The paraganglia of adult man were studied using the formaldehyde-induced fluorescence (FIF) method for histochemical characterization of biogenic monoamines. Microspectrofluorimetry was used to record the emission spectra and fluorescence intensities of the paraganglionic cells. The study of samples from six patients showed that well vascularized paraganglia were widely distributed throughout the retroperitoneal spaces. The paraganglia exhibited strong FIF with the spectral characteristics of monamines. Treatment with HC1 caused an increase in the fluorescence intensity of the paraganglia and a simultaneous shift of the emission maximum from 480--495 nm. This change suggests the presence of high concentrations of tryptophyl-containing peptides and is not due to monoamines. The possibility of a dual endocrine function for the paraganglia is discussed.  相似文献

The distribution and morphology of chromaffin cells in the para-aortic region and in the ganglia of the paravertebral sympathetic chain was studied with fluorescence histochemistry and electron microscopy. 2. Four types of chromaffin cell were distinguished largely on the basis of their vesicular content: Type I cells contain large, electron-dense vesicles (600-7000 A) and are comparable to noradrenaline-containing cells in the adrenal gland, Type II cells contain large, vesicles (600-7000 A) that are filled with a less electron-dense material than that in Type I cells and are comparable to adrenaline-containing cells in the adrenal gland, Type III cells contain smaller vesicles (1000-3000 A) that are incompletely filled with an electron-dense material and may represent cells that have been depleted of their catecholamines by stimulation, Type IV cells are clearly different from the other three cell types with respect to the size and appearance of the vesicles (1000-1500 A), nuclei and rough endoplasmic reticulum and may represent immature sympathetic neurons. 3. Nerve profiles, identified as cholinergic, were found in close apposition with all four cell types. No examples of a close association between processes of chromaffin cells and sympathetic neurons were found.  相似文献

Summary The increase in numbers of extra-adrenal chromaffin cells of abdominal paraganglia in senescent F344 rats was investigated by 5-bromo-2-deoxyuridine immunocytochemistry. A monoclonal antibody raised against 5-bromo-2-deoxyuridine was used to react with tissue-sections of paraganglia taken from 28-month-old animals given weekly injections of the thymidine analog over a 14-week period. No immunoreactivity was detected in the extra-adrenal chromaffin cells, whereas control sections of intestinal epithelium showed abundant immunoreactivity. Also, the profile for immunoreactivity of the glucocorticoid receptor in relation to age was compared between extra-adrenal and adrenal chromaffin cells, which share cytological characteristics, but not the increase associated with senescence. In the extra-adrenal chromaffin cells, the intensity of receptor immunostaining was unchanged, while in the adrenal chromaffin cells it decreased with age. These results indicate that hypertrophy of the paraganglia in aged F344 rats is not due to the proliferation of extra-adrenal chromaffin cells. Instead, they suggest that the chromaffin cell phenotype may be induced in pre-existing cells and that the expression of the glucocorticoid receptor has an intrinsic role in this change.  相似文献

Summary The abdominal vagal paraganglia of the rat consist of small groups of cells, interspersed by blood vessels and nerve bundles and lying close to, or within, the vagus nerve or its branches. Each cell group consists of 2–10 Type I cells incompletely invested by 1–3 satellite cells. Type I cells are characterised by the presence of numerous dense-cored vesicles in their cytoplasm and may exhibit synaptic-like contact with each other.Small efferent nerve endings make synaptic contacts with Type I cells. Larger cup-shaped afferent nerve endings also make synaptic contacts of two kinds with Type I cells. Nerve-nerve synapses are often seen within or close to paraganglia.Attention is drawn to the close similarity of fine structure of abdominal vagal paraganglia, carotid body and small intensely fluorescent cells of the superior cervical ganglion in rats. Possible functional implications of this morphological similarity are discussed.  相似文献

