Plant growth promoting Pseudomonas fluorescens strain 267, isolated from soil, produced pseudobactin A, 7-sulfonic acid derivatives of pseudobactin A and several B group vitamins. In coinoculation with Rhizobium leguminosarum bv. trifolii strain 24.1, strain 267 promoted clover growth and enhanced symbiotic nitrogen fixation under controlled conditions. To better understand the beneficial effect of P. fluorescens 267 on clover inoculated with rhizobia, the colonization of clover roots by mTn5-gusA marked bacteria was studied in single and mixed infections under controlled conditions. Histochemical assays combined with light and electron microscopy showed that P. fluorescens 267.4 (i) efficiently colonized clover root surface; (ii) was heterogeneously distributed along the roots without the preference to defined root zone; (iii) formed microcolonies on the surface of clover root epidermis; (iv) penetrated the first layer of the primary root cortex parenchyma and (v) colonized endophytically the inner root tissues of clover.  相似文献

The expression of nodulation genes inR. trifolii is induced by flavone compounds present in clover root exudates. In the present experiments a bioassay with an indicator strain ofR. trifolii, which contained thelacZ gene fromEscherichia coli fused to theR. trifolii nodA gene, was used to measure the level ofnod gene expression inR. trifolii. Compounds that stimulatednodA gene expression were shown to be present in exudates of white clover (Trifolium repens L.) and nine cultivars of subterranan clover (T. subterraneum L.) seedling sgrown at a range of pH between pH 3.0 and pH 8.0. Thenod gene-induction activity of exudates was, however, reduced when seedlings of all clover species were grown at pH>7.0 and at pH<4.0 and pH<5.0 for white clover and subterranean clover respectively. No major differences were apparent in the activity of exudates from seedlings of the various cultivars of subterranean clover.Nod gene-induction activity of exudates was shown to increase markedly with seedling age. The presence of Ca at concentrations up to 10 mM in seedling culture solutions also resulted in marked increases in thenod gene-induction activity of seedling exudates. Increases in activity due to the presence of Ca were most apparent at low pH where between 5 and 10-fold increases were observed for white clover and subterranean clover respectively. Conversely, the presence of Al at concentrations up to 60 M in seedling culture solutions had no effect on thenod gene-induction activity of seedling exudates.The observations that both low pH and Ca concentrations affected thenod gene-induction activity of seedling exudates suggested that the net presence of stimulatory flavones in root exudates was an important contributing factor to the acid-sensitive step in nodule formation.  相似文献

The suppressive effects of undersown clover on Thrips tabaci Lindeman infestation in leek is known but not the stages in thrips population dynamics that are affected by intercropping and the mechanisms involved. Colonization or settling of adult onion thrips (T. tabaci) in monocropped leek (Allium porrum L.) and in leek intercropped with strawberry clover (Trifolium fragiferum L.) was studied in two potted plant experiments. Potted leek plants with and without undersown clover were placed for a short period (2 and 5 days) in monocropped or intercropped field plots when high thrips infestation was expected. Thrips adults were counted on all leaves and in the shaft during this observation period. Thrips populations were monitored weekly in the field plots throughout the entire growing season.In both potted plant experiments, there were consistently fewer thrips adults on intercropped leek plants than on monocropped plants. The plant growth parameters – number of leaves and stem diameter – were similar in all treatments and thus did not explain differences in thrips adults. Natural enemies were absent on both leek and clover, and thus cannot explain the differences in adult thrips numbers. Furthermore, no adults of T. tabaci were found on the clover in the potted plant experiment and only very few in the field experiment. Thus it was concluded that strawberry clover was not an effective trap crop for thrips.In the second potted plant experiment, clover was removed just before the leek plants were introduced to the field, thus eliminating direct physical, visual and olfactory interference by clover. After the undersown clover was removed, the leek plants harboured only one-third of the number of thrips adults, as compared to the monocropped plants. This study supports the notion that there are subtle links between intercropping and plant quality, and indicates that host-plant quality is an underlying cause of the reduction in adult thrips numbers in leek/clover intercropping.  相似文献

