Effects of cadmium (Cd(2+)) on photosynthetic and antioxidant activities of maize (Zea mays L.) cultivars (3223 and 32D99) were investigated. Fourteen-day-old cultivar seedlings were exposed to different Cd concentrations [0, 0.3, 0.6 and 0.9mM Cd(NO(3))(2).4H(2)O] for 8 days. The results of chlorophyll fluorescence indicated that different levels of Cd affected photochemical efficiency in 3223 much more than that in 32D99. In parallel, the level of Cd at 0.9mM caused oxidative damage but did not indicate cessation of PSII activity of the cultivars; plant death was not observed at highly toxic Cd levels. Additionally, the increase in Cd concentration caused loss of chlorophylls and carotenoid and membrane damage in both cultivars, but greater membrane damage was observed in 3223 than in 32D99. Depending on Cd accumulation, a significant reduction in dry biomass was observed in both cultivars at all Cd concentrations. The accumulation of Cd was higher in roots than in leaves for both cultivars. Nevertheless, cultivar 3223 transferred more Cd from roots to leaves than did 32D99. On the other hand, our results suggest that there were similar responses in SOD, APX and GR activities with increasing Cd concentrations for both cultivars. However, POD activity significantly increased at highly toxic Cd levels in 32D99. This result may be regarded as an indication of better tolerance of the Z. mays L. cultivar 32D99 to Cd contamination.  相似文献

Heavy metal (HM) contamination of the environment is a serious threat to sustainable crop production. Among the HMs, chromium (Cr) is one of the most toxic HMs that is known to negatively affect growth and metabolic activities of diverse crop plants. The present study was designed to investigate the ameliorative role of 5-aminolevulinic acid (ALA) under Cr stress in two maize (Zea mays L.) cultivars showing differential sensitivity to Cr tolerance. ALA is a biosynthesis precursor and it has a dominant regulatory effect related to physiological, respiratory, and photosynthesis processes in various plant species. Three concentrations of Cr (0, 5, and 10 mg kg−1) were tested under the graded levels of ALA application (0, 12.5, and 25 mg L−1). The results indicated that Cr stress differentially reduced plant growth attributes, gas exchange characteristics, photosynthetic pigments, and biomass in both the cultivars. Oxidative stress increased as evidenced in the form of electrolyte leakage, malondialdehyde, and hydrogen peroxide (H2O2) accumulation in plants. The anti-oxidative enzyme activities, that is, catalase (CAT), superoxide dismutase (SOD), and peroxidase (POD) both in the leaves and roots of maize cultivars decreased due to Cr stress. The concentration of Cr increased in roots and shoots of maize under Cr levels without ALA. Under Cr stress, ALA exogenous application markedly enhanced plant growth, photosynthetic pigments, gas exchange capacity, and biomass. Furthermore, ALA application decreased the Cr-induced oxidative stress in maize cultivars by improving the activities of CAT, POD, and SOD in plants. After ALA application, the Cr concentrations and total Cr uptake by plants differently decreased in both cultivars. The 6103 cultivar of maize was found to be a tolerant cultivar against Cr stress due to its strong defensive system with a higher rate of antioxidant enzyme activities. On the other hand, the other maize cultivar (9108) was found to be a sensitive cultivar against Cr stress due to its weak defense system with higher contents of reactive oxygen species. These findings suggest that ALA can play a regulatory role in maintaining optimum plant growth and efficient photosynthetic processes under Cr-challenged habitats in maize. Thus, ALA application may be used as a sustainable remedial strategy to alleviate Cr-induced stress in maize cultivars.

