Abstract Although rice has long been recognized to be uniquely adapted for growth in low oxygen environments of flooded rice fields, rice weeds of the Echinochloa crus-galli complex appear to be at least as well specialized for germination and growth under such unusual biological conditions. Seeds of two varieties of E. crus-galli germinate and grow for prolonged periods in a totally oxygen-free environment. E. crus-galli germinates as well as rice (Oryza sativa) under a total nitrogen atmosphere and produces as large a seedling in spite of its much smaller seed size. Like rice, the seedlings of E. crus-galli are unpigmented, the primary leaves do not emerge from the coleoptile and no root growth occurs without oxygen. Of particular interest is the ultrastructure of mitochondria from anaerobically-grown seedlings. Mitochondrial profiles from the primary leaf of seedlings grown continuously in nitrogen are very similar to those grown aerobically. The size and shape of the mitochondria are similar and the cristae are numerous and normal in appearance. This is in sharp contrast to previous studies of other species which have reported that mitochondria were vesiculate and tended to lose their normal fine-structure after similar periods without oxygen. Finally, based on ultrastructure and 14C labeling studies, anaerobically-grown seedlings are highly active metabolically, which may explain, at least for E. crus-galli var. oryzicola, its ability to germinate and emerge from flooded rice fields.  相似文献

Endogenous hormones, namely cytokinins (CKs), indole-3-aceticacid (IAA) and abscisic acid (ABA) were quantified by specificenzyme-linked immunosorbent assay (ELISA) in the mature seedof normal (cv. Westar) and ogura (ogu) cytoplasmic male sterile(CMS) lines of rapeseed (Brassica napus L.). Dihydrozeatin (DZ)and dihydrozeatin riboside (DZR) were the major CK base andriboside, respectively, in seeds of both the normal and oguCMS lines. The normal seed had more than 4-fold DZ levels incomparison to that of ogu CMS. On the other hand, the ogu CMSseed had higher levels of CK o-glucosides and CK. nucleotidesthan normal seed. Seeds of the normal line contained 5-foldmore IAA but had one-quarter the level of ABA in comparisonto those of the ogu CMS line. The normal line also had greaterseed diameter and weight than the ogu CMS line and the normalseed germinated earlier than the male sterile seed. DZ (10–6M) promoted the germination of ogu CMS seeds, but it was notcomparable to that of the normal line. ABA (10–6 M) inhibitedseed germination of ogu CMS but had little effect on the normalline. The normal seedlings had shorter primary roots, more lateralroots, longer hypocotyls, greater cotyledon fresh weight andhigher chlorophyll levels in comparison to ogu CMS seedlings.Exogenously supplied DZ, IAA and ABA affected the various parametersof both the normal and ogu CMS seedlings, but in most casesdid not fully restore the differences in the two lines. The results presented show that in the ogura cytoplasmic malesterile line of B. napus (1) a number of seed and seedling characteristicsare affected, and (2) the altered seed morphology is accompaniedby changes in the levels of various hormones. High population densities of germinating rice seedlings in initiallyair-saturated sealed aquatic environments exhibited dseedling growth consisting solely of coleoptile emergence inlight and dark environments. Residual oxygen tensions of 17–23%of the initially air-saturated water containing the dseedlings were evident after 15 d in both the light and dark.Coleoptiles of all d seedlings were stark white in appearance,lacked protochlorophyllide, and contained proplastids and amyloplasts,there being no evidence of normal etioplast development in thelight or dark and no chloroplast development in the light. Thus,complete environmental anoxia was observed to be unnecessaryfor inhibiting normal chloroplast photomorphogenesis in coleoptilesof light-germinated rice seedlings. Increasing the oxygen tensionsof the 15-d-old aquatic environments of light- and dark-germinatedd seedlings placed in the light resulted in normal chloroplastphotomorphogenesis in coleoptiles, shoots, and roots. A particulate fraction similar to mitochondria was isolatedfrom germinating lettuce seedlings. The endogenous oxygen uptake of this fraction was very high.It could oxidize most of the Krebs cycle intermediates, theoxidation of l-malate being particularly marked. Glutamic, glycollic, and formic acids could also be oxidizedto a certain extent. Fatty acids inhibited the endogenous respiration,the inhibition increasing with increased concentration. Coumarinand thiouren do not seem to have any direct effect on the respiratoryenzyme systems. They apparently affect the respiration of wholeseeds only indirectly, maybe through some germination regulatingmechanism not yet known.  相似文献