Three experiments are reported which examine the relative roles of host and Rhizobium genotypes as factors limiting clover (Trifolium repens L.) growth at low soil temperatures.In the first experiment un-nodulated clover and perennial ryegrass (Lolium perenne L.) were grown with non-limiting nitrate at root temperatures of 8, 10 and 12°C. The ryegrass had substantially better relative growth rates (RGR) than the clover with the biggest difference occurring at 8°C. Alterations in growth rate with temperature were more marked in clover than in ryegrass but the latter still produced several times more dry matter than clover at each temperature.In the subsequent experiments clover nodulated with different strains of rhizobia was grown with and without non-limiting additions of nitrate at root temperatures of 9, 12 and 15°C. Plants receiving nitrate generally produced more dry matter than those dependent upon Rhizobium for nitrogen but differences in yield between these treatments did not alter with temperature. This suggests that limitations imposed by nitrogen fixation are similar at both high and low temperatures. Indeed, there was some evidence that nitrogen limitations were rather more pronounced at the highest temperature. The first experiment clearly demonstrated that the clover genotype makes particularly poor use of nitrate at low root temperatures when compared to its common companion perennial ryegrass.It can be concluded that improvements in spring growth of clover will rest largely with alterations to the plant genotype and its ability to use combined nitrogen for growth at lower temperatures rather than with changes in rhizobia or any symbiotic characters.  相似文献

Previous experiments have shown that crimson clover (Trifolium incarnatum L.) used as a green manure may supply weed control benefits as well as nitrogen (N) to a subsequent crop of corn (Zea mays L.). In contrast to use of synthetic N fertilizer, use of fresh, incorporated crimson clover residue as an N source has been found to suppress lambsquarters (Chenopodium album L.) aboveground drymatter accumulation but to only temporarily reduce that of sweet corn. One possible cause of the clover's suppressive effect is the initial low availability of N that may occur after residue incorporation in the soil. A factorial treatment combination of +/– crimson clover residue and four rates of N fertilizer was used in two field experiments to further document the clover's influence on early plant growth and development and to test the hypothesis that low initial N availability is responsible for the clover's previously observed suppressive effects. The presence of crimson clover residue was found to reduce total emergence of lambsquarters by 27%, while application of N fertilizer increased lambsquarters emergence by almost 75%. Lambsquarters emergence was also delayed by the residue treatment. Addition of N did not alleviate the clover's suppressive effect on total emergence or emergence rate of lambsquarters. Sweet corn emergence and emergence rate differed by less than 5% in 0 N/+residue and 0 N/–residue treatments. Applications of N to residue plots suppressed rather than enanced sweet corn emergence. Lambsquarters aboveground biomass accumulation was 46% tower in the residue than nonresidue treatments at 23 days after planting (DAP) and remained 26% lower at 53 DAP. Addition of N did not alleviate the suppressive effect of the clover residue on lambsquarters aboveground drymatter accumulation. Sweet corn aboveground biomass accumulation was not affected by the presence of the clover residue. The results of the experiments indicate that the suppressive effect of crimson clover residue on lambsquarters emergence and growth is not attributable to initial low availability of N. However, given the stimulatory effect of N fertilizer on lambsquarters development, use of crimson clover as an N source would appear to provide weed control benefits both as a direct suppressant of weed emergence and growth and as a substitute for fertilizer N.  相似文献

Pseudomonas sp. strain 267 isolated from soil promoted growth of different plants under field conditions and enhanced symbiotic nitrogen fixation in clover under gnotobiotic conditions. This strain produced pyoverdine-like compound under low-iron conditions and secreted vitamins of the B group. The role of fluorescent siderophore production in the beneficial effect of strain 267 on nodulated clover plants was investigated. Several non-fluorescent (Pvd-) Tn5 insertion mutants of Pseudomonas sp. strain 267 were isolated and characterized. The presence of Tn5 insertions was confirmed by Southern analysis of EcoRI digested genomic DNA of each derivative strain. The siderophore-negative mutants were compared to the parental strain with respect to their growth promotion of nodulated clover infected with Rhizobium leguminosarum bv. trifolii 24.1. We found that all isolated Pvd- mutants stimulated growth of nodulated clover plants in a similar manner to the parental strain. No consistent differences were observed between strain 267 and Pvd- derivatives strains with respect to their plant growth promotion activity under gnotobiotic conditions.Dr Deryto died in august 1994  相似文献

