The effect of a series of amino acids on the eutectic behavior of NaCl solutions at isotonic concentration has been studied by differential scanning calorimetry. The inclusion of different amino acids had different effects on eutectic formation. The amino acids were grouped into four categories based on their effect on eutectic formation: category C were amino acids that had no effect on eutectic formation; category D amino acids inhibited eutectic formation; category T amino acids shifted the melting of the eutectic to a lower temperature; category E amino acids caused the formation of a new eutectic with a melting temperature approximately -5 degrees C. The mechanism of these different effects on eutectic behavior is discussed, based on the chemical structure of the amino acids.  相似文献

Stress state in plants caused by salinization conditions is characterized by the disturbance of ionic and osmotic homeostasis. The maintenance of the latter is reached by accumulation of osmolytes including free amino acids and soluble sugars in cells. The free amino acid level in the 8-day-old control seedling leaves was higher, than in the roots, whereas the contrary picture was observed in 17-day-old plant tissues. At the same time 8-day-old seedling roots contained more total sugars, than leaves, although the reduced sugar content was nearly a half of the total sugar content. A decrease of both total and reduced sugar levels was observed in 17-day-old seedling tissues. One-day exposure of 7-day-old seedlings to 0.1 M NaCl increased the free amino acid content especially in roots, than in leaves, and the total sugar content in maize leaves, whereas in roots this level remained without changes. The prolongation of salt exposure to 10 days leads to osmolyte content decrease. The seed treatment with Methyure and Ivine intensified accumulation of free amino acids and soluble sugars in the root and leaf tissues under salinization conditions.  相似文献

NaCl胁迫7d明显抑制了两种南瓜——黑籽南瓜(Cucurbita ficifolia Bouche.)和白籽南瓜(Cucurbita moschata Duch.)幼苗地上部和根系的生长,使地上部相对含水量明显下降。随NaCl胁迫浓度(150、300和500mmol/L)的提高,两种南瓜幼苗根、茎和叶片中Na^+含量明显增加,K^+含量明显下降。Na^+和K^+含量在两个南瓜品种各器官中的分布规律分别是:根〉茎〉叶片和茎〉叶片〉根。NaCl胁迫后黑籽南瓜根、茎和叶片Na^+含量远低于白籽南瓜,而叶片游离脯氨酸含量、可溶性糖含量和地上部相对含水量明显高于白籽南瓜。表明两者在渗透调节方式、离子的吸收和运输能力上有差异,黑籽南瓜耐盐性较强。  相似文献

The antioxidative and prooxidative activities of sugars and sugar analogs were investigated on the autoxidation of methyl linoleate and safflower oil. Autoxidation of methyl linoleate was conducted in either the dry state or aqueous emulsion state. Although all sugars and sugar analogs inhibited the autoxidation of methyl linoleate in the dry system, the reducing sugars accelerated oxidation in the aqueous emulsion system. When glyceraldehyde, dihydroxyacetone and glycerol were added in the oxidation system, glyceraldehyde and dihydroxyacetone with water accelerated the formation and decomposition of methyl linoleate monohydroperoxide. On the other hand, glycerol inhibited the formation and decomposition of hydroperoxide. These results indicate that the carbonyl group of sugars accelerates lipid peroxidation and the hydroxy group of sugars and sugar analogs inhibits the oxidation. The effect of sugars in the safflower oil-sugar-cellulose oxidation system was also examined. Sugars at low humidity inhibited the autoxidation of safflower oil, but reducing sugars at high humidity accelerated the oxidation.  相似文献

