以甘蔗(Saccharum officinarum)优良品种桂糖42号(GT42)为研究材料, 分别于未伸长期(9-10叶龄以前) (Ls1)、伸长初期(12-13叶龄) (Ls2)和伸长盛期(15-16叶龄) (Ls3)取甘蔗第2片真叶(自顶部起)对应的节间组织, 测定其赤霉素(GA)、生长素(IAA)、油菜素甾醇(BR)、细胞分裂素(CTK)、乙烯(ETH)和脱落酸(ABA)的含量, 并通过实时荧光定量PCR (qRT-PCR)分析赤霉素合成途径关键基因GA20氧化酶基因(GA20-Oxidase1)、赤霉素受体基因(GID1)和DELLA蛋白编码基因(GAI)的差异表达。结果表明, 在甘蔗伸长期间, GA和IAA含量呈现上升趋势, CTK和ABA含量呈下降趋势, ETH含量先上升后下降, BR含量则变化不明显; GA20-Oxidase1GID1的表达呈上升趋势, 而GAI的表达则呈下降趋势, 这与相关植物激素的变化基本一致。综上, 甘蔗节间伸长过程主要与GA和IAA相关, 其次为CTK和ABA, 而ETH受到IAA的调控影响节间伸长; 植物激素间通过相互作用调控GA20-Oxidase1GID1GAI的表达, 影响GA含量和GA的信号转导过程, 进而影响甘蔗节间的伸长。该研究揭示了甘蔗节间伸长过程中赤霉素生物合成途径和信号转导关键基因的差异表达及植物激素含量的动态变化规律。  相似文献

Ion compositions of the cytoplasm and the vacuole of Chara australiswere analyzed according to Kishimoto and Tazawa (1964) and Kiyosawa(1979a). The ions in the cytoplasm and the vacuole analyzedwere K+, Na+, Ca2+, Mg2+, Cl, NO3 and H2PO4.Assuming that the volume of the cytoplasm Vp is 10% of thatof the whole cell V, the concentrations of K+, Na+, Ca2+, Mg2+,Cl, NO3 and H2PO4 in the cytoplasm averaged70, 15, 13, 4.6, 31, 2.2 and 16 mM, respectively. If the volumeof the cytoplasm was assumed to be 5% of that of the whole cell,their averaged concentrations were 139, 31, 25, 9.2, 62, 4.4and 33 mM, respectively. The averaged ion compositions of thecell sap were K+, 111; Na+, 47; Ca2+, 4.4; Mg2+, 8.9; Cl,91; NO3, 3.3 and H2PO4, 6.0 mM. These values,taking the concentrations and the charges of the protein (Kiyosawa1979b) and amino acids (Sakano and Tazawa 1984) into accountand assuming the presence of some uni- or oligovalent anionsand/or small nonelectrolyte molecules, could explain fairlywell both the electroneutrality and the osmotic pressure ofthe cell, except when Vp/V = 5%. 以甘蔗品种'新台糖22号'为试验材料,在伸长初期以200 mg/L GA3进行叶面喷施处理,对照喷清水,研究GA3处理后甘蔗节间糖苷酶、过氧化物酶、过氧化氢酶的变化,以揭示赤霉素诱导甘蔗节间伸长与相关酶活性的关系.结果表明:(1)GA3处理的株高在各个时期显著高于对照,而且在处理后7、14和28 d分别比对照提高了17.32%、14.50%和8.35%,GA3处理引起甘蔗植株表现的高度优势一直保持到后期,节间伸长效果主要是在茎的中部(5~10节).(2)GA3处理后α-葡萄糖苷酶和α-甘露糖苷酶的活性较对照显著下降;POD和β-半乳糖苷酶的活性也略有下降;α-半乳糖苷酶、β-N-乙酰氨基已糖苷酶、过氧化氢酶的活性显著提高;β-葡萄糖苷酶的活性也有一定程度提高.由此推测,外源GA3主要通过调节α-葡萄糖苷酶活性、α-甘露糖苷酶、α-半乳糖苷酶、β-N-乙酰氨基已糖苷酶活性和过氧化氢酶,其次是POD、β-半乳糖苷酶和β-葡萄糖苷酶活性,最终达到节间伸长效果.  相似文献

