Summary Sugar in the medium is considered to be an essential cause for the high production costs of plantlets in conventional, heterotrophic micropropagation. Chlorophyllous explants, shoots, and plantlets in vitro have high photosynthetic ability to develop photoautotrophy, but their photosynthetic activity is restricted largely by the low CO2 concentration in the vessel during the photoperiod and in part by the presence of sugar in the medium. The growth of plantlets in vitro is often greater under photoautotrophic conditions than under heterotrophic conditions, provided that the in vitro environment is properly controlled for promoting photosynthesis. The advantages and disadvantages of photoautotrophic micropropagation are discussed.  相似文献

The daily dynamics of CO2 concentration in the culture vessels and the photoautotrophic or photomixotrophic growth capacity of apple (Malus pumila hybrid MM 106 paradisiaca× Northern Spy) cultures were studied. The photoautotrophic cultures were grown on a sugar-free growth medium and submitted (0S+CO2) or not (0S-CO2) to periodic injections of exogenous CO2. The photomixotrophic cultures were grown in the presence of 30 g dm−3 sucrose, with (30S+CO2) or without (30S-CO2) CO2 enrichment. The photosynthetic photon flux density applied was of 210 ± 5 μmol m−2s−1. In the 0S-CO2 treatment, CO2 showed rather uniform and narrow light-dark fluctuations throughout the culturing cycle. In the 30S-CO2 treatment, the daily ratio between CO2 produced during the dark period and that uptaken during the following light period, was almost always above 1 with the only exception of a few days (from the 5th to the 9th day) when the amount of photosynthesised CO2 was equal to or higher than that produced during dark respiration. The 0S+CO2 cultures needed to be enriched all days with exogenous CO2 to avoid periods of gas deficiency while in 30S+CO2 the CO2 injected the first culturing day was uptaken over 5 d; thereafter, daily injections were necessary. Culture fresh and dry mass, number of newly formed shoots and number of nodes per shoot in 0S+CO2 treatment did not statistically differ from the values obtained with 30S−CO2. The highest growth was observed in 30S+CO2 treatment. The increase in culture fresh mass due to 1 μmol of CO2 added to the culture vessels was 1.54 and 1.36 mg for 30S and 0S respectively, while in terms of dry mass the increase was about 2.5 times higher in the sugar-enriched treatment. CO2 enrichment accounted for 77.3 % and 21.2 % of the final fresh mass in 0S+CO2 and 30S+CO2, respectively. An improved micropropagation protocol has been developed for teak (Tectona grandis). Nodal explants placed on MS medium supplemented with 22.2 M benzylaminopurine and then serially transferred to fresh medium after 12, 24, 48 and 72 h gave maximum culture establishment (76.8%). Establishment was reduced when explants were retained in the initial culture medium longer than 12 h. Explants collected in May showed maximum (76.8%) response. Placement of the explants on MS medium supplemented with 22.2 M benzylaminopurine and 0.57 M indole-3-acetic acid resulted in the maximum average number of shoots. In vitro raised micro shoots were rooted ex vitro by dipping in indole-3-butyric acid (9.8 mM) for 2 min followed by planting in polyethylene pots containing a soil:vermiculite (1:1 v/v) mixture. This treatment resulted in 77.9% survival of the plantlets. They were weaned in a glasshouse and finally moved to an agro-net shade house.  相似文献

Plants of Solanum curtilobum (from high altitude) and Solanum tuberosum (from low altitude) were grown in open-top chambers in a greenhouse at either ambient (AC, 360 µmol mol–1) or ca. twice ambient (EC, 720 µmol mol–1) CO2 concentrations for 30 d. CO2 treatments started at the reproductive stage of the plants. There were similar patterns in the physiological response to CO2 enrichment in the two species. Stomatal conductance was reduced by 59 % in S. tuberosum and by 55 % in S. curtilobum, but such a reduction did not limit the net photosynthetic rate (P N), which was increased by approximately 56 % in S. curtilobum and 53 % in S. tuberosum. The transpiration rate was reduced by 16 % in both potato species while instantaneous transpiration efficiency increased by 80 % in S. tuberosum and 90 % in S. curtilobum. Plants grown under EC showed 36 and 66 % increment in total dry biomass, whereas yields (dry mass of tubers) were increased by 40 and 85 % in S. tuberosum and S. curtilobum, respectively. EC promoted productivity by increasing P N. Thus S. tuberosum, cultivated around the world at low altitudes, and S. curtilobum, endemic of the highland Andes, respond positively to EC during the tuberisation stage.  相似文献