NaCl胁迫对超大甜椒种子萌发及幼苗生长的影响   总被引:6,自引:1,他引:5  
以超大甜椒种子和幼苗为材料,采用水培法研究低、中、高浓度(50、150、250 mmol·L-1)NaCl胁迫对其种子萌发和幼苗生长的影响.结果显示:(1)低浓度NaCl处理促进种子萌发,而中、高浓度NaCl处理抑制种子萌发;NaCl处理第18天时,高浓度NaCl处理植株全部死亡,其余各处理植株苗高、叶面积、地上部鲜重和干重均随处理浓度升高而下降,但低浓度NaCl能刺激超大甜椒的根系生长.(2)低、中浓度NaCl处理时,植株叶绿素含量未受到大的影响,类胡萝卜素含量却随胁迫时间延长微量升高;在盐胁迫7 d的周期内,低、中浓度NaCl处理植株MDA含量、SOD活性和可溶性糖含量均随浓度升高及时间延长显著增加,脯氨酸含量在低浓度下变化不明显而中浓度下显著升高;在高浓度NaCl处理3 d中,MDA含量急剧升高,SOD活性先升高后下降,脯氨酸和可溶性糖含量显著增加.研究发现,NaCl胁迫浓度越高对超大甜椒种子萌发和幼苗生长的抑制效应越明显;低中浓度NaCl处理幼苗能通过自身的抗氧化酶清除系统和渗透调节物质来抵抗胁迫引起伤害,类胡萝卜素可能也有一定的抗胁迫作用,而高浓度NaCl处理增加了膜脂过氧化程度,严重影响了活性氧和渗透调节物质的正常代谢.  相似文献

Novel cationic amphiphilic compounds were prepared from vernonia oil, a natural epoxidized triglyceride, and studied with respect to vesicle formation, encapsulation of biomaterials such as DNA, and their physical stability and transport through isolated plant cuticle membranes. The amphiphiles studied were a single-headed compound III (a quaternary ammonium head group with two alkyl chains) and a triple-headed compound IV, which is essentially three molecules of compound III bound together through a glycerol moiety. Vesicles of the two amphiphiles, prepared by sonication in water and solutions of uranyl acetate or the herbicide 2,4-D (2,4-dichloropenoxy acetic acid), were examined by TEM, SEM, AFM, and confocal laser systems and had a spherical shape which encapsulated the solutes with diameters between 40 and 110 nm. Vesicles from amphiphile IV could be made large enough to encapsulate a condensed 5.2kb DNA plasmid (pJD328). Vesicles of amphiphile IV were also shown to pass intact across isolated plant cuticle membranes and the rate of delivery of encapsulated radio-labeled 2,4-D through isolated plant cuticle membranes obtained with these vesicles was clearly greater in comparison to liposomes prepared from dipalmitopyl phosphatidylcholine (DPPC) and the control, nonencapsulated 2,4-D. Vesicles from amphiphiles III and IV were found to be more stable than those of liposomes from DPPC. The data indicate the potential of vesicles prepared from the novel amphiphile IV to be a relatively efficient nano-scale delivery system to transport DNA and other bioactive agents through plant biological barriers. This scientific approach may open the way for further development of efficient in vivo plant transformation systems.  相似文献

Effect of grain soaking presowing in 1 mM salicylic acid (SA) and NaCl (0, 50, 100, 150 and 200 mM) on barley (Hordeum vulgare cv Gerbel) was studied. Increasing of NaCl level reduced the germination percentage, the growth parameters (fresh and dry weight), potassium, calcium, phosphorus and insoluble sugars content in both shoots and roots of 15-day old seedlings. Leaf relative water content (RWC) and the photosynthetic pigments (Chl a, b and carotenoids) contents also decreased with increasing NaCl concentration. On the other hand, Na, soluble sugars, soluble proteins, free amino acids including proline content and lipid peroxidation level and peroxidase activity were increased in the two plant organs with increasing of NaCl level. Electrolyte leakage from plant leaves was found to increase with salinity level. SA-pretreatment increased the RWC, fresh and dry weights, water, photosynthetic pigments, insolube saccharides, phosphorus content and peroxidase activity in the stressed seedlings. On the contrary, Na+, soluble proteins content, lipid peroxidation level, electrolyte leakage were markedly reduced under salt stress with SA than without. Under stress conditions, SA-pretreated plants exhibited less Ca2+ and more accumulation of K+, and soluble sugars in roots at the expense of these contents in the plant shoots. Exogenous application (Grain soaking presowing) of SA appeared to induce preadaptive response to salt stress leading to promoting protective reactions to the photosynthetic pigments and maintain the membranes integrity in barley plants, which reflected in improving the plant growth.  相似文献

