The suitability of capacitance measurements by the Bactometer Monitoring System for a quick differentiation between good, poor and moderate quality of cod fillets, was tested.The application of Brain Heart Infusion broth as growing medium for capacitance measurements gave very small differences between duplicate measurements, if any.Both at 20 and 30°C these measurements correlated very well with the results of conventional methods for the determination of psychrophilic and mesophilic count respectively. The correlation coefficients were -0.93 and-0.95.For raw fish like cod fillets, measurements at 20°C are to be preferred because bacteriological quality defects revealed at 20°C were masked at 30°C in some cases.  相似文献

Monitoring of microwave emission from aqueous solution of horseradish peroxidase (HRP) in the process of the enzyme functioning was carried out. For the monitoring, a system containing HRP, luminol and Н2О2 was employed. Microwave emission measurements were carried out in the 3.4-4.2 GHz frequency range using the active and passive modes (active-mode and passive-mode measurements). In the active mode, excitation of the solution in the pulsed electromagnetic field was accomplished. In the passive mode, no excitation was induced. It appears that the passive-mode measurements taken in the course of the peroxidase reaction in the enzyme system have shown a 0.5 °С increase of the microwave signal. Upon the active-mode measurements, taken in the same reaction conditions, the forced excitation of the solution has also led to the increase (by 2 °С) of the level of the microwave signal – i.e. to its 4-fold enhancement compared to the signal obtained in passive-mode measurements.  相似文献

The author analyses the sexual dimorphism of the innominate bone of Homo sapiens by seven measurements of angles and 15 measurements of distances. It appears that the dimorphism is very significant for the ischium, less for the ilium and is not significant for the pubis. People living on the banks of the Techa river were exposed to 90Sr in the early 1950s. Data obtained by radiochemical measurements of extracted permanent teeth, 90Sr autopsy measurements in bone and tooth samples, in vivo measurements of surface beta activity of the anterior teeth and whole-body counter (WBC) measurements of 90Sr in the skeleton have been analyzed. Surface beta activity measurements indicate a biological half-life of 90Sr of about 35 years in enamel. The WBC measurements have been performed since 1974 and a model for the age-dependent strontium retention in human bone has been used to extrapolate to previous time periods when the other measurement results were obtained. For the first decade after the intake, the ratio of the 90Sr concentrations in teeth and bones were found to decrease with age at the time of major intake, from about 10 for 1-year-old children to about 0.3 for adults. There was a considerable variability of individual data within each age group. For adults, the correlation between 90Sr in skeleton and teeth was not high at 0.47 according to radiochemical data for posterior teeth (molars and premolars) and 0.43 according to measurements of surface beta activity for anterior teeth. For children and adolescents there was no correlation between individual measurements in the skeleton and teeth. The absorbed dose in enamel due to 90Sr in dentine has been calculated by Monte Carlo simulations of the electron transport. The results are in agreement with EPR measurements of the absorbed dose in the enamel of persons exposed, mainly due to 90Sr ingestion. Received: 19 July 1999 / Accepted: 5 May 2000  相似文献

Interpretation of the electrical potential on the surface of plant roots   总被引:1,自引:1,他引:0  
The electrical potential difference (p.d.) between two points on the surface of a plant root is shown to be a measure of the difference between the transmembrane potentials at those two points. More precisely, it is shown that axial differences in electrical potential on the surface of the root, or within several tenths of millimeters of it in the rhizosphere, are primarily a result of axial differences in p.d. across the plasmalemma of cells in the cortex, with an additional small effect from axial differences in p.d. across the plasmalemma of cells in the stele. This conclusion results from a model of the root as a three-conductor electrical transmission line. The model requires the solution of a set of differential equations, but simple algebraic approximations are found to apply over a range of model parameters derived from published data. Given the predictions of the model, it follows that, for many research purposes, microelectrode measurements could be replaced with measurements of p.d. on the surface of the root. Such measurements offer substantial advantages over microelectrode measurements: they are non-invasive; they do not include the potential difference across the tonoplast; they measure a spatial average of many cells, not just a single cell; and the measurement is physically robust. Surface potential measurements do not, however, measure trans-membrane potential at one point, but measure differences in trans-membrane potential.  相似文献

