Artificially prepared cow pats containing Ostertagia ostertagi eggs were deposited on two pasture plots in May, June and July 1986. Half of the cow pats, placed on one plot, were inoculated with 2000 conidia per gram faeces of the predacious fungus Arthrobotrys oligospora. On the other plot fungus-free control cow pats were placed at the same time. In the faeces generally fewer infective O. ostertagi larvae developed in the inoculated than in the control cow pats. On the herbage around the control cow pats deposited in May, June and July a maximum concentration of infective larvae was found at the same time on the 6th of August 1986. At that time the herbage larval infectivity around inoculated cow pats deposited in May, June and July was subject to a reduction of 48%, 89% and 46%, respectively, compared with fungus-free control cow pats. This experiment indicates that a concentration of 2000 A. oligospora conidia per gram faeces results in a significant lowering of the herbage larval infectivity during the grazing season in Denmark.  相似文献

The aim of this present study was to compare the kinetics of nuclear maturation between calf and cow oocytes in order to determine if there are differences between the 2 groups which could explain their disparate developmental capacity. The constitutive and neosynthetic protein patterns of cow and calf oocytes and of their corresponding cumulus cells were also compared during in vitro maturation. A total of 397 calf oocytes and 406 cow oocytes was matured in M199 + 10 ng/mL EGF. The first group of oocytes (n = 30) was immediately fixed and stained after removal from the follicle, and represent 0 h. The remaining oocytes were removed from the maturation medium at 4, 8, 12, 16, 20 and 24 h respectively. Half were denuded, fixed and stained for nuclear status; while the remainder were radiolabeled with methionine-(35S). Immediately after isolation, all the oocytes were at the GV stage. By 8 h, GVBD had occurred in most oocytes (calf: 97%; cow: 100%) and some had reached pro-metaphase I (calf: 49%; cow: 51%). By 12 h, most of the oocytes were at metaphase I (calf: 84%; cow: 94%). By 16 h, 54% of calf oocytes had reached telophase I or beyond compared with 71% of cow oocytes. This difference between the 2 groups became significant by 20 h, with 89% of cow oocytes (P < 0.05) at metaphase II and 71% of calf oocytes. By 24 h of culture, GVBD had occurred in all cases. Most oocytes completed meiosis I and were arrested at metaphase II with the first polar body extruded (calf: 72%; cow: 86%). No differences were noted in the constitutive and the neosynthetic protein profiles of cumulus cells in relation to the age of animal. Changes in neosynthetic protein patterns were observed both in cow and calf cumulus during IVM, and several proteins showed stage-specific synthesis. For the constitutive protein patterns of cow and calf oocytes, there were quantitative (38 and 40 kD) and qualitative (4, 10, 16, 17, 24, 25 and 26 kD) differences between the 2 groups. Only a few differences were observed in neosynthetic proteins between cow and calf oocytes, but there were changes in relation to nuclear status both in cow and calf oocytes. In conclusion, the difference in developmental capacity between cow and calf oocytes may be explained by a difference in the kinetics of nuclear maturation, which was significant at 20 h of culture (with 89% of cow oocytes at metaphase II and 71% of calf oocytes). At the biochemical level, our results indicate that nuclear progression during in vitro maturation of bovine oocytes is linked to changes in protein synthesis by the oocyte itself, while cumulus protein synthesis may either stimulate or modulate the process of oocyte maturation.  相似文献

In Denmark earthworms play an important role in the disappearance of cow pats on pastures. Field investigations were designed to study the influence of earthworms on the transmission of infective Cooperia oncophora larvae from experimental cow pats to grass. Results from the present experiment showed that cow pats, protected from attack by earthworms, disappeared at a much lower rate than unprotected cow pats. Development in faeces and transmission to grass of C. oncophora larvae were disturbed by the disintegration of cow pats. The rapid disintegration of unprotected cow pats resulted in an approximately 50% reduction of infective C. oncophora larvae on grass in the vicinity of these cow pats, as compared with larval contamination of grass around protected cow pats. How earthworms may cause a reduction in the transmission of infective C. oncophora larvae to grass by creating adverse conditions for the survival of parasite larvae in the cow pats, by eating and killing the parasite larvae, and by depositing parasite larvae in the soil is discussed.  相似文献