The combined effect of the sugar nucleotides UDP-D-fucose or UDP-D-glucuronic acid together with the free sugars D-fucose or L-arabinose is the inactivation of the Escherichia coli enzyme UDP-galactose 4-epimerase (EC 5.1.3.2). The sugar nucleotide or the free sugar alone or the sugar nucleotide plus 5'-Ump do not inactivate the enzyme. 2. The inactivation of the enzyme by its substrate UDP-D-glucose was not affected by the presence of free sugar. 3. In all cases the inactivation observed follows pseudo-first-order kinetics. 4. A comparison of various sugar nucleotides indicates that the hydroxymethyl group at position 6 of the sugar moiety of the natural substrates is important for substrate binding.  相似文献

The interfacial behavior of the neoglycolipids formed of Guerbet alcohol (G(28)) bound to a triethylene glycol spacer (E(3)) and to a sugar moiety (alpha- and beta-fucose) spread at the air/water interface has been studied under dynamic conditions of compression. Although the alpha (alpha-FucE3G28)- and beta-fucose (beta-FucE3G28) derivatives possessed the same chemical structure, the positioning of the sugar moiety relative to the whole molecule had a significant influence on the organization of neoglycolipid molecules in the spread monolayers. Thus, beta-fucose molecules exhibited higher compressibilities and larger molecular areas than a alpha/beta (84/16%) mixture (alpha(84)-FucE3G28). The comparison of the compressional behavior of the fucose derivatives with that of Guerbet alcohol in the absence and in the presence of the triethylene glycol spacer shows that the presence of the E(3) chain is necessary to stabilize the lipid at the interface and that the incorporation of a sugar moiety into the molecule resulted in an important expansion of a monolayer. Despite their different interfacial behaviors, the two sugar derivatives formed ideal mixtures when cospread at the air/water interface. Conversely, in the presence of a phospholipid, such as DMPC, repulsive interactions were observed and appeared to be stronger for DMPC/alpha(84)-FucE3G28 mixed monolayers. The membrane fluidity of DMPC liposomes bearing the studied amphiphilic molecules was assessed by fluorescence depolarization measurements. The results reveal that whereas G(28) was deeply inserted into the liposome bilayers, the presence of a E(3) chain and of a sugar moiety in these bilayers induced a transfer of the amphiphilic derivatives from the hydrophobic core towards polar headgroups of phospholipid molecules.  相似文献

The physico-chemical properties of beta-lactosyl-closo-orthocarborane in water solution were investigated by multinuclear NMR, (13)C NMR relaxation, and ab-initio calculations. This molecule represents a potentially selective boron carrier in Boron Neutron Cancer Therapy (BNCT) and exhibits amphiphilic characteristics. Its structural and dynamic features were studied comparing NMR data acquired in both aggregating and non-aggregating conditions. Aggregates are characterized by rapid exchange with the bulk and by high sensitivity to temperature conditions. An unusually stable intra-molecular CH...O hydrogen bond was found to persist in water solution both for the free molecules and after aggregate formation. At the same time, inter-molecular specific CH()O interactions do not seem effective in the aggregate formation process, which appears to take place only on non-specific hydrophobic basis.  相似文献

The alpha,beta-unsaturated carbonyl function occurs in a wide variety of bioactive natural products. It is usually associated with the bioactivities of these compounds and acts as Michael acceptors for the addition of protein nucleophilic groups. The design and synthesis of sugars containing this functionality has provided a wide range of compounds, which can serve as building blocks of high synthetic versatility. This review deals with the chemistry of sugar-based molecules bearing singly linked or fused unsaturated lactones and ketones along with that of pyranoid enones and enonolactones. Examples are given of their syntheses and transformations into a variety of complex sugar derivatives such as branched-chain sugars, C-nucleosides, C-glycosyl derivatives, and various natural products, including selected analogues.  相似文献

