A correct separation of solids from liquid fraction is crucial for a successful treatment of swine manure. For this reason an in-depth study of flocculant addition on different livestock wastewaters was carried out. Two flushed swine manure matrices, namely the mixture from nursery and feeder-to-finish pigs and the feeder-to-finish slurry alone, were tested for solids and nutrients removals from liquid fractions. The separation techniques applied were sieving and flocculation. A range of 80-200 ppm of polyacrylamide (PAM) followed by screening was employed in the case of flocculation treatment. The best results were observed when using the highest PAM dose in the matrix correspondent to the mixture of slurries. The removal rates in the liquid fraction were 73% for total solids, 87% for volatile solids, 98% for suspended total and volatile solids, 71% for chemical oxygen demand, 40% for total Kjeldahl nitrogen, and 34% for soluble phosphorus. Once the best PAM dose (120 ppm) was chosen, an anaerobic biodegradability study was performed in order to check the increase of methane production in the separated fractions by using the flocculant and the screen. The assay determined that the solid fractions biodegradability was constant at 79%. Meanwhile for the liquid fractions, an increase of 9% points was achieved with PAM-amendment when compared with 82% reached for the liquid fraction obtained by screening.  相似文献

Anaerobic digestion plants rely on large-capacity storage tanks to manage the agronomic utilisation of the digestate. As a consequence, many Italian A.D. plants have introduced mechanical separation of the digested slurry to simplify process requirements. This study evaluated the possibility of reusing mechanically-separated solid fraction as a further biomass input anaerobic digestion plants. The effects of storage and thermal pre-treatment on digested solid fraction were assessed through biogas and methane yield measures, and then compared to the yields associated with undigested solid fraction of raw pig slurry. The specific CH4 yields of digested solid fractions ranged between 71.4 and 156.9 lN/kg VS, whereas the biogas yield from undigested solid fractions was 78.7 lN/kg VS. Solid fraction storage showed no significant effect on specific CH4 yields in any of the examined samples. However, in the case of the undigested solid fraction, thermal pre-treatment proved to be an effective method to increase CH4.  相似文献

The aim of the present work was to assess the effect of treatments by acidification, solid–liquid separation or acidification followed by solid–liquid separation on the physical and chemical composition of pig slurry (S) and pig slurry fractions (non acidified and acidified solid (SF and ASF) and liquid (LF and ALF) fractions), as well as on the potential of N mineralization of these pig slurry derived materials. Acidification strongly decrease the inorganic carbon content of S, SF and LF and it also affects the distribution of P, Ca and Mg between the solid and liquid fraction leading to an ALF more equilibrated than LF in terms of nutrients. Acidification increases the potential of organic N mineralization in SF and decreases the potential of N immobilization in S and LF. It can be concluded that the proposed treatment generates valuable slurry fractions with distinct characteristics and potential of N mineralization that may be incorporated to soil at different periods after sowing to comply with plant nutrient requirements.  相似文献

Increased swine production results in concentration of wastes generated within a limited geographical area, which may lead to land application rates exceeding the local or regional assimilatory capacity. This may result in pollutant transfer through surface water or soil-groundwater systems, environmental degradation, and/or odor concerns. Existing swine waste pit storage and lagoon treatment technologies may be inadequate to store or treat waste prior to land application without these concerns resulting. Efficient swine waste solids separation may reduce environmental health concerns and generate a value-added bioresource (solids). This study evaluated the efficiency of a polyacrylamide (PAM) flocculant-aided solids separation treatment to reduce pollution indicator concentrations in raw (untreated) swine waste slurry. Swine waste slurry solids separation efficiency through gravity settling (sedimentation) was evaluated before and after the addition of a proprietary polymeric (PAM) flocculant. Results indicated that polymer amendments at concentrations of 62.5-750 mg/l improved slurry solids separation efficiency and significantly reduced concentrations of other associated aquatic pollution indicators in a majority of analyses conducted (33 of 50 total analyses conducted). Results also suggested that PAM-aided solids separation from swine waste slurry might facilitate further treatment and/or disposal and therefore reduce associated environmental degradation potential.  相似文献

