The hypothesis postulating that the antioxidant and immunological effects of dietary methionine (Met) in young turkeys (1–8 weeks of age) can be differentiated by level and source of Met was investigated in this study. A total of 544 female Hybrid Converter turkeys were divided into four groups and fed diets in which Met content was tailored through supplementation with dl-methionine (dl-Met) or dl-methionine hydroxy analogue (MHA) to levels recommended by NRC (1994) (Groups dl-MetL and MHAL) and exceeding them by 50% (Groups dl-MetH and MHAH). Regardless of its source, the increased dietary Met content led to significantly higher body weight gains but had no effect on feed conversion rates. Moreover, an increased Met content lowered lipid peroxide concentrations in breast meat and increased selected indicators of the plasma antioxidant status like uric acid levels, activity of superoxide dismutase (SOD), glutathione (GSH) concentrations, the ferric-reducing ability of plasma (FRAP), increased immunoglobulin A (IgA) plasma levels and decreased interleukin 6 levels. In comparison with dl-Met, MHA decreased the activities of SOD and catalase, and GSH concentrations in plasma. A dosage by source interaction revealed that the lower MHA content was associated with the lowest plasma GSH concentrations, FRAP values and activities of SOD and catalase. The higher dietary MHA level resulted for most parameters similar values, except for a decrease in lipid peroxide concentrations and an increase in plasma IgA levels. It can be concluded that an increased dietary dl-Met and MHA content (about 150% of the recommendations given by NRC 1994) not only increased the growth rate of young turkeys but also improved their antioxidant status. MHA appears to be a less desirable source of dietary Met for young turkeys when the inclusion level of Met matches the current recommendations. Therefore, a further debate is needed to establish the dietary requirements for Met in poultry.  相似文献

Understanding the relationship between nutrition and satellite cell activity will be beneficial in obtaining optimal muscle growth and meat production. The objective of this study was to evaluate the effect of early post-hatch levels of dietary amino acids+/-0.88 NRC, 1.00 NRC, and 1.12 NRC), and feed deprivation on the satellite cell mitotic activity, pectoralis thoracicus muscle weight, and body weight of male turkeys (Meleagris gallopavo). Birds from each treatment were injected with 5-bromo-2'-deoxyuridine (BrdU) to label mitotically active cells. The right pectoralis thoracicus was harvested 1 h after BrdU injection for immunohistochemical and myofiber diameter analysis. On the third day post-hatch, satellite cell mitotic activity was the highest (P<0.05) in the 0.88 NRC amino acid treatment group and the lowest (P<0.05) in the feed-deprived group. On the fourth day post-hatch, feed-deprived birds exhibited the lowest (P<0.05) satellite cell mitotic activity and muscle weight. At 140 days of age, there were no significant differences (P>0.05) between treatments in body weight or pectoralis thoracicus muscle weight. Research evaluating species-related differences in apoptotic events and in genes regulating cell proliferation may be necessary to devise feeding strategies aimed at obtaining optimal pectoralis thoracicus muscle yield at market age.  相似文献

A total of 490 eight-week-old female Hybrid Converter turkeys (body weight 4.11 ± 0.03 kg) were divided into 5 groups with 7 replicates of 14 birds each. For 8 weeks, basal diets were supplemented with methionine (Met) at following levels (weeks 9–12/weeks 13–16 of age): Group 1 – 0.34/0.29%, Group 2 – 0.39/0.34%, Groups 3 and 4 – 0.45/0.38% and 0.51/0.41%, respectively, Group 5 – 0.58/0.47%. Only in the first feeding phase the body weight gain (BWG) was affected by Met levels with the significantly highest BWG in Group 3. No treatment effects were found for feed conversion ratio, carcass yield, carcass composition and meat colour. The blood superoxide dismutase activity was significantly highest in Groups 2 and 3. The concentrations of reduced glutathione in the liver were linearly increased (p = 0.018), whereas the ratio of reduced glutathione to oxidised glutathione was highest in Group 3 (quadratic contrast, p = 0.004). It can be concluded that turkeys from Group 3 (Met levels age depending 15% and 10% above recommendations by NRC) were characterised by a well-balanced physiological response. Attention should be paid to the immune response of birds to higher dietary Met levels: plasma IgA concentrations decreased, whereas IL-6 and TNF-α levels increased in turkeys fed diets with the highest Met content.  相似文献

