Colorado potato beetle (CPB) is the important pest of potato throughout the world. The study showed the effects of vermicompost on nutritional indices, digestive enzyme activities and intermediary metabolism of the larvae and the adults of CPB. Vermicompost affected significantly the efficiency of conversion of ingested food (ECI) and efficiency of conversion of digested food (ECD) in addition to activities of carbohydrases and proteases in both larvae and adults. Amount of total phenol compounds increased in the leaves of the potatoes grown on the soil containing 30% of vermicompost compared to control and it were amended by 15% of vermicompost. In case of intermediary metabolism, activities of aspartate aminotransferase and γ-glutamyl transferase showed no significant differences in the control and the treated larvae, but those were fluctuated upper and lower when 15% of vermicompost was added into growth plots. The amount of low-density lipoproteins (LDL) and high-density lipoproteins (HDL) in the larvae of CPB showed no significant differences among treatments. However, the amounts of LDL, HDL, glycogen and protein of the adults significantly increased using 30% of vermicompost. Results of the current study clearly revealed significant effects on some physiological processes in CPB fed on the plants grown in the different vermicompost treatments.  相似文献

Quantitative trait locus (QTL) mapping is a valuable new tool for locating genomic regions that underlie variation in important traits such as insecticide resistance. Because QTL mapping complements a candidate gene strategy for understanding the genetic architecture of important traits, it may also facilitate the identification of genes causing important variation. After mapping the QTL locations, markers closely linked to QTL can be used for genetic analysis of population structure and to measure the spread and increase of resistance-causing QTL alleles. In this study, QTL influencing resistance to the pyrethroid insecticide esfenvalerate were mapped in the Colorado potato beetle Leptinotarsa decemlineata (Say) (CPB). Three QTL contributing to esfenvalerate resistance were identified from a mapping population of 79 individuals analyzed at 90 marker loci. One QTL had a large effect and two QTL had smaller effects. The major QTL occurs on the X chromosome, overlapping the position of a candidate gene (Leptinotarsa decemlineata Voltage sensitive sodium channel [LdVssc1]) previously implicated in pyrethroid resistance. Resistance-increasing alleles at the two minor-effect QTL originated with the susceptible parent, suggesting that alleles of small effect may be segregating in susceptible populations. Comparison of the New York population from which the susceptible parent originated with a more-susceptible population from North Carolina suggests that the minor-effect loci identified here may explain some of the variation in tolerance observed among susceptible populations. DNA sequencing of a portion of LdVssc1 shows that the resistance-conferring allele from the resistant parent does not contain the kdr mutation previously found in CPB and typically observed in other insects that are resistant to pyrethroid insecticides because of changes in this gene.  相似文献

The genetic structure of a Colorado potato beetle population from Kiev oblast was examined by cluster analysis of individual RAPD patterns. The obtained clustering indicates that the population is structured. This may be explained by adaptation to pyrethroid insecticides used for controlling the population size of this pest. Micro-evolutionary factors affecting the genetic structure of local populations of Colorado potato beetle are discussed.  相似文献

Restriction endonuclease analyses of mitochondrial DNA (mtDNA) were used to examine genetic variability and population structure inLeptinotarsa decemlineata (Say). A group of three enzymes, EcoRI, HpaI, and PstI, was used to reveal polymorphism both within and among some of the 10 populations tested, yielding 16 haplotypes in combination. The frequencies of these 16 haplotypes differed significantly across geographic regions, indicating some partitioning of mtDNA haplotypes. Estimates of mtDNA sequence divergence (δ) between haplotypes ranged from 0.016 to 0.135%, suggesting local differentiation of mtDNA in some populations. Analysis of these data suggests that Texas was colonized by more than one mtDNA lineage, most likely originating in Mexico. We hypothesize that a larger founder size for the initial introductions or high levels of variability in the parent population at the edge of the CPB expanding range led to the initial partitioning of haplotypes observed in samples from Texas.  相似文献

The Colorado potato beetle (CPB), Leptinotarsa decemlineata Say is one of the most important defoliator pests of potato in the world and it often causes extremely large potato yield losses. Potatoes are the preferred hosts for the pest, but it may feed and reproduce on a number of other plants in the Solanaceae family. Public concern related to pesticides and their residues in and on the foods had prompted a rise of consumer interest in organically produced foods. There have been growing efforts to detect and introduce suitable plant compounds that they have insecticidal properties. However, discovering of plant extracts for possible use in control of this pest requires more studying about plant extracts and compounds. Since resistance of CPB to common chemical insecticides is well documented and potato is one of the most prominent nutritious food products for many people in many countries, we examined the effect of essential oils (EOs) of European pennyroyal, lavander, mint, oregano and savory and methanolic extracts of fumitory, licorice and oregano on the pest. These plants were selected because they have medicinal properties and they are safe to human and environment. Adult CPBs were exposed to mentioned plant extracts and essential oils. LC50 values for EOs of lavander and European pennyroyal were 4154 and 3561 ppm, respectively. The results demonstrated that essential oil of European pennyroyal (Mentha longifolia) would be suitable compound to control the pest, but essential oil of mint (Mentha spicata) was not effective against the pest. Also it is notable that at all treatments, the amount of adult feeding was very low.  相似文献

