This study investigates the impacts of influent ammonium concentrations on the microbial community in immobilized heterotrophic ammonium removal system. Klebsiella sp. FC61, the immobilized species, has the ability to perform simultaneous ammonium removal and Fe3+ reduction. It was found that average ammonium removal rate decreased from 0.308 to 0.157 mg/L/h, as the influent NH4 +-N was reduced from 20 to 10 mg/L. Meanwhile, at a total Fe3+ concentration of 20 mg/L, the average Fe3+ reduction removal efficiency and rate decreased from 44.61% and 0.18 mg/L/h, to 27.10% and 0.11 mg/L/h, respectively. High-throughput sequencing was used to observe microbial communities in bioreactor Samples B1, B2, and B3, after exposure to different influent NH4 +-N conditions. Results show that higher influent NH4 +-N concentrations increased microbial richness and diversity and that Klebsiella sp. FC61 play a functional role in the simultaneous removal of NH4 +-N and Fe3+ reduction in bioreactor systems.

Lipid accumulation by Gordonia sp. DG using sodium gluconate as carbon source in comparison with Rhodococcus opacus PD630 was studied. Maximum lipid content 80% was observed at the beginning of the stationary phase for R. opacus and 72% at the end of stationary phase for Gordonia sp. Different agro-industrial wastes were used as carbon source. The cells of the two organism accumulated lipid more than 50% of the biomass with most tested agro-industrial wastes. The maximum value was in presence of sugar cane molasses (93 and 96%) for R. opacus and Gordonia sp. respectively. Maximum triacyglycerols (TAGs), 88.9 and 57.8 mg/l, was obtained using carob and orange waste by R. opacus and Gordonia sp. respectively. The use of orange waste as carbon source by R. opacus, increased lipid unsaturation with C18:3 as the major unsaturated fatty acid. On the other hand, C22:0 and C6:0 were the dominant fatty acids (54.5% of the total identified fatty acids) produced by Gordonia sp. in presence of orange waste as carbon source. Statistical optimization of the medium revealed that maximum lipid content was achieved with 60% orange waste, 0.05 g/l ammonium chloride and 0.2 g/l magnesium sulphate.  相似文献

The process of leachate denitrification by populations of nitrifying and denitrifying bacteria was investigated. Leachate, derived from a local municipal landfill site, was nitrified in a continuously operating packed-bed biofilm reactor and thereafter denitrified in an activated sludge bioreactor. To follow the progress of nitrogen elimination, ammonium, nitrite and nitrate concentrations were determined at all stages of the process. While the nitrite and nitrate concentrations were measured by conventional colorimetric methods, computer controlled coulometric titration with in situ generated hypobromite was used for ammonium determination, which had previously been selectively separated from the sample matrix by gas dialysis. The detection range of the method was from 1 × 10?6 to 1 × 10?3 M ammonium (relative standard deviation (RSD) = 2%, n = 6). No interference of the complex sample matrix was found in ammonium determination. The average ammonium concentration in the leachate was 409 mg/l (standard deviation (SD) = 142 mg/l, n = 55). The ammonium concentrations decreased to 1–5 mg/l during nitrification under continuous operating conditions. Increased ammonium concentrations after nitrification correlated with a decrease in the efficiency of nitrogen elimination by up to 45% due to the build-up of high concentrations of nitrite. The concentration of sulphides, another source of pollution of the leachate, was also determined by triangle programmed coulometric titration. The average concentration of sulphides in the leachate was 221 mg/l (SD = 374 mg/l; n = 55). The sulphide concentrations decreased to concentrations below the detection limit of the coulometric titration (2 × 10?6M) during nitrification.  相似文献

This study presents the improved biodegradation of Congo red, a toxic azo dye, using mutant Bacillus sp. obtained by random mutagenesis of wild Bacillus sp. using UV and ethidium bromide. The mutants obtained were screened based on their decolorization performance and best mutants were selected for further studies. Better decolorization was observed in the initial Congo red concentration range 100–1000 mg/l for wild species whereas mutant strain was found to offer better decolorization up to 3000 mg/l. Mutant strain offered 12–30% reduction in time required for the complete decolorization by wild strain. The optimum pH and temperature were found to be 7.0 and 37 °C, respectively. Two efficient strains such as Bacillus sp. ACT 1 and Bacillus sp. ACT 2 were isolated from the various mutants obtained. Bacillus sp. ACT 2 showed improved enzymatic production and Bacillus sp. ACT 1 showed improved growth compared to wild strain. The enzyme responsible for the degradation was found to be azoreductase by SDS–PAGE and about 53% increased production of enzyme was achieved with mutant species. The experimental data were modeled using growth and substrate inhibition models.  相似文献

