Summary

The structure of the female gonad has undergone important evolutionary modifications in Platyhelminthes-Turbellaria. The primitive condition consists of single oocytes freely distributed in the parenchyma autosynthetically producing both yolk and eggshell forming granules (e.f.g.) (archoophoran level of organization). A derived condition is the presence of a compact, hetero-cellular gonad enveloped by a cellular tunica and/or an extracellular lamina-like layer and composed of germaria with alecithal oocytes and vitellaria with vitelline or yolk cells producing and accumulating both yolk and e.f.g. (neoophoran level of organization). In the last three decades the female gonad of a number of turbellarian taxa has been investigated by means of electron microscopy and cytochemistry, and several characters of oocytes and vitellocytes have been hypothesized to have a potential phylogenetic value. Some of these characters and their possible phylogenetic implications are briefly reviewed.  相似文献

It has been proposed, on the basis of widespread phylogenetic conservation, that H-Y antigen is the inducer of primary sex, causing the undifferentiated XY gonad to become a testis in male heterogametic species such as the human and bovine. That proposition has withstood extensive testing in vivo and in vitro. Freemartin gonads are H-Y+, for example, and masculinization of the freemartin gonad has been attributed to soluble H-Y, borne and transmitted in the serum of the bull twin, and bound in ovarian receptors of the female. We have applied monoclonal H-Y antibodies to the identification of gender in embryos of the bovine. Our preliminary results imply presence of H-Y in bovine embryos of the morula and blastocyst stages recovered at about 6–12 days of gestation. Assignment of H-Y phenotype -- positive in males and negative in females -- allows selective implantation of male and female during embryo transfer. Thus in an early study, we correctly identified gender in 6 of 7 calves born healthy at term, after transfer of 8 blastocysts.  相似文献

The gonadal anatomy and sexual pattern of Rhabdosargus sarba (Teleostei: Sparidae) was studied to provide some basic structural information for the subsequent investigations on the endocrinology of protandrous hermaphroditism in this fish. Evidence derived from relating sex to differences in body size, from gonadal histology and from biopsy, revealed the occurrence of natural sex reversal from male to female in this species. The gonad of R. sarba possessed topographically distinct male and female zones that were well separated by connective tissue. Based on gross-anatomical and histological observations, four types of gonad were distinguished and were designated as Types I-IV in the present study. Active spermatogenetic tissue was present in the Type I, II and IV gonads. Dormant oogonia and perinucleolar oocytes were found in the Type I (male) and II (intersex) gonads, respectively. In the Type III (female) gonad, a functional ovarian zone was observed and the testicular tissue was vestigial. The existence of ovarian tissue as an oogonial band in the Type IV (male) gonad, which was found more commonly in large specimens, suggested that these functional males might not undergo sex reversal in their life cycle. The interrelationship of the different types of gonad is discussed with reference to protandrous hermaphroditism.  相似文献

湖泊放养长江水系中华绒螯蟹的性成熟规律研究   总被引:11,自引:1,他引:11  
于8-11月份,结合中华绒螯蟹捕捞期,即生死洄游期的到来,通过性腺发育的组织学和细胞学观察,重点观察了长江下游安徽杨赛湖等人工放养长江水系中华绒螯蟹的性成熟规律。结果表明:①在性成熟时期,性腺发育与肝脏变化极为密切,肝脏指数逐渐下降,而性腺指数逐渐上升;②雌、雄蟹的性成熟存在规律性差异,8-9月份,雄蟹的性腺指数和性/肝指数均高于雌蟹,而10-11月份,雌蟹的性腺指数和性/肝指数均高于雄蟹;③经性腺的组织学和细胞学观察,雄蟹的性腺发育略早于雌蟹;④洄游高峰期的出现是河蟹性细胞走向成熟的外在表现。作者建议把肝脏指数7.5作为捕捞长江水系河蟹最佳时机的指标。  相似文献

为探寻中国大鲵(Andrias davidianus)生殖腺胚后发育的特点及规律,采用解剖学与组织学技术对其形态结构变化进行了观察.结果表明,大鲵的原始生殖腺开始出现于出膜28~49 d;出膜133~175 d时一些个体生殖腺内已初步分化出原始卵泡;出膜259 ~343 d时一些个体生殖腺内已初步分化出生精小叶;出膜427 d时,卵巢已明显分化为皮质与髓质,且髓质内出现了卵巢腔,精巢内生精小叶及其内的腔隙、精巢间质等分化已较为明显;出膜511 d时精巢分化为明显的生精小叶和非成熟小叶两个区域.本文认为,大鲵与其他无羊膜类原始生殖腺的分化一般发生在胚后阶段,而且雌性的分化时间早于雄性.  相似文献

