The effect of root colonization by Glomus mosseae on the qualitative and quantitative pattern of essential oils (EO) was determined in three oregano genotypes (Origanum sp.). To exclude a simple P-mediated effect through mycorrhization the effect of P application to plants on the EO accumulation was also tested. In two genotypes the leaf biomass was increased through mycorrhization. Root colonization by the arbuscular mycorrhizal fungus (AMF) did not have any significant effect on the EO composition in oregano; however, in two genotypes the EO concentration significantly increased. As EO levels in P-treated plants were not enhanced, we conclude that the EO increase observed in mycorrhizal oregano plants is not due to an improved P status in mycorrhizal plants, but depends directly on the AMF–oregano plant association.  相似文献

Summary ‘Mendocino’ oregano (Origanum vulgare × applii) cultivated in Argentina has been subject to progressive yield loss as a consequence of continuous vegetative propagation. A method of reducing damage to the shoot tip during sterilization procedures for micropropagation is proposed. Single shoot cuttings are less sensitive to disinfecting substances than green tips. Meristems taken from young plantlets that grew in vitro produced less oxidation during the culture than those dissected immediately after disinfecting from plants grown in the field. This work describes the effects of the growth regulators benzyladenine (BA) and naphthaleneacetic acid (NAA) at various concentrations and combinations on micropropagation of O. vulgare × applii. Treatment with 0.28 μM BA and 0.53 μM NAA gave greatest effiency (mean: 22.2 nodes per plantlet). After 60 d 100% of rooted plantlets could be formed per explant under optimum conditions.  相似文献

The aim of this study was to find an alternative to synthetic fungicides currently used in the control of devastating oomycete pathogen Phytophthora infestans, causal agent of late blight disease of tomato. Antifungal activities of essential oils obtained from aerial parts of aromatic plants such as oregano (Origanum syriacum var. bevanii), thyme (Thymbra spicata subsp. spicata), lavender (Lavandula stoechas subsp. stoechas), rosemary (Rosmarinus officinalis), fennel (Foeniculum vulgare), and laurel (Laurus nobilis), were investigated against P. infestans. Both contact and volatile phase effects of different concentrations of the essential oils used were determined by using two in vitro methods. Chemical compositions of the essential oils were also determined by GC-MS analysis. Major compounds found in essential oils of thyme, oregano, rosemary, lavender, fennel and laurel were carvacrol (37.9%), carvacrol (79.8), borneol (20.4%), camphor (20.2%), anethole (82.8%) and 1,8-cineole (35.5%), respectively. All essential oils were found to inhibit the growth of P. infestans in a dose-dependent manner. Volatile phase effect of oregano and thyme oils at 0.3 μg/ml air was found to completely inhibit the growth of P. infestans. Complete growth inhibition of pathogen by essential oil of fennel, rosemary, lavender and laurel was, however, observed at 0.4–2.0 μg/ml air concentrations. For the determination of the contact phase effects of the tested essential oils, oregano, thyme and fennel oils at 6.4 μg/ml were found to inhibit the growth of P. infestans completely. Essential oils of rosemary, lavender and laurel were inhibitory at relatively higher concentrations (12.8, 25.6, 51.2 μg/ml respectively). Volatile phase effects of essential oils were consistently found to be more effective on fungal growth than contact phase effect. Sporangial production was also inhibited by the essential oil tested. Light and scanning electron microscopic (SEM) observation on pathogen hyphae, exposed to both volatile and contact phase of oil, revealed considerable morphological alterations in hyphae such as cytoplasmic coagulation, vacuolations, hyphal shrivelling and protoplast leakage.  相似文献

