Little information is known on what the magnitude of nitrogen (N) processed by ectomycorrhizal (ECM) fungal species in the field. In a common garden experiment performed in a northern California oak woodland, we investigated transfer of nitrogen applied as 15NH4 or 15NO3 from leaves to ectomycorrhizal roots of three oak species, Quercus agrifolia, Q. douglasii, and Q. garryana. Oak seedlings formed five common ectomycorrhizal morphotypes on root tips. Mycorrhizal tips were more enriched in 15N than fine roots. N transfer was greater to the less common morphotypes than to the more common types. 15N transfer from leaves to roots was greater when , not , was supplied. 15N transfer to roots was greater in seedlings of Q. agrifolia than in Q. douglasii and Q. garryana. Differential N transfer to ectomycorrhizal root tips suggests that ectomycorrhizal morphotypes can influence flows of N from leaves to roots and that mycorrhizal diversity may influence the total N requirement of plants.  相似文献

Two new species of Lirula (L. japonica and L. exigua) on Abies mariesii collected in subalpine areas of northern Japan are described as members of Rhytismatales, Discomycetes. Lirula japonica causes needle cast in fir, but L. exigua seems to occur on the needles of physically damaged twigs. Morphological characteristics of both species are discussed.  相似文献

Summary   Geissois sensu stricto (i.e. excluding the Australian species of this genus) has 19 species restricted to islands in the south-west Pacific and is the sole group in Cunoniaceae in which the stipules are intrapetiolar (axillary) in the adult foliage. In apical buds, paired opposite stipules are coherent round their margins and are either clearly intrapetiolar in relation to the most distal pair of developed leaves (Type I, most species), or they appear interpetiolar due to the abortion of the pair of leaves associated with them and the failure of the internode proximal to them to elongate (Type II, a few species). Stipules are often accrescent and the largest in the family occur in this group, reaching 10 cm long in some species. The ornithophilous racemose inflorescences of members of this group normally consist of either axillary triads (G. hirsuta only) or apparently simple, unbranched racemes which are ramiflorous or occasionally axillary. However, bract scars on the axes proximal to the flowers show that both types of inflorescence module usually consist of two metamers. The structure of the inflorescences and stipules in Geissois sensu stricto supports the distinction between this group and the Australian species traditionally included in Geissois sensu lato.  相似文献

Sugar and sugar alcohol concentrations were analyzed in subcellular compartments of mesophyll cells, in the apoplast, and in the phloem sap of leaves of Plantago major (common plantain), Plantago maritima (sea plantain), Prunus persica (peach) and Apium graveolens (celery). In addition to sucrose, common plantain, sea plantain, and peach also translocated substantial amounts of sorbitol, whereas celery translocated mannitol as well. Sucrose was always present in vacuole and cytosol of mesophyll cells, whereas sorbitol and mannitol were found in vacuole, stroma, and cytosol in all cases except for sea plantain. The concentration of sorbitol, mannitol and sucrose in phloem sap was 2- to 40-fold higher than that in the cytosol of mesophyll cells. Apoplastic carbohydrate concentrations in all species tested were in the low millimolar range versus high millimolar concentrations in symplastic compartments. Therefore, the concentration ratios between the apoplast and the phloem were very strong, ranging between 20- to 100-fold for sorbitol and mannitol, and between 200- and 2000-fold for sucrose. The woody species, peach, showed the smallest concentration ratios between the cytosol of mesophyll cells and the phloem as well as between the apoplast and the phloem, suggesting a mixture of apoplastic and symplastic phloem loading, in contrast to the herbal plant species (common plantain, sea plantain, celery) which likely exhibit an active loading mode for sorbitol and mannitol as well as sucrose from the apoplast into the phloem.  相似文献