Rice seeds were germinated for up to 5 days under water (submerged)and some for another day in air (air-adapted). Control seedswere germinated for 6 days throughout in air. Low-temperaturedifference spectra of shoot mitochondria were compared amongthese three types of seedlings. All cytochromes found in theaerobic seedlings were present in the submerged seedlings. However,there were some differences in the cytochromes b553 and c ofthese two types of seedlings. The cytochrome aa3 peak heightand cytochrome oxidase activity per mitochondrial protein increased1.6- and 2.8-fold, respectively, during air adaptation. Slightlyhigher concentrations of the b-type cytochromes than found inair-adapted mitochondria were already present in submerged mitochondria.The computed difference between the dithionite-reduced differencespectra of mitochondria from submerged seedlings before andafter air adaptation, showed that cytochromes aa3 and c hadincreased more than cytochrome b557 during air adaptation. Germination of strawberry seed was accelerated by natural andartificial infections of Ulocladium charatarum and other fungi,including the Arthrinium state of Apiospora montagnei and aCladosporium sp. Germination was variously promoted by fungiboth in light and darkness, on damp filter paper, water andmalt agar plates and on horticultural compost. Germinated seedlingswere apparently undamaged by these infections, although whencultured on malt agar the seedlings were overgrown by the fungus.Several other fungi promoted germination, but three of themlater killed the seedlings. Germination of seed of all threecultivars that were tested was promoted by fungal infectionwhich probably promoted germination by hastening breakdown ofthe seed coat. The influence of prehydration in water or priming in –1.5 MPa polyethylene glycol 8000 solution for various periods,followed by redrying, on germination rate and longevity of lettuce(Lactuca sativa L.) seeds (achenes) was determined during controlleddeterioration at 10% moisture content (fresh weight basis) and40°C. Short prehydration treatments (up to 1 h) had littleeffect on either germination rate or longevity, but significantlyimproved root growth rates. Increasing durations of prehydrationor priming reduced the mean time to germination by up to 61%relative to untreated seeds, but also reduced mean seed longevityby as much as 84% Prehydration and priming altered the relationshipsbetween germination rate and viability and between normal andabnormal seedlings during ageing. Prehydration in abscisic acidor at a temperature inhibitory to germination did not preventthe reduction in longevity under controlled deterioration conditions.While prehydration or priming treatments effectively acceleratesubsequent germination rates of lettuce seeds, the redried seedsare nonetheless highly susceptible to deterioration in storage. Some workers have reported that the breaking of seed dormancyin rice (O. sativa L.) is usually enhanced by higher oxygentension, whereas others have shown that rice seed dormancy canbe broken by incubation under anaerobic conditions. This articleaims to clarify this paradox. The results show that high oxygentensions inhibit seed germination for a certain period afterharvest in Japonica rice, whereas Indica rice cultivars arenot inhibited by oxygen at any stage. Respiratory activities were compared among rice seedlings germinated in air for 6 days (aerobic seedlings), those germinated under water for 5 days (submerged seedlings), and those grown in air for 1 day after 5 days' submerged germination (air-adapted seedlings). The respiratory activity of the submerged seedlings increased rapidly on transfer to air and reached a plateau at 16 hours in air. Respiration of the submerged seedlings was as sensitive to cyanide as those of aerobic and air-adapted seedlings. 2,4-Dinitrophenol had no effect on the respiration of the submerged seedlings, but stimulated those of the other two types of seedlings. Mitochondria from three types of seedlings did not differ in the ADP/O ratio and the respiratory control ratio (RCR) when succinate was oxidized. However, mitochondria from submerged seedlings (submerged mitochondria) showed poor RCR of about unity when malate was oxidized. Both the rate of succinate oxidation and succinate dehydrogenase activity were low in submerged mitochondria, but increased during air adaptation. Although submerged mitochondria oxidized malate very slowly, this activity increased after exposure to air without any increase in malate dehydrogenase activity. When NAD+ was added to submerged mitochondria, oxidation of malate was restored to the level of the aerobic controls. Addition of NAD+ enhanced the state 3 rate in submerged mitochondria, and RCR recovered to nearly the same value as that of the aerobic controls. Similar effects of NAD+ on 2-oxoglutarate oxidation were observed. All these defects in submerged mitochondria were repaired during air adaptation. These results suggest that NAD+-linked substrate oxidation was low in submerged mitochondria because of NAD+ deficiency, and that the oxidation increased with an increasing level of NAD+ during air adaptation.  相似文献