The aim of this study was to investigate the effect of dietary replacement of soya bean meal (SBM) with faba bean (FB) and a blend of non-starch polysaccharide (NSP) degrading enzymes on the gastrointestinal function, growth performance and welfare of young turkeys (1 to 56 days of age). An experiment with a 2×2 factorial design was performed to compare the efficacy of four diets: a SBM-based diet and a diet containing FB, with and without enzyme supplementation (C, FB, CE and FBE, respectively). In comparison with groups C, higher dry matter content and lower viscosity of the small intestinal digesta were noted in groups FB. The content of short-chain fatty acids (SCFAs) in the small intestinal digesta was higher in groups FB, but SCFA concentrations in the caecal digesta were comparable in groups C and FB. In comparison with control groups, similar BW gains, higher feed conversion ratio (FCR), higher dry matter content of excreta and milder symptoms of footpad dermatitis (FPD) were noted in groups FB. Enzyme supplementation increased the concentrations of acetate, butyrate and total SCFAs, but it did not increase the SCFA pool in the caecal digesta. The enzymatic preparation significantly improved FCR, reduced excreta hydration and the severity of FPD in turkeys. It can be concluded that in comparison with the SBM-based diet, the diet containing 30% of FB enables to achieve comparable BW gains accompanied by lower feed efficiency during the first 8 weeks of rearing. Non-starch polysaccharide-degrading enzymes can be used to improve the nutritional value of diets for young turkeys, but more desirable results of enzyme supplementation were noted in the SBM-based diet than in the FB-based diet.  相似文献

To determine the parameters of non-specific immunity in rabbits infected with 10 biologically different strains of the rabbit haemorrhagic disease (RHD) virus (BS89, Hagenow, Rainham, Frankfurt, Asturias, Vt97, Triptis, Hartmannsdorf, Pv97, 9905 RHDVa) the following indices were assessed: polimorphonuclear cell (PMN) adherence capacity, absorption index, percentage of absorbing cells, spontaneous, stimulated as well as spectrophotometric test for reduction of the nitroblue tetrazolium (NBT), metabolic activity coefficient of neutrophilic granulocytes, stimulation index, myeloperoxidase activity, lysozyme concentration and activity index. The symptoms and the percentage of mortality in animals infected were recorded. On the basis of the estimated parameters, the present study confirmed the existence of immunotypes among the strains of the RHD virus. However, the results did not indicate a connection between biological property of the RHD virus (haemagglutination capacity and generation of antigenic variants) and the immunological profile of the strains.  相似文献

Aging has been defined as the changes that occur in living organisms with the passage of time that lead to functional impairment and ultimately to death. Free radical-induced oxidative damage has long been thought to be the most important consequence of the aging process. In the present study, an attempt has been made to study the salubrious effects of dietary supplementation of chitosan on glutathione-dependent antioxidant defense system in young and aged rats. The dietary supplementation of chitosan significantly reduced the age-associated dyslipidemic abnormalities noted in the levels of total cholesterol, HDL-cholesterol, and LDL-cholesterol in plasma and heart tissue. Its administration significantly (P < 0.05) attenuated the oxidative stress in the heart tissue of aged rats through the counteraction of free radical formation by maintaining the enzymatic [glutathione peroxidase (GPx) and glutathione reductase (GR)] and non-enzymatic [reduced glutathione (GSH)] status at levels comparable to that of normal young rats. Our results conclude that dietary intake of chitosan restores the depleted myocardial antioxidant status and suggest that it could be an effective therapeutic agent in treatment of age-associated disorders where hypercholesterolemia and oxidative stress are the major causative factors.  相似文献

