HDACs(Histone deacetylase)家族蛋白质负责组蛋白H3K4和H4K19脱乙酰化,并参与植物生长和应激反应的信号转导过程。茉莉酮酸酯Jasmonates(JA)是一种重要的天然植物激素,不仅调节植物生长和发育而且还参与植物对多种逆境胁迫响应的信号转导和调控过程。但是,HDACs在植物中参与JA信号转导的具体机制目前还不是很清楚。该研究以HDA19(Histone deacetylase 19)为对象,探讨了HDACs在植物JA信号转导中的功能和作用。结果表明:HDA19的T-DNA插入纯合突变体在JA处理条件下没有出现明显的JA根长反应。在相同处理条件下,hda19的不同突变体株系与相同生态型背景的野生型植株(WT)花色素苷含量无显著差异,但下游JAZ1、VSP1等JA信号通路的标记基因都显著上调表达。同时,hda19相比于WT对真菌Botrytis cineara的抗性显著增强,且hda19中下游基础防御标记基因PDF1.2、Thi2.1、ERF1等的表达水平显著高于WT。基于上述研究结果,该研究认为HAD19通过JA信号通路负调控拟南芥对真菌B.cineara的防御反应。  相似文献

以云南省瑞丽市勐秀林场扦插种植的薇甘菊为试材,采用液相色谱串联质谱(LC-MS/MS)技术对花芽未分化期和花序原基分化期花芽中的生长素(IAA)、赤霉素(GA)、脱落酸(ABA)、反式玉米素(tZ)、异戊烯腺嘌呤(IP)、1-氨基环丙烷羧酸(ACC)、茉莉酸(JA)和水杨酸(SA)含量进行定量分析,同时基于转录组基因功能注释数据对内源激素合成、代谢及信号转导途径相关基因进行表达分析,以探讨不同内源激素对薇甘菊花芽形成的调控作用,以及内源激素合成和信号转导途径相关基因调控薇甘菊花芽分化的机制,为后期通过外源激素调控薇甘菊内源激素水平的方式来控制薇甘菊的有性繁殖提供理论和技术支持。结果表明:(1)薇甘菊未分化期花芽中GA15、GA19、GA20、GA24、IAA、ABA和ETH含量低于花序原基分化期,而未分化期花芽中两种细胞分裂素tZ和IP含量显著高于花序原基分化期。(2)基于RNA-seq测序结果,在薇甘菊两个花芽分化时期共获得7 116个差异表达基因(DEGs),其中上调3 907个,下调3 209个。(3)在内源激素合成方面,参与GA15、GA19、GA20、GA24、IAA、ABA和ACC合成的大量DEGs在花序原基分化期上调表达,这与它们在薇甘菊花序原基分化期的高含量趋势相一致;参与IAA合成的YUCCA基因家族和ACC合成的ACS基因在花序原基分化期的高表达也可能参与促进薇甘菊花芽分化。(4)在植物激素转导途径中,在花序原基分化期,生长素信号转导途径通过AUX/IAA(gene-E3N88_07743)的下调表达和ARF(gene-E3N88_41119)的上调表达,乙烯信号转导途径通过ERF(gene-E3N88_41547)的上调表达,赤霉素信号转导途径通过GID1(gene-E3N88_19448)基因的上调表达,细胞分裂素信号转导途径通过B-ARR(gene-E3N88_28086)和A-RRR(gene-E3N88_40764)基因的下调表达,脱落酸途径通过AREB(gene-E3N88_18558)基因的上调表达,茉莉酸信号转导途径通过JAZ(gene-E3N88_05628)的上调表达和MYC2(gene-E3N88_32405)的下调表达来调控薇甘菊花芽分化。研究发现,高水平的GA15、GA19、GA20、GA24、IAA、ABA和ACC有利于薇甘菊的花芽分化;薇甘菊在花芽分化过程中通过改变不同种类内源激素合成、代谢基因的表达来调控激素浓度,而激素又通过信号转导途径引起下游基因的表达,进而调控薇甘菊的花芽分化。  相似文献

Although a wealth of information is available regarding resistance induced by plant growth-promoting rhizobacteria (PGPR), not much is known about plant growth-promoting fungi (PGPF). Hence, the goal of the present research was to provide more information on this matter. In Arabidopsis thaliana L., root colonizing PGPF Penicillium sp. GP16-2 or its cell free filtrate (CF) elicited an induced systemic resistance (ISR) against infection by Pseudomonas syringae pv. tomato DC3000 (Pst), leading to a restriction of pathogen growth and disease development. We demonstrate that signal transduction leading to GP16-2-mediated ISR requires responsiveness to JA and ET in a NPR1-dependent manner, while CF-mediated ISR shows dispensability of SA, JA, ET and NPR1-dependent signaling (at least individually). In addition, root colonization by GP16-2 is not associated with a direct effect on expression of known defense-related genes, but potentiates the activation of JA/ET-inducible ChitB, which only becomes apparent after infection by Pst. However, CF-mediated ISR was partly associated with the direct activation of marker genes responsive to both SA and JA/ET signaling pathways and partly associated with priming, leading to activation of JA-/ET-inducible ChitB and Hel genes. These suggest that CF may contain one or more elicitors that induce resistance by way where at least SA, JA and ET may play a role in defense signaling in Arabidopsis. Therefore, defense gene changes and underlying signaling pathways induced by Penicillium sp. GP16-2 root colonization and its CF application are not the same and only partially overlap.  相似文献

