The nature of the lack of oxygen inhibition of C3-photosynthesisat low temperature was investigated in white clover (Trifoliumrepens L.). Detached leaves were brought to steady-state photosynthesisin air (34 Pa p(CO2), 21 kPa p(O2), balance N2) at temperaturesof 20°C and 8°C, respectively. Net photosynthesis, ribulose1,5-bisphosphate (RuBP) and ATP contents, and ribulose 1,5-bisphosphatecarboxylase/oxygenase (RuBPCO) activities were followed beforeand after changing to 2·0 kPa p(O2). At 20°C, lowering p(O2) increased net photosynthesis by37%. This increase corresponded closely with the increase expectedfrom the effect on the kinetic properties of RuBPCO. Conversely,at 8°C net photosynthesis rapidly decreased following adecrease in p(O2) and then increased again reaching a steady-statelevel which was only 7% higher than at 21 kPa p(O2). The steady-staterates of RuBP and associated ATP consumption were both estimatedto have decreased. ATP and RuBP contents decreased by 18% and33% respectively, immediately after the change in p(O2) suggestingthat RuBP regeneration was reduced at low p(O2) due to reducedphotophosphorylation. Subsequently, RuBP content increased again.Steady-state RuBP content at 2·0 kPa p(O2) was 24% higherthan at 21 kPa p(O2). RuBPCO activity decreased by 22%, indicatingcontrol of steady-state RuBP consumption by RuBPCO activity. It is suggested that lack of oxygen inhibition of photosynthesisat low temperature is due to decreased photophosphorylationat low temperature and low p(O2). This may be due to assimilateaccumulation within the chloroplasts. Decreased photophosphorylationseems to decrease RuBP synthesis and RuBPCO activity, possiblydue to an acidification of the chloroplast stroma. The Km(CO2) ancl Vmax of ribulose 1,5-bisphosphate (RuBP) carboxylaseand its protein ratio to total soluble protein from Oryza speciesincluding cultivars (25 varieties) and wild types (11 species,21 strains) were surveyed. Their variabilities among cultivarsof O. sativa were very small. The averages of the Km(CO2) andVmax values and the ratio of carboxylase to soluble protein,and their standard errors were 10.2?1.0µM, 1.72?0.13units.mg–1(pH 8.0 and 25?C) and 52?2%, respectively. However, some differencesseemed to exist based on genome constitution in the Oryza genus.RuBP carboxylases from the species with the AgAg genome, O.graberrima and O. breviligulate, exhibited low Km(CO2) values(8.0?0.8 µM). High Vmax was associated with the CC genome,O. eichingeri and O. officinalis (2.08?0.15 units.mg–1).A higher ratio of RuBP carboxylase protein to soluble proteinwas found for the AA genome, O. sativa and O. perennis. Bot. 38: 1937–1948. Associations between photosynthesis, the activation state ofRuBP carboxylase and the contents of photosynthetic intermediateswere compared in soybean and wheat leaves before and after exposureto photoinhibitory treatments in the presence of atmosphericO2. Exposing attached leaves to a supra-saturating irradiance(3 800 µmol quanta m– 2 s–1) for 2 h in CO2-freeair decreased carboxylation efficiency and the light-saturatedphotosynthetic rate in air by approximately 50%. Exposure tothe photoinhibitory treatment for periods in excess of 2 h didnot cause a further decrease of photosynthesis in soybean. Althoughphotosynthesis was reduced, the initial and total (fully-activated)activities of ribulose 1,5-bisphosphate carboxylase (RuBPCase)in leaf extracts were unaltered in each species by the photoinhibitorytreatment. This was true for leaves sampled under both air andat a rate-limiting intercellular CO2 partial pressure (Ci) of75 µPa Pa–1. The contents of ribulose l,5-bisphosphate(RuBP) and 3-phosphoglyceric acid (3-PGA) were reduced by thephotoinhibitory treatment in soybean leaves sampled in air andat a rate-limiting Ci, although the RuBP/3-PGA ratio was unaffected.The relative reduction of RuBP content in soybean leaves atrate-limiting C1 was similar to the corresponding reductionof carboxylation efficiency. For wheat,the relative reductionof RuBP content at rate-limiting Ci (–19%) caused by thephotoinhibitory treatment was considerably less than the correspondingdecrease of carboxylation efficiency (–49%).The RuBP/3-PGAratio of wheat was also increased significantly by the photoinhibitorytreatment The significance of these observations to the regulationof CO2-limited photosynthesis in leaves experiencing photoinhibitionunder atmospheric oxygen is discussed. Consideration is