Elevated levels of both ozone and UV-B radiation are typical for high-altitude sites. Few studies have investigated their possible interaction on plants. This study reports interactive effects of O3 and UV-B radiation in four-year-old Norway spruce and Scots pine trees. The trees were cultivated in controlled environmental facilities under simulated climatic conditions recorded on Mt Wank, an Alpine mountain in Bavaria, and were exposed for one growing season to simulated ambient or twice-ambient ozone regimes at either near ambient or near zero UV-B radiation levels. Chlorotic mottling and yellowing of current year needles became obvious under twice-ambient O3 in both species at the onset of a high ozone episode in July. Development of chlorotic mottling in relation to accumulated ozone concentrations over a threshold of 40 nL L–1 was more pronounced with near zero rather than ambient UV-B radiation levels. In Norway spruce, photosynthetic parameters at ambient CO2 concentration, measured at the end of the experiment, were reduced in trees cultivated under twice-ambient O3, irrespective of the UV-B treatment. Effects on photosynthetic capacity and carboxylation efficiency were restricted to trees exposed to near zero levels of UV-B radiation, and twice-ambient O3. The data indicate that UV-B radiation, applied together with O3, ameliorates the detrimental effects of O3. The data also demonstrate that foliar symptoms develop more rapidly in Scots pine than in Norway spruce at higher accumulated ozone concentrations. Formation of pinosylvin (PS) and pinosylvin 3-O-monomethyl ether (PSM), as well as the activities of stilbene synthase (STS) and S-adenosyl-l-methionine (SAM):pinosylvin O-methyltransferase (PMT), were induced strongly in needles of Scots pine seedlings upon ozone treatment, as well as in cell suspension cultures of Scots pine upon fungal elicitation. A SAM-dependent PMT protein was purified and partially characterised. A cDNA encoding PMT was isolated from an ozone-induced Scots pine cDNA library. Southern blot analysis of the genomic DNA suggested the presence of a gene family. The deduced protein sequence showed the typical highly conserved regions of O-methyltransferases (OMTs), and average identities of 20–56% to known OMTs. PMT expressed in Escherichia coli corresponded to that of purified PMT (40 kDa) from pine cell cultures. The recombinant enzyme catalysed the methylation of PS, caffeic acid, caffeoyl-CoA and quercetin. Several other substances, such as astringenin, resveratrol, 5-OH-ferulic acid, catechol and luteolin, were also methylated. Recombinant PMT thus had a relatively broad substrate specificity. Treatment of 7-year old Scots pine trees with ozone markedly increased the PMT mRNA level. Our results show that PMT represents a new SAM-dependent OMT for the methylation of stress-induced pinosylvin in Scots pine needles.  相似文献

Treatment of Pinus sylvestris L. cell-suspension cultures with an elicitor preparation from the pine needle pathogen Lophodermium seditiosum, resulted in a severalhundredto thousandfold accumulation of the stilbenes pinosylvin and pinosylvin 3-O-methyl ether in methanolic cell extracts. There was a simultaneous induction of the biosynthetic enzymes phenylalanine ammonia-lyase (E.C. 4.3.1.5.) and stilbene synthase (pinosylvin-forming, E.C. 2.3.1.146). To estimate the susceptibility of conifer seedlings to aphids under future tropospheric ozone levels, Scots pine (Pinus sylvestris L.) and Norway spruce (Picea abies (L.) Karst.) seedlings were exposed to ambient and elevated ozone levels in an open-air exposure system. Growth and reproduction of the aphids Schizolachnus pineti and Cinara pinea on Scots pine and Cinara pilicornis on Norway spruce were monitored. Levels of free amino acids in foliage and young shoots were used as indicators of host plant quality. In elevated treatment plots the ozone doses were between 1.2 and 1.7 times the dose in ambient plots in 1990–93. Half of the seedling material in 1992–93 was subjected to nitrogen fertilization treatment to evaluate the effects of increased N deposition. In 1990, population density of S. pineti on pine did not differ between ambient and elevated ozone treatments during growing season, but remained higher in the elevated ozone plot than in the ambient plot at the end of the growing season. This was associated with elevated levels of glutamic acid in foliage. In August 1992, the numbers of S. pineti were consistent between the two ambient ozone plots, but deviated highly between the two ozone-fumigated plots. Glycine concentration in pine foliage was elevated by ozone, but free amino acid concentrations were not related to aphid performance. In 1993, ozone and nitrogen did not significantly affect the relative growth rate (RGR) of S. pineti or C. pinea nymphs on Scots pine, but glutamic acid concentration in foliage was increased by nitrogen fertilization. On Norway spruce, fecundity of C. pilicornis females was higher in elevated ozone treatment, but RGR of nymphs was not affected in 1992. In 1993, RGR of C. pilicornis nymphs was increased by nitrogen fertilization in June, but not affected by ozone. Nitrogen fertilization increased the levels of total free amino acids, aspartic acid, glutamic acid and proline in elongating shoots of Norway spruce, and ozone reduced the concentrations of valine and γ-butyric acid. Our results suggest that availability of nitrogen from soil has a stronger impact on the concentrations of free amino acids in conifer seedlings than ozone. Some episodes of high ozone concentration may increase free amino acids in foliage. Aphid response to ozone was extremely variable, in agreement with previous laboratory experiments. The expected 20–70% increase in ambient concentrations of tropospheric ozone may in some occasions enhance aphid performance on Scots pine and Norway spruce seedlings, but in most cases the ozone effect on the susceptibility of conifer seedlings to sucking insect pests is not important.  相似文献

