Synthetic genes were constructed based on the known sequence of the spider dragline silk protein MaSp 2. The genes had 8, 16, or 32 contiguous units of the consensus repeat sequence of the protein. These artificial genes were constructed using a strategy involving compatible but nonregenerable restriction sites, which allowed construction of very large inserts in a precisely controlled manner. This strategy should have general utility in the controlled construction of repetitive proteins composed of identical or different repeat units. The protein from the 16-unit repeat was produced inEscherichia coliat levels up to 10 mg/g wet wt of cells although yields of 1–2 mg/g were more typical. The protein was easily purified with high recovery using an affinity column. The purified protein had the predicted amino acid composition and N-terminal sequence after cleavage of a leader sequence. The methodology described will allow production of sufficient quantities of protein for basic structure/function studies including production of synthetic fibers.  相似文献

Predator–prey relationships are generally based on arm-race. Wasps and spiders are both predators, which could be potential prey for each other. The orb weaver spider Zygiella x-notata is sometimes a prey for the wasp Vespula germanica. We observed the wasp hunting behaviour under natural conditions, and we tested the influence of the spider’s behaviour on the wasp attack success. Wasps were active predators during the reproductive period of the spider. Results showed that wasps located more easily male spiders than females particularly when they were engaged in mate guarding. Female location depended on the presence of a web, but also of prey or prey remains in the web. On the other hand, their location depend neither on the characteristics and the position of the retreat in the environment nor on the size of the web. After location, males were more often captured than females whatever their behaviour (mate guarding or not). Presence of prey remains or prey in the web did not increase the risk for the spider to be captured. There was also no influence of the retreat’s characteristics or of its position in the habitat on the risk for the spider to be captured; but wasp successful attacks were less numerous when silk was present around the entrance of the retreat or when the spider was completely inside. As prey and prey remains favoured location of spiders by the wasps, we tested spider web cleaning behaviour as a response to wasp predatory pressure. By throwing small polystyrene pellets in the webs, we observed that more 80% of the spiders rejected the pellets in less than one minute. Our data indicated that wasps were significant predators of Z. x-notata and wasp attack could have been a selective pressure that had favoured spider defensive behaviours such as web cleaning.  相似文献

A trap-nesting study provided the first documentation of brood parasitism in Dipogon nagasei and in D. iwatai. Dipogon nagasei was found to brood parasitize D. sperconsus, D. conspersus, D. inconspersus, and D. bifasciatus. Dipogon iwatai brood parasitized D. sperconsus, D. conspersus, D. romankovae, and Auplopus carbonarius. Both brood parasitic species are with the subgenus Nipponodipogon, whereas all five Dipogon hosts are in the subgenus Deuteragenia. Comparison of their ecological features revealed that brood parasitism in D. nagasei is considerably more derived than in D. iwatai. Of particular note is the fact that in D. nagasei the female routinely lays up to five eggs on a single host spider, all of which develop into adult wasps without larval cannibalism; almost all Pompilidae previously studied lay only one egg on a host spider.  相似文献

本文记述了分别采自云南省和湖南省的拟平腹蛛科斯托蛛属Storenomorpha 2新种:云南斯托蛛,新种S.yunnan sp.nov.和S.yizhang sp.nov..云南斯托蛛腹部背而两侧斑纹三角形,前后连接成行.粗壮的后胫节突,前端有2个距,其中背距长而有1扭曲.外雌器中隔宽大,交媾腔分为两侧腔,较大,纳精囊位于近生殖厣后缘,交媾管粗,由后向前,在交媾腔前方有1个拐点,扭曲向后与纳精囊相连.宜章斯托蛛腹部背面两侧斑纹4对,彼此分离不连成线,后胫节突和腹距长于背距,插入器鞭状,较云南斯托蛛长,末端超出生殖球一段距离.2新种形态结构有差异.两者以其后跗节突远端2个距的长度明显不同以及腹斑部背斑各异与模式种S.comottoi Simon,1884都不相同.模式标本云南斯托蛛正模、部分副模保存于湖南师范大学(HNU),部分副模将保存于美国加州科学学院(CAS).宜章斯托蛛模式种保存于湖南师范大学(HNU).文中量度单位为mm.  相似文献

通过对两种果外产卵非传粉榕小蜂Philotrypesissp.和Sycoscaptersp.产卵行为的详细观察,发现两种小蜂产卵期都集中在榕果发育的间花期,并且只在进过传粉榕小蜂的榕果内产卵。这两种小蜂的产卵行为基本都可以分为寻找产卵位点、刺壁、产卵和回收产卵器等步骤。落在果面上的两种小蜂的繁殖雌蜂都通过触角敲击果面寻找产卵位点。产卵结束后,Philotrypesissp.大多数情况在原位收回产卵针,而Sycoscaptersp.必须向前爬行才能将产卵针从果内收回。为了争夺产卵位点,在同一榕果产卵的Philotrypesissp.繁殖雌蜂之间会进行打斗。而Sycoscaptersp.繁殖雌蜂之间未观察到打斗行为。两种小蜂产卵器长度虽显著长于各自产卵时榕果果壁厚度,但却显著短于其产卵时榕果果壁和子房层的总厚度,说明这两种小蜂采用产卵针直接刺穿小花子房的产卵模式。  相似文献

