One hundred and seventy Holstein Friesian cows were randomly assigned to receive either 500 ug cloprostenol or saline placebo on Day 26 postpartum followed by 500 ug cloprostenol or saline on Day 40 postpartum. Four treatment groups were formed: Group 1 - saline (Day 26)/saline (Day 40); Group 2 - cloprostenol/(Day 26) saline (Day 40); Group 3 - saline (Day 26)/cloprostenol (Day 40); Group 4 - cloprostenol (Day 26)/cloprostenol (Day 40). Double blind techniques were used in administering treatments and in assessing the response to treatment. Palpation of the reproductive tract per tectum and uterine biopsies were performed on 92 cows prior to each treatment at Day 26 and Day 40 postpartum. Progesterone concentrations were determined on milk samples collected prior to treatment. There were no significant differences among treatment groups with respect to services per conception, number of heats detected before first service and culling for infertility. Cloprostenol treatment at Day 26 appeared to delay the first estrus, but it reduced the number of days to conception after the first service. Cows receiving cloprostenol at Days 26 and/or 40 had a decreased calving-to-conception interval compared to controls (P = 0.01). Sequential therapy with two doses of cloprostenol resulted in slightly better reproductive performance than either treatment on Day 26 or 40 alone. Treatment with cloprostenol resulted in a decrease in the subsequent incidence of pyometra (P < 0.05). It is concluded that in the herd studied, cloprostenol therapy at Day 26 and/or 40 postpartum was beneficial to reproductive performance. Although it was anticipated that cloprostenol would be more effective in cows with elevated progesterone levels, the opposite was observed at the Day 26 cloprostenol treatment. Uterine biopsy at Days 26 and/or 40 had a detrimental effect on subsequent reproductive performance.  相似文献

Fifty-one cyclic beef cows were mated with fertile bulls. At 36 h after the start of oestrus, cows were assigned to receive sesame oil (controls) or progesterone (100 mg) on Days 1, 2, 3 and 4 of pregnancy. Peripheral plasma concentration of progesterone was measured until slaughter on Days 5 or 14. Cows were randomly assigned to be slaughtered on Days 5 or 14 or remain intact and palpated per rectum on Day 40 to verify pregnancy. Uteri on Days 5 and 14 were flushed for recovery of luminal protein and conceptus tissue. Conceptus and endometrial tissues were cultured with [3H]leucine and submitted to two-dimensional-PAGE and fluorography. Administration of progesterone increased peripheral plasma progesterone concentration on Day 2-5. Conceptuses recovered from progesterone-treated cows on Day 14 were advanced in development compared to conceptuses from control cows. Conceptuses recovered from progesterone-treated cows were viable as polypeptides associated with maintenance of pregnancy in cattle were synthesized and released at an earlier time and pregnancy was maintained beyond Day 40. Early progesterone stimulation altered the synthesis and release of polypeptides from endometrial explant cultures on Day 5. Results indicate a role of progesterone in the maternal regulation of conceptus growth and development in early pregnancy of cattle.  相似文献

Three experiments were conducted, using multiparous crossbred beef cows, to test the ability of exogenous prostaglandin F(2alpha) (PGF) and/or naloxone to reduce the duration of the postpartum interval to estrus and to improve subsequent reproductive performance. In each experiment, postpartum cows were assigned to treatments by calving date. In Experiment 1, cows (n=44) were assigned to 1 of 4 treatment groups: 1) control, 2) PGF on Day 25 post partum, 3) 400 mg naloxone (3 doses) at 12-h intervals on Day 30 post partum, and 4) PGF on Day 25 followed by 3 400-mg doses naloxone at 12-h intervals on Day 30 post partum. In Experiment 2, cows (n=126) were assigned either to 1) control or 2) PGF on Day 30 post partum In Experiment 3, cows (n=67) were again assigned to 1 of 4 treatments 1) control, 2) PGF on Day 30 post partum, 3) PGF on Day 40 post partum, and 4) PGF on Day 30 and 40 post partum. Serum progesterone was used to determine the postpartum interval to estrus in Experiments 1 and 3. In all 3 experiments, serum progesterone was used to determine the proportion of cows that had reestablished estrous cycles at the start of breeding. Pregnancy rate and calving interval were analyzed for all trials. Naloxone had no effect (P > 0.20) on any reproductive variable measured. The postpartum interval to estrus was similar (P > 0.30) for PGF-treated and control cows in Experiments 1 and 3. The proportion of cows cycling at the start of breeding and the calving interval were not affected (P > 0.20) by PGF treatment in any of the experiments. Only the administration of PGF on Day 40 post partum in Experiment 3 improved (P=0.04) the subsequent pregnancy rate. Analysis of data pooled across experiments showed that the pregnancy rate was higher (P=0.03) for cows treated with PGF than for control cows (91.4 and 72.9%, respectively). It was concluded that administration of PGF during the early postpartum period improves subsequent reproductive function in beef cows.  相似文献