The effects of low‐level ozone exposure and suppression of natural mycorrhizas on the above‐ground chemical quality of Scots pine (Pinus sylvestris L.) needles and insect herbivore performance were studied in a two‐year field experiment. Seedlings were fumigated with the ozone doses 1.5–1.7 times the ambient, and natural mycorrhizal infection level was about 35% reduced in roots with fungicide propiconazole. On ozone‐exposed seedlings the mean relative growth rate (MRGR) of Lygus rugulipennis Popp. nymphs was lower than on ambient ozone seedlings, but Gilpinia pallida Klug sawfly larvae grew better on elevated ozone seedlings than on ambient ozone seedlings. MRGR of Schizolachnus pineti Fabr. and Cinara pinea L. aphid nymphs or Neodiprion sertifer Geoffr. sawfly larvae or the oviposition of L. rugulipennis and N. sertifer were not affected by ozone exposure. Although ozone exposure did not affect total phenolics, total terpene, total or individual resin acid, total free amino acid, nutrient or sugar concentrations in needles, MRGR of L. rugulipennis positively correlated with total terpenes and MRGR of G. pallida positively with total amino acids. In addition, ozone exposure increased serine and proline concentration and marginally also starch concentration in needles. When mycorrhizas were reduced with fungicide, only MRGR of L. rugulipennis nymphs increased, but performance of other insect herbivores studied was not changed. However, number of L. rugulipennis eggs correlated positively with mycorrhizal infection level and also with total sugars. Reduction of mycorrhizas did not strongly affect the concentrations of analysed compounds in needles, because only phosphorus and potassium and some individual resin acids were reduced by fungicide treatment. These results suggest that low‐level ozone exposure and moderately declined mycorrhizal infection do not drastically affect either the above‐ground chemical quality of Scots pine seedlings or performance of studied insect herbivores.  相似文献

