Abstract— An immunoprecipitation technique has been employed to measure the rate of synthesis of tyrosine hydroxylase in organ cultures of rat superior cervical ganglia and the effect of nerve growth factor on that rate. Ganglia which have been maintained in culture for 16 h without nerve growth factor synthesize tyrosine hydroxylase; the hydroxylase comprises approx 0.2% of the newly synthesized soluble protein. While the total amount of tyrosine hydroxylase synthesized de novo increases in the presence of physiological levels of nerve growth factor, the differential rate of tyrosine hydroxylase synthesis is essentially unchanged. At higher levels of nerve growth factor (3–10 μg/ml) there is a small increase in the differential rate of tyrosine hydroxylase synthesis. The major action of nerve growth factor appears to be on the survival of the tissue, but a small effect on the induction of tyrosine hydroxylase is evident at high levels of nerve growth factor.  相似文献

Abstract— Phenylethanolamine N -methyltransferase (PNMT) and adrenaline (A) have been studied in organ cultures of neonatal rat sympathetic ganglia. Organ culture for 2 days without added nerve growth factor (NGF) caused a fall in noradrenaline (NA) and PNMT contents but there was no change in dopamine (DA) or A contents compared to controls. However, in the presence of dexamethasone, there was a marked increase in both PNMT activity and A content, but no change in NA or DA content. Addition of NGF to cultures stimulated with dexamethasone caused no further significant change in PNMT activity or A content, whereas both NA and DA were increased. Prolonged culture without NGF, in the presence of dexamethasone resulted in reductions in both NA and DA content, but the high levels of PNMT activity and A content were sustained. The results are consistent with the hypothesis that both PNMT and A are not contained in the noradrenergic cell bodies but are located chiefly within the small intensely fluorescent cells in sympathetic ganglia.  相似文献

The activity of RNA polymerase in the superior cervical ganglia of the neonatal rat has been studied. The characteristics of the activity with respect to enzyme concentration, temperature, time, and ion concentrations are presented. Ionic conditions of the assay favoring polymerase I and polymerase II were each studied in the presence of specific inhibitors of DNA-dependent RNA synthesis. The Km for GTP under either polymerase I or polymerase II conditions was found to be 10?5M, and the relative amounts of poly(A)-containing RNA synthesized under both conditions was 4-5%. A decrease in the activity of RNA polymerase with age has been observed. The time course of increased activity following treatment of the animals in vivo or treatment of the isolated ganglia in organ culture with nerve growth factor is presented. The increase in activity observed after the administration of nerve growth factor is discussed in terms of possible mechanisms of action of nerve growth factor in this tissue.  相似文献

—The period during which trans-synaptic stimulation is required by the rat superior cervical ganglion for induction of tyrosine hydroxylase by reserpine has been studied. Ganglia were decentralized on one side at various times before or after an injection of reserpine. The tyrosine hydroxylase activity of the denervated and control ganglia was assayed 72 h after drug treatment. When decentralization was performed 8 h after an injection of reserpine the increase in tyrosine hydroxylase activity was blocked in the denervated ganglia. Decentralization 12 h after reserpine treatment or later had no effect on the enzyme induction. The actual increase in tyrosine hydroxylase activity occurred between 24 and 48 h after injection of reserpine.  相似文献

Small amounts of nerve growth factor (NGF) were present in the superior cervical ganglion and the iris of the rat. The observations that NGF content in each of the tissues was depleted during organ culture and that more NGF appeared in the media than was originally present in the tissues indicated that synthesis or activation of NGF had occurred in organ culture. Antibody to NGF or the depletion of endogenous NGF retarded growth of new sympathetic axons into irides in organ culture. Exogenously added NGF appeared to enhance the initiation of axonal sprouting and the rate of the ramification of nerve fibres.  相似文献

Surgical decentralization of the superior cervical ganglion (SCG) in rats and mice led to a fall in ganglionic tyrosine hydroxylase (T-OH) activity, and a loss of more than 90 per cent of the preganglionic neurone marker, choline acetyl transferase. T-OH activity was reduced by more than 50 per cent in mice SCG ten days after surgery, but fell by only 25 per cent in rat SCG after 21 days. The surgical procedure did not cause obvious histo-logical damage or loss of SCG cells in either species. Both T-OH and choline acetyl transferase activities in rat and mouse SCG recovered to normal three months after surgery. Reserpine treatment was more effective in rats in causing increased ganglionic T-OH activity than in mice. Neither decentralization nor reserpine treatment caused any changes in DOPA-decarboxylase or monoamine oxidase activities in rat SCG. These results demonstrate that T-OH activity in SCG is subject to trans-synaptic regulation in both rats and mice; this regulation does not apply to DOPA-decarboxylase or monoamine oxidase. Differences in basal sympathetic tone may explain the different results obtained in mice and rats.  相似文献