At high ambient temperature (Ta=35°C) weight gain and feed intake declined significantly. At 15°C weight gain was similar to that at 25°C, at the cost of increased feed intake. Under diurnal cyclic temperature, weight gain and feed intake were significantly lower than in the average corresponding temperature.In all treatments the turkeys’ body temperature (Tb) was at the lower level of normothemia known for broiler chickens at a similar age.The blood system compensated for changes in Ta by increasing hematocrit, hemoglobin concentration and heart muscle weight at low Ta, and by plasma expansion and increased panting at high Ta.Plasma (T3) concentration was positively correlated with feed intake and weight gain.  相似文献

Immune response and antioxidant status of immune organs in juvenile Jian carp (Cyprinus carpio var. Jian) fed graded levels of methionine hydroxy analogue (MHA) (0, 5.1, 7.6, 10.2, 12.7, 15.3 g kg(-1) diet) for 60 days were investigated. Results indicated that head kidney index, spleen index, red and white blood cell counts significantly increased with increasing MHA levels up to a point (P < 0.05), whereupon decreased (P < 0.05). Glutathione reductase activity in head kidney and spleen, anti-hydroxy radical and glutathione-S-transferase activities in spleen, catalase activity and GSH content in head kidney significantly increased by MHA supplement, while malondialdehyde content, anti-superoxide anion, superoxide dismutase, glutathione peroxidase activities in head kidney and spleen, protein carbonyl content and catalase activity in spleen, anti-hydroxy radical activity in head kidney significantly decreased by MHA supplement. However, protein carbonyl content and glutathione-S-transferase activity in head kidney, GSH content in spleen remained unaffected. After 60-day feeding trial, a challenge study was conducted by injection of Aeromonas hydrophila for 17 days. Results showed that survival rate, leukocytes phagocytic activity, lysozyme activity, acid phosphatase activity, total iron-binding capacity, haemagglutination titre, complement 3, 4 and immunoglobulin M contents significantly increased by optimal dietary MHA supplement (P < 0.05). These data suggested that MHA affected antioxidant status of immune organs and promoted immune response in juvenile Jian carp.  相似文献

The effect of melatonin on the intensity of free radical processes and activities of superoxide dismutase (SOD, EC 1.15.1.1.) and catalase (EC 1.11.1.6) has been investigated in liver and blood serum of rats with type II diabetes mellitus. According to results of this study the development of diabetes was accompanied by the increase in biochemiluminescence parameters and the enzyme activities studied. Melatonin administration changed the parameters studied towards control values and this is obviously associated with realization of the antioxidant potential of this hormone.  相似文献

The effect of dietary manganese from two different sources on chicks (uninfected and infected with Ascaridia galli) was studied. Chick diet was supplemented with 0.9 g Mn2+ kg-1 food either in the form of MnSO4.H2O or 2Gly. MnCl2.2H2O for 20 days. Chicks were divided into six groups: group 0, control; group 1, control + MnSO4.H2O; group 2, control + 2Gly.MnCl2.2H2O; group 3, infected with A. galli; group 4, infected with A. galli + MnSO4.H2O; and group 5, infected with A. galli + 2Gly.MnCl2.2H2O. Body weight, mortality, worn burden, and liver manganese content were investigated. Excess dietary manganese increased weights and manganese level, but mortality and worm burden were unaffected. A greater bioavailability of manganese from 2Gly.MnCl2.2H2O was established.  相似文献