Salt-induced changes in growth, photosynthetic pigments, various gas exchange characteristics, relative membrane permeability (RMP), relative water content (RWC) and ion accumulation were examined in a greenhouse experiment on eight sunflower (Helianthus annuus L.) cultivars. Sunflower cultivars, namely Hysun-33, Hysun-38, M-3260, S-278, Alstar-Rm, Nstt-160, Mehran-II and Brocar were subjected to non-stress (0 mM NaCl) or salt stress (150 mM NaCl) in sand culture. On the basis of percent reduction in shoot biomass, cvs. Hysun-38 and Nstt-160 were found to be salt tolerant, cvs. Hysun-33, M-3260, S-278 and Mehran-II moderately tolerant and Alstar-Rm and Brocar salt sensitive. Salt stress markedly reduced growth, different gas exchange characteristics such as photosynthetic rate (A), water-use efficiency (WUE) calculated as A/E, transpiration rate (E), internal CO2 concentration (C i) and stomatal conductance (g s) in all cultivars. The effect of 150 mM NaCl stress was non-significant on chlorophyll a and b contents, chlorophyll a/b ratio, RWC, RMP and leaf and root Cl, K+ and P contents; however, salt stress markedly enhanced C i /C a ratio, free proline content and leaf and root Na+ concentrations in all sunflower cultivars. Of all cultivars, cv. Hysun-38 was higher in gas exchange characteristics, RWC and proline contents as compared with the other cultivars. Overall, none of the earlier-mentioned physiological attributes except leaf K+/Na+ ratio was found to be effective in discriminating the eight sunflower cultivars as the response of each cultivar to salt stress appraised using various physiological attributes was cultivar-specific.  相似文献

Gibberellic acid (GA3) is one of the plant growth regulators which improve salt tolerance and mitigate the salt stress impact on plants. The extant analysis was carried out to study the effect of GA3 and different salt concentrations on seed germination and physiological parameters of oat cultivars. Oats is substantially less tolerant to salt than wheat and barley. Experimentation was conducted as factorial with Completely Randomized Block Design with three replicates. Different concentration of NaCl salt ((25, 50, 75 and 100 mM) were used in test control group and 100 and 150 ppm of GA3 were used in two group by pre-treated (after 24 h of the seed soaking) and plants were analyzed on 15th day. Results indicate that increasing salinity would decrease the germination percentage and growth parameter in three oat cultivars. Quotes data indicating a 13%, 19.9% and 32.48% in cultivars NDO-2, UPO-212 and UPO-94 germination reduction when soil salinity reaches 50 mM. A 36.02%, 47.33% and 56.365 reduction in germination is likely when soil salinity reaches 100 mM respectively same cultivars. Seeds treated with GA3 significantly promoted the percentage of germination, shoot and root length, total fresh and dry weight of seedling, tissue water content and seedling vigor index by NDO-2 and UPO-212 under different saline concentration. The maximum average of germination and growth parameters were observed from 150 ppm GA3 treated seeds. But this concentration was significantly inhibited root length in sensitive cultivar UPO-94 at 75 and 100 mM salt as compared to 100 ppm. We observed that, the high concentration of GA3 was not suitable for sensitive oat cultivars. Because the plant root are the real workforce behind any plants success. Thus, it may be concluding that, GA3 treatment could curtail the toxic effect of salinity by increasing germination percentage and shoot and root length, total fresh and dry weight, tissue water content and seedling vigor index in tolerant cultivar.  相似文献

The effect of growing five different cultivars of pepper plants (Capsicum annuum L.) with CdCl2 concentrations ranging from 0.125 to 0.5 mM on different physiological parameters, and antioxidative enzyme activities of leaves was studied. On the basis of growth parameters, pepper plants were relatively tolerant to cadmium, although metal concentrations higher than 0.125 mM produced a significant inhibition of growth, net photosynthesis, and water use efficiency. Different sensitivities to Cd++ ions were observed among cultivars, Abdera being the most resistant to cadmium stress, while Mondo and Herminio were the most sensitive cultivars. Cadmium concentrations of 0.5 mM produced an increase in the activity of glutathione reductase, and guaiacol peroxidase in most cultivars, while catalase and superoxide dismutase (SOD) were slightly depressed. The analysis of the SOD activity pattern by native-PAGE showed the presence in most cultivars of four SODs which were identified as Mn–SOD, Fe–SOD, CuZn–SOD I and CuZn–SOD II. However, the two CuZn–SODs were absent in the Cd-sensitive cv. Herminio. The growth of pepper plants with 0.5 mM cadmium inhibited the activity of CuZn–SODs in all cultivars, while the activity of Mn- and Fe–SOD was enhanced. The activity of NADPH-dehydrogenases (glucose-6-P-dehydrogenase, 6-phosphogluconate dehydrogenase, NADP–isocitrate dehydrogenase and malic enzyme) showed a Cd-dependent enhancement in most cultivars, the highest increase being observed in the tolerant cv. Abdera. These results suggest that in pepper plants the tolerance to Cd toxicity is more dependent on the availability of NADPH than on its antioxidant capacity.  相似文献