Changes in pinosylvin concentration and distribution across the sapwood/heartwood boundary were studied on Scots pine (Pinus sylvestris L.) tree stems to detect seasonal activity in heartwood formation. Pinosylvin concentrations were measured with FT-(NIR) Raman spectroscopy for a total of 96 trees equally distributed on 16 different sampling occasions. In another experiment, cores were sampled every month from six individual Scots pine trees from June to October and analysed for pinosylvin. There was a great between-tree variation in concentration and spatial distribution of pinosylvin. No seasonal trend in the distribution pattern or concentration of pinosylvin was found. Absorption or screening of ultraviolet-B (UV-B) radiation by the epidermis may be an important protective method by which plants avoid damage upon exposure to potentially harmful UV-B radiation. In the present study we examined the relationships among epidermal screening effectiveness, concentration of UV-absorbing compounds, epidermal anatomy and growth responses in seedlings of loblolly pine (Pinus taeda L.) and sweetgum (Liquidambar styraciflua L.). Seedlings of each species were grown in a greenhouse at the University of Maryland under either no UV-B radiation or daily supplemental UV-B radiation levels of 4, 8 or 11 kJ m?2 of biologically effective UV-B (UV-BBE) radiation. Loblolly pine seedlings were subsequently grown in the field under either ambient or supplemental levels of UV-B radiation. At the conclusion of the growing season, measurements of epidermal UV-B screening effectiveness were made with a fiber-optic microprobe. In loblolly pine, less than 0.5% of incident UV-B radiation was transmitted through the epidermis of fascicle needles and about 1% was transmitted in primary needles. In contrast, epidermal transmittance in sweetgum ranged from about 20% in leaves not preconditioned to UV-B exposure, to about 10% in leaves grown under UV-B radiation. The concentration of UV-absorbing compounds was unaffected by UV-B exposure, but generally increased with leaf age. Increases in epidermal thickness were observed in response to UV-B treatment in loblolly pine, and this accounted for over half of the variability in UV-B screening effectiveness. In spite of the low levels of UV-B penetration into the mesophyll, delays in leaf development (both species) and final needle size (loblolly pine) were observed. Seedling biomass was reduced by supplemental UV-B radiation in loblolly pine. We hypothesize that the UV-induced growth reductions were manifested by changes in either epidermal anatomy or epidermal secondary chemistry that might negatively impact cell elongation.  相似文献

Scots pine (Pinus sylvestris L.) seedlings were fumigated with 1.2–1.5 x ambient ozone (cumulative exposure) over 2 seasons in an open-air experiment. Starch and fatty acid concentrations were analyzed in needle and root tissue in the summer, autumn and early winter. Seedling growth was determined by measuring the height of the stem and the total shoot and root biomass. Significant decreases in growth were found in exposed seedlings, even though visible symptoms were lacking. Almost significant reductions in needle and root starch concentrations were found. In the ozone treated foliage, significant increases in myristic acid (140) were detected, but the major fatty acids remained unchanged. Fatty acid ratios showed that the degree of unsaturation decreased in treated needles in the summer. In the roots of ozone treated seedlings, changes in fatty acids were different from those in the foliage. Decreases of the main root fatty acids (160, 180, 181, 18:2, 183) were detected in the summer. These results show that Scots pine is susceptible to enhanced levels of ozone. If the tropospheric ozone levels continue to increase it may have deleterious effects on Scots pine forests in Finland.  相似文献

Projected depletions in the stratospheric ozone layer will result in increases in solar ultraviolet-B radiation (290–320 nm) reaching the earth's surface, These increases will likely occur in concert with other environmental changes such as increases in atmospheric carbon dioxide concentrations. Currently very little information is available on the effectiveness of UV-B radiation within a CO2-enriched atmosphere, and this is especially true for trees. Loblolly pine (Pinus taeda L.) seedlings were grown in a factorial experiment at the Duke University Phytotron with either 0, 8.8 or 13.8 kJ m−2 of biologically effective UV-B radiation (UV-BBE). The CO2 concentrations used were 350 and 650 μmol mol−1. Measurements of chlorophyll fluorescence were made at 5-week intervals and photosynthetic oxygen evolution and leaf pigments were measured after 22 weeks, prior to harvest. The results of this study demonstrated a clear growth response to CO2 enrichment but neither photosynthetic capacity nor quantum efficiency were altered by CO2. The higher UV-B irradiance reduced total biomass by about 12% at both CO2 levels but biomass partitioning was altered by the interaction of CO2 and UV-B radiation. Dry matter was preferentially allocated to shoot components by UV-B radiation at 350 μmol mol−1 CO2 and towards root components at 650 μmol mol−1 CO2. These subtle effects on biomass allocation could be important in the future to seedling establishment and competitive interactions in natural as well as agricultural communities.  相似文献

Formation of the stilbenes pinosylvin and pinosylvin 3-methyl ether, as well as the activity of the biosynthetic enzyme stilbene synthase (pinosylvin-forming), were induced several hundred- to thousandfold in primary needles of 6-week-old pine (Pinus sylvestris L.) seedlings upon exposure to a single pulse of ozone of at least 0.15 microliters per liter. The seedlings required 4 hours of exposure as a minimum for the induction of stilbene biosynthesis when exposed to 0.2 microliters per liter ozone. Both stilbene synthase activity and stilbene accumulation increased with the duration of ozone treatment. The activity of phenylalanine ammonia-lyase and the activity of chalcone synthase, a key enzyme of the flavonoid pathway that uses the same substrates as stilbene synthase, were also stimulated about twofold by ozone. Stilbene biosynthesis appears to represent the first example of a dose-dependent biochemical response to ozone in a conifer species and may serve as a useful biomarker to study stress impacts on pine trees.  相似文献