According to published data, it is well known that essential oils from plants possess antimicrobial activity against a wide range of pathogens, including Salmonella strains. The influence of the administration of essential oils from oregano (Origanum vulgare) and thyme (Thymus vulgaris) as well as Lactobacillus fermentum on crop, caecum, liver and spleen colonization by Salmonella enterica var. Düsseldorf in chicks was investigated in this study. For the experiment, one hundred 1-day old chicks were used, and they were divided into four groups: (i) untreated (C); (ii) treated with L. fermentum (L); (iii) treated with a mixture of oregano and thyme essential oils (OT); and (iv) treated with a combination of L. fermentum and a mixture of oregano and thyme essential oils (LOT). Essential oils from oregano and thyme were mixed with commercial poultry diet, which was offered ad libitum to chicks in appertaining groups. L. fermentum was added daily to drinking water. After 3 days all the chicks were challenged orally with S. enterica var. Düsseldorf. The crops, ceca, spleens and livers of the birds were examined for S. enterica var. Düsseldorf colonization 5 days after the challenge. Our results showed that a combined administration of L. fermentum and essential oils (oregano and thyme) in group with combined application of essential oils and lactobacillus strain reduced the percentage of colonized crops and ceca when compared to the control group without any treatment.  To obtain homozygous genotypes of apple, we have induced haploid development of either the female or the male gametes by parthenogenesis in situ and anther culture, respectively. Of the shoots obtained, which were mainly of a non-haploid nature, some could be derived from fertilised egg cells or from sporophytic anther tissue. In order to select the shoots having a true haploid origin, and thus homozygotes, we decided to use the single multi-allelic self-incompatibility gene as a molecular marker to discriminate homozygous from heterozygous individuals. The rationale behind this approach was that diploid apple cultivars contain 2 different alleles of the S-gene and therefore the haploid induced shoots obtained from them should have only one of the alleles of the single parent. The parental cultivars used were ‘Idared’ (parthenogenesis in situ) and ‘Braeburn’ (androgenesis), and their S-genotypes were known, except for 1 of the ‘Braeburn’S-alleles. To stimulate parthenogenetic development ‘Idared’ styles were pollinated with irradiated ‘Baskatong’ pollen, the S-alleles of the latter (2n) cultivar were also unknown. The cloning and sequence analysis of these 3 unidentified S-alleles, 1 from ‘Braeburn’ and 2 from ‘Baskatong’ is described, and we show that they correspond to the S 24 -, S 26 - and S 27 -alleles. We have optimised a method for analysis of the S-alleles of ‘Idared/Baskatong’- or ‘Braeburn’-derived in vitro plant tissues and have shown that this approach can be applied for the screening of the in vitro shoots for their haploid origin. There are some recommendations for the use of species diversity indices in a paper recently published in this journal by M. Ohsawa on the species richness and composition of weevils in five forest types in the middle region of Japan. Because several factors, such as small sample size, calculation of Simpson’s diversity index by the use of the original equation of Simpson’s measure of concentration D, and a weak point in the Shannon–Wiener diversity index H′, may have led to biased estimates, I recalculated these indices using combined species diversity values of the five forest types. As the general tendencies of 1−D and H′ values calculated here were similar to those in Ohsawa’s paper, there is no need to propose any change to his view. However, these recalculated diversity indices indicated that they are values which are more suitable for use. It should be noted that the characteristics and weak point associated with the diversity indices need to be taken into account in future studies.  相似文献

Phytophthora fragariae var. fragariae is the causal agent of red stele (red core) root rot in strawberry (Fragaria spp.). The inheritance of resistance to one isolate of this fungus was studied in 12 segregating populations of F.×ananassa derived from crosses between four resistant cultivars (‘Climax’, ‘Redgauntlet’, ‘Siletz’, and ‘Sparkle’) and three susceptible cultivars (‘Blakemore’, ‘Glasa’, and ‘Senga’ Sengana’). The analysis clearly supports the hypothesis of a single segregating dominant resistance gene. It is proposed that this gene be designated Rpf2. Sulfur is the oldest and most widely used fungicide in the vineyards of California, where it is used for control of powdery mildew (Uncinula necator [Schw.] Burr). For decades, sulfur use has been associated with outbreaks of Tetranychus pacificus McGregor (Acari: Tetranychidae) on cultivated grapes in the San Joaquin Valley. I undertook large-scale field studies to test this association, to evaluate the impact of sulfur on Galendromus occidentalis (Nesbit) (Acari: Phytoseiidae), a major predator of T. pacificus, and to determine if timing of sulfur applications with respect to grape bloom has an impact on T. pacificus density. The studies took place in a 32 ha vineyard in Fresno County, and all fungicide applications were made with commercial-scale equipment. In 1998 a ‘high sulfur’ treatment, a combination of wettable sulfur and sulfur dust, was compared to ‘low sulfur,’ in which demethylation inhibitor (DMI) fungicides partially substituted for sulfur. In 1999 treatments were ‘sulfur,’ ‘DMI,’ ‘sulfur pre-bloom’ (here sulfur was applied prior to grape bloom, in late May, and then DMIs were applied until mid-season) and ‘sulfur post-bloom’ (the reverse of ‘sulfur pre-bloom’). In each year, the T. pacificus population increase came after the end of fungicide applications, and results clearly show a relationship between sulfur use and T. pacificus density. In 1998, mean T. pacificus density was 2.7 times higher and mean G. occidentalis density 2.5 times higher in ‘high sulfur’ compared to ‘low sulfur.’ In 1999, the highest T. pacificus counts were in the ‘sulfur’ and ‘sulfur pre-bloom’ treatments, 4.8 times higher than ‘sulfur post-bloom’ and 2 times higher than ‘DMIs.’ Density of G. occidentalis was 2.3 times as high in ‘sulfur’ or ‘sulfur pre-bloom’ than ‘DMIs.’ The predator/prey ratio was not significantly different among treatments in 1998, but in 1999 it was highest in the ‘sulfur pre-bloom’ treatment. In 1999, density of Homeopronematus anconai (Baker) (Acari: Tydeidae) was 2.7 times higher in ‘sulfur pre-bloom’ compared to ‘sulfur,’ and higher by 2.7 times in ‘DMI’ compared to ‘sulfur post-bloom,’ suggesting a negative effect of sulfur on this tydeid. These results do not support the hypotheses that the cause of the increase in T. pacificus density is due to negative effects of sulfur on phytoseiids or tydeids. Rather, it appears that a plant-based explanation is likely, first, because of the differences in pre-bloom versus post-bloom sulfuring, and second, because of the long lag time between the end of the sulfur applications and the corresponding increase in spider mite density.  相似文献