Specialized cells of the somatic gonad primordium of nematodes play important roles in the final form and function of the mature gonad. Caenorhabditis elegans hermaphrodites are somatic females that have a two-armed, U-shaped gonad that connects to the vulva at the midbody. The outgrowth of each gonad arm from the somatic gonad primordium is led by two female distal tip cells (fDTCs), while the anchor cell (AC) remains stationary and central to coordinate uterine and vulval development. The bHLH protein HLH-2 and its dimerization partners LIN-32 and HLH-12 had previously been shown to be required for fDTC specification. Here, we show that ectopic expression of both HLH-12 and LIN-32 in cells with AC potential transiently transforms them into fDTC-like cells. Furthermore, hlh-12 was known to be required for the fDTCs to sustain gonad arm outgrowth. Here, we show that ectopic expression of HLH-12 in the normally stationary AC causes displacement from its normal position and that displacement likely results from activation of the leader program of fDTCs because it requires genes necessary for gonad arm outgrowth. Thus, HLH-12 is both necessary and sufficient to promote gonadal regulatory cell migration. As differences in female gonadal morphology of different nematode species reflect differences in the fate or migratory properties of the fDTCs or of the AC, we hypothesized that evolutionary changes in the expression of hlh-12 may underlie the evolution of such morphological diversity. However, we were unable to identify an hlh-12 ortholog outside of Caenorhabditis. Instead, by performing a comprehensive phylogenetic analysis of all Class II bHLH proteins in multiple nematode species, we found that hlh-12 evolved within the Caenorhabditis clade, possibly by duplicative transposition of hlh-10. Our analysis suggests that control of gene regulatory hierarchies for gonadogenesis can be remarkably plastic during evolution without adverse phenotypic consequence.  相似文献

The left gonad from female chick embryos at 4–12 days of incubation was cultured in vitro as pieces of intact gonad, pieces of isolated cortex, and groups of pure germ cells. All cultures were maintained for a time equal to 17 days in ovo. At the end of the culture period, a cytological and quantitative study was made on the germ cells.The results show that some germ cells in pieces of intact 6-day gonad and pieces of 6-day cortex complete their normal developmental sequence and enter zygotene. This shows that the factors that control the differentiation of the germ cells reside in the cortex of the gonad and their expression does not depend upon the pituitary and the medullary estrogens after 6 days of incubation.Germ cells that are cultured as isolated cells do not attach to the tissue culture substrate, do not divide mitotically, and do not enter zygotene. Evidence is presented that suggests 12-day germ cells do enter zygotene when cultured with pieces of 12-day cortex. These data suggest the differentiation of the female germ cells is regulated by the somatic cells of the cortex.  相似文献

In the chicken, serological H-Y antigen is specific for the female sex. Male gonad differentiation can be experimentally influenced by estrogens, resulting in the transient formation of an ovotestis. The sex-inverted gonad becomes positive for H-Y antigen. Therefore, the question arises whether, in normal gonadogenesis also, the female gonad at the indifferent stage, before estrogens are produced, is negative for H-Y antigen. Here we show that this is indeed the case. The female gonad becomes positive for H-Y antigen when the ovary starts its organotypic differentiation at about day 6 1/2 of embryonal development. It is assumed that estrogens are responsible for the occurrence of H-Y antigen. This finding supports the view that H-Y antigen plays a role in primary ovogenesis in the chicken.  相似文献

A novel form of mitochondrial DNA (mtDNA) inheritance has previously been documented for the blue mussel (Mytilus edulis). Female mussels inherit their mtDNA solely from their mother while males inherit mtDNA from both their mother and their father. In males, the paternal mtDNA is preferentially amplified so that the male gonad is highly enriched for the paternal mtDNA that is then transmitted from fathers to sons. We demonstrate that this mode of mtDNA inheritance also operates in the closely related species M. galloprovincialis and M. trossulus. The evolutionary relationship between the male and female mtDNA lineages is estimated by phylogenetic analysis of 455 nucleotides from the large subunit ribosomal RNA gene. We have found that the male and female lineages are highly divergent; the divergence of these lineages began prior to the speciation of the three species of blue mussels. Further, the separation between the male and female lineages is estimated to have occurred between 5.3 and 5.7 MYA.   相似文献