The effects of thiamin (Thi) applied as seed soaking or foliar spray on some key physiological parameters were investigated in two differentially salt-responsive maize (Zea mays L.) cultivars, DK 5783 and Apex 836 F1, exposed to saline stress in two different experiments. An initial experiment (germination experiment) was designed to identify appropriate doses of Thi which could lessen the deleterious effects of salt on plants and screen all available maize cultivars for their differential tolerance to salt stress (100 mM NaCl). The seeds of nine maize cultivars were soaked for 24 h in solutions containing six levels of Thi (25, 50, 75, 100, 125 and 150 mg l?1). Based on the results obtained from the germination experiment, maize cultivar DK 5783 was found to be the most salt tolerant and Apex 836 as the most sensitive cultivar. Also, of six Thi levels used, two levels (100 and 125 mg l?1) were chosen for subsequent studies. In the second experiment (glasshouse experiment), two maize cultivars, DK 5783 (salt tolerant) and Apex 836 (salt sensitive) were subjected to saline regime (100 mM NaCl) and two levels of Thi (100 and 125 mg l?1) applied as foliar spray. Salt stress markedly suppressed shoot and root dry mass, total chlorophylls (“a” + “b”), leaf water potential and maximum fluorescence yield (Fv/Fm) in the plants of both maize cultivars, but it increased proline accumulation, leaf osmotic pressure, malondialdehyde (MDA) and hydrogen peroxide (H2O2) concentrations, electrolyte leakage (EL) as well as activities of some key antioxidant enzymes, superoxide dismutase (SOD; EC. 1.15.1.1), peroxidase (POD; EC. 1.11.1.7) and catalase (CAT; EC. 1.11.1.6). Salt-induced reduction in plant growth parameters was higher in the salt-sensitive cultivar, Apex 836, which was found to be associated with relatively increased EL, and MDA and H2O2 levels, and decreased activities of the key antioxidant enzymes. Application of Thi as seed soaking or foliar spray partly mitigated the deleterious effects of salinity on plants of both maize cultivars. The most promising effect of Thi on alleviation of adverse effects of salt stress on maize plants was found when it was applied as foliar spray at 100 mg l?1. Thiamin application considerably reduced tissue Na+ concentration, but improved those of N, P, Ca2+ and K+ in the salt-stressed maize plants. Exogenously applied thiamin-induced growth improvement in maize plants was found to be associated with reduced membrane permeability, MDA and H2O2 levels, and altered activities of some key antioxidant enzymes such as CAT, SOD and POD as well as increased photosynthetic pigment concentration under saline regime.  相似文献

The changes in growth and photosynthetic performance of two wheat (Triticum aestivum L.) cultivars (Bolal-2973 and Atay-85) differing in their sensitivity to boron (B) toxicity were investigated under toxic B conditions. Eight-day old seedlings were exposed to highly toxic B concentrations (5, 7.5, and 10 mM H3BO3) for 5 and 9 days. Fast chlorophyll a fluorescence kinetics was determined and analysed using JIP test. Growth parameters, tissue B contents, and membrane damage were measured at two stress durations. The photochemical performance of PSII was hindered more in the sensitive cultivar (Atay-85) than that of the tolerant one (Bolal-2973) under B toxicity. The increase in the B concentration and stress duration caused membrane leakage in both cultivars. However, higher membrane damage was observed in Atay-85 compared to Bolal-2973. Additionally, significant reduction of growth parameters was observed in both cultivars at toxic B concentrations. The accumulation of B was higher in shoots than in roots of both cultivars. Nevertheless, Atay-85 translocated more B from roots to leaves compared to Bolal-2973. The advantages of certain JIP test parameters were demonstrated for evaluation of PSII activity in plants exposed to B stress. Evaluation of photosynthetic performance by JIP test as well as assessment of growth and tissue B content might be used to determine the effects of B toxicity in wheat. The results indicated lesser sensitivity to B toxicity in Bolal-2973 compared to Atay-85.  相似文献