The aim of this study is to ascertain the relationship between ingestion of raw cow liver and Toxocara canis infection. A total of 150 apparently healthy adults were divided into 2 groups; 1 group consisted of 86 adults with positive results of Toxocara ELISA, and the other group of 64 adults with negative results. One researcher collected the history of ingestion of raw cow liver within 1 year and recent history of keeping dogs. Among 86 seropositive adults for T. canis, 68 (79.1%) had a recent history of ingestion of raw cow liver. Multivariate statistical analysis showed that a recent ingestion of raw cow liver and keeping dogs were related to an increased risk of toxocariasis (odds ratios, 4.4 and 3.7; and 95% confidence intervals, 1.9-10.2 and 1.2-11.6, respectively). A recent history of ingestion of raw cow liver and keeping dogs was significantly associated with toxocariasis.  相似文献

A 3-D theoretical model that characterizes the flow field around a single cow or multiple cows was developed. A general computer code that simulates the flow field that takes into account the shape of a cow and the boundary conditions of the enclosure was developed. A system of visualization of flow field that provides theoretical basis for the study of heat and mass transfer between a cow and its environment was developed. Qualitative and quantitative analyses were performed to determine the effect of cow orientation with respect to direction of airflow on convective heat exchange between the cow and its environment. The direction of air movement is not an important factor in influencing convective heat transfer between a cow and its environment.  相似文献

Births and postnatal behaviour were observed at a breeding assembly of Grey seals on North Rona. Birth is extremely rapid and the cow immediately turns to smell the pup. Smelling continues at intervals, particularly during the first 10 min after birth and allows the cow to establish the identity of her pup. She also smells the placenta and defends it against attack by gulls. Disturbance by gulls, neighbouring seals and human intrusion may lead to the cow failing to establish a bond with the pup and consequently abandoning it.  相似文献

Horse and cow dung were tested as substrates for oviposition by the stable fly Stomoxys calcitrans (L) (Diptera: Muscidae) in laboratory cages. Odour alone from either horse or cow dung was sufficient to attract flies for oviposition. This was confirmed in wind tunnel experiments, where both horse and cow dung were shown to attract gravid stable flies. However, when S. calcitrans was offered a choice between these two oviposition substrates, flies always chose horse dung over cow dung, both when allowed to contact the substrates and when relying on dung odour alone. Analyses of volatile compounds emanating from horse and cow dung by gas chromatography linked antennogram recordings from S. calcitrans antennae revealed no differences in the chemostimuli released from the two substrates. The predominant chemostimulant compounds in both substrates were carboxylic acids (butanoic acid), alcohols (oct-1-en-3-ol), aldehydes (decanal), ketones (octan-3-one), phenols (p-cresol), indoles (skatole), terpenes (beta-caryophyllene) and sulphides (dimethyl trisulphide). Higher levels (20-40 p.p.m.) of carbon dioxide were recorded over horse dung compared with cow dung, a factor that may contribute to the preference exhibited by S. calcitrans for this substrate for oviposition.  相似文献

A new dicentric Robertsonian translocation is described in a Holstein cow. The translocation appears to have arisen spontaneously from the centric fusion of autosomal acro centrics 14 and 28 which resulted in a diploid chromosome number of 59. 1. The serum lipoprotein pattern was studied in four horses, four ponies and in three high producing lactating and three non-lactating cows. The lipoprotein pattern was estimated with a combination of the preparative ultracentrifugation and the heparin-manganese precipitation technique. 2. The lipid composition of the lipoproteins of horse, pony, lactating cow and non-lactating cow was determined. 3. In all three species more than 50% of serum total lipids was found in the HDL fraction. 4. The mean chylomicron fraction in horse and pony was 3.1%. In the cow it varied from 1.5 to 2.5%. 5. Between the lactating cow and the non-lactating cow there were substantial differences in the concentration of LDL and HDL. 6. The cholesterylester concentration in VLDL, LDL and HDL was clearly higher in the lactating cow than in the non-lactating cow.  相似文献

