The adult house fly Musca domestica (L.) (Diptera: Muscidae) can disseminate bacteria from microbe‐rich substrates to areas in which humans and domesticated animals reside. Because bacterial abundance fluctuates widely across substrates, flies encounter and ingest varying amounts of bacteria. This study investigated the dose‐dependent survival of bacteria in house flies. Flies were fed four different ‘doses’ of green fluorescent protein (GFP)‐expressing Escherichia coli (GFP E. coli) (very low, low, medium, high) and survival was determined at 1, 4, 10 and 22 h post‐ingestion by culture and epifluorescent microscopy. Over 22 h, the decline in GFP E. coli was significant in all treatments (P < 0.04) except the very low dose treatment (P = 0.235). Change in survival (ΔS) did not differ between flies fed low and very low doses of bacteria across all time‐points, although ΔS in both treatments differed from that in flies fed high and medium doses of bacteria at several time‐points. At 4, 10 and 22 h, GFP E. coli ΔS significantly differed between medium and high dose‐fed flies. A threshold dose, above which bacteria are detected and destroyed by house flies, may exist and is likely to be immune‐mediated. Understanding dose‐dependent bacterial survival in flies can help in predicting bacteria transmission potential.  相似文献

The sterile insect technique (SIT), based on the principles of population and behavioral ecology, is widely used to suppress or eradicate target pest insect populations. The effectiveness of SIT depends on the ability of released sterile males to mate with and inseminate wild females; however, the use of gamma radiation to induce sterility negatively affects both somatic cells as well as reproductive cells. Consequently, sterilization by irradiation drastically diminishes mating performance over time. It is well known that fractionated‐dose irradiation, in which a sterilizing dose is delivered via a series of smaller irradiations, reduces radiation damage. In the present study, we evaluated the effect of fractionated‐dose irradiation on fertility, longevity, and mating propensity in Cylas formicarius (Summers) (Coleoptera: Brentidae) for 16 days after irradiation. Fractionated‐dose irradiation with 200 Gy induced full sterility regardless of the number of radiation doses. Although the mating propensity of males sterilized by a single 200 Gy dose (the current standard of the Okinawa Prefecture SIT program) was equal to that of non‐irradiated weevils for the first 6 days, the mating propensity of males sterilized by a series of three doses was maintained for at least the first 12 days. These results demonstrated that fractionated‐dose irradiation can be highly advantageous in C. formicarius eradication programs.  相似文献