This study was conducted to evaluate the use of prostaglandin F2alpha (PGF2alpha) in the initial treatment of ovarian cysts in dairy cattle. Two hundred and sixty three cows diagnosed cystic on palpation per rectum were randomly assigned to one of three treatment groups (A, B or C). Cows in Groups A and B were treated with 25 mg i.m.of PGF2alpha at the time of diagnosis (Day 0), while cows in Group C received 100 mug of GnRH. Seven days following initial treatment (Day 7), cows from Group A that were not observed in estrus were treated with GnRH. Cows from Groups B and C were not treated. On Day 14, all cows that had not been inseminated received PGF2alpha. A blood sample was obtained from all cows on Days 0, 7 and 14 and was analyzed for progesterone (P4) using radioimmunoassay. Incidences of estrus were recorded and cows that were more than 60 d in milk at the time of diagnosis were bred when observed in estrus. The incidence of follicular cysts on Day 0 (as defined as P4 <0.5 ng/ml) was similar between groups and constituted about 40% of all cysts. There were significantly more cows pregnant to insemination within 7 d of initial treatment in Group B than in Groups A and C (P<0.05). After Day 14, the pregnancy rate was not statistically different between Group B and C, but Groups B and C had a statistically higher pregnancy rate than Group A from Day 21 to Day 35. At the end of the study, there was no statistical difference for the pregnancy rate between groups. We concluded that treatment of ovarian cysts diagnosed by per rectum examination with prostaglandin (at time of diagnosis and 14 d later for cows that were not inseminated) was as effective as initial treatment with GnRH followed by prostaglandins 14 d later for cows that were not inseminated previously. Cows that were initially treated with prostaglandins also tended to become pregnant sooner.  相似文献

GnRH treatment (250 ug) 48 h after prostaglandin F(2alpha) in 40 superovulated cows induced a release of LH (increment > 5 ng/ml) in only 13 of the older cows. Eleven of these cows did not yield viable embryos. Thirty-two of 75 cows had preovulatory surge levels of LH 48 h after prostaglandin treatment. Plasma progesterone concentrations were determined in 140 cows at the time that superovulation was initiated. Eighty-four of these donors were superovulated with 40 mg of FSH and 56 donors with 48 mg of FSH. There was no relationship (P > 0.05) between the concentration of progesterone at the start of superovulation with either ovulation rate determined by palpation per rectum or the number of viable embryos per flush. These parameters were also unaffected (P > 0.05) by age of the donor or the dose of FSH. In another group of donors, treatment with 40 mg FSH was compared over a 3-d (n = 28) and a 4-d (n = 18) interval. The donors treated with FSH over a 3-d period had similar ovulation rates but yielded less viable embryos (1.5 v 5.8, P < 0.05). The fertility rate of 33 cows, inseminated 60 and 72 h after prostaglandin, was comparable to the fertility rate of 18 cows inseminated at 60, 72 and 84 h after prostaglandin treatment.  相似文献

Pregnancy failure during placentation in lactating dairy cows was associated with low concentrations of serum progesterone. Beef cows have greater serum progesterone and less pregnancy failure. Experiment 1 determined that reduction of serum progesterone affected late embryonic/early fetal loss in suckled beef cows. Cows (n = 40) received progesterone from two new or used controlled internal drug releasing devices, replaced every 5 d, beginning on Day 28 of gestation (mating = Day 0); CL were enucleated on Day 29. Retention of pregnancy was 77% in treated cows and 97% in 78 control cows (P < 0.05). Experiment 2 determined how pregnant, lactating dairy cows with high or low progesterone concentrations during Days 28-34 differed in luteal function or in serum progesterone during replacement therapy. Luteal tissue from such cows was assayed for progesterone and expression of mRNA for genes of endothelin and prostaglandin (PG) systems. Secretion of progesterone and prostaglandins by dispersed luteal cells was determined during incubation with LH, endothelin-1, or arachidonic acid. Neither luteal progesterone nor mRNAs for endothelin or prostaglandin systems differed. Endothelin-1 inhibited secretion of progesterone more (P < 0.05) in luteal cells from cows with low versus high serum progesterone, when incubated with arachidonic acid. Secretion of prostaglandin F2α was increased and that of 6-keto-PGF1α decreased by endothelin-1 in vitro. Serum progesterone during replacement was lower (P < 0.05) for cows with low than high serum progesterone at lutectomy. Thus, clearance, more than luteal production, determined peripheral progesterone in pregnant, lactating dairy cows.  相似文献

