The role of mycorrhizal infection in the resistance of Vaccinium macrocarpon to manganese was investigated in perlite culture containing nutrient solution amended with Mn at 0, 250, 500 or 1000 g/ml. Shoot and root dry weights of the mycorrhizal plants were higher than nonmycorrhizal plants. The mycorrhizal plants produced significantly longer main roots than the nonmycorrhizal plants. Differences between shoot and root Mn concentrations of mycorrhizal and nonmycorrhizal plants arose by reduction of Mn in the leaves of mycorrhizal plants and a corresponding increase in root tissues.  相似文献

The majority of cultivated cranberry varieties were selected from native populations in the 1800s and early 1900s from sites in Massachusetts, New Jersey, and Wisconsin. Since their initial selections 100–150 years ago, varietal identities have become increasingly confused; primarily the result of there being a paucity of qualitative markers to effectively distinguish among varieties. Random amplified polymorphic DNA (RAPD) technology has the potential for allowing a more definitive classification of varieties and was used in this study to characterize 22 cranberry varieties. Twenty-two decamer primers amplified 162 scorable DNA fragments, of which 66 (41%) were polymorphic. On the basis of these 66 silver-stained RAPDs (ssRAPDs), 17 unique profiles were identified rather than the expected 22. Fourteen varieties had unique ssRAPD profiles, while the remaining 8 were represented by 3 ssRAPD profiles. Permuational analyses of the data suggest that the observed ssRAPD profile duplications are examples of varietal misclassification. Further analyses identified 2 ssRAPD markers that were found only in Eastern varieties (from Mass. and N. J.) and not in Wisconsin varieties. With varieties differing on average by 22 bands, ssRAPDs are shown to be effective in varietal identification and the assessment of genetic diversity in cranberry.New Jersey Agricultural Experiment Station, Publ No. D-12163-8-93, Supported by State funds.  相似文献

Despite the ubiquitous presence of ericoid mycorrhizal (ERM) fungi in cranberry (Vaccinium macrocarpon), no prior studies have examined the effect of ERM colonization on NO(3)(-) influx kinetics. Here, (15)NO(3)(-) influx was measured in nonmycorrhizal and mycorrhizal cranberry in hydroponics. Mycorrhizal cranberry were inoculated with the ERM fungus Rhizoscyphus (syn. Hymenoscyphus) ericae. (15)NO(3)(-) influx by R. ericae in solution culture was also measured. Rhizoscyphus ericae NO(3)(-) influx kinetics were linear when mycelium was exposed for 24 h to 3.8 mm NH(4)(+), and saturable when pretreated with 3.8 mm NO(3)(-), 50 microm NO(3)(-), or 50 microm NH(4)(+). Both low-N pretreatments induced greater NO(3)(-) influx than either of the high-N pretreatments. Nonmycorrhizal cranberry exhibited linear NO(3)(-) influx kinetics. By contrast, mycorrhizal cranberry had saturable NO(3)(-) influx kinetics, with c. eightfold greater NO(3)(-) influx than nonmycorrhizal cranberry at NO(3)(-) concentrations from 20 microm to 2 mm. There was no influence of pretreatments on cranberry NO(3)(-) influx kinetics, regardless of mycorrhizal status. Inoculation with R. ericae increased the capacity of cranberry to utilize NO(3)(-)-N. This finding is significant both for understanding the potential nutrient niche breadth of cranberry and for management of cultivated cranberry when irrigation water sources contain nitrate.  相似文献

Patterns of resource allocation to growth, current reproduction, and potential future reproduction were quantified in six genetically distinct cultivars ofVaccinium macrocarpon. For all cultivars (genotypes), vegetative size is positively correlated with some measures of current reproduction (fruit and flower number) but negatively correlated with others (seed number per fruit, seed weight per fruit). Vegetative growth in the current year is significantly related to the production of reproductive terminal buds, a measure of the potential for reproduction in the following year. Stems with low levels of current reproduction — lower flower number, fruit number, and seed weight — were more likely to form reproductive terminal buds than stems with higher levels of current reproduction. Individual genotypes differed significantly for vegetative size, fruit number, fruit weight, seed number, and seed weight, as well as for the frequency of fruiting stems and reproductive terminal buds produced. Genotypes were segregated in principal component space, indicating overall differences between them in allocation to the suite of variables measured. These results indicate the possibility of fitness differences among cultivars due to genetically determined allocation strategy, which has implications for fitness differences among genotypes within natural populations.  相似文献

