Many bacteria control gene expression in response to cell population density, and this phenomenon is called quorum sensing. In Gram-negative bacteria, quorum sensing typically involves the production, release and detection of acylated homoserine lactone signalling molecules called autoinducers. Vibrio harveyi, a Gram-negative bioluminescent marine bacterium, regulates light production in response to two distinct autoinducers (AI-1 and AI-2). AI-1 is a homoserine lactone. The structure of AI-2 is not known. We have suggested previously that V. harveyi uses AI-1 for intraspecies communication and AI-2 for interspecies communication. Consistent with this idea, we have shown that many species of Gram-negative and Gram-positive bacteria produce AI-2 and, in every case, production of AI-2 is dependent on the function encoded by the luxS gene. We show here that LuxS is the AI-2 synthase and that AI-2 is produced from S-adenosylmethionine in three enzymatic steps. The substrate for LuxS is S-ribosylhomocysteine, which is cleaved to form two products, one of which is homocysteine, and the other is AI-2. In this report, we also provide evidence that the biosynthetic pathway and biochemical intermediates in AI-2 biosynthesis are identical in Escherichia coli, Salmonella typhimurium, V. harveyi, Vibrio cholerae and Enterococcus faecalis. This result suggests that, unlike quorum sensing via the family of related homoserine lactone autoinducers, AI-2 is a unique, 'universal' signal that could be used by a variety of bacteria for communication among and between species.  相似文献

Quorum sensing is a type of bacterial communication mediated by chemical signaling molecules called autoinducers (AIs). The production of AI-2 and AI-3 is dependent on the luxS gene in Escherichia coli O157:H7. A luxS mutation caused a minimal decrease (about 2-fold) in Shiga toxin (Stx) production in in vitro cultures using Luria-Bertani broth. The effect of a luxS mutation on the virulence of E. coli O157:H7 was examined by using germfree mice. There were no differences between the luxS mutant and the wild-type in the bacterial counts in feces shedding, Stx production, or the survival of the mice. The treatment of ciprofloxacin decreased the bacteria in feces but increased the Stx production. However, even treatment with ciprofloxacin did not make any difference between the luxS mutant and the wild-type in animal experiments.  相似文献

Quorum sensing is a process by which bacteria communicate using secreted chemical signaling molecules called autoinducers. In this study, the opportunistic plant pathogen Erwinia carotovora ssp. carotovora was observed to secrete type II signaling molecules. A homolog of luxS, the gene required for AI-2 synthesis in Vibrio harveyi, was isolated from the genome of the pathogen. To determine the potential role of AI-2 in virulence, an isogenic luxS- (ECC) mutant was constructed and tested for its ability to cause tissue maceration. The findings reported here demonstrate that the LuxS-dependent signaling affects the progression of disease symptoms during the early stages of infection by modulating the expression of pectinolytic enzymes.  相似文献

Cell-to-cell communication in bacteria is a process known as quorum sensing that relies on the production, detection, and response to the extracellular accumulation of signaling molecules called autoinducers. Often, bacteria use multiple autoinducers to obtain information about the vicinal cell density. However, how cells integrate and interpret the information contained within multiple autoinducers remains a mystery. Using single-cell fluorescence microscopy, we quantified the signaling responses to and analyzed the integration of multiple autoinducers by the model quorum-sensing bacterium Vibrio harveyi. Our results revealed that signals from two distinct autoinducers, AI-1 and AI-2, are combined strictly additively in a shared phosphorelay pathway, with each autoinducer contributing nearly equally to the total response. We found a coherent response across the population with little cell-to-cell variation, indicating that the entire population of cells can reliably distinguish several distinct conditions of external autoinducer concentration. We speculate that the use of multiple autoinducers allows a growing population of cells to synchronize gene expression during a series of distinct developmental stages.  相似文献

Bacterial populations use cell-cell communication to coordinate community-wide regulation of processes such as biofilm formation, virulence, and bioluminescence. This phenomenon, termed quorum sensing, is mediated by small molecule signals known as autoinducers. While most autoinducers are species specific, autoinducer-2 (AI-2), first identified in the marine bacterium Vibrio harveyi, is produced and detected by many Gram-negative and Gram-positive bacteria. The crystal structure of the V. harveyi AI-2 signaling molecule bound to its receptor protein revealed an unusual furanosyl borate diester. Here, we present the crystal structure of a second AI-2 signal binding protein, LsrB from Salmonella typhimurium. We find that LsrB binds a chemically distinct form of the AI-2 signal, (2R,4S)-2-methyl-2,3,3,4-tetrahydroxytetrahydrofuran (R-THMF), that lacks boron. Our results demonstrate that two different species of bacteria recognize two different forms of the autoinducer signal, both derived from 4,5-dihydroxy-2,3-pentanedione (DPD), and reveal new sophistication in the chemical lexicon used by bacteria in interspecies signaling.  相似文献