The objective of this study was to determine the efficacy of PGF2 alpha treatment on pregnancy and calving rates in autumn-calving suckler beef cows synchronized with progesterone and eCG. The population studied consisted of 124 Charolais and 130 Limousin cows in 13 and 12 beef herds, respectively. In each herd, pairs of cows were formed according to parity, body condition score and calving difficulty. Group 1 received a progesterone releasing intravaginal device (PRID) for 12 d with a capsule containing 10 mg estradiol benzoate at implant insertion and 500 IU eCG at PRID removal (Day 0). Group 2 received the same treatment plus 25 mg i.m. dinoprost at Day -2. Each cow was artificially inseminated 56 h after PRID removal (Day 3). Plasma progesterone concentrations were measured to determine cyclicity prior to treatment in samples take on Days -22 and -12, to confirm the occurrence of ovulation (Day 13) and to determine the early pregnancy rate (Day 26). Serum pregnancy-specific protein B (PSPB) concentrations were determined to assess pregnancy rate at Day 39. The effects of variation factors on pregnancy and calving rates after treatment were studied using logistic mixed models and a Cox model, respectively. There were no significant differences between groups or breeds for the rate of cyclicity before treatment nor for ovulation rate (means, 74.1 and 95.7%, respectively). Cyclicity was, however, influenced by individual factors such as body condition score (OR = 3.36, P = 0.001), parity (OR = 5.4, P = 0.001) and herd factors such as stocking rate (OR = 5.62, P = 0.001). The use of a prostaglandin injection increased pregnancy rate at Day 26 (71.7 vs 56.7%, P = 0.01) and at 39 d (67.7 vs 54.3%, P = 0.02) and the calving rate at induced estrus (64.5 vs 48.5%, P = 0.01). We observed 9 twin calvings (5.6%) which occurred in cyclic cows only before treatment. Cows in Group 2 had a 1.5 greater chance of calving before 300 d following the first AI than cows in Group 1 (P = 0.03). In conclusion, the addition of PGF2 alpha injection, 48 h before PRID removal, increased reproductive efficiency in autumn-calving Charolais and Limousin suckler beef cows compared to a classical estrus synchronization treatment using a PRID + eCG.  相似文献