Summary Iron tolerance of rice (Oryza sativa L.) was investigated using an oxygen depleted hydroculture system. Treatment with high concentrations of Fe2+ induced yellowing and bronzing symptoms as well as iron coatings at the root surface. Root and shoot growth were inhibited by increasing iron concentration in the medium. All symptoms were more pronounced in an iron sensitive cultivar (IR 64) compared to an iron tolerant one (IR 9764-45-2). Superoxide dismutase and peroxidase activity of root extracts of IR 97 were about twice that of IR 64 in untreated control plants. No significant increase of peroxidase activity was detected with increasing iron concentration in the medium. The interaction between Glomus intraradices and the root-lesion nematode Pratylenchus vulnus was studied on micropropagated BA-29 quince rootstock during one growing season. Inoculation with G. intraradices significantly increased growth of plants in low P soil and was more effective than P fertilization at increasing top-plant development. In the presence of the nematode, mycorrhizal plants achieved higher values in all growth parameters measured. P. vulnus caused a significant decrease in the percentage of root length colonized by G. intraradices and fewer internal vesicles were formed within the host roots. Enhanced root mass production accounted for the twofold increase in final nematode population recovered from plants with combined inoculations of pathogen and symbiont. Low levels were found of Al, Fe, Mn and Zn in nonmycorrhizal nematode-infected plants in low P soil. G. intraradices-inoculated plants reached the highest foliar levels of N, Ca, Mg, Mn, Cu and Zn. Mycorrhizal plants infected with P. vulnus maintained normal to high levels of Mn, Cu, and Zn. Inoculation with G. intraradices favours quince growth and confers protection against P. vulnus by improving plant nutrition.  相似文献

Ericoid mycorrhizal fungi are symbiotically associated with the roots of members of the Ericaceae which include genera such as Calluna, Epacris, Erica, Rhododendron and Vaccinium. These ericoid mycorrhizal associations have adapted to a broad range of habitats, from mor humus soils of the northern hemisphere to sandy soils occurring in the southern hemisphere. They also play an important part in enabling plants like Calluna vulgaris (L.) Hull in the northern hemisphere to colonize mine spoils which are inhospitable environments of toxic waste for growth of most plants. The mechanisms of utilizing complex forms of nitrogen and phosphorus and providing protection against toxic metals are described. These mechanisms carried out by ericoid mycorrhizal associations enable host plants to establish in diverse habitats.  相似文献

In high-nutrient low-chlorophyll areas, bacterial degradation of organic matter may be iron-limited. The response of heterotrophic bacteria to Fe addition may be directly controlled by Fe availability and/or indirectly controlled through the effect of enhanced phytoplankton productivity and the subsequent supply of organic matter suitable for bacteria. In the present study, the role of Fe on bacterial carbon degradation was investigated through regrowth experiments by monitoring bacterial response to organic substrates derived from Phaeocystis antarctica cultures set up in <1 nM Fe (LFe) and in Fe-amended (HFe) Antarctic seawater. Results showed an impact of Fe addition on the morphotype dominance (colonies vs. single cells) of P. antarctica and on the quality of Phaeocystis-derived organic matter. Fe addition leaded to a decrease of C/N ratio of Phaeocystis material. The bacterial community composition was modified as observed from denaturing gradient gel electrophoresis (DGGE) profiles in LFe as compared to HFe bioassays. The percentage of active bacteria as well as their specific metabolic activities (ectoenzymatic hydrolysis, growth rates and bacterial growth efficiency) were enhanced in HFe bioassays. As a consequence, the lability of Phaeocystis-derived organic matter was altered, i.e., after seven days more than 90% was degraded in HFe and only 9% (dissolved) and 55% (total) organic carbon were degraded in LFe bioassays. By inducing increased bacterial degradation and preventing the accumulation of dissolved organic carbon, the positive effect of Fe supply on the carbon biological pump may partly be counteracted.  相似文献

Nitrosomonas europaea, as an ammonia-oxidizing bacterium, has a high Fe requirement and has 90 genes dedicated to Fe acquisition. Under Fe-limiting conditions (0.2 μM Fe), N. europaea was able to assimilate up to 70% of the available Fe in the medium even though it is unable to produce siderophores. Addition of exogenous siderophores to Fe-limited medium increased growth (final cell mass). Fe-limited cells had lower heme and cellular Fe contents, reduced membrane layers, and lower NH3- and NH2OH-dependent O2 consumption activities than Fe-replete cells. Fe acquisition-related proteins, such as a number of TonB-dependent Fe-siderophore receptors for ferrichrome and enterobactin and diffusion protein OmpC, were expressed to higher levels under Fe limitation, providing biochemical evidence for adaptation of N. europaea to Fe-limited conditions.  相似文献