Mechanical separation of liquid animal manure can be an effective technique for production of a liquid and a nutrient-rich solid fraction. The efficiency of separators depends on the physical and chemical composition of the animal manure. Therefore, the particle size composition was measured for different types of manure before treatment with a decanting centrifuge and a screw press. Storage of pig manure reduces the total dry matter content, and the content of small particles (< 0.0016 mm) is reduced more than the content of large particles. In consequence, the proportion of large particles will increase, while the portion of small particles will decrease. The separation efficiency of the screw press was found to be low, as this separator only retains particles > 1 mm. The decanter centrifuge retained all the particles > 0.02 mm and was therefore much more efficient than the screw press. Separation efficiency was also found to be highly dependent on the type of manure used.  相似文献

Liquid livestock waste can be managed by separating liquid and solid fractions then treating each separately by applying best available technology, such as anaerobic digestion for the solid fraction. There is an increasing use of polyacrylamide (PAM) as a flocculant agent to improve solid-liquid separation. In the present work, the anaerobic toxicity of PAM residues and the optimal range of total solids concentration for maximum methane production were studied as a function of PAM dosage. Results showed that dry matter and its volatile solids content increased significantly with increasing PAM dosage. Batch anaerobic tests showed that methane yield decreased linearly with increasing total solids, while the methane production per unit of raw substrate reached a maximum at 16.4% total solids. No PAM toxicity was measured for PAM concentrations below 415 g/kg total solids, but some indirect inhibitory phenomena were observed, such as a limited hydrolysis rate due to particle aggregation, and inhibition of methanogenesis by high ammonia concentration.  相似文献

The aim of this study was to compare the environmental impact of the straw-flow system for fattening pigs with the slatted-floor system by measuring pollutant gas emissions such as ammonia (NH3), nitrous oxide (N2O), methane (CH4) and carbon dioxide (CO2), manure nitrogen (N) content and emissions of water vapour (H2O). Three successive batches of 32 pigs were fattened. For each batch, pigs were allotted to two groups raised in separated rooms fitted either with a concrete totally slatted-floor system (0.75 m2 per pig) or with a straw-flow system (0.79 m2 per pig). With this last system, pigs were kept on a sloped floor, straw being provided daily at the top of the pen. Throughout the fattening period, about 34.4 kg of straw were supplied per pig. The straw, mixed with dung, travelled down the slope by pig motion and went out of the pen to a scraped passage. The solid fraction was scraped every day, stored in a heap in the room and removed every month, 1 week before each period of gaseous emission measurement. The liquid fraction was automatically pumped from the scraped passage into a hermetic tank, which was emptied at the end of each fattening period. Rooms were ventilated mechanically in order to maintain a constant ambient temperature. Once a month, the emissions of NH3, N2O, CH4, CO2 and H2O were measured hourly for 6 consecutive days via infrared photoacoustic detection. Mean daily emissions per pig fattened on the slatted floor or on the sloped floor were, respectively, 4.98 and 13.31 g NH3, 0.67 and 0.68 g N2O, 15.2 and 8.88 g CH4, 548 g and 406 g CO2 equivalents, 1.61 and 1.77 kg CO2 and 2.33 and 2.95 kg H2O. Except for N2O emissions, all the differences were statistically significant (P < 0.001). From the slatted-floor system, the amount of slurry removed per fattening period was on average 256 kg per pig. From the straw-flow system, solid manure amounted on average to 209 kg per pig and liquid manure to 53 kg per pig. The total N-content of the manure was 2.23 kg N per pig with the straw-flow system (solid and liquid manure) v. 3.26 kg N per pig for slurry from the slatted-floor system. This reduction of 30% observed with the sloped floor was mainly explained by the higher level of NH3-N emissions.  相似文献