alsogiven to the previous contention that contemporary measurementsof initial activity in crude extracts may provide a spuriousindication of the amount of the enzyme-CO2-Mg2 + form of RuBPcarboxylase present in the leaf. Changes in the rates of gas exchange and the amount of ribulose1,5-bisphosphate (RuBP) carboxylase protein were determinedin the 12th leaf blades of rice during the reproductive stages.RuBP carboxylase exhibited a large change similar to that inthe assimilation rate at 2% O2 throughout the leaf's life, butits decrease during senescence was barely faster than the decreasein the assimilation rate. Consequently, the overall relationshipwas slightly curvilinear. By contrast, leaf conductance decreasedmore slowly than the assimilation rate which resulted in theintercellular CO2 concentration increasing during senescence. In order to determine the maximum activity of RuBP carboxylaseat the intercellular CO2 concentration, the kinetic parametersand their pH response were determined using purified, and completelyactivated, rice RuBP carboxylase. The maximum carboxylase activityat the intercellular CO2 concentration was linearly correlatedwith the assimilation rate at 2% O2 (r=0.989), and was veryclose to that needed to account for the assimilation rate. We conclude that changes in both the amount of RuBP carboxylaseprotein and leaf conductance reflect the change in photosynthesisduring the life span of the leaf. Changes in the levels of 14C-labelled metabolites were monitoredin Chlorella pyrenoidosa cells during a transition from highto low irradiance, i.e., from 700 to 430 µmol quanta (400–700nm) m–2 s–1. Chlorella cells assimilated 14CO2 photosynthetically(steady-state 14C-labelling) for 12 min at the high irradianceand then 10 min at the low irradiance. With the transition tolow light, the level of 14C-labelled ribulose 1,5- bisphosphate(RuBP) did not decrease, even though the rate of total 14C-incorporationdecreased by 80%. The data suggest that RuBP carboxylase deactivatesrapidly (within 1 or 2 min) on exposure to low light, causingRuBP pool sizes to be maintained (or even increased) in spiteof a decreased rate of RuBP regeneration. There was also evidenceof light modulation of other enzymes, including some enzymesinvolved in sucrose synthesis. Photosynthetic and photorespiratoryenzymes in widely divergent plant species with special referenceto the moss Ceratodon purpureus: Properties of ribulose bisphosphatecarboxylase/oxygenase, phosphoenolpyruvate carboxylase and glycolateoxidase.—J. exp. Bot. 36: 1677–1684. Km(CO2) values and maximal velocities of ribulose bisphosphatecarboxylase/oxygenase (E.C. 4.1.1.39 [EC] ) were determined for sixplant species growing in the wild, consisting of a moss, a fernand four angiosperms. The maximum velocities of the RuBP carboxylasesvaried from 0.13 to 0.;62 µmol CO2 fixed min–1 mg–1soluble protein and the Km(CO2) values from 15 to 22 mmol m–3CO2. The highest Km(CO2) values found were for the moss, Ceratodonpurpureus, and the grass, Deschampsia flexuosa. These plantsalso had the highest ratios of the activities of RuBP carboxylaseto RuBP oxygenase. Glycolate oxidase (E.C. 1.1.3.1 [EC] ) activitieswere slightly lower in D.flexuosa, but not in C. purpureus,than for typical C3 species. Phosphoenolpyruvate carboxylase(E.C. 4.1.1.31 [EC] ) was not involved in the photosynthetic carboxylationby these two plants. However, another grass, Phragmites australis,was intermediate in PEP carboxylase activity between C3 andC4 plants The properties of RuBP carboxylase/oxygenase are discussedin relation to the activities of PEP carboxylase and glycolateoxidase and to the internal CO2 concentration. The reaction of spinach RuBisCO activated with CO2 and Mg2+proceeded in two phases, an initial burst for a few minutesand the subsequent linear phase, in the presence of saturatingconcentrations of CO2, ribulose 1,5-bisphosphate (RuBP), andMg2+. The percentage of the activity in the linear phase tothat in the initial burst was 55% with RuBisCO prepared withpolyethylene glycol, and very close to the value with the enzymereleased immediately from isolated chloro-plasts. RuBisCO preparedwith ammonium sulfate had a much larger decrease of the activityin the linear phase. The Euglena enzyme had a linear courseof reaction with time for up to 20 minutes. The Km for CO2 of spinach RuBisCO activated beforehand was 20µM in the initial burst, and 28 µM in the linearphase. In the