Low‐dose (≤0.1 Gy) radiation‐induced adaptive responses could protect cells from high‐challenge dose radiation‐induced killing. The protective role is believed to promote the repair of DNA double‐strand breaks (DSBs) that are a severe threat to cell survival. However, it remains unclear which repair pathway, homologous recombination repair (HRR) or non‐homologous end‐joining (NHEJ), is promoted by low‐dose radiation. To address this question, we examined the effects of low‐dose (0.1 Gy) on high‐challenge dose (2–4 Gy) induced killing in NHEJ‐ or HRR‐deficient cell lines. We showed that 0.1 Gy reduced the high‐dose radiation‐induced killing for wild‐type or HRR‐deficient cells, but enhanced the killing for NHEJ‐deficient cells. Interestingly, low‐dose radiation also enhanced the killing for wild‐type cells exposed to high‐challenge dose radiation with high‐linear energy transfer (LET). Because it is known that high‐LET radiation induces an inefficient NHEJ, these results support that the low‐dose radiation‐stimulated protective role in reducing high‐challenge dose (low‐LET)‐induced cell killing might depend on NHEJ. In addition, we showed that low‐dose radiation activated the DNA‐PK catalytic subunit (DNA‐PKcs) and the inhibitor of DNA‐PKcs destroyed the low‐dose radiation‐induced protective role. These results suggest that low‐dose radiation might promote NHEJ through the stimulation of DNA‐PKcs activity and; therefore, increase the resistance of cells to high‐challenge dose radiation‐induced killing. Summary Previous studies by Prehn demonstrated a direct correlation between the dose of carcinogen used for tumor induction and the immunogenicity of the resulting tumors. The purpose of the present study was to determine the role of the host's immune response and the influence of the carcinogen on immune function in this relationship. For that reason, a comparison was made of the immunogenicities of tumors induced with two doses of carcinogen in immunologically normal mice and in mice immunodepressed by adult thymectomy and irradiation. If the direct relationship between dose and immunogenicity demonstrated in normal mice was due to the degree of immunosuppression produced by the carcinogen, this correlation should not be apparent in mice already immunosuppressed. Although there was some increase in the immunogenicity of tumors induced in the immunosuppressed mice, the same relationship between carcinogen dose and immunogenicity was observed in both groups of mice. These results indicate that the degree of immunogenicity of tumors induced with both high and low doses of carcinogen was influenced by immunoselection, but in addition another, non-immunologic factor was significant in the relationship between carcinogen dose and immunogenicity. Pupae of the Queensland fruit fly or Q‐fly Bactrocera tryoni (Froggatt) are irradiated routinely to induce reproductive sterility in adults for use in sterile insect technique programmes. Previous studies suggest that adult sexual performance and survival under nutritional and crowding stress are compromised by the current target dose of radiation for sterilization (70–75 Gy), and that improved mating propensity and survival under stress by irradiated males may be achieved by reducing the target sterilization dose without reducing the level of induced sterility. This raises the question of the amount by which the irradiation dose can be reduced before residual fertility becomes unacceptable. The present study measures the levels of residual fertility in male and female irradiated Q‐flies at different irradiation doses (20, 30, 40, 50, 60 and 70 Gy), and investigates the possibility that fecundity and fertility increase between 10–15 and 30–35 days post emergence. Male flies require a higher dose than females to induce sterility, with no residual fertility found in females irradiated at doses of 50 Gy or above, and no residual fertility found in males irradiated at doses of 60 Gy or above. Irradiated females are more fecund at 30–35 days post emergence than at 10–15 days. However, fertility does not increase between 10 and 15 days post emergence and 30–35 days, even at doses below 50 Gy. The present study shows that there is scope to reduce the target sterilization dose for Q‐flies below that of the current dose range (70–75 Gy) at the same time as retaining an adequate safety margin above radiation doses at which residual fertility can be expected.  相似文献

A model is described in which damage to a single intracellular locus can lead to a tumorigenic transformation. Assuming a large number of independent intracellular loci to be at risk and assuming that damage to a locus sufficient to cause a tumorigenic transformation occurs with probability greater than zero for all doses greater than zero, leads to the use of the Weibull distribution to characterize the probability of a nonspontaneous tumorigenic cellular transformation occurring after exposure to a given dose of carcinogen. The excess lifetime tumor incidence (i.e., the proportion of tumor bearers) above the spontaneous incidence is used as an estimate of the non-spontaneous incidence and is characterized by a tumor incidence function that represents the probability of occurrence of one or more non-spontaneous tumorigenic cellular transformations amongN(D) independent surviving cells per individual, after exposure to a doseD of carcinogen. The tumor incidence function is fitted to published data for the excess tumor incidence after exposure of animals or humans to ionizing radiation and after exposure of animals to chemical carcinogens.  相似文献

Summary Human peripheral blood was treated with ultrasound either before or after irradiation, and chromosome aberrations in lymphocytes of peripheral blood cultures compared to those resulting from an equivalent dose of irradiation given alone.When peripheral blood is sonicated at a high intensity (3 W/cm2) for 10 min after irradiation, there is an increase in aberration frequency as compared to control samples receiving the equivalent radiation dose alone. However, should the blood be sonicated at the same frequency and for the same time period before radiation there is no significant increase in total chromosome aberrations over the irradiated controls. On the contrary a significant decreases occurs in certain classes of aberration.When sonification with a lower intensity (20 mW/cm2) was used in combination with irradiation the reverse effect was noted. Ultrasound administered for 10 min after radiation caused no significant increase in aberrations. On the contrary increasing the period of sonification to one hour resulted in a lowering of all types of aberration, significant in the case of dicentrics and total chromosome aberrations, when compared to irradiation alone. Reversing the order of treatment again resulted in the opposite effect to that achieved with comparable experiments at high intensities of sonification. Ultrasound before radiation did not produce lower breakage rates. Instead, when the period of sonification was increased to one hour, the number of aberrant cells, fragments, and total aberrations rose significantly over controls.It is suggested that sonification produces chemical changes affecting cellular repair systems, which when combined with ionising radiation, results in an increased or decreased repair effect depending on the dose, duration, and order of treatment.Dedicated to Prof. Dr. Felix Mainx on the occasion of his 80th birthday  相似文献