Quantitative cell biology requires precise and accurate concentration measurements, resolved both in space and time. Fluorescence correlation spectroscopy (FCS) has been held as a promising technique to perform such measurements because the fluorescence fluctuations it relies on are directly dependent on the absolute number of fluorophores in the detection volume. However, the most interesting applications are in cells, where autofluorescence and confinement result in strong background noise and important levels of photobleaching. Both noise and photobleaching introduce systematic bias in FCS concentration measurements and need to be corrected for. Here, we propose to make use of the photobleaching inevitably occurring in confined environments to perform series of FCS measurements at different fluorophore concentration, which we show allows a precise in situ measurement of both background noise and molecular brightness. Such a measurement can then be used as a calibration to transform confocal intensity images into concentration maps. The power of this approach is first illustrated with in vitro measurements using different dye solutions, then its applicability for in vivo measurements is demonstrated in Drosophila embryos for a model nuclear protein and for two morphogens, Bicoid and Capicua.  相似文献

Truss analysis and length measurements were made on 168 striped red mullet Mullus surmuletus. Multivariate statistical analyses with principal component analysis and partial redundancy analysis (pRDA) were used on these measurements to evaluate the influence of maturity, sex and geographical area distribution on body shape. Truss measurements were important to quantify and discriminate changing body shape, presumably due to changing environmental conditions. Sexual dimorphism was not observed and juveniles could be distinguished from adults based on their body shape. More importantly, M. surmuletus occurring in different geographical areas could be differentiated using this method. Based on pRDA, a significant difference of head morphological dimensions was observed between populations occurring in the eastern English Channel and those occurring in the Bay of Biscay, suggesting that fish from these areas could represent two subpopulations.  相似文献

Soft tissue artefact affects the determination of skeletal kinematics. Thus, it is important to know the accuracy and limitations of kinematic parameters determined and modelled based on skin marker data. Here, the curvature angles, as well as the rotations of the lumbar and thoracic segments, of seven healthy subjects were determined in the sagittal plane using a skin marker set and compared to measurements taken in an open upright MRI scanner in order to understand the influence of soft tissue artefact at the back. The mean STA in the flexed compared to the extended positions were 10.2±6.1 mm (lumbar)/9.3±4.2 mm (thoracic) and 10.7±4.8 mm (lumbar)/9.2±4.9 mm (thoracic) respectively. A linear regression of the lumbar and thoracic curvatures between the marker-based measurements and MRI-based measurements resulted in coefficients of determination, R2, of 0.552 and 0.385 respectively. Skin marker measurements therefore allow for the assessment of changes in the lumbar and thoracic curvature angles, but the absolute values suffer from uncertainty. Nevertheless, this marker set appears to be suitable for quantifying lumbar and thoracic spinal changes between quasi-static whole body postural changes.  相似文献

Impedance measurements are necessary to determine the passive electrical properties of cells including the equivalent circuits of the several pathways for current flow. Such measurements are usually made with microelectrodes of high impedance (some 15 MΩ) over a wide frequency range (1-10,000 Hz) and so are subject to many errors. An input amplifier has been developed which has negligible phase shift in this frequency range because it uses negative feedback to keep tiny the voltage on top of the microelectrode. An important source of artifact is the extracellular potential produced by capacitive current flow through the wall of the microelectrodes and the effective resistance of the bathing solution. This artifact is reduced some 10 times by shielding the current microelectrode with a conductive paint. The residual artifact is analyzed, measured, and subtracted from our results. The interelectrode coupling capacitance is reduced below 2 × 10-17 F and can be neglected. Phase and amplitude measurements are made with phase-sensitive detectors insensitive to noise. The entire apparatus is calibrated at different signal to noise ratios and the nature of the extracellular potential is investigated. The phase shift in the last 5-20 μm of the microelectrode tip is shown to be small and quite independent of frequency under several conditions. Experimental measurements of the phase characteristic of muscle fibers in normal Ringer are presented. The improvements in apparatus and the physiological significance of impedance measurements are discussed. It is suggested that the interpretation of impedance measurements is sensitive to small errors and so it is necessary to present objective evidence of the reliability of one's apparatus and measurements.  相似文献

Laser spectroscopy measurements of the effective temperature of Ar1+ ions in the PNX-U multipole trap, in which argon plasma is ionized and heated by microwaves under electron-cyclotron-resonance conditions, are performed using a narrow-band tunable dye laser. The absorption profile of the 611.5-nm line is examined. In a microwave power range of 5–50 kW, the observed behavior of the effective temperature of argon ions Ti, eff indicates an anomalous mechanism for ion heating. It is shown that certain information about the electron temperature can be deduced from measurements by the laser-induced fluorescence (LIF) technique. The measurements performed also serve to test the laser technique and apparatus that is presently being developed for diagnosing additives to the ITER divertor plasma by the LIF method.  相似文献