The oxytocin-induced uterine prostaglandin (PG) F2 alpha response and the levels of endometrial oxytocin receptors were measured in ovariectomized ewes after they had been given steroid pretreatment (SP) with progesterone and estrogen to induce estrus (day of expected estrus = Day 0) and had subsequently been treated with progesterone over Days 1-12 and/or PGF2 alpha over Days 10-12 postestrus. The uterine PGF2 alpha response was measured after an i.v. injection of 10 IU oxytocin on Days 13 and 14, using the PGF2 alpha metabolite, 13,14-dihydro-15-keto-PGF2 alpha (PGFM), as an indicator for PGF2 alpha release. The levels of oxytocin receptors in the endometrium were measured on Day 14. During the treatment with progesterone, the peripheral progesterone concentrations were elevated and remained above 1.8 ng/ml until the morning of Day 14. The PGFM responses to oxytocin in untreated controls and SP controls were low on both Days 13 and 14 whereas the levels of endometrial oxytocin receptors in the same ewes were high. Treatment with progesterone either alone or in combination with PGF2 alpha significantly (p less than 0.04) increased the PGFM response on Day 14 and reduced the levels of endometrial oxytocin receptors; treatment with PGF2 alpha alone had no effect. It is concluded that progesterone promotes the PGFM response to oxytocin while simultaneously suppressing the levels of endometrial oxytocin receptors. PGF2 alpha treatment had no effect on either the uterine secretory response to oxytocin or the levels of oxytocin receptors in the endometrium.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Involvement of the plasminogen activation system in cow endometritis   总被引:2,自引:0,他引:2  
The objectives of this study were to investigate the: (a) presence and activity of components of the "plasminogen activators/plasmin" system in dairy cows with or without endometritis; (b) variations in enzyme activity according to the degree of endometritis; and (c) associations between these enzymes and changes in endometrial histology after intrauterine antibiotic treatment. Endometrial biopsies were collected from anestrus (no palpable ovarian structures and milk progesterone <1 ng/ml) Holstein cows, 30-40 days postpartum. On the basis of a vaginoscopic examination, rectal palpation of the cervix and uterus, and endometrial histology, there were 92 cows with endometritis and 20 cows without endometritis. After biopsy collection, each cow was given an intrauterine infusion of 1.5x10(6) IU of procaine penicillin G. In cows with endometritis, genital tract examinations and biopsies were repeated 2 weeks later. Both plasminogen activators (PAs), tissue type (t-PA) and urokinase (u-PA), were immunologically identified in all uterine biopsies. Plasminogen activator activity (PAA) increased, whereas plasminogen activator inhibition (PAI) and plasmin inhibition (PI) decreased in proportion to the degree of inflammation. Two weeks after intrauterine treatment, PAA had decreased significantly in all cows that had reduced severity of endometrial inflammation and had increased significantly in all cows with increased severity of inflammation. The change in the degree of inflammation depended upon plasminogen activator activity; cows with higher PAA were more likely to improve. In conclusion, there was evidence for a role of the plasminogen activation proteolytic system in bovine endometritis.  相似文献

Doses of 125, 250 or 500 micrograms LH-RH were injected i.m. into suckled beef cows on approximately Day 11 of an oestrous cycle synchronized by prostaglandin treatment. There was a positive linear relationship between dose of LH-RH and the area under the measured LH peak. Administration of 500 micrograms LH-RH as a single injection to suckled cows 13-32 days post partum resulted in LH release but failed to induce normal ovarian activity. A small transient rise in plasma progesterone for 6--9 days occurred at the expected time after injection in 50% of animals. Administration of 500 micrograms LH-RH to suckled beef cows approximately 20--30 days post partum and a second injection approximately 10 days later at the time when the resulting transient rise in plasma progesterone had returned to basal values induced normal cyclic activity (as shown by progesterone concentrations and observed oestrus) at 35 days compared with 70 days for untreated controls. Pituitary responsiveness to LH-RH, as assessed by LH levels, was found to increase up to 20 days post partum.  相似文献

The effect of plane of nutrition on progesterone receptor (PR) and estrogen receptor alpha (ERalpha) expression in ovine endometrium was investigated. Rasa Aragonesa ewes (n=26) were fed diets to provide either 1.5 (Group C) or 0.5 (Group L) times the daily maintenance requirement and were slaughtered at Days 5 or 10 of the estrous cycle (Day 0=estrus). PR and ERalpha immunoreactivity were analyzed in eight endometrial cell compartments, defined by cell type and location. Group L had less PR immunostaining on Day 5 (P<0.05), which is consistent with lesser endometrial content of progesterone found in such animals. Most cell types of Group C had down regulation of PR at Day 10, but in Group L, this pattern was observed only in three cell compartments. The lesser PR contents found at Day 5 in Group L ewes may explain the lack of inhibition of PR. No effect of treatment or day of the estrous cycle was observed in ERalpha. Results indicate that endometrial PR is affected in a cell type, in specific manner, by plane of nutrition.  相似文献

The influence of temporary cooling on pregnancy rate and ovarian secretion of cortisol, estradiol and progesterone in Holstein cows was evaluated during the months of June to September. Cows were randomly assigned to one of five treatment groups: 1) evaporative cooling, 8 d; 2) evaporative cooling, 16 d; 3) refrigerated air conditioning, 8 d; 4) refrigerated air conditioning, 16 d and 5) controls maintained outdoors with access to shade. Estrus was synchronized by giving two injections of prostaglandin (25 mg). Cows in the cooled groups were placed in box stalls at the time of the second prostaglandin injection, allowing them to be cooled for approximately 3 d prior to breeding. Blood samples were taken on days 0, 5, 10, 15, 20 and 25 postbreeding and serum was analyzed for estradiol, cortisol, progesterone and a pregnancy-specific protein. Pregnancy rates as determined by palpation per rectum at 40 to 60 d post breeding were not different between cows in cooled treatments, regardless of duration or type of cooling. Pregnancy rate was higher (P<0.05) in cooled as compared to control cows. Serum progesterone concentrations were higher on Day 15 in cooled cows as compared to control cows. No differences in serum estradiol or cortisol concentrations were observed between cooled and control cows. Pregnancy-specific protein determinations indicated that 50% of both cooled and control cows conceived. Results indicate that temporary cooling increased pregnancy rates by increasing embryonic survival rates.  相似文献

