The growth patterns of plants subjected to phosphorus starvation resemble those caused by treatment with ABA, suggesting that ABA could mediate the response of the plant to phosphorus starvation. We examined the role of ABA in phosphorus stress by comparing growth and biochemical responses of Arabidopsis thaliana ABA mutants aba-1 and abi2-1 to those of wild-type plants. We first characterized acid phosphatase production of wild-type Arabidopsis in response to phosphorus starvation. We found that several acid phosphatase isozymes are present in roots and shoots, but only a subset of these isozymes are induced by phosphorus stress, and they are induced in both organs. Production of acid phosphatase in response to phosphorus stress was not affected by the aba-1 or abi2-1 mutations. Low phosphorus also resulted in decreased growth of both wild-type and ABA mutant plants, and the root-to-shoot ratio was increased in both wild type and mutants. Anthocyanins accumulated in response to phosphorus stress in both wild-type and mutant plants, but the increase was reduced in the aba-1 mutant. Thus, two different ABA mutants responded normally in most respects to phosphorus stress. Our data do not support a major role for ABA in coordinating the phosphorus-stress response.  相似文献

Salt stress decreases plant growth prior to significant ion accumulation in the shoot. However, the processes underlying this rapid reduction in growth are still unknown. To understand the changes in salt stress responses through time and at multiple physiological levels, examining different plant processes within a single set-up is required. Recent advances in phenotyping has allowed the image-based estimation of plant growth, morphology, colour and photosynthetic activity. In this study, we examined the salt stress-induced responses of 191 Arabidopsis accessions from 1 h to 7 days after treatment using high-throughput phenotyping. Multivariate analyses and machine learning algorithms identified that quantum yield measured in the light-adapted state (Fv′/Fm′) greatly affected growth maintenance in the early phase of salt stress, whereas the maximum quantum yield (QYmax) was crucial at a later stage. In addition, our genome-wide association study (GWAS) identified 770 loci that were specific to salt stress, in which two loci associated with QYmax and Fv′/Fm′ were selected for validation using T-DNA insertion lines. We characterized an unknown protein kinase found in the QYmax locus that reduced photosynthetic efficiency and growth maintenance under salt stress. Understanding the molecular context of the candidate genes identified will provide valuable insights into the early plant responses to salt stress. Furthermore, our work incorporates high-throughput phenotyping, multivariate analyses and GWAS, uncovering details of temporal stress responses and identifying associations across different traits and time points, which are likely to constitute the genetic components of salinity tolerance.  相似文献

Boron (B) is an essential micronutrient for higher plants. There is wide genetic variation in the response to B deficiency among plant species and cultivars. The objective of this study was to identify quantitative trait loci (QTL) that control B efficiency in natural Arabidopsis accessions. The B efficiency coefficient (BEC) and seed yield under low B conditions (SYLB) were investigated by solution culture in two separate experiments in an Arabidopsis recombinant inbred line (RIL) population. Both of the traits studied exhibited high transgressive variation in the RIL population, and, in total, five and three QTL were identified for BEC and SYLB, respectively. Three of the five QTL, including the QTL, AtBE1-2, that has a large effect on the BEC, were found at the interval of the corresponding QTL for SYLB in both experiments. The close genetic relationship between BEC and SYLB was further confirmed by conditional QTL mapping in the RIL population and unconditional QTL mapping in an AtBE1-2-segregated F(2) population. Epistatic interactions for the tested traits were analysed, and were found to be widespread in the detected QTL of Arabidopsis in the RIL population. Comparison of the QTL interval for B efficiency with reported B-related genes showed that 10 B-related genes, together with one BOR1 homolog (BOR5, At1g74810) were located in the QTL region of AtBE1-2. These results suggest that natural variation in B efficiency in Arabidopsis has a complex molecular basis. They also provide a basis for fine mapping and cloning of the B-efficiency genes, with the ultimate aim of discovering the physiological mechanism of action of the genes.  相似文献