The objectivity of several anthropometric measurements were tested in two institutes on the same subjects. In many cases systematic differences are to be expected: 1. Girth measurements tend to differences in the same direction. 2. The comparability of skeletal measurements is often uncertain; some measurements tend to give lower, other ones higher figures. Biacromial diameter and radio-ulnar epicondylar width do not reveal systematic differences. 3. Skinfold thicknesses measured do not reveal systematic differences.  相似文献

For spectral‐domain optical coherence tomography (SD‐OCT) studies of neurodegeneration, it is important to understand how segmentation algorithms differ in retinal layer thickness measurements, segmentation error locations and the impact of manual correction. Using macular SD‐OCT images of frontotemporal degeneration patients and controls, we compare the individual and aggregate retinal layer thickness measurements provided by two commonly used algorithms, the Iowa Reference Algorithm and Heidelberg Spectralis, with manual correction of significant segmentation errors. We demonstrate small differences of most retinal layer thickness measurements between these algorithms. Outer sectors of the Early Treatment Diabetic Retinopathy Study grid require a greater percent of eyes to be corrected than inner sectors of the retinal nerve fiber layer (RNFL). Manual corrections affect thickness measurements mildly, resulting in at most a 5% change in RNFL thickness. Our findings can inform researchers how to best use different segmentation algorithms when comparing retinal layer thicknesses.  相似文献

Green photosynthetic bacteria harvest light and perform photosynthesis in low-light environments, and contain specialized antenna complexes to adapt to this condition. We performed small-angle neutron scattering (SANS) studies to obtain structural information about the photosynthetic apparatus, including the peripheral light-harvesting chlorosome complex, the integral membrane light-harvesting B808-866 complex, and the reaction center (RC) in the thermophilic green phototrophic bacterium Chloroflexus aurantiacus. Using contrast variation in SANS measurements, we found that the B808-866 complex is wrapped around the RC in Cfx. aurantiacus, and the overall size and conformation of the B808-866 complex of Cfx. aurantiacus is roughly comparable to the LH1 antenna complex of the purple bacteria. A similar size of the isolated B808-866 complex was suggested by dynamic light scattering measurements, and a smaller size of the RC of Cfx. aurantiacus compared to the RC of the purple bacteria was observed. Further, our SANS measurements indicate that the chlorosome is a lipid body with a rod-like shape, and that the self-assembly of bacteriochlorophylls, the major component of the chlorosome, is lipid-like. Finally, two populations of chlorosome particles are suggested in our SANS measurements.  相似文献

The optical pathlength amplification (ß factor), inducedby multiple scattering in glass-fibre filters used for lightabsorption measurements of aquatic particles, has been measuredby the transmittance-reflectance (T-R) method for algal specieswith cell diameter in the range from 1 to 5 µm. Theßfactors obtained showed a much lower dispersion than that observedin similar measurements carried out by the standard transmittancemethod, suggesting that this could be due, at least in part,to the incomplete account of the scattering losses taken bythe latter method. Homodyne and heterodyne measurements have been made of the spectrum and intensity of laser light scattered from solutions of skeletal muscle myosin at high salt concentrations. The spectral broadening and intensity measurements are consistent with the hypothesis that a myosin monomer-dimer equilibrium exists in these solutions. Values have been calculated for molecular weight and radius of gyration of the myosin monomer, virial coefficients, and diffusion constants of both monomer and dimer, and equilibrium constant of the reaction to form dimer. Diffusion constant measurements yield an approximate length of 1481 Å for the monomer and 2121 Å for the dimer. The significance of these lengths is discussed in terms of the models of Huxley and Pepe for the structure of the myofilament.  相似文献

PurposeThis work describes the integration of the M6 Cyberknife in the Moderato Monte Carlo platform, and introduces a machine learning method to accelerate the modelling of a linac.MethodsThe MLC-equipped M6 Cyberknife was modelled and integrated in Moderato, our in-house platform offering independent verification of radiotherapy dose distributions. The model was validated by comparing TPS dose distributions with Moderato and by film measurements. Using this model, a machine learning algorithm was trained to find electron beam parameters for other M6 devices, by simulating dose curves with varying spot size and energy. The algorithm was optimized using cross-validation and tested with measurements from other institutions equipped with a M6 Cyberknife.ResultsOptimal agreement in the Monte Carlo model was reached for a monoenergetic electron beam of 6.75 MeV with Gaussian spatial distribution of 2.4 mm FWHM. Clinical plan dose distributions from Moderato agreed within 2% with the TPS, and film measurements confirmed the accuracy of the model. Cross-validation of the prediction algorithm produced mean absolute errors of 0.1 MeV and 0.3 mm for beam energy and spot size respectively. Prediction-based simulated dose curves for other centres agreed within 3% with measurements, except for one device where differences up to 6% were detected.ConclusionsThe M6 Cyberknife was integrated in Moderato and validated through dose re-calculations and film measurements. The prediction algorithm was successfully applied to obtain electron beam parameters for other M6 devices. This method would prove useful to speed up modelling of new machines in Monte Carlo systems.  相似文献