Pathogens may reach agricultural soils through application of animal manure and thereby pose a risk of contaminating crops as well as surface and groundwater. Treatment and handling of manure for improved nutrient and odor management may also influence the amount and fate of manure-borne pathogens in the soil. A study was conducted to investigate the leaching potentials of a phage (Salmonella enterica serovar Typhimurium bacteriophage 28B) and two bacteria, Escherichia coli and Enterococcus species, in a liquid fraction of raw pig slurry obtained by solid-liquid separation of this slurry and in this liquid fraction after ozonation, when applied to intact soil columns by subsurface injection. We also compared leaching potentials of surface-applied and subsurface-injected raw slurry. The columns were exposed to irrigation events (3.5-h period at 10 mm h−1) after 1, 2, 3, and 4 weeks of incubation with collection of leachate. By the end of incubation, the distribution and survival of microorganisms in the soil of each treatment and in nonirrigated columns with injected raw slurry or liquid fraction were determined. E. coli in the leachates was quantified by both plate counts and quantitative PCR (qPCR) to assess the proportions of culturable and nonculturable (viable and nonviable) cells. Solid-liquid separation of slurry increased the redistribution in soil of contaminants in the liquid fraction compared to raw slurry, and the percent recovery of E. coli and Enterococcus species was higher for the liquid fraction than for raw slurry after the four leaching events. The liquid fraction also resulted in more leaching of all contaminants except Enterococcus species than did raw slurry. Ozonation reduced E. coli leaching only. Injection enhanced the leaching potential of the microorganisms investigated compared to surface application, probably because of a better survival with subsurface injection and a shorter leaching path.  相似文献

Individual stereoisomers of the phosphinic pseudodipeptide hPhepsi[P(O)(OH)CH(2)]Phe were obtained by stereoselective liquid chromatographic separation as N- and C-terminally protected derivative on quinidine carbamate modified silica stationary phase. The stereoisomeric purity, exceeding 95% for each fraction, was determined before and after deprotection using two independent methods. The absolute configuration was rationally assigned by application of enantiomerically pure phosphinic acid substrates in the synthetic procedure and correlation with biological activity of the products. Substantial differences in inhibition of leucine aminopeptidase by the individual isomers revealed novel insights into potency of the recently developed and remarkably effective compound.  相似文献

通过对黑土稻田CH4和N2O排放的观测,发现水稻生长季CH4和N2O排放量低于全国其它地区稻田CH4和N2O排放之间存在互为消长关系(r=-0.513,P<0.05),但在同样施肥水平条件下,间歇灌溉与长期淹灌相比,CH4排放明显减少而N2O略有增加,其相对综合温室效应被大大减少且水稻产量未受影响。为此,间歇灌溉可作为减少稻田温室气体排放的水分管理措施。另外,通过对CH4和N2O排放的相关微生物过程探讨,揭示产甲烷菌数与CH4排放问呈显著性正相关(R2=0.82,P<0.05),硝化菌数和反硝化菌数与N2O排放有重要关系。  相似文献

One hundred million tons of farm stalk waste and livestock and poultry excrement are used every year in China for the production of clean energy (biogas) by anaerobic digestion. Consequently, a large amount of fermented liquid is produced, and if disposed of improperly, it will result in secondary pollution. Agricultural application of this anaerobic slurry as a liquid fertilizer would reduce possible eutrophication of water sources from random slurry discharge and supply a superior organic fertilizer for farming. This study investigated the effect of applying anaerobic digestion slurry as a liquid fertilizer on the methane emitted from a paddy field. A two‐year (2008–2009) field experiment replacing chemical fertilizer with liquid fertilizer from anaerobically digested pig manure slurry was conducted in a paddy field in Yixing, Jiangsu, China. A static closed chamber method was used to measure methane fluxes over the period from June 2008 to October 2009. All fertilizer treatments increased methane emissions relative to untreated controls, with increases in methane ranging from 40–70% in 2008 to 48–84% in 2009. Paddy fields treated with anaerobically digested pig manure slurry had greater methane emissions (8–84% in 2008 and 3–26% in 2009) than those treated with chemical fertilizer. This suggests that the anaerobic digestion slurry would increase methane emission and so is unsuitable as a liquid fertilizer in paddy fields without development of cultivation practices to limit these emissions.  相似文献