The allocation, external morphology, and ultrastructure of various types of sensilla on the antennae have been studied in the larvae of the Colorado potato beetle (Leptinotarsa decemlineata Say) using scanning and transmission electron microscopy. It has been found that the larval antenna bears 11 sensilla of four morphological types: trichoid (five), basiconic (four), styloconic (one), and antennal cone (one). It has been shown that the sensilla are innervated by 2?C7 receptor cells: trichoid, by 2?C4 receptor cells; basiconic, by 4?C7 receptor cells; and styloconic, by 6 receptor cells. Judging by the specifics of their ultrastructural organization, the function of these sensilla is gustatory. The antennal cone is innervated by two receptor cells, and its function is olfactory.  相似文献

为了进一步明确温度对马铃薯甲虫Leptinotarsa decemlineata (Say)生长发育的影响, 在恒温条件, 研究了温度对马铃薯甲虫实验种群生长发育的影响。结果表明:温度对马铃薯甲虫各虫态的发育历期、存活率及种群的繁殖力有显著的影响。发育历期随温度的升高而缩短, 发育速率与温度呈显著的正相关。马铃薯甲虫世代存活率由大到小的顺序为27℃23℃19℃31℃15℃27℃时成虫的产卵量最高, 单雌平均卵量为729.7粒;其次为23℃, 单雌平均卵量为639.2粒。并测得马铃薯甲虫卵、1龄幼虫、2龄幼虫、3龄幼虫、4龄幼虫、幼虫期、蛹期及全世代的发育起点温度分别为9.14, 10.50, 8.17, 10.28, 9.04, 9.59, 10.23和10.90℃, 有效积温分别为73.26, 43.22, 39.23, 34.05, 161.97, 273.02, 100.38和542.58日·度。据此认为温度对马铃薯甲虫实验种群的生长发育、存活和繁殖影响明显。  相似文献

马铃薯甲虫Leptinotarsa decemlineata (Say)是马铃薯Solanum tuberosum等茄科作物上的一种毁灭性害虫。为了探明寄主植物对新疆马铃薯甲虫种群生长发育的影响, 本研究通过非选择性试验测定了马铃薯、 茄子Solanum melongena和番茄Lycopersicon esculentum 3种栽培寄主及野生寄主中亚天仙子Hyoscyamus pusillus对马铃薯甲虫种群生长发育、 存活、 繁殖及生命表参数的影响。结果表明: 马铃薯甲虫幼虫虽然能够取食番茄, 但幼虫的发育历期长、 存活率低且蛹不能羽化, 表明番茄不是新疆马铃薯甲虫种群的适宜寄主。马铃薯、 茄子、 中亚天仙子3种寄主植物对该虫卵、 幼虫、 蛹的存活率和发育历期及成虫产卵前期没有显著影响, 但对蛹重和繁殖力影响显著。取食不同寄主植物后, 该虫蛹重和繁殖力从大到小的次序为: 马铃薯>茄子>中亚天仙子; 种群的净增殖率(R0)、 内禀增长率(rm)和种群趋势指数(I)从大到小依次为: 马铃薯>茄子>中亚天仙子。因此马铃薯为新疆马铃薯甲虫种群的最适宜寄主, 其次为茄子和中亚天仙子, 而番茄为不适宜寄主。  相似文献

Summary

The Daudi cell line, a male human line from Burkitt's lymphoma, possesses the peculiarity of releasing H-Y antigen into its culture medium. Daudi-conditioned medium was injected into Colorado potato beetles either during the blastoderm stage, when individualization of the pole cells occurs, or later, during gonadal differentiation. Ovarian and testicular sections examined at hatching showed that only ovarian differentiation was affected by the Daudi conditioned medium, which, irrespective of the day of injection, reduced the number of the terminal filaments of the future lateral ovarioles. Furthermore, in some cases sexual differentiation was blocked altogether. When H-Y antigen was precipitated from the conditioned medium with specific H-Y antisera, the effectiveness of Daudi-conditioned medium was partially destroyed. These results suggest that mammalian H-Y antigen inhibits morphogenetic events leading to ovarian differentiation in the Colorado potato beetle.  相似文献