Measurement of thyroid size and volume is a useful clinical parameter in both human and veterinary medicine, particularly for diagnosing thyroid diseases and guiding corrective therapy. Procuring a fully-equipped clinical ultrasound unit (FCUS) may be difficult in most veterinary settings. The present study evaluated the inter-equipment variability in dolphin thyroid ultrasound measurements between a portable ultrasound unit (PUS) and a FCUS; for both units, repeatability was also assessed. Thyroid ultrasound examinations were performed on 15 apparently healthy bottlenose dolphins with both PUS and FCUS under identical scanning conditions. There was a high level of agreement between the two ultrasound units in dolphin thyroid measurements (ICC = 0.859–0.976). A high intra-operator repeatability in thyroid measurements was found (PUS: ICC = 0.854–0.984, FCUS: ICC = 0.709–0.954). As a conclusion, no substantial inter-equipment variability was found between PUS and FCUS in dolphin thyroid size measurements under identical scanning conditions, supporting further application of PUS for quantitative analyses of dolphin thyroid gland in both research and clinical practices at aquarium settings.  相似文献

BackgroundLimb-salvage surgery for primary bone sarcomas are preceded by X-ray and MRI for surgical planning. However, the accuracy of X-ray and MRI predicted margins are not well described. Our study examined these questions: (1) How accurately do X-ray and MRI margin measurements reflect the true margin on pathology reports? (2) Do X-ray or MRI margin measurements have smaller differences compared to pathology reports? (3) How many X-ray or MRI margin measurement differences were greater than 1 cm, 2 cm, and 3 cm from pathology reports? (4) Is there an X-ray or MRI view that consistently results in a smaller difference from pathology reports?MethodsThis retrospective chart review examined patients with primary bone sarcoma treated with limb-salvage surgery. Reviewers used electronic measurement tools to determine margins from X-ray or MRI based on the resection length of the pathologic specimen. Mean differences of margin measurements to pathology reports were calculated. We determined outliers of imaging margin measurements at 1 cm, 2 cm, and 3 cm differences to pathology reports.ResultsIn the total cohort of 39 patients, the mean difference of X-ray and MRI margins compared to pathology reports were 1.09 cm (st dev 0.79 cm) and 0.71 cm (st dev 0.70 cm), respectively. MRI margin measurements had smaller differences compared to pathology reports than X-ray in 32 of 38 cases (84%) with complete imaging. X-ray outliers at 1 cm, 2 cm, and 3 cm differences were 36, 14 and 2 respectively for 70 margin measurements and MRI outliers at 1 cm, 2 cm, and 3 cm differences were 17, 6, and 0 respectively for 66 margin measurements. The views with the smallest difference were anterior-posterior X-rays and MRI views with the closest predicted margin.ConclusionElectronic MRI margin measurements with the closest predicted margin provided the smallest differences with pathology reports and are therefore the most accurate for preoperative planning. When there is adequate residual diaphysis for reconstructive fixation, surgeons should plan for a 3 cm bone margin using MRI measurements to ensure complete removal of the intramedullary extent of sarcoma.Level of Evidence: IV  相似文献

Quantum yield measurements were made with the red alga Porphyridium cruentum, cultured so as to give different proportions of chlorophyll and phycobilins. Totally absorbing suspensions were used so that there was no uncertainty in the amount of energy absorbed. These measurements have shown that chlorophyll, in this alga, has a photosynthetic efficiency as high as in other algal groups, and higher than the phycobilins—at least at wave lengths shorter than about 650 mµ. Wave lengths longer than this are beyond the range of maximum efficiency of chlorophyll. Under specified conditions of temperature and supplementary light full efficiency may be extended to longer wave lengths. The results of these measurements have made it unnecessary to suppose that in red algae chlorophyll plays a minor role while the phycobilins are the photosynthetic sensitizers of primary importance.  相似文献