Some new field observations on West African pottos are presented and compared with previously published information on substrate use and diet. This evidence suggests that the predominant means by which pottos find food is by carefully searching branches in the forest canopy with their noses and that gums and slow-moving invertebrates found on branch surfaces are major components of their diet. When fruits are scarce, gums may be the potto’s chief source of energy. It is argued that the potto’s deliberate locomotion and some of its anatomical peculiarities are related primarily to this diet and foraging behavior, rather than to concealment from predators or the capture of birds, two factors previously suggested to have been important in the evolution of the potto’s locomotor technique. The potto’s niche is compared to that of other lorisid primates and is found to resemble closely that of Galago crassicaudatusin several respects.  相似文献

In 1749, Linnaeus presided over the dissertation “Oeconomia Naturae,” which argued that each creature plays an important and particular role in nature’s economy. This phrase should be familiar to readers of Darwin, for he claims in the Origin that “all organic beings are striving, it may be said, to seize on each place in the economy of nature.” Many scholars have discussed the influence of political economy on Darwin’s ideas. In this paper, I take a different tack, showing that Darwin’s idea of an economy of nature stemmed from the views of earlier naturalists like Linnaeus and Lyell. I argue, in the first section of the paper, that Linnaeus’ idea of oeconomia naturae is derived from the idea of the animal economy, and that his idea of politia naturae is an extension of the idea of a politia civitatis. In the second part, I explore the use of the concept of stations in the work of De Candolle and Lyell – the precursor to Darwin’s concept of places. I show in the third part of the paper that the idea of places in an economy of nature is employed by Darwin at many key points in his thinking: his discussion of the Galapagos birds, his reading of Malthus, etc. Finally, in the last section, I demonstrate that the idea of a place in nature’s economy is essential to Darwin’s account of divergence. To tell his famous story of divergence and adaptation, Darwin needed the economy of nature.  相似文献

Summary An inorganic (Bainsville) and an organic (Farnham) soil were hydrolyzed by continuous and stepwise hydrolysis with hot 3N HCl for 1, 2, 3, 4, 11, 15, 18 and 24 h and continuously with hot 6N HCl for 24 h. The following nitrogen forms were determined: total N, hydrolyzable-N, amino acid-N, amino sugar-N and ammonia-N. Proportions of ‘unknown’ N were computed from these data. Continuous hydrolysis with 3N HCl yielded more amino acid-N and less ‘unknown’ N than did stepwise hydrolysis with the same acid strength. But continuous hydrolysis for 24 h with 6N HCl produced more amino acid-N and less ‘unknown’ N than did hydrolysis with 3N HCl by either method. It was estimated that 33 and 54% of the total N in the inorganic and organic soil, respectively, was protein-N. The ‘unknown’ N in the inorganic and organic soil constituted 51 and 37% of the total N, respectively. From our work it appears that the ‘unknown’ N is not proteinaceous. It can be readily degraded chemically and microbiologically to NH3 and N-gases. More attention needs to be given to identifying the ‘unknown’ N which constitutes a large portion of the total soil-N. A more adequate knowledge of the chemical constitution of the ‘unknown’ soil N may lead to the development of technologies that will make more efficient use of the N in soils.  相似文献

