There is very little information on the capacity of freshwater carnivorous fish to biosynthesize highly unsaturated fatty acids (HUFA). The striped snakehead fish (Channa striata) is a carnivorous species cultured inland of several Southeast Asian countries due to its pharmaceutical properties in wound healing enhancement. We described here the full-length cDNA cloning of a striped snakehead fatty acid desaturase (fads), which is responsible for desaturation of unsaturated fatty acids in the HUFA biosynthesis. Bioinformatics analysis reveals a protein coding region with length of 445 amino acids containing all characteristic features of desaturase enzyme, including a cytochrome b5-domain with the heme-binding motif, two transmembrane domains and three histidine-rich regions. The striped snakehead fads amino acid sequence shares high similarity with known fads of other teleosts. The mRNA expression of striped snakehead fads also showed an ontogenic-related increase in expression in 0-20 days after hatch larva. Using ISH, we localized the presence of fads in larva brain, liver and intestinal tissues.  相似文献

The biochemical toxicity of arsenic trioxide (AsIII) in a freshwater edible fish Channa punctatus has been studied on exposures ranging from 7 to 90 d. The arsenic concentration increased exponentially in liver, kidney, gills, and muscles of fish up to 60 d of exposure to arsenic. However, arsenic concentration in these tissues declined at 90 d of exposure. This relationship between period of exposure and concentration of arsenic in selected tissues suggests an adaptive response of fish to arsenic. Furthermore, exposure to arsenic-induced lipid peroxidation in these organs increased initially at 7 d of exposure; however, it decreased up to 60 d of exposure but increased again at 90 d of treatment. Values of reduced glutathione (GSH) reflected the observations of lipid peroxidation. The role of GSH in this adaptive response has been discussed.  相似文献

Response of Channa punctatus to acidic water was studied by exposing fishes to pH 3.5, 4.5, 5.5 and 6.5 for 6 weeks. Growth and mortality data indicated increasing stress as the acid level in the ambient water increased. While no mortality was recorded at pH 6.5, a distinct loss of weight compared to continuous gain in body weight in control fish indicated stress. As the pH level decreased, the rate of loss in body weight increased accompanied by mortality which rose to as high as 60% within 3 weeks in fishes exposed to water at pH 3.5.
Haematological investigations confirmed the general stress indicated by growth and mortality data. Thus. RBC and related values indicated overall polycythemia. However, eosinophils, basophils. and large and small lymphocytes showed a distinct fall in number as compared to the control.
Correlated haematopoietic studies revealed that both the initial and penultimate stages in RBC and neutrophil development recorded an increase parallel to that observed in peripheral blood, but intermediate stages, probably because they were unable to keep pace with the fast turnover, showed a relative decrease.
Biochemical investigations showed an increase not only in blood glucose level but also in liver glycogen content. However, there was a significant decrease in muscle glycogen reserves.

1. The phylogeography of freshwater taxa is often integrally linked with landscape changes such as drainage re‐alignments that may present the only avenue for historical dispersal for these taxa. Classical models of gene flow do not account for landscape changes and so are of little use in predicting phylogeography in geologically young freshwater landscapes. When the history of drainage formation is unknown, phylogeographical predictions can be based on current freshwater landscape structure, proposed historical drainage geomorphology, or from phylogeographical patterns of co‐distributed taxa. 2. This study describes the population structure of a sedentary freshwater fish, the chevron snakehead (Channa striata), across two river drainages on the Indochinese Peninsula. The phylogeographical pattern recovered for C. striata was tested against seven hypotheses based on contemporary landscape structure, proposed history and phylogeographical patterns of co‐distributed taxa. 3. Consistent with the species ecology, analysis of mitochondrial and microsatellite loci revealed very high differentiation among all sampled sites. A strong signature of historical population subdivision was also revealed within the contemporary Mekong River Basin (MRB). Of the seven phylogeographical hypotheses tested, patterns of co‐distributed taxa proved to be the most adequate for describing the phylogeography of C. striata. 4. Results shed new light on SE Asian drainage evolution, indicating that the Middle MRB probably evolved via amalgamation of at least three historically independent drainage sections and in particular that the Mekong River section centred around the northern Khorat Plateau in NE Thailand was probably isolated from the greater Mekong for an extensive period of evolutionary time. In contrast, C. striata populations in the Lower MRB do not show a phylogeographical signature of evolution in historically isolated drainage lines, suggesting drainage amalgamation has been less important for river landscape formation in this region.  相似文献

