Populus euphratica has been used as a plant model to study resistance against salt and osmotic stresses, with recent studies having characterized the tonoplast and the plasma membrane ATPases, and two Na+/H+ antiporters, homologs of the Arabidopsis tonoplast AtNHX1, were published in databases. In the present work we show that P. euphratica suspension-cultured cells are highly tolerant to high salinity, being able to grow with up to 150 mM NaCl in the culture medium without substantial modification of the final population size when compared to the control cells in the absence of salt. At a salt concentration of 300 mM, cells were unable to grow but remained highly viable up to 17 days after subculture. The addition of a 1-M-NaCl pulse to unadapted cells did not promote a significant loss in cell viability within 48 h. In tonoplast vesicles purified from cells cultivated in the absence of salt and from salt-stressed cells, vacuolar H+-pyrophosphatase (V-H+-PPase) seemed to be the primary tonoplast proton pump; however, there appears to be a decrease in V-H+-PPase activity with exposure to NaCl, in contrast to the sodium-induced increase in the activity of vacuolar H+-ATPase (V-H+-ATPase). Despite reports that in P. euphratica there is no significant difference in the concentration of Na+ in the different cell compartments under NaCl stress, in the present study, confocal and epifluorescence microscopic observations using a Na+-sensitive probe showed that suspension-cultured cells subject to a salt pulse accumulated Na+ in the vacuole when compared with control cells. Concordantly, a tonoplast Na+/H+ exchange system is described whose activity is upregulated by salt and, indirectly, by a salt-mediated increase of V-H+-ATPase activity.  相似文献

Genes for V-H+-ATPase subunits were identified and cloned from the salt-tolerant wheat mutant RH8706-49. Sequences of these genes are highly conserved in plants. Overexpression of these genes in Arabidopsis thaliana improved its salt tolerance, and increased the activities of V-H+-ATPase and Na+/H+ exchange, with the largest increase in plants carrying the c subunit of V-H+-ATPase. Results from quantitative RT-PCR analysis indicated that the mRNA level of each V-H+-ATPase subunit in the Arabidopsis increased under salt stress. Overall, our results suggest that each V-H+-ATPase subunit plays a key role in enhancing salt tolerance in plants.  相似文献

We investigated the effects of silicon (Si) on time-dependent changes in root tonoplast H+-ATPase and H+-PPase activities, membrane fatty acid compositions and tonoplast fluidity in two barley (Hordeum vulgare L.) cultivars differing in salt tolerance. Plants were grown in NaCl-free (control) and NaCl-supplied (60 and 120 mM, respectively) nutrient solutions with or without 1.0 mM Si. Plant roots were harvested to isolate tonoplast vesicles for assay of H+-ATPase and H+-PPase activities at days 2, 4, and 6 after treatment in the first experiment and for analysis of membrane fatty acid composition and fluidity at day 4 after treatment in the second experiment. The results showed that tonoplast H+-ATPase and H+-PPase activities in roots of salt-treated plants increased at day 2, which was more obvious at 60 mM NaCl in the salt-tolerant cultivar than in the salt-sensitive cultivar, and then decreased at day 4 and onward. These enzyme activities decreased consistently from days 2 to 6 for treatment with 120 mM NaCl. However, inclusion of 1.0 mM Si significantly enhanced both H+-ATPase and H+-PPase activities in roots of salt stressed barley, which was irrespective of NaCl level or cultivar used. The ratio of unsaturated to saturated fatty acids (U/S) increased under salt stress for both cultivars. Addition of Si to salt treatment increased the ratio of U/S in salt-tolerant cultivar but it did not in salt-sensitive cultivar compared to non-Si-amended salt treatment. Salt treatment decreased tonoplast fluidity of roots of barley significantly compared with control treatment. However, root tonoplast fluidity was significantly lower in the Si-amended salt treatment than in the non-Si-amended salt treatment. These results were in line with the previous findings that Si could help increase antioxidative defense and reduce membrane lipid oxidative damage in barley under salt stress. The possible mechanisms involved in Si-enhanced salt tolerance were discussed with respect to cell membrane integrity, stability and function in barley.  相似文献