The influence of pre-sowing seed treatment with polyamines (2.5 mM putrescine, 5.0 mM spermidine and 2.5 mM spermine) on growth, photosynthetic capacity, and ion accumulation in two spring wheat (Triticum aestivum L.) cultivars MH-97 (intolerant) and Inqlab-91 (tolerant) was examined. The primed seeds of each treatment and non-primed seeds were sown in a field containing 15 dS m−1 NaCl. Although all three polyamines were effective in improving shoot growth and grain yield in both cultivars under saline conditions, the effect of spermine was very pronounced particularly in improving grain yield. Different priming agents did not affect the net CO2 assimilation rate and transpiration rate of either cultivar. However, pre-treatment with spermidine increased stomatal conductance (gs) in the tolerant cultivar, whereas with spermine stomatal conductance decreased in the intolerant cultivar under salt stress. Priming agents had different effects on the accumulation of different ions in wheat plant tissues. When spermidine and distilled water were used as priming agents, they were effective in reducing shoot [Na+] in the tolerant and intolerant cultivars, respectively under saline conditions. Although all priming agents caused an increase in shoot [K+], distilled water was more effective in improving shoot [K+] in both cultivars under salt stress. Pre-treatment with spermidine was very effective in reducing shoot [Cl] under saline conditions particularly in the tolerant cultivar. However, the pattern of accumulation of different ions in roots due to different seed priming treatments was not consistent in either cultivar except that root Na+ decreased due to priming with spermine and spermidine in the intolerant and tolerant cultivars under saline conditions. In conclusion, although all three priming agents, spermine, spermidine and putrescine, were effective in alleviating the adverse effect of salt stress on wheat plants, their effects on altering the concentration of different ions and growth were different in the two cultivars differing in salt tolerance.  相似文献

Potato (Solanum tuberosum) is a major crop world-wide and the productivity of currently used cultivars is strongly reduced at high soil salt levels. We compared the response of six potato cultivars to increased root NaCl concentrations. Cuttings were grown hydroponically and treated with 0 mM, 60 mM and 180 mM NaCl for one week. Growth reduction on salt was strongest for the cultivars Mozart and Mona Lisa with a severe senescence response at 180 mM NaCl and Mozart barely survived the treatment. The cultivars Desiree and Russett Burbank were more tolerant showing no senescence after salt treatment. A clear difference in Na+ homeostasis was observed between sensitive and tolerant cultivars. The salt sensitive cultivar Mozart combined low Na+ levels in root and stem with the highest leaf Na+ concentration of all cultivars, resulting in a high Na+ shoot distribution index (SDI) for Mozart as compared to Desiree. Overall, a positive correlation between salt tolerance and stem Na+ accumulation was found and the SDI for Na+ points to a role of stem Na+ accumulation in tolerance. In stem tissue, Mozart accumulated more H2O2 and less proline compared to the tolerant cultivars. Analysis of the expression of proline biosynthesis genes in Mozart and Desiree showed a clear reduction in proline dehydrogenase (PDH) expression in both cultivars and an increase in pyrroline-5-carboxylate synthetase 1 (P5CS1) gene expression in Desiree, but not in Mozart. Taken together, current day commercial cultivars show promising differences in salt tolerance and the results suggest that mechanisms of tolerance reside in the capacity of Na+ accumulation in stem tissue, resulting in reduced Na+ transport to the leaves.  相似文献

Among trace metals, lead is a highly toxic contaminant, being hazardous to humans and animals. Application of maize plants for phytoremediation of polluted soils and waters has recently been of particular interest. The aim of this work is to investigate the Pb-phytoextraction potential of the maize cv. Tzariza used widely in Eastern European agriculture. Maize seedlings were exposed in a nutrient solution to 1–10000 µM of Pb2+ for 21 days. Lead accumulated mostly in conductive tissues and shoots at 0.1 mM and higher concentrations of Pb in growth medium. Pb at concentrations of 1 and 10 mM caused an increase in the superoxide anion level and the catalase activity in maize leaves. Lead ions were tolerable to maize seedlings within a concentration range up to 1000 µM of Pb2+. The levels of lead in the nutrient solution above 1 mM resulted in inhibition of the growth of axial organs, decrease in leaf area, inhibition of water absorption, and reduction in accumulation of biomass. Theoretical considerations indicate that in the temperate climates of the phytoremediation with maize may allow annual removal up to 90 kg of Pb per km2, depending on the initial level of soil contamination.  相似文献