Abstract: The fatty acid composition of phosphatidylethanolamine (PE), ethanolamine plasmalogens (EPs), phosphatidylserine (PS), phosphatidylcholine (PC), and sphingomyelin was studied in 22 human forebrains, ranging in age from 26 prenatal weeks to 8 postnatal years. Phospholipids were separated by two-dimensional TLC, and the fatty acid methyl esters studied by capillary column GLC. Docosahexaenoic acid (22:6n-3) increased with age in PE and PC, whereas arachidonic acid (20:4n-6) remained quite constant. In EP, 22:6n-3 increased less markedly than 20:4n-6, adrenic (22:4n-6) and oleic (18:1n-9) acids being the predominant fatty acids during postnatal age. In PS, 18:1n-9 increased dramatically throughout development, and 20:4n-6 and 22:4n-6 increased only until ∼6 months of age. Although 22:6n-3 kept quite constant during development in PS, its percentage decreased due to the accretion of other polyunsaturated fatty acids (PUFAs). As a characteristic myelin lipid, sphingomyelin was mainly constituted by very long chain saturated and monounsaturated fatty acids. Among them, nervonic acid (24:1n-9) was the major very long chain fatty acid in Sp, followed by 24:0, 26:1n-9, and 26:0, and its accretion after birth was dramatic. As myelination advanced, 18:1n-9 increased markedly in all four glycerophospholipids, predominating in EP, PS, and PC. In contrast, 22:6n-3 was the most important PUFA in PE in the mature forebrain.  相似文献

The chemical composition of phloem sap from the uppermost internodeof rice plants (Oryza sativa L., var. Kantou), one week afteranthesis, was compared with that of phloem sap from the leafsheath of a young seedling. The pure phloem sap from rice plantswas collected by an insect laser technique. The phloem sap from the uppermost internode contained a highlevel of sucrose (573.8 mM) which was the only sugar detected.The concentrations of total amino acids, potassium and ATP were124.8 mM, 40.4 mM and 1.76 mM, respectively. The concentrationof sucrose was three times higher and the potassium level wasone third as high in the internode sap as in the phloem sapfrom the leaf sheath. The total concentration of amino acidswas almost the same, but the relative amount of each amino acidwas quite different. The ratios of levels of Glu to Gln andof levels of Asp to Asn in the phloem sap from the uppermostinternode were smaller than those in the phloem sap from theleaf sheath. The adenylate energy charge was 0.92–0.93in both types of phloem sap. The amino acid composition of the phloem sap from the uppermostinternode was compared with that of the phloem sap of the flagleaf and the endosperm sap of the same plant, one week afteranthesis. Sugarcane borer (Diatraea saccharalis) is a major pest of sugarcane (Saccharum spp. hybrids), across the Americas. The insect is partially controlled by biological and chemical means, but still causes significant economic losses to sugarcane growers and processors. Proteins derived from Bacillus thuringiensis, Bt proteins, have been used to control sugarcane borer in maize (Zea mays) for the past decade. In sugarcane, the expression of individual Bt proteins has been reported several times. However practical use of Bt proteins requires their use as part of an Integrated Pest Management (IPM) system that includes the delivery of high doses of protein and the use of a refuge to slow the evolution of insect resistance to the protein. Here we demonstrate the feasibility of using Bt proteins to protect sugarcane from sugarcane borer in a commercial setting. We have expressed two Bt proteins with differing modes of action (Cry1Ab and Cry2Ab) in commercial sugarcane varieties, demonstrated efficacy against sugarcane borer in the field and describe a strategy for trait deployment in this tropical crop with complex genetics that limits trait introgression by backcrossing.  相似文献

Changes in both cell wall and water-soluble polysaccharide compositionduring the growth of kiwifruits [Actinidia deliciosa (A. chev.) C. F. Liang and A. R. Ferguson var. deliciosa ‘Hayward’]were investigated. Cellulose was the major wall polysaccharide,with galactose and uronics the main non-cellulosic sugars. Muchsolubilization of cell wall pectic polysaccharides was detected.While wall-galactose solubilization started 3 months after anthesis,polyuronide degradation did not start until the fifth month,1 month prior to the harvest date. Parallel to these processes,a linear increase in water-soluble polysaccharides was detected.These mainly comprised galactose-rich polymers in the first3 months and little-branched polyuronides after the fifth month.Two different mechanisms for galactose and uronic acid solubilizationfrom kiwifruit cell walls during fruit development are proposed. Abstract: Ganglioside composition of rat trigeminal nerve was studied during development in order to understand the changes that occur as a result of cellular differentiation in the nerve. The ganglioside composition of the trigeminal nerve was entirely different from that of brain. The major gangliosides in adult trigeminal nerve were GM3, GD3, and LM1 (sialosyl-lactoneotetraosylceramide or sialosylparagloboside). The structure of LM1 and other gangliosides was established by enzymatic degradation and by analysis of the products of acid hydrolysis. At 2 days after birth, when the Schwann cells were immature, GM3 and GD3 were the major gangliosides in the nerve, 50 and 18 mol %, respectively. As the nerve developed and Schwann cells proliferated and myelinated the axons, the mol % of GM3 and GD3 reduced and that of LM1 steadily increased. Polysialogangliosides did not change drastically with nerve development. The rate of deposition of LM1 in the nerve with age was very similar to that of myelin marker lipids, cerebrosides, and sulfatides; thus, deposition appears to be localized mainly in the rat nerve myelin. LM1 also had long-chain fatty acids 22:0 and 24:0, which are not usually found in CNS gangliosides. The ganglioside pattern of the rat trigeminal nerve was very similar to that of rat sciatic nerve, but was different from that of rabbit and chicken sciatic nerve. The activity of the two key enzymes involved in the metabolism of GM3, viz., CMP-N-acetylneuraminic acid:lactosylceramide sialyltransferase and UDP-N-acetylgalactosamine:GM3-N-acetylgalactosaminyltransferase, was also studied during development of the nerve and brain. The developmental profiles of both enzymes were consistent with the amounts of GM3 present in the nerve.  相似文献

