Previous percentage labelled mitoses studies in acutely uraemic mice have demonstrated a lengthening of the cell cycle and the DNA synthetic phase of ileal epithelium. The mitotic index was unaltered. Further studies have been performed to obtain an estimate of mitotic duration. Acute renal failure was produced by urinary outflow obstruction in male mice. Controls were subjected to sham operation. The mean number of cells per crypt cell column, number of mitoses present per crypt section and differential mitotic stage count were assessed 18 hr after operation for uraemic and control mice. The mean number of metaphases accumulated per crypt section over a 2 hr interval following colchicine injection was obtained in other groups of mice and the mitotic duration calculated. The mean number of mitoses per crypt section was 1.30 ± 0.46 for the controls and 1.48 ± 0.66 for the uraemic group. No evidence for a block in mitosis was indicated by the differential mitotic stage count. After applying Tannock's correction factor the mitotic duration was estimated to be 0.91 ± 0.18 hr for the control group and 2.81 ± 0.89 hr for the uraemic group. The difference in duration between the groups, 1.90 ± 0.91 hr, was significant (P≤0.05). Reduction in cell proliferation may explain the development of uraemic lesions in the gastrointestinal tract.  相似文献

Rats were treated with guanethidine-sulphate every 48 hr from birth until 15 days and then maintained until young adulthood. Sympathectomy was verified by dissection and light microscopic preparation of the superior cervical and coeliac ganglia which showed at least a 78% reduction in the number of perikarya. The effect of the chemical sympathectomy was a decrease in the amplitude of the circadian mitotic rhythm from 44·7 to 27·1%, 67·0 to 25·3% and 54·9 to 24·7%, in the duodenum, jejunum, and ileum, respectively. The shape of the mitotic index curve was altered and the mean mitotic index was significantly decreased (P < 0·01) in all three segments of the small intestine. The mean mitotic index of control intestinal epithelium was 3·2 ± 0·1%, 3·6 ± 0·1% and 4·0 ± 0·1% in the duodenum, jejunum, and ileum, respectively, and 2·3 ± 0·1%, 2·4 ± 0·1%, and 2·5 ± 0·1% in guanethidine-treated rats. Stathmokinetic estimates of cycle time were obtained by use of the metaphase arrest agent, colchicine. The longest cell generation cycle time (Tc) and lowest mitotic index occurred between 12.00 and 16.00 hours and the shortest Tc and highest mitotic index occurred between 00.00 and 04.00 hours, in all three segments of the small intestine. Guanethidine-treatment lengthens Tc throughout the small intestinal epithelium and reduces the range of variation in Tc over a 24-hr period. It is suggested that norepinephrine depletion induced by guanethidine may be the cause of the inhibition in circadian periodicity and that norepinephrine and the sympathetic nervous system may be essential for the maintenance of circadian rhythms in mitotic activity.  相似文献

The ileum of suckling rats contains a high level of copper, most of which is concentrated within cytoplasmic vesicles of the enterocytes. Intestinal closure, 20-21 days after birth, results in the replacement of enterocytes by cells devoid of these vesicles and there is a concomitant fall in the level of copper in the ileum. Administration of cortisone acetate (0.5 mg/g body wt) to 5-day-old rats results in premature loss of copper-laden ileal enterocytes and an 80-90% decrease in the ileal copper concentration. The loss of copper is mainly from the soluble fraction of the tissue and is proportionally greater from the high molecular weight-protein fraction rather than from the copper complex.  相似文献

Both the latent period and the volume doubling time of 20-methylcholanthrene-induced tumours in the skin of SAS/TO mice have been found to be longer for plucked skin than for unplucked skin, even though the cell cycle time of the basal layer cells of the epithelium is much shorter after plucking (50 ± 4 hr) than in unplucked skin (110 ± 10 hr).  相似文献