The last decades, the research on bacterial cell-cell communication or quorum sensing has been quite intense. Quorum sensing allows bacteria to coordinate their behavior and to act as one entity. Quorum sensing controls microbiological functions of medical, agricultural and industrial importance and a better understanding of the underlying mechanisms and the conditions under which the signaling occurs, offers possibilities for new applications. In this article a dynamic model for diauxic growth, overflow metabolism and AI-2-mediated cell-cell communication of Salmonella Typhimurium is presented. The growth, and the production and uptake of the AI-2 signaling molecule of S. Typhimurium are investigated in a controlled environment (bioreactor). In a first stage a model is developed to describe diauxic growth and overflow metabolism. This model is extended in a second stage to describe AI-2 dynamics of S. Typhimurium in relation to the growth kinetics and biomass concentration. It is illustrated how this model can be employed to test hypotheses concerning AI-2 dynamics on the basis of macroscopic data.  相似文献

Quorum sensing is a process of bacterial cell-cell communication that uses small diffusible molecules to coordinate diverse behaviors in response to population density. The only quorum-sensing system shared by Gram-positive and Gram-negative bacteria involves the production of autoinducer-2 (AI-2). The AI-2 synthase LuxS is widely distributed among the Bacteria, which suggests that AI-2 is a language for interspecies communication. However, LuxS is also an integral component of the activated methyl cycle in bacteria. LuxS-based quorum sensing has been intensively studied in the past decade, mostly in relation to the AI-2 molecule and the downstream effects of luxS knockouts; few studies have focused on the gene and protein activity itself. Ongoing attempts to dissect the metabolic and signaling roles of LuxS leave little doubt that unraveling the regulation of luxS expression and cellular LuxS activity is the key to understanding LuxS-based quorum sensing.  相似文献

Quorum sensing is a process that enables bacteria to communicate using secreted signaling molecules called autoinducers. This process enables a population of bacteria to regulate gene expression collectively and, therefore, control behavior on a community-wide scale. Quorum sensing is widespread in the bacterial world and, generally, processes controlled by quorum sensing are unproductive when undertaken by an individual bacterium but become effective when undertaken by the group. Cell-cell communication can occur within and between bacterial species, and between bacteria and their eukaryotic hosts, which suggests that the chemical lexicon is complex. Prokaryotic and eukaryotic mechanisms for enhancing and inhibiting quorum sensing have been identified, which suggests that manipulation of quorum-sensing-controlled processes could be common in bacterial-bacterial and bacterial-eukaryotic associations.  相似文献

In processes regulated by quorum sensing (QS) bacteria respond to the concentration of autoinducers in the environment to engage in group behaviours. Autoinducer-2 (AI-2) is unique as it can foster interspecies communication. Currently, two AI-2 receptors are known, LuxP and LsrB, but bacteria lacking these receptors can also respond to AI-2. In this work, we present an efficient and reproducible synthesis of a novel chemical probe, d-desthiobiotin-AI-2. This probe binds both LuxP and LsrB receptors from different species of bacteria. Thus, this probe is able to bind receptors that recognise the two known biologically active forms of AI-2, presenting the plasticity essential for the identification of novel unknown AI-2 receptors. Moreover, a protocol to pull down receptors bound to d-desthiobiotin-AI-2 with anti-biotin antibodies has also been established. Altogether, this work highlights the potential of conjugating chemical signals to biotinylated derivatives to identify and tag signal receptors involved in quorum sensing or other chemical signalling processes.  相似文献

In a process called quorum sensing, bacteria communicate with one another by exchanging chemical signals called autoinducers. In the bioluminescent marine bacterium Vibrio harveyi, two different auto inducers (AI-1 and AI-2) regulate light emission. Detection of and response to the V.harveyi autoinducers are accomplished through two two-component sensory relay systems: AI-1 is detected by the sensor LuxN and AI-2 by LuxPQ. Here we further define the V.harveyi quorum-sensing regulon by identifying 10 new quorum-sensing-controlled target genes. Our examination of signal processing and integration in the V.harveyi quorum-sensing circuit suggests that AI-1 and AI-2 act synergistically, and that the V.harveyi quorum-sensing circuit may function exclusively as a 'coincidence detector' that discriminates between conditions in which both autoinducers are present and all other conditions.  相似文献