Wheat vacuolar H+-ATPases (V-ATPase) subunit B, named TaVB, was isolated from the salt-tolerant wheat RH8706-49 and used to transform Arabidopsis plants. TaVB-expressed Arabidopsis has a higher germination rate, root length, V–H+-ATPase activity, and overall salt tolerance than the wild type, indicating that expression of the gene can affect salt tolerance of the transgenic plants. Under salt stress, intracellular Na+ levels in transgenic plants are significantly lower than the control. These results suggest that expression of the wheat TaVB gene may enhance plant tolerance to salt stress.  相似文献

Two barley cultivars (Hordeum vulgare L., cvs. Elo and Belogorskii) differing in salt tolerance were used to study 22Na+ uptake, expression of three isoforms of the Na+/H+ antiporter HvNHX1-3, and the cellular localization of these isoforms in the elongation zone of seedling roots. During short (1 h) incubation, seedling roots of both cultivars accumulated approximately equal quantities of 22Na+. However, after 24-h incubation the content of 22Na+ in roots of a salt-tolerant variety Elo was 40% lower than in roots of the susceptible variety Belogorskii. The content of 22Na+ accumulated in shoots of cv. Elo after 24-h incubation was 6.5 times lower than in shoots of cv. Belogorskii and it was 4 times lower after the salt stress treatment. The cytochemical examination revealed that three proteins HvNHX1-3 are co-localized in the same cells of almost all root tissues; these proteins were present in the tonoplast and prevacuolar vesicles. Western blot analysis of HvNHX1-3 has shown that the content of isoforms in vacuolar membranes increased in response to salt stress in seedling roots and shoots of both cultivars, although the increase was more pronounced in the tolerant cultivar. The content of HvNHX1 in the seedlings increased in parallel with the enhanced expression of HvNHX1, whereas the increase in HvNHX2 and HvNHX3 protein content was accompanied by only slight changes in expression of respective genes. The results provide evidence that salt tolerance of barley depends on plant ability to restrict Na+ transport from the root to the shoot and relies on regulatory pathways of HvNHX1-3 expression in roots and shoots during salt stress.  相似文献

The effects of osmotic stress on H+-ATPase and H+-PPase activities and the levels of covalently conjugated polyamines (CC-PAs) and noncovalently conjugated polyamines (NCC-PAs) were investigated using tonoplast vesicles isolated from the roots of wheat (Triticum aestivum L.) seedlings differing in drought-tolerance. The results showed that after polyethylene glycol (PEG) 6,000 (–0.55MPa) treatment for 7 days, seedling leaf relative water content (LRWC), relative dry weight increase rate (RDWIR) and root H+-ATPase and H+-PPase activities from the drought-sensitive cultivar Yangmai No. 9 decreased more markedly than those from the drought-tolerant cultivar Yumai No. 18. At the same time, the increase of the NCC-spermidine (NCC-Spd) and CC-putrescine (CC-Put) levels in root tonoplast vesicles from Yumai No. 18 was more obvious than that from Yangmai No. 9. Exogenous Spd treatment alleviated osmotic stress injury to Yangmai No. 9 seedlings, coupled with marked increases of tonoplast NCC-Spd levels and H+-ATPase and H+-PPase activities. Treatments with methylglyoxyl bis (guanyl hydrazone) (MGBG), an inhibitor of S-adenosylmethionine decarboxylase (SAMDC), and phenanthrolin, an inhibitor of transglutaminase (TGase), significantly inhibited the osmotically induced increases of NCC-Spd and CC-Put levels, respectively, in root tonoplast vesicles from Yumai No. 18 seedlings. Both MGBG and phenanthrolin treatments markedly promoted osmotically induced decreases of tonoplast H+-ATPase and H+-PPase activities and osmotic stress tolerance of seedlings of this cultivar. These results suggest that the NCC-Spd and CC-Put present in tonoplast vesicles isolated from wheat seedling roots might enhance the adaptation of seedlings to osmotic stress via maintenance of tonoplast H+-ATPase and H+-PPase activities.  相似文献