Newborn rats were injected with guanethidine-sulfate (20 μg/g body weight) every 48 hr from 12 hr after birth until day 14 (eight injections per animal). The guanethidine treatment resulted in an 86% absolute reduction in cell number in the superior cervical ganglia of 15 day old rats. The cells which remained after guanethidine treatment showed destruction of mitochondria and an extensive decrease in endoplasmic reticulum. Chemical sympathectomy with guanethidine induced a 3.1 hr lengthening of the acinar cell generation cycle time (17.4 hr to 20.5 hr), resulting from a longer G1 period (6.9 hr in the control group as compared to 10.5 hr in the guanethidine-treated group), as well as a decrease in the mean percentage of [3H]thymidine-labeled acinar cells (22.3 ± 0.5% to 19.3 ± 0.5%) and mean acinar cell mitotic index (2.6 ± 0.2% to 2.1 ± 0.1%). A circadian rhythm was found to exist in parotid gland acinar cell mitotic activity of 15 day old rats and the amplitude of the rhythm was reduced from 26.5% to 14.9% in guanethidine-treated rats. This study indicates that the diminution of sympathetic influence on the developing parotid gland results in a slight, but significant alteration in acinar cell proliferation.  相似文献

1. Using standard autoradiographic techniques, the percentage of labelled mitoses and the numbers of labelled cells in the various layers of the epithelium of the hamster cheek pouch have been studied at various times after a single injection of [3H]thymidine. 2. A new method for determining the cell cycle time of the basal layer cells is described and compared with the techniques previously used. This method is based on the rate at which labelled cells leave the basal layer and enter the spinous layer above it. 3. Using this technique, the mean cell cycle time of the basal cells has been estimated as 130 ± 7 hr; a value which compares favourably with the estimate of 135 ± 17 hr determined with similar assumptions from the length of the S period and the labelling index. 4. The technique enables an estimate to be made of the fraction of cells in the basal layer that are proliferating (the growth fraction). This is found to be indistinguishable from 1. 5. The mean time taken by cells in travelling from the basal layer to the keratin, i.e. the transit time through the spinous layer, has been found to be approximately 186 hr.  相似文献

The duration of cell cycle parameters in control mouse jejunum has been compared with that found following induction of a graft-versus-host reaction (GvHR) during the first 3 weeks of postnatal life. Values for tc and tG1 were found to decrease progressively during normal development: estimates for the whole crypt column in 21-day-old mice were approximately half to one quarter those found 6 days after birth 12.1 +/- 0.5 hr and 24.2 +/- 0.3 hr for tc; 2.8 +/- 0.3 hr and 12.1 +/- 0.3 hr for tG1 respectively; (means +/- SE). tS and tG2 were found to remain approximately constant during this period of neonatal development. Injecting foreign spleen cells into 3-day-old mice produced no effect on crypt cell proliferation or cell cycle parameters measured 3 days later. GvHR mice studied 8 days after spleen cell injection, however, showed both an increase in crypt cell proliferation and decreases in the values for tc and tG1 to levels similar to those normally found in 21-day-old control animals (tc 12.4 +/- 0.4 hr and tG1 5.4 +/- 0.4 hr for 11-day-old GvHR mice). The possible mechanism leading to these changes is discussed. The ability of GvHR to stimulate cell proliferation is used in the present work to test the hypothesis that the total number of cell divisions taking place after birth determines the temporal sequence of changes in disaccharidase content produced during neonatal development.  相似文献

It was previously reported that in rats hypophysectomized at 6 days of age, the apicotubular system that permits the entrance of macromolecules closes spontaneously in the duodenal epithelium. Extending these observations to the ileum, where pinocytosis is nutritionally important in the suckling rat, now reveals that in the absence of the hypophysis, closure of the surface does not occur. Hence, the failure of growth that sets in during the fourth postnatal week appears not to be due to malnutrition consequent on the cessation of intracellular digestion. Treatment of the hypophyseoprivic sucklings with either thyroxine or cortisone from 15 to 23 days causes the disappearance of the apical infoldings, vesicles, and giant vacuole characteristic of the suckling stage. Instead, the cells exhibit all of the ultrastructural features typical of mature ileal epithelium. The effects of the two hormones differed in that cortisone caused a slightly greater lengthening of the microvilli. These results further extend the evidence indicating that either thyroid hormone or glucocorticoid can activate the complete regulatory program that brings about the redifferentiation of the intestinal epithelium from the suckling to the mature type.  相似文献