In response to insect attack and mechanical wounding, plants activate the expression of genes involved in various defense-related processes. A fascinating feature of these inducible defenses is their occurrence both locally at the wounding site and systemically in undamaged leaves throughout the plant. Wound-inducible proteinase inhibitors (PIs) in tomato (Solanum lycopersicum) provide an attractive model to understand the signal transduction events leading from localized injury to the systemic expression of defense-related genes. Among the identified intercellular molecules in regulating systemic wound response of tomato are the peptide signal systemin and the oxylipin signal jasmonic acid (JA). The systemin/JA signaling pathway provides a unique opportunity to investigate, in a single experimental system, the mechanism by which peptide and oxylipin signals interact to coordinate plant systemic immunity. Here we describe the characterization of the tomato suppressor of prosystemin-mediated responses8 (spr8) mutant, which was isolated as a suppressor of (pro)systemin-mediated signaling. spr8 plants exhibit a series of JA-dependent immune deficiencies, including the inability to express wound-responsive genes, abnormal development of glandular trichomes, and severely compromised resistance to cotton bollworm (Helicoverpa armigera) and Botrytis cinerea. Map-based cloning studies demonstrate that the spr8 mutant phenotype results from a point mutation in the catalytic domain of TomLoxD, a chloroplast-localized lipoxygenase involved in JA biosynthesis. We present evidence that overexpression of TomLoxD leads to elevated wound-induced JA biosynthesis, increased expression of wound-responsive genes and, therefore, enhanced resistance to insect herbivory attack and necrotrophic pathogen infection. These results indicate that TomLoxD is involved in wound-induced JA biosynthesis and highlight the application potential of this gene for crop protection against insects and pathogens.  相似文献

The nuclear protein ETHYLENE INSENSITIVE2 (EIN2) is a central component of the ethylene signal transduction pathway in plants, and plays an important role in mediating cross-links between several hormone response pathways, including abscisic acid (ABA). ABA mediates stress responses in plants, but there is no report on the role of EIN2 on plant response to salt and osmotic stresses. Here, we show that EIN2 gene regulates plant response to osmotic and salt stress through an ABA-dependent pathway in Arabidopsis. The expression of the EIN2 gene is down-regulated by salt and osmotic stress. An Arabidopsis EIN2 null mutant was supersensitive to both salt and osmotic stress conditions. Disruption of EIN2 specifically altered the expression pattern of stress marker gene RD29B in response to the stresses, but not the stress- or ABA-responsive genes RD29A and RD22, suggesting EIN2 modulates plant stress responses through the RD29B branch of the ABA response. Furthermore, disruption of EIN2 caused substantial increase in ABA. Lastly, our data showed that mutations of other key genes in ethylene pathway also had altered sensitivity to abiotic stresses, indicating that the intact ethylene may involve in the stress response. Taken together, the results identified EIN2 as a cross-link node in ethylene, ABA and stress signaling pathways, and EIN2 is necessary to induce developmental arrest during seed germination, and seedling establishment, as well as subsequent vegetative growth, thereby allowing the survival and growth of plants under the adverse environmental conditions. 茉莉酸是植物伤反应的特异激素, 在植物伤反应中具有核心作用, 其下游调控机制已经比较清晰。在番茄(Lycopersicon esculentum)伤反应中, 系统素和茉莉酸协同启动相关基因的表达, 行使系统性防御功能。拟南芥(Arabidopsis thaliana)信号肽是新发现的一类信号物质, 可以激活植物的初始免疫反应, 但其在伤反应中的作用机制有待进一步研究。脱落酸位于茉莉酸上游, 单独或者协同茉莉酸参与植物的防御反应。另外, 植物中还存在以核糖核酸酶为代表的且不依赖于茉莉酸的伤反应信号转导途径。该文对植物伤反应的防御机制和信号转导做了详细概述。  相似文献