The effect of three sugars and their amino derivatives on violaxanthin cycle enzymes activity was investigated in duckweed (Lemna trisulca), a model water-plant. No effect of sugars and amino sugars on violaxanthin de-epoxidase was observed independent of incubation time; however, epoxidation of zeaxanthin to violaxanthin was inhibited. The minimum amino sugar concentrations causing maximum inhibition of zeaxanthin epoxidation have been estimated. Amino sugars but not sugars caused more than a 50% inhibition of zeaxanthin epoxidation in duckweed after a 24h incubation when applied at a concentration of 0.5%. Incubation with amino sugars under a 6d photoperiod enhanced the inhibitory effect. Zeaxanthin epoxidation was completely inhibited under such conditions, whereas only a minor inhibitory effect was observed in sugar treated plants. The strong amino sugar inhibition of zeaxanthin epoxidase activity represents additional evidence for the creation of an unstable carotenoid carbocation in the molecular mechanism of epoxidation.  相似文献

To study the effects of the sugar structure on the activity of anthracycline against cancer cells, six daunorubicin analogs containing different uncommon sugars were synthesized. Their cytotoxicities were tested against colon cancer cells by MTS assay. The results showed that the aglycon without sugar moiety has 70-100-fold lower activity against cancer cells than daunorubicin derivatives with various uncommon sugars. It suggests that the sugar structure in daunorubicin plays a critical role in determining its anticancer activity. In the compounds with various sugars, the 4'-OH of the sugar is an important determinant for their activity, while the axial-3'-substituent in the sugar interferes with the binding of daunorubicins to DNA. Therefore, 2,6-dideoxy sugars are a better choice for generating biologically active daunorubicin analogs than 6-deoxysugars, 2,3,6-trideoxysugars, or 2,3,4,6-tetradeoxysugars.  相似文献

以杂交酸模(Rumex K-1)为试材,研究了不同浓度(100~300 mmol·L-1) KCl和NaCl胁迫对杂交酸模幼苗叶片光合活性及渗透调节的影响.结果表明:200 mmol·L-1浓度的NaCl对杂交酸模幼苗叶片光合活性的抑制作用大于KCl;当浓度增大到300 mmol·L-1时,KCl对杂交酸模叶片光合活性的抑制作用显著大于NaCl.300 mmol·L-1 KCl和NaCl处理植株的叶片水势分别为-0.93 MPa和-1.05 MPa,渗透势分别为-1.43 MPa和-1.10 MPa,说明KCl对杂交酸模植株过多的伤害不是渗透胁迫造成的;经过300 mmol·L-1KCl胁迫后,杂交酸模叶片中Na+含量急剧降到对照植株的11.4%,而补充25 mmol·L-1 NaCl可以明显缓解KCl对杂交酸模光合活性的伤害,说明Na+的亏缺和高浓度K+的积聚可能是导致高浓度KCl对杂交酸模光合活性的伤害比NaCl更严重的主要原因.  相似文献

The sugar contents of emergent macrophytes from a freshwater lake, a freshwater swamp, and a salt marsh in the southeastern United States were examined together with the dissolved free sugars produced during macrophyte degradation and in natural water samples collected adjacent to macrophyte stands. Simultaneous separation of up to 13 neutral and 2 amino sugars together with 3 uronic acids and muramic acid was achieved by anion-exchange high-performance ion chromatography. As little as 10 pmol or a concentration of 20 nM sugar can be detected by pulsed amperometry, a greater sensitivity for sugar quantification than that of previously reported detection techniques used in conjunction with either gas or liquid chromatographic systems. Optimum conditions for hydrolysis of plant material by using trifluoroacetic acid were determined, and internal standards were used to quantify losses due to matrix effects and solid-phase extraction of samples. Our data demonstrate that ratios of certain indicator sugars in undegraded macrophytes differ significantly from ratios of dissolved free sugars formed during macrophyte degradation, reflecting the complex processes (biological and physical) involved in vascular plant degradation in aquatic ecosystems. Natural water samples collected adjacent to macrophyte beds contained dissolved free sugars at concentrations of 620 nM (lake), 890 nM (freshwater swamp), and 2,300 nM (salt marsh). Sugar signatures of these natural water samples were similar to those of macrophyte degradation products.  相似文献

