安徽大别台区虫生真菌区系的物种多样性研究   总被引:7,自引:0,他引:7  
对安徽大别山区虫生真菌区系的物种多样性进行了研究。结果表明该地区虫生真菌资源十分丰富 ,共有虫生真菌 5 0种 ,隶属于 4目 4科 16属。科从大到小依次为麦角菌科 (Clavicipitaceae) (2 7种 ,占 5 4 % )、丝孢科 (Hy phomycetaceae) (15种 ,占 30 % )、虫霉科 (Entomophthoraceae) (5种 ,占 10 % )、束梗孢科 (Stilbellaceae) (3种 ,占 6 % )。含 3个种以上的优势属依次为虫草属 (Cordyceps) (2 6种 ,占 5 2 % )、拟青霉属 (Paecilomyces) (6种 ,占 12 % )、白僵菌属 (Beauveria) (3种 ,占 6 % ) ,共计 35种 ,占总种数的 70 % ;含 2个种的属为刺束梗孢属 (Akanthomyces)和虫瘟霉属 (Zoophthora) ;其余 11个属皆为单种属。优势种依次为粉拟青霉 (Paecilomycesfarinosus)、细脚拟青霉 (P .tenuipes)、下垂虫草 (Cordycepsnutans)、球孢白僵菌 (Beauveriabassiana)、金龟子绿僵菌小孢变种 (Metarhiziumanisopliaevar.anisopliae)。区系地理成分划分为 7大类型 :世界分布种 (42 .11% )、欧亚大陆分布种 (10 .5 3% )、亚热带—热带分布种 (5 .2 6 % )、东亚分布种 (7.89% )、东亚—新几内亚分布种 (5 .2 6 % )、中国—日本分布种 (5 .2 6 % )和特有成分 (2 3.6 8% ) ,表现出明显的东亚区系特征 ,而且本区及  相似文献

We investigated the occurrence of entomopathogenic fungi in 1080 soil samples representing multiple locations and conditions in Korea. Entomopathogenic fungi were isolated from soils using a selective medium containing dodine and antibiotics. Following an initial identification based on morphology, the fungal isolates were more precisely identified by the sequence of their nuclear ribosomal RNA (rRNA) internal transcribed spacer (ITS) regions. As a result, entomopathogenic fungi were found to occur in 32% (342 isolates) of the soil samples studied. The most abundant species were Beauveria spp. (125 isolates) and Metarhizium spp. (82 isolates). Entomopathogenic fungi were more often recovered from natural mountain and riparian soils than from agricultural habitats. The pathogenicity of isolated fungi was evaluated by using wax moth Galleria mellonella L. (Lepidoptera: Pyralidae) larvae. It was determined that 60% (207 isolates) of the isolates were pathogenic using this model. These entomopathogenic fungi may, therefore, have potential use against a variety of agricultural pests. This is the first study of the isolation and distribution of entomopathogenic fungi in representative sampling locations throughout Korea.  相似文献

应用ISSR分子标记对安徽大别山区的球孢白僵菌遗传多样性进行了研究。从33个引物中筛选出12个多态性高、稳定性好的ISSRs用于正式的扩增分析,在2个自然保护区、3个不同季节和3个不同海拔梯度采集的48个菌株中共扩增出84条带,其中73条为多态性条带,多态性为81%,平均每个引物扩增出7条(2~11)。群体的多态位点百分率(PPL)达81%,Nei’s基因多样性(H)为0.3187,Shannon信息指数(I)为 0.4782。居群间的基因分化系数较小(Gst)0.1028。以上结果表明:安徽大别山区球孢白僵菌有较高的遗传多样性, 居群间遗传变异较小,居群内表现出较高水平的遗传分化。  相似文献