Among 67 endophytic fungi isolated from Quercus variabilis, 53.7% of endophytic fungal fermentation broths displayed growth inhibition on at least one test microorganism, such as pathogenic fungi (Trichophyton rubrum, Candida albicans, Aspergillus niger, Epidermophyton floccosum, Microsporum canis) and bacteria (Escherichia coli, Bacillus subtilis, Pseudomonas fluorescens). Moreover, 19.4% of strains showed a broader antimicrobial spectrum, such as Aspergillus sp., Penicillium sp., Alternaria sp., 20.9% of strains showed strong inhibition (+++) to pathogenic bacteria, while only 7.5% displayed that to test fungi. The most active antifungal strain I(R)9-2, Cladosporium sp. was selected and fermented. From the broth, a secondary metabolite, brefeldin A was obtained. This is the first report on the antimicrobial potentials of endophytic fungi residing in Q. variabilis and isolation of brefeldin A produced by Cladosporium sp.  相似文献

Anagrus epos Girault (Hymenoptera: Mymaridae) is a candidate for a classical biological control program targeting the glassy-winged sharpshooter (GWSS), Homalodisca vitripennis (Germar) (Hemiptera: Cicadellidae), in California. Because mass production of GWSS is expensive and labor-intensive, a factitious host that is more economical to produce is desirable to mass produce A. epos for colonization and augmentation efforts. Here, we report the results of host specificity tests and potential rearing techniques for A. epos under laboratory conditions. Females discriminated and oviposited into eggs of seven cicadellid species: H. vitripennis, Circulifer tenellus (Baker), Erythroneura variabilis Beamer, Amblysellus grex (Oman), Graphocephala atropunctata (Signoret), Macrosteles severini Hamilton, and H. liturata Ball, and two cerambycid species: Phoracantha recurva Newman and P. semipunctata (F.). Anagrus epos successfully completed development in the eggs of H. vitripennis, C. tenellus, E. variabilis, A. grex, G. atropunctata, M. severini, and H. liturata. The use of a factitious host and potential nontarget effects of this generalist parasitoid are discussed.  相似文献

Two species of Leptographium were isolated from blue-stained sapwood of Pinus khasya and bark beetle galleries in pine trees near Chiang Mai, Thailand. Based on morphological observations, these two species were identified as L. pini-densiflorae and L. yunnanense. This is the first record of these fungi in Thailand. Leptographium yunnanense appeared to be associated with Polygraphus major. A powdery mildew fungus on leaves of Dalbergia cultrata var. cultrata (Fabaceae) collected at the Queen Sirikit Botanical Garden in northern Thailand is proven to be a new species of the genus Brasiliomyces and is described as B. chiangmaiensis sp. nov. with light and SEM micrographs. Differences in known Brasiliomyces species are discussed, and a key to species of this genus is provided.  相似文献

We examined the floral morph of tetraploid Ophiorrhiza japonica Blume var. amamiana Hatus. and diploid O. japonica var. japonica to elucidate the association of distyly and ploidy levels. Chloroplast DNA phylogeny was reconstructed to determine the number of tetraploidization events and floral morph shifts in O. japonica. All individuals of O. japonica var. amamiana proved to be long-homostylous, whereas O. japonica var. japonica was distylous with typical long- and short-styled flowers. Distyly is related to the ploidy level. The bagging treatment of flowers indicated that O. japonica var. amamiana is self-compatible and potentially automatically self-pollinating. In cpDNA sequencing analysis, no haplotype was shared between the two varieties. The cpDNA haplotype network displayed the monophyly of O. japonica var. amamiana, suggesting a single origin of this variety. Hence, both tetraploidization and the breakdown of distyly to homostyly in O. japonica var. amamiana likely occurred just once. Because O. japonica var. amamiana having the morphological and cytological entity is recognized as a single lineage and clearly separated from O. japonica var. japonica, this variety can be considered to be a distinct species. We therefore propose to raise O. japonica var. amamiana to the rank of species. The anamorphic fungus Sporothrix inflata, known as a soil-borne fungus with worldwide distribution, was isolated for the first time from the cortex and central cylinder of living and dead roots of healthy and diseased oak trees (Quercus robur and Q. petraea). Isolation frequencies of S. inflata from oak roots varied according to the health status of trees, oak species, study sites, soil depth and root diameter. Colony morphology and growth rate of isolates are influenced by colony age and type of culture medium.  相似文献