An Arthrobacter sp. (strain 9006), isolated from lake water, accumulated nitrite up to about 15 mg N/l, but no nitrate. In a mineral medium supplemented with tryptone, yeast extract, acetate and ammonium, the cells released nitrite into the medium parallel to growth or when growth had virtually ceased. The nitrite formed was proportional to the initial acetate concentration, indicating an involvement of acetate metabolism with nitrification. The organism grew with a wide variety of organic carbon sources, but washed cells formed nitrite from ammonium only in the presence of citrate, malate, acetate or ethanol. Magnesium ions were required for nitrification of ammonium and could not be replaced by other divalent metal ions. Analysis of the glyoxylate cycle key enzymes in washed suspensions incubated in a minimal medium revealed that isocitrate lyase and malate synthase were most active during the nitrification phase. Nitrite accumulation but not growth was inhibited by glucose, tryptone and yeast extract. Ammonium at low concentrations caused a rapid and effective inhibition of nitrate utilization in the light by the cyanobacterium Anacystis nidulans without affecting the cellular level of nitrate reductase activity. The inhibition was reversible, and the ability of the cells to utilize nitrate was restored immediately after ammonium had been exhausted. The inhibitory effect was dependent on consumption by the cells of the added ammonium which was rapidly incorporated into amino acids. In the presence of L-methionine-d,l-sulfoximine (MSX) or azaserine, inhibitors of the glutamine synthetase-glutamate synthase pathway, ammonium did not exhibit any inhibitory effect on nitrate utilization. Ammonium assimilation, rather than ammonium itself, seems to regulate nitrate utilization in A. nidulans. Heterotrophic carbon utilizing microbes were acclimatized in the laboratory by inoculating sludge collected from the waste discharge pond of a small-scale rural abattoir in India in a nutrient solution intermittently fed with glucose and ammonium chloride. Cultures of 10 well-developed isolates were selected and grown in a basal medium containing glucose and ammonium chloride. Culture supernatants were periodically analyzed for ammonium nitrogen (NH4 +-N) and chemical oxygen demand (COD). Polyphasic taxonomic study of the most active nitrifier (S18) was done. Half saturation concentration (K s), maximum rate of substrate utilization (k), yield coefficient (Y) and decay coefficient (K d) were determined from the Lineweaver–Burk plot using the modified Monod equation. S18 was able to remove 97 ± 2% of (NH4 +-N) and 88 ± 3% of COD. Molecular phylogenetic study supported by physiological and biochemical characteristics assigned S18 as Achromobacter xylosoxidans. Nitrification activity of A. xylosoxidans was demonstrated for the first time, while interestingly, the distinctive anaerobic denitrification property was preserved in S18. K s values were determined as 232.13 ± 1.5 mg/l for COD reduction and 2.131 ± 1.9 mg/l for NH4 +-N utilization. Yield coefficients obtained were 0.4423 ± 0.1134 mg of MLVSS/mg of COD and 0.2461 ± 0.0793 mg of MLVSS/mg of NH4 +-N while the decay coefficients were 0.0627 ± 0.0013 per day and 0.0514 ± 0.0008 per day, respectively. After a contact period of 24 h, 650 ± 5 mg/l solids were produced when the initial concentration of COD and NH4 +-N were 1820 ± 10 mg/l and 120 ± 5.5 mg/l, respectively. This is the first report on the kinetic coefficients for carbon oxidation and nitrification by a single bacterium isolated from slaughterhouse wastewater.  相似文献