Summary The ultrastructure of seven types of neurosecretory cells (NSC) in the medial and lateral groups of the protocerebrum is described. The differences among cell types established earlier by light microscopy parallel differences in size and appearance of the neurosecretory particles observed in electron micrographs. No relationship was found between the affinity for Gomori's paraldehyde fuchsin stain and the nature of the particles.The secretions of the A-, A1-, and C-types of NSC of the medial group are characterized by electron-dense neurosecretory granules of 1250 Å dia., medium-dense granules of 2100 Å, and electron-lucent vesicles of 1700 Å, respectively. The L-type NSCof the lateral group contain smaller (1300 Å) or larger (1700 Å) neurosecretory granules. The medial B- and E-types of NSC and the lateral LB-type contain granulated vesicles (1200 Å) of the same appearance. In this study, we report for the first time the presence of an antidiuretic factor in the head of the Colorado potato beetle (Leptinotarsa decemlineata (Say)) which acts directly on Malpighian tubules. Biologically active fractions were isolated from the head and separated using molecular weight filtration and high-performance liquid chromatography (HPLC). The resulting fractions were tested for their antidiuretic activity on single isolated Malpighian tubules. Antidiuretic activity was found in the 25% acetonitrile Sep-Pak fraction and the Cn-2 (3000-10,000 MW) and Cn-3 (<3000 MW) fractions, suggesting that the antidiuretic factor was probably a peptide of 25 to 50 amino acids. The antidiuretic factor was very potent, since after five successive fractionations on two different HPLC columns, a high level of inhibition (63%) of fluid secretion by Malpighian tubules could be observed at low dose (0.14 head-equivalent/microl). The antidiuretic factor isolated from the head of the Colorado potato beetle was not affected by repeated freezing and thawing but was sensitive to heat. The differences observed between the Colorado potato beetle antidiuretic factor and other insect diuretic and antidiuretic factors may indicate the possibility of a novel family of water regulation hormones in insects.  相似文献

  • 1 Orientation of second‐ and fourth‐instar Colorado potato beetle (CPB) Leptinotarsa decemlineata (Say) (Coleoptera: Chrysomelidae), to volatiles emitted from a solanaceous host, potato, and seven synthetic blends or three individual chemicals emitted by potato plants were investigated in laboratory bioassays.
  • 2 Both second‐ and fourth‐instar CPB were attracted to intact and mechanically damaged (MD) potato foliage. When offered a choice between intact and MD foliage, no preference was observed.
  • 3 Among seven synthetic blends tested (of which six are attractive to adult CPB), second‐ and fourth‐instar CPB were attracted only to a single three‐component blend comprising (±)‐linalool, methyl salicylate, and (Z)‐3‐hexenyl acetate. Individual compounds and two‐component blends were inactive. No significant difference was noted between larval responses to the attractive synthetic blend vs. MD potato foliage.
  • 4 Second‐and fourth‐instar larvae had similar thresholds for behaviour for the three‐component blend (50 µg source load). Female CPB were attracted to source loads 10× below the larval threshold (5 µg). Male CPB were the most sensitive life form tested with a behavioural threshold at 0.5 µg source load which was 10× and 100× below female and larval thresholds, respectively
  • 5 This is the first report of a synthetic chemical attractant for CPB larvae. As both larval and adult CPB are attracted to a single chemical blend, the usefulness of the attractant as a component of an attracticide or ‘push‐pull’ strategies for management of pestiferous populations is enhanced.

Many representatives of the beetle family Chrysomelidae exhibit a distinctive sexual dimorphism in the structure of adhesive tarsal setae. The present study demonstrates the influence of surface roughness on the friction force of Leptinotarsa decemlineata males and females. The maximum friction force of individual beetles was measured on epoxy resin surfaces (smooth and with asperities ranging from 0.3 to 12.0 microm) using a centrifugal force tester. On the smooth surface, no considerable differences between males and females were found, whereas on rough surfaces, females attached significantly (up to two times) stronger than males. Clawless beetles generated lower forces than intact ones, but demonstrated similar differences between males and females. The results indicate that the female adhesive system has its main functional trait in a stronger specialisation to rough plant surfaces whereas the adhesive system of males possess a certain trade-off between attachment to rough plant surfaces during locomotion on vegetation and to the smooth surface of the female elytra, while mating.  相似文献

In laboratory and greenhouse studies, the invading ability, virulence, and mortality caused by Stinernema feltiae and Heterorhabditis bacteriophora were compared. After one and two days of exposure to either nematode species, the mortality of Colordo potato beetle (CPB) Leptinotarsa decemlineata larvae at different instars, third and fourth, was recorded and the number of nematodes invading cadavers was more than the number of nematodes inside the larvae at the late last instar (one day before pre-pupa). Two concentrations, 250 and 500 IJs/dish, infective juvenile nematodes/0.5 ml were tested on different CPB larval instar. S. feltiae was more effective, with fourth instar rather than third and late last instar. On the other hand, H. bacteriophora showed a very weak effect with L. decemlineata. Also it was clear that S. feltiae was more effective and faster than H. bacteriophora: more than 70% of larvae were killed within 24 hours compared with H. bacteriophora which killed 40% of larvae within 48–72 hours. A significant difference in invading efficiency was observed with concentration 2500 IJs/pot in the greenhouse test. The number of adult females found in the cadavers of L. decemlineata larvae was always higher than the number of males. Foliage application of S. feltiae and H. bacteriophora resulted in a significant reduction of the number of damaged leaves and a lower index of damage compared with that in the control. We conclude that S. feltiae has significant potential and can help in the management of the Colorado potato beetle.  相似文献

