Sequence analysis of five of the six endopolygalacturonase-encoding genes ( Bcpg1 , Bcpg2 , Bcpg3 , Bcpg4 , Bcpg5 ) from 32 strains of Botrytis cinerea showed marked gene to gene differences in the amount of among-strains diversity. Bcpg4 was almost invariable in all strains; Bcpg3 and Bcpg5 showed a moderate variability, similar to that of non-pathogenicity-associated genes examined in other studies. Conversely, Bcpg1 and Bcpg2 were highly variable and were shown to be under positive selection based on the McDonald–Kreitman test and likelihood ratio test. The evolution of the five endopolygalacturonase genes is explained by their different ecophysiological role. Diversification and balancing selection, as detected in Bcpg1 and Bcpg2 , can be used by the pathogen to escape recognition by the host and delay plant reaction in the early phases of infection. The analysis of the polymorphisms and the location of the sites with high probability of being positively selected highlighted the relevance of variability of the BcPG1 and BcPG2 proteins at their C-terminal end. By contrast, the absence of variability in Bcpg4 suggests that the efficiency of the product of this gene is critical for B. cinerea growth in late phases of infection or during intraspecific competition, thus markedly affecting strain fitness.  相似文献

Zebrafish were intraperitoneally injected with 10(6)CFU (LD50) Listonella anguillarum. Three inoculated and control fish were collected at 1, 2, 4, 6 and 22h post infection (hpi) and the expression of genes related to the immune response (il1b, cebpb, tfa, mpx, tnfa, nitr9, tlr22, hsc70, cp, mrlp1, c3b and lyz) in each fish was monitored by means of real-time RT-PCR. A similar experiment was performed considering an intermediate time point at 15 hpi. Different relative levels of expression were found among genes. Also, wide interindividual variation in gene expression for most genes was detected among fish, inoculated or not. A steady increase of expression starting from the initial stages of the interaction was found for interleukin-1beta. An initial increase in levels of gene expression was found for the genes coding for the CCAAT/Enhancer Binding Protein subunit beta and the Novel Immune-Type Receptor 9, although their levels decreased later on and were indistinguishable from the controls at 22 hpi. Finally, some genes (Transferrin, Myeloid-specific Peroxidase and Tumour Necrosis Factor alpha) were upregulated at 22 hpi. Taken together, our results show an induction in gene expression of genes involved in the inflammatory and immune response upon L. anguillarum infection but also reveal the existence of a wide variation in the levels of expression of the studied genes in the zebrafish population.  相似文献

Fast recognition of host signals and early activation of infection mechanisms in Plasmopara viticola are decisive for successful infestation of Vitis vinifera. To better understand interactive processes at the first front line of combat between the pathogen and its host, a specific pre-infective stage was generated in a host-free system. Zoospore encystment was triggered within minutes after treatment with CaCl2. Subsequently, high rates of germ tube formation occurred in a synchronized manner. This method was employed to compare development-related gene expression in strains of different virulence. Soon after germination, spores showed strong up-regulation of two effector genes, PvRxLR18 and PvRxLR28, particularly in the high virulence strain. On infected grapevine leaf-discs of cultivars with different susceptibility, a similar up-regulation was found at 6 hours post inoculation (hpi). This effect was much more evident in the high virulence than in the low virulence strain and was significantly higher on leaves of the tolerant cultivar Regent than on Müller-Thurgau. In addition, PvRxLR67 was up-regulated 24 hpi in the high virulence strain indicating that different effectors are active in later infection stages. Differences in the expression pattern of RxLR effector genes between the two strains corroborated with infection symptoms visible by sporulation.  相似文献