The present study was developed to determine if administration of progesterone, early in the estrous cycle of the cow, stimulated an advanced pulsatile release of PGF2 alpha from the uterine endometrium resulting in a decreased interestrous interval. Twenty-three cyclic beef cows were randomly assigned to receive either sesame oil or progesterone (100 mg) on Day 1, 2, 3 and 4 of the estrous cycle. Peripheral plasma concentrations of progesterone and the metabolite of prostaglandin F2 alpha, 15-keto-13,14-dihydro-prostaglandin F2 alpha (PGFM) were measured by radioimmunoassay. Administration of exogenous progesterone increased peripheral plasma concentration of progesterone in treated (3.67 ng/ml) compared to control (1.28 ng/ml) cows from Day 2 through 5 of the estrous cycle. Progesterone administration shortened the interestrous interval (16.7 d) compared to controls (21.6 d). The shortened interestrous intervals in treated cows resulted from an earlier decline in peripheral plasma progesterone. Decline of peripheral plasma progesterone concentrations is coincident with an increased pulsatile release of PGFM in both progesterone treated and control cows. Results indicate that administration of exogenous progesterone stimulates an earlier maturation of endometrial development, causing an advanced release of PGF2 alpha which shortens the interestrous interval of the cow.  相似文献

Luteal progesterone was removed by an injection of prostaglandin F(2alpha) or bilateral ovariectomy on Day 12 of pregnancy in pony mares. The embryonic vesicle remained mobile in the uterus until loss occurred on Days 13, 13, 15, or 19 in four prostaglandin-treated mares and Days 15, 17, 19, or 26 in four ovariectomized mares. Exogenous progesterone given daily, starting on Day 12, maintained pregnancy until Day 40 in five of five prostaglandin-treated and three of four ovariectomized mares. During two-hour mobility trials on Day 14, embryonic vesicles in mares without luteal or exogenous progesterone (n = 9) moved to a different uterine segment less frequently (mean number of location changes per two-hour trial: 7.2 +/-1.0 vs 10.4 +/-1.1, P < 0.05) and were observed more often in the uterine body (14.9 +/-2.9 vs 8.9 +/-1.3, P < 0.10) compared to vesicles in mares with a progesterone influence (n = 15). Of mares that still had a vesicle present on Day 18, fixation occurred by Day 17 in all (12 12 ) mares under the influence of luteal or exogenous progesterone but failed to occur in the three mares that were not under progesterone influence. Progesterone replacement was started on Day 16 in three mares that received prostaglandin F(2alpha) on Day 12 and still had a vesicle on Day 16. The vesicle was maintained and continued to develop in all three mares, indicating that the vesicles were viable four days after PGF(2alpha) treatment. However, fixation tended to be delayed (P < 0.15) and orientation of the embryo proper was altered (P < 0.005) compared to mares that were continuously under the influence of progesterone. The results demonstrated the importance of luteal progesterone to mobility, fixation, orientation, and survival of the embryonic vesicle.  相似文献