The recent introduction and rapid spread of the tomato leaf miner Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) throughout Europe, Africa and the Middle East poses a severe threat to future cultivation of tomato and other Solanaceae. Among the best practicable means for effective and sustainable control of this invasive insect pest are entomopathogenic fungi, which can either prevent a further spread of this insect to new areas or keep population densities below an economic threshold level. Here, we report on the efficacy of a commercially available mycoinsecticide based on the entomopathogen Beauveria bassiana (Bals.) Vuill. (Ascomycota: Hypocreales) against all four larval stages of T. absoluta. In bioassays, high mortality rates and significantly reduced longevity of larvae were obvious when larvae had fed for a period of around 15 days on leaves with B. bassiana propagules present on the surface as an epiphyte with mortality values corrected for variance in control mortality of 90–100%. In addition, a second mode of action of B. bassiana against T. absoluta larvae was evident in bioassays in the form of an endophytic establishment of this fungus in treated tomato plants. Longevity of T. absoluta L4 larvae was significantly lower in individuals which had fed on surface‐sterilized tomato leaves obtained from plants treated 18 days before the bioassay with a B. bassiana suspension compared to larvae feeding on control plants treated with water. Corrected mortality reached values between 30% and 50% for all larval instars. Growth of tomato plants was not inhibited due to colonization by endophytic B. bassiana and a weak systemic translocation of fungal propagules in non‐treated leaves was evident in the assays. Accordingly, entomopathogenic fungi like B. bassiana express different modes of action again target insect pests, which is of particular relevance for the design of efficient management strategies for invasive pests like T. absoluta.  相似文献

高黎贡山南段立体气候明显、生境复杂多样, 生物多样性较高, 但目前对其昆虫病原真菌资源的关注较少。为了解高黎贡山南段昆虫病原真菌的物种多样性及分布格局, 本研究在该区域不同海拔生境中进行了昆虫病原真菌资源的调查。于2019-2021年在该区域沿海拔(700-3,100 m)梯度每上升100 m设置样地, 采集土样和受感染昆虫标本, 分离昆虫病原真菌菌株和鉴定物种; 将样区分为5个海拔段(I-V), 用α多样性分析昆虫病原真菌的物种多样性和分布特征, 用β多样性分析其群落结构, 通过Pearson相关性和冗余分析(redundancy analysis, RDA)分析生境生态因子对其物种多样性的影响。结果表明, 从高黎贡山南段共获得昆虫病原真菌菌株2,175株, 隶属于5科20属78种, 其中从土壤样品中获得24种1,331株昆虫病原真菌, 从昆虫样品中获得66种844株昆虫病原真菌。中海拔段III (中山湿性常绿阔叶林)中昆虫病原真菌物种丰富度(49种)和菌株数(766株)最高; 中海拔段II (亚热带雨林)和Ⅲ的Shannon-Wiener多样性指数(分别为2.686和2.677)和Simpson多样性指数(分别为0.884和0.876)最高; 高海拔段V的Simpson优势度指数(0.615)最高; 低海拔段I (干热河谷)的Pielou均匀度指数(0.790)最高。海拔对两种来源(土壤和昆虫)的昆虫病原真菌物种多样性均有显著影响; 来自土壤的昆虫病原真菌受土壤温度的影响较大, 而来自昆虫的昆虫病原真菌受空气湿度的影响较大; 土壤pH值对该区域昆虫病原真菌多样性的影响较小。研究结果表明, 高黎贡山南段的昆虫病原真菌资源丰富, 广泛分布于各海拔段。  相似文献

Four isolates of the entomopathogenic fungus Metarhizium anisopliae were tested for their potential to control the biting midge Culicoides brevitarsis, the principal vector of bluetongue virus in Australia. Adult C. brevitarsis died three to eight days after walking on paper substrate treated with 0.7 g/m2 conidia of any of the isolates, indicating that M. anisopliae has potential as a surface treatment or topical application control strategy. Incorporation of the fungus into freshly excreted cattle dung at rates of between 0.25 and 1 g conidia/kg reduced the emergence of adult midges by up to 98.5% compared to untreated dung indicating that M. anisopliae has the potential to control C. brevitarsis larvae in cattle dung. Three of the isolates produced similar mortality rates on adult and immature C. brevitarsis while the fourth isolate produced lower, but still significant, mortality rates on adult and immature stages.  相似文献