Methanol extracts from light- and dark-grown sunflower seedlings,Helianthus annuus L., var. ‘Inra No. 6501’, weresubjected to solvent partition. The neutral diethyl ether fractionfrom the light-grown seedlings contained material(s) which inhibitedcress seed germination. Such inhibition was scarcely detectablein extracts of the etiolated seedlings. Sometimes inhibitionalso occurred in the petroleum spirit fraction. A mixture ofcis, trans- and trans, transxanthoxin showed the same partitioncharacteristics as the inhibiting substance(s); the latter co-chromatographedwith xanthoxin in all paper, thin layer, and high pressure liquidchromatography separation procedures tried. The inhibition wasalso detectable with the Avena colcoptile straight growth bioassay. It is concluded that xanthoxin is formed during illuminationof sunflower seedlings; its role in the phototropic responseof these seedlings is discussed.  相似文献

The synthetic methyl ester of GA73 (GA73-Me) and the naturalantheridiogen of Lygodium japonicum showed almost the same activityto induce the formation of antheridia in dark-grown protonemataof L. japonicum at concentrations of 10-14 M and higher. Thus,it appears that the principal antheridiogen in L. japonicumis GA73-Me. GA73-Me inhibited formation of ar-chegonia in light-grownprothallia of L. japonicum at concentrations of 10-11 M andhigher and induced germination of spores in the dark in thisspecies at the same range of concentrations. GA73(free acidform) promoted growth of seedlings of dwarf rice and hypocotylsof cucumber seedlings at dosages of and above 1 and 100 ng/plant,respectively. Eight compounds related to GA73-Me, includingantheridiogens of Anemia phyllitidis and Anemia mexicana, wereactive in inducing an-theridial formation in L. japonicum, althoughtheir activities were considerably lower than that of GA73-Me. Observations by scanning electron microscopy revealed that rhizobiaattach to the surface of rice protoplasts with regenerated cellwalls, isolated mesophyll cells of asparagus, and root hairsof rice and oat seedlings. Those strains of rhizobia, namelyRhizobium leguminosarum biovar trifolii, Bradyrhizobium japonicumand Bradyrhizobium sp., attach to the cells of these monocotsin the same manner as they attach to the host dicots tested.Escherichia coli did not attach. These results suggest thatthe attachment of rhizobia is not a host-specific process. Whenoat seedlings were infected by R. l. trifolii, hair curlingwas observed. Causes of cytoplasmic male sterility (CMS) in plants have beenstudied for two decades, and mitochondrial chimeric genes havebeen predicted to induce CMS. However, it is unclear what happensafter CMS-associated proteins accumulate in mitochondria. Inour previous study of microarray analysis, we found that 140genes are aberrantly regulated in anthers of CW-type CMS ofrice (Oryza sativa L.). In the present study, we investigatedDCW11, one of the down-regulated genes in CW-CMS encoding aprotein phosphatase 2C (PP2C). DCW11 mRNA was preferentiallyexpressed in anthers, with the highest expression in maturepollen. As predicted by the N-terminal sequence, DCW11 signalpeptide–green fluorescent protein (GFP) fusion proteinwas localized in mitochondria. Knockdown of DCW11 in wild-typerice by RNA interference caused a major loss of seed-set fertility,without visible defect in pollen development. Since this knockdownphenotype resembled that of CW-CMS, we concluded that the down-regulationof DCW11 is correlated with CW-CMS. This idea was supportedby the up-regulation of alternative oxidase 1a (AOX1a), whichis known to be regulated by mitochondrial retrograde signaling,in DCW11 knockdown lines. Down-regulation of DCW11 and up-regulationof AOX1a were also observed in two other types of rice CMS.Our result indicates that DCW11 could play a role as a mitochondrialsignal transduction mediator in pollen germination.  相似文献