Small granule chromaffin cells (SGC cells) were identified in the adrenal medulla of adult dogs. They were small in size and usually showed a high nucleo-cytoplasmic ratio. Cytoplasmic projections were occasionally observed in some of these cells. They contained a variable number of small secretory granules with diameters ranging from 70 to 300 nm, but mostly from 100 to 200 nm. The densities of the secretory granules were variable, ranging from highly dense to less dense. These adrenal SGC cells were rich in free ribosomes and polysomes, but were relatively poor in other cell organelles. Chromaffin cells which were intermediate in their characteristics (IM cells) between the SGC cells and the typical A and N cells were also identified. These IM cells contained both highly electron dense and less dense granules in various proportions. The IM cells were classified into two subgroups, according to the proportions of adrenaline type granules and noradrenaline type granules. One group resembled A cells (IM-A cells) and the other resembled N cells (IM-N cells). Light microscopic histochemical studies of A cells stained with the ammoniacal silver solution demonstrated that they contained a small number of darkly stained granules. Electron microscopic cytochemistry revealed that the electron dense granuls in the SGC cells, IM cells and A cells reacted positively with both the potassium dichromate solution at pH 4.1 and the ammoniacal silver solution.  相似文献

Only limited information is available on the stereochemistry of the in vivo distribution of beta-receptor-blocking drugs. In this study we determined the levels of the propranolol enantiomers in plasma, cerebrospinal fluid (CSF) and central nervous system (CNS), and peripheral tissues in the dog following an intravenous dose of a deuterium-labeled pseudoracemate. The appearance of the propranolol enantiomers in the CSF was rapid and nonstereoselective, with maximum concentrations reached at 15 min after dosing. The levels of the enantiomers in both CSF and plasma then declined in a parallel biphasic fashion, with a terminal t1/2 of about 125 min. Except for an early high CSF/plasma concentration ratio of 0.35, the CSF propranolol levels corresponded to the unbound concentration in plasma, CSF/plasma 0.20. All areas of the brain showed a similar uptake of propranolol, with a tissue concentration exceeding that in plasma about 10-fold during the terminal phase of elimination. The uptake of propranolol by peripheral tissues varied widely, ranging from a 50-fold accumulation by the lungs compared to plasma to no accumulation by adipose tissue. However, as for the CSF, there was no evidence of stereoselective uptake of propranolol by any CNS or peripheral tissue except for the liver. A significantly higher level of (+)-vs. (-)-propranolol in liver tissue presumably was a reflection of stereoselective hepatic metabolism of (-)-propranolol by this tissue. The slight stereoselectivity in plasma binding of propranolol known to exist in the dog had no significant influence on tissue or CSF distribution.  相似文献

In this study, pollen grains and anatomical features of Turkish lilies were investigated under the electron (SEM) and light (LM) microscope. LM and SEM observations showed that the pollen grains are monosulcate, heteropolar, elliptical in polar view and oblate. Numerical results based on combined palynological and anatomical characters were discussed and compared with traditional taxonomic treatments. It was found that the midrib shape, mesophyll type, P/E (polar/equatorial), sulcus length, and lumina width are the most valuable traits in separating the examined taxa. The numerical analysis showed that Lilium candidum L. differs from the rest Turkish Lilium and also confirmed a close relationship between L. szovitsianum Fisch. & Avé-Lall. and L. armenum Miscz. ex Grossh. Also this study is the first report dealing with anatomical and palynological features of all Turkish lilies.  相似文献

An inventory of the distribution of thoracic glomus tissue (thoracic chemoreceptor tissue, aortic bodies) has been derived from serial sections in foetal, newborn and adult rats. In contrast to other common experimental species, such as the cat and dog, aortic bodies are an inconstant feature and, on the whole, are scarce in the rat. These morphological findings correlate well with experimental studies, indicating an absence or paucity of thoracic chemoreceptor activity in the rat. The results are discussed in terms of their physiological and pathophysiological implications and a revised model of the peripheral arterial chemoreceptor system is presented.  相似文献

Glandular structures at the sternites of 31 species of Paederinae (Coleoptera : Staphylinidae) are described in detail for the first time. A median gland of unknown biological significance is situated at the front margin of the 4th sternite in Paederus and Paederidus (Paederus s.l.). A gland tissue at the posterior border of the antecosta opens into a reservoir situated between the antecosta and the intersegmental membrane. Each of the numerous glandular units obviously consist of 2 secretory cells that are drained by a tubule. They are thus related to dermal glands. Two types of pores, single pores and pore plates, are dispersed over the body, but are aggregated in the vicinity of the aperture of the reservoir. In Rugilus, 2 hitherto unknown glands of the same shape are located at the front margins of sternites 4 and 5. In contrast to the glands of Paederus s.l., there are big pores under the gland tissue and the opening area lies in a pit. One further species, Stilicosoma rufipes, was recently classified in the genus Rugilus, but does not have any glands, and therefore the genus Stilicosoma Casey, which was once named for S. rufipes, is re-established. Ochthephilum fracticorne does not have a reservoir, but otherwise combines features of both the glands of Paederus s. l. and those of Rugilus. This link points out the possible derivation of these glands from a common origin. All species of Lathrobiina have accumulations of tubules at the front margins of at least the 4th sternites, and thus represent a rather primitive stage in the development of the glands, while the lack of glandular peculiarities at the 4th sternites in Medonina is plesiomorphic. With respect to a special type of dermal tubules, the genera Medon, Pseudomedon, and Sunius are distinguished from Lithocharis, hence denoting a clear dividing line between the subtribes Medonina and Lithocharina. These features as well as the glands themselves, provide the basis for clarifying the largely unknown phylogeny of Paederinae.  相似文献