Vine mealybug, Planococcus ficus, is a major pest of grapevine, which is present in at least 39 countries. According to American Vineyard Foundation, P. ficus is in the top ranks among major insect-pests of grapevine. It is the ‘top priority concerns’ by grape growers and a ‘threat to the sustainability of wine industry’ demanding a ‘high priority research’. In Douro vineyards, it is considered as an occasional insect-pest; however, its importance is increasing in some localities. The present study investigates the occurrences of P. ficus-associated fungi. Vine mealybugs were observed in two of the four surveyed farms. Out of the 183 collected mealybugs, 58 were dead of which 25 had symptoms of mycosis and 13 were parasitised. Subculturing cadavers and subsequent pathogenicity test yielded 22 entomopathogenic fungi (EPF) including yeasts. The yeast Meyerozyma (=Pichia) guilliermondii, and the EPF Sarocladium kiliense and Purpureocillium lilacinum were the most abundant, i.e. representing 18.18% (N?=?4), 13.64% (N?=?3) and 13.64% (N?=?3) of the isolates, respectively. Considering biological affinities, fungal families Nectriaceae and Microascaceae had the most similar count-data profiles. To our knowledge, this work reports the first isolations of EPF from vine mealybug worldwide; and Pseudocosmospora rogersonii in Europe and as EPF worldwide. The mortality rate originated by mycoses on P. ficus was significantly higher than by its parasitoids, suggesting that fungi as P. ficus biocontrol agents are relatively more important than considered before. Overall, this report provides new insights into the development of mycoinsecticides and conservation biocontrol strategies for P. ficus pest management.  相似文献

在室内评价了球孢白僵菌对烟蚜茧蜂生命参数及控害效果的影响。分别在烟蚜茧蜂寄生桃蚜后不同时间进行高剂量(1900孢子/mm2)接菌,检测蚜虫感病率和寄生蜂形成的僵蚜率及僵蚜出蜂率。结果表明,球孢白僵菌对僵蚜率和僵蚜出蜂率的影响随接菌时间不同而变化。在烟蚜茧蜂寄生前1d、寄生当天和寄生后3d接菌,蚜虫感病率分别为59.6%、56.2%和34.8%;与对照相比,僵蚜率分别下降94%、59%和47%,僵蚜出蜂率分别减少83%、54%和49%。在寄生后5d或7d接菌,僵蚜率和僵蚜出蜂率不受明显影响,但蚜虫感病率降低到8.2%以下。对蚜尸内白僵菌菌体含量检测表明,随着烟蚜茧蜂寄生后接菌时间的推移,菌体数量迅速下降。寄生蜂寄生后5d或7d接菌,蚜尸内几乎检测不到菌体。直接喷雾接菌烟蚜茧蜂,成蜂寿命缩短4d左右,且81.8%的蜂尸受白僵菌感染。接菌后的寄生蜂对蚜虫寄生率几乎无影响,但寄生蜂在蚜虫体内的存活时间缩短了27.8%。  相似文献

Abstract The sweetpotato whitefly, Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) remains a serious threat to crops worldwide. The damaging B‐biotype is of specific economic concern because it is an effective vector of over 111 viruses from several families, particularly geminiviruses. Bemisia tabaci is regularly intercepted on plants coming into the UK where it is subjected to a policy of eradication. The UK maintains Protective Zone status against this pest. A main pathway of entry of B. tabaci into the Protected Zone involves propagating material, especially Poinsettia (Euphorbia pulcherrima). With increased insecticide resistance continuously being recorded, B. tabaci is becoming more difficult to control/eradicate. Recent research involving both entomopathogenic nematodes and fungi is showing much potential for the development of control programs for this pest. Both the nematode Steinernema feltiae and the fungus Lecanicillium muscarium have been shown to be most effective against second instar B. tabaci. Fine‐tuning of the environmental conditions required has also increased their efficacy. The entomopathogens have also shown a high level of compatibility with chemical insecticides, all increasing their potential to be incorporated into control strategies against B. tabaci.  相似文献

自柑橘木虱Diaphorina citri体上分离虫生真菌,通过形态特征和ITS序列分析,鉴定虫生真菌的种类;通过致病性测定,明确昆虫病原真菌种类。结果发现,自死体柑橘木虱上分离到18个虫生真菌菌株,隶属4种真菌;自活体上分离到985个虫生真菌菌株,隶属25种真菌。致病性测定结果发现,仅4种真菌对柑橘木虱成虫有致病性,包括刀孢蜡蚧菌Lecanicillium psalliotae、球孢白僵菌Beauveria bassiana、爪哇虫草Cordyceps javanica和淡紫紫孢菌Purpureocillium lilacinum,该4种真菌的分生孢子悬浮液(浓度5×10^7个孢子/mL)接种柑橘木虱成虫后10 d的累计死亡率分别为100%、100%、98.89%和43.33%。其中爪哇虫草仅自死体柑橘木虱上分离到,刀孢蜡蚧菌仅在活体上分离到,球孢白僵菌和淡紫紫孢菌在死体和活体上均被分离到。可见,柑橘木虱活体上的虫生真菌种类丰富。生产上应加强保护和利用昆虫病原真菌,提高其对柑橘木虱的自然抑制力。  相似文献

