辽宁省主栽水稻品种抗稻瘟病基因的鉴定及分析   总被引:1,自引:0,他引:1  
鉴定主栽水稻品种及育种骨干亲本的抗瘟基因型,有助于了解不同抗病基因在品种中的分布,为抗病品种选育及品种布局提供参考。本研究选取辽宁省24份水稻材料,根据7个抗稻瘟病基因的保守区设计引物,扩增各品种的编码区序列,对扩增的序列进行对比分析,鉴定各基因在24个品种中的分布情况。结果表明:辽宁省24个主栽水稻品种均不携带Pi21、Pi36、Pi37或其抗病等位基因,而Pid2、Pid3、Pita和Pik/Piks/Pikm/Pikp在24个品种中以不同突变类型及不同频率出现,其中,在2个品种中检测到Pid2抗病基因及Pid3的抗病等位基因;4个品种检测到Pita的抗病等位基因,Pita的等位基因中新发现的几处碱基突变并未影响抗病基因的功能;所有品种中均无与Pik及其复等位基因完全一致的序列,但利用携带AVR-pik的稻瘟病菌接种鉴定结果表明,辽粳454和沈农265携带的Pik等位基因可能仍具有抗病基因功能。  相似文献

以褐飞虱-水稻为模式,研究虫害胁迫下植物的化学防御生理生态特征,测定了可溶性糖含量、叶绿素含量、光合速率、蒸腾速率、气孔导度以及细胞间隙二氧化碳浓度、防御酶的时间变化特征.结果表明,经虫害诱导,不同品种水稻可溶性糖含量降低;同时,光合速率以及叶绿素含量也相应下降;蒸腾速率、气孔导度和细胞间隙二氧化碳浓度的变化不显著。虫害对水稻叶片多酚氧化酶、过氧化物酶和脂氧合酶均具有诱导作用.这种诱导作用具有时间效应,且不同品种诱导作用不一致.  相似文献

Rice is an important food crop, but it is a poor source of essential micronutrients such as iron and zinc. In order to improve the metal ion content of rice grains through breeding or biotechnology, more information is needed on the molecular players that help mobilize metals from leaves to developing seeds. To profile several genes simultaneously, a cDNA macroarray was developed using 36 metal-related genes from rice, including ZIPs, NRAMPs, and YSLs (coding for known or potential metal transporters), as well as NAS, FER, FRO, NAAT, FDH, GSTU, and PDR (involved in metal homeostasis). Because flag leaves are the major source of phloem-delivered photoassimilates and remobilized metals for developing seeds, we analyzed the expression of these metal-related genes in flag and non-flag leaves of four different rice cultivars (Cocodrie, Taipei 309, IR58, and IR68144) during the period of mid-grain fill. Genes (24 of 36) exhibited low to non-detectable signals in the macroarray, while 12 genes (OsIRT1, OsZIP1, OsZIP5, OsZIP8, OsYSL5, OsYSL6, OsYSL7, OsYSL8, OsYSL18, OsNRAMP2, OsNRAMP4 and OsNRAMP7) were found to be highly expressed in both flag and non-flag leaves of all four cultivars. Additional expression analysis using semi-quantitative or quantitative PCR provided results that were generally consistent with the macroarray, but semi-quantitative PCR confirmed that OsFDH, OsFER1, OsNAAT, OsNAS1, OsPDR9, OsYSL12, OsYSL13, OsZIP7, and OsZIP10 were also expressed in leaves. This specialized macroarray has provided a short list of potential candidate genes, expressed in leaves, which might contribute to the process of metal transport to distant sinks, such as seeds.  相似文献

Probes isolated from DR and DQ beta cDNA and genomic clones were used in hybridizations to restriction enzyme-digested DNA from human homozygous typing cells (HTC) as well as other DR homozygous cells in order to estimate the number of beta genes in the DR/DQ class II region. Varying numbers of DR beta genes were found in HTC of different DR specificities, from possibly one in DR 8 cells to three in cells of DR 2 to 7. The DR beta genes of different specificities seem to be related to one another in a distinct fashion. In contrast, all HTC contain two DQ beta genes per chromosome. The restriction site polymorphism of DQ beta genes is considerably more extensive than that of DQ serology, although one of the genes seems to be nonpolymorphic. In addition to the two DP beta genes identified previously, a minimum of three to five DQ and DR beta genes exist in the human haploid genome.  相似文献

The metabolic changes of bacterial blight-resistant line C418/Xa23 generated by molecular marker-assisted selection (n= 12), transgenic variety C418-Xa21 generated by using the Agrobacterium-mediated system (n= 12), and progenitor cultivar C418 (n= 12) were monitored using gas chromatography/mass spectrometry. The validation, discrimination, and establishment of correlative relationships between metabolite signals were performed by cluster analysis, principal component analysis, and partial least squares-discriminant analysis. Significant and unintended changes were observed in 154 components in C418/Xa23 and 48 components in C418-Xa21 compared with C418 (P< 0.05, Fold change > 2.0). The most significant decreases detected (P< 0.001) in both C418/Xa23 and C418-Xa21 were in three amino acids: glycine, tyrosine, and alanine, and four identified metabolites: malic acid, ferulic acid, succinic acid, and glycerol. Linoleic acid was increased specifically in C418/Xa23 which was derived from traditional breeding. This line, possessing a distinctive metabolite profile as a positive control, shows more differences vs. the parental than the transgenic line. Only succinic acid that falls outside the boundaries of natural variability between the two non-transgenic varieties C418 and C418/Xa23 should be further investigated with respect to safety or nutritional impact.  相似文献