Effects of fusaproliferin (FUS) on membrane potential (E M), electrolyte leakage, enzymes activity and respiration of roots, were studied in two maize cultivars (Zea mays L.), differing in their susceptibility to this toxin. In short-term experiments (≤ 6 h), E M has been rapidly and significantly depolarized by FUS. The rapidity of E M depolarization in tolerant cv. Lucia was more expressive in comparison with susceptible cv. Pavla, but the extent of E M depolarization was higher in cv. Pavla. In both maize cultivars, higher depolarization of E M was registered in cells of root zone I. In long-term experiments after the first E M depolarization, which occurred during the first 6 h of FUS treatment, gradual depolarization continued up to 24 h and was represented not only by the active component (E P) but also by the passive component (E D) of E M. The decrease in E M and E D was followed by a loss of K+ ions from FUS treated roots of both cultivars. The leak of K+ ions from the root cells of both root zones as well as both maize cultivars increased with the time of FUS treatment and was significantly higher in susceptible cv. Pavla than in tolerant cv. Lucia. FUS treatment of maize roots resulted in a significant decrease of root respiration which was higher in susceptible cv. Pavla than in tolerant cv. Lucia.  相似文献

Aluminum toxicity is an important stress factor in acid soils. Growth, respiration and permeability properties of root cells were studied in five cultivars of Lotus corniculatus subjected to aluminum (Al) or low pH stress. The cultivars showed significant differences in root elongation under stress conditions, which correlated with changes in membrane potential (EM) of root cortical cells. A pH drop from 5.5 to 4.0 resulted in significant membrane depolarization and root growth inhibition. The strongest inhibition was observed in cv. São Gabriel (33.6%) and least in cv. UFRGS (25.8%). Application of an extremely high Al concentration (2 mM) stopped the root growth in cv. INIA Draco, while inhibition in cv. UFRGS reached only 75%.The EM values of cortical cells of Lotus roots varied between −115 and −144 mV. Treatment with 250 μM of AlCl3 (pH 4) resulted in rapid membrane depolarization. The extent of the membrane depolarization ranged between 51 mV (cv. UFGRS) and 16 mV (cv. INIA Draco). The membrane depolarization was followed by a loss of K+ from Al-treated roots (2 mM Al) and resulted in a decrease of the diffusion potential (ED). The total amount of K+ in Al-treated roots dropped from 31.4 to 16.8 μmol g−1 FW in sensitive cv. INIA Draco, or from 26.1 to 22.7 μmol g−1 FW in tolerant cv. UFGRS. The rate of root respiration under control conditions as well as under Al treatment was higher in cv. INIA Draco than in cv. UFRGS. Al-induced inhibition of root respiration was 21–34% of the control.  相似文献

Salinity, as a serious and prevalent abiotic stress, causes widespread crop losses by restricting plant growth and production throughout the world. In this study, the biochemical and molecular responses of the pistachio (Pistacia vera L.) plant were studied under NaCl and salicylic acid (SA) treatments using hydroponically grown salt tolerant (Ghazvini) and salt sensitive (Sarakhs) pistachio cultivars. NaCl treatment (250 mM) increased the production of hydrogen peroxide (H2O2) and malondialdehyde (MDA) and the activity of antioxidant enzymes in both cultivars. In the sensitive cultivar, the H2O2 content was higher than the tolerant cultivar, especially in the roots. SA application to both salt-stress-treated cultivars resulted in an increase in photosynthetic pigment contents and antioxidant enzyme activity and a decrease in the H2O2 and MDA contents. After NaCl treatment, the isochorismate synthase (ICS) gene was upregulated in Ghazvini which leads to an increase in the SA content of the salt tolerant pistachio cultivar. In contrast, the salt treatment downregulated the expression of the ICS gene in Sarakhs. The ICS gene expression was positively regulated by SA treatment under the salt stress condition. Our results suggest that Ghazvini has higher salinity tolerance than Sarakhs due to its higher antioxidant capacity, photosynthetic pigment content, and the cultivar-specific expression pattern of the ICS gene. In this study, the potential alleviative effects of SA on the adverse effect of salt stress in P. vera (Pistacia vera) were also identified and highlighted.