Abstract— Nerve-growth factor (NGF) or dexamethasone administered to newborn rats result in increased levels of phenylethanolamine-N-methyl transferase (PNMT) activity in the superior cervical ganglia. Treatment with dexamethasone, but not NGF, also results in increased levels of epinephrine which parallel the increased PNMT in the ganglia. Dexamethasone slows the decrease in PNMT levels after cessation of NGF treatment, suggesting that dexamethasone decreases the rate of PNMT destruction. Examination of the ganglia by histofluorescence indicates that NGF increases the bundles of fibres while dexamethasone increases the number of small intensely fluorescent cells.  相似文献

Abstract— Tyrosine hydroxylase (TH) activity was measured in the carotid body. superior cervical ganglion and adrenal glands of the rat under normal conditions and at 48 h following exposure of the animals for 1-3 h in a low O2 atmosphere. Basal TH levels were 5-6 nmol/h/mg tissue for both the carotid body and the ganglion. Forty-eight hours after hypoxia, there was an increase in enzyme activity in both tissues which paralleled the severity of the hypoxia but was greater in the carotid body than the superior cervical ganglion. Thus, following exposure to 5% O2 in N2 for two 30-min periods (20-min interim), TH activity had increased by 50% in the carotid body and 33% in the ganglion; after exposure to 10% O2 in N2 for 3 h (continuous), TH levels were increased by 37% in the carotid body and 12% in the ganglion. In the adrenal gland, basal TH activity was 3.42 ± 1.87 nmol/h/mg tissue, and this value was unchanged following either level of hypoxia.  相似文献

Abstract— After previous studies had shown that nerve growth factor produces a very similar change in the enzyme pattern of adrenergic neurons as does an increased activity of the preganglionic cholinergic nerves, the present experiments revealed that the nerve growth factor-mediated selective induction of TH and DBH is enhanced by glucocorticoids in a way similar to that mediated by acetylcholine via nicotinic receptors. Corticosterone (5 μM) produced not only an increase in the maximal response to NGF but shifted the concentration response curve of TH to NGF to the left. The potentiation effect was shown to be specific for glucocorticoids, since other steroid hormones like testosterone, β-estradiol and progesterone had no effect. Moreover, the glucocorticoid effect could be antagonized by cortexolone, suggesting an effect via glucocorticoid receptors. In addition to the potentiation of the nerve growth factor-mediated enzyme induction, glucocorticoids reduced the exposure time to NGF, necessary to initiate maximal TH induction, from 4 h to 10 min. The glucocorticoid potentiation of NGF-mediated specific enzyme induction is discussed in relation to the site and mechanism of action of NGF.  相似文献

Daily injections of 100 μg/kg of d -lysergic acid diethylamide (LSD) for 14 days produced a significant decrease in the dopamine level in rat brain corpus striatum which was still apparent 15 days after the last LSD treatment. Further LSD injections did not change the amount of dopamine depletion. In cerebral cortex, 14 days of LSD injections produced a significant decrease in the norepinephrine level and a significant increase in tyrosine hydroxylase activity. The elevated tyrosine hydroxylase activity was still present 15 days after the final LSD injection but only in those animals receiving daily vehicle injections during this period. Pre-treatment of rats with daily saline injections for 2 weeks before the 2 week period of LSD treatment prevented both the reduced norepinephrine content and elevated tyrosine hydroxylase activity usually found 24 h after the last LSD injection.  相似文献

Abstract— The effect of the Nerve Growth Factor on the polysomal pattern of chick embryo sensory ganglia cultured in vitro has been studied. A two-fold stimulation of polysome formation in NGF treated ganglia with respect to the control has been found after 3 to 15 hours of incubation. Double labelling experiments performed in order to detect qualitative changes of the polysomal pattern showed no differences between NGF treated and untreated ganglia after a 3-hour incubation. Slight but significant differences in the poly-somal-monomer ratio (higher in the NGF treated ganglia) were found only for longer incubation time. Similar proportions of free and membrane bound polysomes have been found in NGF-treated and control ganglia.  相似文献