At least three sources of resistance to the watermelon strain of Papaya ringspot virus (PRSV-W) have been identified in cucumber (Cucumis sativus L.) including: ’TMG-1’, an inbred line derived from the Taiwanese cultivar, ’Taichung Mou Gua’; ’Dina-1’, an inbred line derived from the Dutch hybrid ’Dina’; and the South American cultivar ’Surinam’. In this investigation we sought to determine the inheritance of resistance to PRSV-W in ’Dina-1’, the allelic relationships among the three sources of PRSV-W resistance, and the relationship between PRSV-W resistance and known resistances to other cucurbit potyviruses. Like ’Surinam’ and ’TMG-1’, resistance in ’Dina-1’ is controlled by a single gene. Despite differences in dominance vs recessive performance and patterns of virus accumulation, all three sources of resistance complemented each other. ’TMG-1’ and ’Dina-1’ also possess co-segregating, single-gene resistances to Zucchini yellow mosaic virus (ZYMV), Watermelon mosaic virus and Moroccan watermelon mosaic virus. Sequential inoculations and F3 family analysis indicated that resistance to PRSV-W completely co- segregated with resistance to ZYMV in ’TMG-1’. Although PRSV-W resistances are at the same locus in both ’TMG-1’ and ’Surinam’, ’Surinam’ is only resistant to PRSV-W, and progeny of ’TMG-1’×’Surinam’ were resistant to PRSV-W but susceptible to ZYMV. Susceptibility to ZYMV and resistance to PRSV-W in ’Surinam’ was not influenced by co-inoculation or sequential in- oculations of the two viruses. Collectively, the co- segregation of resistances to PRSV-W, ZYMV, WMV and MWMV in ’TMG-1’ (within 1 cM), allelism of PRSV-W resistances in ’TMG-1’ and ’Surinam’, and resistance to only PRSV-W in ’Surinam’, suggest that multiple potyvirus resistance in cucumber may be due to different alleles of a single potyvirus resistance gene with differing viral specificities, or that the multiple resistances are conferred by a tightly linked cluster of resistance genes, of which ’Surinam’ only possesses one member. Males of tuco-tucoCtenomys talarum Thomas, 1898 use particular burrow’s entrances to emit their territorial vocalization. Therefore we studied the internal structure of these entrances and the possible effect on the emission and propagation of airborne sounds. Externally, the burrow entrances used by tuco-tucos males to vocalize were characterized by the absence of sand mounds around their openings. Internally, most of the burrow’s entrances consisted of a main, relatively straight, tunnel of 30–40 cm length, with a diameter of 5.7–6.4 cm. After passing through the burrow’s entrance, the low-frequency components of an artificial signal played back inside the tunnel were not only less attenuated but also amplified (measured at 10–30 cm from the burrow opening). Therefore, the emission of territorial vocalizations inside the particular burrow’s entrances may be considered as a complex adaptative behavior, in which burrow structure improves the signal emission and propagation. Moreover, this work also showed thatC. talarum’s territorial vocalization seems to be adequate for long, inter-burrow communication, since its physical characteristics (high amplitude and low main frequency) are concomitant with the frequencies that are better transmitted in the natural habitat of this species of subterranean rodent.  相似文献

The fronds of Rumohra adiantiformis, also known as ‘7-weeks-fern,’ ‘leatherleaf’ or ‘samambaia-preta,’ are used worldwide as florists’ greenery. Costa Rica and the USA cultivate and export R. adiantiformis, whereas in South Africa and Brazil this trade is based on extractivism. In Brazil, R. adiantiformis is harvested in the Atlantic Forest biome; 50% of the production comes from the slopes of the ‘Serra Geral’ (state of Rio Grande do Sul), where some 2000 families harvest and trade R. adiantiformis as their main source of revenue. However, harvesting, trading and transporting wild ornamental species is illegal in the Atlantic Forest, This study evaluates R. adiantiformis extractivism in hilly areas of Rio Grande do Sul, from an ethnobotanical/ethnoecological perspective. The study reveals that emic perceptions on the sustainability of R. adiantiformis extractivism are in agreement with relevant ecological data. The regeneration areas on the slopes of the Serra Geral have the highest plant density ever reported for R. adiantiformis; its extractivism has low environment impact and generates significant income for local communities. Further studies of these areas are important in order to support neoextractivism as a suitable form of use of the state's biodiversity.  相似文献

