The maps of the lampbrush chromosomes of Triturus marmoratus oocytes were constructed on the basis of their lengths and major morphological characters such as giant fusing loops, dense matrix loops, lumpy objects, axial granules, lateral globules and reflected fusions; a nucleolus organizing region occurs subterminally on the right side of chromosome X. — Bivalent I appears morphologically asymmetrical, its two partners being of different lengths and bearing heteromorphic loops and other heterozygous structures: this heteromorphism may indicate that the two partners of bivalent I represent the ZW heterochromosomes of the species. Finally, an autoradiographic study has been performed in order to ascertain the pattern of 3H-uridine incorporation shown by the most typical landmarks and nucleoli.This work was financially supported by C.N.R., Roma.  相似文献

Eleven microsatellites have been characterized for Triturus macedonicus. Nine loci showed different variation patterns in a sample of 40 individuals from a single breeding pond in Zagori province (Greece), with an average number of 4.2 alleles per locus and an expected heterozygosity ranging from 0.1199 to 0.8079. Distinct cross-priming amplification rates were recovered on four additional crested newt species. Two monomorphic T. macedonicus loci were polymorphic in other Triturus species. The microsatellites developed herein could be a useful intraspecific genetic tool to undertake fine-scale population genetic analyses as well as in the study of contact zones between crested newt species.  相似文献

We characterized the adult body form of the crested newt (Triturus cristatus superspecies) and explored its evolution. From seven morphometric traits, we determined that body size, interlimb distance and head width define the body form. None of the morphometric traits showed a phylogenetic signal. Three body‐shape morphotypes (Triturus dobrogicus + T. cristatus, Triturus carnifex + Triturus macedonicus and Triturus karelinii + Triturus arntzeni) and three body‐size morphotypes (T. dobrogicus, T. cristatus and all other crested newts) could be recognized. The ancestral phenotype (a large body with a short trunk and a wide head) characterized T. karelinii and T. arntzeni. Triturus carnifex and T. macedonicus had a somewhat different phenotype (large body and wide head, accompanied by mild body elongation). The most derived phenotype included body size reduction and more pronounced body elongation in T. cristatus and, especially, in T. dobrogicus. Body elongation occurred by trunk lengthening but not head and tail lengthening. Additionally, contrary to other tetrapods, evolutionary axis elongation in crested newts was followed by a decrease in body size. We advocate the hypothesis that ecology drives the evolution of body form in crested newts.  相似文献

Summary Endocrine-like cells (ELC) scattered in the basal part of the ciliated epithelium were identified in the lungs of the newt, Triturus alpestris. These ELC have a clear cytoplasm containing large amounts of secretory vesicles (69–180 nm in diameter), especially in their basal parts, but do not display formaldehyde-induced fluorescence. Larvae of the urodeles, Triturus cristatus, can be taught a simple discrimination task for food reward. The preference established by training is retained by the adult newts after metamorphosis. No significant change in the natural preference of these animals occurs after metamorphosis. Adult animals appear to be more difficult to train than the larvae.  相似文献

Mitotic metaphase chromosomes of cold-treated Triturus cristatus show a characteristic pattern of constrictions, most of which lie close, though not immediately adjacent, to the centromeres. The chromatin in these cold-induced constrictions stains intensely with Giemsa. Cold-treated spermatogonia show spiral structure throughout the metaphase chromatids; the packing of chromatin fibrils is much tighter in the constricted regions than elsewhere, and the gyres in the constricted regions are narrower and of shorter pitch.  相似文献

To increase the number of markers available for study of phylogeny and phylogeography in the newt genus Triturus, we developed and tested 59 primer pairs using three different techniques. Primers were obtained from published sources, by designing exon-primed intron-crossing primers and from randomly cloned anonymous nuclear DNA fragments. Successful polymerase chain reaction products were cloned and sequenced. Five fragments were successfully amplified and sequenced for six species of Triturus: intron 7 of the β-fibrinogen gene (βfibint7), third intron of the calreticulin gene (CalintC), the 11th intron of the α-subunit of the platelet derived growth factor receptor (PDGFRα) and two anonymous markers (Cri1 and Cri4). The average percentage species divergence across all the markers is low (c. 3%), compared to what has been found in mitochondrial DNA (25-30%).  相似文献