We have identified two types of invertases, one bound ionically and the other covalently to the particulate fraction in grains of heat tolerant C 306 and heat susceptible WH 542 cultivars of wheat (Triticum aestivum L.). The cell walls contained a high level of invertase activity, of which 79.2–72.8% was extractable by 2 M NaCl and 14.9–21.1% by 0.5% EDTA in C 306 and WH 542, respectively. The NaCl-released invertase constituted the predominant fraction. Using 5–100 mM sucrose and pH range of 4.0–7.0, the apparent Michaelis constant (K m, enzyme substrate affinity measure) of enzyme ranged from 5.73 to 16.06 mM for C 306 and from 6.08 to 19.86 mM for WH 542. The V max (maximum catalytic rate) values at these pH were higher in C 306 (0.63–11.04 μg sucrose hydrolysed min−1) than WH 542 (0.51–8.73 μg sucrose hydrolysed min−1). By employing photo-oxidation and by studying the effect of pH on K m and V max, the involvement of histidine and α-carboxyl groups at the active site of the enzyme was indicated. The two cultivars also showed differential response in terms of thermodynamic properties of the enzyme i.e. energy of activation (E a), enthalpy change (ΔH) and entropy change (ΔS). NaCl-released invertase showed differential response to metal ions in two cultivars suggesting their distinctive nature. Mn2+, Cu2+, Hg2+, Mg2+, Zn2+ and Cd2+ were strong inhibitors in WH 542 as compared to C 306 while K+, Ca2+ were stimulators in both the cultivars. Overall the results suggest that genetic differences exist in wall bound invertase properties of wheat grains as evident in its altered kinetic behaviour.  相似文献

The influence of NaCl and Na2SO4 on growth of two maize cultivars (Zea mays cv. Pioneer 3906 and cv. Across 8023) differing in Na+ uptake was investigated in two green-house experiments. Na+ treatment with different accompanying anions (Cl?/SO42?) showed that ion toxicity was caused by Na+. While shoot growth of the two cultivars was markedly affected by salt in comparison to the control during the first 2–3 weeks, there were only slight differences between the cultivars. The shoot Ca2+ concentration was reduced in both cultivars, and the youngest leaves contained an even lower concentration compared with the rest of the shoot. During this first phase, Across 8023 tended to have higher concentrations of Ca2+ than Pioneer 3906. The Na+-excluding cultivar Pioneer 3906 showed continuous, although reduced, growth compared with the control, while the Na+ concentration in the shoot decreased until flowering. Cultivar Across 8023 accumulated Na+ until flowering: the reduction in the growth of stressed plants was greater than that for Pioneer 3906. Leaves of cultivar Across 8023 showed clear toxic symptoms, while those of the more salt-tolerant cultivar Pioneer 3906 did not. It is concluded that Na+ exclusion contributes to the salt tolerance of maize.  相似文献

Hydrolysis of Al3+ was performed in the presence of isolated root cell walls from a series of wheat cultivars (Triticum aestivum L.) known to have differential tolerance to Al contamination. Aluminium speciation was dependent on the cell wall concentration. At low cell wall concentrations, significant amounts of the very toxic Al13 species were formed. At higher cell wall concentrations, formation of the tridecamer was hindered or completely inhibited. The sensitive wheat cultivars displayed a higher affinity for aluminium than the tolerant cultivars. A possible Al tolerance mechanism based on cell wall permeability is discussed.  相似文献