Summary Phenotypic plasticity may be an ecologically important evolutionary response to natural selection in multiple environments. I have determined the effect of diet-induced developmental plasticity in the head size of grasshoppers (Melanoplus femurrubrum) onfeeding performance on two types of plants. Full-sib families were divided and raised on either red clover, Trifolium repens, or rye grass, Lolium perenne. In three different stages of ontogeny, grasshoppers raised on rye grass had significantly larger heads, relative to body size, than full-sibs raised on clover. A principal components analysis indicated that two to five relative head size characters covaried as a block in their plastic response to the feeding environment. Regressions of adjusted consumption rates (mg/sec) against relative head size revealed that larger head sizes, induced by the rye grass diet, enhanced consumption rates of rye grass, but not clover. Unexpectedly, a similar positive association was observed between head size and consumption rate for grasshoppers raised on clover when they were feeding on clover. These results support the inference that grasshoppers exhibit adaptive phenotypic plasticity. However, the unexpected influence of head size on consumption rates of clover indicates that the functional relationship between head morphology and feeding performance is complex and that variation in this relationship among plant environments is not sufficient to explain the evolution of diet-induced phenotypic plasticity.  相似文献

Summary Collections of the slug Deroceras reticulatum were made from grassland sites containing contrasting frequencies of the cyanogenic morph of white clover, Trifolium repens. In choice chamber experiments, slugs obtained from sites with a low frequency of cyanogenic clover showed a significantly greater degree of selective eating of acyanogenic morphs than slugs taken from a site containing a high frequency of cyanogenic clover. Differences in selectivity between populations were caused both by differences in the rate of initiation of feeding on cyanogenic morphs, and by differences in the extent of damage once feeding had been initiated. The implications of these results for the cyanogenic polymorphism of T. repens are discussed.  相似文献

A series of laboratory and field studies were done to evaluate a range of leguminous plant species for their feeding potential by adult weevils of the genusSitona Germar. (Coleoptera: Curculionidae). Three species ofSitona, S. lineatus L.,S. flavescens Marsh. andS. hispidulus F. all of which are found commonly on white clover (Trifolium repens L.) in the UK were offered a range of 11 legume species,T. repens (white clover, cv. Olwen),T. pratense L. (red clover, cv. Marcom),T. fragiferum L. (strawberry clover, cv. Palestine),T. hybridum L. (hybrid clover, cv. Tetra),T. incarnatum L (crimson clover),T. dubium Sibth. (lesser yellow trefoil),Lotus corniculatus L. (birdsfoot-trefoil, cv. Leo),L. uliginosus Schkuhr. (large birdsfoot-trefoil),Melilotus alba Desr. (white melilot),Medicago sativa L. (lucerne, cv. Europe) andM. lupulina L. (black medick) in two laboratory experiments. The weevils were offered a choice of these legumes in one experiment whilst in the other they did not have a choice of food material. These legumes were also sown in the field and a number of measurements of damage, together with counts ofSitona spp., were made. In the laboratoryS. lineatus andS. hispidulus favoured some of the legumes to a greater or lesser extent than white clover.S. flavescens was more restricted in its feeding than the other two weevil species. In the field studyS. lineatus invaded the experimental area quickly and tended to favourMedicago spp. andMelilotus spp. Later in the yearS. flavescens dominated the sitona fauna on the experiment, with the exception of aggregations ofS. lineatus onM. sativa andM. alba. In a separate screen of 5 varieties of white clover (cvs Donna, Menna, Kersey, Olwen and Grasslands Huia), cv. Olwen appeared to be the most susceptible to sitona attack.  相似文献