The sterile insect technique relies on sterilization of males using ionizing radiation. Life cycle stage, and the environmental conditions under which irradiation is carried out are crucial to the provision of good‐quality insects. To identify an optimal radiation strategy for Glossina pallidipes Austen, 1903, 13‐day‐old males were irradiated at different doses in a nitrogen atmosphere. The following day the males were mated with 8‐day‐old virgin females. Pupal production of mated females was monitored for 6 weeks, and induced sterility was determined by probit analysis. Survival of the males that mated was also monitored. At least 95% sterility of irradiated males was achieved with a 158 Gy dose in nitrogen and a 125 Gy in air. Irradiation significantly lowered the probability of survival between 30 and 100 days of age (especially flies irradiated in air), but probabilities of survival were similar outside this period for irradiated and unirradiated flies. Exposure of 2‐ or 13‐day‐old males to sterilizing radiation induced similar levels of sterility in both air and nitrogen.  相似文献

Adult mammals have experience‐dependent plasticity in visual system, but it is unclear whether adult insects also have this plasticity after the critical period of visual development. Here, we have established a modified Y‐maze apparatus for investigating experience‐dependent plasticity in Drosophila. Using this setup we demonstrate that flies after the critical period have bidirectional modifications of the phototaxis preference behavior (PPB) induced by visual deprivation and experience: Visual deprivation decreases the preference of flies for visible light, while visual experience exerts the opposite effect. We also found an age‐dependent PPB plasticity induced by visual deprivation. Molecular and cellular studies suggest that the N‐methyl‐ d ‐aspartate receptors (NMDARs) mediate ocular dominance plasticity in visual cortex in mammals, but direct behavioral evidence is lacking. Here, we used the genetic approaches to demonstrate that NMDAR1, which is NMDARs subunit in Drosophila, can mediate PPB plasticity in young and adult flies. These findings provide direct behavioral evidence that NMDAR1 mediates PPB plasticity in Drosophila. Our results suggest that mammals and insects have analogous mechanisms for experience‐dependent plasticity and its regulation by NMDAR signaling.  相似文献

The sterile insect technique (SIT) was developed to eradicate the new world screwworm from the southern United States and Mexico, and became a component of many area‐wide integrated pest management programs, particularly useful in managing tephritid fruit flies. SIT is based on the idea of rearing and sterilizing male pests, originally by ionizing radiation, and then releasing into field, where they compete for and mate with wild females. Mating with sterile males leads to reduced fecundity to lower pest populations. There are concerns with the use and distribution of radioisotopes for SIT programs, which have led to developing X‐ray irradiation protocols to sterilize insects. We considered the possibility that X‐ray irradiation exerts sublethal impacts aside form sterilizing insects. Such effects may not be directly observable, which led us to the hypothesis that X‐ray irradiation in one life stage creates alterations in biological fitness and protein expression in the subsequent stage. We tested our hypothesis by irradiating larvae of Bactrocera dorsalis. There are two major points. One, exposing larvae to X‐ray treatments led to reduced adult emergence, fecundity, fertility, and flight capacity from the corresponding pupae and emerged adults. Two, the X‐ray treatments led to substantial expression changes in 27 pupal proteins. We assorted the 67 spots representing these proteins into three groups, metabolism, development, and structure. Our interpretation is these X‐ray induced changes in biological performance and protein expression indicate their adult counterparts may be disabled in their abilities to successfully compete for and mate wild females in native habitats.  相似文献