The tonoplast and plasma membrane localized sodium (potassium)/proton antiporters have been shown to play an important role in plant resistance to salt stress. In this study, AtNHX1 and AtNHX3, two tonoplast Na+(K+)/H+ antiporter encoding genes from Arabidopsis thaliana, were expressed in poplar to investigate their biological functions in the resistance to abiotic stresses in woody plants. Transgenic poplar plants expressing either gene exhibited increased resistance to both salt and water-deficit stresses. Compared to the wild type (WT) plants, transgenic plants accumulated more sodium and potassium ions in the presence of 100 mM NaCl and showed reduced electrolyte leakage in the leaves under water stress. Furthermore, the proton-translocating and cation-dependent H+ (Na+/H+ or K+/H+) exchange activities in the tonoplast vesicles isolated from the leaves of transgenic plants were higher than in those isolated from WT plants. Therefore, constitutive expression of either AtNHX1 or AtNHX3 genetically modified the salt and water stress tolerance of transgenic poplar plants, providing a potential tool for engineering tree species with enhanced resistance to multiple abitotic stresses.  相似文献

The changes in transport activity of tonoplast proton pumps under the influence of exogenous NO donator and modulation of Ca2+ concentration jointly and separately were investigated at different stages of ontogenesis and under hyperosmotic stress. The results suggest that both exogenous NO donator and Ca2+ ions can influence the activity of transport processes related to tonoplast and this influence is especially evident in the period of growth and accumulation of metabolites. Under hyperosmotic stress, H+-pyrophosphatase plays a more important role than H+-ATPase: the activity of the former increases 2.3-fold compared to the control osmotic conditions, whereas the activity of H+-ATPase is practically unchanged. H+-pyrophosphatase was more responsive to the presence of exogenous NO donator and to variations in Ca2+ concentration. The effects of exogenous NO donator on tonoplast proton pumps depended on the concentration of Ca2+, which apparently can mediate NO action.  相似文献

A tonoplast protein of 31 kDa apparent molecular mass (TpP 31) was isolated from two-dimensional gels. Amino acid sequences were determined from LysC endoproteinase-peptide fragments. Using degenerate oligonucleotides, a corresponding cDNA clone of 1034 bp was isolated from a barley leaf cDNA library. It encodes for subunit E of the vacuolar H+-ATPase, the first one identified in plants so far. The open reading frame extends over 681 bp, encoding a gene product of 227 amino acids and a calculated molecular weight of 26 228 g mol?1. Northern and Western blot analysis indicates constitutive expression of subunit E in all plant organs with only small effects of salt stress. Localization of TpP 31 at the tonoplast was confirmed in fractions of purified vacuolar membrane obtained by free-flow electrophoresis. Immunoprecipitation of newly synthesized 35S-labelled membrane proteins with anti-TpP 31 gave two additional bands with apparent molecular masses of about 53 and 62 kDa. Gel filtration after mild solubilization showed co-purification of TpP 31 with the 55 kDa subunit of the H+-ATPase. Both results provide evidence beyond the sequence homology that TpP 31 is a structural component of the vacuolar H+-ATPase.  相似文献

Plants of the facultative halophyte and CAM species Mesembryanthemum crystallinum L. (Aizoaceae) were stressed for 8 d with 400 mol m−3 NaCl in the root medium. NaCl was then removed from the substratum, and the plants were watered again with NaCl-free solution. A second set of plants was maintained as controls. A small degree of CAM, as indicated by day-night changes in malate levels, was expressed during ageing of the plants. Salinity-stress-dependent CAM induction was reversible by the removal of salt, as indicated by similar Δ malate levels in previously salt-stressed plants and in non-stressed plants on day 19 of the experiment. Tonoplast vesicles were isolated from leaves during the time-course of stress application, stress removal and ageing. Parameters of the tonoplast H+-ATPase were correlated to the application of salinity, the expression of CAM and ageing. It was concluded, first, that a pronounced increase in the amount of tonoplast H+-ATPase is related to salinity per se and a smaller increase to ageing; secondly, that there is an increase in the specific activity of the enzyme related to ageing; thirdly, that the induction of two new polypeptides with molecular masses of 32 and 28 kDa is correlated in time with the expression of CAM, and, fourthly, that the two new polypeptides are part of the tonoplast H+-ATPase holoenzyme.  相似文献