Unannotated mammalian genome databases (dog, cow, opossum) were searched for candidate connexin genes, using sequences from annotated genomes (man, mouse). All 18 'multi-species' connexin genes, i.e., orthologs of connexin26 , 29/31.3 (duplicated in opossum), 30, 30.2/31.9, 30.3, 31, 31.1, 32, 36, 37, 39/40.1, 40, 43, 45, 44/46, 47, 50, and 57/62 , were found in dog, cow and opossum. Connexin25 and 58 have been considered specific for man, but evident orthologs of connexin25 were found in dog, cow and opossum, and orthologs of connexin58 were found in dog and cow. Moreover, a connexin43 -like sequence (approx. 80% identical to connexin43 ) was found in man, chimpanzee, dog and cow. In the three former species, the sequences were located on the X chromosome. In man, chimpanzee and cow, there were stop codons in all reading frames; these sequences are therefore judged as pseudogenes, called here Cx43pX . In the dog, the sequence contained an open reading frame for a protein of 35.7 kDa (connexin35.7). We suggest that these sequences are orthologs of connexin33 , previously considered as a rodent-specific connexin gene. Thus, connexin25 , 33 and 58 are not species-specific genes. However, the opossum may possess a candidate, connexin39.2 , without obvious orthologs in other mammals. Furthermore, pseudogenes of primate connexin31.3 and opossum connexin35 (one of the two orthologs of primate connexin31.3) were detected. These results suggest that the structure of the mammalian connexin gene family should be revised, especially with regard to the so-called 'species-specific' connexins .  相似文献

Pten作为抑癌基因,参与调控细胞生长、粘附、凋亡以及其它细胞活动.目前,国内外关于Pten在奶牛乳腺发育过程中表达及调节的研究鲜有报道.为了揭示Pten的表达与奶牛乳腺发育与泌乳之间的关系,本研究应用qRT-PCR技术检测Pten在不同泌乳时期和不同乳品质的奶牛乳腺组织中的表达差异,进而应用脂质体转染方法,通过siRNA介导的RNA干扰技术改变Pten基因在奶牛乳腺上皮细胞中的表达量,CASY法检测细胞活力,用ELISA试剂盒检测细胞分泌β-酪蛋白的含量,采用qRT-PCR、Western 印迹等技术检测Pten对奶牛乳腺上皮细胞中乳蛋白相关信号通路基因表达的影响.结果显示,泌乳期高乳品质奶牛乳腺组织中Pten表达水平显著低于泌乳期低乳品质及干乳期奶牛;Pten基因沉寂后,细胞活力提高,β-酪蛋白质量浓度增加,CSN2、AKT、MTOR、STAT5表达量增加.研究表明,Pten可通过抑制细胞活力和乳蛋白分泌而影响泌乳.  相似文献

Myostatin is a paracrine/autocrine factor that inhibits muscle growth, and mutations that affect myostatin activity or expression produce dramatic increases in muscle mass in several species. However, at present it is less clear whether differences in myostatin expression or activity exist between species with differing body sizes. Here we demonstrate that mouse muscle expresses far greater levels of myostatin mRNA than cow. In addition, activity of a 1200 bp mouse myostatin promoter construct was significantly greater than that of a 1200 bp cow myostatin promoter construct in C2C12 myotubes. In contrast, activity of reporter constructs flanked by one or both untranslated regions (UTRs) was not significantly different between the two species. Sequence analysis identified a number of promoter regions which differed between larger species (cow, pig, goat, sheep, human) and smaller (mouse, rat), including a TATA-box sequence, a CACCC box, two AT-rich regions (AT1 and AT2), and a palindromic sequence (PAL). We therefore used mutagenesis to alter the mouse sequence for each of these elements to that of the cow. Mutagenesis of the TATA, CACC, and AT1 sequences of the mouse to those of the cow significantly decreased activity of the mouse myostatin promoter compared to the wild type mouse promoter, while mutation of the AT2 and PAL sequences tended to increase promoter activity. Finally, the cow myostatin promoter was less responsive to FoxO signaling than the mouse myostatin promoter. Together these data support the hypothesis that differences in promoter activity between mouse and cow may contribute to differences in expression of the myostatin gene between these species.  相似文献