The effect of γ‐irradiation on the in vivo‐measured chlorophyll ?uorescence in a pumpkin leaves (Cucurbita pepo) has been investigated. Plants were grown in the same environment, then divided into several groups and irradiated at ambient conditions at small dose levels (up to 13.4 Gy) with 60Co γ‐rays. The post‐irradiation effect on chlorophyll status in the leaves was examined by measuring chlorophyll ?uorescence 2 days (48 h) after exposure. It is undoubtedly found that the value of ?uorescence intensity ratio (FIR) at 690 nm and 735 nm (F690/F735) depends upon the ionizing radiation dose. Even with the smallest dose of 3.35 Gy, ionizing radiation notably altered the ?uorescence spectra of leaves. The spectra difference was manifested by decrease of FIR due to changed chlorophyll luminescence, the possible reason for which could be increase of chlorophyll concentration during the recovery process of the plant. The potential implications of these results for plant physiological status monitoring, as well as for pollution detection and assessment, are discussed in brief. In this study, the effects of gamma irradiation on the resistance of pear fruit against Penicillium expansum, the causal agent of blue mould disease, were investigated. A low dose of gamma irradiation for 14 days increased the disease resistance and firmness of pear fruits. Remarkably, exposure to 200 Gy of gamma irradiation significantly maintained fruit firmness, markedly reduced disease incidence and enhanced the activity of defence‐related enzymes (e.g., β‐1,3‐glucanase, phenylalanine ammonia lyase, peroxidase and polyphenol oxidase) and expression of pathogenesis‐related (PR) genes (e.g., PR‐1, PR‐3 and PR‐4). Therefore, the gamma irradiation‐induced resistance against P. expansum involves both metabolic changes and the induction of expression of defence‐related genes. In addition, scanning electron microscopic analysis revealed that gamma irradiation significantly inhibits the growth of P. expansum. These results suggest that exposure of mature harvested pear fruits to artificial gamma irradiation confers fungal disease resistance; therefore, gamma irradiation represents an important strategy for controlling postharvest diseases in pear fruit.  相似文献

ADSCs (adipose‐derived mesenchymal stem cells) are candidate adult stem cells for regenerative medicine. Notch signalling participates in the differentiation of a heterogeneous ADSC population. We have isolated, human adipose tissue‐derived single‐cell clones using a cloning ring technique and characterized for their stem cell characteristics. The role of Notch signalling in the differentiation capacity of these adipose‐derived single‐cell‐clones has also been investigated. All 14 clones expressed embryonic and mesenchymal stem cell marker genes. These clones could differentiate into both osteogenic and adipogenic lineages. However, the differentiation potential of each clone was different. Low adipogenic clones had significantly higher mRNA expression levels of Notch 2, 3 and 4, Jagged1, as well as Delta1, compared with those of high adipogenic clones. In contrast, no changes in expression of Notch signalling component mRNA between low and high osteogenic clones was found. Notch receptor mRNA expression decreased with the adipogenic differentiation of both low and high adipogenic clones. The γ‐secretase inhibitor, DAPT (N‐[N‐(3,5‐difluorophenacetyl)‐l ‐alanyl]‐(S)‐phenylglycine t‐butyl ester), enhanced adipogenic differentiation. Correspondingly, cells seeded on a Notch ligand (Jagged1) bound surface showed lower intracellular lipid accumulation. These results were noted in both low and high adipogenic clones, indicating that Notch signalling inhibited the adipogenic differentiation of adipose ADSC clones, and could be used to identify an adipogenic susceptible subpopulation for soft‐tissue augmentation application.  相似文献

The effects of elevated UV‐B (280–315 nm) radiation on the long‐term decomposition of Quercus robur leaf litter were assessed at an outdoor facility in the UK by exposing saplings to elevated UV‐B radiation (corresponding to a 30% increase above the ambient level of erythemally weighted UV‐B, equivalent to that resulting from a c. 18% reduction in ozone column) under arrays of cellulose diacetate‐filtered fluorescent UV‐B lamps that also produced UV‐A radiation (315–400 nm). Saplings were also exposed to elevated UV‐A radiation alone under arrays of polyester‐filtered fluorescent lamps and to ambient solar radiation under arrays of nonenergized lamps. After 8 months of irradiation, abscised leaves were placed into litter bags and allowed to decompose in the litter layer of a mixed deciduous woodland for 4.08 years. The dry weight loss of leaf litter from saplings irradiated with elevated UV‐B and UV‐A radiation during growth was 17% greater than that of leaf litter irradiated with elevated UV‐A radiation alone. Annual fractional weight loss of litter (k), and the estimated time taken for 95% of material to decay (3/k) were respectively increased and decreased by 27% for leaf litter exposed during growth to elevated UV‐B and UV‐A radiation, relative to that exposed to UV‐A alone. The present data corroborate those from a previous study indicating that UV‐B radiation applied during growth accelerates the subsequent decomposition of Q. robur leaf litter in soil, but indicate that this effect persists for over four years after abscission.  相似文献