Fungi were isolated from young, serial-washed roots of Calluna sampled from a Danish heathland, Hjelm Hede. Of the 626 isolates, those that were dark, sterile and septate were divided into 13 morphological groups based on their appearance in culture on malt agar. Mycorrhizal synthesis in vitro showed that several groups formed typical ericoid mycorrhiza with seedlings of Calluna; these ericoid mycorrhizal fungi were morphologically similar to Hymenoscyphus ericae. The identities of the other dark, septate fungi are uncertain. Oidiodendron spp. were isolated in a very low frequency; these fungi also formed typical ericoid mycorrhiza. The Calluna root system on Hjelm Hede demonstrated a high morphological diversity among the associated dark, septate fungi suggesting that more than one fungus could coexist in the same host root system.  相似文献

Although boron (B) is a micronutrient essential for the growth of vascular plants, it reduces growth and seed yield when present in excessive amounts. A hydroponic assay of nineteen Brassica rapa genotypes resulted in the identification of two tolerant genotypes, WWY Sarson and Local at a range of boron concentrations (15–165 μM). The most tolerant and sensitive genotypes were assessed for shoot boron concentrations in a soil assay with 4, 29 and 54 mg B kg−1 soil. The soil assay confirmed the results of the hydroponic screening. Shoot boron uptake was at least three times lower and shoot boron concentrations about 10 times lower in the tolerant than sensitive genotypes, indicating that boron tolerance involved boron exclusion from the shoot.  相似文献

Anthocyanin (Acy) contents and GST activities of bilberry (Vaccinium myrtillus L.) were investigated in two experiments conducted in June (Exp. I: active growth) and in August September (Exp. II: beginning of frost hardening) in Northern Finland (65 degrees N). Bilberry plants were subjected to +2 degrees C and +18 degrees C in Exp. I or +5/0 degrees C (day/night) and +18/+13 degrees C (day/night) in Exp. II. GST activities were assessed using either 1-chloro-2,4-dinitrobenzene (CDNB) or trans-cinnamic acid (tCA) as substrates. We found temperature to have no effect on Acy during either active growth or frost hardening. Acy increased several-fold from active growth to the beginning of frost hardening, but no increment was observed during the development of frost hardening. This suggests a role of Acy in photoprotection at low temperatures rather than their direct involvement in the development of freezing tolerance. The lack of response of GST activity to frost hardening and to temperature in autumn may indicate an indirect role of GSTs in frost hardening as protective enzymes. GST activity was the same with the two substrates studied (CDNB, tCA), supporting the assumption that GSTs could catalyze reactions with endogenous phenylpropanoids.  相似文献

Phosphorus is the major nutrient limiting plant growth in a Costa Rican silvopastoral system located on an acid, high P-retaining, volcanic soil. We investigated plant responsiveness to vesicular-arbuscular mycorrhizal (VAM) inoculation using the leguminous tree species Erythrina berteroana Urban, and the two dominant grass species Paspalum conjugatum Berg and Homolepsis aturensis Chase of this silvopastoral system. We grew grass seedlings in the greenhouse for 15 weeks in a methyl bromide-sterilized study soil to which either mixed-species VAM inoculum (Theobroma cacao feeder roots) or autoclave-sterilized cacao roots (non-inoculated control) were added. E. berteroana was grown from both seedlings and vegetative stakes (40 cm long) for 30 and 19 weeks, respectively. Upon harvest, we measured above and below ground biomass, N and P content, rootshoot ratio, legume nodulation, and VAM infection levels. The total above-ground and root biomass of mycorrhizae-inoculated P. conjugatum seedlings were 2.5 and 2.8 times greater than those of noninoculated seedlings. In contrast, VAM-inoculated seedlings of H. aturensis produced 8.4 and 5.9 times more total above-ground and root mass than noninoculated seedlings. Mycorrhizae-inoculated E. berteroana seedlings produced 10.6 times greater shoot biomass for inoculated versus noninoculated seedlings, while E. berteroana vegetative stakes exhibited a negative growth response to VAM inoculation (an approximately 16% decrease in shoot biomass for VAM-inoculated cuttings). The difference in responsiveness between Erythrina growth forms is hypothesized to reflect the cost-benefit relationship between plant host and fungal symbiont for energy and nutrient reserves.  相似文献