Plants exposed to stress pass the memory of exposure to stress to the progeny. Previously, we showed that the phenomenon of transgenerational memory of stress is of epigenetic nature and depends on the function of Dicer-like (DCL) 2 and DCL3 proteins. Here, we discuss a possible role of DNA methylation and function of small RNAs in establishing and maintaining transgenerational responses to stress. Our new data report that memory of stress is passed to the progeny predominantly through the female rather than male gamete. Possible evolutionary advantages of this mechanism are also discussed.Key words: transgenerational response to stress, Arabidopsis thaliana, maternal inheritance, methylation changes, homologous recombination frequency, genome instability, adaptive response, dcl2, dcl3Plants are sedentary organisms and thus can not respond to rapidly changing growth conditions by escaping to new environments as animals usually do. Moreover, since seed dispersal is rather limited in the vast majority of plants, the progeny is very likely to grow under the same environmental growth conditions as its parents did. The memory of pre-existing growth conditions can be advantageous for plant survival. The environmental experience of parents can be recorded in the form of induced epigenetic modifications that occur in somatic cell lineages. The very late, almost at the end of plant development, separation of germline cells from somatic tissues enables incorporation of acquired epigenetic changes in the gametes. Indeed, previous reports suggested that the progeny of exposed plants might have an advantage while growing in the same environment as its parents.13 Despite a growing number of experimental evidences that support the existence of the phenomenon of memory of stress, the data on adaptive changes in the progeny of stressed plants are scarce.Parental exposure to stress may not only lead to adaptive effects in progeny but also introduce a certain degree of changes in genome stability.49 Our early report showed that the progeny of tobacco plants infected with tobacco mosaic virus had an increased meiotic recombination frequency.8 A more recent report demonstrated that these progeny plants had a higher frequency of rearrangements at the loci carrying the homology to N-gene-like R-gene loci, allowing speculations about a possible role of these rearrangements in pathogen resistance evolution.9 Similarly, a study of Molinier et al. (2006) showed that the progeny of plants exposed to UVC or flagellin had an increased frequency of somatic homologous recombination events (HRF).4 The authors demonstrated that an increase in HRF triggered by a single exposure to UVC was maintained for five consecutive generations in the absence of stress. In contrast, our most recent reports demonstrated that maintaining an increase in HRF caused by ancestral exposure to heat, cold, flood, UVC or salt required exposure to stress in subsequent generations: if F1 plants were propagated for one more generation without stress, the effect diminished and HRF returned back to the level observed in the progeny of untreated plants.6,7 This scenario seems to be more probable from an evolutionary point of view. Within a given environmental niche, plants establish certain genetic and epigenetic traits needed to cope with the expected growth conditions. Drastic environmental changes or new unusual stresses may trigger a cascade of gene expression changes in attempt to survive and adapt to new conditions. Some of these potentially advantageous changes are most probably recorded in the form of DNA methylation and chromatin modifications and are passed to progeny as memory of stress exposure.It can be further hypothesized that if these new environmental conditions are no longer present during the lifespan of future generations, the newly established methylation patterns and chromatin organization will return to the original epigenetic landscape that was the most adequate fit for this environmental niche. If the same new stresses occur in consecutive generations, the newly established epigenetic changes will be maintained and possibly stabilized after many generations of exposure.  相似文献

The effects of coarse vs. fine-grained variation in water regime on 13 accessions of Arabidopsis thaliana were compared and it was found that the scale of environmental variation can lead to differences in which traits are affected and in the magnitude and direction of effect. Coarse-grained (between individual) variation was studied by comparing plants that received high or low amounts of water on a daily basis. Fine-grained (within individual) variation was distributed across the coarse-grained treatments; plants in the fine-grained treatment received the same amount of water per week as the corresponding high and low coarse-grained treatments; however, in the fine-grained treatments, plants received their entire weekly water allotment over 2 days. The number of leaves produced, days to bolting, length of bolt, number of basal and lateral branches, and survival to 30 days after bolting were measured and analyzed by ANOVA. Plants grown in high water generally produced more leaves, taller main axes, more branches and had increased survival, relative to those in low water. Bolting date showed a significant genotype specific effect of water level. Plants grown under fine-grained variation in watering had taller main axes relative to constant water. There were significant interactions between both types of variation for bolting date, main axis height, and survival. Principal components for all traits were loaded such that the components themselves were also independently affected by genotype, water level, variability of water level (grain), and level-by-grain interactions in ways that were largely predicted by their component traits. Overall, the effects of fine-grained variation were subtler than those of coarse-grained variation: fewer traits responded, and the magnitude of the responses was smaller. Nevertheless, the responses to fine-grained variation are distinct from those of coarse-gained variation, differing with respect to which traits respond and in the direction of response, and must therefore be treated as independent sources of environmental variation.  相似文献