Plant tissue culture technology offers a solution for meeting the increasing commercial demand on economically important plants such as rice, a widespread dietary staple. However, significant genotype-specific morphogenetic responses constitute a considerable on rice regeneration in plant biotechnology contexts. Aside from genotype dependency, the components of the nutrient media including gelling agents have an important impact on regeneration efficiency. The current study explores the effect of different gelling agents on various stages of rice regeneration in two Egyptian rice cultivars-Sakha104 and Giza178. Media solidified with varying concentrations of a variety of gelling agents (agar, bacto agar, gelrite and phytagel) were tested for their impact on the frequency of callus induction, shoot regeneration and rooting. The results indicated gellan gum (gelrite and phytagel) was superior to agar products (agar and bacto agar) for callus induction. By contrast, no significant differences were found between different gelling agents for shoot regeneration. Gellan gum and media solidified with bacto agar were found to lead to significantly higher root regeneration than agar. The Sakha104 cultivar showed better responses than Giza 178 for callus induction and similar performance to the Giza 178 cultivar for root regeneration irrespective of the gelling agent. This work provides insights into the impact of different gelling agents on the morphogenetic response of two rice cultivars and can be used to help maximize the frequency of rice regeneration.  相似文献

The probability distribution and moments of the number of alleles present in a sample of homologous chromosomes are studied. It is assumed that there are multiple copies of the gene on each chromosome. When there are only two copies per chromosome or when there are only two or three chromosomes, it is possible to use analytic methods to tackle the problem. Jasmonate is an important endogenous chemical signal that plays a role in modulation of plant defense responses. To understand its mechanisms in regulation of rice resistance against the fungal pathogen Magnaporthe oryzae, comparative phenotype and proteomic analyses were undertaken using two near‐isogenic cultivars with different levels of disease resistance. Methyl‐jasmonate (MeJA) treatment significantly enhanced the resistance against M. oryzae in both cultivars but the treated resistant cultivar maintained a higher level of resistance than the same treated susceptible cultivars. Proteomic analysis revealed 26 and 16 MeJA‐modulated proteins in resistant and susceptible cultivars, respectively, and both cultivars shared a common set of 13 proteins. Cumulatively, a total of 29 unique MeJA‐influenced proteins were identified with many of them known to be associated with plant defense response and ROS accumulation. Consistent with the findings of proteomic analysis, MeJA treatment increased ROS accumulation in both cultivars with the resistant cultivar showing higher levels of ROS production and cell membrane damage than the susceptible cultivar. Taken together, our data add a new insight into the mechanisms of overall MeJA‐induced rice defense response and provide a molecular basis of using MeJA to enhance fungal disease resistance in resistant and susceptible rice cultivars.  相似文献

Two numerical taxonomic methods, cluster analysis and minimum spanning tree, were applied to classify 20 Nigerian cultivars of yellow yam (Dioscorea cayenensis). Cultivars were grown for 3 yr and 49 plant characters observed. The results were summarised as a dendrogram and an undirected minimum spanning tree. The classifications from both methods were complementary and facilitated a better understanding of the phenetic similarity between cultivars. Four main clusters were revealed with different intra-cluster variation. The clustering pattern was not remarkably affected by the two classificatory methods employed.  相似文献

Abstract

Chrysanthemum is a leading ornamental species worldwide and demonstrates a wealth of morphological variation. Knowledge about the genetic basis of its phenotypic variation for key horticultural traits can contribute to its effective management and genetic improvement. In this study, we conducted a genome-wide association study (GWAS) based on two years of phenotype data and a set of 92,617 single nucleotide polymorphisms (SNPs) using a panel of 107 diverse cut chrysanthemums to dissect the genetic control of three inflorescence-related traits. A total of 81 SNPs were significantly associated with the three inflorescence-related traits (capitulum diameter, number of ray florets and flowering time) in at least one environment, with an individual allele explaining 22.72–38.67% of the phenotypic variation. Fifteen highly favorable alleles were identified for the three target traits by computing the phenotypic effect values for the stable associations detected in 2 year-long trials at each locus. Dosage pyramiding effects of the highly favorable SNP alleles and significant linear correlations between highly favorable allele numbers and corresponding phenotypic performance were observed. Two highly favorable SNP alleles correlating to flowering time and capitulum diameter were converted to derived cleaved amplified polymorphic sequence (dCAPS) markers to facilitate future breeding. Finally, six putative candidate genes were identified that contribute to flowering time and capitulum diameter. These results serve as a foundation for analyzing the genetic mechanisms underlying important horticultural traits and provide valuable insights into molecular marker-assisted selection (MAS) in chrysanthemum breeding programs.

Analytic expressions for the expectations and variance of the number of alleles with gene frequencies in a specified class are derived in the entire population as well as in a random sample of genes drawn from the population. The correlation of this quantity with heterozygosity at the locus is also obtained. The derivations are given in details for a steady state population of finite size under the infinite allele model of selectively neutral alleles. The results are extended to include weak selection pressures and non-stationarity of the population. The relevance of the correlation of heterozygosity and the number of rare alleles in connection with the neutralist-selectionist controversy is also discussed.  相似文献

Summary Rice (Oryzae sativa L.) cultivars differ widely in their susceptibility to zinc (Zn) dificiency. Excised root apices of cv IR26 actively absorbed Zn at a rate twice that of cv M101 roots. This difference in Zn uptake rates could not be attributed to greater root surface area in cv IR26 as compared to cv M101. The maximum rates of Zn uptake (Vmax) and the Km values also differed markedly between these two cultivars. Roots of cv M101 have a two-fold greater affinity for Zn than do those of cv IR26. Leaf blade tissues of IR26 and M101 rice absorbed Zn at similar rates. Rice cv IR26 readily develops Zn deficiency symptoms in hydroponic culture but cv M101 rarely does so.  相似文献