carboxylase reaction initiated with inactive enzyme,the activity was initially negligible, but in 5 minutes increasedto the level observed in the linear phase of the activated enzyme.The Km for CO2 in the linear phase of the pre-inactivated enzymewas 70 µM. The concentration of RuBP was the immediate cause of the two-phasiccourse of the carboxylase reaction of spinach RuBisCO. The curvatureof the time course was not observed below 35 µM RuBP.The enzyme required over 88 µM RuBP for the conventionaltwo-phasic course. Further increase of the concentration ofRuBP increased the extent of the curvature, but did not startthe curvature sooner after the start of the reaction. Even ifspinach RuBisCO was in the linear phase, dilution of RuBP orits consumption by the enzymatic reaction to less than 30 µMcaused the enzyme to show the resumed biphasic reaction courseafter addition of a high concentration of RuBP. 1This paper is the twenty-fourth in a series on PhotosyntheticCarbon Metabolism in Euglena gracilis. Chloroplast envelopes were isolated from chloroplasts purifiedfrom Spinacea oleracea L. (C3), Panicum miliaceum L. (NAD-malicenzyme-type C1), Digitaria sanguinalis (L.) Scop. (NADP-malicenzyme-type C4), Kalanchoe daigremontiana Hamet et Perrier (constitutiveCAM), and from Mesembryanthemum crystallinum L. (inducible CAM)performing either C3 photosynthesis or Crassulacean acid metabolism(CAM). For each species, methods were developed to isolate chloroplastenvelopes free of thylakoid contamination. The polypeptidesof ribulose bisphosphate (RuBP) carboxylase which has been consistentlyreported in envelope preparations of spinach were not foundin envelope preparations of C4 mesophyll chloroplasts. Silverstaining of envelope polypeptides resolved electrophoreticallyon sodium dodecylsulfate polyacrylamide gradient slab gels produceda more complex profile than did Coomassie staining which haspreviously been used with C3 envelope preparations, even thoughsilver reacted poorly with polypeptides corresponding to thesubunits of RuBP carboxylase. All of the plants examined possesseda major polypeptide of 27 to 29 kilodaltons (kD) which was previouslysuggested to be the phosphate translocator in spinach. WithC3 M. crystallinum, the 29 kD polypeptide stained most intensely.After induction of CAM, a 32 kD polypeptide also stained intensely,giving a profile similar to that obtained with the constitutiveCAM species. A 32 kD polypeptide was also prominent in C4 envelopepreparations, suggesting that a 32 kD polypeptide may be a translocatorprotein which is required in Crassulacean acid metabolism andC4 photosynthesis, but not in C3 photosynthesis. RuBP carboxylase-oxygenase protein in three C3 species (Nicotianatabacum L., Solanum tuberosum L., Triticum aestivum L.) andthree C4 species (Panicum miliaceum L., Panicum texanum Buckl.,Zea mays L.) was quantitatively determined by sucrose densitygradient centrifugation and by immunochemical assay using antibodyraised to crystallized tobacco leaf RuBP carboxylase-oxygenase.The C3 species had 3- to 6-fold higher concentrations of RuBPcarboxylase-oxygenase than the C4 species when expressed oneither a chlorophyll or a leaf area basis. The C3 species alsoallocated a higher fraction of their total soluble protein tothis enzyme (from 25 to 60% for the C3 species compared to 8to 23% for the C4 species). There was no RuBP carboxylase-oxygenaseprotein or activity in the C4 mesophyll cells, while the enzymeconstituted from 20 to 40% of the total soluble protein in theC4 bundle sheath cells. A close correlation (r = +0·91)was found between catalytic activity and level of the enzymeprotein in the species examined.  相似文献

Decreasing relative water content (RWC) of leaves progressivelydecreases stomatal conductance (gs), slowing CO2 assimilation(A) which eventually stops, after which CO2 is evolved. In somestudies, photosynthetic potential (Apot), measured under saturatingCO2, is unaffected by a small loss of RWC but becomes progressivelymore inhibited, and less stimulated by elevated CO2, below athreshold RWC (Type 1 response). In other studies, Apot andthe stimulation of A by elevated CO2 decreases progressivelyas RWC falls (Type 2 response). Decreased Apot is caused byimpaired metabolism. Consequently, as RWC declines, the relativelimitation of A by gs decreases, and metabolic limitation increases.Causes of decreased Apot are considered. Limitation of ribulosebisphosphate (RuBP) synthesis is the