Summary An indirect gold-labeling method utilizing the lectin from Limax flavus was employed to characterize the subcellular distribution of sialic acid in glycoconjugages of the salamander olfactory mucosa. The highest density of lectin binding sites was in secretory vesicles of sustentacular cells. Significantly lower densities of lectin binding sites were found in secretory granules of acinar cells of both Bowman's and respiratory glands. Lectin binding in acinar cells of Bowman's glands was confined primarily to electron-lucent regions and membranes of secretory granules. In the olfactory mucus, the density of lectin binding sites was greater in the region of mucus closest to the nasal cavity than in that closest to the epithelial surface. At the epithelial surface, the density of lectin binding sites associated with olfactory cilia was 2.4-fold greater than that associated with microvilli of sustentacular cells or non-ciliary plasma membranes of olfactory receptor neurons, and 7.9-fold greater than non-microvillar sustentacular cell plasma membranes. Lectin binding sites were primarily associated with the glycocalyx of olfactory receptor cilia. The cilia on cells in the respiratory epithelium contained few lectin binding sites. Thus, sialylated glycoconjugates secreted by sustentacular cells are preferentially localized in the glycocalyx of the cilia of olfactory receptor neurons.  相似文献

The distribution of glutamate-like immunore-activity in the thoracic and abdominal ganglia of the locust was studied using two polyclonal antibodies against glutamate. Because glutamate is a precursor of the inhibitory transmitter -amino butyric acid (GABA) the distribution of immunostaining by antibodies against glutamate and GABA was closely compared in adjacent serial sections. When the antibodies were used at optimal dilutions there was no overlap in the distribution of immunostaining for glutamate and GABA. In the pro- and mesothoracic ganglia 360–400 somata are immunoreactive for glutamate, while in the metathoracic ganglion about 600 somata were stained. These range in diameter from 10–100 m in diameter and include the majority of the large somata in these ganglia. Bundles of primary neurites emerging from these large somata can be traced through the neuropile. Most of the bundles correspond to the known paths of motor neurone primary neurites. In addition the T-tract is also immunolabelled. The free abdominal ganglia each contain 80–100 somata ranging in size from 10–45 m while the terminal ganglion contains about 250 somata, 10–60 m in diameter.  相似文献

Studies on the collections of the Musée Royal de l'Afrique Centrale and the Institut Royal des Sciences Naturelles de Belgique have shown that Xenopus kigesiensis Tinsley, 1973 is synonymous with X. vestitus Laurent, 1972. Previously unrecognized representatives of the species have been found in the museum collections and these considerably enlarge the known geographical distribution of X. vestitus. Samples have been recorded principally from highland areas adjacent to the western Rift in Zaire, Uganda and Ruanda; comprehensive morphological analysis demonstrates relatively limited intraspecific variation throughout the known range.
The distribution of X. vestitus overlaps with that of three other Xenopus taxa: X. laevis victorianus, X. bunyoniensis and a new Xenopus species. There is evidence from a large number of collections from widely separated localities that X. vestitus is commonly sympatric with X. l. victorianus. On the other hand, it is postulated that X. vestitus has recently invaded Lake Mutanda, Uganda, and has replaced the original X. bunyoniensis population during the past 30 years. The museum record provides a potential origin of this invasion: X. vestitus populations are well documented at Rutshuru, only 25 km downstream from Lake Mutanda.
A comparison of the morphological characters of X. vestitus, X. l. victorianus and X. bunyoniensis is presented to provide information on the systematic relationships and possible ecological adaptations of the overlapping species.  相似文献