The experiment investigated the effect of dietary bovine lactoferrin (bLF) on growth performance and antioxidant systems of piglets. Duroc–Landrace–Yorkshire crossbred female piglets (n = 120, 35 days of age, live-weight 9.70 ± 0.71 kg) were fed a diet containing 0, 1250 or 2500 mg/kg bLF for 30 days. After completion of the feeding experiment, eight animals from each treatment were randomly selected to determine malondialdehyde (MDA) and total antioxidant capacity (TAOC), copper–zinc–superoxide dismutase (CuZnSOD), glutathione peroxidase (GPx), catalase (CAT) activities in serum and longissimus muscle, and CuZnSOD, GPx and CAT mRNA levels in longissimus muscle. Results showed that supplementation with bLF improved average daily gain (ADG), average daily feed intake (ADFI) and reduced diarrhea ratios of piglets with the linear and quadratic polynomial contrasts being significant (P<0.01). The highest weight gain and the best feed conversion occurred at 2500 mg/kg bLF. Serum and longissimus muscle CuZnSOD and GPx, and serum TAOC linearly and quadraticly increased (P<0.01), serum CAT and longissimus muscle CAT and TAOC linearly increased (P<0.01), whereas MDA concentrations in serum and longissimus muscle linearly decreased (P<0.01) as dietary bLF supplementation increased. Increasing dietary bLF levels improved (P<0.01) mRNA expression of CuZnSOD and CAT in longissimus muscle of piglets. The results indicated that exogenous antioxidant bLF was effective in improving growth performance and enhancing antioxidant enzymes activity and mRNA levels of piglets.  相似文献

Two types of fat, represented by cod liver oil and lard, were added at various levels to the diet of rainbow trout. The food intake of the trout and their growth rate were measured. The results indicated that when cod liver oil was used as the fat supplement, the amount and the water temperature affected food intake and weight gain; fish fed on lard showed only a slight loss of appetite.  相似文献

Attention has recently been focused on endothelial function after a single high-fat meal, i.e. on the anticipated direct atherogenic effect of triglyceride-rich lipoproteins. Our study was designed to investigate the effect of a low-fat diet given for four weeks followed by a high-fat diet for another four weeks. At the end of each dietary period, a noninvasive ultrasound investigation of endothelial function of the brachial artery was performed along with laboratory tests. Endothelial function was measured immediately before the dietary load and after three and six hours in 11 healthy volunteers. The results were expressed as percentage of the changes in artery diameter at rest and during hyperemia; the data were processed using computer technology. When compared to the low-fat regimen, the total cholesterol content rose after the high-fat diet from 4.28 mmol/l to 5.15 mmol/l (p<0.05) in the whole group of volunteers. There was no difference between both dietary regimens in baseline triglycerides. The brachial artery dilatation under basal conditions was 5.26+/-2.88 mm after the high-fat diet compared with the value of 3.13+/-3.01 mm (p<0.05) after the low-fat diet. When measured individually endothelial function in the whole group of volunteers in the course of the day, the degree of arterial dilatation after one month on low-fat diet was 3.13+/-3.0%, 3.88+/-2.5% and 5.23+/-3.3% at single measurement. When comparing arterial dilatation at two closest measurements, a non-significant trend, p>0.05 was seen in either case. The following values were obtained after one month on the high-fat diet: 5.26+/-2.9%, 4.47+/-1.7%, and 6.2+/-3.6%; again showing a non-significant trend of p>0.05. In this study, a single high-fat meal at the different dietary regimen did not significantly influence the vasoreactivity of the brachial artery in young volunteers.  相似文献

An increasing body of evidence substantiating the effectiveness of balneotherapy has accumulated during recent decades. In the present study, 42 ambulatory patients (23 males and 19 females, mean age 59.5 years) with degenerative musculoskeletal disease were randomised into one of two groups—bathing in tap water or in mineral water at the same temperature—and subjected to 30-min balneotherapy sessions on 15 occasions. Study parameters comprised serum levels of sensitised C-reactive protein (CRP), plasma lipids, heat shock protein (HSP-60) and total antioxidant status (TAS). In both groups, CRP levels followed a decreasing tendency, which still persisted 3 months later. At 3 months after balneotherapy, serum cholesterol levels were still decreasing in patients who had used medicinal water, but exhibited a trend towards an increase in the control group. Triglyceride levels followed a decreasing trend in both patient groups. TAS showed a declining tendency in both groups. No changes of HSP-60 levels were observed in either group. Balneotherapy with the thermal water from Hajdúszoboszló spa had a more pronounced physiological effect compared to that seen in the control group treated with tap water in a 3 month period.  相似文献