The production of paecilotoxin from various isolates of Paecilomyces lilacinus was studied using three different media and high performance liquid chromatography (HPLC). Alkaline medium was found to be suitable for the production of the toxins. Among 20 strains tested, 19 including four clinical isolates were found to produce the toxins. Production patterns of paecilotoxins were very similar in each strain and the main toxins were A and B.  相似文献

利用黄粉虫Tenebrio molitorL.作为寄主从土壤中诱感并分离昆虫病原真菌。结果显示,从吉林市的18个土样中分离出球孢白僵菌(Beauveria bassiana)、金龟子绿僵菌(Metarhiziumanisopliae)和玫烟色拟青霉(Paecilomyces fumosoroseus)。土样中的昆虫病原真菌检出率为77.78%。这表明利用黄粉虫分离土壤中的昆虫病原真菌是一种简单、有效的方法。  相似文献

Aim:  To examine whether isolates of the entomopathogenic fungus Beauveria bassiana are more closely associated to their summer hosts compared with overwintering hosts, with recently developed molecular tools based on mitochondrial regions. Methods and Results:  Primers for the traditional ITS1‐5·8S‐ITS2 region and two mitochondrial intergenic regions, namely, nad3‐atp9 and atp6‐rns, were used. All amplified products were sequenced, aligned and Neighbour‐Joining (NJ), parsimony and Bayesian phylogenetic inference analyses were performed. The isolates examined were grouped with very good support into three distinct groups, two of them showed geographical correlation, but no clear association to their host. Conclusions:  The mitochondrial intergenic regions used were more informative than the nuclear ITS1‐5·8S‐ITS2 sequences. The sequence variability observed, that allowed the phylogenetic placement of the isolates into distinct groups, depended on the geographical origin of the isolates and can be exploited for designing group‐specific and isolate‐specific primers for their genetic fingerprinting. No clear associations with summer Sunn Pest populations were observed. Significance and Impact of the Study:  Studies on the genetic variability of biocontrol agents like B. bassiana are indispensable for the development of molecular tools for their future monitoring.  相似文献

The influence of six chemical insecticides on growth, sporulation, conidial germination and cuticle-degrading enzyme production by Isaria fumosorosea were investigated under laboratory conditions. Maximum reduction in vegetative growth, sporulation and conidial germination in relation to the control treatment was observed for Chloranthraniliprole whereas Indoxacarb proved to be the safest insecticide causing lowest reduction in these parameters. Chloranthraniliprole, Chlorpyrifos and Chlorfenapyr caused higher reduction in enzyme activities (chitinase, Pr1, Pr2 and lipase) at all three concentrations whereas very low reduction in enzyme activities was caused by Hubendamide+ Avermectin and Indoxacarb when used at 10 µg/ml. The data presented can be used for future recommendations of these insecticides in IPM programmes where I. fumosorosea is an important control agent.  相似文献

根据几种丝状真菌FUS3/KSS1类MAPK的保守序列设计简并引物,从昆虫病原真菌球孢白僵菌中扩增出MAPK基因的部分片段,进而利用YADE法延伸该片段的上、下游邻接序列,获得MAPK基因的全长序列,命名为BbMPK1。用3′RACE扩增出BbMPK1的全长cDNA序列,该序列含有一个1071bp的ORF,编码356个氨基酸的多肽,推测分子量为41.2kDa,等电点为6.61。BbMPK1含有11个MAPK共有的蛋白激酶区域和1个MAPK激酶作用的磷酸化位点区域(TEY),其氨基酸序列与丝状真菌的TMKA、PMK1、CMK1、FMK1和BMP1等MAPK高度同源。系统聚类结果表明,BbMPK1属于酵母FUS3/KSS1类MAPK。Southern杂交表明,BbMPK1在球孢白僵菌基因组中以单拷贝形式存在。RT-PCR分析表明BbMPK1在分生孢子休眠阶段、萌发阶段和菌丝生长时期均表达。研究结果为阐明酵母FUS3/KSS1类MAPK同源基因在球孢白僵菌中的作用奠定了基础。  相似文献