This paper reports a detailed electron microscopic study of the ventricular cells in the growing spinal cord during the first two months of tail regeneration in Triturus vulgaris and T. cristatus. In the proliferating ependyma some cells undergo a neural differentiation in order to produce "Cerebro-Spinal-Fluid-Contacting-Neurons (CSFCNs). These round or pear-like cells store dense core vesicles, 80-100 nm in diameter, and show less electron-density than ependymal elements. The stereociliar apparatus is the last cytological differentiation of these elements and after two months of regeneration, synapse-like endings were occasionally found.  相似文献

A reconstruction is presented of the phylogenetic relationships withing Triturus . Two different data sets are used. Protein electrophoretic data were scored for 19 presumptive gene loci in nine species (Rafinski & Arntzen, 1987) and a set of 16 behaviour characters from the courtship of Triturus was scored for the same nine species. When these are analysed in concernt, a single, fully resolved and robust solution is found. Consequently, two subgenera are recognized: Triturus and Palaeotriton , the former including all te medium- and large-bodied species: T. alpestris, T. vittatus, T. marmoratus, T. cristatus and its three former subspecies ( T. carnifex, T. dobrogicus and T. Karelini ); the latter comprising the small-bodied species: T. bascai, T. italicus, T. helveticus, T. vulgaris and T. montandoni . Within the subgenus Triturus, T. alpestris and T. vittatus are sister taxa to the remaining members of the subgenus. Similarly, T. boscai and T. italicus are a monophyletic group, sister to T. vulgaris, T. helveticus and T. montandoni . Alternative solutions may be found on the basis of a quantitative analysis of the behavioural characters employed in this study. An alternative phylogenetic tree is included as an example.  相似文献

Allozymic variation in 22 loci in several Yugoslavian populations of four subspecies of the smooth newt Triturus vulgaris, has been analyzed. The frequency of private alleles and the coefficient of genetic differentiation, FST, give very different indirect estimates of the effective number of migrants per generation, Nm. However, such Nm estimates, in most cases higher than 1, imply that gene flow between populations is large enough as to prevent differentiation by random drift. In the case of T.v. vulgaris, of which sixteen populations amply distributed through Yugoslavia were sampled, there is evidence that frequent extinction and recolonization processes might be responsible for the observed genetic structure. This conclusion has been reached after testing the correlation between genetic, environmental and geographical matrices.  相似文献

Autoradiographic studies combined with digestion tests of incorporated 3H-uridine showed that the peripheral nerve of Triturus contains ribonucleic acid. Localization studies revealed the presence of RNA in the axon, in the myelin and Schwann sheath, and in the Schwann cell body. Similar experiments on nerve separated by transection from its neuronal cell bodies yielded the same results. They showed that RNA of the nerve can be synthesized without the intervention of the neuronal cell body. The results strongly suggest that the radioactive substance, precursor or RNA, is transported inward from the Schwann cell to be deposited in the myelin sheath and axon. The route of passage and the possible sites of origin of the RNA in the nerve are discussed. A significant role is suggested for the Schmidt-Lantermann cleft because of its relations with the adaxonal layer of Schwann cytoplasm and with the myelin leaflets.  相似文献

The cyclic adenosine 3′-5′-monophosphate content of four regenerate stages in the forelimb of the newt, Triturus viridescens, was assayed using the Gilman method and compared to the content in the normal, unamputated, forelimb. The concentration was found to be highest in the earliest stages of regeneration, followed by a sharp drop and then a rise to a plateau approximately that of the unamputated limb. The possibility that cyclic AMP acts as a second messenger for nervous and hormonal influences on regeneration is discussed.  相似文献