(1) The pattern of absolute atid relative gonad growth during the life-span and the seasonal gotiad cycle were determined from monthly samples of chub taken from two tributaries of the Herefordshire Wye. (2) A marked change in the pattern of absolute gonad growth was thought to indicate the attainment of sexual maturity. This was found to occur synchronously with corresponding changes in the pattern of body growth described previously and was in agreement with the age at which a seasonal gonad cycle was detectable. (3) Sexual maturity was attained during the eighth year of life (7+) in female chub and in males from the Afon Llynfi, but most males from the River Lugg matured a year earlier. (4) The chub gonads continued to increase in relative size after the attainment of sexual maturity. This was more evident in female chub. (5) Seasonal changes in the gonad condition and weight were followed. Gonad maturation occurred during spring and the gonad mass increased rapidly just prior to spawning. The rabbit is an attractive species for the study of gonad differentiation because of its 31-day long gestation, the timing of female meiosis around birth and the 15-day delay between gonadal switch and the onset of meiosis in the female. The expression of a series of genes was thus determined by qPCR during foetal life until adulthood, completed by a histological analysis and whenever possible by an immunohistological one. Interesting gene expression profiles were recorded. Firstly, the peak of SRY gene expression that is observed in early differentiated XY gonads in numerous mammals was also seen in the rabbit, but this expression was maintained at a high level until the end of puberty. Secondly, a peak of aromatase gene expression was observed at two-thirds of the gestation in XX gonads as in many other species except in the mouse. Thirdly, the expression of STRA8 and DMC1 genes (which are known to be specifically expressed in germ cells during meiosis) was enhanced in XX gonads around birth but also slightly and significantly in XY gonads at the same time, even though no meiosis occurs in XY gonad at this stage. This was probably a consequence of the synchronous strong NANOS2 gene expression in XY gonad. In conclusion, our data highlighted some rabbit-specific findings with respect to the gonad differentiation process.  相似文献

Levels of prostaglandins E(2) and F(2alpha) were measured in male and female gonadal portions of the functional hermaphroditic scallop Argopecten purpuratus, at the time of spawning. For these measurements, mature scallops were stimulated to initiate their chain of spawning events, and prostaglandins (PGs) were analyzed in samples before stimulation, when animals appeared stimulated to release sperm, during the release of sperm, when animals appeared stimulated to release oocytes, when most of the oocytes had been released, and 24 hr after the end of spawning. The experiment was run twice, once in winter and once in spring. An additional in vitro experiment was carried out to analyze activity of the monoamines dopamine and serotonin on the levels of these prostaglandins. For this, minced tissue of the female portion of the gonad was incubated for different time periods with each of the amines at 10(-5) M. PGs were determined in the tissue samples by radioimmunoanalysis. The results showed that the amounts of the PGs increased significantly in both parts of the gonad during the global process of spawning and decreased when it had been completed. Incubation of minced gonad with the amines affected the time course of PGs variations, i.e., the effect of these compounds depended on the incubation time used. The results support a model for the regulation of spawning which assumes the occurrence of a stimulus which is detected by receptors, processed by nerve cells, and sent to the gonad where intermediation by amines (changing the time course of the process) induces liberation of gametes, in some way modulated by prostaglandins.  相似文献

A temperature-sensitive mutation, isx-1(hc17), is reported in the nematode Caenorhabditis elegans which alters the sexual phenotypes of both genotypic sexes. At the restrictive temperature, XX animals are functionally female rather than hermaphroditic due to the absence of spermatogenesis, and XO animals develop as intersexes. These intersexes have normal male head and tail structures and exhibit some mating behavior, but possess hermaphrodite-like gonads which produce no sperm and usually contain a few oocytes. An abortive vulva is usually present and evidence is presented which suggests that the formation of the vulva by the hypodermis is induced by the underlying gonad. The direct effects of the mutation are confined to the descendants of four primordial gonad cells. Gametogenesis and gonad sheath development do not seem to be tightly coupled and are shown to differ in their responses to X-chromosome dosage. The interaction of the intersex mutation with mutant alleles of two transformer genes tra-1 and tra-2 is discussed and a model for the action of these genes in gonad development and sex determination is proposed.  相似文献

Abstract. The development and sexual differentiation of gonads in female rat embryos and fetuses between the ages of 11 and 17 days was studied by immunocytochemical analysis of intermediate filament proteins and laminin by light and electron microscopy. In the 11-day-old pregonadal embryo, the surface epithelial cells in the ventral cortex of the mesonephros contained desmin but not cytokeratin or vimentin. The development of the gonad began on the following day by proliferative growth of the mesonephric surface cells, which like the subepithelial cells soon expressed vimentin in addition to desmin. The differentiation continued by formation of separate epithelial cell clusters, which joined into cords, irregular in shape and size. Desmin disappeared from the cord cells and cytokeratins appeared while vimentin remained in all somatic cell types. Desmin was especially abundant in some stromal cells adjacent to the epithelial tissues. After the segration of the basic ovarian tissues, vimentin and desmin decreased and cytokeratins appeared in the surface epithelial cells. New changes in cytokeratin expression appeared with the differentiation of the embryonic cords in a sex-specific manner with gradual decrease of reactivity for cytokeratin 18. No immunoreaction to the neurofilament proteins was found at the present ages, and the germ cells were negative for intermediate filaments. The results show that desmin is expressed in several primitive ovarian and mesonephric cells even though they are not myogenic. The sexual differences emerge after the incipient formation of the genetically female gonad, as different organization of the internal epithelial tissue with different timing of changes in intermediate filament proteins when compared with the male gonad.  相似文献