To elucidate the mechanism of irradiance-dependent adjustments in the chlorophyll antenna size of photosynthesis, we addressed the regulation of expression of genes encoding a variety of chlorophyll biosynthesis enzymes and that of the Lhcb genes in the model organism Dunaliella salina. Among the chlorophyll biosynthesis enzymes tested, only the chlorophyll a oxygenase (CAO) gene responded to changes in the level of irradiance with substantial mRNA level and kinetics of change that were similar to those of the Lhcb genes. Evidence is presented for the operation of a cytosolic signal transduction pathway for the rapid (order of minutes) regulation of both CAO and Lhcb gene expression by irradiance. Inhibitor studies and transient activation of Ca2+-dependent kinase suggested phopholipase-C activation to Ca2+ release, and activation of a specific Ca2+/CaM-dependent protein kinase in this cytosolic signal transduction pathway. The redox state of the plastoquinone pool also serves to regulate CAO and Lhcb gene expression on a slower time scale (hours) and probably serves as a plastidic-origin signal that acts coordinately with the cytosolic signal transduction pathway. It is proposed that irradiance-dependent adjustments in the chlorophyll antenna size occur by coordinate regulation of CAO and Lhcb gene expression via two distinct signal transduction pathways in photosynthetic organisms.  相似文献

The simple gaseous compound ethylene (ET) has long been recognized as a common component of plant responses to insect feeding and pathogen attack. However, it is presently uncertain whether it plays a role in host–plant resistance to piercing–sucking insects such as aphids. In these experiments, we investigated the expression of key ET‐associated genes in resistant and susceptible interactions in two model systems: the tomato‐MiMacrosiphum euphorbiae (Thomas) (Hemiptera: Aphididae: Macrosiphini) system and the melon‐virus aphid transmission gene (Vat)‐Aphis gossypii Glover (Hemiptera: Aphididiae: Aphidini) system. We examined expression patterns of genes associated with ET synthesis, perception, signal transduction, and downstream response. When compared with control plants, plants infested with aphids showed marked differences in gene expression. In particular, ET signaling pathway genes and downstream response genes were highly upregulated in the resistant interaction between A. gossypii and Vat+, indicating ET may play a role in Vat‐mediated host–plant resistance. A key integrator between the ET and jasmonic acid pathways (Cm‐ERF1) showed the strongest response.  相似文献

Jasmonates (JAs), which include jasmonic acid and its cyclopentane derivatives are synthesized from the octadecanoid pathway and widely distributed throughout the plant kingdom. JAs modulate the expression of numerous genes and mediate responses to stress, wounding, insect attack, pathogen infection, and UV damage. They also affect a variety of processes in many plant developmental processes. The JA signal pathway involves two important events: the biosynthesis of JA and the transduction of JA signal. Several important Arabidopsis mutants in jasmonate signal pathway were described in this review.  相似文献

Jasmonates (JAs) and salicylic acid (SA) are plant hormones that play pivotal roles in the regulation of induced defenses against microbial pathogens and insect herbivores. Their signaling pathways cross-communicate providing the plant with a regulatory potential to finely tune its defense response to the attacker(s) encountered. In Arabidopsis thaliana, SA strongly antagonizes the jasmonic acid (JA) signaling pathway, resulting in the downregulation of a large set of JA-responsive genes, including the marker genes PDF1.2 and VSP2. Induction of JA-responsive marker gene expression by different JA derivatives was equally sensitive to SA-mediated suppression. Activation of genes encoding key enzymes in the JA biosynthesis pathway, such as LOX2, AOS, AOC2, and OPR3 was also repressed by SA, suggesting that the JA biosynthesis pathway may be a target for SA-mediated antagonism. To test this, we made use of the mutant aos/dde2, which is completely blocked in its ability to produce JAs because of a mutation in the ALLENE OXIDE SYNTHASE gene. Mutant aos/dde2 plants did not express the JA-responsive marker genes PDF1.2 or VSP2 in response to infection with the necrotrophic fungus Alternaria brassicicola or the herbivorous insect Pieris rapae. Bypassing JA biosynthesis by exogenous application of methyl jasmonate (MeJA) rescued this JA-responsive phenotype in aos/dde2. Application of SA suppressed MeJA-induced PDF1.2 expression to the same level in the aos/dde2 mutant as in wild-type Col-0 plants, indicating that SA-mediated suppression of JA-responsive gene expression is targeted at a position downstream of the JA biosynthesis pathway.  相似文献

EIN2 (ethylene insensitive 2) is a very important component in the ethylene signal transduction pathway. Recently, the genomic DNA and full-length cDNA of LeEIN2 (tomato EIN2) have been isolated in our laboratory. To reveal the function of LeEIN2, transgenic tomato plants with reduced expression levels of LeEIN2 were produced. The fruit ripening and expressions of ripening-related genes encoding polygalacturonase and TomLoxB were inhibited in the LeEIN2-silenced transgenic plants compared to the wild-type Ailsa Craig. In the seedling ethylene response assay, the transgenic tomato plants with reduced LeEIN2 expression exhibited ethylene insensitivity. These results indicate that LeEIN2 plays a critical role in regulating tomato fruit ripening and is a positive regulator in ethylene signal transduction pathway.  相似文献