The effects of acute and subacute thiamine deficiency on jejunal and ileal epithelial cells were studied in rats, using crypt and villus cell population, crypt cell production per crypt (CCPC), crypt growth fraction (Ip) and crypt cell cycle time (Tc) as parameters. In acute thiamine deficiency there was marked jejunal hypoplasia of the crypt and villus, but in the ileum there was hypoplasia only of the crypt. The jejunal epithelium of the subacute thiamine deficiency (STD) group showed no morphometric changes. In contrast, in the ileal epithelium of STD rats there was decreased crypt depth and villus cell population. Thiamine deficiency had no significant effect on CCPC, Ip and Tc.  相似文献

1. Proteolytic (substrate nitrocasein), tripeptidase (substrate glycylglycylgly-cine) and aminopeptidase (substrate leucyl-beta-naphthylamide) activities were studied in homogenates of jejunal and ileal mucosa of 7-, 10-, 14-, 21-, 35- and 60-day-old rats. 2. Proteolytic activity was practically the same in jejunum of 7-, 10-, 14- and 21-day-old rats, but after day 21 a significant increase was observed. The activity of the ileum changed very little during postnatal development and was always higher than that of the jejunum. 3. Tripeptidase activity was low in the jejunum of 7- and 14-day-old rats, an increase was observed between day 14 and 21, but later no substantial changes were found. There were no changes in the ileum. The activity in the jejunum of 7- and 14-day-old rats was lower than in the ileum, but later the jejunum was more active than the ileum. 4. Aminopeptidase activity had a similar developmental pattern to tripeptidase activity. A low activity was found in the jejunum of 7- and 14-day-old rats, the maximum was in 21-day-old rats and then a decrease was observed, though values for 60-day-old rats were still higher than for 7- and 10-day-old rats. The activity in the ileum was practically the same in all age groups studied except in 14- and 21-day-old rats, where a transient peak was observed.  相似文献

1. The developmental pattern and effect of cortisone on acid beta-galactosidase and neutral beta-galactosidase were studied in postnatal rats by a recently proposed method for their independent determination. 2. After birth the acid beta-galactosidase activity increases in the ileum, whereas it decreases slightly in the jejunum. On day 16 after birth the activity in the ileum decreases and in 20-day-old rats activity in both parts of the intestine decreases to adult values. In suckling animals the activity in the ileum exceeds the jejunal activity severalfold and in adult animals the activity in the jejunum is slightly higher than that in the ileum. 3. Neutral beta-galactosidase activity is high after birth and decreases in both jejunum and ileum after day 20 after birth. In 12-20-day-old rats activity in both parts is essentially the same, but in adult animals jejunal activity exceeds ileal activity four-to five-fold. 4. Cortisone (0.5, 2.0 or 5.0mg/100g body wt. daily for 4 days) does not influence the activity of either enzyme in 60-day-old rats. Acid beta-galactosidase activity is decreased after cortisone treatment in 8-, 12-, 16-and 18-day-old rats, with sensitivity to cortisone increasing with the approach of weaning. No effect of cortisone on acid beta-galactosidase is seen in 8-day-old rats. Neutral beta-galactosidase activity is increased in the ileum of 8-, 12-, 16- and 18-day old rats, but only in the jejunum of 8-and 12-day-old rats.  相似文献