Ammonium-induced inhibition of nitrate utilization was monitored in cell suspensions of the unicellular alga Cyanidium caldarium. It was found that the inhibition followed an exponential pattern with a t 1/2 value of about 1.5 min in cells previously grown under conditions of excess nitrate, and of about 15 min in cells grown under conditions of severe nitrate limitation. In the latter cells only, a pretreatment with cycloheximide greatly increased the t 1/2 value of inhibition. Also the resumption of nitrate utilization when ammonium was depleted followed an exponential pattern with a t 1/2 value of about 4.5 min.Our results are consistent with the hypothesis that inhibition of nitrate utilization occurs at the level of nitrate reductase activity.  相似文献

In an attempt to establish the nature of the ammonium-assimilation products which mediate the inhibition by ammonium of nitrate uptake in cyanobacteria, the effect of different amino acids on nitrate utilization by intact Anacystis nidulans cells has been assayed. To exclude an indirect inhibition of nitrate uptake through the ammonium which the amino acids might release, the cells were pretreated with l-methionine-d,l-sulfoximine (MSX), a potent inactivator of glutamine synthetase. Under these conditions, several l-amino acids, but not the corresponding d-isomers, affected nitrate utilization to a variable extent, causing inhibitions ranging between 20 and 80% when added at 20 mM concentration.For most of the inhibitory amino acids, including l-isoleucine, l-leucine and l-valine, a correlation was found between their ability to act as amino group donors to -ketoglutarate, in reactions catalyzed by A. nidulans cell-free extracts, and their inhibitory effect on nitrate utilization. l-Glutamine, l-asparagine and glycine, being effective inhibitors of nitrate utilization, were poor substrates for the transaminating activity to -ketoglutarate, however. An ureolytic ammonium-oxidizing chemolithotroph belonging to the genus Nitrosospira was shown to nitrify at pH 4.5 in a pH-stat with urea as a substrate. With ammonium as the sole substrate nitrification did not occur at pH values below 5.5. Nitrosomonas europaea ATCC 19718 and Nitrosospira briensis ATCC 25971 did not possess urease activity. The results indicate that in acid soils nitrification by ureolytic ammonium-oxidizing chemolithotrophs may not be restricted to microsites of neutral pH.  相似文献

Alcaligenes faecalis sp. No. 4, that has the ability of heterotrophic nitrification and aerobic denitrification in high-strength ammonium at about 1200 mg-N/l, converted about one-half of removed NH 4+-N to intracellular nitrogen and nitrified only 3% of the removed NH4+. From the nitrogen balance, 40–50% of removed NH4+-N was estimated to be denitrified. Production of N2 was confirmed by GC-MS and 90% of denitrified products was N2. The maximum ammonium removal rate, 29 mg-N/l h and its denitrification rate in aerated batch experiments, were 5–40 times higher than those of other bacteria with the same ability.  相似文献

In recent years, more and more attentions are put on the remediation of Cr(VI) contamination with chromate resistant bacteria. Leucobacter sp. CRB1 was a novel chromate reducing bacteria isolated from the soil of chromite ore processing residue (COPR) disposal site in Changsha, China. The objectives of this study were to evaluate the Cr(VI) tolerance of Leucobacter sp. CRB1 as well as its tolerant mechanism, and Cr(VI) reduction ability. The results showed that Leucobacter sp. CRB1 was able to tolerate 4,000 mg/l of hexavalent chromium with 34.5% reduction efficiency. At the optimum pH 9.0, the maximum concentration of chromate be reduced completely was 1,818 mg/l in growing cells and 2,100 mg/l in resting cells. Scanning electron microscopy (SEM) and energy dispersive X-ray analysis (EDAX) showed that extracellular Cr(VI) reduction of Leucobacter sp. CRB1 contributed to its high tolerance and high reduction ability. With repeating spiking, 2,490 mg/l hexavalent chromium was reduced totally within 17 h. The results suggest Leucobacter sp. CRB1 has potential application for remediation of high concentration of Cr(VI) contamination.  相似文献

Summary A pot experiment with lettuce involving three N forms each at six application levels, showed that lettuce can be grown satisfactorily with a very low nitrate content when supplied with ammonium sulphate and a nitrification inhibitor. For plants growing on nitrate N, the optimum midrib sap nitrate concentration as maturity approached was about 1400 mg/1 NO3-N. Large losses of mineral N were observed from the peat medium, even in the absence of plants. A relationship is presented which would enable a lettuce grower to estimate whole-shoot nitrate concentration from a quick test of midrib sapi.e. NO3-N (mg/kg in fresh shoot) =0.14×NO3-N (mg/l in sap). Tipburn was worst at intermediate levels of applied N, and was less serious with pure ammonium nutrition than with nitrate.  相似文献