likely cause of decreasedApot at low RWC, not inhibition or loss of photosynthetic carbonreduction cycle enzymes, including RuBP carboxylase/oxygenase(Rubisco). Limitation of RuBP synthesis is probably caused byinhibition of ATP synthesis, due to progressive inactivationor loss of Coupling Factor resulting from increasing ionic (Mg2+)concentration, not to reduced capacity for electron or protontransport, or inadequate trans-thylakoid proton gradient (pH).Inhibition of Apot by accumulation of assimilates or inadequateinorganic phosphate is not considered significant. DecreasedATP content and imbalance with reductant status affect cellmetabolism substantially: possible consequences are discussedwith reference to accumulation of amino acids and alterationsin protein complement under water stress.  相似文献

A spectrophotometric assay has been used to measure the activityof PEP carboxylase and RuBP carboxylase in the epidermal andmesophyll tissue of Commelina communis. On both a chlorophylland protein basis the PEP carboxylase activity was always greaterin the epidermis than in the mesophyll, whereas RuBP carboxylaseactivity was always highest in the mesophyll. PEP carboxylaseactivity in epidermal extracts was lost very slowly and itspH optimum was a broad one in the range 7·5–8·0.The Km values for PEP carboxylase in the epidermis and mesophyllobtained from light- and dark-treated plants were not very differentalthough its Vmax was much lower in dark-treated tissue. Thesedata are discussed in relation to the possible role of PEP carboxylasein guard cell metabolism.  相似文献

Photosynthetic rates of outdoor-grown soybean (Glycine max L.Merr. cv. Bragg) canopies increased with increasing CO2 concentrationduring growth, before and after canopy closure (complete lightinterception), when measured over a wide range of solar irradiancevalues. Total canopy leaf area was greater as the CO2 concentrationduring growth was increased from 160 to 990 mm3 dm–3.Photosynthetic rates of canopies grown at 330 and 660 mm3 CO2dm–3 were similar when measured at the same CO2 concentrationsand high irradiance. There was no difference in ribulose bisphosphatecarboxylase/oxygenase (rubisco) activity or ribulose 1,5-bisphosphate(RuBP) concentration between plants grown at the two CO2 concentrations.However, photosynthetic rates averaged 87% greater for the canopiesgrown and measured at 660 mm3 CO2 dm–3. A 10°C differencein air temperature during growth resulted in only a 4°Cleaf temperature difference, which was insufficient to changethe photosynthetic rate or rubisco activity in canopies grownand measured at either 330 or 660 mm3 CO2 dm–3. RuBP concentrationsdecreased as air temperature during growth was increased atboth CO2 concentrations. These data indicate that the increasedphotosynthetic rates of soybean canopies at elevated CO2 aredue to several factors, including: more rapid development ofthe leaf area index; a reduction in substrate CO2 limitation;and no downward acclimation in photosynthetic capacity, as occurin some other species. This study was done to explore an enzymatic mechanism for thephotosynthetic carbon reduction cycle whereby the rate of synthesisof ribulose 1,5-bisphosphate (RuBP) could be changed while thelevels of intermediates other than 3-phosphoglycerate and RuBPwere kept constant. Chloroplast aldolase was purified to homogeneityfrom spinach leaves. When the enzyme was assayed in the directionof fructose 1,6-bisphosphate synthesis in the presence of theconcentrations of the substrates reported in vivo, the activitywas severely inhibited by physiological concentrations of RuBP.The aldolase reaction proceeded with a sequential mechanism.The Km for dihydroxyacetone phosphate and D-glyceraldehyde 3-phosphatewere 0.45 mM and 40 µM, respectively. The activity wascompetitively inhibited by RuBP with respect to dihydroxyacetonephosphate. The KI was 0.78 mM. The maximum activity of aldolasein spinach leaves was calculated as 1,360µmol (mg Chl)–1h–1 An equation to express the reaction for the synthesisof fructose 1,6-bisphosphate by aldolase was constructed topredict the metabolic rate of this reaction in vivo. The intraseminal embryo of Cyamopsis tetragonaloba is greenfrom the globular stage onwards. The chloroembryos do not photosynthesizein vivo but are capable of in vitro photosynthesis at a ratewhich is similar to that of leaf tissue and they utilize bicarbonateions in this process. Both ribulose bisphosphate (RuBP) andphospho-enol-pyruvate (PEP) carboxylases are functional in CO2fixation by the embryos. The influence of salinity in the growing media on ribulose-1,5-bisphosphate (RuBP) carboxylase and on CO2 fixation by intactsugar beet (Beta vulgaris) leaves was investigated. RuBP carboxylase activity was mostly stimulated in young leavesafter exposure of plants for 1 week to 180 mM NaCl in the nutrientsolution. This stimulation was more effective at the higherNaHCO2 concentrations in the reaction medium. Salinity also enhanced CO2 fixation in intact leaves mostlyat rate-limiting light intensities. A 60 per cent stimulationin CO2 fixation rate was obtained by salinity under 450 µEm–2 s–1. At quantum flux densities of 150 µEm–2 s–1 (400–700 nm) this stimulation was280 per cent. Under high light intensities no stimulation bysalinity was found. In contrast, water stress achieved by directleaf desiccation or by polyethylene glycol inhibited enzymeactivity up to fourfold at –1.2 MPa. Chlorella vulgaris 11h cells grown in air enriched with 4% CO2(high-CO2 cells) had carbonic anhydrase (CA) activity whichwas 20 to 90 times lower than that of algal cells grown in ordinaryair (containing 0.04% CO2, low-CO2 cells). The CO2 concentrationduring growth did not affect either ribulose 1,5-bisphosphate(RuBP) carboxylase activity or its Km for CO2. When high-CO2 cells were transferred to low CO2 conditions,CA activity increased without a lag period, and this increasewas accompanied by an increase in the rate of photosynthetic14CO2 fixation under 14CO2-limiting conditions. On the otherhand, CA activity as well as the rate of photosynthetic 14CO2fixation at low 14CO2 concentrations decreased when low-CO2cells were transferred to high CO2 conditions. Diamox, an inhibitor of CA, at 0.1 mM did not affect photosynthesisof low-CO2 cells at high CO2 concentration (0.5%). Diamox inhibitedphotosynthesis only under low CO2 concentrations, and the lowerthe CO2 concentration, the greater was the inhibition. Consequently,the CO2 concentration at which the rate of photosynthesis attainedone-half its maximum rate (Km) greatly increased in the presenceof this inhibitor. When CO2 concentration was higher than 1%, the photosyntheticrate in low-CO2 cells decreased, while that in high-CO2 cellsincreased. Fractionation of the low-CO2 cells in non-aqueous medium bydensity showed that CA was fractionated in a manner similarto the distribution of chlorophyll and RuBP carboxylase. These observations indicate that CA enhances photosynthesisunder CO2-limiting conditions, but inhibits it at CO2 concentrationshigher than a certain level. Cytochrome aa3 was found to occur in Halobacterium halobiumat an early exponential phase of growth. Some of its spectralproperties were determined with a solubilized membrane fractionof the bacterium. Cytochrome c oxidizing activity of the cytochromeaa3 was not dependent on salt concentration, but decreased steeplywith increasing buffer concentration of the reaction mixture,just as the activity of cytochrome aa3 from non-halophilic bacteria. We have previously shown that inorganic orthophosphate (Pi)uptake by Catharanthus roseus cells proceeds by a proton/Picotransport mechanism [Sakano (1990) Plant Physiol. 93: 479]that acidifies the cytoplasm [Sakano et al. (1992) Plant Physiol.99: 672]. In the present study, we analyzed changes in the contentof endogenous organic acids, carbon dioxide evolution, and oxygenconsumption upon Pi application. The results are consistentwith the operation of the biochemical pH-stat mechanism [Davies(1986) Physiol. Plant. 67: 702] during and after Pi uptake. The effect of photosynthetic photon flux density (PPFD) on carboxylationefficiency, estimated as the initial slope (IS) of net CO2 assimilationrate versus intercellular CO2 partial pressure response curve,as well as on ribulose-1, 5-bisphosphate carboxylase (Rubisco)activation was measured in Trifolium subterraneum L. leavesunder field conditions. The relationship between IS and PPFDfits a logarithmic curve. Rubisco activation accounts for theIS increase only up to a PPFD of 550 µmol photons m-2s-1. Further IS increase, between 550 and 1000 µmol photonsm-2 s-1, could be related to a higher ribulose fcwphosphate(RuBP) availability. The slow, but sustained IS increase above1000 µmol photons m-2 s-1 could be explained by the mesophyllCO2 diffusion barriers associated with the high chlorophylland protein content in field developed leaves.