记述采自贵州省施秉县一洞穴小弱蛛属Leptonetela一新种,施秉小弱蛛Leptonetela shibingensis sp.nov.。该新种近似于查氏小弱蛛Leptonetela zhai WangLi,2011,主要区别有:眼退化,仅留眼丘,后者6眼完全;雄蛛触肢器中突(median apophysis,M)小,三角形,而后者中突大,近掌状;雄蛛触肢器胫节基刺(basal spine of male palpal tibia,TBS)短而粗,末端向内弯曲。模式标本保存于贵州科学院生物研究所。  相似文献

Rhizobia are the common bacterial symbionts that form nitrogen-fixing root nodules in legumes. However, recently other bacteria have been shown to nodulate and fix nitrogen symbiotically with these plants. Neptunia natans is an aquatic legume indigenous to tropical and subtropical regions and in African soils is nodulated by Allorhizobium undicola. This legume develops an unusual root-nodule symbiosis on floating stems in aquatic environments through a unique infection process. Here, we analyzed the low-molecular-weight RNA and 16S ribosomal DNA (rDNA) sequence of the same fast-growing isolates from India that were previously used to define the developmental morphology of the unique infection process in this symbiosis with N. natans and found that they are phylogenetically located in the genus Devosia, not Allorhizobium or Rhizobium. The 16S rDNA sequences of these two Neptunia-nodulating Devosia strains differ from the only species currently described in that genus, Devosia riboflavina. From the same isolated colonies, we also located their nodD and nifH genes involved in nodulation and nitrogen fixation on a plasmid of approximately 170 kb. Sequence analysis showed that their nodD and nifH genes are most closely related to nodD and nifH of Rhizobium tropici, suggesting that this newly described Neptunia-nodulating Devosia species may have acquired these symbiotic genes by horizontal transfer.  相似文献

4种胡蜂前溶血肽原基因的克隆与序列比较   总被引:4,自引:1,他引:4  
从雌性亚非马蜂、额斑黄胡蜂、墨胸胡蜂、大胡蜂毒腺中抽提总RNA,通过RT-PCR方法扩增得到4种胡蜂蜂毒前溶血肽原的cDNA,再将扩增产物克隆到pGEM⑥-Teasy vector上。测序结果表明:扩增得到的片段长度均为213bp,系蜂毒前溶血肽原编码区的cDNA。经序列比较,4种胡蜂蜂毒前溶血肽原之间的氨基酸序列同源性都超过95%。亚非马蜂、额斑黄胡蜂、墨胸胡蜂、大胡蜂各自与意大利蜜蜂蜂毒前溶血肽原氨基酸序列的同源性分别为95.8%、100%、97.2%、97.2%。结果表明:蜂毒前溶血肽原一级结构序列具有很高的保守性,尽管胡蜂和蜜蜂属于膜翅目不同的总科,但它们的前溶血肽原基因却非常相似。  相似文献

1998~1999年、1999~2000年在武汉地区使用陷阱法系统采样,研究了大麦田蜘蛛群落结构,比较了不同的冬天气候因素对麦田蜘蛛群落结构的影响.使用Berger-Parker优势度指数分析了调查时间段大麦田的地面优势种蜘蛛.结果显示,1998~1999年的优势种蜘蛛为皿蛛科蜘蛛,隆背微蛛Erigone prominens Boes. et Str.在大麦的全生长期均为优势种.1999~2000年,主要的优势种蜘蛛为狼蛛科的类水狼株Pirata piratoides Boes.et Str.、沟渠豹蛛Pardosa laura Karsch和拟环纹豹蛛Pardosa pseudoannulata (Boes.et Str.).另外,通过各优势种蜘蛛雄蛛的个体数变化,探讨了各优势种蜘蛛在田间的交配繁殖情况.结果表明,冬天气候对蜘蛛的交配时期有明显影响.在暖冬天气,大多数种类的交配比冷冬提前.一般情况下,各优势种蜘蛛能在麦田繁殖1代.另外,还对平腹蛛科的2种蜘蛛的雄蛛在田间的活动情况进行了描述.  相似文献

Mycobacterium abscessus subsp. massiliense, a rapidly growing mycobacteria (RGM) that is becoming increasingly important among human infectious diseases, is virulent and pathogenic and presents intrinsic resistance to several antimicrobial drugs that might hamper their elimination. Therefore, the identification of new drugs to improve the current treatment or lower the risk of inducing resistance is urgently needed. Wasp venom primarily comprises peptides that are responsible for most of the biological activities in this poison. Here, a novel peptide Polydim-I, from Polybia dimorpha Neotropical wasp, was explored as an antimycobacterial agent. Polydim-I provoked cell wall disruption and exhibited non-cytotoxicity towards mammalian cells. Polydim-I treatment of macrophages infected with different M. abscessus subsp. massiliense strains reduced 40 to 50% of the bacterial load. Additionally, the Polydim-I treatment of highly susceptible mice intravenously infected with M. abscessus subsp. massiliense induced 0.8 to 1 log reduction of the bacterial load in the lungs, spleen, and liver. In conclusion, this is the first study to show the therapeutic potential of a peptide derived from wasp venom in treating mycobacteria infections. Polydim-I acts on the M. abscessus subsp. massiliense cell wall and reduce 40–90% of the bacterial load both in vitro and in vivo. The presented results encourage further studies on the use of Polydim-I as one of the components for M. abscessus subsp. massiliense treatment.  相似文献