Evidence of adaptation in Arabidopsis thaliana (Brassicaceae) phenotypic traits has rarely been shown. We demonstrate latitudinal clines in two A. thaliana traits: hypocotyl responses to red and far-red light. Natural populations of A. thaliana were sampled along a latitudinal gradient from southern to northern Norway. Seeds from maternal families within each population were subjected to 1 wk of constant red, far-red, blue, white, and dark treatments. Hypocotyl lengths were measured for each maternal family within each population. Significant variability within and among populations in hypocotyl responses for the various treatments was found. There was a significant latitudinal cline in hypocotyl responses for red and far-red treatments, with northern populations being more de-etiolated than southern populations. These results suggest that northern populations are more responsive to red and far-red light than southern populations. Thus, differentiation of seedling traits in natural populations of A. thaliana seems in part to be mediated by the phytochrome pathway. There was no correlation between hypocotyl responses and flowering time for any treatment. This suggests that flowering time variability and variability in hypocotyl responses may not be governed by genes shared between the pathways, such as those involved in photoreception or the circadian clock.  相似文献

Autophagy is a dynamic process that involves the recycling process of the degradation of intracellular materials. Over the past decade, our molecular and physiological understanding of plant autophagy has greatly been increased. Most essential autophagic machineries are conserved from yeast to plants. The roles that autophagy-related genes (ATGs) family play in the lifecycle of the Arabidopsis are proved to be similar to that in mammal. Autophagy is activated during certain stages of development, senescence or in response to starvation, or environmental stress in Arabidopsis. In the progression of autophagy, ATGs act as central signaling regulators and could develop sophisticated mechanisms to survive when plants are suffering unfavorable environments. It will facilitate further understanding of the molecular mechanisms of autophagy in plant. In this review, we will discuss recent advances in our understanding of autophagy in Arabidopsis, areas of controversy, and highlight potential future directions in autophagy research.  相似文献

Irradiation of Arabidopsis thaliana ecotypes C24, Wassilewskija (Ws) and Columbia-0 (Col-0) with supplementary ultraviolet-A+B (UV-A+B) radiation revealed ecotype-specific differences in expression of the gene for the pathogenesis-related protein PR-5. C24 showed an increased expression level of PR-5 (5- and 20-fold higher compared with Col-0 and Ws, respectively). Expression of other molecular markers such as CHS (encoding chalcone synthase), MEB5.2 [encoding a gene strongly up-regulated by ultraviolet-B (UV-B)] and PYROA [encoding a pyridoxine (Vitamin B6) biosynthesis enzyme] only showed slight differences between ecotypes. Oxidative stress during UVA+B exposure was monitored by staining for H2O2. This analysis also revealed important ecotype-specific differences. 'H2O2 hot spots' were found in C24, whereas an even distribution of H2O2 was found in Ws and Col-0. Necrotic lesions also appeared on C24 leaves after prolonged UV-B exposure. There was a reverse correlation between the H2O2 steady-state concentration and the PR-5 gene expression; Ws showed the highest level of H2O2 accumulation but the lowest expression level of the PR-5 gene. Furthermore, application of paraquat on the rosettes led to similar PR-5 expression and H2O2 accumulation patterns as were found after UV-A+B irradiation. The observed ecotypic differences were also reflected in a statistically significant UV-B-dependent decrease in biomass, rosette size and leaf area for Ws, but not for C24 and Col-0. Our results show that a significant ecotype-specific genetic variability in general UV-B responses in Arabidopsis exists. Moreover, the signal transduction or gene regulation pathway for PR-5 differs from the other molecular markers used in this study.  相似文献

Subcellular events of Erysiphe cichoracearum infections of epidermal cells were visualized in living tissues of Arabidopsis plants carrying various green fluorescent protein (GFP)-tagged organelles via laser scanning confocal microscopy. Early in the infection sequence, cytoplasm and organelles moved towards penetration sites and accumulated near penetration pegs. Peroxisomes appeared to accumulate preferentially relative to the cytoplasm at penetration sites. Another early event, which preceded haustorium formation, was the aggregation of some GFP-tagged plasma membrane marker proteins into rings around penetration sites, which extended across cell-wall boundaries into neighboring cells. This feature localized to sites where papillae were deposited. The extrahaustorial membrane (EHM) encases the fungal feeding structure, the haustorium, separating it from the host cytoplasm. Eight plasma membrane markers were excluded from the EHM and remained in a collar-like formation around the haustorial neck. These observations support the suggestions that the EHM is a unique, specialized membrane and is different from the plasma membrane. Our results suggested two possibilities for the origin of the EHM: invagination of the plasma membrane coupled with membrane differentiation; or de novo synthesis of the EHM by targeted vesicle trafficking.  相似文献