The widespread use of bisphenol A (BPA) has led to its ubiquity in the natural environment. It is extensively incorporated into different industrial products and is associated with deleterious health effects on both public and wildlife. The current trial was conducted to determine the toxic potential of bisphenol A using various parameters viz haematological, biochemical, and cytological in freshwater fish Channa punctatus. For this purpose, fish were exposed to 1.81 mg/l (1/4 of LC50) and 3.81 mg/l (1/2 of LC50) of BPA along with positive (acetone) and negative controls (water) for 96 h. The blood samples were collected at 24, 48, 72, and 96 h post-exposure. Compared to the control group, fish after acute exposure to BPA showed a significant decrease in HB content, number of red blood cells, PCV values whereas a significant increase in WBCs count was recorded with an increase in the exposure period. Besides, oxidative stress (determined as malondialdehyde content) increased as BPA concentration increased. Further, the activity of different antioxidant enzymes like catalase, and superoxide dismutase decreased significantly after treatment. Results also showed significantly increased frequency of morphological alterations, nuclear changes, and increased DNA damage potential of BPA in red blood cells. Further structural analysis of erythrocytes in maximally damaged group using Scanning Electron Microscopy was performed. The study concludes that BPA exhibits genotoxic activity and oxidative stress could be one of the mechanisms leading to genetic toxicity.  相似文献

Results of experiments on fishes maintained at three different temperatures (25, 30 and 35° C), compared with a control group maintained at 14° C, are presented. Fishes showed obvious signs of stress at 30 and 35° C, as indicated by loss of weight and increase in mortality rate. On the other hand, those at 14 and 25° C gained weight steadily and showed normal growth.
Biochemical studies confirm the stress symptoms, as there was a steady, statistically significant fall in blood glucose level and depletion of glycogen reserves in liver and muscle within the first week at 35° C and by the fourth week at 30° C.
These results were further confirmed by haematological investigations. All the parameters tested indicated deviation from the normal healthy conditions seen at 14 and 25° C. The deviations in most of the parameters at 35° C were statistically significant from about the second week, and at 30° C by the fourth week. Erythrocytic polycythemia accompanied by an increase in haemoglobin content and haematocrit values were indicative of thermal stress. Leucopenia was mainly contributed by neutrophil decrease, but a relative increase in thrombocytes was recorded.
Correlated erythropoietic studies indicated an increase both in the initial (small lymphoid haemoblast) and penultimate (mature reticulocyte) stages in red cell development, although the intermediate stages decreased in number. Similar trends in the different stages of neutrophil development were observed. The significance of these changes in the context of homeostatic phenomena is discussed.  相似文献

Plasma and tissue metabolite levels were measured in the air-breathing Channa maculata during acute and prolonged exposure to normoxic and hypoxic water. Exposure of the fish to hypoxic water (water oxygen partial pressure, PwO 2= 50 mmHg) for 1 h caused increases in plasma glucose and lactate, liver and brain lactate, liver a-amino acid, heart and brain alanine and brain succinate levels. The metabolic changes in heart, brain and muscle could only be detected when Pw O2 was 30 or 10 mmHg. Heart glycogen and liver lipid decreased during acute exposure. Prolonged exposure to hypoxic water ( Pw O2= 30 mmHg) for 3 days caused an increase in plasma glycerol and liver lactate dehydrogenase activity, and a depletion of glycogen store in all tissues investigated. However, metabolite levels which had been elevated during acute hypoxic exposure were observed to return to their normoxic values after prolonged exposure. It was concluded that anaerobic metabolism was triggered by acute exposure to hypoxic water. Prolonged exposure to hypoxic water induced a metabolic readjustment involving mobilisation of lipid and glycogen stores, which is probably a reflection of the high metabolic load of aerial respiration imposed on the fish during exposure to hypoxic water.  相似文献

Genetic variability and differences in wild striped snakehead Channa striata from Malaysia were analysed by genotyping nine novel nuclear microsatellite loci. Analysis revealed moderate‐to‐high genetic diversity in most of the populations, indicative of large effective population sizes. The highly diversified populations are admixed populations and, therefore, can be recommended as potential candidates for selective breeding and conservation since they each contain most of the alleles found in their particular region. Three homogenous groups of the wild populations were identified, apparently separated by effective barriers, in accordance with contemporary drainage patterns. The highest population pairwise FST found between members of the same group reflects the ancient population connectivity; yet prolonged geographical isolation resulted in adaptation of alleles to local contemporary environmental change. A significant relationship between genetic distance and geographical isolation was observed (r = 0·644, P < 0·01). Anthropogenic perturbations indicated apparent genetic proximity between distant populations.  相似文献