In order to determine the relative importance of ionic toxicity versus the osmotic component of salt stress on germination in durum wheat (Triticum durum Desf.), seeds of three cultivars differing in their salt and drought resistance (Omrabi-5, drought-resistant; Belikh, salt-resistant and Cando, salt-sensitive) were incubated in various iso-osmotic solutions of NaCl, mannitol and polyethylene-glycol (PEG) (osmotic potential of –0.15 (control solution) –0.58, –1.05 or –1.57 MPa). Moderate stress intensities only delayed germination, whereas the highest concentration of NaCl and PEG reduced final germination percentages. PEG was the most detrimental solute, while mannitol had no effect on final germination percentages. All osmotica reduced endosperm starch and soluble sugars content as well as -amylase activities recorded after 48 h of treatment while -amylase activities were, in contrast, slightly stimulated in all cultivars. Deleterious effects of NaCl and PEG were higher on isolated embryos germinated onto an in vitro Linsmaier and Skoog (LS) medium comparatively to whole seeds. All PEG-treated embryos, however, recovered after the stress relief while NaCl-treated embryos exhibited a lower rate of recovery and some extent of abnormal germination after rinsing. It was concluded that stress inhibition of germination could not be attributed to an inhibition of mobilisation of reserves and that the main effect of PEG occurred via an inhibition of water uptake while detrimental effects of NaCl may be linked to long-term effects of accumulated toxic ions. The behaviour of the three cultivars during germination did not fully reflect their mean level of putative stress resistance in field conditions and germination is, therefore, not recommended as a reliable selection criterion for breeding purposes.  相似文献

An enzyme system from fruits of the flax plant is described that catalyzes the synthesis of the sugar nucleotide guanosine 5'-diphosphate l-fucose from guanosine 5'-diphosphate d-mannose with the intermediate formation of guanosine 5'-diphosphate 4-keto-6-deoxy-d-mannose. Tritium from-[(3)H]H(2)O was incorporated into the l-fucose portion of the sugar nucleotide in the course of the reaction, and tritium at the 3,5-carbons of the d-mannose moiety of GDP-d-mannose was exchanged with protons in the medium. These results support a mechanism of synthesis analogous to that proposed for the formation of l-rhamnose and other 6-deoxy sugars.  相似文献

The phase behaviour, particularly the fluidity within each phase state and the transitions between them, of lipopolysaccharides and of their lipid moiety, free lipid A, of various species of Gram-negative bacteria, especially of Salmonella minnesota and Escherichia coli, has been investigated by applying mainly Fourier-transform infrared spectroscopy and differential scanning calorimetry. For enterobacterial strains, the transition temperatures of the gel----liquid crystalline (beta----alpha) phase transition of the hydrocarbon chains in dependence on the length of the sugar moiety are highest for free lipids A (around 45 degrees C) and lowest for deep rough mutant lipopolysaccharides (around 30 degrees C). Evaluating certain infrared active vibration bands of the hydrocarbon moiety, mainly the symmetric stretching vibration of the methylene groups around 2850 cm-1, it was found that, in the gel state, the acyl chains of lipopolysaccharides and free lipid A have a higher fluidity as compared with saturated and the same fluidity as compared with unsaturated phospholipids. This 'partial fluidization' of lipopolysaccharide below the transition temperature correlates with its reduced enthalpy change at that temperature compared to phospholipids with the same chain length. The fluidity depends strongly on ambient conditions, i.e. on the Mg2+ and H+ content: higher Mg2+ concentrations and low pH values make the acyl chains of free lipid A and lipopolysaccharide preparations significantly more rigid and also partially increase the transition temperature. The influence of Mg2+ is highest for free lipid A and decreases with increasing length of the sugar side chain within the lipopolysaccharide molecules, whereas the effect of a low pH is similar for all preparations. At basic pH, a fluidization of the lipopolysaccharide and lipid A acyl chains and a decrease in transition temperature take place. Free lipid A and all investigated rough mutant lipopolysaccharides exhibit an extremely strong lyotropic behaviour in the beta----alpha melting enthalpy but not in the value of the transition temperature. The phase transition is distinctly expressed only at water concentrations higher than 50-60%. A further increase of the water content still leads to an increase in the phase-transition enthalpy, particularly for lipopolysaccharides with a more complete sugar moiety. The fluidity of the hydrocarbon chains is shown to be an important parameter with respect to the expression of biological activities.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献
