments, confirm the known Novikoff theory reffering to a close connections between the morphological and functional interrelation between the lysosomes and the Golgi zone.  相似文献

Synopsis The fine structure and cytochemistry of the intestinal epithelial cell of the fowl have been investigated. The fine structure of the mature absorptive cell of the fowl duodenum was very similar to that described for man and other mammals. Minor differences were the thinner microvillous glycocalyx, the unusual length of the cells and their microvilli, and the wide distribution of lysosomal bodies. The membrane-associated enzymes alkaline phosphatase, ATPase (pH 7.2) and leucine naphthylamidase were mainly associated with the brush border; this organelle also gave positive reactions for mucopolysaccharides and phospholipids. No enzyme activities were found in the terminal web.The distribution of lysosomes between the terminal web and the Golgi apparatus was correlated with the granular localization of the lysosomal enzymes acid phosphatase, -glucuronidase and non-specific esterase. The mitochondrial enzyme succinate dehydrogenase was seen to be localized in rod-like dots which marked the distribution of mitochondria in the absorptive cell. The localization of mitochondrial ATPase (pH 9.4) was not clearly demonstrated because of diffusion artifacts. The region of the Golgi apparatus gave a strong reaction for thiamine pyrophosphatase, together with weak reactions for acid and alkaline phosphatases after extensive overincubation.The endoplasmic reticulum-associated enzymes glucose-6-phosphatase and nonspecific esterase were distributed throughout the absorptive cell, with a maximum activity apical to the Golgi apparatus. Additionally, the jejunal absorptive cells showed endoplasmic reticulum-as well as lysosomal-associated -glucuronidase.  相似文献

Summary A range of enzymatic activities in cervical mucus-secreting, ciliated and subcolumnar basal cells were assessed using light and electron microscopic cytochemical techniques. Enzymes detected in all three cell types were those of the tricarboxylic acid cycle, pentose-phosphate and glycolytic pathways, other mitochondrial associated enzymes (NADH and NADPH dehydrogenase), acid phosphatase and non-specific esterase. Mucus-secreting and ciliated cells exhibited thiamine pyrophosphatase and 5 nucleotidase activities while leucine aminopeptidase was most convincingly demonstrated in mucus-secreting cells. Alkaline phosphatase, on the other hand, was detected only in mucus-secreting and subcolumnar basal cells. The profile of enzymatic activities in subcolumnar basal cells closely resembles that of mature lining cells and further supports the hypothesis that these cells differentiate into functioning columnar cells.  相似文献

Summary New light microscopic visualization methods were developed for the histochemical detection of non-specific alkaline and acid phosphatase, Mg-, Ca-and Na, K-dependent adenosine triphosphatase, myosin adenosine triphosphatase, glucose-6-phosphatase, 5-nucleotidase and thiamine pyrophosphatase with cerium ions as trapping agents in cryostat and plastic sections. The techniques are based on the conversion of cerium phosphate into cerium perhydroxide by H2O2 which decomposes at 55°–60° C into cerium hydroxide and oxygen radicals. These radicals are able to oxidize diaminobenzidine (DAB) to DAB brown. Addition of nickel ions to the DAB-H2O2 mixture generates bluish-black stained nickel-DAB complexes. Compared with the classical metal precipitation, azo, azoindoxyl and tetrazolium procedures the H2O2-DAB and especially the H2O2-DAB-nickel methods provided identical or superior results in catalytic phosphatase histochemistry and immunohistochemistry when using non-specific alkaline phosphatase as the enzyme label.  相似文献

In the rat adenohypophyseal mammotrophs and somatotrophs, activities of acid phosphatase, thiamine pyrophosphatase and aryl sulphatase were defined after the incubation of the pituitary gland in the medium containing 1.65 M sucrose. Following 60--120 minutes of the incubation the above enzymes were found, in addition to their usual sites, in all cisternae of the Golgi apparatus and the rough endoplasmic reticulum as well as within the perinuclear cisternae. The mechanism of the above alterations is discussed, and a suggestion is put forward that the emergence of enzymatic activities in the above structures may reflect their biogenetic pathways in the cell.  相似文献