Very little is known about the capacity of soil chytrids to withstand freezing in the field. Tolerance to freezing was tested in 21 chytrids isolated from cropping and undisturbed soils in temperate Australia. Samples of thalli grown on peptone–yeast–glucose (PYG) agar were incubated for seven days at −15 °C. Recovery of growth after thawing and transferring to fresh medium at 20 °C indicated survival. All isolates in the Blastocladiales and Spizellomycetales survived freezing in all tests. All isolates in the Chytridiales also survived freezing in some tests. None of the isolates in the Rhizophydiales survived freezing in any of the tests. However, some isolates in the Rhizophydiales recovered growth after freezing if they were grown on PYG agar supplemented with either 1 % sodium chloride or 1 % glycerol prior to freezing. After freezing, the morphology of the thalli of all isolates was observed under LM. In those isolates that recovered growth after transfer to fresh media, mature zoosporangia were observed in the monocentric isolates and resistant sporangia or resting spores in the polycentric isolates. Encysted zoospores in some monocentric isolates also survived freezing. In some of the experiments the freezing and thawing process caused visible structural damage to the thalli. The production of zoospores after freezing and thawing was also used as an indicator of freeze tolerance. The chytrids in this study responded differently to freezing. These data add significantly to our limited knowledge of freeze tolerance in chytrids but leave many questions unanswered.  相似文献

Summary One purpose of this study was to determine whether an increase in plant density would result in a decrease in response to mycorrhizal infection (particularly as measured by phosphorus content). Increases in plant density generally result in increases in root density in the volume of soil occupied by the plants. Root density, in turn, largely determines phosphorus uptake. If mycorrhizal plants had significantly higher effective root densities than non-mycorrhizal plants due to the fungal hyphae and thus were more thorough in exploiting a given volume of soil for phosphorus, then a given increase in root density might result in a greater proportional increase in phosphorus uptake for non-mycorrhizal plants than for mycorrhizal plants. Two experiments were performed in which mycorrhizal infection and available soil volume per plant were manipulated; one in which the number of plants within a given pot size was varied (experiment 1), and another in which single plants were grown in pots of differing volume (experiment 2). The two experiments yielded similar results but for apparently different reasons. In the first experiment, for a given increase in root density, non-mycorrhizal plants had a greater proportional increase in phosphorus uptake than mycorrhizal plants. Thus, as predicted, response to mycorrhizal infection was greatest at the lowest planting density (highest available soil volume per plant, lowest root density). In experiment 2, response to infection was also greatest at the highest available soil volume per plant (largest pot), but pot size did not influence root density. These results show that the benefit from mycorrhizal infection may be partly determined by root density and they suggest that plants either occurring in patches of contrasting root density in a given community, or occurring in different communities with inherently different root densities may differ in their reliance upon mycorrhizal fungi for phosphorus uptake.  相似文献

Swamp dodder (Cuscuta gronovii) is a parasitic plant detrimental to cranberries. Observation of emergence of dodder seeds collected from a cultivated cranberry bog in Massachusetts revealed 2 or more peak emergence times during 4 consecutive growing seasons. Molecular methods were used to investigate genetic variation among the emerging dodder seedlings. On emergence, dodder seedlings were collected and analyzed for DNA sequence diversity in thetrnL (UAA) intron, a noncoding region of chloroplast DNA. DNA sequence analysis of 87 seedlings collected during the 1999 and 2000 growing seasons revealed the presence of 2 dodder ecotypes, designated A and B. Comparative DNA sequence analysis indicated that in thetrnL (UAA) intron, the sequence of ecotype A is identical to that ofCuscuta gronovii, whereas the sequence of ecotype B is closest to that ofCuscuta attenuata (99.3% sequence identity; 293 bases considered). ABg/II restriction enzyme cut site was identified that distinguished between thetrnL (UAA) introns of ecotypes A and B. Restriction fragment length polymorphism (RFLP) was used to analyze the sequences of 100 seedlings collected during the growing seasons of 2001 and 2002. Only 10 of the 187 samples were ecotype A, all of which emerged on or before May 7 in the growing seasons. Therefore, the predominant dodder haplotype found in this study may be a close relative ofC. attenuata and notC. gronovii, the common species found in cranberry bogs.  相似文献

Summary Plant yield of mycorrhizal and non-mycorrhizalFestuca rubra L. was linearly decreased with increasing zinc concentrations in nutrient solution. In all cases, non-mycorrhizal plant growth was significantly greater than that of mycorrhizal plants. Zinc and phosphorus concentrations of root and shoot of mycorrhizal plants were greater in all zinc treatments while mycorrhizal plants showed equal or lower tolerance indices to zinc than non-mycorrhizal plants. Yield depressions of mycorrhizal plants may be the result of enhanced zinc and phosphorus concentrations combined with the cost for growth and maintenance of the mycorrhizal fungi.  相似文献