As a consequence of the ongoing reduction of the stratospheric ozone layer, the vegetation is exposed to increasing levels of UV-B radiation (280–320 nm). In addition ozone in the troposphere is a pollutant and also capable of affecting the photosynthetic machinery. In this study, 5-year-old European beech trees were exposed from 1 July to October 1993 to two levels of UV-B radiation and two levels of ozone, alone and in combination, in open-top chambers equipped with lamps. The simulated UV-B levels corresponded to either clear sky ambient level or a 14% decrease in the stratospheric ozone column over eastern Denmark, resulting in a 23% difference in biologically effective UV-B (UV-BBE) irradiance. The maximum UV-Bbe given was 8.61 kJ m−2 day−1. The ozone levels were either the ambient (average 32 nl l−1) or ambient with ozone addition (average resulting concentration 71 nl l−1). Compared to the control treatment (ambient UV-B, ambient O3) the elevated levels of UV-B and O3 affected the trees negatively, expressed as declines in net photosynthesis (Pn), stomatal conductance (gs), chlorophyll fluorescence (Fv/Fm) and acceleration of senescence, measured as yellowing of the leaves. The UV-B treatment induced stomatal closure before the other treatments did. The magnitude of the decreases in Pn and Fv/Fm occurred in the order: control 3 3. Compared to the control, the combination treatment with high levels accelerated the visual senescence processes by ca 27 days, while for high UV-B and O3 alone, there was an acceleration by 14 and 21 days, respectively. UV-B and O3 in combination enhanced the negative effects compared with UV-B and O3 alone. The Pn and Fv/Fm results could be related to this acceleration process. The chamber effect was investigated by comparing the control plots with a plot without open-top chamber. The trees in the chambers showed a higher Pn and Fv/Fm and a 14-day delayed senescence compared to the trees outside, probably caused by higher temperatures, a more protected environment and altered conditions inside the chambers.  相似文献

Elevated atmospheric carbon dioxide (CO2) and ozone (O3) concentrations have both been shown to affect plant tissue quality, which in turn could affect litter decomposition and carbon (C) and nutrient cycling. In order to evaluate effects of climate change on litter chemistry, needle litter was collected from Scots pine (Pinus sylvestris L.) saplings exposed to elevated CO2 or O3 concentration and their combination over three growing seasons in open‐top chambers. The decomposition of needle litter was followed for 19 months in a pine forest. During decomposition, needle samples for secondary compound analysis were collected and the mass loss of needles was followed. Main nutrients and total phenolics were analysed from litter in the beginning and at the end of the experiment. After 19‐month decomposition, the accumulated mass loss was about 34%; however, no significant differences were found in decomposition rates of needle litter between various treatments. Concentrations of total monoterpenes were about 4%, total resin acids 21% and total phenolics 14% of the initial concentrations in litter after 19‐month decomposition. In the beginning of litter decomposition, concentrations of individual monoterpenes –α‐pinene and β‐pinene – were significantly higher in needle litter grown under elevated CO2. However, concentrations of total monoterpenes during the whole decomposition period were not significantly affected by CO2 or O3 treatments. Concentrations of some individual and total resin acids were higher in needle litter grown under elevated CO2 or O3 than under ambient air. There were no significant differences in concentrations of total phenolics as well as nitrogen (N) and the main nutrient concentrations between treatments during decomposition. High concentrations of monoterpenes and resin acids in needles might slightly delay C recycling in forest soils. It is concluded that elevated CO2 and O3 concentrations do not have remarkable impacts on litter decomposition processes in Scots pine forests.  相似文献