Seven polymorphic microsatellite loci were isolated and characterized for the snakehead murrel, Channa striata (Channidae), a valuable tropical freshwater fish species. Among 25 specimens collected from Kedah state in Malaysia, the number of alleles per locus ranged from 2 to 7. Observed and expected heterozygosities ranged from 0.120 to 0.880 and 0.117 to 0.698, respectively. A single locus (CS1-C07) was significantly deviated from Hardy-Weinberg equilibrium after Bonferroni correction. These novel markers would be useful for population genetic studies of the C. striata.  相似文献

Channa gachua were monthly sampled throughout a year and the histological analysis of their ovaries was done to determine the changes occurring in ovarian development. Based on histological examination of the ovaries, the oogenic process of C. gachua undergoes distinct cyclic and seasonal morphological changes. Five different developmental stages were identified under three major categories: pre-spawning (immature, maturing, mature), spawning (ripe-running) and post-spawning (spent). The peak spawning period of C. gachua was noticed during December - February. The gonadosomatic index (GSI) and ova diameter ranged from 0.79 to 3.61% and 543–1123 μm respectively. The highest mean GSI (3.61 ± 0.16) and oocyte diameter (1123 ± 55 μm) were observed in December indicating that during this month the gonadal development reached maturity.  相似文献

Abstract

Dimethoate is one of the organophosphate insecticides widely used in agriculture throughout the world and is an inhibitor of cholinesterase in animals. The objective of the present study was to detect oxidative stress and DNA damage induced by dimethoate in the freshwater fish Channa punctatus (C. punctatus). The LC50-96 h value of technical grade dimethoate was estimated at 19.10 μg L-1 in a semi-static system and, on the basis of the LC50 value, three concentrations were determined. The fish were exposed to these concentrations of dimethoate for 96 h and samplings were done at 24 and 96 h for assessment of oxidative stress and DNA damage. After exposure to dimethoate, the level of superoxide dismutase declined while lipid peroxidation, glutathione, induction of micronucleus and DNA damage were increased in C. punctatus as the concentration and exposure time increased. Thus our results suggest that dimethoate induces genotoxic effects which invariably accompanied and correlated with oxidative stress.  相似文献

Resting metabolism was measured in immature mandarin fish Siniperca chuatsi weighing 42.1-510.2 g and Chinese snakehead Channa argus weighing 41.5-510.3 g at 10, 15, 20, 25, 30 and 35 degrees C. Heat increment of feeding was measured in mandarin fish weighing 202.0 (+/-14.0) g and snakehead weighing 200.8 (+/-19.3) g fed swamp loach Misgurnus anguillicaudatus at 1% body weight per day at 28 degrees C. In both species, weight exponent in the power relationship between resting metabolism and body weight was not affected by temperature. The relationship between resting metabolism and temperature could be described by a power function. The temperature exponent was 1.39 in mandarin fish and 2.10 in snakehead (P<0.05), indicating that resting metabolism in snakehead increased with temperature at a faster rate than in mandarin fish. Multiple regression models were used to describe the effects of body weight (W, g) and temperature (T, degrees C) on the resting metabolism (R(s), mg O(2)/h): lnR(s)=-5.343+0.772 lnW+1.387 lnT for the mandarin fish and lnR(s)=-7.863+0.801 lnW+2.104 lnT for the Chinese snakehead. The proportion of food energy channelled to heat increment was 8.7% in mandarin fish and 6.8% in snakehead.  相似文献

In five species of fish from the Family Sciaenidae, collected from marine, brackish and fresh-water environments, the following parameters were studied: haemoglobin concentration, haematocrit, number of red blood cells, mean corpuscular volume, mean corpuscular haemoglobin, mean corpuscular haemoglobin concentration, blood pH, oxygen affinity of blood and stripped haemolysate, Root effect, the number of haemoglobins separated by polyacrilamide electrophoresis, iron concentration and osmotic pressure of the serum. Plagioscion squamosissimus , the only freshwater species studied, clearly separated from the other four species; it exhibited the highest haemoglobin number, haemotocrit, number erythrocytes, of oxygen affinity of the haemolysate, and the lowest oxygen affinity of the blood, iron concentration, pH and osmotic pressure.  相似文献