Protein in white clover (Trifolium repens L.) is poorly utilised by ruminants because of its extensive degradation to ammonia in the rumen. However, white clover produces condensed tannins (CT) in its flowers, which can reduce rumen proteolysis. Effects of increasing proportions of clover dry matter (DM) as flowers (and therefore floral CT) on soluble protein, ammonia and volatile fatty acid (VFA) concentrations were determined with in vitro incubations. Minced mixtures of 0, 250, 500, 750 and 1000 g/kg of DM as white clover flower (F) with the remainder as white clover leaf, were incubated in vitro and sampled after 0, 2, 4, 8, 12 and 24 h. Treatments contained 0, 13, 26, 39 and 52 g CT/kg DM, respectively. A further treatment with 500 g/kg DM as flower and 500 g/kg DM as leaf had polyethylene glycol added to remove effects of CT. Increasing the proportion of white clover as flowers from 0 to 1000 g/kg DM reduced net conversion of plant N to ammonia N from 290 to 120 mM/M at least partly due to reduced solubility of the protein. Treatments with 750 g/kg DM or more as clover flowers reduced ammonia concentrations to levels likely to limit microbial growth. Total VFA production was not affected by flower content, although the proportion of acetate to propionate increased. The contribution of CT to treatment effects was small compared to effects attributed to difference in chemical composition between flowers and leaves.  相似文献

A field study was carried out near Zürich (Switzerland) to determine the yield of symbiotically fixed nitrogen (15N dilution) from white clover (Trifolium repens L.) grown with perennial ryegrass (Lolium perenne L) and from red clover (Trifolium pratense L.) grown with Italian ryegrass (Lolium multiflorum Lam.). A zero N fertilizer treatment was compared to a 30 kg N/ha per cut regime (90 to 150 kg ha−1 annually). The annual yield of clover N derived from symbiosis averaged 131 kg ha−1 (49 to 227 kg) without N fertilization and 83 kg ha−1 (21 to 173 kg) with 30 kg of fertilizer N ha−1 per cut in the seeding year. Values for the first production year were 308 kg ha−1 (268 to 373 kg) without N fertilization and 232 kg ha−1 (165 to 305 kg) with 30 kg fertilizer N ha−1 per cut. The variation between years was associated mainly with the proportion of clover in the mixtures. Apparent clover-to-grass transfer of fixed N contributed up to 52 kg N ha−1 per year (17 kg N ha−1 on average) to the N yield of the mixtures. Percentage N derived from symbiosis averaged 75% for white and 86% for red clover. These percentages were affected only slightly by supplemental nitrogen, but declined markedly during late summer for white clover. It is concluded that the annual yield of symbiotically fixed N from clover/grass mixtures can be very high, provided that the proportion of clover in the mixtures exceeds 50% of total dry mass yield.  相似文献

The temporal N-uptake patterns of white clover (Trifolium repens L.) mixed with perennial ryegrass (Lolium perenne L.) and of red clover (Trifolium pratense L.) mixed with Italian ryegrass (Lolium multiflorum Lam.) were determined in successive harvests of herbage within the growth cycles of a ley established near Zürich (Switzerland). Rooting patterns were examined by injecting15N-fertilizer at soil depths ranging from 10 to 40 cm. The results were analyzed to determine the effect of variations in time and depth of N-uptake on the15N-based measurement of N from symbiosis (Nsym) and N from transfer (Ntrans).Grasses in mixture appeared to have deeper rooting systems than grass monocultures, which led to an overestimation of N transfer from white clover to perennial ryegrass if15N was spread on the soil surface.White clover generally lagged behind grass in soil N- uptake. Soil N-uptake of red clover slowed down before that of the grass because % Nsym almost reached 100% during the second half of each growth cycle. However, the effect of these dissimilarities on the seasonal average of %Nsym did not exceed 2%.It is concluded that at the observed high levels of N2 fixation, failure to account for the N-uptake patterns of the test and reference crops only slightly affected the estimates of % Nsym and % Ntrans, and did not invalidate the observed differences between species.  相似文献