A previous study showed that noncyclic dairy cows treated with 10 microg of GnRH and a progesterone-releasing CIDR insert on Day 0, 25 mg of PGF2alpha and CIDR removal on Day 7, followed by 1 mg estradiol benzoate on Day 9 for those cows that still had not shown estrus (CGPE program) had higher conception rate (47% vs. 29%) than cows treated only with CIDR and estradiol benzoate as above (CE program). This study was to investigate the mechanisms by which the CGPE program improved conception rate compared with the CE program. Sixteen noncyclic Holstein-Friesian cows were randomly assigned to 2 groups balanced for the size and growth pattern of the dominant follicles, which were determined by ultrasonography over a 3-d period. One group received the above CGPE treatment, and the other group received the CE treatment. Follicular and luteal development were monitored by daily ultrasonography. Blood samples were collected daily from Day -2 to Day 11, and thereafter milk samples were collected thrice weekly for a further 24 d. Blood and milk samples were analyzed for progesterone. The GnRH treatment induced ovulation in 7 of 8 cows, resulting in elevated (P<0.05) progesterone concentrations between Days 4 and 7 for cows in the CGPE group. All induced CL underwent luteolysis by 24 h after PGF2alpha treatment. Within 5 d of CIDR removal, 7 of 8 cows in both the CE and CGPE groups ovulated. The interval from emergence of the ovulatory follicle to ovulation was similar (P=0.32) but less (P<0.05) variable for the CGPE group (9.0+/-0.3 d) compared with the CE group (10.3+/-1.2 d). Progesterone concentration in milk samples was similar between the two groups up to 10 d after ovulation. In summary, the GnRH treatment induced ovulation or turnover of dominant follicles, induced a synchronized initiation of a new follicular wave, and increased the progesterone concentration from 4 d after treatment. These could be the reasons for the increased conception rate of cows treated with the CGPE program.  相似文献

Practical estrus synchronization schemes are needed for mares. The Ovsynch synchronization protocol for cattle involves the administration of gonadotropin-releasing hormone (GnRH) to induce ovulation or luteinization of dominant follicles during the luteal phase and prostaglandin 7 days later to cause regression of any luteal tissue and development of a preovulatory follicle. An Ovsynch-type synchronization program potentially could be developed for horses if luteinization or ovulation of diestrous follicles occurred in response to GnRH treatment. The objective of this study was to determine if administration of the GnRH agonist, deslorelin acetate, on Day 8 or 12 postovulation would induce luteinization or ovulation of diestrous follicles in the mare. The model used was cycling mares maintained in an artificial luteal phase by administration of a synthetic progestin following prostaglandin-induced luteal regression. On the day of ovulation, 21 light horse mares were randomly assigned to one of three groups: (1) no GnRH, altrenogest from Days 5 to 15 postovulation with prostaglandin on Day 15; (2) GnRH on Day 8, altrenogest from Days 5 to 15 with prostaglandin given on Day 6 to induce luteolysis of the primary corpus luteum, an implant containing 2.1mg of deslorelin acetate inserted on Day 8 and removed on Day 10, with a second prostaglandin treatment on Day 15; (3) GnRH on Day 12, altrenogest from Days 9 to 19, prostaglandin on Day 10, a deslorelin acetate implant injected on Day 12 (subsequently removed on Day 14), and a second dose of prostaglandin administered on Day 19. Follicular development was monitored every other day from Day 5 until a 30-mm sized follicle was observed, and then daily to detection of ovulation. Serum progesterone concentrations were determined daily for 12 consecutive days. Progesterone concentrations in Group 1 remained elevated until approximately Day 12 postovulation. Prostaglandin administration on Day 15 resulted in complete luteolysis in all seven mares. In Group 2, progesterone concentrations in six of seven mares declined to baseline after prostaglandin treatment. No increase in serum progesterone was noted in any of the six mares that were given GnRH on Day 8, including three mares that had diestrous follicles > or =30mm in diameter at the time of treatment. Similarly, progesterone concentrations in six of seven mares in Group 3 declined to baseline after prostaglandin and there was no increase in progesterone after administration of GnRH on Day 12. No ultrasound evidence of luteinization or ovulation of diestrous follicles were noted after GnRH administration in any mares of Group 2 or 3. In conclusion, administration of the GnRH agonist deslorelin acetate to mares failed to induce luteinization or ovulation of diestrous follicles. The effects of diet on mineral concentrations in serum, liver and endometrium were determined at points in the reproductive cycle in heifers. Dietary treatments extended 134 d and included feeding a basal hay (negative control), basal hay with concentrate (feed control), feed control with phosphorus and feed control with both phosphorus and trace minerals. Samples of serum and liver were taken at the beginning and end of the trial. Within an estrous cycle during the trial (endometrial biopsy) the cows were sampled either at Day 1 or Day 12 for determining progesterone levels and mineral elements in the blood, liver and endometrial tissue. Trace element concentrations in serum and liver did not differ among collection periods pretrial, endometrial biopsy and post breeding nor among treatment groups. However, endometrial tissue concentrations of copper, manganese and zinc were higher at Day 1 than at Day 12 (P < 0.05) in reverse of serum progesterone, which was higher at Day 12 (P < 0.05). Supplemental trace minerals appeared to increase concentrations of copper (P < 0.20), manganese (P < 0.10) and zinc (P < 0.20) at Day 1 but decrease concentrations of these same elements at Day 12 (P < 0.05, P < 0.10 and P < 0.05, respectively). The large differences in trace element concentrations observed in endometrial tissue at the estral phases and under different diets suggest the possible importance of trace elements and trace element nutrition in fertilization and (or) embryo survival.  相似文献