The present study investigates the effect of soil amended with sugarcane straw leachate and its constituents on root elongation of weed and crop plants. The influence of soil biotic and abiotic factors on plant growth were also evaluated through assays in both autoclaved soil and sand. In unsterile soil, straw leachates stimulated root growth of some test plants at 6 g dry straw ls−1 and inhibited root elongation at higher concentrations. A bioassay guided process of the bioactive leachate constituents led to the isolation of vanillic, syringic and ferulic acids. These compounds were also assayed on the test plants in the three mentioned substrates. In unsterile soil, phenolics stimulated the growth of some species at 19 mg l−1. Higher phenolics concentrations were inhibitory. The concentration needed to inhibit 25% root elongation (EC25) was calculated from the dose–response curves. The concentration of phenolics in the leachate (64 g dry straw l−1) was estimated to be 187 mg l−1 (ferulic acid), 131 mg l−1 (vanillic acid) and 20 mg l−1 (Syringic acid). In general, these concentrations were below the EC25 values determined in unsterile soil indicating that these compounds cannot completely explain the strong inhibitory activity of sugarcane straw leachates. The role of soil factors on phytotoxicity of sugarcane straw leachate and its identified growth regulators is also discussed.  相似文献

Change in the planting arrangement can provide roots a different environment that alters access to soil resources, which can subsequently impact the productivity of above ground crop compartments. Therefore, the aim of this study was to assess the sugarcane yield as well as the distribution and biomass accumulation of roots across different spacing configuration treatments, in the absence of the confounding effect of machine traffic between the crop rows, in two areas with contrasting soil conditions. Two experiments were conducted under field conditions, in clayey and sandy soil. The experiments were conducted across two crop seasons and incorporated a randomized block design that tested six planting configurations: CS, conventional spacing (1.50 m); AS, alternated spacing (0.90?×?1.50 m); TS, triple spacing (0.75?×?0.75?×?1.50 m); PP 1.0 m, precision planting (1.0?×?1.0 m); PP 0.75 m, precision planting (0.75?×?0.75 m); and PP 0.50 m, precision planting (0.5?×?0.5 m). Results showed, in clayey soil, higher sugarcane yield for the PP 0.75 m spacing in the two crops evaluated; while, the PP 1.0 m spacing had the higher production in sandy soil, but only during the first ratoon. In relation to the total sugarcane production per unit area, the yield gains from reduced spacing were most evident for the first two crop cycles. The distribution and root biomass accumulation of sugarcane did not explain the above ground productivity in planting configurations.  相似文献

Lignocellulosic biomass from sugarcane (Saccharum spp. hybrids) could potentially be a major feedstock for second-generation biofuel production. Consequently, selecting sugarcane varieties with favorable biomass characteristics, typically less enzymatic recalcitrance and better saccharification yield without sugar-yield penalty, will be important in sugarcane breeding. Economical and high-throughput techniques for profiling the major biomass components of this complex system will facilitate selection of clones with ideal lignocellulosic composition from large numbers of genotypes in breeding programs. We used a combined high-throughput profiling approach to evaluate the biomass composition of samples from a sugarcane germplasm collection. This employed near-infrared (NIR) spectroscopy for fiber characterization and high-performance liquid chromatography (HPLC) for determining the sugar content in juice. The results for 331 samples, from a diverse sugarcane population of 186 genotypes, derived from 143 parents of different genetic backgrounds, showed that high-quality NIR spectroscopic predictions were feasible for cellulose, hemicellulose, lignin, and extractives values in fiber, and sugars in juice were suitably analyzed by HPLC. The analysis of total biomass indicated that this NIR- and HPLC-based high-throughput method allowed a robust phenotypic assessment of a large number of samples for the key biomass traits in the sugarcane system, including total dry biomass, fiber, sugar content, and theoretical ethanol yields, and could potentially become the method of choice for sugarcane germplasm screening in breeding programs targeting the support of biofuel production.  相似文献

