水稻T-DNA插入突变体库的构建及突变类型的分析   总被引:7,自引:2,他引:7  
利用农杆菌介导的转化系统转化中花11成熟胚愈伤组织,获得1489个独立转化的T-DNA插入再生株系。PCR和Southern杂交的结果表明,69.8%转化株系被整合了T-DNA,通过Tail-PCR也从转化植株中扩增出T-DNA侧翼序列。同时对1066个T1转化株系的抽穗期、株高、单株穗数的调查结果表明,不同株系中分离出了突变植株。  相似文献

采用ATMT技术建立大丽轮枝菌落叶型菌株XJ2008菌株的T-DNA插入突变体文库,共获得6 043个突变体。从中随机挑选104个突变体,以野生型XJ2008菌株为参照,评价其致病性、菌落生长速率、分生孢子及微菌核的产生能力等。结果表明,有12.5%的突变体丧失产孢能力,4.8%的突变体的生长速率显著减慢,8.7%的突变体的生长速率显著加快,12.5%的突变体丧失产生微菌核的能力,47.1%的突变体的致病性显著低于野生型菌株XJ2008,且突变体2-736、2-740、2-745的病情指数分别约为野生型菌株XJ2008的0.184、0.168和0.197倍。该突变体库突变体遗传稳定性好,性状多样性丰富。  相似文献

利用功能互补,对拟南芥AtMGT9基因的Mg2+转运功能进行了研究.同时对AtMGT9的两个T-DNA插入突变体株系(mgt9-1,mgt9-2)进行表型分析,mgt9-1只能得到杂合体植株,有花粉败育现象,mgt9-2有纯合体,但没有可见表型.基因序列分析表明,mgt9-1中T-DNA插入位点在起始密码子后,mgt9-2中T-DNA插入位点在靠近C端第2个跨膜区中间位置.功能互补和63Ni2+示踪实验证明截短的mgt9-2蛋白能够互补细菌镁离子转运突变体MM281, 具有全序列基因一样的转运特性,而mgt9-1不具有转运功能,是一个功能缺失突变体.这为进一步研究AtMGT9蛋白的结构与功能的关系奠定了基础.  相似文献

Two alleles at a new locus, central pair–associated complex 1 (CPC1), were selected in a screen for Chlamydomonas flagellar motility mutations. These mutations disrupt structures associated with central pair microtubules and reduce flagellar beat frequency, but do not prevent changes in flagellar activity associated with either photophobic responses or phototactic accumulation of live cells. Comparison of cpc1 and pf6 axonemes shows that cpc1 affects a row of projections along C1 microtubules distinct from those missing in pf6, and a row of thin fibers that form an arc between the two central pair microtubules. Electron microscopic images of the central pair in axonemes from radial spoke–defective strains reveal previously undescribed central pair structures, including projections extending laterally toward radial spoke heads, and a diagonal link between the C2 microtubule and the cpc1 projection. By SDS-PAGE, cpc1 axonemes show reductions of 350-, 265-, and 79-kD proteins. When extracted from wild-type axonemes, these three proteins cosediment on sucrose gradients with three other central pair proteins (135, 125, and 56 kD) in a 16S complex. Characterization of cpc1 provides new insights into the structure and biochemistry of the central pair apparatus, and into its function as a regulator of dynein-based motility.  相似文献

稻瘟病菌T-DNA插入方法优化及其突变体分析   总被引:10,自引:0,他引:10  
优化了农杆菌介导转化稻瘟病菌获得T-DNA插入突变的条件,包括选择转化子的潮霉素B用量,抑制农杆菌的抗生素头孢噻肟钠和羧苄青霉素的配比,不同转化阶段培养基的选择等。转化1×106个孢子平均可获得约500个左右的转化子,PCR和TAILPCR检测表明约85%转化子中含T-DNA插入。对1520个突变体进行形态变异观察,发现菌落颜色突变的有15个;随机取58个突变体进行比较,发现产孢量减少的4个,孢子萌发率降低的8个,附着胞形成率降低的9个;还获得对水稻品种C101LAC(Pi-1)和751127(Pi-9)致病的突变体,为进一步克隆相应的无毒基因奠定了基础。  相似文献