Axenic seedling bioassays were performed on white clover, vetch, and alfalfa to assess the variety and dose responses of biological activities exhibited by membrane chitolipooligosaccharides (CLOSs) from wild type Rhizobium leguminosarum bv. trifolii ANU843. Subnanomolar concentrations of CLOSs induced deformation of root hairs (Had) and increased the number of foci of cortical cell divisions (Ccd) in white clover, some of which developed into nodule meristems. In contrast, ANU843 CLOSs were unable to induce Had in alfalfa and required a 104-fold higher threshold concentration to induce this response in vetch. Also, ANU843 CLOSs were not mitogenic on either of these non-host legumes. In addition, CLOS action also increased chitinase activity in white clover root exudate. Thus, the membrane CLOSs from wild type R. leguminosarum bv. trifolii are fully capable of eliciting various symbiosis-related responses in white clover in the same concentration range as extracellular CLOSs of other rhizobia on their respective legume hosts. These results and our earlier studies indicate that membrane CLOSs represent one of many different classes of bioactive metabolites made by R. leguminosarum bv. trifolii which elicit more intense symbiosis-related responses in white clover than in other legumes. Parasitism of the cabbage root fly, Delia radicum (L.) by the staphylinid Aleochara bilineata Gyllenhal and the cynipid Trybliographa rapae Westwood was examined in a cabbage monoculture and a mixed stand of cabbage undersown with white clover. Number of overwintering cabbage root fly pupae per plant was consistently reduced in the mixed stand, and the incidence of plants attacked by cabbage root fly was either reduced or not different in the mixed stand compared to cabbage monoculture. For both parasitoids, the probability of D. radicum attacked plants having at least one parasitized pupa increased with density of cabbage root fly pupae around the plant. For A. bilineata, this positive relation between presence of parasitism and host density was consistently stronger in cabbage monoculture than in cabbage undersown with clover. Location of a host plant by T. rapae was not consistently affected by the presence of clover. D. radicum attacked plants situated in the cabbage and clover mixture were found by T. rapae as easily as in cabbage monoculture. Overall, the total risk of parasitism for a cabbage root fly pupa by A. bilineata was reduced in the mixed stand compared to the cabbage monoculture, whereas the risk of parasitism by T. rapae was not consistently affected by clover. For both parasitoids, intensity of parasitism showed a variable relationship with host density on individual plants attacked by the cabbage root fly. Overall, in spite of consistently lower total density of pupae in the mixed cabbage—clover than in cabbage monoculture, the density of unparasitized pupae was reduced by the presence of non-host plants only in two of the four experiments. The results emphasize the need to include not only herbivore and crop, but also other plant species as well as natural enemies when evaluating management methods.  相似文献

The combined effects of soil water deficit and above and below ground interspecific plant competition on the growth, water-use efficiency (WUE), and measured carbon isotopic composition (δ13C) values of white clover and ryegrass were studied. White clover and ryegrass were grown in specially designed crates 1) individually; 2) in shoot competition; or 3) in shoot + root competition and either well-watered or at a moderate or severe soil water deficit. The effects of shoot + root competition on shoot dry matter growth were substantial and benefited both white clover and ryegrass when well-watered or at a moderate soil water deficit, while severely reducing white clover shoot dry matter growth at severe soil water deficit. Plant competition did not affect the WUE of white clover or ryegrass. As soil water deficit increased, the WUE of white clover did not change whereas the WUE of ryegrass increased and was greater than that of white clover. This was attributed to the lower leaf water conductance of ryegrass which conserved water and maintained growth longer compared to white clover. A stronger correlation existed between soil water deficit and measured δ13C values for ryegrass at each plant competition level (P<0.001) than existed for white clover (individual: P<0.01; shoot + root: P<0.001; shoot: P<0.10). Unlike white clover, the relationship between measured δ13C values and shoot dry matter growth indicated that C assimilation for ryegrass was dependent on type of plant competition. That WUE remained constant for white clover while measured δ13C values increased as soil water deficit increased, suggests that the role below ground respiration rate played in determining δ13C values increased. The WUE of white clover appears to be independent of the nature of the competition between plants and the soil water deficit level at which it is grown, whereas for ryegrass, the addition of root competition to shoot competition should lead to increases in its WUE. During two consecutive years the effects of intercropping fresh market white cabbage with two species of clover on pest populations and yield were studied. White cabbage cv. Minicole was intercropped withTrifolium repens (white clover) andTrifolium subterraneum (subterranean clover) as compared to the monocrop. During the season observations were made on pest population developments, especially ofMamestra brassicae L. (cabbage moth),Brevicoryne brassicae L. (cabbage aphid),Delia brassicae L. (cabbage root fly), and evaluation of caterpillar feeding injury. At harvest the yield in quantity and quality was determined to be able to assess the gross financial result. Intercropping effects in terms of suppression of oviposition and larval populations of various pests were found. Although no pesticides were used and competition reduced the weight, the quality of the intercropped cabbages lead to a better financial result compared to the monocropped cabbage crop. The results are discussed in the perspective of the practical implications in the context of IPM.  相似文献