The monophyly of European newts of the genus Triturus within the family Salamandridae has for decades rested on presumably homologous behavioral and morphological characters. Molecular data challenge this hypothesis, but the phylogenetic position of Triturus within the Salamandridae has not yet been convincingly resolved. We addressed this issue and the temporal divergence of Triturus within the Salamandridae with novel Bayesian approaches applied to DNA sequence data from three mitochondrial genes (12S, 16S and cytb). We included 38 salamandrid species comprising all 13 recognized species of Triturus and 16 out of 17 salamandrid genera. A clade comprising all the "Newts" can be separated from the "True Salamanders" and Salamandrina clades. Within the "Newts" well-supported clades are: Tylototriton-Pleurodeles, the "New World Newts" (Notophthalmus-Taricha), and the "Modern Eurasian Newts" (Cynops, Pachytriton, Paramesotriton=together the "Modern Asian Newts", Calotriton, Euproctus, Neurergus and Triturus species). We found that Triturus is a non-monophyletic species assemblage, which includes four groups that are themselves monophyletic: (i) the "Large-Bodied Triturus" (six species), (ii) the "Small-Bodied Triturus" (five species), (iii) T. alpestris and (iv) T. vittatus. We estimated that the last common ancestor of Triturus existed around 64 million years ago (mya) while the root of the Salamandridae dates back to 95 mya. This was estimated using a fossil-based molecular dating approach and an explicit framework to select calibration points that least underestimated their corresponding nodes. Using the molecular phylogeny we mapped the evolution of life history and courtship traits in Triturus and found that several Triturus-specific courtship traits evolved independently.  相似文献

Two non-glial phagocytes were found to participate along with ependymoglial cells in Wallerian degeneration of the severed optic nerve of the newt (Triturus viridescens). The first type of non-glial cell (polymorphonuclear phagocyte) was positively identified as a neutrophil and participates in the early stages of degeneration. Cells of this type make a brief appearance, reaching a peak by the second postoperative day (2 p.o.d.), and quickly diminish until few can be found by 4 p.o.d. Neutrophils invade the degenerating optic nerve from surrounding connective tissue spaces, most likely, through channels which penetrate the nerve parenchyma. The second type of non-glial cell is an invading mononuclear phagocyte which exhibits characteristics of microglial cells reported in other vertebrate species. Such cells appear in the nerve much later than the neutrophils and towards the end of Wallerian degeneration (6-10 p.o.d.). Their mode of entry and exit appears to be the same as that reported for neutrophils. The neutrophils and microglial-like, mononuclear phagocytes may serve to supplement the histolytic action of the ependymoglial cells, picking up scattered fragments of degenerating myelin and axons.  相似文献

We assess the role of the Carpathians as an extra‐Mediterranean glacial refugium for the crested newt Triturus cristatus. We combine a multilocus phylogeography (one mitochondrial protein‐coding gene, three nuclear introns, and one major histocompatibility complex gene) with species distribution modelling (projected on current and Last Glacial Maximum climate layers). All genetic markers consistently show extensive genetic variation within and genetic depletion outside the Carpathians. The species distribution model suggests that most of the current range was unsuitable at the Last Glacial Maximum, but a small suitable area remained in the Carpathians. Triturus cristatus dramatically expanded its postglacial range, colonizing much of temperate Eurasia from a glacial refugium in the Carpathians. Sexual selection has traditionally been investigated assuming that male quality is as skewed as patterns of male reproductive success can sometimes be. Recently, female choice has been investigated under the model of genetic compatibility, which assumes that each individual female has her own 'best' mate and there is no overall optimal choice for all females. We investigated female mate choice in the newt species Triturus alpestris, a member of a genus where female choice has been investigated only within the context of the optimal male (female choice for condition-dependent traits). We provided females with two males that differed in one condition-dependent trait (body size) and overall genetic composition. Both male body size and female body size did not influence paternity, but the degree of genetic relatedness between females and potential mates did. Two components of fitness (fecundity and hatching success) did not differ between singly and multiply sired clutches, indicating that females do not employ polyandry as a means of increasing offspring fitness through genetic bet-hedging. Instead, we hypothesize that females may mate initially for fertility assurance, but prefer less-related males as the most genetically compatible mates.  相似文献