We studied the effects of dietary inclusion of freeze-dried goat and cow milk on the utilization of copper, zinc and selenium, and on the metabolic fate of copper and zinc, in rats using a standard (non-milk) control diet recommended by the American Institute of Nutrition and diets based on goat or cow milk. For animals given the goat milk diet, the apparent digestibility coefficient (ADC) of copper is similar to that obtained with the standard diet and higher than that in animals given the cow milk diet. The copper balance was higher among the rats given the goat milk and the standard diets than among those given cow milk. The ADC and retention of zinc and selenium were higher for the goat milk diet than for the other two diets. The copper content in the kidneys and in the femur was greater when the animals consumed a goat milk diet than a cow milk diet. Zn deposits in femur, testes, liver, kidney, heart and longissimus dorsi muscle were greatest with the goat-milk diet, followed by the standard diet and were lowest for the rats given cow-milk diet. This study shows that the goat-milk has an important and beneficial effect on the bioavailability of copper, zinc and selenium.  相似文献

Sheep milk, compared with cow and goat milk, had a protective effect on Gram-negative bacteria and Listeria spp. heated at 65°C in a test-tube method. This effect was not solely due to fat content as cow milk artificially reconstituted to 10% homologous fat was not as protective. Listeria monocytogenes in whole sheep, cow and goat milks at an inoculum level of 1 times 106 cfu ml-1 was heated at 68°C for 15 s in the plate pasteurizer and survival was only detected in whole sheep milk after heating. Whole sheep, cow and goat milks containing high levels of L. monocytogenes (1 times 106 cfu ml-1) could not survive the current HTST plate pasteurization protocol.  相似文献

为了解决养牛业中粪便污染问题,本研究利用亮斑扁角水虻(黑水虻)来转化利用牛粪,探讨了饲养密度对新鲜牛粪的转化效率。本试验以亮斑扁角水虻为研究对象,选择设置每20.0 kg牛粪投入3500头、8750头、17500头4日龄幼虫3个处理密度,在每个密度日均1.0 kg等量饲喂条件下,分析亮斑扁角水虻幼虫百虫重、粗蛋白、粗脂肪、牛粪转化率指标的差异,探索一种适于亮斑扁角水虻幼虫处理新鲜牛粪的饲养密度。结果表明:百虫重、粗蛋白、粗脂肪3个指标在3组处理之间都存在极显著差异。百虫重和粗脂肪两个指标和试验组密度情况在0.01的水平上显著负相关,但3个处理中转化率最高的为8750头的饲养密度。综合评价认为亮斑扁角水虻4日龄幼虫8750头/20.0 kg牛粪的投入量为实验范围内最佳饲养密度。  相似文献

Plasma hormone levels during the estrous cycle of the cow, ewe,and sow have been measured, and the patterns of secretion ofestrogens, progesterone, and luteinizing hormone during thecycle have been related to ovarian changes and other informationconcerning the cycle for each species. Peripheral plasma progesteroneand LH levels are generally inversely related during the cyclein each species, and it seems clear that progesterone exertsa negative feedback on LH secretion in all three species, atleast insofar as the cyclic release of preovulatory amountsof LH is concerned. Peak plasma progesterone levels are highestin the sow, lowest in the ewe, and intermediate in the cow.Plasma LH levels at estrus are highest in the ewe, lowest inthe sow, and intermediate in the cow. Sharp peaks in plasmaLH occur at the onset of estrus in the cow, and a few hoursafter the onset of estrus in the ewe and sow; these peaks areof about 6–8 hr duration. LH exerts a luteotrophic actionon the corpora lutea of all three species, and verylow levelsof LH secretion appear capable of maintaining the corpus luteumin the ewe and cow. There is no good evidence that prolactinis luteotrophic in any of these species. Three peaks of plasmaestrogen levels are seen in the ewe and the cow and these appearrelated to periods of accelerated follicle growth. One peakoccurs early in the cycle and before plasma progesterone levelsrise appreciably and another occurs during the luteal phasejust prior to corpus luteum regression. The third peak occursafter plasma progesterone levels decline and is associated withgrowth of the ovulating follicle. The luteal phase estrogenpeak has not been found in the sow. The rapid rise in bloodestrogens after the corpus luteum regresses facilitates thepreovulatorysurge of LH in all three species. Cyclical regressionof the corpus luteum in all three speciesappears to be underlocal control of the adjacent horn of the uterus. Exogenousestrogens are luteolytic in the cow and ewe, but luteotrophicin the sow. The ovaries of all three species contain very poorlydeveloped interstitial tissue probably because of the neailycomplete dedifftrentiation of the thecal cells during atresia.Thus, these animals lack an important source ofsteroid hormonespresent in the lodents and certain other species.  相似文献