Chalcone synthase (CHS) and stilbene synthase (STS) are closely related polyketide synthases which are key enzymes in the biosynthesis of flavonoids and stilbenes. Scots pine (Pinus sylvestris) is an interesting plant for a direct comparison of the enzymes. It not only contains the usual flavonoids, but also an unusual chalcone derivative (pinocembrin), and it synthesizes stilbenes of the pinosylvin type. We analysed a CHS and a STS by molecular cloning and functional expression in Escherichia coli. The CHS was active not only with 4-coumaroyl-CoA (to naringenin chalcone), but also with cinnamoyl-CoA (leading to pinocembrin). The STS was identified as dihydropinosylvin synthase, because it preferred dihydrocinnamoyl-CoA to cinnamoyl-CoA. The protein deviated in 47 positions from the CHS consensus. It had 73.2% identity with the CHS from P. sylvestris and only 65.3% with a STS from peanut (Arachis hypogaea). We also investigated the regulation of both enzyme types in P. sylvestris plantlets exposed to stress. CHS was present in non-stressed plantlets, and induction led to a transient increase with a peak after 16 h. STS1 type activities were regulated differently and were absent in non-stressed plantlets. Increases were observed after a lag period of at least 6 h, and highest activities were obtained after 30 h. The analysis of the reactions in the plant extracts and the substrate specificity of the cloned STS indicate that the plants contain at least two different types of STS: the cloned dihydropinosylvin synthase and a pinosylvin synthase which preferentially utilizes cinnamoyl-CoA as substrate.  相似文献

Summary The effect of ozone, needle age, and season on the pH of homogenate and acid contents of Scots pine and Norway spruce needles is presented. In addition enzyme activities of cytochrome C-oxidase (cyt. C-ox), phosphoenolpyruvate-carboxylase (PEPC), shikimic acid-dehydrogenase (SHDH) and malate-dehydrogenase (MDH) were measured in Scots pine needles. In freshly sprouted spruce needles the level of quinic acid is high and the pH of the needle homogenate is low. Shikimic acid starts at low levels, increases with increasing needle age and becomes dominant, whereas the quinic acid content decreases. Malic acid has a marked seasonal trend; no trend was found in citric acid. Ozone (200 g/m3) decreased shikimic acid and quinic acid, whereas pH, malic acid and citric acid increased. Ozone (100 g/m3) had a similar effect, except in the current-year spruce needles. In Scots pine needles ozone led to increased enzymatic activities of cyt. C-ox, PEPC and SHDH, and a decrease in the activity of MDH. This effect was more pronounced in summer than in autumn, but the visible damage was greater in autumn. These effects can be found with other stresses and are not specific for ozone.  相似文献

Scots pine (Pinus sylvestris L.) and Norway spruce (Picea abies Karst.) seedlings were exposed to realistically elevated O3 levels in open‐air experiments over three growing seasons. The total O3 exposure doses were 1.2 × (1991), 1.5 × (1992) and 1.7 × (1993) ambient levels. During the 1992 and 1993 growing seasons pine and spruce seedlings received two different levels of nitrogen supply. Effects on growth, mycorrhiza formation, needle ultrastructure, primary and secondary compounds were studied. Ozone exposure had only slight effects on biomass production, growth height and nutrient content of studied conifers. Higher nitrogen availability improved growth of the seedlings and resulted in higher concentration of nitrogen in needles. In Scots pine O3 exposure did not have effects on quantity of total mycorrhizas and short roots, while higher nitrogen availability decreased quantity of mycorrhizas and short roots. In both tree species O3 exposure induced O3‐related ultrastructural symptoms, e.g. granulation and dark staining of the chloroplast stroma in the needle mesophyll cells, at both nitrogen availability levels. Ozone exposure and nitrogen availability did not have significant effects on starch concentrations in either tree species. Concentrations of some individual terpenes were higher in O3‐exposed needles, while concentrations of individual and total resin acids, total phenolics and catechins were not affected by O3 exposure. Nitrogen availability did not have substantial effects on concentrations of monoterpenes. By contrast, concentrations of some individual and total resin acids were lower in pine needles and higher in spruce needles with higher nitrogen availability, while phenolic concentration in spruce needles decreased at higher nitrogen availability. The results suggest that realistically elevated levels of O3 in the field can have some negative effects on the mesophyll ultrastructure of conifer needles, but carbon allocation to root and shoot growth and secondary metabolites are not affected substantially.  相似文献