Summary.  Wistar rats were fed with different diets with or without supplement coenzyme Q10 (CoQ10) and with oil of different sources (sunflower or virgin olive oil) for six or twelve months. Ubiquinone contents (CoQ9 and CoQ10) were quantified in homogenates of livers and brains from rats fed with the four diets. In the brain, younger rats showed a 3-fold higher amount of ubiquinone than older ones for all diets. In the liver, however, CoQ10 supplementation increased the amount of CoQ9 and CoQ10 in both total homogenates and plasma membranes. Rats fed with sunflower oil as fat source showed higher amounts of ubiquinone content than those fed with olive oil, in total liver homogenates, but the total ubiquinone content in plasma membranes was similar with both fat sources. Older rats showed a higher amount of ubiquinone after diets supplemented with CoQ10. Two ubiquinone-dependent antioxidant enzyme activities were measured. NADH-ferricyanide reductase activity in hepatocyte plasma membranes was unaltered by ubiquinone accumulation, but this activity increased slightly with age. Both cytosolic and membrane-bound dicumarol-sensitive NAD(P)H:(quinone acceptor) oxidoreductase (DT-diaphorase, EC 1.6.99.2) activities were decreased by diets supplemented with CoQ10. Animals fed with olive oil presented lower DT-diaphorase activity than those fed with sunflower oil, suggesting that the CoQ10 antioxidant protection is strengthened by olive oil as fat source. The aim of this study was to investigate the effect of brachytherapy on lipid peroxidation and antioxidant status in patients with uterine cervix cancer. The study was conducted on 84 uterine cervix cancer patients from the Brachytherapy Department of the Regional Centre of Oncology in Bydgoszcz. Patients with uterine cervix cancer were found to have elevated levels of lipid peroxidation and antioxidant defence system impairment relative to healthy females. The results of the study indicate that brachytherapy has no direct effect on the antioxidant system of patients with uterine cervical carcinoma. However, the normalisation of catalase and glutathione peroxidase activity and erythrocyte TBARS level observed six months after the end of therapy may be due to the arrest of the progression of the disease.  相似文献

The only method of assessing the virulence of myxoma virus is to record survival times of rabbits inoculated with the virus. This raises ethical concerns about using animals in experiments where death is the end point. We investigated whether or not the opioid analgesic buprenorphine could be used in rabbits without compromising the myxoma virus virulence assay and on the presumption that animals may suffer pain during the course of the disease. Thirty, 5-month-old New Zealand White rabbits were divided into two groups stratified for weight and gender, and inoculated intradermally with 100 pfu of the Standard Laboratory Strain (SLS) of myxoma virus. At day 6 post infection (p.i.), when eyelid swelling was first seen, each animal in one group was treated with 0.03 mg/kg buprenorphine, subcutaneously, morning and evening until death. Animals in the other group were untreated. Animals were weighed daily and rectal temperatures taken morning and evening. Intake of food and water was assessed as was general demeanor including respiratory effort. There was no significant difference in mean survival time, weight change, or demeanor between the two groups. Increased respiratory effort was seen from day 10 p.i. in animals surviving up to and beyond that time but again there was no difference between groups. Animals treated with buprenorphine refused food and water a day earlier than untreated animals, and had lower temperatures immediately prior to death. It was concluded that the opiate analgesic buprenorphine can be used without compromising the current virulence assay for the SLS of myxoma virus in New Zealand White rabbits but that the clinical signs of myxomatosis that could be attributed to pain were not abrogated.  相似文献