采用羧甲基纤维素钠筛选培养基,对黑曲霉(Aspergillus niger)T-DNA突变子文库进行筛选,分离到一株纤维素酶分泌水平较低的菌株AN-108,为野生型菌株的83.3%。进一步测定该突变子固体发酵的纤维素酶活力,与野生型菌株相比没有明显差别,推测与固体发酵培养基中含有的天然糖类有关。在添加不同糖类的CMC-Na平板上培养该突变子,菌落周围均出现较明显的水解圈,结果显示糖类可能作为诱导物克服突变带来的影响。为了确定突变子AN-108中何种基因被阻断,采用反向PCR方法分析了T-DNA插入位点的序列,获得序列经过比对分析发现,该序列与黑曲霉An14g03730同源程度达90%,编码富含脯氨酸蛋白(proline-rich protein,PRP)。  相似文献

Brachypodium distachyon is a promising model system for the structural and functional genomics of temperate grasses because of its physical, genetic and genome attributes. The sequencing of the inbred line Bd21 ( http://www.brachypodium.org ) started in 2007. However, a transformation method remains to be developed for the community standard line Bd21. In this article, a facile, efficient and rapid transformation system for Bd21 is described using Agrobacterium -mediated transformation of compact embryogenic calli (CEC) derived from immature embryos. Key features of this system include: (i) the use of the green fluorescent protein (GFP) associated with hygromycin selection for rapid identification of transgenic calli and plants; (ii) the desiccation of CEC after inoculation with Agrobacterium ; (iii) the utilization of Bd21 plants regenerated from tissue culture as a source of immature embryos; (iv) the control of the duration of the selection process; and (v) the supplementation of culture media with CuSO4 prior to and during the regeneration of transgenic plants. Approximately 17% of CEC produced transgenic plants, enabling the generation of hundreds of T-DNA insertion lines per experiment. GFP expression was observed in primary transformed Bd21 plants (T0) and their progeny (T1). The Mendelian inheritance of the transgenes was confirmed. An adaptor-anchor strategy was developed for efficient retrieval of flanking sequence tags (FSTs) of T-DNA inserts, and the resulting sequences are available in public databases. The production of T-DNA insertion lines and the retrieval of associated FSTs reported here for the reference inbred line Bd21 will facilitate large-scale functional genomics research in this model system.  相似文献

With the completion of the rice (Oryza sativa L.) genome‐sequencing project, the rice research community proposed to characterize the function of every predicted gene in rice by 2020. One of the most effective and high‐throughput strategies for studying gene function is to employ genetic mutations induced by insertion elements such as T‐DNA or transposons. Since 1999, with support from the Ministry of Science and Technology of China for Rice Functional Genomics Programs, large‐scale T‐DNA insertion mutant populations have been generated in Huazhong Agricultural University, the Chinese Academy of Sciences and the Chinese Academy of Agricultural Sciences. Currently, a total of 372,346 mutant lines have been generated, and 58,226 T‐DNA or Tos17 flanking sequence tags have been isolated. Using these mutant resources, more than 40 genes with potential applications in rice breeding have already been identified. These include genes involved in biotic or abiotic stress responses, nutrient metabolism, pollen development, and plant architecture. The functional analysis of these genes will not only deepen our understanding of the fundamental biological questions in rice, but will also offer valuable gene resources for developing Green Super Rice that is high‐yielding with few inputs even under the poor growth conditions of many regions of Africa and Asia.   相似文献