This study presents evidence, using sequences of ribosomal 16S and COI mtDNA, for the presence of two mitochondrial genomes in Perumytilus purpuratus. This may be considered evidence of doubly uniparental mtDNA inheritance. The presence of the two types of mitochondrial genomes differentiates females from males. The F genome was found in the somatic and gonadal tissues of females and in the somatic tissues of males; the M genome was found in the gonads and mantle of males only. For the mitochondrial 16S region, ten haplotypes were found for the F genome (nucleotide diversity 0.004), and 7 haplotypes for the M genome (nucleotide diversity 0.001), with a distance Dxy of 0.125 and divergence Kxy of 60.33%. For the COI gene 17 haplotypes were found for the F genome (nucleotide diversity 0.009), and 10 haplotypes for the M genome (nucleotide diversity 0.010), with a genetic distance Dxy of 0.184 and divergence Kxy of 99.97%. Our results report the presence of two well-differentiated, sex-specific types of mitochondrial genome (one present in the male gonad, the other in the female gonad), implying the presence of DUI in P. purpuratus. These results indicate that care must be taken in phylogenetic comparisons using mtDNA sequences of P. purpuratus without considering the sex of the individuals.  相似文献

分析家蚕Bombyx mori雌雄生殖腺细胞蛋白质,有利于发现性别分化相关的功能蛋白质,探讨生殖腺发育相关基因的表达调控机理。本研究利用蛋白质双向凝胶电泳和图像分析技术,分析家蚕蛹期第2天的雌雄生殖腺细胞蛋白质。结果表明: 在雄蚕生殖腺蛋白质电泳图谱中共检测到435个蛋白斑点,其中特异性蛋白斑点73个,占总蛋白斑点数的16.8%;雌蚕生殖腺的电泳图谱中有417个蛋白斑点,其中特异性蛋白斑点55个,占总蛋白斑点数的13.2%。雌雄能匹配的蛋白斑点有362对,匹配率达85.0%。  相似文献

The objective of this study was to determine the sexual pattern of the Indian dascyllus Dascyllus carneus . After an initially undifferentiated state, gonads of D. carneus developed an ovarian lumen and primary growth stage oocytes, and subsequently cortical-alveolus stage oocytes. From ovaries with cortical-alveolus stage oocytes and from more developed ovaries, some gonads redifferentiated into testes. From a sample of 163 individuals, two had a gonad containing degenerating vitellogenic oocytes and proliferating spermatogenic tissue, nine had a gonad containing degenerating cortical-alveolus stage oocytes and spermatogenic tissue, and five had a gonad with degenerating primary growth stage oocytes and spermatogenic tissue. The size of these individuals overlapped greatly with the size range of mature females, suggesting that at least in some individuals, redifferentiation toward a testis occurred after spawning as females. This indicates that D. carneus is a functional, diandric protogynous hermaphrodite. Removal of a dominant male(s) did not induce a sex change in any of the ranking females in the laboratory and field groups. There was no difference in the number of chases and signal jumps performed by the ranking female between control and experimental field groups, or before and after removal of the male. However, the sizes of the ranking females were at or beyond the size range of individuals with a mixed-stage gonad, suggesting that the developmental window for female-to-male sex change may not be open ended. In 41 of 43 field groups, in which sex of fish was determined histologically or by the shape of the urogenital papilla, one to several highest size ranks were occupied by males, followed by one to several females. Mature males, however, were not limited to the highest ranks and occurred at various lower size ranks within groups. Individuals with a mixed-stage gonad also occupied various size ranks within groups.  相似文献

Pupae of the olive fruit fly, Dacus oleae (Gmelin) 1 to 2 days before adult emergence were irradiated with the suitable sterilizing dose of 80 Gy gamma rays. At intervals of 5, 10, 15, 20, 25 and 30 days after adult emergence, anatomical and biometrical studies were performed to determine the extent of recovery of D. oleae gonads during one month of adult life. There were some indications of gonad recovery after two weeks. This recovery was observed as a decrease in the percentage deviation from the corresponding controls of 20-day-old adult gonad (especially those of males). Generally, female gonads are more sensitive to gamma-rays than those of males.  相似文献