Myelin proteolipid protein (PLP) is modified after translation by the attachment of long-chain fatty acids to several cysteine residues. In this study, the amount and pattern of fatty acids covalently bound to rat PLP were determined during brain development and in myelin subfractions. For this purpose, PLP was isolated by gel-filtration chromatography in organic solvents, subjected to alkaline methanolysis, and the released fatty acid methyl esters were analyzed by gas-liquid chromatography. At all ages examined, PLP had the same amount of covalently-bound fatty acids (3–4% w/w) and palmitate, oleate and stearate were always the major acyl chains. In contrast to myelin lipids, the fatty acid composition of PLP showed only minor changes between 15-days and 90-days of age. The amount and pattern of fatty acids bound to PLP prepared from three myelin subfractions were also indistinguishable. The conservation of a characteristic PLP-fatty acid make-up during brain development and in various myelin compartments suggests that this post-translational modification is essential for the normal functioning of the protein.  相似文献

Abstract: The phospholipid profile of different chick embryo brain regions was studied from 11 to 21 days of development, revealing interesting changes in content and distribution. Total phospholipid phosphorus (P), in micrograms of P per microgram of DNA, increases significantly during development of cerebral hemispheres (CHs), optic lobes (OLs), and brainstem (BS). Compared with CH and OL, the BS shows at all stages a significantly higher concentration of phospholipid P, which in contrast decreases in the cerebellum (CB) during development. Moreover, the data show interesting differences between the right and the left portion of the brain. The distribution of phospholipid P and the fatty acid composition of phospholipids were asymmetric between left and right OL and CH, as were the concentrations of DNA and cholesterol, demonstrating lateralized neurochemical development in these structures, i.e., left OL, right OL, left CH, and right CH. The data are discussed also in relation to the potential importance of neurochemical lateralization for determining lateralized embryonic and postnatal behavior of this species.  相似文献

The influence of the Na and Le genes in peas on gibberellin (GA) levels and metabolism were examined by gas chromatographic-mass spectrometric analysis of extracts from a range of stem-length genotypes fed with [13C, 3H]GA20. The substrate was metabolised to [13C, 3H]GA1, [13C, 3H]GA8 and [13C, 3H]GA29 in the immature, expanding apical tissue of all genotypes carrying Le. In contrast, [13C, 3H]GA29 and, in one line, [13C, 3H]GA29-catabolite, were the only products detected in plants homozygous for the le gene. These results confirm that the Le gene in peas controls the 3-hydroxylation of GA20 to GA1. Qualitatively the same results were obtained irrespective of the genotype at the Na locus. In all Na lines the [13C, 3H]GA20 metabolites were considerably diluted by endogenous [12C]GAs, implying that the metabolism of [13C, 3H]GA20 mirrored that of endogenous [12C]GA20. In contrast, the [13C, 3H]GA20 metabolites in na lines showed no dilution with [12C]GAs, confirming that the na mutation prevents the production of C19-GAs. Estimates of the levels of endogenous GAs in the apical tissues of Na lines, made from the 12C:13C isotope ratios and the radioactivity recovered in respective metabolites, varied between 7 and 40 ng of each GA per plant in the tissue expanded during the 5 d between treatment with [13C, 3H]GA20 and extraction. No [12C]GA1 and only traces of [12C]GA8 (in one line) were detected in the two Na le lines examined. 利用改进的冰冻切片法结合间接免疫荧光标记技术对甘蔗茎尖细胞有丝分裂过程中微管骨架的变化进行了研究。结果表明, 在甘蔗茎尖细胞有丝分裂过程中存在4种循序变化的典型微管列阵,即周质微管、早前期微管带、纺锤体微管及成膜体微管。同时, 还观察到在各种典型微管列阵相互转变过程中存在各种微管列阵的过渡状态。甘蔗茎尖正在伸长的幼叶部位细胞的周质微管主要为与细胞伸长轴相垂直的横向周质微管; 茎尖幼叶部位伸长缓慢细胞的微管主要为纵向及斜向排列的周质微管,在甘蔗茎尖幼叶基部初生增粗分生组织处, 横向、斜向、纵向及随机排列的周质微管列阵均有分布。在少数分裂前期的细胞中, 发现细胞具有2条早前期微管带, 其具体功能还不清楚。表明甘蔗茎尖细胞微管列阵的变化与许多双子叶植物及部分单子叶植物具有共同的变化规律, 进一步证明微管骨架的周期性变化在植物中具有普遍性。  相似文献