Generation and flanking sequence analysis of a rice T-DNA tagged population   总被引:26,自引:0,他引:26  
Insertional mutagenesis provides a rapid way to clone a mutated gene. Transfer DNA (T-DNA) of Agrobacterium tumefaciens has been proven to be a successful tool for gene discovery in Arabidopsis and rice (Oryza sativa L. ssp. japonica). Here, we report the generation of 5,200 independent T-DNA tagged rice lines. The T-DNA insertion pattern in the rice genome was investigated, and an initial database was constructed based on T-DNA flanking sequences amplified from randomly selected T-DNA tagged rice lines using Thermal Asymmetric Interlaced PCR (TAIL-PCR). Of 361 T-DNA flanking sequences, 92 showed long T-DNA integration (T-DNA together with non-T-DNA). Another 55 sequences showed complex integration of T-DNA into the rice genome. Besides direct integration, filler sequences and microhomology (one to several nucleotides of homology) were observed between the T-DNA right border and other portions of the vector pCAMBIA1301 in transgenic rice. Preferential insertion of T-DNA into protein-coding regions of the rice genome was detected. Insertion sites mapped onto rice chromosomes were scattered in the genome. Some phenotypic mutants were observed in the T1 generation of the T-DNA tagged plants. Our mutant population will be useful for studying T-DNA integration patterns and for analyzing gene function in rice.Electronic Supplementary Material Supplementary material is available in the online version of this article at .Communicated by D. 植物中不同转基因方法转化外源基因的T-DNA整合特征既具有共性,又具有特性,使得转基因的遗传在各独立转化体间呈现多样性,另外多种遗传因子和限制因素使受体植物中外源基因的表达存在下降,甚至出现基因沉默等复杂现象。本文主要对农杆菌介导及裸露DNA直接转化转基因植物中T-DNA的分子特征和转基因表达的影响因子进行了介绍和概述。转化体中转基因的遗传稳定性和表达主要取决于转基因在植物基因组中的整合位置、拷贝数及组成结构。因而,通过对具有表达水平各异的转化体进行深入的遗传分析和分子生物学研究以及转化体之间进行的比较研究,将对转基因技术自身的完善、定点整合以及更有效的利用转基因技术都具有十分重要的意义。  相似文献

We have previously generated a large pool of T-DNA insertional lines in rice. In this study, we screened those T-DNA pools for rice mutants that had defective chlorophylls. Among the 1,995 lines examined in the T2 generation, 189 showed a chlorophyll-deficient phenotype that segregated as a single recessive locus. Among the mutants, 10 lines were beta-glucuronidase (GUS)-positive in the leaves. Line 9-07117 has a T-DNA insertion into the gene that is highly homologous to XANTHA-F in barley and CHLH in ARABIDOPSIS: This OsCHLH gene encodes the largest subunit of the rice Mg-chelatase, a key enzyme in the chlorophyll branch of the tetrapyrrole biosynthetic pathway. In the T2 and T3 generations, the chlorina mutant phenotypes are co-segregated with the T-DNA. We have identified two additional chlorina mutants that have a Tos17 insertion in the OsCHLH gene. Those phenotypes were co-segregated with Tos17 in the progeny. GUS assays and RNA blot analysis showed that expression of the OsCHLH gene is light inducible, while TEM analysis revealed that the thylakoid membrane of the mutant chloroplasts is underdeveloped. The chlorophyll content was very low in the OschlH mutants. This is the first report that T-DNA insertional mutagenesis can be used for functional analysis of rice genes.  相似文献

在构建由农杆菌介导的玉米Ds转座因子插入的水稻转化群体中,得到了一个茎秆等组织发生脆性突变的株系。理化指标定量测定表明,脆性株系的载荷强度和纤维素含量都比正常植株低很多,可溶性糖含量略有减少。对这个突变株的分子检测结果表明Ds因子在脆性株系中为单位点插入。检测了自前3代(T1,T2,T3)植株中T-DNA(Ds)插入与脆性表型的共分离关系。初步结果表明这个突变是T-DNA(Ds)的插入造成的,这个突变基因可能与水稻纤维素合成有关。  相似文献