After 80 years of the commercial application of artificial insemination (AI) in the cow, the method still has numerous benefits over natural insemination including worldwide gene improvement. The efficiency of insemination depends, among many other factors, on the delivery of an appropriate number of normal spermatozoa to the appropriate reproductive tract site at the appropriate time of estrus. The metabolic clearance of steroid hormones and pregnancy associated glycoproteins and the negative effects of different types of stress related to high milk production makes the high-producing dairy cow a good animal model for addressing factors affecting fertility. Nevertheless, extensive studies have shown a positive link between high milk production in an individual cow and high fertility. When a cow becomes pregnant, the effect of pregnancy loss on its reproductive cycle is also a topic of interest. This paper reviews the factors of a noninfectious nature that affect the fertility of lactating dairy cows following AI. Special attention is paid to factors related to the cow and its environment and to estrus confirmation at insemination. Pregnancy maintenance during the late embryonic/early fetal period is discussed as a critical step. Finally, the use of Doppler ultrasonography is described as an available research tool for improving our current understanding of the health of the genital structures and conceptus.  相似文献

Abstract: When a fistulated cow was fed an all forage diet, ruminal pH remained more or less constant (6.7 to 6.9). The ruminal pH of a concentrate-fed cow decreased dramatically in the period soon after feeding, and the pH was as low as 5.45. Mixed ruminal bacteria from the forage-fed cow converted CO2 and H2 to methane, but the ruminal fluid from the concentrate-fed cow did not produce methane. When the pH of the ruminal fluid from the concentrate-fed cow was adjusted to pH 7.0, methane was eventually detected, and the absolute rate constant of methane production was as high as the one observed with ruminal fluid from the forage fed cow (0.32 h−1). Based on the zero-time intercepts of methane production, it appeared that the concentrate-fed cow had fewer methanogens than the forage-fed cow. When the mixed ruminal bacteria were incubated in a basal medium containing 100 mM acetate, methanogenesis was pH-dependent, and no methane was detected at pH values less than 6.0. Because the removal of acetic acid completely reversed the inhibition of methanogenesis, it appeared that volatile fatty acids were causing the pH-dependent inhibition. Based on these results, concentrate diets that lower ruminal pH may provide a practical means of decreasing ruminal methane production.  相似文献

A Bayesian analysis of longitudinal mastitis records obtained in the course of lactation was undertaken. Data were 3341 test-day binary records from 329 first lactation Holstein cows scored for mastitis at 14 and 30 days of lactation and every 30 days thereafter. First, the conditional probability of a sequence for a given cow was the product of the probabilities at each test-day. The probability of infection at time t for a cow was a normal integral, with its argument being a function of "fixed" and "random" effects and of time. Models for the latent normal variable included effects of: (1) year-month of test + a five-parameter linear regression function ("fixed", within age-season of calving) + genetic value of the cow + environmental effect peculiar to all records of the same cow + residual. (2) As in (1), but with five parameter random genetic regressions for each cow. (3) A hierarchical structure, where each of three parameters of the regression function for each cow followed a mixed effects linear model. Model 1 posterior mean of heritability was 0.05. Model 2 heritabilities were: 0.27, 0.05, 0.03 and 0.07 at days 14, 60, 120 and 305, respectively. Model 3 heritabilities were 0.57, 0.16, 0.06 and 0.18 at days 14, 60, 120 and 305, respectively. Bayes factors were: 0.011 (Model 1/Model 2), 0.017 (Model 1/Model 3) and 1.535 (Model 2/Model 3). The probability of mastitis for an "average" cow, using Model 2, was: 0.06, 0.05, 0.06 and 0.07 at days 14, 60, 120 and 305, respectively. Relaxing the conditional independence assumption via an autoregressive process (Model 2) improved the results slightly.  相似文献