The effect of a single systemic injection of reserpine on tyrosine hydroxylase activity in the locus coeruleus, cerebellum, hypothalamus, and hippocampus was examined. Increases in enzyme activity were seen in all four brain areas; the time-course of the changes, however, was different in each case. In the locus coeruleus the maximum change in enzyme activity was seen 3 days after drug administration; in the cerebellum, 7-11 days; in the hypothalamus, 8-11 days; and in the hippocampus, 21 days. Since tyrosine hydroxylase in the cerebellum and hippocampus is present in terminals of neurons whose cell bodies are located in the locus coeruleus, the delayed increase in enzyme activity in cerebellum and hippocampus probably depends upon the slow rate of transport of TH molecules in these neurons.  相似文献

Myelinating organ cultures of rat cerebellum were maintained in vitro for up to 130 days. Extensive myelination took place between 7 DIV (days in vitro) and 28 DIV. Centrifugation of a crude culture myelin fraction on a discontinuous gradient yielded three layers termed light myelin, heavy myelin and membrane fraction, which exhibited an ultrastructure virtually identical to that of comparable layers prepared from surviving littermates. However, culture myelin layers showed a gross deficiency of galactolipids with a relative increase in phospholipids. The 2,3′-cyclic nucleoside-monophosphate phosphodiesterase (CNP) activity was decreased in the culture myelin layers, but not to an extent comparable to the cerebroside deficiency. A form of “slow myelin maturation” takes place in vitro with both myelin cerebrosides and sulphatides increasing in cultures older than 60 DIV. The results indicate that CNS myelination comprises at least two phases, and that the second phase involving galactolipid enrichment of myelin can, under experimental conditions, be partly uncoupled from the first phase without affecting the morphology or ultrastructure of the sheaths.  相似文献

—A method is described for the preparation of relatively pure cholinergic synaptosomes from bovine cervical sympathetic ganglia. After dispersion of the tissue by incubation with collagenase, differential centrifugation yielded a crude synaptosomal fraction (P2), which was further purified by centrifugation on sucrose or Ficoll-sucrose density gradients. Assay of acetylcholine and choline acetyltransferase, and electron microscopy confirmed that presynaptic nerve endings survived the incubation procedure, appeared in the P2 fraction and thereafter sedimented to a density of approx. 1.13 in sucrose and 1.06 in Ficoll-sucrose. The washed P2 fraction had an acetylcholine content of 2.3 nmol/mg of protein, which is at least three times higher than the corresponding fraction from brain.  相似文献

本研究以成人正中神经切割伤后2~3个月的神经干为材料,冰冻切片,用免疫双重染色技术显示了神经生长因子与睫状节神经营养(诱向)因子在再生的周围神经组织中的表达与分布。神经生长因子选用APAAP法.其阳性产物呈红色;睫状节神经营养(诱向)因子选用ABC系统,4氯-1-萘酚显色,阳性产物为褐色。光镜下观察:神经生长因子的阳性反应产物出现在正中神经切割伤后再生的神经纤维中,高倍镜下可见其阳性产物分布在轴索,而在雪旺氏细胞中没能见到呈红色的阳性反应产物;睫状节神经营养(诱向)因子分布在一些细胞体积大、核大呈增生活跃状态的雪旺氏细胞中。红与褐双色反应产物色调清晰,效果较好。研究结果提示:睫状节神经营养(诱向)因子与神经生长因子在人周围神经再生过程中起着十分重要的作用。  相似文献

Abstract— The development of tyrosine hydroxylase (tyrosine 3-hydroxylase, EC 1.14.3.a) activity has been examined in whole rat brain and in various regions and subcellular fractions thereof. The specific activity of tyrosine hydroxylase increased almost 15-fold from 15 days of gestation to adulthood. With maturation, those regions of the brain that contain only terminals of the catecholaminergic neurons showed the greatest increases in enzyme activity. There was a shift in the subcellular distribution of tyrosine hydroxylase from the soluble fraction in the fetal brain to the synaptosomal fraction in the adult brain. Tyrosine hydroxylase, dopamine hydroxylase (EC 1.14.2.1) and the specific uptake mechanism for norepinephrine appear to develop in a coordinated fashion.  相似文献