The proliferation parameters of the Walker carcinoma were estimated from both in vivo and in vitro measurements. The transplantable Walker carcinoma 256 was grown in male inbred BD1 rats. During exponential growth, 5-6 days after transplantation, a PLM curve was performed, yielding estimates of Tc ? 18.0 hr, Ts ? 6.4 hr, TG2+M? 4.1 hr. With the double labelling technique in vitro under 2.2 atm oxygen we obtained: Tc ? 18.2hr, Ts ? 8.2 hr, TG2+M? 2.0hr. From pulse cyto-photometry DNA content histograms the fractions of cells in the cell cycle phases were calculated using a computer program: fG1? (47.6 ± 1.1)%, fs? (34.1 ± 1.0)%, fG2+M? (18.3 ± 1.5)%. These fractions remained constant between the fifth and the twelfth day after transplantation. At that time the tumour growth had already slowed down appreciably. The growth fraction determined by repetitive labelling was 0.96 on the fifth and 0.93 on the seventh and eleventh day. The cell loss factor was φ? 17% during exponential tumor growth and increased to about 100% between the tenth and twelfth day. The agreement of the cell kinetic data determined by autoradiography from solid tumours in vivo (PLM, continuous labelling) and autoradiography as well as pulse cytophotometry from in vitro experiments (excised material) was satisfactory.  相似文献

The effect of estradiol-17β on the length of the various phases of the cell cycle was studied in the neonatal mouse uterine, and cervical epithelium. A double labelling method was used, and in addition labelled mitoses were counted. In the uterus proper, estradiol shortens the length of the total cell cycle, Tc, from 17-9 hr to 15-7 hr, and the duration of S phase, Ts, from 6–7 to 5-1 hr 6 hr after estradiol treatment. 12 hr after estradiol treatment, Tc is shortened to 7-4 hr and Ts to 4–5 hr. The shortening of Tc at 12 hr is mainly due to an effect on TG1, which is shortened from 8–55 hr in untreated animals to 1–8 hr in estradiol treated animals. The Tc of cervix epithelium cells in untreated animals was found to be 21-8 hr. After treating the mice for 6 hr with estradiol the Tc was now increased to 47 hr and further to 61 -2 hr following 12 hr treatment with the hormone. Ts increases from 8-3 hr to 15-2 hr following 6 hr estradiol treatment, and to 15-4 hr after 12 hr treatment. The effect is most pronounced in TG1, which is lengthened from 10–95 hr in untreated animals to 28-1 hr and 43 hr, respectively, in animals treated for either 6 or 12 hr with estradiol.  相似文献

Neonatal hypoxia increases aldosterone production and plasma lipids. Because fatty acids can inhibit aldosterone synthesis, we hypothesized that increases in plasma lipids restrain aldosteronogenesis in the hypoxic neonate. We exposed rats to 7 days of hypoxia from birth to 7 days of age (suckling) or from 28 to 35 days of age (weaned at day 21). Plasma was analyzed for lipid content, and steroidogenesis was studied in dispersed whole adrenal glands untreated and treated to wash away lipids. Hypoxia increased plasma cholesterol, triglycerides, and nonesterified fatty acids in the suckling neonatal rat only. Washing away lipids increased aldosterone production in cells from 7-day-old rats exposed to hypoxia, but not in cells from normoxic 7-day-old rats or from normoxic or hypoxic 35-day-old rats. Addition of oleic or linolenic acid to washed cells inhibited both aldosterone and corticosterone production, although cells from hypoxic 7-day-old rats were less sensitive. We conclude that hypoxia induces hyperlipidemia in the suckling neonate and that elevated nonesterified fatty acids inhibit aldosteronogenesis.  相似文献