A cDNA (SsCAX1) encoding a tonoplast-localised Ca2+/H+ exchanger was isolated from a C3 halophyte Suaeda salsa (L.). To clarify the role of SsCAX1 in plant salt tolerance, Arabidopsis plants expressing SsCAX1 were treated with NaCl. Transgenic Arabidopsis plants displayed decreased salt tolerance. Although Na+ content was close to wild-type plants, transgenic plants accumulated more Ca2+ and retained less K+ in leaves than the wild-type plants in salinity. Furthermore, transgenic lines held higher leaf membrane leakage than wild-type lines under NaCl treatment. In addition, transgenic plants showed a 23% increase in vacuolar H+-ATPase activity compared with wild-type plants in normal condition. But the leaf V-H+-ATPase activity had subtle changes in transgenic plants, while significantly increased in wild-type plants under saline condition. These results suggested that regulated expression of Ca2+/H+ antiport was critical for maintenance of cation homeostasis and activity of V-H+-ATPase under saline condition.  相似文献

High salinity is the one of important factors limiting plant growth and crop production. Many NHX-type antiporters have been reported to catalyze K+/H+ exchange to mediate salt stress. This study shows that an NHX gene from Arachis hypogaea L. has an important role in K+ uptake and transport, which affects K+ accumulation and plant salt tolerance. When overexpressing AhNHX1, the growth of tobacco seedlings is improved with longer roots and a higher fresh weight than the wild type (WT) under NaCl treatment. Meanwhile, when exposed to NaCl stress, the transgenic seedlings had higher K+/H+ antiporter activity and their roots got more K+ uptake. NaCl stress could induce higher K+ accumulation in the roots, stems, and leaves of transgenic tobacco seedlings but not Na+ accumulation, thus, leading to a higher K+/Na+ ratio in the transgenic seedlings. Additionally, the AKT1, HAK1, SKOR, and KEA genes, which are involved in K+ uptake or transport, were induced by NaCl stress and kept higher expression levels in transgenic seedlings than in WT seedlings. The H+-ATPase and H+-PPase activities were also higher in transgenic seedlings than in the WT seedlings under NaCl stress. Simultaneously, overexpression of AhNHX1 increased the relative distribution of K+ in the aerial parts of the seedlings under NaCl stress. These results showed that AhNHX1 catalyzed the K+/H+ antiporter and enhanced tobacco tolerance to salt stress by increasing K+ uptake and transport.  相似文献

Nitraria tangutorum Bobr. is a typical halophyte with superior tolerance to salinity. However, little is known about its physiological adaptation mechanisms to the salt environment. In the present study, N. tangutorum seedlings were treated with different concentrations of NaCl (100, 200, 300 and 400 mmol L?1) combined with five levels of Ca2+ (0, 5, 10, 15 and 20 mmol L?1) to investigate the effects of salt stress and exogenous Ca2+ on Na+ compartmentalization and ion pump activities of tonoplast and plasma membrane (PM) in leaves. Na+ and Ca2+ treatments increased the fresh weight and dry weight of N. tangutorum seedlings. The absorption of Na+ in roots, stems and leaves was substantially increased with the increases of NaCl concentration, and Na+ was mainly accumulated in leaves. Exogenous Ca2+ reduced Na+ accumulation in roots but promoted Na+ accumulation in leaves. The absorption and transportation of Ca2+ in N. tangutorum seedlings were inhibited under NaCl treatments. Exogenous Ca2+ promoted Ca2+ accumulation in the plant. Na+ contents in apoplast and symplast of leaves were also significantly increased, and symplast was the main part of Na+ intracellular compartmentalization. The tonoplast H+-ATPase and H+-PPase activities were significantly promoted under salt stress (NaCl concentrations ≤300 mmol L?1). PM H+-ATPase activities gradually increased under salt stress (NaCl concentrations ≤200 mmol L?1) followed by decreases with NaCl concentration increasing. The tonoplast H+-ATPase, H+-PPase and PM H+-ATPase activities increased first with the increasing exogenous Ca2+ concentration, reached the maximums at 15 mmol L?1 Ca2+, and then decreased. The tonoplast and PM Ca2+-ATPase activities showed increasing trends with the increases of NaCl and Ca2+ concentration. These results suggested that certain concentrations of exogenous Ca2+ effectively enhanced ion pump activities of tonoplast and PM as well as promoted the intracellular Na+ compartmentalization to improve the salt tolerance of N. tangutorum.  相似文献