A strain YZN-001 was isolated from swine manure effluent and was identified as Pseudomonas stutzeri. It can utilise not only nitrate and nitrite, but also ammonium. The strain had the capability to fully remove as much as 275.08 mg L−1 NO3–N and 171.40 mg L−1 NO2–N under aerobic conditions. Furthermore, At 30 °C, the utilization of ammonium is approximately 95% by 18 h with a similar level removed by 72 h and 2 weeks at 10 and 4 °C, respectively. Triplicate sets of tightly sealed serum bottles were used to test the heterotrophic nitrifying ability of P. stutzeri YZN-001. The results showing that 39% of removed NH4+–N was completely oxidised to nitrogen gas by 18 h. Indicating that the strain has heterotrophic nitrification and aerobic denitrification abilities, with the notable ability to remove ammonium at low temperatures, demonstrating a potential using the strain for future application in waste water treatment.  相似文献

The performance of two laboratory‐scale horizontal subsurface‐flow constructed wetlands(CWs) treating artificial sewage in response to varying influent components and loads was investigated. Acidification with a pH of 3.0 was detected under an organic carbon load of 100 mg/day, which further inhibited the activity of the denitrification process. With an increase in the carbon load to 240 mg/day, the pH was significantly elevated to 6.0. However, a negative effect of sulphide as a product of sulphate reduction was observed on the removal of ammonium, the plants (Juncus effusus), and the organic carbon degradation. With a produced sulphide concentration of about 3.5 mg/L, the ammonium removal decreased from 100% to 30% under an inflow load of 100 mg/day, and the number of healthy stalks of J. effuses was reduced from about 14 000/m2 to less than 6000/m2. The removal of organic carbon decreased from 94% to 68% under an influent load of 240 mg/day, when the sulphide concentrations reached up to about 8–10 mg/L. The production of sulphide was not immediately controlled by stopping the inflow sulphate load to remove the negative effect of the sulphide toxicity, thus indicating an immobilization of the deposited reduced sulphur compounds. Moreover, the effect of a nitrate dosage on the sulphide control was also examined, but was shown to be only evident under the conditions of a low organic carbon input.  相似文献

It has been shown previously that added ammonium salts cause a cessation of nitrate utilization in some Chlorella species. It has also been shown that Chlorella vulgaris can form an inactivated nitrate reductase which is an HCN complex. In the present study, a comparison has been made of the rate of nitrate utilization and the rate of nitrate reductase inactivation in Chlorella vulgaris in response to the addition of ammonium salts and light-dark changes. The rate of formation of HCN-inactivated enzyme is too slow to account for the prompt inhibition of nitrate utilization caused by adding ammonium. In contrast, when nitrate utilization is inhibited by addition of ferricyanide to intact cells, the HCN-inactivated enzyme is promptly formed in vivo, and might account for the inhibition of nitrate utilization, though inhibition of nitrate uptake can not be excluded.  相似文献

Eight bacterial isolates closely related to Diaphorobacter sp. were isolated from activated biomass surviving on wastewater laden with dyes and nitro-substituted chemicals and were identified by 16S rDNA sequence analysis. The isolates showed sequence similarity of 99–100% to other Diaphorobacter strains such as ZY 2006b, F2, NA5, PCA039, D. nitroreducens KSP4, and KSP3 and 98–99% sequence homology to D. nitroreducens NA10B (type strain JCM 11421). Neighbor-joining tree revealed that all the eight strains formed tight cluster together and also showed close clustering with other Diaphorobacter strains. Isolates demonstrated the ability to perform simultaneous nitrification and denitrification under aerobic conditions. Strains HPC 805, 815, 821, and 856 gave highest chemical oxygen demand removal (85–93%) and ammonia removal (92–96%), which correlated well with higher growth rates of the cultures. Simultaneously, complete removal of nitrate supplied in the medium in presence of ammonium and acetate (electron donor) was observed in addition to aerobic nitrite release from ammonium. Thus, the above strains showed ability to perform partial nitrification followed by further aerobic removal of common intermediate nitrite, which indicated their potential application in treatment systems for treatment of high-nitrogen-containing wastewaters.  相似文献