Ruminant husbandry constitutes the most important source of anthropogenic methane (CH4). In addition to enteric (animal-derived) CH4, excreta are another source of CH4, especially when stored anaerobically. Increasing the proportion of dietary concentrate is often considered as the primary CH4 mitigation option. However, it is unclear whether this is still valid when diets to be compared are energy-balanced. In addition, non-structural carbohydrates and side effects on nitrogen (N) emissions may be important. In this experiment, diet types representing either forage-only or mixed diets were examined for their effects on CH4 and N emissions from animals and their slurries in 18 lactating cows. Apart from a hay-only diet, treatments included two mixed diets consisting of maize stover, pelleted whole maize plants and gluten or barley straw and grain and soy bean meal. The diets were balanced in crude protein and net energy for lactation. After adaptation, data and samples were collected for 8 days including a 2-day CH4 measurement in respiratory chambers. Faeces and urine, combined proportionately according to excretion, were used to determine slurry-derived CH4 and N emissions. Slurry was stored for 15 weeks at either 14°C or 27°C, and temperatures were classified as 'cool' and 'warm', respectively. The low-starch hay-only diet had high organic matter and fibre digestibility and proved to be equally effective on the cows' performance as mixed diets. The enteric CH4 formation remained unaffected by the diet except when related to digested fibre. In this case emission was lowest with the hay-only diet (61 v. 88 to 101 g CH4/kg digested NDF). Feeding the hay diet resulted in the highest slurry-CH4 production after 7 weeks of storage at 14°C and 27°C, and after 15 weeks at 14°C. CH4 emissions were, in general, about 10-fold higher at 27°C compared with 14°C but only after 15 weeks of storage. Urinary N losses were highest with the barley diet and lowest with the maize diet. There was a trend towards similar differences in N losses from the slurry of these cows (significant at 14°C). However, contrary to CH4, slurry-N emissions seemed to be temperature-independent. In conclusion, energetically balanced diets proved to be widely equivalent in their emission potential when combining animal and their slurry, this even at a clearly differing forage : concentrate ratio. The variation in CH4 emission from slurry stored shortly or at cold temperature for 15 weeks was of low importance as such conditions did not support methanogenesis in slurry anyway.  相似文献

The production of energy crops for farm-scale anaerobic digestion (AD) can affect emissions of greenhouse gases (GHG) in a number of ways. Some examples are: fugitive CH4 emissions from the digester and the storage of the digestate, emissions of N2O from soil and emissions of CO2 from farm machinery. Moreover, uptake of AD may be accompanied by changes in the way the farm is operated, which may affect GHG emissions. The scale of these emissions was assessed from published data for the biogas feedstocks cattle slurry and grass silage. Emissions were compared to references representing current farm operation and energy generation by fossil fuels. Feeding the digester with cattle slurry for the entire year did not result in reduced emissions due to relatively high emissions from stored raw slurry in summer. If grass was used for digester feedstock, the level of N2O emissions from the crop was the most important factor for the GHG balance of farm-scale AD. If N2O emissions were low, biogas realised substantial savings of GHG in the order of 1 t CO2 equivalents per hectare per year. At a high level of N2O emissions, energy cropping might even result in increased GHG emissions compared to fossil fuels.  相似文献