Young Scots pine trees naturally established at a pine heath were exposed to two concentrations of CO2 (ambient and doubled ambient) and two O3 regimes (ambient and doubled ambient) and their combination in open-top field chambers during growing seasons 1994, 1995 and 1996 (late May to 15 September). Filtered ozone treatment and chamberless control trees were also included in the treatment comparisons. Root ingrowth cores were inserted to the undisturbed soil below the branch projection of each tree at the beginning of the fumigation period in 1994 and were harvested at the end of the fumigation periods in 1995 and 1996. Root biomasses were determined from different soil layers in the ingrowth cores, and the infection levels of different mycorrhizal types were calculated. Elevated O3 and CO2 did not have significant effects on the biomass production of Scots pine coarse (Ø > 2 mm) or fine roots (Ø < 2 mm) and roots of grasses and dwarf shrubs. Elevated O3 caused a transient stimulation, observable in 1995, in the proportion of tuber-like mycorrhizas, total mycorrhizas and total short roots but this stimulation disappeared during the last study year. Elevated CO2 did not enhance carbon allocation to root growth or mycorrhiza formation, although a diminishing trend in the mycorrhiza formation was observed. In the combination treatment increased CO2 inhibited the transient stimulating effect of ozone, and a significant increase of old mycorrhizas was observed. Our conclusion is that doubled CO2 is not able to increase carbon allocation to growth of fine roots or mycorrhizas in nutrient poor forest sites and realistically elevated ozone does not cause a measurable limitation to roots within a period of three exposure years.  相似文献

Terpene, resin acid and total phenolic concentrations in five‐year‐old Scots pine (Pinus sylvestris L.) seedlings were analysed after exposure to ambient and realistically elevated (2 × ambient) O3 and CO2 concentrations and their combination in open‐top chambers during two growing seasons. Under O3 exposure, limonene concentration in needles and isopimaric concentration in stems decreased significantly. As a response to elevated CO2, α‐pinene and total phenolic concentrations in needles increased significantly, while bornyl acetate concentration in needles and palustric + levopimaric and neoabietic acid concentrations in stems decreased significantly. Some terpenes and resin acids were found at lower concentrations in the combined O3 and CO2 treatment than in O3 exposure or elevated CO2. A negative chamber effect was found: seedlings growing inside the chambers with ambient air had significantly lower concentrations of some terpenes and resin acids than seedlings growing outside the chambers. There was a lot of between‐tree variation in terpene and resin acid concentrations, which is typical of open‐pollinated populations. The results of this study suggest that, at least in short‐term experiments, Scots pine secondary metabolites are relatively insensitive to climate change factors. Total phenolics in the needles were the most responsive group showing about 25% increase in elevated CO2, and O3 exposure did not mitigate this CO2 effect. Terpenes and resin acids were less responsive, although some individual compounds showed notable responses, e.g. α‐pinene in needles, which increased about 50% in response to elevated CO2. As a consequence, although there were only slight effects on total pools of needle secondary metabolites, considerable O3 and CO2 effects on certain individual compounds might have ecological significance via trophic amplification, e.g. in decomposing processes of needle litter.  相似文献

The aim of this work was to test our hypothesis that pine needles protect themselves against UV-B radiation via anatomical changes in the epidermal layer. This could lead to needle growth reductions if large quantities of assimilates are allocated for the epidermal protective mechanisms at the expense of photosynthetic area. Effects of enhanced UV-B radiation on the needle ontogeny of mature Scots pines (Pinus sylvestris L.) were studied during the second season of a field experiment. Depending on the season and the time of the year (1996-1997), the enhanced UV-B irradiance varied from 0.92 to 5.09 kJ m-2 day-1 UV-BBE compared to 0.54-2.44 kJ m-2 day-1 UV-BBE of ambient radiation. It was found that UV-B treatment accelerated the early development of needles. In 6-day-old enhanced UV-B-treated needles, mesophyll and hypodermic cells were fully differentiated, whereas in ambient-treated needles, no lobate mesophyll cells were seen and hypodermic cells had not yet developed. In fully grown needles, no accelerated differentiation was seen, except that the epidermal cross-sectional area was smaller. The continuation of the experiment will show if such a significant difference only occurs irregularly and incidentally or if it is of consistent significance for needles.  相似文献