Drosophila melanogaster is attracted to chemicals produced by fermentation and it is abundantly found in rotten fruits. Considering its habitat, the fruit fly is reported to be tolerant to environmental chemicals. Quantitative real‐time polymerase chain reaction was employed to investigate the expression pattern and physiological function of genes putatively involved in chemical detoxification. In quantitative real‐time polymerase chain reaction assays, normalization of target gene expression with internal reference genes is required. These reference genes should be stably expressed during chemical exposure and in chemical‐free conditions. In this study, therefore, we used two programs (geNorm and BestKeeper) to evaluate the expression stability of five reference genes (nd, rpL18, ef1β, hsp22 and tbp) in female adult flies exposed to various concentrations of methanol and ethyl acetate. Four genes (nd, rpL18, ef1β and tbp) were found to be suitable for use as reference genes in methanol‐treated flies and three genes (ef1β, nd, tbp) were found to be suitable for use as reference genes in ethyl acetate‐treated flies. These results suggested that a combination of two genes among these stably expressed genes can be used for accurate normalization of target gene expression in quantitative real‐time polymerase chain reaction‐based determination of gene expression profiles in D. melanogaster treated with both chemicals.  相似文献

Flesh flies of the genus Sarcophaga (Diptera: Sarcophagidae) are carrion‐breeding, necrophagous insects important in medical and veterinary entomology as potential transmitters of pathogens to humans and animals. Our aim was to analyse the diversity of gut‐associated bacteria in wild‐caught larvae and adult flesh flies using culture‐dependent and culture‐independent methods. Analysis of 16S rRNA gene sequences from cultured isolates and clone libraries revealed bacteria affiliated to Proteobacteria, Actinobacteria, Firmicutes and Bacteroidetes in the guts of larval and adult flesh flies. Bacteria cultured from larval and adult flesh fly guts belonged to the genera Acinetobacter, Bacillus, Budvicia, Citrobacter, Dermacoccus, Enterococcus, Ignatzschineria, Lysinibacillus, Myroides, Pasteurella, Proteus, Providencia and Staphylococcus. Phylogenetic analysis showed clone sequences of the genera Aeromonas, Bacillus, Bradyrhizobium, Citrobacter, Clostridium, Corynebacterium, Ignatzschineria, Klebsiella, Pantoea, Propionibacterium, Proteus, Providencia, Serratia, Sporosarcina, Weissella and Wohlfahrtiimonas. Species of clinically significant genera such as Ignatzschineria and Wohlfahrtiimonas spp. were detected in both larvae and adult flesh flies. Sequence analysis of 16S rRNA gene libraries supported culture‐based results and revealed the presence of additional bacterial taxa. This study determined the diversity of gut microbiota in flesh flies, which will bolster the ability to assess microbiological risk associated with the presence of these flies. Drosophila bang‐sensitive (bs) mutants exhibit a stereotypic seizure and paralysis following exposure to mechanical shock. In a physiological preparation, seizures and failures corresponding to the defective behavior are observed in response to high frequency stimulation. The amplitude of the stimulus necessary to produce bs behavior, or seizure threshold, varies with bs mutant and its gene dosage. In many respects, the bs defects are similar to those observed in mammalian seizure disorders. Antiepileptic drugs (AEDs) were administered by feeding to easily shocked2 (eas2), a representative bs mutant. The mean recovery times of treated flies were examined in comparison to control cultures. Some of the drugs administered, including carbamazeprine, ethosuximide, and vigabactrin, had little or no effect on the bs behavior of eas2. Gabapentin, however, showed a reduction in mean recovery time with chronic drug exposure. Phenytoin also had a significant effect on the bs behavior of treated flies. There was a reduction of both mean recovery time and the percentage of flies that displayed bang‐sensitive behavior with both acute and chronic treatment. The adult giant fiber preparation was used to examine the effects of phenytoin physiologically. Treated eas2 flies showed changes in their response to normal stimulation as well as alterations in seizure threshold in response to high frequency stimulation. Gabapentin was also effective against two other bs mutants, bangsenseless1 and slamdanceiso7.8, at strain‐specific concentrations, while phenytoin also reduced bang‐sensitive behaviors in bangsenseless1 in a dose dependent manner. AEDs, therefore, can be used to dissect aspects of bs behavior and this model may be useful in understanding the underlying basis of seizure disorders. © 2003 Wiley Periodicals, Inc.