AIMS: To examine the prevalence and diversity of bacterial faecal pathogens in unseparated slurry, separated solids and liquid fractions from a commercial pig farm. METHODS: A total of 43 stored slurry specimens originating from a fattening house over the period February-April 2002 were analysed, consisting of unseparated (n = 14) slurry, separated solids (n = 16) and separated liquid (n = 13). Specimens were examined for the presence of five bacterial pathogens including Salmonella spp., Shigella spp., Campylobacter spp., Escherichia coli O157 and Yersinia enterocolitica. Selective enrichment and plating methods were employed for detection of Salmonella spp. and Campylobacter spp. and conventional selective plating techniques for the remaining genera. Antibiogram profiles to 12 antibiotic agents were obtained for all Salmonella isolates obtained. RESULTS: Salmonella spp. were identified in all components of the slurry specimens, whereas Campylobacter spp. was only recovered from the unseparated and separated liquid fractions. In both cases, the separated liquid fraction had the highest prevalence of pathogens and the separated solid fraction had the lowest prevalence. None of the slurry specimens examined were positive for E. coli O157:H7, Shigella spp. or Y. enterocolitica. Twenty-nine isolates of Salmonella were recovered from the slurry specimens, comprising seven serovars, of which Salmonella manhattan was the most prevalent, accounting for over half [15 of 29 (51.7%)] of all Salmonella isolates. Salmonella anatum, Salm. derby, Salm. give, Salm. heidelberg, Salm. simi and Salm. stanley serovars were also recovered. All Salmonella isolates were sensitive to ampicillin, augmentin (amoxicillin/clavulanic acid), chloramphenicol, ciprofloxacin, gentamicin, kanamycin and trimethoprim, but has variable resistance to tetracycline (100%), sulphonamides (84.6%), furazolidone (38.5%), nalidixic acid (15.4%) and streptomycin (15.4%). The majority (57.7%) of isolates displayed antibiotic resistance to at least two antibiotic agents, followed by 34.6% of isolates being resistant to three agents and the remainder (7.7%) being resistant to four antibiotics. SIGNIFICANCE AND IMPACT OF THE STUDY: This study demonstrated a marked reduction in the prevalence of Campylobacter and Salmonella in the solids component of separated pig slurry. The adoption of control processes such as aeration of slurry prior to its spread onto agricultural land and newer approaches to pathogen reduction should be investigated, to reduce the transmission of pathogens from pig slurry to the environment.  相似文献

A novel proteomic approach for probing cell and tissue proteome, which combines liquid phase protein separations with microarray technology has been developed. Proteins in cell and tissue lysates or in cellular subfractions are separated using any one of a number of separation modes which may consist of ion exchange liquid chromatography (LC), reverse phase LC, carrier ampholyte based separations, e.g. the use of Rotofor, affinity based separations, or gel based separations. Each first-dimension fraction obtained using one separation mode can be further resolved using one or more of the other separation modes to yield either purified protein in solution or liquid fractions with substantially reduced protein complexity. The advantage of a liquid based separation system is that proteins in hundreds of individual fractions can be arrayed directly and used as targets for a variety of probes. Constituent proteins in reactive fractions are identified by mass spectrometry and may be further resolved to determine the nature of the reactive protein(s). We present in this report initial data based on microarray analysis of individual Rotofor fractions obtained from lung adenocarcinoma cell line A549 lysates which have been probed with antibodies against specific proteins.  相似文献

The haemolytic activity of the total Complement (CH50) and the fractions C3 and C4 were assayed in rats transplanted with Yoshida's tumor and then treated with hCG, LH-FSH and PGE2. A relevant increase, only concerning the values of the CH50 and C3 fraction, was observed in all animals in the early days after the transplantation, probably due to a sort of stress "by transplantation". Afterwards, hCG and PGE2 induced an increase in CH50 and C3 values, but not in the C4 fraction. Treatment with LH and FSH led to a very slight increase in the CH50 and C3. In the following days, as a consequence of the cachexia, a progressive reduction of the values of the Complement was observed in all animals. Those treated with hCG also showed a little increase of survival. The authors suggest that the increase in CH50 and C3 fraction induced by the treatment with hCG and PGE2 could be an expression of increase of the aspecific humoral immunity, as a compensatory mechanism of the cell-mediated immunological depression which occurs during neoplasias.  相似文献