Marker-assisted selection (MAS) offers quick and reliable prediction of the phenotypes of seedlings in large populations and thus opens new approaches for selection to breeders of apple (Malus x domestica Borkh.). The development of framework maps enables the discovery of genetic markers linked to desired traits. Although genetic maps have been reported for apple scion cultivars, none has previously been constructed for apple rootstocks. We report the construction of framework genetic maps in a cross between ‘M.9’ (‘Malling 9’) and ‘R.5’ (‘Robusta 5’) apple rootstocks. The maps comprise 224 simple sequence repeat (SSR) markers, 18 sequence-characterised amplified regions, 14 single nucleotide polymorphisms and 42 random amplified polymorphic DNAs. A new set of 47 polymorphic SSRs was developed from apple EST sequences and used for construction of this rootstock map. All 17 linkage groups have been identified and aligned to existing apple genetic maps. The maps span 1,175.7 cM (‘M.9’) and 1,086.7 cM (‘R.5’). To improve the efficiency of mapping markers to this framework map, we developed a bin mapping set. Applications of these new genetic maps include the elucidation of the genetic basis of the dwarfing effect of the apple rootstock ‘M.9’ and the analysis of disease and insect resistance traits such as fire blight (Erwinia amylovora), apple scab (Venturia inaequalis) and woolly apple aphid (Eriosoma lanigerum). Markers for traits mapped in this population will be of direct use to apple breeders for MAS and for identification of causative genes by map-based cloning.  相似文献

 Spot form of net blotch (SFNB) (Pyrenophora teres f maculata) is an economically damaging foliar disease of barley in many of the world’s cereal growing areas. The development of SFNB-resistant cultivars may be accelerated through the use of molecular markers. A screen for SFNB resistance in 96 lines identified four new sources of resistance, including a feed variety, ‘Galleon’, for which a fully mapped doubled haploid population was available. Segregation data indicated SFNB resistance was conferred by a single gene in the ‘Galleon’בHaruna Nijo’ cross, positioned on the long arm of chromosome 7H. This gene is designated Rpt4 and is flanked by the RFLP loci Xpsr117(D) and Xcdo673 at distances of 6.9 cM and 25.9 cM, respectively. The marker Xpsr117(D) was validated using another population segregating for Rpt4, correctly predicting SFNB resistance with more than 90% accuracy. Partial genomic and cDNA sequences of the RNase alleles S1, S7, S8 and Sf were obtained from Prunus dulcis cvs ’Ne Plus Ultra’, ’Ferragnes’ and ’Nonpareil’ 15–1, and IRTA Selection 12–2. Total DNA was extracted from leaves, and cDNA was prepared from total RNA extracted from styles. The partial cDNA sequences of the S1 allele from ’Ferragnes’, and the S7 and S8 alleles from ’Nonpareil’ 15–1, matched those reported in the literature for the alleles Sb, Sc and Sd respectively. The sequences of the S1, S7, S8 and Sf alleles found in genomic DNA contained introns of 562, 1,530, 2,208 and 689 bp respectively. The exon/intron splice junction sites of all alleles followed the GT/AG consensus sequence rule, and the sequences were found to be highly conserved. Self-compatibility has become the primary objective of most almond (Prunus amygdalus Batsch) breeding programmes in order to avoid the problems related to the gametophytic self-incompatibility system present in almond. The progeny of the cross ‘Vivot’ (S 23 S fa) × ‘Blanquerna’ (S 8 S fi) was studied because both cultivars share the same S f allele but have a different phenotypic expression: active (S fa) in ‘Vivot’ and inactive (S fi) in ‘Blanquerna’. In addition, the microscopic observation of pollen tube growth after self-pollination over several years showed an unexpected self-incompatible behaviour in most seedlings of this cross. The genotypes of this progeny showed that the S fi pollen from ‘Blanquerna’ was not able to grow down the pistils of ‘Vivot’ harbouring the S fa allele, confirming the active function of this allele against the inactive form of the same allele, S fi. As self-compatibility was observed in some S 8 S 23 and S 8 S fa individuals of this progeny, the S f haplotype may not always be linked to the expression and transmission of self-compatibility in almond, suggesting that a modifier locus may be involved in the mechanism of self-incompatibility in plants.  相似文献