One strategy for improving fertility in cattle is mid-cycle administration of GnRH to increase progesterone secretion and delay luteolysis. This strategy might be especially useful during hot weather because heat stress increases uterine prostaglandin release and reduces development of the elongating embryo. A series of experiments was conducted to test the efficacy of GnRH for increasing fertility. There was no effect of administration of 100 microg GnRH at Day 11 after anticipated ovulation on pregnancy rates in virgin heifers subjected to timed artificial insemination (TAI) during the summer. Similarly, there was no beneficial effect of administration of GnRH at Day 11 after anticipated ovulation on pregnancy rates of lactating cows subjected to TAI in summer and winter. Three experiments tested effects of injection of GnRH at Days 14 or 15 after anticipated ovulation on pregnancy rates of lactating cows. The first experiment used 477 lactating cows subjected to TAI. Cows receiving GnRH at Day 14 had higher pregnancy rates in both summer and winter than cows receiving vehicle (20.3 versus 12.7%, P<0.02). When this experiment was repeated during summer with 137 cows, there was a negative effect of GnRH treatment at Day 14 on pregnancy rate. In the third experiment, lactating cows during summer were inseminated at detected estrus and cows were assigned to treatment with either GnRH or vehicle at Days 14 or 15 after insemination. Pregnancy rates were 25.6% (32/125) for cows receiving vehicle, 20.7% (19/92) for cows receiving GnRH at Day 14, and 20.3% (16/79) for cows receiving GnRH at Day 15. In conclusion, GnRH administration at Days 11-15 after anticipated ovulation or estrus did not consistently increase pregnancy rates in either cool or warm seasons.  相似文献

The levels of progesterone and estrogen secretion were studied in relationship to the superovulatory response in Jersey cows. Progesterone and estrogen concentrations were measured in superovulated Jersey cows with the objective of correlating the patterns of steroid secretion with embryo yield and quality. Pregnant mare serum gonadotropin (PMSG) was used in combination with prostaglandin F(2) alpha analogue to induce superovulation in 18 multiparous, cyclic cows. Serum progesterone and estradiol levels from cows which exhibited estrus within 24 to 48 h after prostaglandin administration (n=13) were used to estimate the superovulatory response. Sex steroid concentrations at the day of estrus (Day 0) was a strong indicator of embryo yield. Progesterone was negatively (r=-0.56) and estrogen positively (r=0.80) correlated to the number of embryos collected. Dramatic increase in progesterone from Day 0 to Day 7 was a significant indicator of embryo yield. A higher rise of estrogen in the follicular phase was an indicator of a larger number of growing follicles and, consequently, better superovulatory response. Nonresponding animals did not show any significant change in the hormonal profile from the day of PMSG treatment to the day of embryo collection. The estimation of progesterone and estradiol concentrations, simultaneously, gave a more objective prediction of embryo yield.  相似文献

Patterns of uterine microbial recovery and serum concentrations of prostaglandin F(2alpha) metabolite (PGFM), cortisol, luteinizing hormone (LH) and progesterone (P4) were determined in normal postpartum dairy cows (n = 12) and in postpartum cows that developed cystic ovaries (n = 12). Palpation per rectum, ultrasound examinations of the reproductive tract and endometrial swab harvest were carried out once every 4 d during the first month postpartum. Daily blood samples were collected during the first 30 d postpartum and every other day until Day 60 postpartum for hormone assay. Bacterial growth densities of cultures of uterine swabs were high in cows that developed cystic ovaries. Cysts were detected between Day 8 and 16 postpartum and persisted for a period of 18.6 +/- 9.9 d (range 8 to 41 d), followed by spontaneous regression and ovulations. Serum PGFM and cortisol were elevated for several days prior to detection of cystic ovaries but not prior to first postpartum ovulations. Serum LH concentrations in cystic cows were at basal concentrations prior to discovery of cysts. The results suggest that postpartum intrauterine infections may provoke increased secretion of prostaglandin F(2alpha) and cortisol associated with the formation of cystic ovaries in dairy cows.  相似文献