Agriculture is an important source of ammonia (NH3), which contributes to acidification and eutrophication, as well as emissions of the greenhouse gases nitrous oxide (N2O) and methane (CH4). Controlling emissions of one of these pollutants through application of technical measures might have an impact (either beneficial or adverse) on emissions of the others. These side effects are usually ignored in policy making. This study analyses cost-effectiveness of measures to reduce acidification and eutrophication as well as agricultural emissions of N2O and CH4 in Europe, taking into account interrelations between abatement of NH3, N2O, and CH4 in agriculture. The model used is based on the RAINS (Regional Air pollution INformation and Simulation) model for air pollution in Europe, which includes emissions, abatement options, and atmospheric source-receptor relationships for pollutants contributing to acidification and eutrophication. We used an optimisation model that is largely based on the RAINS model but that also includes emissions of N2O and CH4 from agriculture and technical measures to reduce these emissions. For abatement options for agricultural emissions we estimated side effects on other emissions. The model determines abatement strategies to meet restrictions on emission and/or deposition levels at the least cost. Cost-effective strategies to reduce acidification and eutrophication in Europe were analysed. We found that NH3 abatement may cause an increase in N2O emissions. If total agricultural N2O and CH4 emissions in Europe were not allowed to increase, cost-effective allocation of emission reductions over countries in Europe changed considerably.  相似文献

通过盆栽试验,研究了脲酶抑制剂氢醌(HQ)、硝化抑制剂双氰胺(DCD)及二者的组合(HQ+DCD)对种稻土壤N2O和CH4排放的影响.结果表明,在未施麦秸粉时,所有施抑制剂的处理均较单施尿素的能显著减少水稻生长期供试土壤N2O和CH4的排放.特别是HQ+DCD处理,其N2O和CH4排放总量分别约为对照的1/3和1/2.而在施麦秸粉后,该处理的N2O排放总量为对照的1/2,但CH4排放总量却较少差别.不论是N2O还是CH4的排放总量,施麦秸粉的都比未施的高出1倍和更多.因此,单从土壤源温室气体排放的角度看,将未腐熟的有机物料与尿素共施,并不是一种适宜的施肥制度.供试土壤的N2O排放通量,与水稻植株的NO-3N含量和土表水层中的矿质N量分别呈显著的指数正相关和线性正相关;CH4的排放通量则与水稻植株的生长量和土表水层中的矿质N量呈显著的线性负相关.在N2O与CH4的排放间,未施麦秸粉时存在着定量的相互消长关系;施麦秸粉后,虽同样存在所述关系,但难以定量化.  相似文献

A field trial was carried out on a 15 year old Miscanthus stand, subject to nitrogen fertilizer treatments of 0, 63 and 125 kg‐N ha?1, measuring N2O emissions, as well as annual crop yield over a full year. N2O emission intensity (N2O emissions calculated as a function of above‐ground biomass) was significantly affected by fertilizer application, with values of 52.2 and 59.4 g N2O‐N t?1 observed at 63 and 125 kg‐N ha?1, respectively, compared to 31.3 g N2O‐N t?1 in the zero fertilizer control. A life cycle analyses approach was applied to calculate the increase in yield required to offset N2O emissions from Miscanthus through fossil fuel substitution in the fuel chain. For the conditions observed during the field trial yield increases of 0.33 and 0.39 t ha?1 were found to be required to offset N2O emissions from the 63 kg‐N ha?1 treatment, when replacing peat and coal, respectively, while increases of 0.71 and 0.83 t ha?1 were required for the 125 kg‐N ha?1 treatment, for each fuel. These values are considerably less than the mean above‐ground biomass yield increases observed here of 1.57 and 2.79 t ha?1 at fertilization rates 63 and 125 kg‐N ha?1 respectively. Extending this analysis to include a range of fertilizer application rates and N2O emission factors found increases in yield necessary to offset soil N2O emissions ranging from 0.26 to 2.54 t ha?1. These relatively low yield increase requirements indicate that where nitrogen fertilizer application improves yield, the benefits of such a response will not be offset by soil N2O emissions.  相似文献