Treatment of intact potato (Solanum tuberosum L., cv. Nevskii) tubers with 24-epibrassinolide (EB) resulted in prolonged deep dormancy, increased production of ethylene and higher contents of free and bound abscisic acid (ABA) in buds. EB at the most efficient concentration 0.021 mg dm–3, applied immediately after tuber harvest, inhibited sprouting by 36 – 38 d, increased ethylene formation after 1 and 7 d of storage by almost 300 and 150%, respectively, and increased the content of both free and bound ABA during the whole period of storage (on average by about 80%). Electron microscopic and morphometric studies showed that EB brings about a decrease in cell volume in tunica and all types of meristems and an increase in the number of vacuoles, accompanied by a decrease in their volume.  相似文献

Chrysanthemum plantlets were cultivated in vitro on media with 2.0, 0.3, or 0 % sucrose, or photoautotrophically without an organic carbon source but with supplementation of the culture vessel atmosphere with 2 % CO2. The photoautotrophically cultivated plantlets showed a better growth and multiplication, higher contents of chlorophyll (Chl) and carotenoids, higher Chl a/b ratio, net photosynthetic rate and ribulose-1,5-bisphosphate carboxylase/oxygenase and phosphoenolpyruvate carboxylase activities than plantlets grown on the medium with sucrose. The effect of storage conditions on the serological activity of two isolates of potato virus YNTN strain (PVYNTN) was studied by ELISA. Purified virus, intact and homogenized infected leaves stored freeze-dried and frozen at various temperatures were tested. Purified virus was the most stable at +4 °C and non-purified virus was best preserved as a freeze-dried leaf homogenate at –20 °C. Population dynamics of Meloidogyne chitwoodi were studied for 2 years in a commercial potato field and microplots. Annual second-stage juvenile (J2) densities peaked at harvest in mid-fall, declined through the winter, and were lowest in early summer. In the field and in one microplot study, population increase displayed trimodal patterns during the 1984 and 1985 seasons. Overwintering nematodes produced egg masses on roots by 600-800 degree-days base 5 C (DD₅) after planting. Second-generation and third-generation eggs hatched by 950-1,100 DD₅ and 1,500-1,600 DD₅, respectively, and J2 densities rapidly increased in the soil. A fourth generation was observed at 2,150 DD₅ in 1985 microplot studies. Tubers were initiated by 450-500 DD₅, but J2 were not observed in the tubers until after the second generation hatched at 988-1,166 DD₅. A second period of tuber invasion was observed when third generation J2 hatched. The regional variation in M. chitwoodi damage on potato may be explained by degree-day accumulation in different potato production regions of the western United States.  相似文献

A temporary immersion system for potato microtuber production was designed using 4-l vessels. This culture technique showed several advantages compared to solid cultures: i.e., three fold increase in shoot length, more internodes per plant and improved vigor. In the tuber induction stage, microtubers can be induced at all plant nodes, indicating that the tuberization is not restricted to specific regions. For both cultivars tested, Desiree and Atlantic, an average of 3.1 and 2.8 tubers per single node cutting was achieved after 9 weeks in culture. The size and weight of the tubers were higher than on solid media. Scale up was performed with cv. Atlantic in 10-l polycarbonate flasks and 12 units were mounted containing 150 single nodal cuttings each. An average of 2.6 tubers per inoculated cutting was obtained, with 1.3 g fresh weight per microtuber. Temporary immersion is a valuable option for potato microtuber production, as well as for shoot production during the planting season. 以云南马铃薯品种‘剑川红’植株1个浆果中的天然实生种子产生的70个单株以及B20[CIP010(♀)×CIP004(♂)]杂交组合后代100个实生苗单株为材料,采用12对SSR引物对自交种和杂交实生群体的遗传差异性进行分析,旨在从后代群体中找到与母本(亲本)在分子水平上表现一致的植株,为种质资源的长期保存提供依据。结果表明:(1)‘剑川红’自交群体的多态性比率为81.6%,比杂交组合B20实生群体的多态性比率72.8%略高,说明2个群体的多态性比率均较高。(2)聚类分析结果显示,自交后代和杂交后代群体的遗传相似系数均较高,变化范围均在0.74~0.96之间,说明2个群体均发生了不同程度的遗传分离,但分离的程度较小,绝大多数条带表现一致。(3)在所有供试材料中,同一浆果中均未发现与‘剑川红’母株在分子水平上表现完全一致的单株。研究认为,在分子水平上寻找完全不分离的实生群体难度非常大,需进一步评价与母株(亲本)在分子水平上相似株系的田间表现,从而确定是否可以通过相近或极相近株系来恢复种源。  相似文献

