Fungal mycelia are exposed to heterogenic substrates. The substrate in the central part of the colony has been (partly) degraded, whereas it is still unexplored at the periphery of the mycelium. We here assessed whether substrate heterogeneity is a main determinant of spatial gene expression in colonies of Aspergillus niger. This question was addressed by analyzing whole-genome gene expression in five concentric zones of 7-day-old maltose- and xylose-grown colonies. Expression profiles at the periphery and the center were clearly different. More than 25% of the active genes showed twofold differences in expression between the inner and outermost zones of the colony. Moreover, 9% of the genes were expressed in only one of the five concentric zones, showing that a considerable part of the genome is active in a restricted part of the colony only. Statistical analysis of expression profiles of colonies that had either been or not been transferred to fresh xylose-containing medium showed that differential expression in a colony is due to the heterogeneity of the medium (e.g., genes involved in secretion, genes encoding proteases, and genes involved in xylose metabolism) as well as to medium-independent mechanisms (e.g., genes involved in nitrate metabolism and genes involved in cell wall synthesis and modification). Thus, we conclude that the mycelia of 7-day-old colonies of A. niger are highly differentiated. This conclusion is also indicated by the fact that distinct zones of the colony grow and secrete proteins, even after transfer to fresh medium.  相似文献

Conidia of Aspergillus niger 20 Osm producing extracellular inulinase were immobilized on pumice stones or polyurethane sponge and used in repeated-batch processes. Some factors affecting inulinase biosynthesis by the mycelium A. niger immobilized on pumice stones were investigated. Maximal inulinase production occurred in 50 ml of medium containing 0.5 g of carrier at 30 °C, pH 6.0 and at an agitation speed of 200 rpm. This procedure enabled repeated-batch enzyme production and as many as six subsequent 24 h batches could be fermented by using the same carrier. This is the first report on inulinase biosynthesis by mycelium of A. niger immobilized on polyurethane sponge using unconventional oxygenation of culture which ensures that the dissolved oxygen concentration remains constant.  相似文献

Lytic enzyme activity in autolysing mycelium of Aspergillus niger   总被引:1,自引:0,他引:1  
We have studied changes in the activity of some lytic enzymes contained in mycelium of Aspergillus niger in cultures relative to the autolytic phase of growth. Acid phosphatase, polygalacturonidase and alpha-amylase activity reached its highest level (40.7, and 8 U/sample, respectively) at the initiation of the autolytic phase of growth. 1.3-beta-Glucanase and beta-N-acetylglucosaminidase reached its highest level (3.5 and 2 U/sample, respectively) during the first days of autolysis. Alkaline phosphatase, cellulase, invertase, esterase, chitinase and proteolytic activity is also present in autolysing mycelium of A. niger, though comparatively low. Their maximum activity coincided with the beginning of the autolytic phase of growth. In all enzymes studied here, as autolyis proceeded, enzyme activity decreased by about 90%. Only esterase activity remained nearly constant throughout the whole period of autolysis described here.  相似文献

A simple method is described for the immobilization of Aspergillus niger GIV-10 which produces an extracellular glucose oxidase. A. niger conidia were immobilized on sintered glass Raschig rings, pumice stones or polyurethane foam. Mycella growing out from the spores produced extracellular glucose oxidase: the highest production was with the pumice stone carrlers. This technique facilitates the growth of the filamentous cultures in the spongy structure of a support with continuous accumulation of biomass. After 24 to 36 h, a culture liquid with 2.7 to 3.1 U of glucose oxidase/ml was obtained. This procedure also made possible repeated batch enzyme production and as many as 25 subsequent 24-h batches could be fermented by using the same carrier with only a small loss of glucose oxidase activity.The authors are with the Institute of Microbiology, M. Aspergillus niger is an important cell factory for the industrial production of enzymes. These enzymes are released into the culture medium, from which they can be easily isolated. Here, we determined with stable isotope dimethyl labeling the secretome of five concentric zones of 7-day-old xylose-grown colonies of A. niger that had either or not been treated with cycloheximide. As expected, cycloheximide blocked secretion of proteins at the periphery of the colony. Unexpectedly, protein release was increased by cycloheximide in the intermediate and central zones of the mycelium when compared to nontreated colonies. Electron microscopy indicated that this is due to partial degradation of the cell wall. In total, 124 proteins were identified in cycloheximide-treated colonies, of which 19 secreted proteins had not been identified before. Within the pool of 124 proteins, 53 secreted proteins were absent in nontreated colonies, and additionally, 35 proteins were released ≥4-fold in the central and subperipheral zones of cycloheximide-treated colonies when compared to nontreated colonies. The composition of the secretome in each of the five concentric zones differed. This study thus describes spatial release of proteins in A. niger, which is instrumental in understanding how fungi degrade complex substrates in nature.  相似文献

Summary The immobilization in polyacrylamide gel (PAAG) of the Aspergillus niger mycelium, which has the activity of hydroxylating indolyl-3-acetic acid (IAA) at 4-, 5-, and 6-positions of the indole nucleus, was studied. To preserve the hydroxylating activity, the immobilization should be performed at 5°C–10°C for 5–10 min. The hydroxylating activity of the A. niger mycelium entrapped in PAAG attained 70%–80% of that of free cells. The IAA transformation in the presence of sodium desoxycholate, polyethylene-glycol-400 (PEG-400), Span-60 or preincubation of granules entrapping mycelia in the presence of Tween-80 or PEG-400 not only double the hydroxylation rate but stabilize the activity as well. Gels entrapping mycelia may be used five or six times without altering activity. Incubation of gels with mycelium in the nutrient medium also increases and stabilizes the hydroxylating activity. In aerated columns, it is possible to obtain continuous hydroxylation of IAA, at a concentration of 0.5 g/l, by the immobilized mycelium of A. niger. The yield of hydroxy derivatives reached 70%, the activity remaining unaltered during 15 days' operation of the column.  相似文献