Heterotrophic nitrification and aerobic and anaerobic denitrification byAlcaligenes faecalis strain TUD were studied in continuous cultures under various environmental conditions. Both nitrification and denitrification activities increased with the dilution rate. At dissolved oxygen concentrations above 46% air saturation, hydroxylamine, nitrite and nitrate accumulated, indicating that both the nitrification and denitrification were less efficient. The overall nitrification activity was, however, essentially unaffected by the oxygen concentration. The nitrification rate increased with increasing ammonia concentration, but was lower in the presence of nitrate or nitrite. When present, hydroxylamine, was nitrified preferentially. Relatively low concentrations of acetate caused substrate inhibition (KI=109 M acetate). Denitrifying or assimilatory nitrate reductases were not detected, and the copper nitrite reductase, rather than cytochrome cd, was present. Thiosulphate (a potential inhibitor of heterotrophic nitrification) was oxidized byA. faecalis strain TUD, with a maximum oxygen uptake rate of 140–170nmol O2·min-1·mg prot-1. Comparison of the behaviour ofA. faecalis TUD with that of other bacteria capable of heterotrophic nitrification and aerobic denitrification established that the response of these organisms to environmental parameters is not uniform. Similarities were found in their responses to dissolved oxygen concentrations, growth rate and ammonia concentration. However, they differed in their responses to externally supplied nitrite and nitrate.  相似文献

Nitrite accumulation can be undesirable in nitrifying reactors used for the biological elimination of nitrogen from wastewaters because the ammonium oxidation process was seen to be inhibited. There is a need to better understand the effects of nitrite on both ammonium and nitrite oxidizing processes. In this paper, the effect of nitrite on the nitrifying activity of a sludge produced in steady-state nitrification was evaluated in batch cultures. At 25 mg N/l of added nitrite, nitrification was successfully carried out. Addition of higher nitrite concentrations to nitrifying cultures (100 and 200 mg N/l) provoked inhibitory effects on the nitrification respiratory process. Nitrite at 100 and 200 mg N/l induced a significant decrease in the values for nitrate yield (−20% and −34%, respectively) and specific rate of nitrate formation (−26% and −67%, respectively), while the ammonium consumption efficiency kept high and the specific rate of ammonium oxidation did not significantly change. This showed that the nitrite oxidizing process was more sensitive to the presence of nitrite than the ammonium oxidizing process. These results showed that as a consequence of nitrite accumulation in nitrification systems, the activity of the nitrite oxidizing bacteria could be more inhibited than that of the ammonium oxidizing bacteria, provoking a higher accumulation of nitrite in the medium.  相似文献

Summary The aim of this work was to investigate the effect of nitrogen starvation and subsequent fentilization with nitrate or ammonium on nitrate content and nitrate reductase activity of Rumex obtusifolius L. under natural conditions.When plants were transplanted to nitrate-poor media, endogenous nitrate was reduced within a few days. In parallel, nitrage reductase activities dropped to about 25% of the initial values. As a consequence of nitrate fertilization (1; 10 or 100 mmol KNO3/l substrate), endogenous nitrate content of the plant abruptly increased within one day. In extreme cases, nitrate concentrations of up to 10% of plant dry weight could be observed without being lethal. High external nitrate concentrations caused an inhibition of nitrate reductase within the leaves, while low external concentrations provoked an increase in the enzyme activity of about 450% within one day. Ammonium fertilization (5 mmol (NH4)2SO4/l substrate) also caused an increase in nitrate reductase activity and nitrate content within leaf blades. This observation indicates a rapid nitrification of ammonium in the substrate. When plants were fertilized with ammonium plus nitrate (2.5 mmol (NH4)2SO4+ 5 mmol KNO3/l substrate), an extremely high and long term increase in nitrate reduction could be observed. Due to an intensive enzymatic nitrate turnover, the nitrate content of leaf blades then remained relatively low. Our observations do not point to an inhibition of nitrate reductase activity in leaves of Rumex obtusifolius by ammonium. Despite temporarily high endogenous nitrate concentrations, Rumex obtusifolius may not be termed as a nitrate storage plant, since the accumulation of nitrate is a short term process only.  相似文献