It was the aim of this study to determine the way in which low temperature modifies the effect of a competing grass on nitrogen fixation of a forage legume. White clover (Trifolium repens L.) was grown in monoculture or in different planting ratios with timothy (Phleum pratense L.) or perennial ryegress (Lolium perenne L.) in growth chambers at either 7.5/5°C (LoT) or 15/10°C (HiT) average day/night temperatures, and with 2.5 or 7.5 mM 15N-labelled nitrate in the nutrient solution.Competition with grass led to a marked increase in the proportion of clover nitrogen derived from symbiosis (% Nsym). This increase was slower at LoT where % Nsym was reduced considerably; it was closely related to the reduction in the amount of available nitrate as a result of its being utilized by the grass.Nitrogen concentration in white clover herbage and dry matter yield per clover plant were reduced, for the most part, when a competing grass was present. The amount of nitrogen fixed per plant of white clover decreased markedly with temperature. Low temperature consequently accentuated competition for nitrate. The capacity of white clover to compete successfully was limited by its slower growth and nitrogen accumulation.  相似文献

The consumotion, utilization and growth of third-instar larvae of black beetle, Heteronychus arator (F.) feeding on roots of ryegrass (Lolium perenne), paspalum (Paspalum dilatatum) and white clover (Trifolium repens) were studied. Consumption was greater on the grasses than on white clover when assessed by gravimetric methods. Larvae had a higher growth rate on the grasses. Utilization of the roots of the three species was similar indicating that poor growth on white clover is the result of low consumption probably induced by the presence of feeding deterrents in the roots.This work is part of a study carried out by the senior author for the degree of D. A system was established for introducing cloned genes into white clover (Trifolium repens L.). A high regeneration white clover genotype was transformed with binary Agrobacterium vectors containing a chimaeric gene which confers kanamycin resistance. Transformed kanamycin resistant callus was obtained by culturing Agrobacterium inoculated stolon internode segments on selective medium. The kanamycin resistance phenotype was stable in cells and in regenerated shoots. Transformation was confirmed by the expression of an unselected gene, nopaline synthase in selected cells and transgenic shoots and by the detection of neomycin phosphotransferase II enzymatic activity in kanamycin resistant cells. Integration of vector DNA sequences into plant DNA was demonstrated by Southern blot hybridisation.  相似文献

Water extracts of the compost produced from activated sludge and coffee residue were found to be selectively inhibitory to seed germination of some legumes. Germination rate of white clover (Trifolium repens L.), red clover (Trifolium pratense L.) and alfalfa (Medicago sativa L.) seeds were reduced to 2, 29 and 73% of the control, respectively, by water extracts of the compost (20 g l–1). However, the extracts did not show any inhibition to seed germination of sorghum (Sorghum bicolor Moench), African millet (Eleusine coracana Gaertn.), and Komatsuna (Brassica rapa L.) at the same concentration. The inhibitors in the compost extracts were separated by ion-exchange chromatography and reverse-phase high performance liquid chromatography (HPLC) and the inhibitory activities of seed germination were tested with white clover seeds. Five inhibitors were isolated and identified as 3,4-dichlorophenylacetic acid (3,4-DCP), 3,4-dichlorobenzoic acid (3,4-DCB), 3,4,5-trichlorophenylacetic acid, 3,4,5-trichlorobenzoic acid and mono-2-ethylhexylphthalate by 1H-, 13C-NMR spectroscopy and mass spectrometry. The inhibitory activities of some authentic chemicals of the inhibitors and the related compounds were compared. The results indicated that the main inhibitor in the compost could be 3,4-DCB, which was contained at the concentration of 6.58 mg kg–1 compost and showed the strongest inhibitory effect on seed germination of white clover among the tested compounds.  相似文献