【目的】昆虫气味结合蛋白(OBPs)在昆虫嗅觉行为中发挥着重要作用。马铃薯甲虫Leptinotarsa decemlineata是马铃薯上一种最主要的毁灭性害虫。为阐明该虫嗅觉识别分子机制,本研究对马铃薯甲虫26个OBP基因序列特征及组织表达谱进行研究。【方法】基于马铃薯甲虫触角转录组测序数据,利用生物信息学方法及qRT-PCR技术,分别对马铃薯甲虫26个LdecOBPs (LdecOBP1-LdecOBP26)的系统进化及基因的组织表达谱进行分析。【结果】除LdecOBP26基因外,其余25个LdecOBPs基因序列均具有完整的开放阅读框,编码120~255个氨基酸残基,预测的蛋白分子量为13.66~29.38 kD,等电点为4.12~8.42,它们属于两个亚家族,其中13个为Classical-C OBPs, 12个为Minus-C OBPs。除LdecOBP3和LdecOBP26外,其他24个LdecOBPs的N端均由16~23个氨基酸组成的信号肽序列。不同的OBPs亚家族均具有各自典型保守的Cys残基。LdecOBPs之间高度分化,氨基酸序列一致性在3.20%~41.91%。系统进化树分析表明,LdecOBPs与沙葱萤叶甲Galeruca daurica的GdauOBPs亲缘关系最近。基因表达谱分析显示,26个LdecOBPs基因在马铃薯甲虫的不同组织中表达,其中有12个LdecOBPs基因(LdecOBP2, LdecOBP4, LdecOBP6, LdecOBP9, LdecOBP10, LdecOBP12, LdecOBP13, LdecOBP16, LdecOBP20-22和LdecOBP24)在触角中高表达,2个LdecOBPs基因(LdecOBP5和LdecOBP17)在足中高表达,其他12个LdecOBPs基因(LdecOBP1, LdecOBP3, LdecOBP7, LdecOBP8, LdecOBP11, LdecOBP14, LdecOBP15, LdecOBP18, LdecOBP19, LdecOBP23, LdecOBP25和LdecOBP26)在触角、头(去除触角)、胸、腹、足和翅这些组织中均表达。【结论】本研究结果为进一步研究马铃薯甲虫嗅觉识别分子机制奠定了基础。  相似文献

Feeding behavior, in an ad libitum situation on potato plants in the laboratory, was continuously observed for approximately 7 h/day on 2 successive days for 18 adult femaleLeptinotarsa decemlineata. Additional behaviors were also recorded including resting, walking, biting, local movements, grooming, defecating, and regurgitating. These data were used to calculate a time budget for the various behaviors. The feeding data were analyzed to describe the structure of feeding for young adult females on their normal host plant. The criterion for a meal (minimum intermeal interval) was determined to be 286 s. This criterion was used to distinguish between intra- and intermeal interruptions in feeding for all subsequent analyses. Meals taken from leaves that were young, medium aged, or old did not differ, but on average beetles took 60% of their meals from young leaves. Meal size and meal duration were equally good predictors of when a meal would end. Feeding from stems was a prominent feature for most beetles. The stem meals had much longer durations than leaf meals, but stem feeding did not affect subsequent leaf feeding. The structure of feeding by these beetles is compared with that found in other insects, especiallyLocusta migratoria.  相似文献

Quiescence (death feigning) was induced in the Colorado potato beetle, Leptinotarsa decemlineata by mechanical disturbance of live and artificial plants on which beetles were walking, feeding or grooming. The response consisted of a sudden cessation of activity lasting 12 to 500 s. Recovery from this immobile state was usually preceded by either antennal or leg movement. The time taken for beetles to come out of the immobile state (i.e. recovery time) was not dependent on age or sex of beetles or on stimulus duration, but was dependent on degree of displacement and on satiation level. Repeated stimulations led to longer recovery times and to a smaller proportion of responding beetles. Stimulation within the immobile state also prolonged recovery time. The most effective frequency of mechanical disturbance for startling beetles was 30 Hz. This response is presumed to be part of the extensive defense repertoire of this beetle, but we have so far been unable to discover a natural enemy that will elicit it.  相似文献