Chloroplast DNA polymorphism in four oak species (Quercus serrata, Q. mongolica var. crispula, Q. dentata and Q. aliena) was studied using collections from a total of 127 localities in Japan and South Korea on the basis of five intergenic spacers (trnD-trnT, trnT-trnL, rps14-psaB, trnS-trnT and trnQ-trnS). Although no variation existed in sequences among the four species, a single nucleotide (T/C) substitution in the trnQ-trnS intergenic spacer was found in all the four species, resulting in two haplotypes (T- and C-type). Phylogenetic analyses of the four species and related species showed that the C-type is derived and even likely of monophyletic origin, while the T-type is ancestral. Geographically, the T-type is widespread from South Korea to Japan, whereas the C-type is restricted to eastern Japan with rare exceptions. Eastern Japan approximately coincides with the distribution range of the boreal conifer forest during the last glacial maximum. Overall evidence suggests that the mutation from T- to C-type occurred in an individual of one of the four oak species and then was transferred to all the species by hybridization in eastern Japan, and that the Kanto District provided individuals with the C-type with a refugium during the last glacial maximum.Electronic Supplementary Material Supplementary material is available in the online version of this article at  相似文献

The entire (e) locus of tomato (Solanum lycopersicum L.) controls leaf morphology. Dominant E and recessive e allele of the locus produce pinnate compound and complex reduced leaves. Previous research had indicated that SlIAA9, an Aux/IAA gene, was involved in tomato leaf morphology. Down-regulation of SlIAA9 gene by antisense transgenic method decreased the leaf complex of tomato and converted tomato compound leaves to simple leaves. The leaf morphology of these transgenic lines was similar with leaf morphology of tomato entire mutant. In this paper, we report that a single-base deletion mutation in the coding region of SlIAA9 gene results in tomato entire mutant phenotypes.  相似文献

The maT clade of transposons is a group of transposable elements intermediate in sequence and predicted protein structure to mariner and Tc transposons, with a distribution thus far limited to a few invertebrate species. We present evidence, based on searches of publicly available databases, that the nematode Caenorhabditis briggsae has several maT-like transposons, which we have designated as CbmaT elements, dispersed throughout its genome. We also describe two additional transposon sequences that probably share their evolutionary history with the CbmaT transposons. One resembles a fold back variant of a CbmaT element, with long (380-bp) inverted terminal repeats (ITRs) that show a high degree (71%) of identity to CbmaT1. The other, which shares only the 26-bp ITR sequences with one of the CbmaT variants, is present in eight nearly identical copies, but does not have a transposase gene and may therefore be cross mobilised by a CbmaT transposase. Using PCR-based mobility assays, we show that CbmaT1 transposons are capable of excising from the C. briggsae genome. CbmaT1 excised approximately 500 times less frequently than Tcb1 in the reference strain AF16, but both CbmaT1 and Tcb1 excised at extremely high frequencies in the HK105 strain. The HK105 strain also exhibited a high frequency of spontaneous induction of unc-22 mutants, suggesting that it may be a mutator strain of C. briggsae.  相似文献

Haploid sporophytes of Osmunda claytoniana (2n = x = 22) were apogamously produced from calli when cultivated on a hormone-free medium. Flow cytometric analysis showed that ploidy chimeras were spontaneously produced in a haploid sporophyte of O. claytoniana and those of O. japonica that were obtained in the previous study. In the haploid sporophyte of O. claytoniana, a diploid pinnule and a partially diploid terminal segment were produced in a haploid pinna. In O. japonica, a haploid sporophyte yielded a diploid pinna in a haploid frond, and another haploid sporophyte yielded a diploid pinnule in a haploid pinna. Diploid chimeras were large in size and could be readily distinguished from other haploid parts of the fronds. It is likely that the chimeras were produced clonally from a single diploid cell that established chromosome doubling.  相似文献