Summary With the objective of using microtubers for conservation of potato germplasm, the main effects of genotype, abscisic acid (ABA), and sucrose level, and of their interactions on biomass production, microtuberization, microtuber dormancy, and dry matter content, were studied. ABA decreased both microtuber production and microtuber dormancy, whereas higher concentrations (60–80 gl−1) of sucrose promoted biomass production, microtuber production as well as microtuber dry matter content. Microtubers stored under diffused light had longer dormancy than those kept continuously in the dark. Interactions among various factors conditioned the main effects for some characters. In vitro performance of the genotypes studied was related to their known performance under in vivo conditions for most of the characters. Microtubers produced on media devoid of ABA and containing high sucrose concentrations and N6-benzyladenine (44.38 μM) could be stored for 12 mo. under diffused light at 6±1°C.  相似文献

Three clones of Coriandrum sativum L. shoots were obtained from three seedlings and micropropagated alternately on modified MS media containing kinetin only and kinetin plus indolyl-3-acetic acid (IAA). During the first 9 months of culture the shoots possessed the juvenile phenotype after which a sharp transition to mature phenotype occurred. In 15–17 months this was followed by shoot necrosis and decrease in number of shoots in the clones, leading to death of the clones.Conditions of in vitro culture tripled the length of the juvenile period. Mature phase of the shoots was stable in that no reversion to the juvenile phase was observed. Partial rejuvenation of mature shoots took place owing to formation of adventitious shoots in the callus formed at the shoot base. However maturation of such rejuvenated adventitious shoots took place much more rapidly in comparison with micropropagated juvenile shoots derived from seedlings. Reduction of the morphogenic potential of the mature shoots after 15–17 months of subculturing, an increase in number of abnormal shoots and shoot necrosis indicated physiological ageing of the clones.Data presented in the paper provide evidence of the clone ageing phenomenon during prolonged subculture in vitro.  相似文献

Sago, a processed (gelatinized) edible starch, was successfully used as a gelling agent in culture medium. The efficacy of sago-gelled (80 g dm–3) medium was studied in ten potato (Solanum tuberosum L.) genotypes during micropropagation and minimal growth conservation. Sago starch provided a firm gelling surface throughout the entire culture period, and fostered optimum plantlet growth in terms of shoot height, number of nodes per plant, number of leaves and fresh mass. No softening of the sago-gelled medium occurred over prolonged (six months) storage. The study showed that sago starch could be used as a substitute to agar in culture medium to substantially reduce the medium cost.  相似文献

The potassium uptake by potato tuber discs tissues freshly cut and after 24 h of ageing in the presence or not of abscisic acid was investigated. Uptake kinetics revealed a biphasic dependence on external K+ concentrations. At concentration less than 10 mM, uptake was mediated by a saturable component and a linear component became apparent at higher concentrations. At low K+ concentrations (lmM), the capacity of K+ uptake diminished by 2 times after ageing. Treatment of tissues with ABA increased the rate of K+ uptake. In both fresh and aged tissues the uptake was strongly enhanced by fusicoccin and decreased by several metabolic inhibitors and ATPase inhibitors, underlying the active nature of uptake and suggesting the involvement of a plasmalemma H+-ATPase in K+ transport system. Summary Biotechnology has become an important tool to produce plant secondary metabolites and proteases are among them. Although pineapple plants have been found to produce proteases, most of the biotechnological investigations on this crop have been focused on propagation. The procedure involving the use of temporary immersion bioreactors is one of the most outstanding because of its high multiplication rate. We previously recorded specific protease activity in the culture medium during the pre-elongation step of this protocol. Therefore we decided to modify this phase, looking for an increase of protease excretion. Three independent experiments were performed to evaluate the effects of culture duration, and levels of gibberellic acid (GA) and 6-benzyladenine (BA). The following indicators were recorded: shoot fresh mass per bioreactor; and protein concentration, proteolytic activity, and specific protease activity in culture media. As happens in investigations focused on protease production, the specific protease activity was the most important indicator recorded here. It maximized at 21 d of culture. Moreover, GA (4.2 μM) increased specific activity in the culture medium while BA produced a negative effect. Results shown here demonstrate that conditions adquate for propagation purposes (15-d pre-elongation phase; 2.8 μM GA; 2.2 μM BA) are not necessarily adequate for protease excretion.  相似文献