Using everted sac technique we demonstrated the transfer of 125I-mEGF across the jejunal and ileal walls of suckling, weanling and adult rats. The transfer by the suckling rat jejunum and ileum was significantly inhibited by the presence of dinitrophenol and sodium azide or by the replacement of sodium with potassium or choline. RP-HPLC analysis detected carboxy-terminal processing of 125I-mEGF in suckling and adult rat jejunum and ileum. Suckling rat jejunum produced 125I-des(53)mEGF and 125I-des(49–53)mEGF, whereas 125I-des(48–53)mEGF was detected in suckling rat ileum or adult rat jejunum and ileum. All three forms of 125I-mEGF bound to anti-EGF antibody and EGF receptors. The receptor binding of 125I-des(53)mEGF was higher than that of 125I-mEGF, but those of 125-des(49–53)mEGF and 125I-des(48–53)mEGF were greatly diminished. Results indicate a carboxy-terminal processing of mouse EGF during uptake and transfer in the small intestine of developing and adult rats, and the resulting products showed altered receptor binding. An identical amino acid sequence of the C-terminal pentapeptide of EGF from mouse, human and possibly rat may suggest a biological significance of C-terminal processing of EGF in the small intestine.  相似文献

The cycle of the seminiferous epithelium was studied in Nelore zebu bulls 4–6 years old. The stages of the cycle were determined according to the shape and position of spermatid nuclei and the presence of meiotic divisions in cross-sections of seminiferous tubules. The relative frequencies of stages 1 to 8 were, respectively: 31.3 ± 0.5, 12.2 ± 0.7, 21.2 ± 0.5, 8.8 ± 0.6, 4.2 ± 0.4, 5.7 ± 0.6, 6.3 ± 0.5 and 10.3 ± 0.5. The duration of the cycle was estimated by autoradiography using tritiated thymidine injected directly into the testes. The mean duration of one cycle was estimated to be 14.0 ± 0.4 days.  相似文献

Epithelial cell kinetics were studied in an ileal segment after transposition to proximal jejunum. The number of cells per villus column in the transposed ileum increased after 4-7 days to reach values normal for jejunum after 14-30 days. This increase was accompanied by a simultaneous increase in the number of cells per crypt column up to 130% of values in jejunum and ileum in situ. The percentage of labelled crypt cells, after labelling with 3H-thymidine, and the relative size of the proliferative cell compartment in the crypt in the transposed ileum did not differ from values in the ileum in situ at any time interval after surgery. The total proliferative activity per crypt, which was determined by scintillation counting of isolated crypts after 3H-thymidine labelling, increased two-fold from 7 days after surgery. Cell migration studies showed that the increase in the number of villus cells was probably not caused by a change in the life span of the epithelial cells. It seems that the increase in the number of villus cells in ileal epithelium after transposition to proximal jejunum is brought about by an enlargement of the crypt, while the relative size of the proliferative cell compartment in the crypt remains unchanged.  相似文献

The increase of sucrase activity in homogenates of jejunum and ileum of suckling rats after cortisone administration has been investigated. Serial tissue sections of villi and crypts were also assayed for sucrase activity and these results were compared with the migration of cells labelled with [(3)H]thymidine along the villus. By using a low dose of cortisone (0.5mg/day per 100g body wt.) it was found that the sensitivity of the small intestine producing system to cortisone stimulation increased during the suckling period. On the other hand, 5mg of cortisone/day per 100g body wt. produced practically the same increase of sucrase during the entire suckling period. Sucrase activity in homogenates of the entire small-intestinal wall was first detected 24h after the first injection of cortisone (5mg/day per 100g body weight) to 9-day-old animals and maximum activity both in the jejunum and ileum was reached by 120h. Jejunal activity was greater than ileal activity, but the rate of the increase was similar. The half-time of the increase was 23-27h, whereas enterocytes migrate from the base to the tip of the villi in approximately 72h. Comparison of sucrase activity in serial tissue sections of villi and crypts at various times after cortisone treatment showed that the leading edge of sucrase activity proceeds toward the tip of the villi at the same rate as the advancing edge of newly formed cells. Sucrase activity increased in the newly induced cells as they migrated to the tip of the villi. It was concluded that the increase of sucrase activity in suckling rats after cortisone stimulation is due to at least three factors: (1) increase of activity in newly differentiating cells, (2) increased percentage of villus cells with sucrase activity and (3) continued production or activation of sucrase activity as the cells migrate along the villi.  相似文献