P transposons belong to the eukaryotic DNA transposons, which are transposed by a cut and paste mechanism using a P-element-coded transposase. They have been detected in Drosophila, and reside as single copies and stable homologous sequences in many vertebrate species. We present the P elements Pcin1, Pcin2 and Pcin3 from Ciona intestinalis, a species of the most primitive chordates, and compare them with those from Ciona savignyi. They showed typical DNA transposon structures, namely terminal inverted repeats and target site duplications. The coding region of Pcin1 consisted of 13 small exons that could be translated into a P-transposon-homologous protein. C. intestinalis and C. savignyi displayed nearly the same phenotype. However, their P elements were highly divergent and the assumed P transposase from C. intestinalis was more closely related to the transposase from Drosophila melanogaster than to the transposase of C. savignyi. The present study showed that P elements with typical features of transposable DNA elements may be found already at the base of the chordate lineage. Fecundity and feeding of two introduced sibling biological control species, Galerucella calmariensis and G. pusilla (Coleoptera: Chrysomelidae) on purple loosestrife, Lythrum salicaria L. (Lythraceae) were compared at constant temperatures of 12.5, 15, 20, 25, and 27.5 °C. Larval feeding was also carried out at 30 °C, but at this temperature, larvae developed only to the L2 stage and none pupated. Thus, data for this temperature were not used in the analysis. There were significant species × temperature interactions in fecundity. Of the two species, Galerucella pusilla laid more eggs. Although egg production of both species was lowest at 12.5 °C and increased to 20 °C, at higher temperatures, the two species reacted differently. From 25 to 27.5 °C, egg production decreased for G. pusilla, but G. calmariensis fecundity peaked at 27.5 °C. Significant temperature × species × life-stage interactions were also observed in feeding. For each species, the amount of feeding varied with temperature and stage of development. Galerucella pusilla adults consumed more foliage at 15, 20, and 27.5 °C. However, at 12.5 °C G. calmariensis adults fed more than G. pusilla. G. pusilla larvae consumed an average of 25% less foliage than G. calmariensis. The lower larval consumption of G. pusilla suggests that when food is limited, G. pusilla larvae may have a higher survival rate because of its ability to complete larval development with less food and produce more progeny due to its greater fecundity. When food is not limited neither species would have a competitive advantage and both species could coexist temporally and spatially. However, since G. calmariensis larvae consumed more leaf material, the larval stage of this species would have a greater impact on purple loosestrife than G. pusilla.  相似文献

The objectives of this study were to clarify the influences of nitrogen (N) load on the growth and photosynthetic responses of Quercus serrata seedlings to O3 and to obtain basic data for evaluating the critical levels of O3 for protecting Q. serrata forests in Japan. The effects of O3 and/or N load on growth and photosynthetic activity of Q. serrata seedlings were investigated during the two growing seasons. Two-year-old seedlings were assigned to 12 experimental treatments, which were comprised of the combination of four gas treatments (charcoal-filtered air and three levels of O3 at 1.0, 1.5 and 2.0 times ambient concentration) and three N treatments (0, 20 and 50 kg ha−1 year−1). During the second growing season, no significant interactive effects of O3 and N load on the growth and net photosynthetic rate of the seedlings were detected. Threrfore, we concluded that N supply to the soil at ≤50 kg ha−1 year−1 does not significantly influence the growth and photosynthetic responses of Q. serrata seedlings to O3. Based on the O3 exposure-response relationships for the whole-plant growth of the seedlings, the critical level of O3 for Q. serrata was estimated to be approximately 36 nmol mol−1 as the average 15-h O3 concentration during the one growing season.  相似文献

Flower-visiting beetles belonging to three species of Cetoniidae were collected on three mountains near Beijing, China, and yeasts were isolated from the gut of the insects collected. Based on the 26S rDNA D1/D2 domain and internal transcribed spacer (ITS) region sequence analysis and phenotypic characterization, four novel anamorphic yeast species located in the Candida albicans/Lodderomyces elongisporus clade were identified from 18 of the strains isolated. The new species and type strains are designated as Candida blackwellae AS 2.3639T (=CBS 10843T), Candida jiufengensis AS 2.3688T (=CBS 10846T), Candida oxycetoniae AS 2.3656T (=CBS 10844T), and Candida pseudojiufengensis AS 2.3693T (=CBS 10847T). C. blackwellae sp. nov. was basal to the branch formed by C. albicans and C. dubliniensis with moderately strong bootstrap support. The closest relative of C. oxycetoniae was L. elongisporus. C. jiufengensis sp. nov. and C. pseudojiufengensis sp. nov. were closely related with each other and formed a branch in a subclade represented by C. parapsilosis and L. elongisporus.  相似文献