The vessels of the forelimb stump and regenerate were perfused with Prussian blue and studied as whole mounts and in histological sections to reveal the condition and disposition of the blood vessels in various stages of forelimb regeneration in the adult newt, Triturus viridescens. The development of the vessels in the regenerate seemed to be comparable in all its essential features to that which has been described for the normal developing limb in urodele, chick and pig embryos. The first signs of regeneration of the vessels are seen during wound healing when fine sprouts appear from the old vessels near the amputation wound. These grow and anastomose, but are limited to the transition region between old and new tissues and avoid the growing blastema during the early stages of regeneration. As the regenerate enlarges into a conical structure vessels invade the proximal part of the growth and avoid the distal regions. It is only during the stages of histogenesis and morphogenesis that vessels grow into more distal regions. The regions of most active enlargement of the early or later regenerate are those most poorly vascularized. These results are discussed against the background of the activity of certain enzymes during regeneration. In the advanced regenerate, preferential channels are consolidated until in the palette and digital stages the pattern of the blood vessels resembles that of the normal limb.  相似文献

The behaviour in groups of both sexes of marbled newts (Triturus marmoratus) from Sintra, Portugal, kept for a few weeks in a large aquarium, is described. Behaviour patterns described for this species and for other species in this genus were recognised. The following behaviours are described for the first time: Walk-over, Land-over, Stand-on, Show, Erect-body, Yawn, Circle, and Dig. Females seem to have a more active role in courtship than is usually considered. The behaviour frequencies for each sex were analysed using simulation statistics. When specimens of the newt Triturus carnifex, under anaesthesia by submersion in a 0.2% chlorbutol solution for 25 min, are isolated in a respiratory chamber at 18 degrees C containing water with only 1.3 ppm of oxygen, they consume the oxygen completely in about 3 hr, but they can stay alive for many more hours and wake up with no apparent exterior consequences. Hypoxia induces rapid onset of hepatic steatosis and melanosis, as well as a controlled haemolytic process involving a pool of red blood cells of the same order of size as that held as a reserve in the spleen by animals in an aerial habitat. At the origin of the phenomena is an intense response by the hypophysis, histologically detectable 1 hr from the onset of treatment and confirmed 2 hr later by a highly significant increase in the plasma thyroidstimulating hormone (TSH) concentration compared with the controls (41.5 +/- 13.7 microU/L vs. 15.5 +/- 6.2; P < 0.005). The thyroid follicles react by reabsorbing their colloid, but instead of an increase in the plasma free T3 and T4 concentrations, fT3 falls significantly (1.5 +/- 0.3 pg/mL vs., the 2.4 +/- 0.7; P < 0.05), whereas fT4 remains stationary (4.0 +/- 0.5 pg/mL vs. 4.6 +/- 0.8; N.S.). After 6 hr, the plasmatic TSH concentration is still higher than in the controls (27.0 +/- 3.0 microU/L vs. 15.5 +/- 6.2; P < 0.05), whereas fT3 and fT4 remain stable (1.5 +/- 0.3 and 4.4 +/- 0.5 pg/mL, respectively). If T3 or T4 labelled with 125I is administered prior to hypoxia, after 6 hr of treatment the radioactivity is found to be limited exclusively to the liver and kidney; the thyroid, gall bladder and gut result negative, and this does not agree with hypotheses of hormone inactivation by deiodination, sulphation or glucuronidation. This apparently peculiar endocrine path has not been observed in previous studies on hypoxia in vertebrates, because the experiments were always designed to analyse plasma hormone levels after at least 24 hr of hypoxia or during chronic treatments, losing the most interesting phases of the endocrine response. The possibility that the hypoxic newt possesses alternative or complementary metabolic pathways to anaerobic glycolysis to sustain steatogenesis and melanogenesis and maintain the same cardiac activity as the controls is briefly discussed.  相似文献