Snakehead Channa striata is an important freshwater food fish in many Southeast Asian countries. Three monoclonal antibodies (C9, C10 and D10) were developed against purified serum immunoglobulins of Channa striata (Cs-Ig) and characterized. C9 and D10 MAbs were specific to heavy chain, while C10 MAb detected only unreduced Cs-Ig in western blotting. In competitive ELISA, C9 and C10 MAbs were specific to C. striata Ig and showed no cross reactivity with serum Ig of other fish species i.e. Channa punctatus, Channa marulius, Clarias batrachus and Labeo rohita. D10 MAb showed reactivity to serum Ig of C. striata and C. marulius. In FACS analysis of gated lymphocytes, the percentage of Ig+ cells detected by C9 MAb was 18.2%, 27.7% and 10.3% in blood, spleen and kidney, respectively (n=3, body weight 500-600 g). However, only a few cells (0.5%) were found to be Ig+ in thymus (n=5). C9 MAb was also successfully employed to demonstrate Ig+ cells in blood smears and formalin fixed sections of spleen and kidney. These findings suggest that the spleen plays an important role in humoral immunity as compared to head kidney. Further, these MAbs can be useful immunological tool in monitoring health status of cultured C. striata.  相似文献

The control of the cardiovascular system with particular emphasis on the regulation of blood distribution in the gills and air-breathing organ was studied in the air-breathing teleost Channa argus. Perfused head preparations were used in addition to experiments with isolated strip preparations of arteries and heart chambers. The distribution of adrenergic nerves was investigated using Falck-Hillarp fluorescence histochemistry. This preliminary study shows an adrenergic control system composed of chromaffin cells and adrenergic nerves similar to that found in other teleosts investigated, although the systemic arteries (coeliac artery, dorsal aorta and the vasculature of the air-breathing organ) appear to lack an adrenergic innervation. The reactions of isolated artery strip preparations to acetylcholine and adrenaline resemble those seen in other teleosts, and there is a prominent inhibitory effect of L-isoprenaline suggestive of arterial beta-adrenoceptors. The general vascular resistance of the gill apparatus-air-breathing organ increases in response to acetylcholine or adrenaline, and there is a redistribution of perfusion flow from the air-breathing organ circuit (anterior venous outflow from the first and second pair of gills and the air-breathing organ) to the general systemic circuit (dorsal aortic outflow from the third and fourth pair of gills). Stimulation of the vagal branch entering the air-breathing organ mimics the effects of acetylcholine or adrenaline. This innervation is probably non-adrenergic since no adrenergic nerve fibres could be demonstrated in the vasculature of the air-breathing organ using the histochemical technique. An adrenergic control of the vasculature of the air-breathing organ is not likely, since the concentration of adrenaline needed to affect the vasculature is not reached in the plasma even during "stress".  相似文献

The present study was undertaken with the objective to investigate the intraspecific variation of Channa punctatus on the basis of morphometric characters using the truss network system that was constructed from the fish body. Fish samples were collected from the Narora (n = 62) and Kanpur sites (n = 46) of the Ganga River, the Firozabad site in the Yamuna River (n = 66) and at the Lucknow site of the Gomti River (n = 60). Data were subjected to principal component analysis, discriminant function analysis and univariate analysis of variance. The first principal component (PC I) analysis explained 47.75% of total variation while PC II and PC III explained 8.18 and 7.48%, respectively. The step‐wise discriminant function analysis retained seven variables that significantly discriminated the populations. Using these variables 81.85% of the original groups were classified into their correct samples. Misclassification was higher for the samples from Ganga River (19.6% at the Kanpur site and 19.4% for the Narora site). Of the total 27 transformed morphometric measurements, 23 exhibited significant differences among the populations. These findings indicate the presence of different stocks of fish from the three rivers.  相似文献

Over the years, indiscriminate usage of pesticides has resulted in situations which are not conducive for a good environment. In aquatic toxicology, fishes have been developed as established models for studying toxic responses of xenobiotics including pesticides. Pendimethalin (PD), an herbicide, is widely present in the aquatic environment, but little is known regarding its potential neurotoxicity in fish. The present study was conducted on Channa punctata Bloch exposed to sub-lethal doses of PD (0.5 and 0.8 ppb) for 96 h. The exposure resulted in alterations in epinephrine levels in the fish brain. Epinephrine levels decreased significantly in a dose dependent manner with increase in the PD exposure. A marked decrease in the activity of acetylcholinesterase along with reduction in Na+-K+-ATPase and monoamine oxidase activity was also observed. In comparison with the corresponding controls, the sub-lethal doses of PD also caused significant changes in the oxidative stress markers (lipid peroxidation and carbonyl derivatives of protein oxidative destruction levels) and antioxidant defenses (reduced glutathione levels, catalase, glutathione-s-transferase activity) in brain tissue. Our results reflect that a detailed investigation is warranted regarding the toxicity potential of PD.  相似文献