Forty-four genotypes from the rice germplasm were identified under photoinhibition/photooxidation and shade conditions and divided into four basic types: (1) cultivars tolerant to both photooxidation and shading, (2) cultivars tolerant to photooxidation but sensitive to shading, (3) cultivars tolerant to shading but sensitive to photooxidation, and (4) cultivars sensitive to both photooxidation and shading. Photosynthetic characteristics of a cultivar tolerant (cv. Wuyugeng 3) and a cultivar sensitive (cv. Xiangxian) to photooxidation and shading were compared. The photochemical efficiency (Fv/Fm) of photosystem 2 (PS2) and the content of PS2-D1 protein in the tolerant cultivar Wuyugeng 3 decreased less under photooxidative conditions as compared with Xiangxian. Under similar conditions, superoxide dismutase was induced rapidly to a higher activity and the active oxygen (O) built up to a lower level in Wuyugeng 3 than in Xiangxian. Net photosynthetic rate (P N) decreased by 23 % in Wuyugeng 3 vs. 64 % in Xiangxian. Shading (80 %) during the booting stage caused only small decreases (7–13 %) in ribulose-1,5-bisphosphate carboxylase activity and P N in Wuyugeng 3 but severe decreases (57–64 %) were observed in Xiangxian which corresponded to the decreases in grain yield of the two cultivars (38 and 73 %, respectively). We described a simple and effective screening method and physiological basis for breeding crops for enhanced tolerance to both high and low irradiance.  相似文献

Boron (B) is an essential micronutrient for plants, which when occurs in excess in the growth medium, becomes toxic to plants. Rapid inhibition of root elongation is one of the most distinct symptoms of B toxicity. Hydrogen sulfide (H2S) is emerging as a potential messenger molecule involved in modulation of physiological processes in plants. In the present study, we investigated the role of H2S in B toxicity in cucumber (Cucumis sativus) seedlings. Root elongation was significantly inhibited by exposure of cucumber seedlings to solutions containing 5 mM B. The inhibitory effect of B on root elongation was substantially alleviated by treatment with H2S donor sodium hydrosulfide (NaHS). There was an increase in the activity of pectin methylesterase (PME) and up-regulated expression of genes encoding PME (CsPME) and expansin (CsExp) on exposure to high B concentration. The increase in PME activity and up-regulation of expression of CsPME and CsExp induced by high B concentration were markedly reduced in the presence of H2S donor. There was a rapid increase in soluble B concentrations in roots on exposure to high concentration B solutions. Treatment with H2S donor led to a transient reduction in soluble B concentration in roots such that no differences in soluble B concentrations in roots in the absence and presence of NaHS were found after 8 h exposure to the high concentration B solutions. These findings suggest that increases in activities of PME and expansin may underlie the inhibition of root elongation by toxic B, and that H2S plays an ameliorative role in protection of plants from B toxicity by counteracting B-induced up-regulation of cell wall-associated proteins of PME and expansins.  相似文献

The inevitable exposure of crop plants to salt stress is a major environmental problem emerged from the presence of excess NaCl radicals in the soil. Handling the problem in maize plants using a biological agent was the main interest of the present study. The non-pathogenic, halophytic, facultative aerobic bacterium Geobacillus caldoxylosilyticus IRD that was isolated from Marakopara pond in the Atoll Tikehau (French Polynesian, 2005) and found tolerant to salt stress until 3.5% NaCl (w/v). An artificial symbiosis was achieved by inoculating Geobacillus sp. into 5-day-old maize cultivars of triple hybrids (321 and 310) and singlet hybrids (10 and 162). Thereafter, maize seedlings were exposed to 350 mmol NaCl for 10 days. The data revealed that Geobacillus sp. had interacted with salinized maize and improved maize overall growth, dry weight and relative water content. Na+ accumulation was six times less and Cl accumulation was 13 times less in the tips of salinized maize seedlings upon Geobacillus sp. inoculation. Salinized maize without Geobacillus viewed decayed cortical cells of seedlings. In addition, proline content was two times higher in salinized seedlings lacking Geobacillus. Photosynthetic pigments and antioxidant enzymes were significantly regulated upon inoculation. Beyond this study, we presented a novel insight into a possible role of Geobacillus caldoxylosilyticus bacteria in controlling/protecting maize plants against high salt stress.  相似文献