Quorum sensing regulates cell density-dependent phenotypes and involves the synthesis, excretion and detection of so-called autoinducers. Vibrio harveyi strain ATCC BAA-1116 (recently reclassified as Vibrio campbellii), one of the best-characterized model organisms for the study of quorum sensing, produces and responds to three autoinducers. HAI-1, AI-2 and CAI-1 are recognized by different receptors, but all information is channeled into the same signaling cascade, which controls a specific set of genes. Here we examine temporal variations of availability and concentration of the three autoinducers in V. harveyi, and monitor the phenotypes they regulate, from the early exponential to the stationary growth phase in liquid culture. Specifically, the exponential growth phase is characterized by an increase in AI-2 and the induction of bioluminescence, while HAI-1 and CAI-1 are undetectable prior to the late exponential growth phase. CAI-1 activity reaches its maximum upon entry into stationary phase, while molar concentrations of AI-2 and HAI-1 become approximately equal. Similarly, autoinducer-dependent exoproteolytic activity increases at the transition into stationary phase. These findings are reflected in temporal alterations in expression of the luxR gene that encodes the master regulator LuxR, and of four autoinducer-regulated genes during growth. Moreover, in vitro phosphorylation assays reveal a tight correlation between the HAI-1/AI-2 ratio as input and levels of receptor-mediated phosphorylation of LuxU as output. Our study supports a model in which the combinations of autoinducers available, rather than cell density per se, determine the timing of various processes in V. harveyi populations.  相似文献

The LuxS enzyme, catalyzing the synthesis of AI-2, is conserved in a wide diversity of bacteria. AI-2 has therefore been suggested as an interspecies quorum sensing signal. To investigate the role of endogenous AI-2 in protein expression of the Gram-negative pathogen Salmonella enterica serovar Typhimurium (S. Quorum sensing is used by a large variety of bacteria to regulate gene expression in a cell-density-dependent manner. Bacteria can synchronize population behavior using small molecules called autoinducers that are produced by cognate synthases and recognized by specific receptors. Quorum sensing plays critical roles in regulating diverse cellular functions in bacteria, including bioluminescence, virulence gene expression, biofilm formation, and antibiotic resistance. The best-studied autoinducers are acyl homoserine lactone (AHL) molecules, which are the primary quorum sensing signals used by Gram-negative bacteria. In this review we focus on the AHL-dependent quorum sensing system and highlight recent progress on structural and mechanistic studies of AHL synthases and the corresponding receptors. Crystal structures of LuxI-type AHL synthases provide insights into acyl-substrate specificity, but the current knowledge is still greatly limited. Structural studies of AHL receptors have facilitated a more thorough understanding of signal perception and established the molecular framework for the development of quorum sensing inhibitors.  相似文献

Quorum sensing is an intricate example of "social" behavior in microbial communities mediated by small secreted molecules (autoinducers). The mechanisms of membrane permeation remain elusive for many of them. Here we present the assessment of membrane permeability for three natural autoinducers and four synthetic analogues based on their polarity, surface activity, affinity for lipid monolayers, and ability to induce lateral pressure changes in the inner E. coli membrane sensed by the bacterial tension-activated channel MscS. AI-1 (N-(3-oxodecanoyl)-l-homoserine lactone) is surface-active, and it robustly inserts into lipid monolayers, indicating strong propensity toward membranes. When presented to membrane patches from the cytoplasmic side, AI-1 transiently shifts MscS's activation curve toward higher tensions due to intercalation into the cytoplasmic leaflet followed by redistribution to the opposite side. Indole showed no detectable surface activity at the air-water interface but produced a moderate increase of lateral pressure in monolayers and was potent at shifting activation curves of MscS, demonstrating transients on sequential additions. AI-2 (4,5-dihydroxy-2,3-pentanedione, DPD) showed little activity at the interfaces, correspondingly with no effect on MscS activation. After chemical modification with isobutyl, hexyl, or heptyl chains, AI-2 displayed strong surface activity. Hexyl and especially heptyl AI-2 induced robust transient shifts of MscS activation curves. The data strongly suggest that both AI-1 and indole are directly permeable through the membrane. AI-2, more hydrophilic, shows low affinity toward lipids and thus requires a transport system, whereas alkyl analogues of AI-2 should permeate the membrane directly.  相似文献