J. FIEDUREK, J. SZCZODRAK AND J. ROGALSKI. 1995. A simple method for the immobilization of Aspergillus niger mycelium producing polygalacturonase (PG) and pectinesterase (PE) is described. Fungal conidia were immobilized on wheat, rye, barley, peas, buckwheat and mustards seeds. Spongy mycelia overgrowing the seed surfaces on mineral medium with pectin produced extracellular PG and PE; the highest production was reached on the wheat carrier. Some of the variables influencing the enzymatic activity have been optimized. After every 24 h, a culture liquid with 6˙8–7˙8 U of PG ml−1 and 7˙0–10˙1 U of PE ml−1 was obtained. This procedure also made possible repeated batch enzyme production and, as many as eight subsequent 24-h batches could be fermented by using the same carrier without any loss of PG activity. The addition of sodium orthovanadate (1 mmol) into the medium with pectin caused a significant increase in PG and PE activity produced by free cells of A. niger (by 1˙59-fold and 1˙67-fold respectively), and only 0˙47-fold of PG activity in case of the immobilized mycelium.  相似文献

Adaptation to a changing environment is essential for the survival and propagation of sessile organisms, such as plants or fungi. Filamentous fungi commonly respond to a worsening of their growth conditions by differentiation of asexually or sexually produced spores. The formation of these specialized cell types is, however, also triggered as part of the general life cycle by hyphal age or density. Spores typically serve for dispersal and, therefore, translocation but can also act as resting states to endure times of scarcity. Eukaryotic differentiation in response to environmental and self-derived signals is commonly mediated by three-tiered mitogen-activated protein (MAP) kinase signaling cascades. Here, we report that the MAP kinase Fus3 of the black mold Aspergillus niger (AngFus3) and its upstream kinase AngSte7 control vegetative spore formation and secondary metabolism. Mutants lacking these kinases are defective in conidium induction in response to hyphal density but are fully competent in starvation-induced sporulation, indicating that conidiation in A. niger is triggered by various independent signals. In addition, the mutants exhibit an altered profile of volatile metabolites and secrete dark pigments into the growth medium, suggesting a dysregulation of the secondary metabolism. By assigning the AngFus3 MAP kinase pathway to the transduction of a potentially self-derived trigger, this work contributes to the unraveling of the intricate signaling networks controlling fungal differentiation. Moreover, our data further support earlier observations that differentiation and secondary metabolism are tightly linked in filamentous fungi.  相似文献

The formation of hyphae that grow solely by apical extension is a defining feature of filamentous fungi. Hyphal morphogenesis involves several key steps, including the establishment and maintenance of a stable polarity axis, as well as cell division via the deposition of septa. Several filamentous fungi have been employed in attempts to decipher the mechanisms underlying these steps. Amongst these fungi, Aspergillus nidulans has proven to be a particularly valuable model. The genetic tractability of this fungus coupled with the availability of sophisticated post-genomics resources has enabled the identification and characterization of numerous genes involved in hyphal morphogenesis. Here, we summarize current progress towards understanding the function of these genes and the mechanisms involved in polarized hyphal growth and septation in A. nidulans. The effect of different carbon sources on the sugar concentration and composition of theAspergillus niger mycelium has been studied at different days of incubation. No positive correlation could be drawn among dry weight, growth rate, and concentration of sugar and with substrate and sugar composition of mycelium. Glucose was the predominant sugar in all the cases.

Summary The possibility that adenosine 3,5 monophosphate exerts an effect on citric acid production by Aspergillus niger by influencing pellet morphology has been investigated. The effect of pH and inoculum size on pellet formation, citric acid production, and intracellular and extracellular cAMP levels were studied. High levels of intracellular and extracellular cAMP in the later stages of the fermentation, the period of maximum citric acid formation, were associated with those treatments which gave pellets of intermediate size. The highest cAMP levels were associated with those treatments which gave the highest citric acid titre. It was concluded that high cAMP levels are principally associated with an optimum physiological state for citric acid production and that cAMP levels do not vary directly with pellet size.  相似文献

Gluconic acid was produced in repeated batch processes with Aspergillus niger AM-11, immobilized in pumice stone particles using an unconventional oxygenation of culture media based on the addition of H2O2, decomposed by catalase to O2 and water. The highest gluconic acid productivity of 8.2 g l–1 h–1 was reached with 30 g immobilized mycelium per 150 ml, 10% (w/v) glucose, at 24 °C and pH 6.5, with O2 at 100% saturation. The immobilized mycelium was successfully reused up to 8 times in 1-h batches with only a slight loss (11%) of gluconic acid productivity.  相似文献

A. niger C grown on wheat bran extract medium provided culture supernatants with the highest enzymatic activities. Aspergillus niger produces two extracellular glucoamylases (GAI of Mr 85 300 and GAII of Mr 77 600) separable on DEAE-cellulose. The enzymes differes in electrophoretic mobility, thermostability and substrate specificity. The GAI/GAII ratio depends on the concentration and form of nitrogen (nitrate or ammonium) in the culture medium. Proteinase VIII from Bacillus subtilis converts GAI to a form showing properties similar to those of GAII. Possible proteolytic degradation of GAI to GAII by Asp. niger endogenous proteinase(s) is suggested.  相似文献