Salinity stress is one of the most serous factors limiting the productivity of agricultural crops. Previous studies have shown that exogenous fatty acids (EFAs) enhanced plant performance in saline environment. However, the mechanisms remained unclear. This study aimed to investigate whether EFAs (palmitic and linoleic acids) had ameliorating effects on salt injury in NaCl-treated barley (Hordeum vulgare L.) seedlings, and to explore the possible mechanisms by determining tonoplast composition and function. The results showed that linoleic acid at 1 mmol l−1 in culture solution possessed protective effects on root tonoplast function against salt stress in the barley seedlings; this was accompanied with a significant suppression of the degradation of phospholipids and PAs in tonoplast vesicles. Moreover, these salt-ameliorating effects of linoleic acid on tonoplast function were also indicated by the increase in H+-ATPase and H+-PPase activities. In response to the changes in membrane bound enzyme activities, an augmentation in the activity of a vacuolar Na+/H+ antiport was occurred by the application of linoleic acid under saline conditions. These findings suggested that the application of linoleic acid exhibited protective effects on tonoplast function in the barley seedlings under salt stress, perhaps due partly to suppress the degradation of phospholipids and PAs in tonoplast vesicles, thus leading partial restorations in the activities of vacuolar H+-ATPase, H+-PPase and Na+/H+ antiport.  相似文献

According to sequences of several vacuolar Na+/H+ antiporter genes from Xinjiang halophytic plants, a new vacuolar Na+/H+ antiporter gene (HcNHX1) from the halophyte Halostachys caspica was obtained by RACE and RT-PCR using primers corresponding to conserved regions of the coding sequences. The obtained HcNHX1 cDNA was 1,983 bp and contained a 1,656 bp open reading frame encoding a deduced protein of 551 amino acid residues. The deduced amino acid sequence showed high identity with other NHX1 we have cloned previously from halophyte in Xinjiang desert area. The phylogenetic analysis showed that HcNHX1 formed a clade with NHX homologs of Chenopodiaceae. Expression profiles under salt treatment and ABA induction were investigated, and the results revealed that expression of HcNHX1 was induced by NaCl and ABA. To compare the degree of salt tolerance, we over-expressed HcNHX1 in Arabidopsis. Two transgenic lines grew more vigorously than the wild type (WT) under salt stress. The analysis of ion contents indicated that under salt stress, the transgenic plants compartmentalized more Na+ in the leaves compared with wild-type plants. Together, these results suggest that the products of the novel gene HcNHX1 from halophyte Halostachys caspica is a functional tonoplast Na+/H+ antiporter.  相似文献

选用两个耐盐性强弱不同的大麦(Hordeumvulgare L.)品种,研究了NaCl胁迫下其幼苗根中ATP和焦磷酸(PPi)含量的变化以及PPi对液泡膜H -ATP酶活性的影响.结果表明:在含NaCl 200mmol/L的1/2 Hoagland溶液中处理2 d,耐盐品种(滩引2号)根中液泡膜H -ATP酶活性增加,然后逐渐下降,而H -PPi酶活性在NaCl处理9 d中'直下降.盐敏感品种(科品7号)在NaCl胁迫下根中H -ATP酶和H -PPi酶活性都下降(图1).与对照相比较,NaCl胁迫下耐盐品种根中ATP含量2 d时增加,4 d后下降;盐敏感品种根中ATP积累受NaCl胁迫的抑制(图2).NaCl胁迫下,两品种的PPi含量皆略有增加(图3).PPi对液泡膜H -ATP酶活性有竞争性抑制作用(图4).结果表明:ATP积累是NaCl胁迫下液泡膜H -ATP酶活性增加的原因之一,NaCl胁迫下大麦品种根中ATP含量下降和PPi对液泡膜H -ATP酶的抑制使该酶活性下降.  相似文献