A deviation from usually found characteristics of stomata in Wrightia tomentosa was noted during in vitro propagation. Increase in stomatal frequency in leaves of plants grown in vitro was observed with 29.4 % malformed stomata. The stomata were spherical, wide open, did not close in detached leaves even after 3 h. The leaves exhibited 93.4 % total water loss during 3-h period. Stomatal frequency, percentage of malformed stomata and rate of water loss declined in subsequent rooting phase. Nevertheless, for high survival rate plantlets were hardened under gradually decreasing air humidity either in partially opened glass bottles containing Soilrite™ moistened with 1/4 Murashige and Skoog nutrients or in pots covered with polyethylene bags. The stomatal characteristics of hardened plants were comparable to seedlings. Survival rate was more than 95 %.  相似文献

Pathogenesis of M. chitwoodi associated with potato (Solanum tuberosura cv. Russet Burbank) followed a pattern characteristic of root-knot nematodes. Giant ceils developed in the phloem tissues of roots, stolons, and tubers and appeared to arise by hypertrophy and karyokinesis rather than cellular fusion. Gall formation was a function of parasite density and developed by hypertrophy of cortical cells. Brownish lesions which are symptomatic of tuber infection resulted from lignification of cortical cell walls in contact with egg matrix.  相似文献

Multiple shoots were obtained from nodal and shoot tip segments of 10 to 15-day-old seedlings of Syzygium cuminii L. on Murashige & Skoog (MS) revised medium supplemented with 6-benzyladenine (BA) (0.23–8.90 M) singly or in combination with -naphthaleneacetic acid (NAA), indole-3-acetic acid (IAA) or indole-3-butyric acid (IBA). Excised shoots were placed for root induction on MS medium containing NAA and/or IBA and then transferred to MS basal medium to form complete plantlets. 该研究采用同源克隆与PCR扩增方法,从马铃薯品种‘Desiree’中克隆植物磺肽素受体基因StPSKR1和StPSKR2的全长cDNA,并对其进行生物信息学分析及亚细胞定位分析,为深入研究StPSKR1和StPSKR2基因在马铃薯生长发育和生物胁迫中的作用提供理论依据。结果发现:(1)通过同源克隆与PCR扩增获得StPSKR1和StPSKR2的全长cDNA片段,并将其克隆到pGWB5-GFP载体;测序结果显示这2个基因编码的蛋白质与数据库给定的蛋白质序列保持一致,表明成功克隆到StPSKR1和StPSKR2基因。(2)StPSKR1位于马铃薯1号染色体上,cDNA全长3 042 bp,编码1 013个氨基酸,预测蛋白相对分子质量为112.16 kD,理论等电点6.27;StPSKR2位于7号染色体,cDNA全长3 135 bp,编码1 044个氨基酸,相对分子量为114.99 kD,理论等电点6.19。(3)生物信息学分析显示,StPSKR1和StPSKR2都属于跨膜蛋白。(4)亚细胞定位结果显示,StPSKR1和StPSKR2均定位于细胞膜上。  相似文献

Summary Successful fundamental or basic research, while being stimulated by applied studies, provides the development of new technologies for the benefit of mankind. Photoautotrophic micropropagation or micropropagation using sugar-free medium is no exception from this generalization. The concept of photoautotrophic micropropagation is derived from research that revealed the relatively high photosynthetic abilities of chlorophyllous cultures such as leafy explants and plantlets in vitro. To meet the ever-increasing demand for quality transplants, the scaling-up of photoautotrophic micropropagation systems, for commercialization, has become necessary. This article reviews the recent advancement in the development and utilization of large culture vessels for photoautotrophic micropropagation with special emphasis on the feasibility of the system for the commercial-scale propagation. The review also includes choices for supporting material, ventilation type, planting density, vessel volume, and vessel sterilization procedure, and problems and solutions to achieve uniform growth in a large culture vessel. A case study of the commercial application of a photoautotrophic micropropagation system using large culture vessles, which recently has been established in Kunming, China, is also presented in this article.  相似文献