When seeds of Chenopodium album are imbibed in 0–01 Mpotassium nitrate solution in the light at constant temperature,percentage germination increases to an optimum at 24 °C,above which it decreases. These relationships are linear ifpercentage germination values are transformed to normal deviates.At supra-optimal temperatures, alternating temperatures havelittle or no effect on this basic relationship. However, atsub-optimal temperatures normal deviate germination increaseslinearly with amplitude at constant mean temperature and theincreases are relatively greater at lower mean temperatures.The effect of amplitude is also greater when more time is spentat the wanner temperature in the diurnal cycle. Seeds of Panicummaximum show very similar responses except that the effect ofamplitude is greater when a shorter time is spent at the warmertemperature in the diurnal cycle. These observations form thebasis of a quantitative model which, at sub- and supra-optimaltemperatures, respectively, accounts for 90 and 75% of the variationin germination of C. album seeds subject to a very wide rangeof thermal environments on a two-dimensional temperature gradientplate. The data presented for P. maximum are less comprehensive,but again the model accounts for 80% of the variation. The relevanceof the same model to two unrelated species from different climatesand of different ecological behaviour suggests that it may begenerally useful in determining optimum temperatures for seedviability testing regimes and ultimately for predicting fieldbehaviour These results allow the producers to improve the productionof homogeneous populations of cyclamen seedlings Chenopodium album L., Panicum maximum Jacq., seed dormancy, germination, alternating temperatures, temperature gradient plate  相似文献

In this study the effects of seed size variation on germinationand seedling vigour have been investigated within and betweenploidy levels of diploid and related autotetraploid Dactylixglomerata. Rates of seed germination and seedling growth werecompared in two contrasting environments using diploid and tetraploidseeds of equal and also different biomass. Within each ploidylevel, seed biomass had no effect on either the overall percentagenor the rate of germination. In contrast, the comparison ofseeds of equal biomass but differing in ploidy level showedthat seeds from tetraploid plants germinated faster and to ahigher percentage than those from diploid plants. With respectto seedling growth, heavier seeds from the tetraploid genotypesgave seedlings of significantly higher biomass than those fromlighter tetraploid and diploid seeds throughout the 2 monthsof study. Interestingly, seeds of equal biomass but from differentploidy levels produced seedlings more similar than those fromthe extreme seed weight categories. These differences were maintainedin two different environmental conditions. Quercus floribunda Lindl. (tilonj oak), a late successionalspecies, and Cupressus torulosa D. Don. (surai), an early successionalspecies, were compared in terms of seed germination and seedlinggrowth. In Petri dish germination tests in which light quality,temperature and osmotic water stress were varied, the germinationof both species was reduced by continuous dark or far-red lightand by osmotic water stress. These effects were greater on thegermination of surai than on that of tilonj oak. The growthof seedlings in soil was measured over a twelve-month periodduring which different levels of shading and different wateringregimes were applied. Surai was less affected than tilonj oakby reduced soil moisture, but the effect of deep shade treatmentswas greater on surai than with tilonj oak. At higher soil moisturestress seedlings of surai attained greater height then tilonjoak, but the latter achieved higher dry weight and a higherroot: shoot and lower leaf-weight ratio. Summary Ultrastructural changes of organelles, especially those of mitochondria in rice seedlings germinated under strictly anaerobic conditions were investigated.The embryos of dry seeds had slightly modified mitochondria, characterized by an electron transparent matrix with few cristae and electron opaque patches. These mitochondria developed normally for 48 hours irrespective of whether oxygen was present or not. However, after 72 hours' germination under anaerobic conditions vesiculation of the cristae developed and progressed greatly for the subsequent 24 hours and most of the spaces in the mitochondrion became occupied with vacuolated cristae. Vesiculation seemed to be the effect of cessation of energy supply from the mitochondria themselves.Ultrastructural changes of other organelles such as the plastids and endoplasmic reticulum were also observed after 96 hours under anaerobic conditions.  相似文献
